Scanning electron microscopy (SEM) was used to provide directevidence that shoots produced in vitro from fragmented shootapices of grapevine were adventitious in origin. The effectof temperature on the formation of the adventitious buds wasalso examined using SEM. At 27°C, shoot buds were initiatedby 31 d following fragmentation of the apex, while at 35°Cshoot initiation and multiplication were already well-advancedat only 18 d after the start of culture. At 38°C, apicalfragments quickly browned and died. After 25 d at 35°C,structures resembling inflorescence primordia were also visible.These did not occur in cultures at 27°C. The primordia laterdeveloped into multiple-branched tendrils, structures whichappear to be intermediate between tendrils and inflorescencesand have not been previously described.  相似文献

Correlative Inhibition in the Shoot of Agropyron repens ( L.) Beauv   总被引:1,自引:0,他引:1  
Correlative inhibition was investigated in plants of Agropyronrepens at two temperatures. Reciprocal inhibition ocrurred betweenthe main shoot apex and the outgrowing axillary shoots, withthe balance of inhibition varying with temperature. Apical dominancewas stronger at 10 °C than at 20 °C , but even at 10°C release of apical dominance by decapitation had onlyminor effects on the timing of outgrowth, growth pattern andrate of dry weight aocumulation of the axillary shoots. Dominanceof the main shoot apex by the axillary shoots was stronger at20 °C than at 10 °C. Removal of axillary buds preventeddecline in size and activity of the main shoot apex ard resultedin increased rates of primordium initiation, leaf emergenceand dry weight accumulation in the main shoot. It is suggestedthat a system of reciprocal dominance provides a mechanism formaintaining the characteristic habit of the grass plant andlimits growth in height of vegetative shoots. Irradiating 7-node rhizome fragments of Agropyron repens (L.)Beauv. incubated at 23 °C , with light of increasing far-redto red ratio, increased the incidence of a light-imposed inhibition,which subsequently could be broken by transfer to darkness.Inhibition did not occur when fragments were sprouted in thedark for three days before being exposed to light. Removal ofwavelengths below 600, 690 and 720 nm from tungsten light byfilters, successively lessened the incidence of inhibition comparedwith the unfiltered controls The reassertion of dominance amongst shoots and buds on multi-noderhizome fragments was lessened by light of increasing far-redto red ratios, and dominance was not reasserted under ‘tungstenlights. Wavelengths between 690 and 720 nm appeared to be responsiblefor this effect, but the magnitude of the effects varied seasonally.Dormant (correlatively inhibited) buds on rhizome fragmentswith developed dominant shoots were not reactivated when thefragments were transferred from dark to ’tungsten light.  相似文献

Warm temperatures (35°C day/30°C night) which inhibittuberization in potato (Solanum tuberosum L., cv. Sebago) increasedgibberellin activity in crude extracts from buds, but not frommature leaves, as determined by the lettuce hypocotyl bioassay.Changes in the growth of tubers and stolons indicate the occurrenceof basipetal movement of GA3 applied to the terminal bud ora mature leaf. 14C labelling from GA3 or mevalonic acid injectedjust below the terminal bud was recovered in the lower shoot,stolons and tubers, but the amount transported was greater atcool temperatures (20/15°C). Three-month ‘old’ and ‘fresh’ seedsof Talinum triangulare were subjected to various treatmentsto induce early and rapid germination. Scarification and activated carbon were the most effective treatmentsin improving total germination in fresh seeds, while the 3 and5 per cent thiourea treatments were most effective in improvingtotal germination in old seeds. Activated carbon, scarificationand 5 per cent thiourea treatments enhanced early germinationin both old and fresh seeds. Cumulative percentage germinationwas very high in fresh seeds after scarification or after treatmentwith activated carbon and 5 per cent thiourea, and lowest inseeds treated with 3 per cent thiourea and hot water. In oldseeds, highest cumulative percentage germination was obtainedwith 3 and 5 per cent thiourea treatments and scarification.Generally, higher germination was obtained with fresh seedsthan with old seeds. Partial seed-coat removal and treatment with 5 per cent thiouriaresulted in a higher rate of and cumulative percentage germinationcompared with seeds with the coat partially removed but nottreated with thiourea. Constantly high temperature (34 °C) increased both rateand total germination compared with seeds planted at room temperature(20–23 °C). Treatments that did not induce germinationwere 1 per cent thiourea, H2SO4, cold water soaking at roomtemperature, 6 per cent hydrogen peroxide and soil planting.These treatments effected less than 3 per cent germination. The effects of pruning shoot tops on the synthesis and degradationof ribulose 1,5–Wsphosphate carboxylase (RuBPCase) inleaves on remaining shoots were investigated in mulberry trees.Leucine labelled with 14C was fed to leaf discs from field-grownmulberry trees and 14C incorporation into RuBPCase was examined.Proportion of 14C in RuBPCase to leucine–14C absorbedby leaf discs was remarkably lowered by top-pruning, thoughoccasionally a slight increase was observed soon after pruning.Yet RuBPCase content in leaves on top-pruned shoots became progressivelyhigher than that in leaves on intact shoots. Changes in 14Cin Ru1BPCase in leaves of mulberry saplings previously fed 14CO2were followed. Following 14CO2 feeding, the attainment of themaximal level of 14C in RuBPCase was retarded by top-pruning.The highest level of 14C in RuBPCase was maintained in leaveson top-pruned shoots but decreased in leaves on intact shoots.Specific radioactivity in RuBPCase continued to increase inleaves on top-pruned shoots even after attaining a maximum levelin the control leaves. These facts suggest that the increasein RuBPCase by top-pruning results from a cessation of its degradationfor the remobilization of nitrogen for newly developing leaveson shoot tops. Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. First-year seedlings of Picea sitchensis were induced to setbuds by transferring them from long days (17 h) to short days(10 h at 20 °C, 12 h night at 12 °C). After 20 shortdays, the seedlings stopped growing in height, the shoot apicaldomes began enlarging and the transverse growth rates of theshoot apical dome tissues increased. For about 25 days thereafterprimordia were produced faster within the developing buds thanat the apices of seedlings which had been kept in long daysto prevent bud-set. The morphology of axillary shoots of pea plants (Pisum sativumL. cv. Alaska) was analysed as a function of the position ofthe bud on the plant axis and the stage of plant developmentwhen the buds began to grow. Buds from the three most basalnodes were stimulated to develop by decapitating the main shootwhen buds were still growing (4 d plants), shortly after budsbecame dormant (7 d plants) or after the initiation of floweringon the main shoot (post-flowering plants, about 21 d after sowing).Branch shoots were scored for node of floral initiation (NFI),shoot length, and node of multiple leaflets (NML), a measureof leaf complexity. Shoots that developed spontaneously fromupper nodes (nodes 5-9) on intact post-flowering plants werescored for NFI. NFI for basal buds on 4 and 7 d plants variedas a function of nodal position and ranged from 5 to 6·7nodes. NFI on these plants was not influenced by bud size orwhether a bud was growing or dormant when the plant was decapitated.NFI for shoots derived from basal buds on decapitated post-floweringplants and upper nodes on intact post-flowering plants was about4. Reduced NFI on post-flowering plants may be due to depletionof a cotyledon-derived floral inhibitor. Basal axillary shootson 4 d plants were about 20% longer than those on 7 d plantsand about five times longer than those on post-flowering plants.These differences may be due to depletion of gibberellic acidsfrom the cotyledons. The effects of night-time temperature, leaf-to-air vapour pressuredeficit (VPD) and water stress on CO2 recycling in Bromeliahumilis Jacq. grown under two light and nitrogen regimes wereinvestigated. At night-time temperatures above 30°C, integratednet dark CO2 uptake was severely reduced and CO2 for malatesynthesis was mainly derived from dark respiration. At 35°C,up to 84% of the CO2 liberated by dark respiration was refixedinto malic acid. Below 30 °C only nitrogen deficient plantsshowed significant recycling. No significant differences wereobserved between high and low light grown plants in CO2 recycling.A doubling of leaf-to-air VPD from 7-46 Pa kPa–1 to 15.49Pa kPa–1 resulted in a 2- to 20-fold decrease in leafconductance and about 50 to 65% reduction in integrated darkCO2 uptake. However, about twice as much CO2 was recycled atthe higher VPD as in the lower. Ten days of water stress resultedin 80 to 100% recycling of respiratory CO2. Under high VPD andwater stress treatments, the amount of water potentially savedthrough recycling of CO2 reached 2- to 6-fold of the actualtranspiration. In general, nitrogen deficient plants had higherper cent recycling of respiratory CO2 in response to high night-timetemperature, increased VPD or water stress. Nodulated white clover plants (Trifolium repens L. cv. Huia)were grown for 71 d in flowing nutrient solutions containingN as 10 mmol m–3 NH4NO3, under artificial illumination,with shoots at 20/15°C day/night temperatures and root temperaturereduced decrementally from 20 to 5°C. Root temperatureswere then changed to 3, 7, 9, 11, 13, 17 or 25°C, and theacquisition of N by N2 fixation, NH4+ and NO3 uptakewas measured over 14 d. Shoot specific growth rates (d. wt)doubled with increasing temperature between 7 and 17°C,whilst root specific growth rates showed little response; shoot:root ratios increased with root temperature, and over time at11°C. Net uptake of total N per plant (N2 fixation + NH4++ NO3) over 14 d increased three-fold between 3 and 17°C.The proportion contributed by N2 fixation decreased with increasingtemperature from 51% at 5°C to 18% at 25°C. Uptake ofNH4+ as a proportion of NH4+ + NO3 uptake over 14 d variedlittle (55–62%) with root temperature between 3 and 25°C,although it increased with time at most temperatures. Mean ratesof total N uptake per unit shoot f. wt over 14 d changed littlebetween 9 and 25°C, but decreased progressively with temperaturebelow 9°C, due to the decline in the rates of NH4+ and NO3uptake, even though N2 fixation increased. The results suggestthat N2 fixation in the presence of sustained low concentrationsof NH4+ and NO4 is less sensitive to low root temperaturethan are either NH4+ or NO3 uptake systems. Intact vegetative buds of mulberry (Morus bombycis) attachedto shoot segments were prefrozen, stored in liquid nitrogen,thawed, and the meristems excised for culture on Murashige andSkoog's medium supplemented with 1 mg 1–1 BA to regenerateplants. Either prefreezing at –10 °C or –20°C along with rapid thawing at 37 °C or prefreezingat –20 °C or –30 °C along with slow thawingat 0 °C was a suitable condition for high percentages ofsurvival and shoot regeneration. Potted mulberry plants couldbe finally obtained from the cryopreserved material. The systemusing intact buds as the material for cryopreservation is quitesimple when compared to conventional systems using isolatedmeristems together with cryoprotectants. Cucumber explants including at least part of the cotyledon,a short section of hypocotyl, and the apical bud, are capableof producing multiple axillary buds from the seedling apex andadventitious shoots from the hypocotyl base in a medium whichcontains 2·0 mg dm–3 of kinetin. Removal of theapical bud triples the number of shoots produced from the apexof explants with two intact cotyledons but does not affect shootproduction from explants with some or all of their cotyledonsremoved. The area of intact cotyledon also influences morphogenesis,as explants with both cotyledons removed, failed to produceadventitious shoots from the hypocotyl base. Culture in continuousdarkness entirely prevents shoot development from the explantbase, but has little influence on shoot production from theapex. The influence of endogenous growth regulators and apicaldominance on the morphogenesis of shoots in cucumber seedlingsare discussed. In Cordyline terminalis negatively geotropic leafy shoots and positively geotropic rhizomes develop from single axillary buds on either shoots or rhizomes. All axillary buds have similar morphogenetic potential when released from apical dominance. Experiments in which the orientation of the apex is changed, organs removed, or growth regulators applied indicate that after a rhizome is initiated, it is maintained as a rhizome by auxin originating in the leafy shoot. When auxin levels are lowered by changes in the orientation of the axis or shoot removal, the rhizome apex becomes a shoot apex, which appears to be the stable state of the actively growing apex. Benzyl adenine when applied exogenously to the apex or lateral buds has the same effect as lowering the auxin level. Gibberellic acid has no effect on the apex or lateral buds. High levels of exogenous naphthaleneacetic acid cause bud release and development of rhizomes from previously inhibited axillary buds of the shoot. However, it was not possible to convert a shoot apex into a rhizome apex by auxin treatment. It is suggested that the release of buds on the lower side of horizontal branches and of buds directly above a stem girdle is caused by high auxin levels on the lower side or distal to the girdle. The cell cycle in Silene coeli-rosa shoot apices was measuredto test whether or not early components of the floral stimulus,produced during the 2nd and 3rd long days (LD) of an inductiveLD treatment, resulted in an increase in the duration of G2phase in constant 20–24 h cell cycles. Plants were grownat 20°C in short days (SD) of 8 h light and 16 h darknessfor 28 d (day 0). Starting on day 0, plants were given SD or3 LD each comprising an identical 8 h day and 16 h photo-extension,or 3 dark-interrupted (d.i.) non-inductive LD, interrupted at1700 h of each day with 1 h of darkness. The cell cycle (percentagelabelled mitoses method) and changes in cell number were determinedin the shoot apical meristem. During days 1–2 of the SDtreatment, the cell cycle and mean cell generation time (MCGT)was 18 and 32 h, respectively, giving a growth fraction of 56%.During days 2–3, the cell cycle and MCGT shortened to15 and 23 h, respectively (growth fraction = 65%). During days1–2 of the LD and d.i. LD treatments, cell cycles andMCGTs were 9–10 and 27–29 h, respectively, resultingin smaller growth fractions (about 33%). Thus, shortened cellcycles and altered growth fractions occurred regardless of whetheror not the treatment was inductive. The LD treatment resultedin a marked shortening of G1 and, to a lesser extent, S-phase,whilst G2 remained constant. These changes were consistent withincreases in the proportion of cells in G2 during the photoextensionof each LD which were suppressed during the comparable periodsof the d.i. LD treatment. The latter treatment resulted in eachphase occupying virtually identical proportions of the cellcycle as in the SD treatment. Thus, the unique cell cycle responsesto the initial part of the inductive LD treatment were increasesin the proportion of cells in G2 coupled with G1 and G2 beingof similar duration. Spring wheat was grown to maturity in three growth rooms providing:(a) 18 h of light at 20° C and 6 h of darkness at 15°C (hot long days, HL); (b) 18 h of light at 15° C and 6h of darkness at 15° C (cold long days, CL); (c) 14 h lightat 20° C and 10 h of darkness at 15° C (hot short days,HS). Plants were moved between environments at spikelet initiationand anthesis, so dividing the growth period into three. Meanlengths in days of these periods in the different environmentswere: Period 1: HL 16, CL 18, HS 25; Period 2: HL 42, CL andHS 61; Period3: HL 53, CL 83, HS 63. The length of periods 2and 3 also depended on previous treatments. Grain dry weight was affected by environmental differences inall periods and effects in successive periods were additive.Compared with HL, CL or HS in period I before initiation increasedgrain yield by 6 per cent by increasing grain number per ear,HS in period 2 between initiation and anthesis decreased itby 24 per cent by decreasing the number of grains per spikeletand the proportion of spikelets that contained grain; CL inperiod 2 increased it by 21 per cent by increasing the numberof ears; CL in period 3 after anthesis increased it by 16 percent because leaves died later; HS in period 3 decreased itby 14 per cent because there was less radiation and hence lessphotosynthesis. Dry weight of shoot and root at maturity wasincreased by CL or HS in periods 1 or 2, and increased by CLand decreased by HS in period 3. The effects on final yieldof treatment during periods 1 and 2 were the consequence ofsimilar effects already produced at anthesis, and shoot androot dry weight changed little during period 3. The effects of environmental differences on grain dry weightcould not be explained by differences in leaf-area durationafter anthesis (D3), except that CL in period 3 increased bothyield and D3 but not proportionately, so that, as with HS inthe same period, grain: leaf ratio was decreased. Environmentaldifferences in periods 1 and 2 appeared to affect grain weightby altering the capacity of the ear to accumulate carbohydrates,determined by the number of grains per ear, rather than by alteringthe supply of carbohydrates, determined by D3. There were some interactions between environments in differentperiods which were usually small compared with the main effects.  相似文献

Eleven F1 hybrid genotypes of winter rape (Brassica napus ssp.oleifera) were used in a study of induction and growth of microspore-derivedembryos. Plants of each genotype were grown in controlled environmentsat either a constant 15°C or a constant 20°C, both witha 16 h photoperiod. Equal numbers of buds, approximately 2.5mm in length, containing uninucleate microspores were harvestedfrom each genotype and either pretreated (14 d at 4°C) ordissected immediately after harvest. Anthers were cultured onliquid medium based upon that of Murashige and Skoog (1962)and containing 8% sucrose, 0.5 mg dm–3 naphthylaceticacid and 0.05 mg dm–3 benzylaminopurine. Anthers fromequal samples of buds were incubated at 35°C for 0, 1, 2or 3 d before transfer to 30°C (21 d) and then 25°C.After a total of 42 d incubation, cultures were scored for thepresence of macroscopic embryos (1–2 mm in length) andfor the presence of anthers containing aborted embryoids whichhad not developed further. The results showed first that bud pretreatment completely inhibitedinduction and secondly that anthers of all genotypes had anabsolute requirement for a 35°C treatment (optimal duration2 d) in order to induce embryoid formation. In the great majorityof genotypes plants grown at 15°C provided more productiveanthers than plants grown at 20°C. However, within eachtreatment there were great differences both in the frequencyof anthers showing induced embryoids and of the final yieldof embryos. There was evidence that hybrids with a common parentresponded similarly under certain treatments. This confirmedthe importance of genotypic control for some components of embryoyield. Sporangia of Phytophthora palmivora germinated by either forminggerm tubes or producing zoospores. Two distinct modes of germ-tubedevelopment have been described. Sporangia in distilled waterformed zoospores at 10–34°C with an optimum at 22°Cbut germinated by means of germ tubes at 30 and 34°C only.Zoospore formation was inhibited to varying degrees by cocoapod extract, I.0 per cent (w/v) peptone and yeast extract, 100–500µg1-1 thiamine, and by very low concentrations of severalamino acids, carbohydrates, and inorganic salts. The development of axillary buds, terminal buds, and the shoots extended from them was studied inHydrangea macrophylla. The upper and lower parts in a nonflower-bearing shoot are discernible; the preformed part of a shoot develops into the lower part and the neoformed part into the upper part (Zhou and Hare, 1988). These two part are formed by the different degrees of internode elongation at early and late phases during a growth season, respectively. Leaf pairs in the neoformed part of the shoot are initiated successively with a plastochron of 5–20 days after the bud burst in spring. The upper axillary buds are initiated at approximately the same intervals as those of leaf pairs, but 10–30 days later than their subtending leaves. Changes in numbers of leaf pairs and in lengths of successive axillary buds show a pattern similar to the changes in internode lengths of the shoot at the mature stage. The uppermost axillary buds of the flower-bearing shoot often begin extending into new lateral shoots when the flowering phase has ended. The secondary buds in terminal and lower axillary buds are initiated and developed in succession during the late phase of the growth season. Internode elongation seems to be important in determining the degrees of development of the axillary buds. Pattern of shoot elongation is suggested to be relatively primitive. Significances of apical dominance and environmental conditions to shoot development are discussed.  相似文献

Shoot and root growth rate, carbohydrate accumulation (includingfructan), reducing sugar content and dry matter percentage weremeasured in six wheat cultivars, ranging from winter to springtypes, grown at either 5 or 25 °C. At 5 °C (comparedwith 25 °C), the relative growth rate (RGR) of shoots wassimilarly reduced in all cultivars, but the RGR of shoots wasmore affected in winter wheats. This difference resulted insmaller root:shoot ratios than in spring wheats, which alsodeveloped more first-order lateral roots. A direct relationshipbetween carbohydrate accumulation at low temperatures and reductionin root growth was established.
