Seed germination is greatly influenced by both temperature (T) and water potential (ψ) and these factors largely determine germination rate (GR) in the field. Quantitative information about T and ψ effects on seed germination in lemon balm (Melisa officinalis L.) is scarce. The main objective of this study was to quantify seed germination responses of lemon balm to T and ψ, and to determine cardinal temperatures in a laboratory experiment. A segmented model was used to describe the effects of ψ (i.e., T) on GR and other germination parameters. The segmented model estimates were 7.2 °C for base (T b), 28.9 °C for optimum (T o), 40.1 °C for ceiling temperature (T c) and 1.64 physiological days (f o) (equivalent to a GRmax of 0.610 d?1 and a thermal time of 35.6 °C days) to reach 50 % maximum germination in the control (0 MPa) treatment (R 2 = 0.99, RMSE = 0.005 day?1). The inherent maximum rate of germination (days) was calculated by the [GRmax = 1/f o] model. ψ affected cardinal temperatures. From 0 to ?0.76 MPa, when ψ increased, T b was a constant 7.2 °C to ?0.38 MPa and increased linearly to 20.1 °C as ψ decreased. T o and f o increased linearly from 28.9 to 30 °C, and from 1.64 to 5.4 day?1, respectively as ψ decreased. However, there was no signification difference in T o as ψ decreased nor did T c decrease from 40.1 to 35 °C as ψ decreased. T b, T c and GRmax were the sole parameters affected by ψ and could be used to characterize differences between ψ treatments with respect to GR at various Ts. Therefore, the segmented model and its parameters can be used in lemon balm germination simulation models.  相似文献

Behavioural thermoregulation of 3Pieris butterfly species,P. rapae, P. melete andP. napi, was examined in relation to the intensity of solar radiation. To evaluate solar radiation intensity, the temperature (Twr) was measured with a mercury thermometer whose bulb was covered with white cloth and exposed to direct sunlight. On clear days, the diurnal air temperature was between 16 and 28°C. The Twt varied between 18 and 45°C, while the temperature in the shade was under 25°C. When the Twt was under 28°C, the body temperatures (Th) of butterflies closely coincided with it. Butterflies with Tb's under 26°C were resting, while those with Tb's between 26 and 28°C were basking. When Twr was between 28 and 40°C, the butterflies were active and their Tb's were always lower than Twr, never exceeding 36°C, though body temperatures could be artificially elevated easily up to the level of Twr. When Twr exceeded 40°C, butterflies showed species-specific heat-avoiding behaviour.P. rapae, whose habitat resources exist in the sun, intercepted solar radiation by closing the wings over the body.P. melete andP. napi, however, whose main habitat resources exist in the shade, moved into the shade. Strictly speaking, it is concluded that both butterflies, in many cases, leave shaded habitats for sunny habitats to elevate their Tb rather than enter the shaded habitats for heat-avoiding.  相似文献

NMR relaxation times have been used to characterize molecular motion and intermolecular complexes in the aqueous phase of bovine chromaffin granules. Partially relaxed 13C and proton spectra have been obtained at 3 and 25°C. T1 measurements of five protonated carbons on epinephrine (C2, C5, C6 CHOH and NCH3) give a correlation time of 0.15 (10?9) s at 25°C for the catechol ring and methine carbon, while the effective correlation time for the NCH3 group is somewhat shorter due to its internal degree of rotational freedom. Resonances of protonated carbons on the soluble protein chromogranin give very similar corerlation times: 0.20 (10?9) s for the peptide α-carbon and 0.2 (10?9) s for the methylene sidechain carbons of glutamic acid. The correlation time (τR) of ATP was not measured direrctly using 13C T1 data due to the weakness of its spectrum, but its reorinetation appears to be substantially slower than that of epinephrine or chromogranin. This conclusion is based on three observations: (1) the qualitative temperature dependence of T1 for H2 and H8 on the adenine ring places τR for ATP to the right of the T1 minimum, or τR ? 1.0 (10?9) s; (2) 13C resonances of ATP have anomalously low amplitudes compared with epinphrine resonances, a fact that is readily explained only if ATP undergoes substantially slower reorientation; and (3) a comparision of the T1 data on H8 on chromaffin granules and in a dilute aqueous solution, where ρR for ATP cam be measured directly, indicates that τR ~ 1.0 (10?9 s at 25°C in the granules. The relaxation data are consistent with the concept of a storage complex based on electrostatic interaction between a polyion (chromogranin) and its counterious (ATP and epinephrine), in which ATP cross-links cationic sidechains of the protein.

By means of differential scanning calorimetry, effects of systematic series of Group I and VII ions on the phase state of model multibilayer dimyristoylphosphatidylcholine (di(14:0)PC) membranes have been studied at a lipid/ion molar ratio of 3/1. The sign-changing correlations between the ionic radii of cations and temperature shifts of di(14:0)PC phase transition were obtained. For cosmotropic Li+ and Na+, the observed shifts were positive (LiCl: ΔT m = 0.6°C; ΔT p = 1.9°C), whereas chaotropic K+ and Rb+ presence resulted in negative shifts (RbCl: ΔT m = ?0.3°C; ΔT p = ?2.5°C). The anions (Cl?, Br?, I?) showed a similar effect increasing with the ion chaotropicity. An essentially weaker effect of Cs+ as compared to other alkali metal ions (CsCl: ΔT m ≈ 0°C; ΔT p = ?0.1°C) can be one of the reasons of its accumulation in living organisms. Generalization of all available data allowed us to specify some important factors of lipid-ion interactions that should be taken into account in further investigations in this field.  相似文献

Abstract The germination of Sorghum bicolor seeds of 9 genotypes was tested at temperatures between 8°C and 48°C on a thermal gradient plate. Samples were tested from three regions of the panicle expected to differ in temperature during grain filling. Seeds of a tenth genotype, SPV 354, produced in controlled-environment glasshouses at different panicle temperatures, were tested similarly. In addition, the emergence of SPV 354 was measured from planting depths of 2 and 5 cm at mean soil temperatures of 15, 20 and 25°C. Four methods of calculating mean germination rate for the nine genotypes were compared. Germination characters like base, optimum and maximum temperature (Tb, To, Tm), thermal time (θ)and the germination rate at To(Rmax showed only small differences between methods. There was a range of genotypic variation in all characters: Tb 8.5–11.9°C; To, 33.2–37.5°C; Tm, 46.8–49.2°C; θ, 23.4–38.0°Cd; Rmax, 0.69–1.14-d-1. In contrast, mean germinability (G) was between 90% and 100% over the temperature range 13–40°C. Panicle temperature had no effect on any germination character in SPV 354. However, deeper burial increased θ for emergence and decreased G, irrespective of soil temperature except at 5 cm. Increasing panicle temperature, by reducing seed size, reduced G and increased θ by about 10% only at 15°C and 5 cm depth.  相似文献

The physical properties of the plasma membrane of the aquatic phycomycete Blastocladiella emersonii were investigated, in particular the effects of cations on membrane structure. Intact zoospores and lipid extracts were labelled with the spin-labels 5-nitroxystearate (5-NS), 12-nitroxystearate (12-NS), and 2,2,6,6-tetramethylpiperidine-1-oxyl (Tempo). Electron spin resonance spectroscopy indicated a total of three breaks in plots of the hyperfine splitting parameter, 2T|, order parameter, S, and the partition coefficient, f, vs. temperature. The first and third break points (TL and TH) were found to be independent of the external K+, Ca2+, or Mg2+ concentrations. They were similar to the break points found in aqueous dispersions of lipid extracts and correlate well with the temperature limits for zoospore viability. In contrast, the middle break point (TM) was markedly influenced by the external Ca2+ concentration. Ca2+ increased TM from 12°C (no Ca2+ added) to 22°C (10 mM Ca2+), i.e., growth temperature. K+ reversed this Ca2+ effect, downshifting TM from 22°C to 10°C. A comparison of the physico-chemical effects of these ions on the membrane, as revealed by the cation-induced shift in TM, is closely correlated with the temperature dependence and physiological effects of cations on zoospore differentiation. This suggests that cations may modify the physical state of the plasma membrane and be involved in regulating the initial changes during zoospore encystment.  相似文献

The interaction of the surfactant octyl glucoside (OG) with dimyristoylphosphatidylcholine (DMPC), dipalmitoylphosphatidylcholine (DPPC), distearoylphosphatidylcholine (DSPC), and soy bean phosphatidylcholine (soy bean PC) was studied using high-sensitivity titration calorimetry. We determined the partition coefficient of OG between water and lipid bilayers and the transfer enthalpy of the surfactant by addition of lipid vesicles to OG monomers or vice versa. Comparison with the micellization enthalpy of the surfactant gives information on differences in the hydrophobic environment of OG in a liquid-crystalline bilayer or a micelle. The average partition coefficient P in mole fraction units for xe≈0.12–0.2 decreases slightly from 4152 at 27°C to 3479 at 70°C for DMPC and from 4260 to 3879 for soy bean PC, respectively. The transfer enthalpy ΔHT of OG into lipid vesicles is positive at 27°C and negative at 70°C. Its temperature dependence is larger for the incorporation of OG into DMPC than into soy bean PC vesicles. It is concluded that OG in DMPC vesicles is better shielded from water than in soy bean PC vesicles or in micelles. Titration calorimetry was also used to determine the phase boundaries of the coexistence region of mixed vesicles and mixed micelles in the systems OG/DMPC, OG/DPPC, OG/DSPC, and OG/soy bean PC vesicles at 70°C in the liquid-crystalline phase. DMPC and soy bean PC solubilization was also studied at 27°C to investigate the effect of temperature. The effective surfactant to lipid ratios at saturation, Resat, for all PCs studied are in the range between 1.33–1.72 and the ratios at complete solubilization, Resol, are between 1.79–3.06. At 70°C, the Resat values decrease with increasing chain length of the saturated PC. The ratios depend also slightly on temperature and the degree of unsaturation of the fatty acyl chains. For the OG/soy bean PC system, the coexistence range for mixed vesicles and mixed micelles is larger than for the corresponding PCs with saturated chains.  相似文献

A chiral spin crossover iron(II) complex, fac-Λ-[FeII(HLR)3](ClO4)2·EtOH was synthesized and its crystal structures in both the high-spin (HS) and low-spin (LS) states were determined, where HLR denotes 2-methylimidazol-4-yl-methylideneamino-R-(+)-1-methylphenyl. The complex assumes octahedral coordination geometry of N6 donor atoms by three bidentate ligands HLR. The complex exists as the facial-Λ-isomer of fac-Λ-[FeII(HLR)3]2+ of the possible geometrical fac- and mer-isomers and the Δ- and Λ-enantiomorphs. The X-ray structural analyses revealed that the R-form of the ligand (HLR) induces the fac-Λ-isomer of fac-Λ-[FeII(HLR)3]2+ and the S-form of the ligand (HLS) induces the fac-Δ-isomer of fac-Δ-[Fe(HLS)3]2+. The complex fac-Λ-[FeII(HLR)3](ClO4)2·EtOH shows a complete steep spin crossover between the HS and the LS states at T1/2 = 195 K.  相似文献

Bats in hot roosts experience some of the most thermally challenging environments of any endotherms, but little is known about how heat tolerance and evaporative cooling capacity vary among species. We investigated thermoregulation in three sympatric species (Nycteris thebaica, Taphozous mauritianus and Sauromys petrophilus) in a hot, semi-arid environment by measuring body temperature (T b), metabolic rate and evaporative water loss (EWL) at air temperatures (T a) of 10?C42?°C. S. petrophilus was highly heterothermic with no clear thermoneutral zone, and exhibited rapid increases in EWL at high T a to a maximum of 23.7?±?7.4?mg?g?1?h?1 at T a????42?°C, with a concomitant maximum T b of 43.7?±?1.0?°C. T. mauritianus remained largely normothermic at T as below thermoneutrality and increased EWL to 14.7?±?1.3?mg?g?1?h?1 at T a????42?°C, with a maximum T b of 42.9?±?1.6?°C. In N. thebaica, EWL began increasing at lower T a than in either of the other species and reached a maximum of 18.6?±?2.1?mg?g?1?h?1 at T a?=?39.4?°C, with comparatively high maximum T b values of 45.0?±?0.9?°C. Under the conditions of our study, N. thebaica was considerably less heat tolerant than the other two species. Among seven species of bats for which data on T b as well as roost temperatures in comparison to outside T a are available, we found limited evidence for a correlation between overall heat tolerance and the extent to which roosts are buffered from high T a.  相似文献

The influence of reaction media on the racemic temperature (Tr) in the lipase-catalyzed resolution of ketoprofen vinyl ester was investigated. An effective approach to the control of the enzymatic enantioselectivity and the prediction of the increasing tendency was developed based on the Tr influenced by reaction media. The Tr for the resolution catalyzed by Candida rugosa lipase (CRL) was found at 29 °C in aqueous and S-ketoprofen was obtained predominantly at 40 °C. However, CRL showed R-selectivity at 40 °C in diisopropyl ether because the Tr was changed to 56 °C. CRL, lipase from AYS Amano® and Mucor javanicus lipase were further applied for the investigation of the enzymatic enantioselectivity in dioxane, DIPE, isooctane and their mixed media with water. The effects of the reaction medium on Tr could be related to the solvent hydrophobicity, the lipase conformational flexibility and the interaction between the enantiomers and the lipase.  相似文献

A technique based on homogenisation of rapidly frozen tissue was used to investigate the regulation of intracellular pH (pHi) in freshwater and marine fish from diverse environmental temperatures. The following species were held at ambient temperatures of ca. 1°C (Notothenia coriiceps; Antarctica), 5°C (Pleuronectes platessa, Myoxocephalus scorpius; North Sea), and 26°C (Oreochromis niloticus; African lakes). The effects of seasonal acclimatisation to 4, 11 and 18°C were also examined in rainbow trout in the winter, autumn and summer, respectively. Extracellular (whole blood) pH (pHe) did not follow the constant relative alkalinity relationship, where pH+=pOH for any particular temperature, over a range of 1–26°C (overall δpHeT=0.009±0.002 U °C−1; P<0.001), apparently being regulated by ionic fluxes and ventilation. Intracellular pH (pHi) was also regulated independently of pN(=0.5 pK water) in all species of fish examined. The inverse relationship between pHi and environmental temperature gave an overall δpHiT of −0.010±0.001 U °C−1 (for both white and red muscle) and −0.004±0.003 U °C−1 (cardiac muscle). However, between 1 and 11°C δpHiT was much higher (P<0.001), −0.022±0.003 U °C−1 (white muscle) and −0.022±0.004 U °C−1 (red muscle). The possible adaptive roles for these different acid–base responses to environmental temperature variation among tissues and species, and the potential difficulties of estimating pHi, are discussed.  相似文献

A Gram-negative, non-endospore-forming, rod shaped, strictly aerobic, moderately halophilic bacterium, designated strain M9BT, was isolated from the hypersaline lake Aran-Bidgol in Iran. Cells of strain M9BT were found to be motile and produce colonies with an orange-yellow pigment. Growth was determined to occur between 5 and 20 % (w/v) NaCl and the isolate grew optimally at 7.5–10 % (v/w) NaCl. The optimum pH and temperature for growth of the strain were determined to be pH 7.0 and 35 °C, respectively, while it was able to grow over pH and temperature ranges of 6–8 and 25–45 °C, respectively. Phylogenetic analysis based on the comparison of 16S rRNA gene sequences revealed that strain M9BT is a member of the genus Marinobacter. The closest relative to this strain was found to be Marinobacter hydrocarbonoclasticus MBIC 1303T with a similarity level of 97.7 %. DNA–DNA hybridization between the novel isolate and this phylogenetically related species was 13 ± 2 %. The major cellular fatty acids of the isolate were identified as C16:0, C19:1 ω6c, C18:1 ω9c and C16:1 ω9c. The polar lipid pattern of strain M9BT was determined to consist of phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylserine and three phospholipids. Ubiquinone 9 (Q-9) was the only lipoquinone detected. The G+C content of the genomic DNA of this strain was determined to be 58.6 mol%. Phenotypic characteristics, phylogenetic analysis and DNA–DNA relatedness data suggest that this strain represents a novel species of the genus Marinobacter, for which the name Marinobacter persicus sp. nov. is proposed. The type strain of Marinobacter persicus is strain M9BT (=IBRC-M 10445T = CCM 7970T = CECT 7991T = KCTC 23561T).  相似文献

For the high production of phenylalanine by Escherichia coli, we cloned the pheAFR and aroFFR genes (FR = feedback resistant), which encoded chorismate mutase P-prephenate dehydratase and 3-deoxy-d-arabinoheptulosonate-7-phosphate synthase that are feedback inhibition-free as to the endproducts, into a temperature-controllable expression vector composed of the PR and PL promoter and a temperature sensitive repressor, cI857, of bacteriophage lambda. The plasmid obtained was designated as pSY130-14, and the temperature dependency of expression of the cloned genes and of phenylalanine production was investigated at different temperatures between 30 and 42°C using the strain AT2471 harbouring the plasmid. Above 35°C, the pheAFR gene and aroFFR gene expressions, and activities of both enzymes continued to increase up to 42°C. The cell concentration remained constant up to 38.5°C, but started to decrease sharply above 40°C, while the cell concentration of the host strain, AT2471, remained constant at all temperatures tested. The concentration of phenylalanine also depended on the temperature, and the highest production of phenylalanine, 18.6 g l−1, was obtained from glucose at 38.5°C in a 2.5 1 reactor.  相似文献

(1) The thermal capabilities of Australian silvereyes (Zosterops lateralis, 11 g) were investigated both at low and high ambient temperatures (Ta) during the photophase and scotophase. (2). The peak metabolic rate (PMR) induced by helium–oxygen (79:21 %, He–O2) exposure during the photophase was 15.64±1.55 mL O2 g−1 h−1 at an effective lower survival limit Ta (Tpmr) of −39.7±6.1°C. (3). Above the thermoneutral zone (TNZ), metabolic rate, body temperature (Tb), and thermal conductance increased steeply, but they were able to withstand a Ta of 39°C. (4). Our study shows that silvereyes are able to tolerate an impressive range of Ta from about −42°C to at least +39°C and are able to produce enough heat to maintain a thermal difference between Tb and Ta of up to 80°C.  相似文献

Pulsed NMR techniques have been applied to the study of the relaxation parameters characterizing 23Na within frog striated muscle. Experiments were performed at 3°C, 22–24°C and 39°C at a Larmor frequency of 15.7 MHz; at 22–24°C, measurements were obtained both at 15.7 MHz and at 7.85 MHz.As previously reported, only a single spine-lattice relaxation time (T1) was observed, but both slow (T2)I and fast (T2)II components of the spin-spin relaxation time were measured. The effect of temperature (θ) upon (1/T1) was qualitatively similar to that reported for 23Na in free solution; (θ) did not significantly affect (1/T2) over the range of temperatures studied. (1/T2)I, and to a lesser degreee, (1/T1) exhibited a modest inverse dependence of doubtful significance on the Larmor frequency. The data are examined within the framework of a simple specific model; a conservative values in assumed for the quadrupolar coupling constant characterizing immobilized intracellular Na+. Within this framework, the results suggest that the fraction of bound ions whose molecular tumbling is severely restricted does not exceed some few percent of the total sodium population.  相似文献

Experimental warming of forest ecosystems typically stimulates soil respiration (CO2 efflux), but most warming experiments have been conducted in northern latitudes (>?40°N) with relatively young soils. We quantified the influence of experimental warming on soil respiration (RT) in two adjacent forest habitats—a mature, closed canopy forest and a gap where trees were manually removed— on highly-weathered Ultisols of the southeastern U.S. (33°N). Using temperature variation, both natural and induced by experimental warming, we also quantified the temperature sensitivity of RT, defined as the activation energy, EA in the Arrhenius equation. Experimental warming (either + 3 °C or + 5 °C above ambient) did not significantly increase soil respiration rate or cumulative CO2 loss over the 3 years of the experiment, and did not influence the temperature sensitivity of soil respiration, once the influence of natural temperature variation was taken into consideration. Despite the absence of an experimental warming effect, we observed that EA varied on monthly time scales, and varied differently in each habitat. Soil moisture and habitat also influenced RT, but the effects were not consistent, and varied by month. Our results suggest that although RT does depend on temperature, the sensitivity of RT to temperature variation is influenced primarily by factors like microclimate and plant phenology that can change on relatively short (<?monthly) time scales. Thus, using the temperature sensitivity of RT to predict future CO2 losses due to warming is only reasonable if monthly variation in EA is incorporated into models for lower-latitude subtropical ecosystems with highly weathered soils, such as those in this study. Finally, our results suggest that higher temperatures may not enhance RT in highly-weathered, C-poor soils to the extent that has been reported in prior studies of high-latitude soils, which may constrain ecosystem-atmosphere carbon exchanges and feedbacks to the climate system.  相似文献

Time spent in shade by a group of 6 Brahman (B), 6 Shorthorn (S) and 6 Brahman × Hereford—Shorthorn cross (BX) steers was studied in an experimental shade-yard and subsequently at pasture. B sought shade least and S the most in both situations; the cooling effect of time spent in shade on S is reported. Time spent in shade was positively correlated with mean radiant temperature and solar radiation in both situation. The amount of time animals spent in shade in an experimental shade-yard was highly correlated with the amount of time spent in shade and in grazing at pasture.Mean values of rectal temperature (Tre), respiration rate (Rr), skin temperature (Tsk) and sweating rate (Sr) measured immediately prior to release in the shade-yard were calculated. Tre and Rr were positively correlated with time spent in shade, both in the shade-yard and at pasture, and negatively correlated with time spent grazing. Sr was highly positively correlated with time spent in shade and negatively correlated with time spent grazing. The relative contribution of the physiological parameters to time spent in shade and grazing, were estimated by stepwise multiple regression models. Rr was the best predictor of the time spent in shade in both situations. The only significant improvement in variance accounted for was by the inclusion of Sr in the time spent in shade at pasture. Tre was the best predictor of grazing time and the variance accounted for by the model could not be significantly improved by inclusion of any other physiological factor measured. It is suggested that shade use and respiration rate could be simple and effective methods to select heat tolerant animals in the tropics. Both are easily measured over a wide range of situations and are better correlates of thermoregulatory behaviour than the other physiological factors measured.  相似文献

A novel Gram stain-negative, catalase- and oxidase-positive, strictly aerobic bacterium, designated strain H50T, was isolated from an amphioxus breeding zone in the coastal region of the Yellow Sea, China. Cells were observed to be ovoid or short rods, lacked flagella and were found to contain bacteriochlorophyll a. Poly-beta-hydroxybutyrate was found to be accumulated. The temperature range for growth was determined to be 0–37 °C (optimum 28–37 °C). The halotolerance range for growth is 1–15 % NaCl (optimum 2–7 %). The pH range for growth is 6.0–8.0 (optimum 7.0). The major fatty acids were identified as C18:1ω7c and C16:0. The following polar lipids were found to be present: diphosphatidylglycerol, phosphatidylglycerol and a lipid. The predominant respiratory quinone was determined to be Q-10. DNA G+C content was determined to be 57.7 mol%. Strain H50T exhibited the highest 16S rRNA gene sequence similarity to Pelagicola litoralis DSM 18290T (96.1 %), Roseovarius mucosus DSM 17069T (95.8 %) and Roseovarius tolerans DSM 11457T (95.7 %). In the phylogenetic trees, strain H50T was clustered with the genus Roseovarius but not Pelagicola. On the basis of phenotypic, chemotaxonomic and genotypic data, strain H50T is considered to represent a novel species in the genus Roseovarius, for which the name Roseovarius marisflavi sp. nov. is proposed. The type strain is H50T (=CGMCC 1.10799T=JCM 17553T).  相似文献
