采用光谱法和胰酯酶分解法对大果木姜子油酯中分得的结晶C—I组成进行了分析,结果表明C—I是由11种甘油三酯组成。其中2-位为月硅酸的甘油三酯占91.80%,1,3-位脂肪酸主要为癸酸和月桂酸,甘油三酯的组成中,CLC占62.24%,LLC占24.50%。  相似文献

目的探讨DHA对肝X受体激动剂T0901317诱导的HepG2细胞甘油三酯积聚的影响。方法体外培养HepG2细胞,以50μmol/LDHA、10μmol/LT0901317分别处理细胞以及50μmloL/LDHA和10μmol/LT0901317共同处理细胞48h。油红0染色观察细胞内脂质沉积;氯仿-甲醇抽提细胞总脂质,酶法定量检测细胞甘油三酯含量;实时定量PCR检测与脂肪酸代谢相关基因如SREBP-1c、FAS、SCD-1、PPARa和CD36的mRNA水平。结果与对照组相比,10μmol/LT0901317处理48h后,HepG2细胞内的油红O染色脂滴增多,甘油二酯浓度升高了50%;脂肪酸合成基因:SREBP-1c、FAS和SCD-1及脂肪酸吸收基因CD36的mRNA水平分别升高了9.9、5.2、2.2和1.5倍,而脂肪酸降解基因PPARoz的mRNA无变化。DHA与T0901317共同处理的HepG2细胞内脂滴明显减少;甘油三酯含量比70901317处理组降低了15%:SREBP—1c、FAS、SCD-1和CD36的mRNA水平比T0901317处理组分别降低了92%、31%、46%和60%,而PPARa的mRNA水平比T0901317处理组升高了30%。结论DHA通过降低脂肪酸合成和吸收基因的表达并升高脂肪酸降解基因的表达缓解肝x受体激活所致HepG2细胞内甘油三酯积聚。  相似文献

Roxb.]种子桕脂甘油三酯组成进行了测定,桕脂的甘油三酯是由9-14种甘油三酯所组成,与可可脂相似之处是甘油三酯2-位上的脂肪酸,82-88%为油酸,二饱和酸甘油酯中81-88%,其中以POP为主(74-81%);单饱和酸甘油酯占1.8-6.0%;而三饱和酸甘油酯占9.6-15.6%,主要是PPP(8.5-14.0%)。桕脂经过适当的加工处理,可以做可可脂的代用品,将湖南产的乌桕与广西栽培的4个品种(“大粒鸡爪”,“蜈蚣”,“铜锤”和“小粒鸡爪”)桕脂中的甘油三酯组成比较,以“大粒鸡爪”桕脂中二饱和酸甘油酯的比较最高(88.0%),三饱和酸甘油酯的含量最低(9.6%)。  相似文献

小果微花藤(茶茱萸科)[Iodes vitiginea(Hance)Hemsl.](Icacinaccae)种仁含油39-50%,经气相色谱分析,油的主要脂肪酸为十六碳烯酸和棕捅酸。利用臭氧化反应、色谱-质谱和红外光谱方法,鉴定十六碳烯酸为顺-Δ′-十六碳烯酸。用高效液相色谱从油中分离出13个甘油三酯(峰),其中8种甘油三酯进行了色谱鉴定。总甘油三酯中,主要甘油三酯为 HHH(18.4%),HHP(32.3%),PHP(16.8%)。  相似文献

蛋白核小球藻脂溶性化合物的抑菌活性及成分分析   总被引:9,自引:0,他引:9  
利用纸片法对蛋白核小球藻(Chlorella pyrenoidosa Chick.)脂溶性化合物的粗脂以及经硅胶柱层析分离后的不同组分进行了抑菌实验。结果表明,蛋白核小球藻的粗脂有极强的抑菌活性,其石油醚洗脱组分占总脂的63.9%,抑菌活性最弱;乙醇洗脱级分占总脂的32.2%,抑菌活性最强;苯洗脱组分只占总脂的3.9%,对玉米大斑病菌(Helminthosporium turcicum Pass)有极强的抑制活性。各洗脱组分及粗脂对革兰氏阴性菌均我抑制活性。成分分析结果表明,石油醚洗脱组分以烷烃类化合物为主,其中1-十九烯经烃含量最高;苯洗脱组分以6,10,14-三甲基-2-十五酮、十六碳酮和2-十一烷酮这3种酮类化合物为主;而乙醇洗脱组分含有大量的脂肪酸,其中γ-亚麻酸含量最高(56.673%)。蛋白核小球藻脂溶怀化合物的抑菌活性是其各种化学成分相互综合作用的结果。  相似文献

丹麦植物生理研究所和哥本哈根大学研究了食用植物木薯Manihotesculonta Crantz,即M.utifissima Pohl种子的主要成份。木薯种子的种仁为干种子的57%,并含有占种仁干重47%的丰富类脂化合物。总类脂物质的色层分离表明,其中98%的类脂物质是三脂肪酸甘油脂,2%是由微量的二、单脂肪酸甘油脂,磷脂和醣脂组成。未发现有游离脂肪酸。  相似文献

本语文对中华鳖的稚鳖、一龄鳖、二龄鳖和三龄鳖的肌肉、背甲、裙边和油脂块等不同部位中的水分、灰分、蛋白南含量、脂含量及其肌肉中的脂肪酸的组成进行了分析,结果表明;中华鳖的肌肉、裙边和背甲中的蛋白质的含量高达18.33-23.44%,脂肪含量较低。肌肉中高度不饱和脂肪酸含量占脂肪酸总量的32.69%,DHA与EPA分别占脂肪酸总量的8.30%和6.97%左右。本研究在分析与比较的基础上评价了中华鳖的营  相似文献

本试验对黑穗醋栗种子脂肪酸的组成和含量进行了研究。结果表明,种子中脂肪酸主要由棕榈酸、硬脂酸、亚油酸、亚麻酸和r-亚麻酸等组成,尚有两种性质不清。其中饱和脂肪酸占23.5%,不饱和脂肪酸占72.83%,未知物占3.92%,而r-亚麻酸含量占13.22%,它是抗心血管疾病的药理活性成分。  相似文献

Qualitative and quantitative changes in phosphatidylethanolamine (PE) were analyzed in the eggs, embryos and tadpoles of the Japanese pond frog, Rana nigromaculata, at various stages of development. The weight percentage of PE to total phospholipid and to total lipid was about 15-18% and about 3-4%, respectively, during embryonic life. At all stages from the unfertilized egg to the feeding tadpole, the major fatty acids at the 1-position of PE were palmitic, stearic and oleic acids. At the 2-position, arachidonic, oleic, palmitic, stearic and linoleic acids were present during embryonic life. The most abundant fatty acid at the 2-position was arachidonic acid at the unfertilized egg and hatching embryo stages. However, palmitic acid was the most prevalent 2-fatty acid at the posthatching tadpole and the feeding tadpole stages. Thus, there were marked changes in the positional distribution of the constituent fatty acids in PE during development.  相似文献

Abstract

The nutritive value of genetically modified myristic acid-rich rapeseed, in which a acyl-thioesterase gene inserted, was studied. Crude nutrients, amino acid and fatty acid profiles as well as mineral and glucosinolate contents were determined and compared with those of the non-transgenic parental cultivar. The concentration of crude nutrients, minerals and amino acids were found to be within the range of natural variance. The myristic and palmitic acid content increased from 0.1 – 11.4% and from 3.6 – 20%, respectively, at the expense of oleic acid, which decreased from 68.6 – 42.6% of total fatty acids. The glucosinolate contents increased from 12.4 µmol/g in the parental plant to 19 µmol/g DM in the GM-plant. Full-fat rapeseed of both cultivars was incorporated in pig diets at a level of 15%, and the digestibility and the production efficiency were tested under ad libitum feeding conditions with ten pigs each over the growing finishing period from 32 – 105 kg BW. The experimental diets did not show significant differences in digestibility and energetic feeding value. However, feed intake and weight gain decreased presumably due to the increasing glucosinolate intake associated with the feeding of transgenic rapeseed. The dietary fatty acids profile influenced the fatty acid profile of body fat. Myristic acid accumulated in back fat and intramuscular fat while the oleic acid content decreased. The increased glucosinolate intake affected the weight of thyroid glands and their iodine concentration.  相似文献

The phosphatidylcholine transfer protein (PC-TP) from bovine liver has a binding site for phosphatidylcholine (PC). Structural and molecular characteristics of this site were investigated by binding PC-analogues carrying photolabile, fluorescent and short-chain fatty acids. Analysis of the photolabeled PC/PC-TP adduct showed that the hydrophobic peptide segment Val171-Phe-Met-Tyr-Tyr-Phe-Asp177 is part of the lipid binding site for the 2-acyl chain. This site was further studied by binding PC carrying cis-parinaric acid at the sn-2-position. Time resolved fluorescence anisotropy measurements indicated that the 2-acyl chain was immobilized following the rotation of PC-TP. Similar experiments with PC carrying cis-parinaric acid at the sn-1-position demonstrated that the 1-acyl chain was immobilized as well but at a site distinctly different from that of the 2-acyl chain. Binding sites for the 1- and 2-acyl chain were then explored by use of PC-isomers carrying decanoic, lauric and myristic acid at the sn-1- (or sn-2-)-position and oleic acid at the sn-2- (or sn-1-)-position. Incubation with vesicles prepared of these PC-species indicated that binding to PC-TP diminished with decreasing acyl chain length but more so for species with short-chain fatty acids on the sn-2-position than on the sn-1-position. Transfer experiments confirmed that PC-TP discriminates between PC-isomers of apparently equal hydrophobicity favouring the transfer of these species carrying oleic acid at the sn-2-position.  相似文献

摘要 目的:揭示次氯酸与不饱和脂肪酸的氧化反应机制及转化产物。方法:运用Gaussian 16软件包,采用密度泛函方法M06-2X(D3),结合6-31+G(d)基组,在SMD液相水模型水平下进行计算。结果:次氯酸与单不饱和脂肪酸油酸的氧化反应是先形成氯鎓离子中间体,氯鎓离子再与水分子反应生成氯醇,第一步氯鎓离子的形成是控速步骤,其反应活化自由能~8 kcal/mol。环氧化合物和短链的醛是两种转化产物,前者由氯醇脱氯化氢而来,而后者由环氧化合物和氯醇通过系列与次氯酸根的反应而得到,生成它们的控速步骤的反应活化自由能分别为23 和24 kcal/mol。选取两个乙基为取代基的乙烯为油酸模型,其与次氯酸反应的活化自由能仅比油酸高1 kcal/mol。计算得到次氯酸与亚油酸、顺-9,反-11 亚油酸、梓树酸和花生四烯酸模型氧化反应生成氯醇的活化自由能分别是~10、13、16和14 kcal/mol。结论:氯鎓离子中间体机制是次氯酸与不饱和脂肪酸氧化反应的主要机制,反应的活化自由能通常低于15 kcal/mol,意味着此氧化反应动力学上容易发生。氧化产物氯醇能转化为环氧化合物和短链的醛,但活化自由能较高,约23和24 kcal/mol。选取距离双键3个碳以内的结构为不饱和脂肪酸模型,它能够很好地反映不饱和脂肪酸的反应活性。  相似文献

To improve the performance of yeast surface-displayed Rhizomucor miehei lipase (RML) in the production of human milk fat substitute (HMFS), we mutated amino acids in the lipase substrate-binding pocket based on protein hydrophobicity, to improve esterification activity. Five mutants: Asn87Ile, Asn87Ile/Asp91Val, His108Leu/Lys109Ile, Asp256Ile/His257Leu, and His108Leu/Lys109Ile/Asp256Ile/His257Leu were obtained and their hydrolytic and esterification activities were assayed. Using Discovery Studio 3.1 to build models and calculate the binding energy between lipase and substrates, compared to wild-type, the mutant Asp256Ile/His257Leu was found to have significantly lower energy when oleic acid (3.97 KJ/mol decrease) and tripalmitin (7.55 KJ/mol decrease) were substrates. This result was in accordance with the esterification activity of Asp256Ile/His257Leu (2.37-fold of wild-type). The four mutants were also evaluated for the production of HMFS in organic solvent and in a solvent-free system. Asp256Ile/His257Leu had an oleic acid incorporation of 28.27% for catalyzing tripalmitin and oleic acid, and 53.18% for the reaction of palm oil with oleic acid. The efficiency of Asp256Ile/His257Leu was 1.82-fold and 1.65-fold that of the wild-type enzyme for the two reactions. The oleic acid incorporation of Asp256Ile/His257Leu was similar to commercial Lipozyme RM IM for palm oil acidolysis with oleic acid. Yeast surface-displayed RML mutant Asp256Ile/His257Leu is a potential, economically feasible catalyst for the production of structured lipids.  相似文献

This paper reports the results of our analysis of the impact high levels of de novo fatty acids have on the proportions of essential and non-essential fatty acids in human milk lipids. The data for seven fatty acids (linoleic, alpha-linolenic, arachidonic (AA), docosahexaenoic (DHA), palmitic, stearic and oleic) were derived from several studies conducted in Nigeria. The proportion by weight of each of these fatty acids was plotted versus the proportion of C10-14 fatty acids. As the proportion of C10-14 fatty acids increased from 15 to 65%, there was not a proportional decrease in the percentages of all seven fatty acids, but, instead, preferential incorporation of the essential fatty acids, AA and DHA into the triacylglycerol component of the milk. At the same time, the proportions of stearic and oleic acid declined by 69% and 86%, respectively. However, the proportions of linoleic acid, palmitic acid, DHA, AA and alpha-linolenic acid, in milk lipids decreased by only 44%, 40%, 39%, 28% and 2.3%, respectively. These observations indicate that as the contribution of C10-14 fatty acids increases, essential fatty acids are preferentially incorporated into milk triacylglycerols at the expense of oleic acid and stearic acid.  相似文献

beta-Lactoglobulin isolated from milk of cow, sheep, and goat had about 0.5 mol of fatty acids bound per mol of monomer protein. Fatty acids, mainly palmitic and oleic acids, were the major components (about 75% of total lipids). Albumin isolated from the same samples had about 4.5 mol of fatty acids bound per mol of protein. These two proteins were the only whey proteins able to bind labeled fatty acids in vitro. Interaction of beta-lactoglobulin and albumin with insolubilized fatty acids showed some differences, suggesting different structures of the respective fatty acid binding sites.  相似文献

Interphotoreceptor retinoid-binding protein (IRBP) purified from monkey interphotoreceptor matrix contains relatively high concentrations of endogenous fatty acids, 6.51 mol/mol of protein. Sixty-five percent of the total fatty acid bound to IRBP was found to be noncovalently attached, with the remainder covalently bound. The fatty acids are not residual components of phospholipids or neutral lipids, as judged by microchemical methods. The major fatty acids bound to IRBP are: palmitic (35%), stearic (21%), palmitoleic (7%), oleic (29%), linoleic (6%) and docosahexaenoic acids (2%). These fatty acids account for about 90% of the total fatty acid bound to interphotoreceptor matrix proteins extracted with organic solvents. Thus, IRBP may function as an intercellular fatty acid carrier and may depend on the covalently bound fatty acids for anchoring in the outer leaflet of cell membranes.  相似文献

Qualitative and quantitative changes in phosphatidylinositol (PI) were analyzed in the eggs, embryos and tadpoles of the Japanese pond frog, Rana nigromaculata, at various stages of development. 2. The weight percentage of PI to total phospholipid and lipid was about 8.4-15.2% and 1.4-2.6%, respectively, during embryonic life. 3. At the early stages of the unfertilized egg and the two-cell embryo, the predominant fatty acids are palmitic, stearic, oleic and linoleic acid. From the dorsal lip, early gastrula stage and beyond, the percentage of linoleic acid declines and there is an increase in palmitoleic acid. A relatively large amount of arachidonic acid was noted at the unfertilized egg stage at the 1-position. 4. A large amount of arachidonic acid was also observed at the 2-position of PI in the unfertilized egg, hatching embryo and post-hatching tadpole stages, relative to palmitic and stearic acid. 5. Palmitic and stearic acid were increased at the 2-position of PI in the other embryo and the feeding tadpole stages, relative to arachidonic acid, indicating a shift in these molecular species. 6. Thus, there were marked changes in the positional distribution of the constituent fatty acids in PI during early development of R. nigromaculata.  相似文献

人类对于大型偶蹄类动物的利用已有较多先例,北美麝牛(Ovibous muschtus)近年在加拿大、美国等已经驯养,并以其毛绒作为一种特殊的毛丝工业原料。美洲野牛(Bison Americasus)已与家牛杂交生产肉用牛。我国青藏高原的牦牛(Bos grinus)与黄牛杂交而成的犏牛,已成为藏民的主要肉乳兼用牛。羚牛(Budorcas taxicolor)系稀有动物,为亚洲特产,分布于中国、印度、缅甸及不丹。中国的羚牛分布于川、陕、甘、滇、藏5省区,为分布区最广、数量最多、资源量丰富的国家(Wu,1985)。  相似文献
