Several fixation procedures were studied to determine those most suitable for preservation of seeds during late stages of development and early stages of germination. These are the periods when the tissues are partially dehydrated and are most difficult to fix for electron microscopy. It was found that a prefixation with a mixture of glutaraldehyde, reconstituted formaldehyde (i.e. paraformaldehyde), and acrolein, followed by a postfixation in OsO4 or KMnO4, gives very acceptable images. The results also indicate that glutaraldehyde is necessary for preservation of cell shape, paraformaldehyde for stabilization of reserve proteins, and acrolein for rapid penetration of tissues. Phosphate, cacodylate, and collidine are all acceptable buffers, although collidine gives the most consistent results.  相似文献

In plants and cyanobacteria, the primary step in oxygenic photosynthesis, the light induced charge separation, is driven by two large membrane intrinsic protein complexes, the photosystems I and II. Photosystem I catalyses the light driven electron transfer from plastocyanin/cytochrome c(6) on the lumenal side of the membrane to ferredoxin/flavodoxin at the stromal side by a chain of electron carriers. Photosystem I of Synechococcus elongatus consists of 12 protein subunits, 96 chlorophyll a molecules, 22 carotenoids, three [4Fe4S] clusters and two phylloquinones. Furthermore, it has been discovered that four lipids are intrinsic components of photosystem I. Photosystem I exists as a trimer in the native membrane with a molecular mass of 1068 kDa for the whole complex. The X-ray structure of photosystem I at a resolution of 2.5 A shows the location of the individual subunits and cofactors and provides new information on the protein-cofactor interactions. [P. L-tyrosyl-L-tyrosine crystallizes as a dihydrate in the orthorhombic system, space group C222(1), with a = 12.105(2), b = 12.789(2), c = 24.492(3) A, Z = 8. The structure was solved by direct methods and refined to a final R-value of 0.059 for 1740 observed reflections. The molecule exists as a zwitterion, the peptide unit is trans planar, and the backbone torsion angles correspond to an extended conformation, with psi 1 = 149.4 degrees, phi 2 = -161.2 degrees, psi 2 = 158.3 degrees. The values of the side-chain torsion angles (chi 1, chi 2) are (-58.8 degrees, -63.1 degrees) for the first tyrosine and (-171.7 degrees, -116.5 degrees) for the second. The planes of the aromatic rings are nearly parallel (dihedral angle of 6.1 degrees), and their centers are separated by 10.9 A. The carboxyl plane forms a dihedral angle of 23.8 degrees with the plane of the peptide bond.  相似文献

Seeds from Hypericum species have recently been identified as an interesting source of xanthone derivatives. Extraction of seeds from H. perforatum with MeOH and subsequent concentration via polyamide adsorption yielded a fraction enriched in tetrahydroxyxanthones (THX), which were further semipurified by silica gel chromatography. Based on tentative structure assignment of the two main THX X1 and X2 by NMR a total synthesis was performed for both compounds (THX 1 and 2 , respectively), starting with an Ullmann ether synthesis. The synthesized 1 and 2 were characterized via 1D‐ and 2D‐NMR methods as well as by LC/HR‐MS analysis and proven to be 1,4,6,7‐THX ( 1 ) and 1,2,6,7‐THX ( 2 ). Final structure assignment of the natural Hypericum THX constituents was accomplished by comparing chromatographic and spectroscopic data (LC/MSn and GC/MS) with those of 1 and 2 which were obtained by synthesis. Beyond, investigations into the seeds of H. perforatum and H. tetrapterum by scanning electron microscopy (SEM) provided insights of the structure of the testa (seed coat), which is established by two cell layers, with the lignified sclerenchyma presumably being the depository of the xanthones.  相似文献

Viability of Lettuce Seeds: I. SURVIVAL IN HERMETIC STORAGE   总被引:2,自引:0,他引:2  
Seeds which show orthodox storage characteristics conform toa common pattern of survival over a wide range of storage conditionswhich can be described by a single equation. Two aspects ofthis are that in a constant environment the life-spans of individualseeds in a population are normally distributed, and that thereis negative linear relationship between log moisture contentand log life-span. However, when orthodox seeds are fully hydratedthey survive much longer than would be predicted by extrapolatingfrom lower moisture contents; but the moisture content at whichthe change occurs has not previously been investigated. In this paper the viability of lettuce seeds was examined aftervarious periods of hermetic storage at different moisture contents,temperatures and initial partial pressures of oxygen. At moisturecontents below 15% the pattern of survival is typical of otherorthodox seeds but above this value the responses change infour ways: instead of being deleterious, oxygen becomes beneficialto survival; instead of life-spans being normally distributed,they become skewed; the relative effect of temperature on decreasinglongevity is slightly diminished; and the decrease in eurvivalperiod with increase in moisture content begins to become lessmarked, so that ultimately, above 20% to 30% moisture content,there is no further decrease in longevity. These results, which indicate substantial physiological changesat about 15% moisture content, are discussed in relation tothe hypothesis, postulated by Villiers, that repair and turnovermechanisms are absent from dry seeds but are activated on hydration. From the stem of Barringtonia racemosa two new regioisomeric oleanane-type triterpenes were isolated and characterized as racemosol B (1) and isoracemosol B (2). The structures were elucidated by the use of spectroscopic methods. In addition, the known compounds lupeol, betulinic acid and bartogenic acid were identified. Racemosol B was evaluated for cytotoxicity against a panel of human tumor cell lines, A-549, MDA-MB-231, HeLa, and K-562 and showed moderate but cell-type selective activity.  相似文献

Membrane damage as a result of dehydration was studied in Lotus corniculatus L. cv. Carroll seeds which had been pregerminated for 0, 12, and 24 hours prior to dehydration. During reimbibition, desiccation-tolerant (0- and 12-hour) seeds leaked relatively low quantities of all solutes (total electrolytes, potassium, phosphate, sugar, amino acid, and protein). Desiccation-sensitive (24-hour) seeds leaked higher levels, but evidence of selective permeability remained. Membrane damage was not manifested as a complete removal of the diffusion barrier, although its permeability properties were dramatically altered. Consequently, the plasmalemma was not ruptured or torn by the dehydration treatment, but a more subtle structural alteration occurred.  相似文献

Bioguided-fractionation of the methanol extract of the leaves of Careya arborea led to isolation of a triterpenoid saponin, designated arborenin, and characterized as 3-O-beta-D-glucopyranosyl(1-->2)-beta-D-glucopyranosyl-2 alpha,3beta-dihydroxy-taraxast-20-en-28-oic acid (1), together with desacylescin III (2). The structures were determined on the basis of extensive 2D NMR spectroscopic analysis. The saponin showed in vitro antileishmanial activity against Leishmania donovani (strain AG 83).  相似文献
