Summary The portion of the life cycle ofAcetabularia mediterranea from cyst formation to gamete release is described. During maturation, the cyst nucleus undergoes a series of mitoses in which the nuclear membrane remains intact, and the spindle microtubules are confined to the intranuclear space. There is a dark requirement for the completion of cyst maturation at the end of which the nuclei are in groups and centrioles are present. The final migration of the nuclei, cleavage of the cytoplasm into gametes, and the development of flagella is cxtremely rapid, taking six to twelve hours. Cysts which fail to germinate are blocked at a late stage in the maturation process, either before or after the final nuclear division.The observations are related to mitosis in other green algae (Chlorophyceae) and to gametogenesis inBryopsis hypnoides. Summary InAllomyces neo-moniliformis meiosis takes place during resting sporangium germination. The meiospores are characteristically binucleate and biflagellate as described byEmerson (1938) andTeter (1944). A variation in the number of nuclei and flagella per meiospore from two is correlated with germination of the resting sporangia under reduced oxygen tension. The meiospores are extremely poor swimmers and are typically amoeboid. At encystment the gamma bodies of the cell are mobilized and appear involved in cyst wall synthesis. A single mitotic division of each nucleus gives rise to four nuclei. Gamete cleavage is as described for spore cleavage inBlastocladiella (Lessie andLovett 1968). The assembly of the nuclear cap and side body complex of the spore are extremely late processes in gametogenesis. The gametes are released when the single papilla dissolves. The gametes fuse in pairs and after zygote formation the cell is uninucleate with two flagella. The biflagellate zygote is an active swimming cell. The presence of homothallism or hetero-thallism inA. neo-moniliformis is discussed.  相似文献

Gametogenesis in male and female gametophytes was studied by light microscopy and EM in the dioecious multinucleate green alga Derbesia tenuissima (Moris & De Notaris) P. Crouan & H. Crouan, where male and female gametes differ in size. Gametogenesis was divided into five stages: 32 h (stage 1), 24 h (stage 2), 16 h (stage 3), 8 h (stage 4), and 0.5 h (stage 5) before gamete release. At stage 1, the first sign of gametogenesis observed was the aggregation of gametophyte protoplasm to form putative gametangia. At stage 2, gametangia were separated from the vegetative protoplasm of gametophytes. Morphological changes of nuclei and organelles occurred at this early stage of male gametogenesis, and organelle DNA degenerated. At stage 3, male organelle DNA had completely degenerated, whereas in female gametangia, organelle DNA continued to exist in both chloroplasts and mitochondria. Gametogenesis was almost completed at stage 4 and fully at stage 5. Small male gametes had a DNA‐containing nucleus and a large mitochondrion and one or several degenerated chloroplasts. The mitochondria and plastids were devoid of DNA. The large female gametes had a nucleus and multiple organelles, all of which contained their own DNA. Thus, degeneration of chloroplast DNA along with morphological changes of organelles occurred at male gametogenesis in anisogamous green algae (Bryopsis and D. tenuissima), in contrast with previous studies in isogamous green algae (Chlamydomonas, Acetabularia caliculus, and Dictyosphaeria cavernosa) in which degeneration of chloroplast DNA occurred after zygote formation.  相似文献

Summary Gametes and zygotes of Acetabularia have been studied with the electron microscope. The two flagella of the gamete enter the cell obliquely and the roots run near the surface of the organism. No connexion between the eyespot and the flagella was found nor was there regular relationship between the two eyespots of the zygote. When zygotes were fixed some hours after fusion of the gametes the eyespots appeared to be within the same chloroplast.Some zygotes had two nuclei or dumb-bell shaped nuclei suggesting stages of fusion. The initial contact between nuclei appeared to be through a narrow gap formed by fusion of the two nuclear membranes.  相似文献

Gametogenesis in Calcarea is still poorly known; therefore, in this work, we describe the gametogenesis of Paraleucilla magna (Porifera, Calcarea) using light and electron microscopy. Male and female gametes derived from choanocytes. Oocytes grew inside a follicle-like structure composed of pinacocyte-like cells and choanocytes. They were nourished through nutrient transfer from nurse cells and through bacteria phagocytosis, which was never documented for a calcarean species. Mature oocytes were typical, spherical, with nucleolated nuclei and a cytoplasm filled with different types of inclusions and digestive vesicles. Nuage was also found. Spermatogenesis occurred in the lumen of differentiated choanocyte chambers, and spermatic cysts were never formed. Spherical spermatogonia without flagellum or collar were derived from hourglass-shaped primordial germ cells. Spermatozoa were of the ‘aberrant type’, showed a compact electron-dense nucleus and a homogeneous granulated body in its cytoplasm. The gametogenesis of P. magna is similar to what has been observed for other Porifera and Eumetazoa.  相似文献

Mitosis in Mantoniella squamata (Manton and Parke) Desikachary, a small scale-covered green monad, is presented. Organelle replication precedes nuclear division and begins with the replication of the chloroplast. As the chloroplasts separate, the Golgi and flagellar apparatuses divide. The discoid microbody enlarges and becomes ‘V'-shaped, with the arms extending toward depressions in the pyrenoid stalks of the chloroplasts. At prophase, microtubules produced by an amorphous microtubule organizing center enter the nucleus via polar fenestre. The nuclear membrane remains intact. As the chloroplasts migrate further apart, the spindle pole-to-pole distance increases. By metaphase, daughter-cell lobes are discernible as a cleavage furrow, which appears as early as prophase, and begins to incise the cell. A single Golgi apparatus is situated near the spindle pole; the flagellar apparatus lies adjacent to the pole. The cleavage furrow continues to constrict the cell, resulting in a narrowing isthmus containing the elongate microbody, nucleus and a rootlet system connecting the basal bodies of the daughter flagella. At telophase, no extra-nuclear microtubular systems other than the previously observed rootlet are present and the nuclei remain separated from each other. In cells undergoing multiple divisions to produce more than two daughter cells, the orientation of organelles changes somewhat, with the basal bodies and the Golgi apparatus separating daughter nuclei prior to the onset of cytokinesis. The mechanics of mitosis in Mantoniella are compared with other green monads and the evolutionary implications discussed.  相似文献

SYNOPSIS. During gametogenesis mother individuals of Hastigerina pelagica (d'Orbigny) undergo significant morphological changes. Thirty h before gamete release, the cytoplasm changes from pale orange to bright red, possibly due to transport of stored lipids from the inner region to more peripheral parts of the cytoplasm. During the next 10 to 15 h the bubble capsule which surounds the calcareous shell is discarded. After all bubbles have disappeared, the individual sheds its spines by resorbing the spine bases close to the shell surface. A single mother nucleus divides into some hundreds of thousands of gamete nuclei within a span of ~ 20 h. A bulge of cytoplasm is extruded from the aperture and increases in size during the next 5 to 10 h. This bulge consists of cytoplasmic strands in which gametes and spherical bodies are embedded. The gametes and spherical bodies mature and are released during the afternoon and early evening. The gametes have 2 unequal acronematic flagella. A previously undescribed structure in foraminiferal reproduction is the spherical body which consists of a large vacuole surrounded by a thin cytoplasmic layer in which several nuclei, various typical cell organelles and multiple flagella are present. The spherical bodies are believed to play a role as receptacles of waste material, possibly including residual digestive enzymes, thereby protecting the gametes from lysis during the reproductive process. Fusion of gametes and further development into the next generation have not been observed.  相似文献

The main structural features of male gametogenesis in Fucus serratus L. are described. The antheridial parent cells exhibit meristematic characteristics: large nucleus, dense cytoplasm, chloroplasts with few thylakoids. Synaptonemal complexes are frequently observed in the nuclei at the same time that microtubules are very abundant around the centrioles. Meiotic spindles are intranuclear, as are the mitotic spindles of the subsequent synchronous karyokineses. A nuclear membrane is present in all stages observed. Antheridial maturation is accompanied by multiplication of mitochondria and plastids as well as development of two major kinds of “vacuoles.” Cytokinesis proceeds with the progressive fusion of one of these types of vacuoles: this mode of cytoplasmic cleavage is unique among algae. Formation and differentiation of characteristic spermatozoid organelles (flagella, eyespot, proboscis) are also described.  相似文献

Transmission electron microscopic examination of Cephaleuros virescens Kunze growing on leaves of Camellia sp. indicates that gametes are similar to those of Trentepohlia aurea. The gametes bear two, smooth isokont “keeled” flagella containing typical “9 + 2” axonemes and lacking scales. Flagellar insertion is apical and the parallel basal bodies overlap laterally. Each basal body is associated with a separate multilayered structure and component microtubular spline. The latter extends posteriorly beneath the plasmalemma. A nucleus, mitochondria, chloroplasts, and cytoplasmic haematochrome droplets are present. Pyrenoids and eyespots are absent. The subcellular components of C. virescens gametes are comparable to those found in gametes of T. aurea; however, the arrangement of basal bodies and multilayered structures differs slightly from that in T. aurea. Comparison of the fine structure of gametes from Cephaleuros, Phycopeltis, and Trentepohlia clearly indicates that the (1) mode of flagellar insertion, (2) morphology, number, and arrangement of multilayered structures, and (3) keeled flagella are common to these three genera and, thus far, unique among the green algae. Although flagellar insertion is apical, it is not bilaterally symmetrical (sensu stricto), nor is it asymmetrical (cf. Chara and Nitella sperms). The arrangement may be termed “reversed bilateral symmetry” and standardization of the terminology is recommended.  相似文献

Patterns of reproduction were investigated in some microalgal species of Chlorophyceae (Botryosphaerella sudetica, Neochloris aquatica, Neochloris vigensis, Bracteacoccus minor). Under continuous light, the microalgae reproduced asexually producing autospores. However, appropriate manipulation of external conditions led to a change in the reproduction pattern towards production of zoospores or gametes. Production of zoospores and gametes was inhibited by light; motile cells emerged when microalgae were cultivated in darkness. The period of dark treatment necessary for zoosporogenesis or gametogenesis differed substantially among species that were tested. Sexual reproduction was observed in Neochloris vigensis and Bracteacoccus minor, whose generative life cycle had not been previously reported. The morphology of motile cells, the mode of sexual reproduction, and the efficiency of both the production of motile stages and mating events, were investigated. In order to gain detailed insights into patterns of reproduction, Botryosphaerella sudetica was selected for investigation under different light treatments. Non-actinic red light applied in the early phase of dark cultivation (up to 2 h) suppressed both zoosporogenesis and gametogenesis. However, after a 3-h dark pre-treatment, red light treatment had no effect on zoosporogenesis or gametogenesis. In contrast, non-actinic blue light did not block zoosporogenesis or gametogenesis, regardless of the time of treatment. The possible role of a red-light photoreceptor in zoosporogenesis and gametogenesis is discussed.  相似文献

ABSTRACT Specimens of Pelomyxa palustris from five collecting sites had numerous nonmotile flagella. The structures are called flagella because of morphological similarities to flagella and because P. palustris has affinities with amoeboid flagellates. Flagella were photographed on living cells and studied by transmission and scanning electron microscopy. From 64 to 742 flagella per cell were estimated from scanning electron microscopy of ten cells 204 to 1269 μm in length. The nonmotile flagella arise from basal granules which were, in one strain, surrounded by radiating electron-dense microtubules. This strain also had excess axonemal microtubules. Abundant cytoplasmic microtubules were arranged in several different patterns. In about half of the P. palustris cells in which nuclei were studied, microtubules were either apposed to the nuclear membrane in a parallel alignment (with some also radiating) or radiating from the nuclear membrane (with none parallel). Bacteria associated with nuclei were of three characteristic types: Gram-negative rods, Gram-positive rods, and large rods. All nuclei within a given trophozoite had similar perinuclear features. Recent proposals for separation of Pelomyxa to its own phylum (based on its proposed primitive, unique nature) can not be justified. Pelomyxa is a complex, highly specialized organism adapted to live in a specific fresh-water environment. Mastigamoebid amoeboid flagellates of the genera Mastigamoeba, Mastigella, Mastigina, and possibly Dinamoeba are placed with Pelomyxa within the order Pelobiontida Page, 1976, emend., containing two families. The unicellular green alga Chlamydomonas reinhardtii is able to take up methylammonium/ammonium from the medium at different stages of its sexual life cycle. Vegetative cells and pre‐gametes mostly used a low‐affinity system (LATS) component, but gametes obtained after light treatment of N‐deprived pre‐gametes expressed both LATS and high‐affinity system (HATS) components for the uptake of methylammonium/ammonium. The activity of the LATS component was stimulated by light in only 5 min in a process independent of protein synthesis. By using the lrg6 mutant that produces sexually competent gametes in the dark, light effects on ammonium transport and gamete differentiation have been separately analysed. We have found light regulation of four Amt1 genes: Amt1; 1, Amt1; 2, Amt1; 4 and Amt1; 5. Whereas light‐dependent expression of Amt1; 1, Amt1; 2 and Amt1; 4 was independent of gametogenesis, and that of Amt1; 5 was activated in the lrg6 mutant, suggesting a connection between this transporter and the subsequent events taking place during gametogenesis.  相似文献

The marine centric diatom Biddulphia levis produced uniflagellate fusiform male gametes completely within the parent cell frustule. These gametes lacked both a central pair of microtubules in the flagellar axoneme and chloroplasts but did contain a cone of microtubules which passed posteriorly from the base of the kinetosome along the nuclear envelope. The gametes were released through a specialized pore in the girdle band leaving behind a cytoplasmic mass which contained chloroplasts and other cytoplasmic components. Tubules which resembled the flimmer hairs on the gamete flagellum occurred in cisternae of the cytoplasmic reticulum in the residual cytoplasm and in the nuclear envelope of the gametes. Gametogenesis in B. levis is compared with similar processes in other centric diatoms.  相似文献

Summary The fate of chloroplast nuclei (cp-nuclei) and mitochondrial nuclei (mt-nuclei) was followed during gametogenesis in male and female coenocytic thalli in the anisogamous algaBryopsis maxima by epifluorescence microscopy, after staining with 4,6-diamidino-2-phenylindole (DAPI), by quantification of chloroplast DNA (cp-DNA) by fluorimetry using a video-intensified, photon-counting system (VIMPICS), and by CsCl density gradient centrifugation. The male and female coenocytic thalli, 48 h before the release of gametes, contain a large number of chloroplasts, each of which is larger in size than the cell nucleus and the mitochondria and contains about 150 cp-nuclei. The size of each chloroplast in the female and male gametangia decreases markedly during gametogenesis as a result of continuous divisions till about 10 h before the release of gametes and, eventually, the numbers of cp-nuclei per chloroplast in the male and female gametangia fall to about 20 and 5, respectively. Two hours later, as the preferential digestion of cp-DNA in the male gametangium occurs, the number of cp-nuclei in the chloroplast of each male gamete falls to zero while the number of cp-nuclei in female gamete does not change, even after release of female gametes. Several mt-nuclei are observed in all of the female gametes. By contrast, the mt-nuclei in the bulk of the male gametes disappear but those in a few gametes remain. The profiles after CsCl density gradient centrifugation of DNAs extracted from male and female plants and gametes support the cytological data. The results suggest that the preferential digestion of cp-DNA in male plants occurs about 8 h before the release of gametes and that there is differential digestion of cp-DNA and mitochondrial DNA (mt-DNA).  相似文献

At the ultrastructural level, cell division in Ochromonas danica exhibits several unusual features. During interphase, the basal bodies of the 2 flagella replicate and the chloroplast divides by constriction between its 2 lobes. The rhizoplast, which is a fibrous striated root attached to the basal body of the long flagellum, extends under the Golgi body to the surface of the nucleus in interphase cells. During proprophase, the Golgi body replicates, apparently by division, and a daughter rhizoplast, appears. During prophase, the 2 pairs of flagellar basal bodies, each with their accompanying rhizoplast and Golgi body, begin to separate. Three or 4 flagella are already present at this stage. At the same time, there is a proliferation of microtubules outside the nuclear envelope. Gaps then appear in the nuclear envelope, admitting the microtubules into the nucleus, where they form a spindle. A unique feature of mitosis in O. danica is that the 2 rhizoplasts form the poles of the spindle, spindle microtubules inserting directly onto the rhizoplasts. Some of the spindle microtubules extend from pole to pole; others appear to attach to the chromosomes. Kinetochores, however, are not present. The nuclear envelope breaks down, except, in the regions adjacent, to the chloroplasts; chloroplast ER remains intact throughout mitosis. At late anaphase the chromosomes come to lie against part of the chloroplast ER. This segment of the chloroplast ER appears to be incorporated as part of the reforming nuclear envelope, thus reestablishing the characteristic nuclear envelope—chloroplast ER association of the interphase cell.  相似文献

Sexual reproduction in the green alga, Chlamydomonas, is regulated by environmental conditions and by cell–cell interactions. After gametogenesis, flagellar adhesion between gametes triggers gamete activation, leading to cell fusion and zygote formation. Recent studies have identified new molecular events that underlie signal transduction during Chlamydomonas fertilization, including expression of a sex-determining protein, phosphorylation of a homeodomain protein, activity of a kinesin II and regulated translocation of an aurora/Ip11-like protein kinase from the cell body to the flagella.  相似文献

The relative amounts of Concanavalin A (Con A) bound by gamete and vegetative flagella of both mating types (mt + and mt -) of Chlamydomonas eugametos were determined using 125I-Con A. Con A agglutinated all cell types by cross-linking their flagella in a random manner. No correlation was found between the extent of Con A-binding and Con A-mediated isoagglutination. Con A inhibited the sexual interaction between gametes at various levels. In mt + gametes it blocked sexual agglutination, whereas in mt - gametes it prevented papillar fusion. By SDS-gel electrophoresis nine Con A-binding components were found to be present in flagella. The supply of iron has been reported to affect gametogenesis in the gametophytes of some species of kelps (order Laminariales). Spores of the kelps Alaria marginata Postels & Ruprecht, Dictyoneurum californicum Ruprecht, Egregia menziesii (Turner) Areschoug, Laminaria setchellii Silva, and Macrocystis pyrifera (L.) C. Agardh were cultured in enriched seawater with and without added chelated iron (Fe‐ethylenediaminetetraacetate) to determine the effects of iron on oogenesis. All species showed a decrease in oogenesis without added Fe‐ethylenediaminetetraacetic acid (EDTA). Gametophytes of E. menziesii showed predominant gametogenesis with or without supplied iron, resulting in all cells being converted to gametes so that vegetative growth did not continue. Vegetative gametophytes were obtained in the other species used. Gametophytes of M. pyrifera did not show any oogenesis without added Fe‐EDTA, while those of L. setchellii, A. marginata and D. californicum were intermediate in their response, showing some gametogenesis without added Fe‐EDTA. When Fe‐EDTA supply was delayed by 6, 13 and 20 days with spores of M. pyrifera, the gametophytes produced fewer eggs, with a greater decrease as the delay grew longer. A range of Fe‐EDTA concentrations was investigated using isolated female gametophytes of two strains of M. pyrifera and one of Macrocystis integrifolia Bory. None of these three strains produced gametes without the addition of Fe‐EDTA. Gametophytes of M. integrifolia required the least amount of added Fe‐EDTA to achieve gametogenesis while gametophytes of M. pyrifera required higher amounts, with the two strains showing somewhat different responses. Iron nutrition appears to be an essential factor for gametogenesis in several species of kelps.  相似文献

Behaviors of male and female gametes, planozygotes and their microtubular cytoskeletons of a marine green alga Bryopsis maxima Okamura were studied using field emission scanning electron microscopy, high‐speed video microscopy, and anti‐tubulin immunofluorescence microscopy. After fusion of the biflagellate male and female gametes, two sets of basal bodies lay side by side in the planozygote. Four long female microtubular roots extended from the basal bodies to the cell posterior. Four short male roots extended to nearly half the distance to the posterior end. Two flagella, one each from the male and female gametes, become a pair. Specifically, the no. 2 flagellum of the female gamete and one male flagellum point to the right side of the eyespot of the female gamete, which is located at the cell posterior and which is associated with 2s and 2d roots of the female gamete. This spatial relationship of the flagella, microtubular roots, and the eyespot in the planozygote is retained until settlement. During forward swimming, the planozygote swings the flagella backward and moves by flagellar beating. The male and female flagella in the pair usually beat synchronously. The cell withdraws the flagella and becomes round when the planozygote settles to the substratum 20 min after mixing. The axoneme and microtubular roots depolymerize, except for the proximal part and the basal bodies. Subsequently, distinct arrays of cortical microtubules develop in zygotes until 30 min after mixing. These results are discussed with respect to the functional significance of the spatial relationships of flagellar apparatus‐eyespot‐cell fusion sites in the mating gametes and planozygote of green algae.  相似文献
