Nine species of thermophilic fungi were obtained from retailed packets of imported and locally manufactured brands of cigarettes and from cured tobacco leaves. They include known human pathogens such asThermoascus aurantiacus Miehesensu Apinis,Mucor pusillus Lindt andMucor miehei Cooney and Emerson. Chaetomium thermophile La Touche,Humicola insolens Cooney and Emerson,Thermoascus crustaeus (Apinis and Chesters) Stolk andMucor miehei have not been previously reported on tobacco products.Fewer species were obtained from foreign cigarettes than from locally manufactured brands. A mean moisture content of 19% was recorded for imported cigarettes and 37% for Nigerian brands. The potential health hazards posed to man by the unrestricted use of tobacco products are discussed.  相似文献

The uptakes of high-affinity concentrations (10–8 M) ofmeta-tyramine (m-TA),para-tyramine (p-TA), and dopamine (DA) into rat striatal slices have been shown to be inhibited by DNP and ouabain. We now demonstrate that cocaine (5×10–6 M) and low concentrations of sodium ion (26×10–3 M) also reduced these uptakes. The spontaneous efflux and the release [induced by an elevated concentration of potassium ion (5×10–2 M)] of each of the previously accumulated amines were studied in the presence and absence of added calcium ions. The spontaneous efflux of each amine (especially the tyramines) was enhanced by the absence of calcium ions. Part of this enhancement seemed to be due to an inhibition of a calcium-dependent reuptake. The elevated concentration of potassium ion proved to be an effective releaser of each amine; and for DA, such release was decreased by the removal of calcium. Form- andp-TA, however, the removal of calcium either did not reduce or completely abolished the releases depending upon the duration of the calcium removal. The fungitoxic action of alkoxy ethyl mercuric acetates and phenyl mercuric acetate was studied withAspergillus niger, Penicillium italicum andUstilago maydis as test moulds. Carbonic anhydrase II electrophoretic patterns were investigated in 3113 animals belonging to 12 genera and 24 species of New World primates. Polymorphism was detected in 13 species. A total of 24 different alleles was postulated to explain the variability found; the genusAotus showed the highest (eight) number of such alleles. Three genera of the family Callitrichidae (Callithrix, Saguinus, andCebuella) presented five alleles that were not found among the Cebidae. Important markers at the generic level were observed inCallicebus (CA2 *6 andCA2 *12),Cebus (CA2 *10, CA2*16, andCA2 *21), andAotus (CA2*3, CA2*4, CA2*5, CA2*9, CA2*15, CA2*17, CA2*22, CA2*23). CA2*13 seems to be the most common allele among the Cebidae; six genera of this family showed frequencies higher than 70% of it.  相似文献

Expression of the bacterial luciferase (lux) system in mammalian cells would culminate in a new generation of bioreporters for in vivo monitoring and diagnostics technology. Past efforts to express bacterial luciferase in mammalian cells have resulted in only modest gains due in part to low overall expression of the bacterial genes. To optimize expression, we have designed and synthesized codon-optimized versions of the luxA and luxB genes from Photorhabdus luminsecens. To evaluate these genes in vivo, stable HEK293 cell lines were created harboring wild type luxA and luxB (WTA/WTB), codon-optimized luxA and wild type luxB (COA/WTB), and codon-optimized versions of both luxA and luxB genes (COA/COB). Although mRNA levels within these clones remained approximately equal, LuxA protein levels increased significantly after codon optimization. On average, bioluminescence levels were increased by more than six-fold [5×105 vs 2.9×106 relative light units (RLU)/mg total protein] with the codon-optimized luxA and wild type luxB. Bioluminescence was further enhanced upon expression of both optimized genes (2.7×107 RLU/mg total protein). These results show promise toward the potential development of an autonomous light generating lux reporter system in mammalian cells  相似文献

To clarify the ecological significance of the association of sulfate-reducing bacteria (SRB) with sediment particle size, SRB utilizing lactate (l-SRB), propionate (p-SRB) and acetate (a-SRB) were examined with different sizes of sediment particles in a hypertrophic freshwater lake using the anaerobic plate count method. The numbers ofl-SRB anda-SRB were 104–105 colony forming units (CFU) per ml in the 0–3 cm layer and 102–103 CFU ml−1 in the 10–13 cm layer while the numbers ofp-SRB were one or two orders lower than those ofl-SRB anda-SRB. A sediment suspension was fractionated into four fractions (<1, 1–10, 10–94 and >94 μm). The highest proportions ofl-SRB anda-SRB were found in the 10–94 μm fraction: 66–97% forl-SRB and 53–98% fora-SRB. The highest proportion ofp-SRB was found in the >94 μm fraction (70–74%). These results indicate that most SRB were associated with sediment particles. One isolate from an acetate-utilizing enrichment culture was similar toDesulfotomaculum acetoxidans, a spore-forming sulfate-reducing bacterium. When lactate and sulfate were added to sediment samples,l-SRB anda-SRB in the <10 μm-fraction grew more rapidly than those in whole sediment for the first 2 days. This result suggests that nutrients uptake by free-living and small particle-associated (<10 μm) SRB is higher than that by SRB associated with larger particles.  相似文献

The meiobenthos of five mangrove vegetation types in Gazi Bay,Kenya   总被引:1,自引:0,他引:1  
The vertical distribution of meiofauna in the sediments ofAvicennia marina,Bruguiera gymnorrhiza,Ceriops tagal,Rhizophora mucronata andSonneratia alba at Gazi Bay (Kenya), is described. Seventeen taxa were observed, with highest densities in the sediments ofBruguiera (6707 ind. 10 cm–2), followed byRhizophora (3998 ind. 10 cm–2),Avicennia (3442 ind. 10 cm–2),Sonneratia (2889 ind. 10 cm–2) andCeriops (1976 ind. 10 cm–2). Nematodes accounted for up to 95% of total densities; other common taxa were copepods, turbellarians, oligochaetes, polychaetes, ostracods and rotifers. High densities occurred to about 20 cm depth in the sediment. EspeciallyCeriops sediments show still high densities of nematodes (342 ind. 10 cm–2) and copepods (11 ind. 10 cm–2) in the deepest layer (15–22 cm). Particle size and oxygen conditions were major factors influencing meiobenthic distribution;Uca burrows had a major impact on distribution and abundance of meiofauna.  相似文献

The relative importance of autotrophic flagellates, desmids, cyanobacteria, and ciliates as food forDaphnia magna was examined using cohort life tables. Each cohort was fed a single food type at a given concentration, and comparisons among each type were made. Algal feeding treatments included three levels of young (7 to 14 days old)Chlamydomonas reinhardi (Chlorophyta, Chlamydomonadacae), two levels of senescent (> 14 days old)C. reinhardi, two levels ofCryptomonas sp. (Chlorophyta, Cryptomonadacae), two levels ofStaurastrum sp. (Chlorophyta, Desmidacae), four levels of young (7 to 15 days old) or senescent (> 15 days old)Microcystis aeruginosa (Cyanophyta, Chlorococcacae), and a no-food treatment. The ciliatesCyclidium sp. andParamecium caudatum were also presented at concentrations of 1 or 102 cells/ml, as well as mixtures ofC. reinhardi (103/ml) andCyclidium (1/ml) orP. caudatum (1/ml).Daphnia growth, reproduction, and survivorship were highest whenC. reinhardi orCryptomonas were the food source, while those starved or fedM. aeruginosa had shorter survivorship and lower growth and reproduction.Daphnia grew and had high survivorship when fedP. caudatum, but even though eggs were produced, most were aborted after 2 or 3 days.Staurastrum andCyclidium produced intermediate growth and survivorship, but reproduction was seen only in the 103 Staurastrum/ml treatment. Carbon and nitrogen content were general indicators of nutritional value. However, growth, reproduction, and survivorship were higher in some cohorts fed treatments containing relatively low levels of carbon and nitrogen. Other cohorts were short-lived and did not reproduce, despite being fed much higher levels of carbon and nitrogen. The results also suggest that green algae are nutritionally valuable forDaphnia, whereas cyanobacteria are not. As measured by life-table parameters, the nutritional value of ciliates was variable, with some being poor food sources. Thus, the potential of ciliates as a trophic link between microbial production and higher trophic levels may vary with the ciliate community structure. Our results suggest that ciliates alone were insufficient as a food source to supportDaphnia population growth.  相似文献

This study was performed to validate the shelf life of commercial sachet-packed drinking water produced in the Benin City metropolis, Edo State, Nigeria. Seven brands of sachet-packed water that were freshly produced by manufacturers were collected from respective factories and subjected to standard physicochemical and microbial tests. The colour of all water brands (0·0–5·6 HU) was within the limits recommended by the World Health Organization (WHO) (≤15 HU) while the temperature (26·9–28·4°C) was above the limits recommended by WHO (≤25·0°C). The pH reported in brands 1 and 6 at week 8 of storage was below WHO recommended limits (6·5–9·5). At week 8 of storage, brands 1 and 6 had HPC (3·97–4·70 log10 CFU per ml) that were above WHO/National Agency for Food and Drug Administration and Control (NAFDAC) recommended limits (≤2 log10 CFU per ml) while TCC (<1 MPN 100 ml−1) in all brands were within recommended limits (≤10 MPN 100 ml−1). No thermo-tolerant Coliforms and Cryptosporidium were present in all brands; though, Streptococcus faecalis was detected in brand 6. Based on WHO/NAFDAC specifications for the examined parameters, brands 1 and 6 were inferred to not comply with the recommended shelf life of 2 months (approximately 8 weeks). Hence, there is a need for strict compliance to NAFDAC-specified Good Manufacturing Practice by these processing factories to prevent probable adverse health effects.  相似文献

The selective effect of DCMU on photosynthetic activity and growth rate was examined in several marine unicellular algae:Nannochloropsis sp. (Eustigmatohyceae),Dunaliella salina (Chlorophyceae)Isochrysis galbana (Prymnesiophyceae) andChaetoceros sp. (Bacillariophyceae). DCMU at 10–7 M caused an immediate decrease in the photosynthetic rate ofDunaliella andIsochrysis (about 50% inhibition), whereas 10–6 M imposed 80% inhibition in the photosynthetic rate ofChaetoceros. InNannochloropsis the rate was affected only when DCMU concentration exceeded 10–6M. Cellular growth rate of all studied algae was affected by DCMU in a similar manner to photosynthesis. The differential effect of DCMU was further examined in mixed cultures in whichNannochloropsis was cultivated together with an additional species simulating a contamination situation of aNannochloropsis culture. When DCMU was added at concentrations higher than 10–7 M, the growth of the competing algae significantly decreased, whileNannochloropsis maintained a relatively high growth rate. Consequently, after a growth period of 4 to 7 days a clear domination ofNannochloropsis was observed. These results demonstrate that DCMU and probably other herbicides of similar characteristics can be used effectively as a selective tool to suppress contaminating unicellular algae in open ponds in order to maintain a monoculture ofNannochloropsis.  相似文献

Vast numbers of spores of the thermophilic actinomycetesExcellospora flexuosa,Thermomonospora alba,T. curvata andT. fusca were collected from the air in fermentation tunnels during the spawning of mushroom compost, i.e. over 109 CFU m–3 of air. Five different genera of fungi, namely,Aspergillus,Aureobasidium,Cladosporium,Penicillium andScytalidium, were found at only 103 CFU m–3 of air.Agaricus bisporus, used for spawning, was absent. Sera of 10 mushroom growers affected by Mushroom Worker's Lung (MWL) were tested by a qualitative dot-ELISA for antibodies against the spores of these micro-organisms. All 10 were positive for one or more of the actinomycetesE. flexuosa,T. alba,T. curvata andT. fusca. No antibodies were found againstStreptomyces thermovulgaris,Thermoactinomyces vulgaris andT. sacchari nor against the fungiAspergillus fumigatus,Penicillium brevicompactum,P. chrysogenum,Scytalidium thermophilum andTrichoderma viride. Sera of 11 of 14 workers engaged in routine spawning of compost in tunnels reacted positively with 1 or more of the actinomycetes. Their10log serum titres increased with the duration of employment to an upper limit of 2.5. The sera of 19 non-exposed individuals were negative. Because high numbers of spores ofE. flexuosa,T. alba,T. curvata andT. fusca were present in the air that was used for successful inhalation provocation of mushroom workers with MWL and because of the elevated serum titres of these workers, we presume these organisms to contribute in the occurrence of MWL.  相似文献

The pattern of L-alanine uptake in isolated cells of interscapular brown adipose tissue has been determined. The uptake can be divided into the diffusion component (Kd=0.55 min–1) and a saturable Na+-dependent transport (K M =0.87 mM andV max=155 nmol/min/106 cells). The saturable component can be subdivided into MeAIB-sensitive (K M =1.63 mM andV max=162 nmol/min/106 cells) and MeAIB-insensitive (K M =3.2 mM andV max=39.5 nmol/min/106 cells). The influence of Ca homoionic clay minerals (montmorillonite, illite, and kaolinite) on the activity,K m , andV m values of acid phosphatase was examined in model experiments. At each substrate (p-nitrophenyl phosphate) level tested, the addition of increasing amounts of clays (50, 100, and 150 mg, respectively) decreased the activity and increased theK m value from 1.43×10–3 m PNP (in the soluble state) to 82.3×10–3M (montmorillonite), 8.02×10–3 m (kaolinite), and 7.65×10–3 m (illite) at the 150 mg level. The maximum enzyme reaction velocity (V m ) remained nearly constant at different amounts of kaolinite and illite, but increased remarkably with rising quantities of montmorillonite. Apparently, the substrate affinity of sorbed acid phosphatase is significantly lower with montmorillonite than with kaolinite or illite. This may be ascribed to an intensive sorption of both substrate and enzyme to the surface as well as to interlattice sites of montmorillonite.  相似文献

Antarctic fishes were sampled with 41 midwater and 6 benthic trawls during the 1999–2000 austral summer in the eastern Ross Sea. The oceanic pelagic assemblage (0–1,000 m) contained Electrona antarctica, Gymnoscopelus opisthopterus, Bathylagus antarcticus, Cyclothone kobayashii and Notolepis coatsi. These were replaced over the shelf by notothenioids, primarily Pleuragramma antarcticum. Pelagic biomass was low and concentrated below 500 m. The demersal assemblage was characteristic of East Antarctica and included seven species each of Artedidraconidae, Bathydraconidae and Channichthyidae, ten species of Nototheniidae, and three species each of Rajidae and Zoarcidae. Common species were Trematomus eulepidotus (36.5%), T. scotti (32.0%), Prionodraco evansii (4.9%), T. loennbergii (4.7%) and Chaenodraco wilsoni (4.3%). Diversity indices were highest for tows from 450 to 517 m (H=1.90–2.35). Benthic biomass ranged from 0.7 to 3.5 t km–2. It was generally higher in tows from 450 to 517 m (0.9–2.0 t km–2) although the highest biomass occurred at an inner-shelf station (238 m) due to large catches of T. eulepidotus, T. scotti and P. evansii.  相似文献

The larval stages ofAblattaria arenaria were provided with 4 different snail species:Monacha syriaca (Ehrenberg),Xeropicta derbentina (Krynicki),Candidula sp., andZebrina eburnea (Pfeiffer) to determine if the prey species affected developmental time and food preference of larvae. Functional response of each larval stage ofA. arenaria was also tested for increasing density ofX. derbentina, the most common prey species found in association withA. arenaria locally. The developmental time of each larval stage did not show any statistical difference when fed with different snail species. The total developmental time from egg hatch to adult emergence was 19.0, 19.1, 18.0 and 21.4 days for prey speciesM. syriaca, X. derbentina, Candidula sp., andZ. eburnea, respectively. When prey was offered to larvae either as a single species or as combination of several species,M. syriaca was the most preferred. The prey least consumed wasCandula sp. when prey was given separately, andZ. eburnea was least preferred when other prey species were present in the arena. The 3rd larval stage did not eat anyZ. eburnea if other prey species were present. The amount of prey consumed by the 1st larval stage did not show any statistical differences with increasing density ofX. derbentina. But the response of 2nd and 3rd larval stages was very similar to each other although the amount of prey they consumed was very different. They both showed a rapid increase in consumption rate at early densities, then a negatively but slowly accelerated rise to plateaus at higher densities, a type-2 functional response curve. All larval stages were very sensitive to starvation. Mortality started after the 2nd day, and all individuals of all larval stages were dead by the 5th day.   相似文献

Summary Activation of protein kinase C has been shown to cause both stimulation and inhibition of transport processes in the brush-border membrane and renal tubule. This study was designed to examine the dose-response nature and time-dependent effect of 4 -phorbol-12-myristate-13-acetate (PMA) on the rates of bicarbonate absorption (J HCO3) and fluid absorption (J v) in the proximal convoluted tubule (PCT) of rat kidney. Bicarbonate flux was determined by total CO2 changes between the collected fluid and the original perfusate as analyzed by microcalorimetry. Luminal perfusion of PMA (10–10 10–5 M) within 10 min caused a significant increase ofJ HCO3 andJ v. A peaked curve of the dose response was observed with maximal effect at 10–8 M PMA on both bicarbonate and fluid reabsorption, which could be blocked completely by amiloride (10–3 m) and EIPA (10–5 M). On the other hand, with an increase of perfusion time beyond 15 min, PMA (10–8 and 10–6 M) could inhibitJ HCO3 andJ v. Amiloride (10–3 M) or EIPA (10–5 M) significantly inhibitsJ HCO3 andJ v, while there is no additive effect of PMA and amiloride or EIPA on PCT transport. An inactive phorbol-ester, 4-phorbol, that does not activate protein kinase C, had no effects onJ HCO3 andJ v. Capillary perfusion of PMA (10–8 M) significantly stimulate bothJ HCO3 andJ v; however, PMA did not affect glucose transport from either the luminal side or basolateral side of the PCT. These results indicate that activation of endogenous protein kinase C by PMA could either stimulate or inhibit both bicarbonate and fluid reabsorption in the PCT dependent on time and dose, and these effects are through the modulation of Na+/H exchange mechanism.  相似文献

Fungal particles can be considered a potential risk factor in children causing asthma, rhinitis, and allergy. However, a direct relationship between mold exposure and respiratory symptoms is difficult to establish, particularly if volumetric results are not well documented, as is often the case in Mexico. In order to assess mold exposure in some asthmatic childrens’ homes, eight asthmatic children were selected. For each child, lung function was measured by spirometry and the flow expiratory volume drop in the first second (FEV1) as well as the CVF were estimated at 5, 10 and 15 min, after a running period on a treadmill. Exercise induced asthma (EIA) was defined as a FEV1 drop <10% or FEV1/CVF drop >10%. Atopy was estimated by skin testing of allergen extracts. Each room was volumetrically sampled using Burkard Personal samplers, every week for a month and with different degrees of activity. Fungal composition was mostly dominated by allergenic molds, spores smaller than 10 μm and non-seasonal molds. Concentrations ranged from 0 to 2087 CFU/m3 and 102 553–29 522 200 spores/m3, being significantly higher in indoor air, but the differences between rooms or weeks were not significant, either for CFUs (P>0.27) or for total particles (P>0.80). In general, mold counts were dominated byPenicillium spp. andCladosporium spp. especiallyP. aurantiogriseum andC. cladosporioides. We observed that the median concentration values were in general higher for children with exercise induced asthma (EIA). But the differences were significant only for EIA and degree, of asthma particularly for total (P=0.038), mycotoxin producers (P<0.009), seasonal molds (P<0.006) andPenicillium spp. (P=0.012). Total spore counts showed the highest median values in children without EIA, moderate asthma degree and no atopy. Significant differences were found for almost all spore, type groupings when comparing the presence of EIA (P<0.05) or asthma degree categories (P<0.030), but no differences were observed for atopy (P>0.21). We conclude that respiratory symptoms as described by EIA, asthma severity and atopy may give a good correlation with fungal concentrations and especially comparisons based on their physical and chemical properties.  相似文献

Densities ofAmblyomma americanum (L.) onBos indicus, B. taurus andB. indicus x B. taurus cattle are compared over a 3-year period, and the growth rate (rate of increase or decrease) of parasitic tick populations on each cattle genotype is estimated.Average log10 densities of parasiticA. americanum larvae are significantly (P=0.05) lower onB. indicus cattle than onB. taurus andB. indicus x B. taurus cattle. Average log densities of nymphal and adult ticks onB. taurus cattle are significantly higher than onB. indicus cattle but neither cattle genotype differs in this regard fromB. indicus x B. taurus cattle.Estimated annual tick population growth rates (log10) for parasiticA. americanum are positive onB. taurus cattle (+0.84 larvae, +0.09 nymphs, +0.22 adults calf–1 year–1), but are negative onB. indicus (–0.18 nymphs, –0.14 adults calf–1 year–1) andB. indicus x B. taurus cattle (–0.45 larvae, –0.24 nymphs, –0.14 adults calf–1 year–1). The major histocompatibility complex (Mhc) consists of class I and class II genes. In the humanMhc (HLA) class II genes, nineDRB loci have been identified. To elucidate the origin of these duplicated loci and allelic divergences at the most polymorphicDRBI locus, introns 4 and 5 as well as the 3′ untranslated region (altogether approximately 1,000 base pairs) of sevenHLA-DRB loci, threeHLA-DRBI alleles, and nine nonhuman primateDRB genes were examined. It is shown that there were two major diversification events inHLA-DRB genes, each involving gene duplications and allelic divergences. Approximately 50 million years (my) ago,DRBI *04 and an ancestor of theDRB1 *03 cluster (DRBI *03, DRBI*15, andDRB3) diverged from each other andDRB5, DRB7, DRB8, and an ancestor of theDRB2 cluster (DRB2, DRB4, andDRB6) arose by gene duplication. Later, about 25 my ago,DRBI *15 diverged fromDRBI*03, andDRB3 was duplicated fromDRBI *03. Then, some 20 my ago, the lineage leading to theDRB2 cluster produced two new loci,DRB4 andDRB6. TheDRBI *03 andDRBI *04 allelic lineages are extraordinarily old and have persisted longer than some duplicated genes. The orthologous relationships ofDRB genes between human and Old World monkeys are apparent, but those between Catarrhini and New World monkeys are equivocal because of a rather rapid expansion and contraction of primateDRB genes by duplication and deletion. The ability ofPseudomonas fluorescens, Escherichia coli andAcinetobacter radioresistenns to remove phosphate during growth was related to the initial biomass as well as to growth stages and bacterial species. Phosphate was removed by these bacteria under favourable conditions as well as under unfavourable conditions of growth. Experiments showed a relationship between a high initial cell density and phosphate uptake. More phosphate was released than removed when low initial cell densities (102–105 cells ml–1) were used. At a high initial biomass concentration (108 cells ml–1), phosphate was removed during the lag phase and during logarthmic growth byP. fluorescens. Escherichia coli. at high initial biomass concentrations (107 cells ml–1), accumulated most of the phosphate during the first hour of the lag phase and/or during logarithmic growth and in some cases removed a small quantily of phosphate during the stationary growth phase.Acinetobacter radioresistens, at high initial cell densities (106, 107 cells ml–1) removed most of phosphate during the first hour of the lag phase and some phosphate during the stationary growth phase.Pseudomonas fluorescens removed phosphate more thanA. radioresistens andE. coli with specific average ranges from 3.00–28.50 mg L–1 compared to average ranges of 4.92–17.14 mg L–1 forA. radioresistens and to average ranges of 0.50–8.50 mg L–1 forE. coli.  相似文献
