Phylogenetic relationships between Bothriocephalus species from freshwater and marine teleosts from different geographical regions were studied using internal transcribed spacer-2 and partial 18S ribosomal DNA. The analyses revealed a paraphyly of Bothriocephalus with respect to the genera Polyonchobothrium, Anantrum, and Clestobothrium. The freshwater species Bothriocephalus claviceps, B. acheilognathi, and Bothriocephalus sp. from Dorosoma petenense formed a well-supported monophyletic cluster, with Polyonchobothrium at its base. In contrast, the type species, B. scorpii, clustered within a distinct lineage formed by a heterogeneous assemblage of marine species, Clestobothrium crassiceps and Anantrum tortum, and the freshwater species B. cf. japonicus. This shows that the current morphology-based classification is unlikely to reflect the phylogenetic relationships within this group and will require a thorough revision. 相似文献

Bothriocephalus gadellus n. sp. is described from the intestine of the beardless codling Gadella imberbis (Gadiformes: Moridae) from the southwestern Gulf of Mexico. It resembles B. scorpii, the type-species of Bothriocephalus, and B. manubriformis, the only species of the genus reported from the Gulf of Mexico. B. gadellus n. sp. differs from both species in having four excretory canals on each side of the proglottid (three per side in B. scorpii; two per side anastomosing to form an extensive plexus throughout the medulla in B. manubriformis ), 24-33 testes per proglottid (30-60 in B. scorpii; 60-100 in B. manubriformis), a total length of 31-47 mm (32-950 mm in B. scorpii; 130-1,000 mm in B. manubriformis), and a seminal receptacle and wing-like expansions on each proglottid (absent in B. scorpii and B. manubriformis), as well as lacking two sets of reproductive organs per proglottid (present in B. scorpii) and a vagina with a bulbous sphincter near the opening (present in B. manubriformis). An amendment to the generic diagnosis of Bothriocephalus might be warranted if other species of this genus are found to possess a seminal receptacle. B. gadellus n. sp. appears to be like those species of Bothriocephalus that can utilise two intermediate hosts (copepod and fish). B. ellipticus and B. neglectus are designated as species inquirendae. This is the first report of a cestode from a morid fish in the Gulf of Mexico, and the first report of a parasite from a species of Gadellus. 相似文献

The molecular variation in Bothriocephalus acheilognathi Yamaguti, 1934 from 11 species of freshwater fish collected in Australia, China, the Czech Republic, England and Hawaii was investigated by determining the nucleotide sequences of the internal transcribed spacer region. The length of the first and second internal transcribed spacer sequences of multiple individuals ranged from 553 to 571 bp and 553 to 615 bp, and the G + C content from 53.1 to 53.5%. The percentage sequence divergence varied between 0 and 0.9% in the ITS1 and 0 and 6.6% in the ITS2, respectively, indicating the occurrence of intraspecific variation. It is demonstrated that the fragment length variation resulted primarily from microsatellite polymorphisms present in the ITS region, especially in the ITS2 region. Phylogenetic analyses revealed that B. acheilognathi examined in this study consisted of three closely related genotypes with certain degrees of host-specificity, and the genotype representing isolates from Cyprinus carpio L. was the most common and diverse form within the species B. acheilognathi. 相似文献

Whole specimens and histological and semi-thin sections of Bothriocephalus gregarius adults were exposed to lectins to identify carbohydrates present in the tegument and parenchyma. The sugars N-acetyl glucosamine, N-acetyl galactosamine, galactose, glucose (or mannose) and fucose were detected in the cestode using eight lectins: WGA (Wheat germ agglutinin), HPA (Helix pomatia agglutinin), SBA (Soy bean agglutinin), PHA (Phaseolus vulgaris agglutinin), RCA60 and RCA120 (Ricinus communis toxin and agglutinin), ConA (Concanavalin agglutinin) and UEA-I (Ulex europaeus agglutinin). Combined use of these methodological approaches (whole specimens, paraffin and semi-thin sections) revealed the presence of a gradient in the distribution of most of the sugars over the tegument, with the highest concentrations on the strobila (as shown by most of the lectins). Other sugars were specific for the scolex or strobila (as shown by UEA-I or HPA, respectively). The ultrastructural study showed that the distribution of glycoconjugates was associated with the presence of specific tegumental coats. The significance of this selective distribution and its relevance to cestode physiology and host-parasite relationships are discussed. 相似文献

A taxonomic study of specimens of Bothriocephalus from eels (Anguilla spp.) in Japan has demonstrated the occurrence of two species, B. claviceps (Goeze, 1782) and B. japonicus Yamaguti, 1934. The former species is a parasite of eels (A. anguilla and A. rostrata) in the Holarctic Region and was recently reported from A. marmorata in Japan. The conspecificity of tapeworms newly found in an eel (A. ? japonica) from Lake Biwa, central Japan, with B. claviceps has been confirmed by the great similarity of their ITS-2 gene sequences (similarity 95.3% and 95.2%). However, the sequences of worms identified as B. claviceps from A. marmorata differed considerably from those of B. claviceps from two populations of A. anguilla from Europe and the above-mentioned one from Japan (similarity 66.3%, 67.1% and 65.1 %, respectively), thus indicating that the former cestodes may have been misidentified. This assumption was confirmed by morphological evaluation of a voucher specimen from A. marmorata. The morphology of this cestode, as well as those from A. japonica from two localities in Japan (Lakes Biwa and Suwa), indicates their conspecificity with B. japonicus. The validity of this taxon has been confirmed on the basis of a re-examination of the type-specimens. The two taxa, B. japonicus and B. claviceps, differ from each other in the shape and length of the scolex (619–730 μm in B. japonicus versus 1,180–2,100 μm in B. claviceps), the relative position of the cirro-vaginal and uterine pores (opposite each other in relation to the median line of the body in B. japonicus versus tandem or slightly offset along the median line in the latter species), and the size of the eggs (41–52 × 28–35 μm in B. japonicus versus 50–70 × 31–43 μm in B. claviceps). 相似文献

Karyotypes of Eubothrium salvelini, E. crassum and Eubothrium sp. were studied using conventional Giemsa staining and comparative karyometric analysis. The karyotypes, reported here for the first time, consist of eight chromosome pairs. The two first pairs of homologues are metacentric and markedly larger than the remaining elements. The obvious similarity in karyotype structure does not exclude the possibility of discriminating E. salvelini and E. crassum using karyotypic characters. The best cytogenetic marker is the last pair of chromosomes, which is acrocentric in the karyotype of E. salvelini and metacentric in that of E. crassum. Karyological observations provide strong evidence for assigning Eubothrium sp. from Clupea harengus membras to E. crassum. Comments are made on the karyotypes of these and related species with respect to their phylogenetic links. 相似文献

Two new species of bucephalids (Digenea: Bucephalidae) were found parasitizing freshwater fishes of Argentina. Prosorhynchoides cambapuntaensis sp. n. from Salminus maxillosus (Characidae) is characterised by the tegument covered with two kinds of spines, scale-like spines anteriorly and fine spines posteriorly; uterine loops reaching the vitelline arch in the pre-oral region; and the Laurer's canal short, opening dorsally immediately posterior to the ovary. Rhipidocotyle santanaensis sp. n. from Acestrorhynchus pantaneiro (Acestrorhynchidae) is characterised by the rhynchus with two lateral projections; the testes arranged in tandem or slightly diagonal; and the vitelline follicles forming an arch in the pre-oral region. 相似文献

Beis I. and Theophilidis G. 1982. Phosphofructokinase in the plerocercoids of Schistocephatus solidus (Cestoda: Pseudophyllidea). International Journal for Parasitology12: 389–393. The Phosphofructokinase from the plerocercoids of Schistocephalus solidus was found to be inhibited by ATP. AMP relieves the ATP inhibition and activates the enzyme. In contrast to mammalian phosphofructokinase, the plerocercoid enzyme does not appear to be sensitive to inhibition by citrate at physiological ATP concentrations. Except for AMP and 3'–5' cyclic AMP no other monophosphate nucleotides were found to activate the enzyme. Succinate, α-ketoglutarate, malate, isocitrate, β-hydroxybutyrate, butyrate, acetoacetate and CoA all inhibit the plerocercoid enzyme at the concentrations tested. The significance of these results in the regulation of glycolysis and the tricarboxylic acid cycle in this parasite is discussed. 相似文献

End products of carbohydrate metabolism in B. scorpii were lactic, succinic and volatile fatty acids.
