To investigate the amount and fate of root-derived C and N, often tracer techniques are used, where plants are labelled with isotopes. In the present study, we evaluated the suitability of the cotton wick method for in situ labelling of peas (Pisum sativum L.) and oats (Avena sativa L.) with 13C and 15N simultaneously. With two greenhouse experiments we investigated how the wick method and aqueous urea and sugar solutions at a variety of concentrations affected plant development. In addition, we investigated the distribution of 13C and 15N in plants from column experiments under outdoor conditions. Solution was taken up by the plant from a small vial connected to the stem by a cotton wick which was passed through a hole in the stem of the plants. Generally, solution uptake varied between individual plants and decreased with increasing sugar concentrations. Below-ground, above-ground and total plant dry matter, were not significantly affected by the wick method and the applied solutions. Mixtures of aqueous glucose solutions at 2 to 4% and aqueous urea solutions at 1% are useful carriers of 13C and 15N. However, in the investigated plants isotopes were not homogeneously distributed among plant parts. Above-ground plant biomass was preferentially enriched with 13C and 15N, whereas below-ground plant biomass was generally lower enriched. Moreover, isotope distribution ratio of individual plants varied considerably, independent of plant part or timing of labelling. This must be taken into account when estimating root-derived C and N. Future studies comparing labelling methods need to present the isotope distribution ratios among plant parts to allow a true comparison of the methods and the evaluation of their suitability for estimating rhizodeposition.  相似文献

Haitian Vodou priests (houngans) and priestesses (mambos) use plant remedies to treat many illnesses, including intestinal parasite infections. The present study screened 12 plants used in Vodou treatments for intestinal parasites to detect in vitro activity against infective-stage larvae of the hookworm Ancylostoma caninum. Water-soluble extracts of 4 of the 12 plants inhibited serum-stimulated feeding by larval A. caninum in a dose-dependent manner. All 4 plant extracts inhibited feeding induced by the muscarinic agonist arecoline, suggesting that these plant extracts may inhibit the insulin-like signaling pathway involved in the recovery and resumption of development of arrested A. caninum larvae. These results indicate that at least some of the plants used in traditional Haitian medicine as vermifuges show activity against nematode physiological processes.  相似文献

In laboratory dual-choice assays females of the cabbage root fly, Delia radicum, prefer for oviposition plants with roots damaged by conspecific larvae to undamaged controls. Cauliflower and kale plants were inoculated with root fly eggs (25 per plant) and the hatching larvae were allowed to feed on the roots for various periods of time (1–17 days). After 4 (cauliflower) or 5 (kale) days of larval feeding the oviposition preference was most pronounced and flies laid between 64% and 68% of their eggs near plants with damaged roots. Later, with increasing damage but fewer surviving, and thus actively feeding, larvae, the magnitude of the preference declined. The preference for plants already damaged by conspecific larvae may contribute to the previously observed aggregated distribution of D. radicum eggs in Brassica crop fields.Further experiments revealed that the sensory cues inducing this oviposition preference originate from the complex consisting of the damaged roots, the surrounding substrate (soil) and associated microbes, rather than from the aerial plant parts. In choice assays using the root-substrate complex of damaged and control plants (aerial parts removed), the observed preference for damaged roots was similar to that found for the entire plant but was more pronounced. The damaged roots alone, compared to control roots, received up to 72% (cauliflower) and 75% (kale) of the eggs. By contrast, surrogate leaves sprayed with methanolic leaf surface extracts from the most preferred plants which had been damaged were not discriminated from surrogate leaved sprayed with extracts of the respective control plants. Analysis of glucosinolate levels in methanolic leaf surface extracts revealed that root damage resulted in enhanced concentrations of indole-glucosinolates on the leaf surface in kale but not in cauliflower. Although indole-glucosinolates are oviposition stimulants for the cabbage root fly, the induced changes were apparently too small to influence oviposition behaviour.  相似文献

1. By selective grazing, invasive grazers can alter macrophyte‐herbivore relationships in shallow freshwater bodies. Evaluating the palatability of macrophytes and understanding the determinants of plant palatability can help predict grazing impact. In no‐choice feeding assays, we tested the palatability of 21 species of freshwater macrophytes to the invasive freshwater apple snail Pomacea canaliculata. 2. Daily feeding rate varied greatly with plant species, ranging from 1.1 to 22% of snail body mass. We assessed six plant properties and examined their correlation with feeding rate. Total nitrogen content was positively related, and C:N ratio and dry matter content (DMC) negatively related, to snail feeding rate. There was no significant correlation between snail feeding rate and plant phenolic content, but the feeding rate on Myriophyllum aquaticum (the plant with the highest phenolic content) was very low. 3. We repeated the feeding assays for 15 species that were not palatable as fresh leaves with reconstituted plant tissues formed by mixing ground up dried leaves with agar. The feeding rate still differed greatly among macrophyte species. Phragmites australis and Vallisneria natans (two species with the highest DMC) were eaten much more as reconstituted plant than as fresh leaves, indicating that structure (i.e. DMC) may be important in their defence against snail herbivory. For two plants (M. aquaticum and Alternanthera philoxeroides) that had moderate amounts of nitrogen/phosphorus but were consumed very little as fresh and reconstituted tissues, we incorporated their extracts into a palatable agar‐based food. The extracts from both species greatly reduced snail feeding rate, indicating the presence of chemical defences in these two species. 4. These results indicated that feeding was affected by several plant traits. The snail favoured plants with a high nitrogen content and avoided plants with a high DMC. Only a few plants possessed chemical feeding deterrents that were effective against this snail. Given the invasive spread of P. canaliculata in Asia, ecologists and managers should consider plant palatability when selecting plants for use in wetland restoration and when predicting the impact of further invasion by this species.  相似文献

为探讨毛竹(Phyllostachys edulis)扩张过程中潜在的化感作用,选择苦槠(Castanopsis sclerophylla(Lindl)Schott)为研究对象。采用水浸提的方法,用毛竹茎叶、枯落物和土壤3部分浸提液浇灌苦槠幼苗,以蒸馏水处理作为对照,对比分析质量浓度分别为0.1、0.05、0.02 g/mL的3个浓度梯度浸提液处理下苦槠幼苗生长指标及各项光合生理指标的差异。结果表明,毛竹浸提液对苦槠幼苗苗高、地径和叶绿素相对含量的影响大体上呈现高浓度抑制低浓度促进的双重浓度效应。不同来源毛竹浸提液的化感效应不尽相同,土壤浸提液对苦槠幼苗生长和光合生理均呈现抑制作用,而茎叶、枯落物浸提液低浓度时为促进作用。毛竹潜在的化感作用,在其扩张过程中可能会干扰森林主要树种更新,从而对森林群落产生威胁。  相似文献

Laboratory experiments were conducted to examine the allelopathic potentials of rice plants (Oryza sativa L.) on seed germination and radicle elongation of Monochoria vaginalis (Burm. f.) Presl var. plantaginea (Roxb.) Solms-Laubat. To compare the relative allelopathic potentials of rice plants at different growth stages, aqueous extracts of dried tissues were obtained from the seeds, husks, and seedlings harvested 10, 20, 40 and 120 days after seeding. When M. vaginalis seeds were incubated in solutions containing aqueous extracts, significant increases in the germination rate were observed for all the aqueous extracts except for the extract of 10 d-old shoots at higher dosages. When partitioned with ethyl acetate, the germination-promoter(s) in the aqueous extracts remained in the aqueous phase while some inhibitors were extracted with ethyl acetate. In contrast, aqueous shoot extracts added to the incubation media at the same dosage for promotion of seed germination inhibited radicle elongation of M. vaginalis, indicating that the sensitivity of the weed varies between these two growth stages.  相似文献

Drought events are predicted to increase due to climate change, yet consequences for plant–insect interactions are only partially understood. Drought‐mediated interactions between herbivores and their host plants are affected by a combination of factors, including characteristics of the affected plant, its associated herbivore and of the prevailing drought. Studying the effect of these factors in combination may provide important insight into plant and herbivore responses to drought. We studied drought effects on plant resistance to two leaf‐chewing herbivores by considering differing growth conditions, plant chemistry and insect responses in concert. We exposed Alliaria petiolata plants from several wild populations to different intensities of intermittent drought stress and quantified drought‐mediated changes in plant chemistry. Simultaneously, we assessed behavior (feeding preference) and performance of two lepidopteran herbivores: Pieris brassicae, a specialist, and Spodoptera littoralis, a generalist. Drought led to lowest concentrations of secondary defense compounds in severely stressed plants, without affecting total nitrogen content. Additionally, drought evoked opposite patterns in feeding preferences (plant palatability) between the herbivore species. Pieris brassicae consumed most of well‐watered plants, while S. littoralis preferred severely drought‐stressed plants. Hence, feeding preferences of S. littoralis reflected changes in plant secondary chemistry. Contrary to their feeding preference, P. brassicae performed better on drought‐stressed than on well‐watered plants, with faster development and higher attained pupal mass (plant suitability). Spodoptera littoralis showed retarded development in all treatments. In conclusion, drought caused plant secondary defense compounds to decrease consistently across all studied plant populations, which evoked contrasting feeding preferences of two herbivore species of the same feeding guild. These results suggest herbivore specificity as a possible explanation for herbivore responses to drought and emphasize the importance of herbivore characteristics such as feeding specialization in understanding and predicting consequences of future drought events.  相似文献

When soybean (Glycine max ) nodulation mutant nts 382 was inoculated with Bradyrhizobium japonicum, these plants nodulated significantly more than the parental type Bragg. Nts 382 seedlings displayed wild-type nodulation pattern when aqueous extracts of young Bragg shoots were applied to the cultural medium together with nutrient solution. Application of young nts 382 shoot extracts to Bragg seedlings did not result in any apparent increase in nodule number. In graft experiments, young shoots from mutant nts 382 induced supernodulation on Bragg root stocks, while no supernodulation was observed when Bragg seedlings were used as scion and grafted onto nts 382 root stocks. Further, the effectiveness of Bragg plant extracts to suppress supernodulation on nts 382 seedlings was found to depend on the age of the plant material used, being very ineffective with extracts from 60-day-old plants. The age effect was not observed in graft experiments. These findings suggest that soybean supernodulation phenomenon may be controlled by one or a few unknown chemicals or plant hormones.  相似文献

The response of the forest cockchafer, Melolontha hippocastani F. (Coleoptera, Scarabaeidae), towards volatiles emitted by different host plants and conspecifics was tested in field experiments during the flight period at dusk. Funnel traps containing artificially damaged leaves from the host plants Carpinus betulus L. and Quercus rubra L., as well as from the non‐host plant Prunus serotina Ehrh. caught significantly more beetles than empty control traps. On the other hand, traps baited with undamaged leaves from Q. rubra did not catch significantly more beetles than empty controls. Leaves from C. betulus damaged by beetle feeding did not attract more beetles than artificially damaged leaves. By use of gas chromatography coupled with electroantennographic detection (GC‐EAD) electrophysiological responses of males and females were shown for 18 typical plant volatiles. A synthetic mixture of selected typical green plant volatiles was also highly attractive in the field. A total of 9982 beetles was caught during the field experiments, among them only 33 females. This suggests that attraction to damaged foliage during flight period at dusk is male‐specific. Field experiments testing the attractiveness of female M. hippocastani towards conspecific males by employing caged beetles and beetle extracts indicated that males of M. hippocastani use a female‐derived sex pheromone for mate location. On wired cages containing either unmated feeding females, or unmated females without access to foliage, or feeding males in combination with extracts from unmated females, significantly more males landed during the flight period than on comparable control cages containing feeding males or male extracts. A possible scenario of mate location in M. hippocastani involving feeding‐induced plant volatiles and a female‐derived sex pheromone is discussed.  相似文献

Choice tests with whole plants and leaf discs indicated that fourth instar Spodoptera exigua (Hübner) (Noctuidae: Amphypyrini) were found more frequently and ate significantly more of the weed Chenopodium murale than the associated crop plant Apium graveolens. In order to explain the preference, plant extracts, plant volatiles, soluble protein concentrations, water contents, and leaf toughness of the two plants were investigated. Bioassays of aqueous methanol (90%) and hexane extracts of leaves on cellulose discs indicated that neither attractants in C. murale nor repellents in A. graveolens could account for the observed preference. No significant difference could be found between the effects of plant volatiles from C. murale, A. graveolens and a control on larval dispersal by S. exigua. Selective feeding for higher levels of proteins also was not a factor, because A. graveolens had nearly twice the soluble protein of C. murale. Water content was approximately 6% higher (by weight) in C. murale than A. graveolens but most polyphagous larvae do not typically show compensatory feeding for water alone. However, the potentially related characteristic of leaf toughness was significantly different, with A. graveolens exhibiting 1.53 times the toughness of C. murale. Studies comparing five types of larval behavior on both plant species showed that the time spent in swallowing behavior was significantly greater on the tougher A. graveolens leaves relative to C. murale. To test the hypothesis that leaf toughness was affecting larval host choice, both plants were finely ground and incorporated into agar blocks. No differences in feeding behavior were detected. The implications of leaf toughness for larval diet and host choice are discussed.  相似文献

A bioassay, based on a membrane sachet technique, has been developed to identify antifeeding compounds affecting the redlegged earth mite, Halotydeus destructor (Acari: Penthaleidae). The method consists of counting H. destructor numbers on membrane sachets in choice experiments, which was quicker and more efficient than weighing the mites. Five per cent aqueous glucose solution was used as a feeding stimulant, with Tween 80® at 5% concentration as a solubilising agent for water-insoluble compounds. (+)-Catechin, rutin, biochanin A, formononetin, chlorogenic acid, and gramine acted as feeding deterrents at 1% concentration. Quercetin (1 %) and azadirachtin (100 ppm) had no significant effect. At lower concentrations (0.01%), compounds showed antifeeding (gramine), phagostimulating (quercetin and chlorogenic acid), or no effects on mite numbers. Dose-dependent deterrent effects of plant extracts were demonstrated with the bioassay, which could be used for other mites.  相似文献

The details of a convenient laboratory press are presented anda procedure by which plant sap is readily obtained is described.The pressing procedure has been shown to release 53–74per cent. of the total plant water as expressed sap. Preliminary observations on the uptake and translocation ofantibiotics by higher plants obtained by examining expressedsap are described. It is shown that the assay of expressed sapprovides a measure of the griseofulvin and chloramphenicol concentrationin leaves of plants grown in solutions of these antibioticsthat does not differ significantly from the value obtained bythe assay of water extracts. Extraction of leaves from plants grown in griseofulvin solutionswith organic solvents demonstrates a much greater antibioticcontent than is indicated by the assay of expressed sap. Toexplain this difference a ‘free’ antibiotic fraction,obtainable in expressed sap or water extracts, and a ‘bound’fraction recovered from the leaf only by organic solvent extractionare postulated.  相似文献

利用滇重楼(Paris polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.)种子外种皮和胚乳的水浸液对白菜(Brassica pekinensis(Lour.)Rupr.)、绿豆(Vigna radiata(Linn.)Wilczak)、小麦(Triticum aestivum L.)种子进行处理,研究滇重楼种子水浸液对3种植物种子萌发、幼苗生长和保护酶活性的影响,并利用GC-MS方法对滇重楼种子内源抑制物的成分进行分析。结果显示,不同浓度滇重楼外种皮、胚乳水浸液对上述3种受体植物的发芽率、苗高、根长及鲜重均产生影响,其作用强度和水浸液的浓度有关,总体上表现出低促高抑的双重浓度效应。滇重楼种子水浸液对白菜的影响作用最强,对绿豆的影响作用最弱,且胚乳水浸液的影响较外种皮强。不同浓度滇重楼种子外种皮和胚乳水浸液均能影响3种植物幼苗体内保护酶的活性,随着水浸液浓度的升高,叶片中超氧化物歧化酶(SOD)、过氧化物酶(POD)活性总体增加,与对照相比差异显著。白菜、小麦过氧化氢酶(CAT)活性减少,与对照相比差异显著;绿豆过氧化氢酶(CAT)活性增加,但与对照相比无显著差异。利用GC-MS方法从胚乳和外种皮水浸液中分别检出8种和2种物质。研究结果表明滇重楼种子中存在内源抑制物质,可能是导致种子休眠的原因;种子水浸液可能通过影响植物幼苗保护酶的活性进而影响其正常生长;有机酸类物质可能是滇重楼种子内源抑制物之一。  相似文献

Electrical penetration graph recordings using direct current (DC-EPGs) were used to analyze aspects of the probing behavior of cowpea aphid,Aphis craccivora Koch, on intact plants and on hexane, ethyl acetate, and methanol extracts of leaves of aphid-resistant (ICV-12) and aphid-susceptible (ICV-1) cultivars of cowpeaVigna unguiculata (L.) Walp. In one set of experiments, recordings were done on plants with or without parafilm wrapping, or on plants painted with raw leaf juice and extracts of the two cultivars. In another study, recordings were done on leaf extracts homogenized in water or in 0.5M sucrose solution and then placed in parafilm membrane sachets. Electrodes were inserted into soil mix for the experiments on potted plants or into extract fractions and raw juice for the membrane feeding experiments on leaf extracts in parafilm sachets. Waveform signals were recorded from resistance fluctuations from interactions between aphids and substrates, and electromotive forces generated within each preparation. ICV-12 plants with or without parafilm wrapping, and ethyl acetate extracts and raw juice of that cultivar significantly (P≤0.05) reduced stylet penetration behavior. Thus, antixenosis as manifested by disruption of aphid stylet activity on host substrates, appeared to be a governing modality of aphid resistance in ICV-12.  相似文献

Alcoholic extracts of 288 of plant materials from 199 plant species have been tested for various biological activities including chemotherapeutic and pharmacological screening. Biological activities, ranging from moderate to good degree, have been observed in 61 plants extracts. Follow up studies have been carried out in these extracts and some of them have shown moderate degree of activities at this Institute. However, none of the extracts was found to be good enough for further development. Results of the present studies, along with chemical investigations on different species of similar genera which were screened earlier, are also discussed.  相似文献

Our laboratory found that silverleaf whitefly (SLW; Bemisia argentifolii Bellows & Perring) feeding alters host plant physiology and chemistry. The SLW induces a number of host plant defenses, including pathogenesis-related (PR) protein accumulation (e.g., chitinases, beta-1,3-glucanases, peroxidases, chitosanases, etc.). Induction of the PR proteins by SLW feeding occurs in various plant species and varieties. The extent and type of induction is dependent on a number of factors that include host plant growing conditions, the length of time the host plant is exposed to SLW feeding, the plant variety, and SLW population densities. The appearance of PR proteins correlates well with reduced infestations of conspecific insect herbivore competitors. Greenhouse and field experiments in which herbivore competitors (cabbage looper, Trichoplusia ni; leaf miner, Liromyza trifolii) were placed on plants previously exposed to SLW feeding demonstrated behavioral differences (oviposition, feeding preferences) and reduced survival rates and development times of these insects. The interaction was asymmetrical, i.e., SLW infestations of plants previously exposed to leaf miners had little or no effect on SLW behavior (oviposition). Induction of plant-defensive proteins by SLW feeding was both local (at the feeding site) and systemic (uninfested leaves distant to the feeding site). There are interactions between diseases such as tomato mottle virus (ToMoV; a geminivirus) and the host plant and SLW. PR proteins were induced in tomato plants infected with ToMoV much as they were via non-viruliferous SLW feeding. The presence of ToMoV in tomato plants significantly increased the number of eggs produced by SLW females. Experiments using tomato plants, powdery mildew (PM), and tobacco mosaic virus (TMV) show that whitefly infestations can affect plant pathogen relationships but the effects vary among pathogen types. Enzyme analyses prior to pathogen inoculation showed that whitefly treatment significantly increased the activities of foliar chitinase and peroxidase. Evaluation of pathogen growth 3 weeks after inoculation showed that whitefly feeding significantly reduced the incidence of PM. However, TMV levels evaluated by ELISA were not significantly affected by whitefly feeding. Six weeks after inoculation with pathogens, the chitinase and peroxidase activities were still elevated in plants initially fed on by whiteflies but continuing pathogen infection had no effect on these enzymes. The possibility that geminivirus infection and/or SLW infestations isolate the host plant for the selected reproduction of the virus and the insect is discussed. Multitrophic cascade effects may contribute to the successful eruptive appearance of SLW on various crops, ranking them as a major pest. They may explain the general observation that when SLW infest a host plant there are few if any competing insect herbivores and pathogens found in the host. However, the results indicate that certain SLW-virus relationships could be mutualistic.  相似文献

This study investigated the possible phytotoxicity induced by Phargmites australis on phenotypic and physiological parameters of recipient plants with identification of major inhibitors in the donor plant. This was achieved using aqueous extracts of different organs and root exudates of P. australis in laboratory and greenhouse experiments with Lactuca sativa as the model test plant. The observed reduced liquid imbibition and altered resource mobilization in seeds of L. sativa, in particular an insufficient carbohydrate supply, demonstrated that the onset of germination might be negatively affected by phytotoxicity. Dose-response studies pointed out that oxidative stress through reactive oxygen species production could potentially cause the observed germination and seedling growth reductions. The osmotic effects by mannitol solution on germination as well as growth and physiology at a level of ?0.57 and ?0.45 bar, respectively, demonstrated that the results from aqueous plant extracts were partially induced by the osmotic potential on and above those levels. Overall, the relative strength of inhibition on measured parameters was the highest in leaf extract, followed by rhizome, root, stem, and inflorescence. Root exudates of P. australis also had negative impacts by reducing germination and growth of plant. High-performance liquid chromatography (HPLC) analysis revealed gallic acid, a potent phytotoxin, as a major compound with an order of leaf >inflorescence>rhizome>root>stem.  相似文献
