本实验主要探究发酵乳杆菌AR497对DSS诱导的小鼠炎症性肠病的影响。发酵乳杆菌AR497在低pH、高胆盐浓度、高渗透压等极端条件下仍具有良好的生长能力,对大部分抗生素敏感;在C57BL/6J小鼠中研究其对由葡聚糖硫酸钠(DSS)诱导的结肠炎的缓解作用,发酵乳杆菌AR497处理后抑制小鼠体重减少,降低疾病活动指数,上调紧密连接蛋白基因Claudin 3,ZO-1和E-cadherin 1的表达。实验结果表明发酵乳杆菌AR497具有优良的生物学特性并可通过保护肠道屏障从而缓解DSS诱导的结肠炎。  相似文献

Ferulic andp-coumaric acid can be separated from their corresponding aliphatic methyl esters by capillary zone electrophoresis, which allows the convenient determination of feruloyl andp-coumaroyl esterase activities using synthetic esters as substrates. As a component of the array of enzymes produced by micro-organisms to deconstruct plant cell walls, feruloyl esterases hydrolyze phenolic groups involved in the cross-linking of arabinoxylan to other polymeric structures. This is important for opening the cell wall structure, making material more accessible to glycosyl hydrolases. Here, we describe the first crystal structure of the non-modular type-A feruloyl esterase from Aspergillus niger (AnFaeA) solved at 2.5A resolution. AnFaeA displays an alpha/beta hydrolase fold similar to that found in fungal lipases and different from that reported for other feruloyl esterases. Crystallographic and site-directed mutagenesis studies allow us to identify the catalytic triad (Ser133-His247-Asp194) that forms the catalytic machinery of this enzyme. The active-site cavity is confined by a lid (residues 68-80), on the analogy of lipases, and by a loop (residues 226-244) that confers plasticity to the substrate-binding site. The lid presents a high ratio of polar residues, which in addition to a unique N-glycosylation site stabilises the lid in an open conformation, conferring the esterase character to this enzyme. A putative model for bound 5,5'-diferulic acid-linked arabinoxylan has been built, pointing to the more relevant residues involved in substrate recognition. Comparison with structurally related lipases reveals that subtle amino acid and conformational changes within a highly conserved protein fold may produce protein variants endowed with new enzymatic properties, while comparison with functionally related proteins points to a functional convergence after evolutionary divergence within the feruloyl esterases family.  相似文献

研究植物源性发酵乳杆菌对免疫系统生理活性、抗高血糖和促进肠蠕动的影响.β-内啡肽对免疫系统有重要作用,阿片-促黑素细胞皮质素原(POMC)是β-内啡肽的前体物质,为了探讨发酵乳杆菌对免疫功能的影响,检测小鼠下丘脑和脑垂体中POMC的基因表达水平.通过喂食发酵乳杆菌时间和浓度的不同及测量β一内啡肽表达水平的方式,来分析血液中B-内啡肽的变化情况.此外,测定了三种小鼠模型的血糖变化,即常规的WT小鼠模型、D-葡萄糖诱导的急性高血糖模型和链唑霉素(STZ)诱导的糖尿病模型,以证实发酵乳杆菌的降血糖效果,并对降血糖效果进行了验证.通过检测血糖随喂食发酵乳杆菌浓度和时间而变化的情况,已证实给药浓度和时间对血糖浓度有很好的影响.另外,为证实发酵乳杆菌对肠蠕动的影响测定了胃肠通过时间,证明发酵乳杆菌对肠蠕动有极好的促进作用.通过此项研究,证实发酵乳杆菌对提高人体免疫系统的生理活性、抗高血糖和促进小肠蠕动有极佳的效果.  相似文献

Aims:  To evaluate the free radical-scavenging capacity of Lactobacillus fermentum and its effects on antioxidant enzyme levels in finishing pigs.
Methods and Results:  The free radical-scavenging activity of Lact. fermentum was analysed in vitro . The tested Lactobacillus showed a high scavenging ability against DPPH (1,1-diphenyl-2-picrylhydrazyl), superoxide and hydroxyl radicals which was dose dependent. Subsequently, 108 crossbred pigs weighing 20·67 BW, were allotted to dietary treatments including a basal diet or the basal diet supplemented with either aureomycin or 10·2 × 107  Lact. fermentum CFU g−1 diet. Supplementation of Lact. fermentum increased total antioxidant capacity ( P  < 0·01) in serum from 50 kg pigs, while serum superoxide dismutase ( P  = 0·01) and glutathione peroxidase ( P  < 0·01) increased, and malondialdehyde levels decreased ( P  < 0·01) in 90 kg pigs. Hepatic catalase ( P  = 0·04), muscle superoxide dismutase ( P  < 0·01) and copper–zinc-superoxide dismutase were enhanced ( P  = 0·01), whereas malondialdehyde levels were reduced ( P  = 0·05) by Lact. fermentum .
Conclusions:  The free radical-scavenging capacity of Lact. fermentum was dose dependent and its supplementation improved the antioxidant status of pigs.
Significance and Impact of the Study:  Lactobacillus fermentum could be used to alleviate oxidative stress and increase pig performance and improve pork quality.  相似文献

将foldon结构域与阿魏酸酯酶C-末端进行融合表达并用组氨酸标签对融合蛋白进行纯化。实现基于foldon的寡聚化阿魏酸酯酶及单体阿魏酸酯酶在毕赤酵母GS115中表达,并应用目标蛋白与foldon结构域融合可自发形成三聚体结构的特性对阿魏酸酯酶进行改造,以提高阿魏酸酯酶的催化性能。经纯化获得寡聚化及单体阿魏酸酯酶,寡聚化阿魏酸酯酶表观分子量约为110kDa,单体阿魏酸酯酶表观分子量为40 kDa;寡聚化阿魏酸酯酶的最适反应温度和pH分别为50℃和5.0,而单体阿魏酸酯酶则分别为50℃和6.0。寡聚化阿魏酸酯酶的底物亲和力(K_m)及催化效率(k_(cat)/K_m)较单体阿魏酸酯酶分别提高3.42倍和7.57倍。结果表明,寡聚化及单体阿魏酸酯酶均成功表达,且寡聚化阿魏酸酯酶在底物亲和力和催化效率上具有明显优势,该提高阿魏酸酯酶催化效率的方法简单、高效,有很好的应用前景。  相似文献

【目的】对黑曲霉(Aspergillus niger)阿魏酸酯酶基因进行克隆和密码子优化,使其在毕赤酵母(Pichia pastoris X-33)中高效表达。【方法】以黑曲霉基因组为模板,经重叠延伸PCR扩增得到阿魏酸酯酶基因(Anfae A),并对Anfae A基因进行毕赤酵母密码子偏好性"随机优化"和"一对一优化",全基因合成后分别与表达载体pPICZαA连接,构建表达载体pPICZαA-Anfae A、pPICZαA-op Anfae A I和pPICZαA-op Anfae A II。经Sac I线性化后电转化至P.pastoris X-33中,筛选阳性转化子。摇瓶发酵4.5 d后,测定并比较重组阿魏酸酯酶(re Anfae A)酶活。【结果】密码子优化前阿魏酸酯酶酶活为6.8±0.1 U/m L,基因"一对一优化"和"随机优化"后的重组酶酶活分别为5.2±0.1 U/m L和39.9±0.1 U/m L,"随机优化"后酶活比优化前提高了近6倍,而"一对一优化"后酶活仅为优化前酶活的76.5%。重组阿魏酸酯酶的最适p H为5.5,且在pH 4.5-7.0稳定性较好;最适反应温度50°C,在45-50°C较稳定。【结论】阿魏酸酯酶基因经密码子"随机优化"后进行重组表达,酶活显著提高,对研究阿魏酸酯酶在毕赤酵母及其它宿主中的高效表达具有一定的借鉴意义,也为大规模工业化应用奠定了基础。  相似文献

The addition of enzymes that are capable of degrading hemicellulose has a potential to reduce the need for commercial enzymes during biomass hydrolysis in the production of fermentable sugars. In this study, a high xylanase producing actinomycete strain (Kitasatospora sp. ID06-480) and the first ethyl ferulate producing actinomycete strain (Nonomuraea sp. ID06-094) were selected from 797 rare actinomycetes, respectively, which were isolated in Indonesia. The addition (30%, v/v) of a crude enzyme supernatant from the selected strains in sugarcane bagasse hydrolysis with low-level loading (1 FPU/g-biomass) of Cellic® CTec2 enhanced both the released amount of glucose and reducing sugars. When the reaction with Ctec2 was combined with crude enzymes containing either xylanase or feruloyl esterase, high conversion yield of glucose from cellulose at 60.5% could be achieved after 72 h-saccharification.  相似文献

生物胺是一种存在于发酵食品中的含氮小分子有机化合物,过量摄入可能引起过敏或其他不良反应。利用酶法降解是减少发酵食品中生物胺含量从而保障食品安全的有效方法之一。文中成功克隆了来源于发酵乳杆菌的多铜氧化酶基因,在大肠杆菌中表达的酶活水平为484 U/L。通过镍柱亲和层析方法获得了此多铜氧化酶的纯酶。该多铜氧化酶的最适反应温度为50℃,最适反应pH为3.5,其K_m为1.3 mmol/L,V_(max)为7.67×10~(-2) mmol/(L·min)。对酶的应用特性研究表明,来源于发酵乳杆菌的多铜氧化酶对18%(W/F)NaCl有一定的耐受性,并可降解包括色胺、苯乙胺、腐胺、尸胺、组胺、酪胺和亚精胺在内的7种生物胺。其中它对组胺和酪胺的降解能力最高,分别为51.6%和40.9%。此外,该酶对酱油中的生物胺也有普遍降解作用,使用较低酶量(500 U/L)时,对酱油中总胺的降解率达到10.6%。多铜氧化酶具备降解发酵食品中生物胺的潜力,为进一步实现这类食品酶的实际应用奠定基础。  相似文献

Four esterase genes and general esterase activity were investigated in the gut of the termite Reticulitermes flavipes. Two genes (RfEst1 and RfEst2) share significant translated identity with a number of insect JH esterases. The two remaining genes (RfEst3 and RfEst4) apparently code for much shorter proteins with similarity to fungal phenolic acid esterases involved in hemicellulose solubilization. All four genes showed consistently high midgut expression. This result was further supported by colorimetric activity assays and Native polyacrylamide gel electrophoresis, which showed significant esterase activity and a number of isoforms in the midgut. The greatest esterase activity and isoform composition were detected when α‐naphthyl propionate was used as a substrate. Moreover, esterase activity and diverse isoforms were present in gut mitochondrial, microsomal, and cytosolic sub‐cellular protein fractions, as well as in the hindgut lumen. These findings reveal an agreement between gut esterase gene expression and activity distributions, and support the idea that R. flavipes gut esterase activity is host (not symbiont)‐derived. In addition, these findings support the hypotheses that termite gut esterases may play important roles in lignocellulose digestion and caste differentiation. This study provides important baseline data that will assist ongoing functional‐genomic efforts to identify novel genes with roles in semiochemical, hormone, and lignocellulose processing in the termite gut. © 2009 Wiley Periodicals, Inc.  相似文献

为在毕赤酵母中表达来源于米曲霉Aspergillus oryzae的A型阿魏酸酯酶并研究其水解功能,探讨大孔树脂对其水解产物阿魏酸的纯化条件及纯化效果,以米曲霉A.oryzae CICC 40186总RNA为模板,通过RT-PCR技术克隆出了米曲霉阿魏酸酯酶A(AorFaeA)成熟肽的编码基因AorfaeA,并借助pPIC9K质粒实现了其在毕赤酵母GS115中的异源表达。SDS-PAGE分析结果显示纯化后的重组阿魏酸酯酶(reAorFaeA)为单一条带,其表观分子质量约39.0 kDa。以阿魏酸甲酯为底物,经高效液相色谱法测得该酶的最高酶活为58.35 U/mg。利用reAorFaeA和木聚糖酶复合酶水解去淀粉麦麸制备阿魏酸,用大孔树脂纯化小麦麸皮阿魏酸粗提液,所测树脂中HPD-300型大孔树脂的吸附量和解吸率较高,以50%的乙醇为洗脱液,当流速为1.0 mL/min时洗脱效果较好。在该纯化条件下,阿魏酸的回收率为92%,质量分数由原材料中的0.13%富集提高到10.55%。这些研究为阿魏酸的酶法"绿色生产"及应用奠定了坚实的理论基础。  相似文献

Pentylferulate synthesis was achieved at high yields (50–60%) with Aspergillus niger feruloyl esterase using a water-in-oil microemulsion system. The initial rate of synthesis decreased by 15–20% when the water content of the microemulsion was increased from 1.8 to 2.4% (v/v), although a concomitant decrease in conversion was not observed. The enzyme stability was significantly higher in the microemulsion than in an aqueous solution.  相似文献

AIMS: The aim of this study was to investigate the properties of temperate bacteriophage of Lactobacillus fermentum, based on its morphology, restriction patterns, protein profile and the impact on the growth of host strain. METHODS AND RESULTS: With Mitomycin C, seven temperate phages were induced from Lactobacilli derived from Chinese yogurt. The temperate phages induced belong to the most common Bradley's group B, having hexagonal head and long, noncontractile tail. They were furthermore confirmed to be the same bacteriophage by identical restriction patterns. SDS-PAGE profile showed that the phage studied had one major structure protein about 31.9 kDa. The presence of the prophage influenced the cell shape and colony size of its lysogenic strain. CONCLUSIONS: The phage obtained had similar, but not complete identical properties with other L. fermentum phages reported. It influenced the growth behaviour of its lysogenic strain. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides some information about bacteriophages occurring in the Chinese yoghurt manufacture and contributes to our knowledge on the bacteriophage diversity in the dairy industry.  相似文献

Aim: Lactobacillus fermentum is a widely utilized probiotic compound fed as an alternative to antibiotics for growth promotion in a wide variety of livestock species. The objective of this research is to develop an economical and practical fermentation medium for the growth of Lact. fermentum using response surface methodology. Methods and Results: A two‐level Plackett–Burman design was used to determine which factors in the fermentation medium influence the growth of Lact. fermentum. Under our experimental conditions, peptone, urea and yeast extract were found to be major factors. Then, the steepest ascent method and the central composite design were applied to optimize the culture of Lact. fermentum. The following composition of the fermentation medium was estimated to be the most economical formula (per litre): 30 g corn syrup, 15 g glucose, 14·4 g peptone, 7 g (NH4)2SO4, 0·5 g urea, 3 g sodium acetate, 4 g sodium citrate, 0·1 g MnSO4·4H2O, 0·5 g MgSO4·7H2O, 7·3 g yeast extract, 0·5 g K2HPO4. Conclusion: Based on 10 side‐by‐side comparisons, we found that the yield of Lact. fermentum using our fermentation medium was 64% greater than those using modified de Man, Rogosa and Sharp broth (MRS) medium (1·8 × 109 CFU ml?1vs 1·1 × 109 CFU ml?1, respectively), while the cost was 89% lower than MRS. This research indicates that it is possible to increase bacterial yield by using inexpensive materials. Significance and Impact of the Study: It is more likely that the use of Lact. fermentum as a probiotic will increase. The low cost medium developed in this research can be used for large‐scale, commercial application where economics are quite likely to be important.  相似文献

Aim: Technological properties of Lactobacillus fermentum isolates involved in spontaneous fermentation of dolo and pito wort were examined to select starter cultures. Methods and Results: 264 isolates were screened for antimicrobial activity, acidifying activity, exopolysaccharides (EPSs) and amylase production. An antimicrobial activity was detected for 33·3%, 31·8%, 22·7% and 15·9% of the isolates towards Staphylococcus aureus enterotoxin A producer, Staphylococcus aureus enterotoxin A and B producer, Escherichia coli and Listeria innocua, respectively. A similarity was found between the isolates which were clustered in four groups according to their rates of acidification of sorghum malt broth. The faster acidifying group of isolates (43·48%) had a rate of acidification evaluated as ΔpH of 1·14 ± 0·15 pH unit after 6 h of fermentation, followed by a second group of isolates (38·08%) with a similar rate of acidification after 9 h of fermentation. From the isolates endowed with an antimicrobial activity, 5·76% belonged to the faster acidifying group and 40·38% belonged to the second group. 88·7% of the isolates had the ability for producing EPSs but not amylase. Conclusion: Lactobacillus fermentum ferments dolo and pito wort by lowering the pH and providing organic acids, EPSs and antimicrobial compounds. Significance and Impact of the Study: Strains with a rapid rate of acidification, an antimicrobial activity and producing EPSs are suggested to have potential for starter cultures.  相似文献

Organic co-solvents can expand the use of enzymes in lignocellulose deconstruction through making substrates more soluble and thus more accessible. In choosing the most adequate co-solvent for feruloyl esterases, hydrolysis of methyl p-hydroxycinnamates by three pure enzymes (and a multi-enzyme preparation) was evaluated. Low concentrations of dimethylsulfoxide (DMSO) enhanced hydrolysis by two of the enzymes while at levels >20%, activity was reduced. DMSO also enhanced acetyl esterase-type activity of the enzymes. The co-solvent effect was different for each enzyme-substrate couple, indicating that other factors are also involved. Kinetic studies with a Talaromyces stipitatus feruloyl esterase showed low concentrations of dimethylsulfoxide enhanced the hydrolytic rate while Km also increased. Moreover, long-term incubation (96 h) of an Aspergillus niger feruloyl esterase in dimethylsulfoxide:water provided to the enzyme the ability to hydrolyze methyl p-coumarate, suggesting an active-site re-arrangement. Dimethylsulfoxide (10-30%) is proposed as an adequate co-solvent for feruloyl esterase treatment of water-insoluble substrates.  相似文献

A randomized double-blind, placebo-controlled trial was conducted to ascertain the intestinal microbiota-altering properties of LGG and L. gasseri TMC0356 (TMC0356) in Japanese cedar Cryptomeria japonica pollinosis patients. Fecal bacteria communities were examined before and after fermented milk administration using culture, FISH and T-RFLP methods. Test group subjects showed the presence of LGG and TMC0356 along with a significant increase in fecal lactobacilli ( P < 0.001) after giving LGG and TMC0356 fermented milk. Culture and FISH analysis revealed no significant changes in other intestinal bacterial groups. Each subject exhibited a characteristic T-RFLP profile pattern that varied quantitatively and qualitatively with JCP shedding. Profile changes were observed in 53% of placebo group subjects and in 21% of test group subject's post-administration, indicating that LGG and TMC0356 suppressed intestinal microbiota changes in JCPsis patients. The results suggest that intestinal microbiota might be more sensitive to exposure to environmental allergens than expected from the results of general culture method studies. Stabilization of intestinal microbiota by selected probiotic strains such as LGG and TMC0356 could be beneficial to homeostasis of the intestinal microbiota and useful in the management of JCPsis.  相似文献

Aims: To investigate the influence of heat‐killed Lactobacillus gasseri TMC0356 on changes in respiratory immune function and intestinal microbiota in a diet‐induced obese mouse model. Methods and Results: Male C57BL/6J mice were fed a high‐fat diet for 16 weeks. After 8 weeks, the high‐fat‐diet‐induced obese mice (DIO mice) were randomly divided into two 0067roups, the DIO and DIO0356 groups. DIO0356 group mice were orally fed with heat‐killed TMC0356 every day for 8 weeks, while DIO group mice were exposed to 0·85% NaCl over the same time period as controls. After intervention, the pulmonary mRNA expression of cytokines and other immune molecules in DIO0356 mice compared to those in DIO group mice was significantly increased (P < 0·05, P < 0·01). In faecal bacterial profiles, analysed using the terminal restriction fragment length polymorphism (T‐RFLP) method, T‐RFLP patterns in 75% of the DIO0356 group mice were apparently changed compared with those in control group mice. Conclusion: These results suggest that inactive lactobacilli may stimulate the respiratory immune responses of obese host animals to enhance their natural defences against respiratory infection, partially associating with their potent impact on intestinal microbiota. Significance and Impact of the Study: We have demonstrated that oral administration of inactive lactobacilli may protect host animals from the lung immune dysfunction caused by obesity.  相似文献
