Two hundred and fiftyArthrobacter strains were tested in a basal salts-glucose medium for their ability to produce glutamic acid; 50 strains produced small amounts of glutamic acid and alanine, as well as traces of other amino acids. Five biotin-dependent strains produced extraordinarily large amounts of glutamic acid. One of these, which was identified asA. globiformis, was selected for further study. Glutamic acid was only produced by this organism at biotin levels suboptimal for growth; maximal production (0.45 moles of glutamic acid per mole of glucose consumed) occurred at a biotin level of 10–5 µg/ml. Other factors which markedly influenced glutamic acid production were temperature, (NH4)2SO4 concentration, and pH of the growth medium.The taxonomy of glutamic acid-producing bacteria and the correlation between biotin deficiency and glutamic acid production are discussed.  相似文献

Glutamic acid production with gel-entrapped Corynebacterium glutamicum   总被引:1,自引:0,他引:1  
A glutamic acid producing microorganism (Corynebacterium glutamicum) is entrapped in a polyacrylamide gel. These immobilized microorganisms were used to produce glutamic acid in successive batches of fresh medium. Free microorganisms similarly used produced much less glutamic acid under similar conditions.  相似文献

We review recent cross-disciplinary experimental and theoretical investigations on metabolism of the amino acid neurotransmitters glutamic acid and gamma-aminobutyric acid (GABA) in the brain during hypoxia and hypercapnia and their possible role in central control of breathing. The roles of classical modifiers of central chemical drive to breathing (H+ and cholinergic mechanisms) are summarized. A brief perspective on the current widespread interest in GABA and glutamate in central control is given. The basic biochemistry of these amino acids and their roles in ammonia and bicarbonate metabolism are discussed. This review further addresses recent work on central respiratory effects of inhibitory GABA and excitatory glutamate. Current understanding of the sites and mechanisms of action of these amino acids on or near the ventral surface of the medulla is reviewed. We focus particularly on tracer kinetic investigations of glutamatergic and GABAergic mechanisms in hypoxia and hypercapnia and their possible role in the ventilatory response to hypoxia. We conclude with some speculative remarks on the critical importance of these investigations and suggest specific directions of research in central mechanisms of respiratory control.  相似文献

1. The influence of nutrition and chronic lead (Pb) administration on the level of eicosanoids and their precursors was evaluated in serum and liver from chicks fed defined diets. 2. Dietary Pb consistently increased arachidonic acid, the arachidonate/linoleate ratio, and hepatic non-protein sulfhydryl concentration. 3. Hepatic microsomal fatty acid elongation activity was decreased by Pb. 4. Cottonseed oil, calcium and methionine interacted with Pb, affecting liver and serum fatty acid profiles. 5. Tissue levels of leukotriene C4 rose significantly with Pb or correlated with Pb-compromised body weight in only one experiment. Serum prostaglandins E2 and F2 alpha were unchanged by treatments. 6. The results suggest that an increase in precursors is not consistently reflected by tissue levels of eicosanoids.  相似文献

In this paper we describe the successful isolation of glyT-derived GAA suppressors. A glyT+ strain containing glyV55, the gene for a GGA/G-reading, methane sulfonate and hydroxylamine. The cells were plated to select for reversal of auxotrophy due to a trpA(GAA211) mutation. With either mutagen, greater than 85% of the prototrophs obtained were due to suppressors of the trpA mutation. Approximately 12% of the ethyl methane sulfonate-induced and 37% of the hydroxylamine-induced suppressors were shown to be about 25% cotranscucible with metB, as is glyT. The transfer ribonucleic acid from four metB-linked suppressor strains (two from each mutagen) was examined by reversed-phase column (RPC-5) chromatography. In all four cases, the glycyl-transfer ribonucleic acid profile displayed an alteration of glyT transfer ribonucleic acid. All four suppressors responded to GAG in addition to GAA but did not suppress the known mutant codons of several other trpA mutants. Other properties are discussed, along with possible reasons for our success in obtaining these suppressors.  相似文献

In order to assess the role of the large (200 μmol/g) pool of glutamic acid normally found in wild-typeNeurospora crassa conidia, the levels of glutamic acid and other amino acids were manipulated by employing a strain blocked in glutamate synthesis. The double mutant strainen(am)-2;am, which has a lesion in NADP-glutamate dehydrogenase (am) and is auxotrophic for glutamate, was employed in order to obtain conidia with a decreased level of glutamic acid. When cultures of this strain were grown in the presence of 50 or 20 mM glutamate as sole nitrogen source, conidia were obtained with a glutamate pool of 97 and 47 μmole/g, respectively, which represents approximately 57 and 28% of the control values obtained from a prototrophic strain. These observations are interpreted to indicate that conidiation can occur even without the accumulation of the entire pool of glutamate in the conidia. However, when conidia of this double mutant strain were incubated in a minimal medium lacking glutamate, germ tube formation was greatly delayed. Germ tube formation was normal in the presence of glutamate or alanine. During germination of conidia in minimal medium, the levels of glutamate, glutamine, aspartic acid, and γ-aminobutyric acid (GABA) were measured at different times. The results obtained support the view that glutamic acid was catabolized through the GABA pathway during the very early stages of germination.  相似文献

以释放胞内物质为主要目的的细胞破碎条件,采用均质破碎法、超声波法、加酶法和碱性自溶法等对谷氨酸菌作进行破碎试验,并对其相应的工艺参数进行了优化研究。结果表明,使用碱性自溶法在pH10.0,温度70℃,干菌浓度为10%时,自溶40min后蛋白质的释放率接近80%,表明该法对释放胞内物质的作用效果较理想。  相似文献

Twenty-fiveBacillus isolates comprising eight species fromdaddawa, daddawansoya, oglrl, okplhe andugba (all Nigerian fermented plant proteins) all produced glutamic acid in varying concentrations when grown in a basal medium. Glutamic acid release during the course of termentation indaddawa showed a corresponding release pattern by the fermentingBacillus subtilis isolate in the basal medium.

Le profil de relarguage d'acide glutamique pendant la fermentation dedaddawa était semblable à celui de la fermentation par l'isolate deBacillus subtilis dans le milieu de base.
