The long-term (10–30 day) continuous recording of valve movements in the mussel Mytilus galloprovincialis was carried out in the laboratory under nearly natural conditions. Fourier analysis revealed the circadian (close to the diurnal) rhythm of the valve activity, the phase of which is readily shifted by a shift in the beginning of the daytime, since the light regime is one of the main factors determining the circadian rhythm. The circadian rhythm was manifested in the daily dynamics of mussel valve activity: in the daytime, mussels hold their valves closed more often than at night. This behavior may be a protective response, namely the “shadow reflex”: mussels close their valves upon a sudden decrease in illumination, thus protecting themselves from possible predators. Circadian activity can mask a mussel’s response to environmental pollution; therefore, regular valve closure should be taken into account in early warning systems such as “MusselMonitor®” with a correction for the season of the year, time of day, and other factors. 相似文献

The European bitterling, Rhodeus amarus, is a non-indigenous fish species in British fresh waters. It lays its eggs in unionid mussels which themselves are vulnerable
to fouling by the non-indigenous zebra mussel, Dreissena polymorpha. Observations from an unmanipulated natural system showed that only 27% of zebra mussel-fouled Unio pictorum hosted bitterling, while 47% of unfouled U. pictorum hosted bitterling. We conducted a field experiment in the River Great Ouse catchment, Cambridgeshire, England in May–June
2007 and 2008 to quantify the impact of zebra mussels on bitterling load in host mussels. Zebra mussel-fouled unionids were
significantly less likely to host bitterling than unfouled unionids. The number of unionids hosting bitterling did not differ
significantly whether the zebra mussels fouling the unionid were alive or dead. Bitterling appeared to discriminate against
zebra mussel-fouled unionids less as the 2007 breeding season advanced, potentially because preferred unfouled unionids had
a higher bitterling load, and were therefore relatively lower quality hosts than at the start of the breeding season. 相似文献

We studied the population genetics of two antimicrobial peptide (AMP) loci, called Mytilin B and Mytilus galloprovincialis defensin 2 (MGD2), in the secondary contact mosaic hybrid zone between Mytilus edulis and M. galloprovincialis. The isolation period between the two species was estimated to be ~1 million years (range, 0.5 million to 2 million years)
long. During this period, coevolution between microbes and the immune system has likely occurred. The secondary contact, which
would date back to ~25,000 (0–200,000) years, recently allowed these coadaptations to be rearranged through hybridization.
Distinctive polymorphisms were uncovered in coding sequences of the two AMP loci such as insertion/deletion of codons or bisubstituted
codons. Very low levels of differentiation were observed between populations of the two species at both loci, while other
nuclear loci often showed marked structure among the same samples. The absence of population differentiation proved to be
the consequence of secondary introgression of highly divergent alleles. While only a few recombinants were observed at the
Mytilin B locus, the MGD2 locus showed a high intragenic recombination rate, which increased in the exon coding for the mature peptide. In addition,
standard neutrality tests revealed significant deviations from the mutation-drift equilibrium at both loci. These results
suggest that either balancing or directional selection is likely to play a role in the evolution of the two AMPs and introgression
would be adaptive. However, evidence accumulated at the Mytilin B locus allows neither for identification of the direction of selection nor for any conclusions on whether selection acted
directly on the antimicrobial peptide itself. At the MGD2 locus, a spatial variation of polymorphism patterns along the sequence suggests that selection was direct, although the precise
nature of the selection (directional vs. balancing) remains unclear. This study concurs with previous reports of an effect
of slight selection on AMP genes evolution in other invertebrates, although selection does not necessarily act on the mature
peptides.

Total arsenic concentration in the edible part of mussels Mytilus galloprovincialis was evaluated seasonally in the coastal area of Rijeka Bay (North Adriatic Sea, Croatia). Sampling stations were located close to the City of Bakar with no industrial facilities (site 1), in the vicinity of the oil refinery and oil thermoelectric power plant (Urinj, site 2), and 4 miles away from the Plomin coal thermoelectric power plant (Brse village, site 3). Additionally, the concentration of arsenic in the tail muscle of the lobster Nephrops norvegicus, collected in Rijeka Bay, was studied. During winter at sites 2 and 3, the total arsenic in the edible part of the mussels was 16.4 mg As/kg FW (FW=fresh weight) and 4.38 mg As/kg FW, respectively, and increased during springtime at site 2 (6.5 mg As/kg FW) compared to the rest of the year, when individual total arsenic concentration at all sites ranged from 1.7 to 3.7 mg As/kg FW. In the winter (sites 2 and 3) and springtime (site 2) there was no correlation between the length of the mussel shell and the arsenic concentration in the edible part of the mussels. In the other seasons, at sites 1, 2 and 3, there was a correlation between arsenic in the edible part of mussels and shell length in most cases (correlation coefficients r varied from 0.64 to 0.85; P <0.05 to P <0.01). Correlation between shell length (in the narrow range of shell lengths from 3.4 to 5.0 cm) and arsenic in the edible part of the mussels shows linearity with a high regression coefficient ( r =0.914; P <0.001). The increase of arsenic in the mussels during winter and spring was suggested at least partially as a result of a low nutritional status, i.e. reduced weight of the mussels edible part during winter. In addition, a linear relationship was found between body length and arsenic concentration in the tail muscle (mean 17.11±4.48 mg As/kg FW) of the Norway lobster.Communicated by H.-D. Franke 相似文献

Fluxes of dissolved oxygen and ammonium across the water sediment interface were measured in a control and in an experimental
area farmed with the clam Tapes philippinarum. Young clams were seeded in March 2003 at mean (~500 ind m−2) and high (~1500 ind m−2) densities in a sandy area (2100 m2) of the Sacca di Goro Lagoon, Italy. Approximately every two months, until October 2003, intact sediment cores were collected
and incubated in the light and in the dark and surface sediments (0–2 cm) were analysed for organic matter and nitrogen content.
Clams farming induced pronounced changes in sediment characteristics and metabolism. Oxygen consumption and ammonium production
at the high density area were, on average, 3 to 4 and 1.9 to 4.9 folds higher than those measured in the control field respectively;
rates were positively correlated with clams biomass. Experimental fields resulted “Net and Total Heterotrophyc” in 3 out of
4 sampling dates and clams were the major factor shifting the benthic system towards this status. In only one occasion the
appearance of the macroalgae Ulva spp. pushed the system rapidly towards hyperautotrophic conditions. Our results indicated that clams have the potential to
drive benthic metabolism in farmed areas and to sustain macroalgal growth through regeneration of a limiting nutrient for

We studied the effects of differences in root growth and nutrient pool on the competitive ability of Festuca ovina (short grass), Arrhenatherum elatius and Calamagrostis epigejos (tall grasses) grown in monocultures and in mixtures of homogeneous and heterogeneous environments during two growing seasons.
Analysis of variance revealed a significant effect of plant species on nutrient concentrations in above-ground biomass and
of substrate type on contents of N, K, Ca, Mg in biomass. The ANOVA also confirmed the significant effect of competitive environment
on the concentration of N, K in above-ground biomass. In heterogeneous environments, both tall grasses (in competition with
F. ovina) were able to produce more roots in the nutrient-rich patches and to accumulate more nitrogen in plant tissues, which was
associated with higher yield of their above-ground biomass. Thus, the relative competitive ability for nutrients of both tall
grasses was higher than that of F. ovina. This competitive ability of A. elatius to C. epigejos increased in heterogeneous treatments. 相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes. 相似文献

We have examined the localization of P450scc in different tissues of the mollusc Mytilus galloprovincialis along a gonadal cycle by using a polyclonal antibody against rat cholesterol side-chain cleavage enzyme (cytochrome P450scc). Immunoreactivity specific for P450scc was found only in the cytoplasm of basophilic cells from digestive gland; gonad, gills and kidney appeared to be devoid of P450scc immunoreactivity. SDS-gel electrophoresis and western blot analysis of different subcellular fractions revealed that this protein is mainly located in microsomes, has a molecular weight of 51.4 kDa and is recognized by the antibody against rat P450scc. These results suggest that the digestive gland of M. galloprovincialis could be considered a steroidogenic tissue where the first step in biosynthesis of steroid hormones occurs. Moreover, seasonal slot blot analysis of post-mitochondrial fractions showed a P450scc induction by available nutrients and phytoplankton blooms. 相似文献

Cytokinin dehydrogenase (CKX) is responsible for regulating the endogenous cytokinin content by oxidative removal of the side
chain and seven distinct genes, AtCKX1 to AtCKX7, code for the enzyme in Arabidopsis thaliana. The recombinant enzyme AtCKX2 was produced in Saccharomyces cerevisiae after expressing the corresponding gene from a plasmid (pDR197) or following chromosomal integration, under either the constitutive
promoter PMA1 or the inducible promoter GAL1. The recombinant protein was purified from yeast culture media using a sequence of chromatographic steps. The purified enzyme
had a molecular mass of 61 kDa and a typical flavoprotein spectrum. The specific activity of the enzyme was 87.8 μkat g−1, with isopentenyladenine as a substrate and 2,3-dimethoxy-5-methyl-p-benzoquinone as an electron acceptor. The pH optimum lay between 7.0 and 8.0, depending on the electron acceptor used. AtCKX2
reacts both with isoprenoid and aromatic cytokinins, the activity with isoprenoid cytokinins being two to three orders of
magnitude higher. AtCKX2 prefers p-quinones and the synthetic dye 2,6-dichlorophenol indophenol as electron acceptors, although low reactivity with oxygen can

In contrast with the paradigmatic mammalian metallothioneins (MTs), mollusc MT systems consist at least of a high-cadmium induced form, possibly involved in detoxification, and another isoform either constitutive or regulated by essential metals and probably associated with housekeeping metabolism. With the aim of providing a deeper characterization of the coordination features of a molluscan MT peptide of the latter kind, we have analyzed here the metal-binding abilities of the recombinant MeMT-10-IV isoform of Mytilus edulis (MeMT). Also, comparison with other MTs of this type has been undertaken. A synthetic complementary DNA was constructed, cloned and expressed into two Escherichia coli systems. Upon zinc coordination, MeMT folds in vivo into highly chiral and stable Zn(7) complexes, with an exceptional reluctance to fully substitute cadmium(II) and/or copper(I) for zinc(II). In vivo cadmium binding leads to homometallic Cd(7) complexes that structurally differ from any of the in vitro prepared Cd(7) complexes. Homometallic Cu-MeMT can only be obtained in vitro from Zn(7)-MeMT after a great molar excess of copper(I) has been added. In vivo, two different heterometallic Zn,Cu-MeMT complexes are recovered, which nicely correspond to two distinct stages of the in vitro zinc/copper replacement. These MeMT metal-binding features are consistent with a physiological role related to basal/housekeeping metal, mainly zinc, metabolism, and confirm the correspondence between the MeMT gene response pattern and the functional properties of the encoded protein. 相似文献

Although our understanding of neuronal development in Trochozoa has progressed substantially in recent years, relatively little
attention has been paid to the bivalve molluscs in this regard. In the present study, the development of FMRFamide-, serotonin-
and catecholamine-containing cells in the mussel, Mytilus trossulus, was examined using immunocytochemical and histofluorescent techniques. Neurogenesis starts during the trochophore stage
at the apical extreme with the appearance of one FMRFamide-like immunoreactive (lir) and one serotonin-lir sensory cell. Later, five FMRFamide-lir and five serotonin-lir apical sensory cells appear, and their basal fibres form an apical neuropil. Fibres of two lateral FMRFamide-lir apical cells grow posteriorly and at the time that they reach the developing foot, the first FMRFamide-lir neurons of the pedal ganglia also appear. Subsequently, FMRFamide-lir fibres grow further posteriorly and reach the caudal region where neurons of the developing visceral ganglia then begin to
appear. In contrast, the five apical serotonin-lir neurons do not appear to project outside the apical neuropil until the late veliger stage. Catecholamine-containing cells
are first detected in the veliger stage where they appear above the oesophagus, and subsequently in the velum, foot, and posterior
regions. Though neural development in M. trossulus partly resembles that of polyplacophorans in the appearance of the early FMRFamidergic elements, and of scaphopods in the
appearance of the early serotonergic elements, the scenario of neural development in M. trossulus differs considerably from that of other Trochozoa (bivalves, gastropods, polyplacophorans, scaphopods and polychaetes) studied
to date.

Background
Gonadotropin releasing hormone (GnRH) is responsible for stimulation of gonadotropic hormone (GtH) in the hypothalamus-pituitary-gonadal axis (HPG). The regulatory mechanisms responsible for brain specificity make the promoter attractive for in silico analysis and reporter gene studies in zebrafish (Danio rerio).Results
We have characterized a zebrafish [Trp7, Leu8] or salmon (s) GnRH variant, gnrh 3. The gene includes a 1.6 Kb upstream regulatory region and displays the conserved structure of 4 exons and 3 introns, as seen in other species. An in silico defined enhancer at -976 in the zebrafish promoter, containing adjacent binding sites for Oct-1, CREB and Sp1, was predicted in 2 mammalian and 5 teleost GnRH promoters. Reporter gene studies confirmed the importance of this enhancer for cell specific expression in zebrafish. Interestingly the promoter of human GnRH-I, known as mammalian GnRH (mGnRH), was shown capable of driving cell specific reporter gene expression in transgenic zebrafish.Conclusions
The characterized zebrafish Gnrh3 decapeptide exhibits complete homology to the Atlantic salmon (Salmo salar) GnRH-III variant. In silico analysis of mammalian and teleost GnRH promoters revealed a conserved enhancer possessing binding sites for Oct-1, CREB and Sp1. Transgenic and transient reporter gene expression in zebrafish larvae, confirmed the importance of the in silico defined zebrafish enhancer at -976. The capability of the human GnRH-I promoter of directing cell specific reporter gene expression in zebrafish supports orthology between GnRH-I and GnRH-III.15.
We have investigated the floral ontogeny of Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis (of the eucalypt group, Myrtaceae) using scanning electron microscopy and light microscopy. Several critical characters for establishing relationships between these genera and to the eucalypts have been determined. The absence of compound petaline primordia in Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis excludes these taxa from the eucalypt clade. Post-anthesis circumscissile abscission of the hypanthium above the ovary in Stockwellia, Eucalyptopsis and Allosyncarpia is evidence that these three taxa form a monophyletic group; undifferentiated perianth parts and elongated fusiform buds are characters that unite Stockwellia and Eucalyptopsis as sister taxa. No floral characters clearly associate Arillastrum with either the eucalypt clade or the clade of Stockwellia, Eucalyptopsis and Allosyncarpia.We gratefully acknowledge Clyde Dunlop and Bob Harwood (Northern Territory Herbarium) for collecting specimens of Allosyncarpia, and Bruce Gray (Atherton) for collecting specimens of Stockwellia. A genetic transformation system has been developed for callus cells of Crataegus
aronia using Agrobacterium
tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with
5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different
types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red
colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli
were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this
is the first time to report an Agrobacterium-mediated transformation system in Crataegus

Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner.
The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate
larvae Galleria
mellonella with Pneumocystis proved futile since P. murina neither caused disease nor was able to proliferate within G. mellonella. It did, however, show that the pathogen could be rapidly cleared from the host. 相似文献

Previously, we described the unique feature of telomeric regions in Iberian shrew Sorex granarius: its telomeres have two ranges of size, very small (3.8 kb of telomeric repeats on average) and very large discontinuous telomeres (213 kb) interrupted with 18S rDNA. In this study, we have demonstrated extraordinary replication pattern of S. granarius large telomeres that have not been shown before in other studied mammal. Using the ReD-FISH procedure, we observed prolonged, through S period, large telomere replication. Furthermore, revealed ReD-FISH asymmetric signals were probably caused by partial replication of telomeres within an hour of 5-bromodeoxyuridine treatment due to the large size and special organization. We also found that in contrast to the telomeric halo from primary fibroblasts of bovine, mink, and common shrew, telomere halo of S. granarius consists of multiple loops bundled together, some of which contain rDNA. Here, we suggested several replicons firing possibly stochastic in each large telomere. Finally, we performed the TIF assay to reveal DNA damage responses at the telomeres, and along with TIF in nuclei, we found large bodies of telomeric DNA and ?-H2AX in the cytoplasm and on the surface of fibroblasts. We discuss the possibility of additional origin activation together with recombination-dependent replication pathways, mainly homologous recombination including BIR for replication fork stagnation overcoming and further S. granarius large telomere replication. 相似文献
