effect of oral creatine supplementation (CR; 5 g/day) in conjunction with exercise training on physical fitness was investigated in men between 55 and 75 yr of age (n = 46). A double-blind randomized placebo-controlled (PL) trial was performed over a 6-mo period. Furthermore, a subgroup (n = 20) completed a 1-yr follow-up. The training program consisted of cardiorespiratory endurance training as well as moderate resistance training (2-3 sessions/wk). Endurance capacity was evaluated during a maximal incremental bicycle ergometer test, maximal isometric strength of the knee-extensor muscles was assessed by an isokinetic dynamometer, and body composition was assessed by hydrostatic weighing. Furthermore, in a subgroup (PL: n = 13; CR: n = 12) biopsies were taken from m. vastus lateralis to determine total creatine (TCr) content. In PL, 6 mo of training increased peak oxygen uptake rate (+16%; P < 0.05). Fat-free mass slightly increased (+0.3 kg; P < 0.05), whereas percent body fat slightly decreased (-1.2%; P < 0.05). The training intervention did not significantly change either maximal isometric strength or body weight. The responses were independent of CR. Still, compared with PL, TCr was increased by approximately 5% in CR, and this increase was closely correlated with initial muscle creatine content (r = -0.78; P < 0.05). After a 1-yr follow-up, muscle TCr was not higher in CR than in PL. Furthermore, the other measurements were not affected by CR. It is concluded that long-term creatine intake (5 g/day) in conjunction with exercise training does not beneficially impact physical fitness in men between 55 and 75 yr of age.  相似文献

Thereafter, this increase was maintained during 10 wk of training associated with low-dose creatine intake (5 g/day).Compared with placebo, maximal strength of the muscle groups trained,maximal intermittent exercise capacity of the arm flexors, and fat-free mass were increased 20-25, 10-25, and 60% more(P < 0.05), respectively, duringcreatine supplementation. Muscle PCr and strength, intermittent exercise capacity, and fat-free mass subsequently remained at a higherlevel in the creatine group than in the placebo group during 10 wk ofdetraining while low-dose creatine was continued. Finally, on cessationof creatine intake, muscle PCr in the creatine group returned to normalwithin 4 wk. It is concluded that long-term creatine supplementationenhances the progress of muscle strength during resistance training insedentary females.

Glucose transport in muscle is a function of the muscle metabolic state, as evidenced by the increase in glucose transport which occurs with conditions of altered aerobic metabolism such as hypoxia or contractile activity. The energy state of the muscle can be determined by the muscle phosphocreatine concentration. Dietary supplementation of creatine has been shown to increase both phosphocreatine (PCr) and creatine (TCr) levels in muscle, although not in the same proportion, so that the PCr/TCr ratio falls suggesting an altered energy state in the cell. The purpose of this study was to determine the effect of increased creatine content on glucose uptake in muscle. PCr and TCr were determined in plantaris muscles from rats following five weeks of dietary supplementation of creatine monohydrate (300 mg/kg/day). (3)H-2-deoxyglucose uptake was measured in epitrochlearis muscles incubated in the presence or absence of a maximally stimulating dose of insulin. Despite a significant increase in creatine content in muscle, neither basal nor insulin-stimulated glucose uptake was altered in creatine supplemented rats. Since PCr levels were not increased with creatine supplementation, these results suggest that the actual concentration of PCr is a more important determinant of glucose uptake than the PCr/TCr ratio.  相似文献

The effects of a recovery drink on overreaching induced by high frequency, high power resistance exercise was assessed. Resistance trained men were assigned to a supplemented (SUP, n = 8), placebo (PL, n = 3) or control (CON, n = 6) groups. All groups completed two weeks of familiarization training using the barbell squat. In week three, SUP and PL performed ten sets of five repetitions of speed squats twice daily, for a total of 15 training sessions. CON maintained their prior training schedule. Data were collected before week three (T1), after week three (T2) and after a week of recovery by training cessation (T3). During week three, SUP consumed an amino acid, carbohydrate and creatine monohydrate containing recovery drink immediately after each training bout. PL was provided a drink of similar appearance and taste but containing minimal nutritional value. At T2, both SUP and PL decreased mean squat velocity and power at 70% 1RM. Additionally, SUP and PL decreased muscle β2-adrenergic receptor (β2-AR) expression by 61 and 83%, respectively. Increases in the ratio of nocturnal urinary epinephrine/β2-AR ratio (EPI: β2AR) for SUP and PL suggested impaired sympathetic nervous system sensitivity. SUP demonstrated a smaller decrease in β2-AR expression and a lower EPI: β2AR, suggesting the recovery drink attenuated the detrimental effects of overreaching on the sympathetic activity. In conclusion, high power resistance exercise overreaching can induce performance decrements and impair sympathetic activity, but these effects may be attenuated by supplementation.  相似文献

The effect of 42 g of protein ingested pre- and post-exercise on recovery from an acute resistance exercise session was examined in 15 male strength/power athletes who were randomly divided into a supplement (SUP) or placebo (PL) group. Subjects reported to the Human Performance Laboratory (HPL) on four separate occasions (T1–T4). Maximal strength [one repetition-maximum (1-RM)] testing was performed during T1. During T2 subjects performed four sets of ten repetitions at 80% of their 1-RM in the squat, dead lift and barbell lunge exercises with 90 s of rest between each set. Blood draws occurred at baseline (BL), immediate and 15 min post-exercise to determine testosterone, cortisol and creatine kinase (CK) concentrations. Subjects reported back to the HPL 24 (T3) and 48 h (T4) post-exercise for a BL blood draw and to perform four sets of ten repetitions with 80% of 1-RM for the squat exercise only. No differences in the number of repetitions performed in the squat exercise were seen between the groups at T2. Relative to T2, PL performed significantly (P < 0.05) fewer repetitions than SUP at T3 and T4 (−9.5 ± 5.5 repetitions vs. −3.3 ± 3.6 during T3, respectively, and −10.5 ± 8.2 repetitions vs. −2.3 ± 2.9 repetitions during T4, respectively). No differences in hormonal measures were seen between the groups. CK concentrations were significantly (P < 0.05) elevated at T3 for both groups, but continued to elevate (P < 0.05) at T4 for PL only. No significant group differences were noted for CK at any time point. Results indicate that a proprietary protein SUP consumed before and after a resistance training session significantly contributes to improvements in exercise recovery 24 and 48 h post-exercise.  相似文献

The purpose of this study was to describe the physiological profile of responders (>20 mmol.kg(-1) dry weight [dw] increase in total intramuscular creatine monohydrate [Cr] + phosphorylated creatine [PCr]) versus nonresponders (<10 mmol.kg(-1) dw increase) to a 5-day Cr load (0.3 g.kg(-1).d(-1)) in 11 healthy men (mean age = 22.7 years). Pre-post 5-day cellular measures included total resting Cr content (Cr + PCr), fiber type composition, and fiber type cross-sectional area (CSA) determined from muscle biopsies of the vastus lateralis. Body mass, daily dietary intake, 24-hour urine outputs, urinary Cr and creatinine (CrN), and strength performance measures (1 repetition maximum [1RM] bench and leg press) were also assessed before and after the 5-day loading period. Results indicated that there were 3 levels of response to the 5-day supplementation: responders (R), quasi responders (QR), and nonresponders (NR) with mean changes in resting Cr + PCr of 29.5 mmol.kg(-1) dw (n = 3), 14.9 mmol.kg(-1) dw (n = 5), and 5.1 mmol.kg(-1) dw (n = 3), respectively. The results support a person-by-treatment interaction to acute Cr supplementation with R possessing a biological profile of lowest initial levels of Cr + PCr, greatest percentage of type II fibers, and greatest preload muscle fiber CSA and fat-free mass. Responders also showed improvement in 1RM leg press scores following the 5-day loading period. NR had higher preload levels of Cr + PCr, less type II muscle fibers, small preload muscle CSA, and lower fat-free mass and displayed no improvements in 1RM strength scores. The results suggest that to be considered a responder to acute oral supplementation, a favorable preexisting biological profile may determine the final extent to which an individual responds to supplementation. Physiologic profiles of nonresponders appear to be different and may limit their ability to uptake Cr. This may help partially explain the reported equivocal performance findings in the Cr supplementation literature.  相似文献

Human skeletal muscle satellite cells (SCs) are essential for muscle regeneration and remodeling processes in healthy and clinical conditions involving muscle breakdown. However, the potential influence of protein supplementation on post-exercise SC regulation in human skeletal muscle has not been well investigated. In a comparative human study, we investigated the effect of hydrolyzed whey protein supplementation following eccentric exercise on fiber type-specific SC accumulation. Twenty-four young healthy subjects received either hydrolyzed whey protein + carbohydrate (whey, n = 12) or iso-caloric carbohydrate (placebo, n = 12) during post-exercise recovery from 150 maximal unilateral eccentric contractions. Prior to and 24, 48 and 168 h post-exercise, muscle biopsies were obtained from the exercise leg and analyzed for fiber type-specific SC content. Maximal voluntary contraction (MVC) and serum creatine kinase (CK) were evaluated as indices of recovery from muscle damage. In type II fiber-associated SCs, the whey group increased SCs/fiber from 0.05 [0.02; 0.07] to 0.09 [0.06; 0.12] (p < 0.05) and 0.11 [0.06; 0.16] (p < 0.001) at 24 and 48 h, respectively, and exhibited a difference from the placebo group (p < 0.05) at 48 h. The whey group increased SCs/myonuclei from 4?% [2; 5] to 10?% [4; 16] (p?p < 0.001) and muscle soreness and CK increased (p < 0.001), irrespective of supplementation. In conclusion, whey protein supplementation may accelerate SC proliferation as part of the regeneration or remodeling process after high-intensity eccentric exercise.  相似文献

The aim of this study was to assess the effect of verbal instruction, surface stability, and load intensity on trunk muscle activity levels during the free weight squat exercise. Twelve trained males performed a free weight squat under four conditions: (1) standing on stable ground lifting 50% of their 1-repetition maximum (RM), (2) standing on a BOSU balance trainer lifting 50% of their 1-RM, (3) standing on stable ground lifting 75% of their 1-RM, and (4) receiving verbal instructions to activate the trunk muscles followed by lifting 50% of their 1-RM. Surface EMG activity from muscles rectus abdominis (RA), external oblique (EO), transversus abdominis/internal oblique (TA/IO), and erector spinae (ES) were recorded for each condition and normalized for comparisons. Muscles RA, EO, and TA/IO displayed greater peak activity (39–167%) during squats with instructions compared to the other squat conditions (P = 0.04–0.007). Peak EMG activity of muscle ES was greater for the 75% 1-RM condition than squats with instructions or lifting 50% of 1-RM (P = 0.04–0.02). The results indicate that if the goal is to enhance EMG activity of the abdominal muscles during a multi-joint squat exercise then verbal instructions may be more effective than increasing load intensity or lifting on an unstable surface. However, in light of other research, conscious co-activation of the trunk muscles during the squat exercise may lead to spinal instability and hazardous compression forces in the lumbar spine.  相似文献

Five women and 3 men (29.8 +/- 1.4 yr) performed dynamic knee-extension exercise inside a magnetic resonance system (means +/- SE). Two trials were performed 7-14 days apart, consisting of a 4- to 5-min exhaustive exercise bout. To determine quadriceps cost of contraction, brief static and dynamic contractions were performed pre- and postexercise. (31)P spectra were used to determine pH and relative concentrations of P(i), phosphocreatine (PCr), and betaATP. Subjects consumed 0.3 g. kg(-1). day(-1) of a placebo (trial 1) or creatine (trial 2) for 5 days before each trial. After creatine supplementation, resting DeltaPCr increased from 40.7 +/- 1.8 to 46. 6 +/- 1.1 mmol/kg (P = 0.04) and PCr during exercise declined from -29.6 +/- 2.4 to -34.1 +/- 2.8 mmol/kg (P = 0.02). Muscle static (DeltaATP/N) and dynamic (DeltaATP/J) costs of contraction were unaffected by creatine supplementation as well as were ATP, P(i), pH, PCr resynthesis rate, and muscle strength and endurance. DeltaATP/J and DeltaATP/N were greatest at the onset of the exercise protocol (P < 0.01). In summary, creatine supplementation increased muscle PCr concentration, which did not affect muscle ATP cost of contraction.  相似文献

Background

Therapeutic vaccination directed to induce an anti-tumoral T-cell response is a field of extensive investigation in the treatment of melanoma. However, many vaccination trials in melanoma failed to demonstrate a correlation between the vaccine-specific immune response and therapy outcome. This has been mainly attributed to immune escape by antigen loss, rendering us in the need of new vaccination targets.

Patients and methods

This phase-II trial investigated a peptide vaccination against survivin, an oncogenic inhibitor-of-apoptosis protein crucial for the survival of tumor cells, in HLA-A1/-A2/-B35-positive patients with treatment-refractory stage-IV metastatic melanoma. The study endpoints were survivin-specific T-cell reactivity (SSTR), safety, response, and survival (OS).

Results

Sixty-one patients (ITT) received vaccination therapy using three different regimens. 55 patients (PP) were evaluable for response and survival, and 41/55 for SSTR. Patients achieving progression arrest (CR?+?PR?+?SD) more often showed SSTRs than patients with disease progression (p?=?0.0008). Patients presenting SSTRs revealed a prolonged OS (median 19.6 vs. 8.6?months; p?=?0.0077); multivariate analysis demonstrated SSTR as an independent predictor of survival (p?=?0.013). The induction of SSTRs was associated with gender (female vs. male; p?=?0.014) and disease stage (M1a/b vs. M1c; p?=?0.010), but not with patient age, HLA type, performance status, or vaccination regimen.

Conclusion

Survivin-specific T-cell reactivities strongly correlate with tumor response and patient survival, indicating that vaccination with survivin-derived peptides is a promising treatment strategy in melanoma.  相似文献

The total creatine(TCr) pool of skeletal muscle is composed of creatine (Cr) andphosphocreatine (PCr). In resting skeletal muscle, the ratio ofPCr to TCr (PCr/TCr; PCr energy charge) is ~0.6-0.8, dependingon the fiber type. PCr/TCr is linked to the cellular free energy of ATPhydrolysis by the Cr kinase equilibrium. Dietary Cr supplementationincreases TCr in skeletal muscle. However, many previous studies havereported data indicating that PCr/TCr falls after supplementation,which would suggest that Cr supplementation alters the restingenergetic state of myocytes. This study investigated the effect of Crsupplementation on the energy phosphates of resting skeletal muscle.Male rats were fed either rodent chow (control) or chow supplementedwith 2% (wt/wt) Cr. After 2 wk on the diet, the gastrocnemius andsoleus muscles were freeze clamped and removed from anesthetizedanimals. Cr supplementation increased TCr, PCr, and Cr levels in thegastrocnemius by 20, 22, and 17%, respectively (P < 0.05). A numerical 6% higher mean soleus TCr in Cr-supplemented ratswas not statistically significant. All other energy phosphate concentrations, free energy of ATP hydrolysis, and PCr/TCr were notdifferent between the two groups in either muscle. We conclude that Crsupplementation simply increased TCr in fast-twitch rat skeletal musclebut did not otherwise alter resting cellular energetic state.

Previous studies have shown that creatine supplementation reduces muscle damage and inflammation following running but not following high-force, eccentric exercise. Although the mechanical strain placed on muscle fibers during high-force, eccentric exercise may be too overwhelming for creatine to exert any protective effect, creatine supplementation may protect skeletal muscle stressed by a resistance training challenge that is more hypoxic in nature. The purpose of this study was to examine the effects of short-term creatine supplementation on markers of muscle damage (i.e., strength, range of motion, muscle soreness, muscle serum protein activity, C-reactive protein) to determine whether creatine supplementation offers protective effects on skeletal muscle following a hypoxic resistance exercise test. Twenty-two healthy, weight-trained men (19-27 years) ingested either creatine or a placebo for 10 days. Following 5 days of supplementation, subjects performed a squat exercise protocol (5 sets of 15-20 repetitions at 50% of 1 repetition maximum [1RM]). Assessments of creatine kinase (CK) and lactate dehydrogenase activity, high-sensitivity C-reactive protein, maximal strength, range of motion (ROM), and muscle soreness (SOR) with movement and palpation were conducted pre-exercise and during a 5-day follow up. Following the exercise test, maximal strength and ROM decreased, whereas SOR and CK increased. Creatine and placebo-supplemented subjects experienced significant decreases in maximal strength (creatine: 13.4 kg, placebo: 17.5 kg) and ROM (creatine: 2.4 degrees , placebo: 3.0 degrees ) immediately postexercise, with no difference between groups. Following the exercise test, there were significant increases in SOR with movement and palpation (p < 0.05 at 24, 48, and 72 hours postexercise), and CK activity (p < 0.05 at 24 and 48 hours postexercise), with no differences between groups at any time. These data suggest that oral creatine supplementation does not reduce skeletal muscle damage or enhance recovery following a hypoxic resistance exercise challenge.  相似文献

Based on background concentrations of elements in soils and the sixth population census data of China, this study discussed the distribution characteristics of soil elements and longevity population at province level in China. Percentages of the aging population are high in Southwest China and the eastern coastal region but low in western and northwestern regions. Provinces in South and Southwest China gain a high level of longevity, while the northern part of China has a low level of longevity. The background concentration of Se in soil has a significant positive correlation with longevity index, while Ba and Ni have a significant negative correlation with longevity indexes. By regression analysis, longevity index C/100,000 can be expressed as C/100,000?=?1.679?0.205 Ni?+?0.413 Co?+?0.006 Se (with R 2?=?0.402 and p?<?0.01), C/65+ can be expressed as C/65+=3.425?0.262 Ni?+?0.435 Co?+?0.006 Se (with R 2?=?0.369 and p?<?0.01).  相似文献

Background

High-carbohydrate diets (HCD) are favoured by the aquaculture industry for economic reasons, but they can produce negative impacts on growth and induce hepatic steatosis. We hypothesised that the mechanism behind this is the reduction of hepatic betaine content.

Objective

We further explored this mechanism by supplementing betaine (1%) to the diet of a farmed fish Megalobrama amblycephala.

Methods

Four diet groups were designed: control (CD, 27.11% carbohydrates), high-carbohydrate (HCD, 36.75% carbohydrates), long-term betaine (LBD, 35.64% carbohydrates) and short-term betaine diet (SBD; 12 weeks HCD?+?4 weeks LBD). We analysed growth performance, body composition, liver condition, and expression of genes and profiles of metabolites associated with betaine metabolism.

Results

HCD resulted in poorer growth and liver health (compared to CD), whereas LBD improved these parameters (compared to HCD). HCD induced the expression of genes associated with glucose, serine and cystathionine metabolisms, and (non-significantly, p?=?.20) a betaine-catabolizing enzyme betaine-homocysteine-methyltransferase; and decreased the content of betaine, methionine, S-adenosylhomocysteine and carnitine. Betaine supplementation (LBD) reversed these patterns, and elevated betaine-homocysteine-methyltransferase, S-adenosylmethionine and S-adenosylhomocysteine (all p?≤?.05).

Conclusion

We hypothesise that HCD reduced the content of hepatic betaine by enhancing the activity of metabolic pathways from glucose to homocysteine, reflected in increased glycolysis, serine metabolism, cystathionine metabolism and homocysteine remethylation. Long-term dietary betaine supplementation improved the negative impacts of HCD, inculding growth parameters, body composition, liver condition, and betaine metabolism. However, betaine supplementation may have caused a temporary disruption in the metabolic homeostasis.

Quinone oxidoreductases (NAD(P)H): quinone oxidoreductase 1 (NQO1) and NRH: quinone oxidoreductase 2 (NQO2) are an antioxidant enzyme, important in the detoxification of environmental carcinogens. Methylene-tetra-hydrofolate reductase (MTHFR), plays a role in folate metabolism and may have oncogenic role through disruption of normal DNA methylation pattern, synthesis, and impaired DNA repair. In a case–control study, genotyping was done in 195 PCa and 250 age matched unrelated healthy controls of similar ethnicity to determine variants in NQO1 exon 4 (C?>?T, rs4986998), exon 6 (C?>?T, rs1800566), NQO2 ?3423 (G?>?A, rs2070999) and MTHFR exon 4 (C?>?T, rs1801133) by PCR–RFLP methods. Heterozygous genotype CT and variant allele career genotype (CT?+?TT) of NQO1 exon 4 showed increased risk of PCa (OR?=?2.06, p?=?0.033; OR?=?2.02, p?=?0.027). Variant allele T also revealed increased risk (OR?=?1.87, p?=?0.029). Similarly variant genotype TT (OR?=?2.71, p?=?0.009), combined genotype (CT?+?TT) (OR?=?1.59, p?=?0.019) and T allele (OR?=?1.63, p?=?0.002) of NQO1 exon 6 demonstrated significant risk for PCa. Diplotypes of NQO1 (exon 4 and 6), C–T (OR?=?1.56, Pc?=?0.007) and T–T (OR?=?0.011, Pc?=?3.86) was associated with an increased risk for PCa. NQO2 and MTHFR did not show any risk with PCa. Our results strongly support that common sequence variants and diplotypes of NQO1 exon 4 and 6 genes may have role in PCa risk in the North Indian population, indicating the importance of genes involved in metabolism with respect to PCa risk. Additional studies on larger populations are needed to clarify the role of variation in these genes in PCa carcinogenesis.  相似文献

In this work, the cytotaxonomic implications of the chromosomal characterization of cultivated and native Zephyranthes species described in northeastern Brazil were studied. All individuals had karyotype formed by a set of metacentric chromosomes, in addition to submetacentric and acrocentric chromosomes. In Zephyranthes robusta, 2n?=?12 was observed and karyotype with formula?4M?+?2SM in somatic cells, representing the most symmetric karyotype among the investigated species. Z.?sylvatica showed three different chromosome complement numbers: 2n?=?12 with formula?1M?+?5SM, 2n?=?12?+?1B with 1M?+?5SM?+?(1B), and 2n?=?18 formed by cracks. The cultivated species Z.?rosea Lindl. presented 2n?=?24 with 4M?+?7SM?+?1A, however Z.?grandiflora Lindl. showed the same chromosome number with 2M?+?5SM?+?5A. Zephyranthes aff. rosea Lindl. presented 2n?=?25 with one small metacentric forming a crack in the fourth metacentric pair. Z.?brachyandra has 2n?=?24?+?(1B) and formula?4M?+?3SM?+?5A?+?(1B). Z.?candida Herb. presented 2n?=?38 and karyotype formula?9M?+?10SM. In Habranthus itaobinus numerical variation was observed, with the majority of populations showing a chromosome complement composed of 2n?=?44?+?1B with 5M?+?12SM?+?5A?+?(1B), or 2n?=?44?+?3B in a single population. Mechanisms involved in the formation of these karyotypes from chromosomal imbalance data are discussed. Taken together, data from this study only partially confirm previous counts for epithets and further enhance the cytological variability data previously reported for the genus.  相似文献

Abstract

A balance experiment was carried out to investigate the effects of betaine monohydrate (BET) or betaine derived from condensed molasses solubles (CMS) as a substitute for methionine and choline on nitrogen (N) balance and total tract nutrient digestibility in weaned piglets. The experiment included four treatments with 32 barrows with an average initial body weight (BW) of 13.5 kg. The supplementation of DL-methionine and choline (positive control = PC) to the basal diet, which was deficient in methionine and low in compatible osmolytes in the form of betaine or its precursor choline (negative control = NC) resulted in a significant increase in N retention of 0.8 g/d. The substitution of DL-methionine and choline with BET or CMS did not affect N retention compared to the PC and the NC treatment either. Feeding the PC diet increased the digestibilities of organic matter, NDF, ADF, NFE, crude ash, Ca, P, methionine, tryptophan and cystine by 1.9%, 7.3%, 9.7%, 1.1%, 6.3%, 13.9%, 7.7%, 15.9%, 4.3% and 2.8%, respectively, and tended (p < 0.20) to increase the digestibilities of most other amino acids by 1.6 – 3.4%. Digestibility of CP, EE (HCl), Mg and Na was 3.1% (p = 0.09), 5.1% (p = 0.09), 5.1% (p = 0.06) and 3.3% (p = 0.17) higher, respectively, when compared to the NC treatment. BET and CMS supplementation increased most nutrient digestibilities in the same magnitude as for the PC treatment. In summary, the supplementation of betaine, originating from different sources, to a diet with low contents of compatible osmolytes increased in particular the fermentation of fibre and enhanced mineral absorption. The supplementation of the NC with DL-methionine was more efficient in improving N retention than the replacement of DL-methionine by betaine originating from BET or CMS.  相似文献

This study focused on understanding the signaling mechanisms leading to GLUT-4 translocation and increased skeletal-muscle glucose uptake that follow creatine (Cr) supplementation in type 2 diabetes (n = 10). AMPK-α protein content presented a tendency to be higher (p = 0.06) after Cr supplementation (5 g/d for 12w). The changes in AMPK-α protein content significantly related (p < 0.001) to the changes in GLUT-4 translocation (r = 0.78) and Hb1Ac levels (r = ?0.68), suggesting that AMPK signaling may be implicated in the effects of supplementation on glucose uptake in type 2 diabetes.  相似文献

The purpose was to determine if creatine supplementation, consumed immediately before and immediately after exercise, with different dosing frequency (i.e., 2 or 3 d wk) could enhance the gains in muscle size and strength from resistance training (RT) in young adults. A group of 38 physically active, nonresistance trained university students (21-28 years) was randomly allocated to 1 of 4 groups: CR2 (0.15 g·kg creatine during 2 d wk of RT; 3 sets of 10 repetitions; n = 11, 6 men, 5 women), CR3 (0.10 g·kg creatine during 3 d wk of RT; 2 sets of 10 repetitions; n = 11, 6 men, 5 women;), PLA2 (placebo during 2 d wk of RT; n = 8, 5 men, 3 women), and PLA3 (placebo during 3 d wk of RT; n = 8, 4 men, 4 women) for 6 weeks. Before and after training, measurements were taken for muscle thickness of the elbow and knee flexor and extensor muscle groups (ultrasound), 1-repetition maximumleg press and chest press strength, and kidney function (urinary microalbumin). Repeated-measures analysis of variance showed that strength and muscle thickness increased in all groups with training (p < 0.05). The CR2 (0.6 ± 0.9 cm or 20%; p < 0.05) and CR3 groups (0.4 ± 0.6 cm or 16.4%; p < 0.05) experienced greater change in muscle thickness of the elbow flexors compared to the PLA2 (0.05 ± 0.5 cm or 2.3%) and PLA3 groups (0.13 ± 0.7 cm or 6.3%). Men supplementing with creatine experienced a greater increase in leg press strength (77.3 ± 51.2 kg or 62%) compared to women on creatine (21.3 ± 10 kg or 34%, p < 0.05). We conclude that creatine supplementation during RT has a small beneficial effect on regional muscle thickness in young adults but that giving the creatine over 3 d wk did not differ from giving the same dose over 2 d wk.  相似文献

The purpose of this study was to examine the efficacy of 15 days of betaine supplementation on peak concentric (CON) and eccentric (ECC) force during isokinetic exercise in active college-aged men. Eleven men volunteered for this study (21.7 ± 5.1 years; height: 178.5 ± 6.4 cm; body mass: 79.8 ± 10.3 kg). Subjects were randomly assigned to either a supplement (BET) or placebo (PL) group. Supplementation occurred for 15 days. Subjects reported to the Human Performance Laboratory on 5 occasions during this period, separated by 72 hours, for a testing and exercise session on an isokinetic chest press device. After each exercise protocol, subjects rated their fatigue and muscle soreness on a 15-cm visual analog scale. Subjects then consumed no daily BET for 4 weeks but maintained familiarity with the exercise device once per week. After the washout period, subjects resumed the BET protocol using the opposite drink and repeated the same 15-day protocol. No differences were noted in maximum CON force output between pre (335.9 ± 78.3 and 321.6 ± 63.6 N) and post (330.3 ± 74.8 and 330.2 ± 71.6 N) workouts in both BET and PL, respectively. In addition, no differences were noted in maximum ECC force output between pre (352.0 ± 90.6 and 324.4 ± 85.2 N) and post (353.2 ± 98.2 and 366.9 ± 128.5 N) workouts in BET and PL, respectively. No differences in subjective measures of soreness and fatigue were seen, but a significant reduction in Δ fatigue was observed in BET compared to PL. In conclusion, 15 days of betaine supplementation did not increase peak CON or ECC force outputs during an isokinetic chest press but did appear to reduce subjective measures of fatigue to the exercise protocol.  相似文献
