Arginine decarboxylase (arginine carboxy-lyase EC 4.1.1.19) of Cucumis sativus cotyledons, has a pH optimum of 8.3 and a temperature optimum of 40°. Among the various plant hormones administered to excised cotyledons in culture, benzyladenine and its riboside were most effective in increasing the arginine decarboxylase activity and putrescine content. The enzyme activity and putrescine content were significantly increased on acid feeding of the cotyledons and decreased by KCl treatment. The KCl effect could be only partially reversed by benzyladenine. Abscisic acid inhibited cotyledon growth and also reduced arginine decarboxylase and putrescine levels. This effect was overcome by cytokinins. The half life of the enzyme using cycloheximide was 3.7 hr. Dibutyryl cyclic AMP and 5′-AMP also marginally stimulated the enzyme and putrescine levels. Mixing experiments indicate that there is neither a non-dialysable activator nor inhibitor of the enzyme.  相似文献

In potassium-deficient barley leaves, on the basis of fresh weight, putrescine was 13·3-fold greater and in magnesium deficiency 3-8-fold greater than controls. Putrescine was found to be reduced in phosphorus (× 0·6), sulphur (× 0·5) and nitrogen (× 0·2) deficiencies and on substituting nitrate for ammonium (× 0·2). Calcium deficiency and high salt (KCl) increased putrescine by 1·5-fold. High KCl reduced spermidine levels (× 0·2) without a corresponding reduction in spermine levels. The agmatine content was enhanced in magnesium- (× 2·5) and potassium- (× 5) deficient plants, while the arginine was increased × 1·4 and × 2·0 respectively on a fresh weight basis. Compounds tentatively identified as diaminopropane and 1-(3-aminopropyl)pyrroline were increased significantly with high KCl and low calcium. No homospermidine or diaminodipropylamine could be detected in any of the extracts, but traces of cadaverine may be present.  相似文献

Diamine oxidase activity against putrescine and agmatine is investigated in young decotylized Glycine max plants growing, in the light, under sterile conditions on a liquid medium in the presence of ammonium chloride or nitrates. Optimal conditions for the enzyme activity are determined. Both amines are oxidised more slowly in plants cultivated on ammonium chloride than in plants grown on nitrates.  相似文献

Seasonal cycles of concentrations and microbial uptake of dissolved free amino acids and the polyamine, putrescine, were followed during summer stratification of a coastal salt pond. Stratification began in May and was clearly seen in profiles of temperature, salinity, pH and alkalinity. Primary production exhibited a mid-August maximum and the O2-H2S interface shoaled at that time. POC and phytopigments roughly followed the pattern of primary production. Cycling of putresince, like the amino acids, was strongly influenced by primary production and microbial decomposition. Putrescine concentration profiles appeared to follow the pattern of primary production more closely, while amino acids appeared to follow the pattern of microbial production. The absence of production of putrescine during the decomposition of dissolved ornithine and the correlation of putrescine concentration with primary production suggest a direct source from algae in the water column.Microbial uptake of amino acids and putrescine together accounted for 60–90% of the bacterial C production measured in oxic waters and almost 300% of that measured in the anoxic bottom layer. Since other organic carbon and nitrogen compounds are also being taken up, these data suggest that tracer uptake methods as we used them may overestimate the true microbial uptake rates, or release of other organic compounds by microbes occurs at the same time. Further work on carbon and nitrogen budgets is needed to resolve the apparent imbalance between organic C and N incorporation and bacterial production.  相似文献

In rice seed, polyamine concentration on a fresh weight basis increased for 16 days after fertilization, followed by a gradual decline. Arginine decarboxylase activity also followed the same pattern. On germination, the polyamine concentration was greatest after 24 hr and the arginine decarboxylase showed a peak after 48 hr.  相似文献

A p-fluorophenylalanine- (PFP) resistant cell line of Nicotiana tabacum and wild type cells accumulating high and low levels of cinnamoyl putrescines, respectively, were used to study the formation of putrescine in the biosynthesis of cinnamoyl putrescines. Labelled arginine and ornithine were equally well incorporated into the main conjugates caffeoyl and feruloyl putrescine. Trapping experiments indicated that both amino acids were decarboxylated for putrescine biosynthesis. Nearly all alcohol-extractable radioactivity from the labelled amino acids was found as cinnamoyl putrescines in the PFP-resistant cell line, whereas wild type cells retained significant radioactivity in the amino acids. The enzyme activities of arginine and ornithine decarboxylases in the resistant cell line were increased 3- to 6-fold.  相似文献

In Phaseolus mungo seeds, polyamine content increased during early germination, being maximum after 24 hr; and the arginine decarboxylase showed a peak after 18 hr. During nodule initiation and growth two peaks of polyamine contents were noted-the first being 2 weeks after nodule initiation and a second one after 5 weeks. Arginine decarboxylase activity also followed the same pattern. In the roots the polyamine concentration as well as arginine decarboxylase increased up to week 2 after sowing followed by a gradual decrease. Estimation of RNA, DNA and protein contents showed a pattern similar to that of the polyamines.  相似文献

Putrescine, spermidine and spermine of high vigour, low vigour and non-viable (classes 1, 2 and 3 respectively) seeds of Oryza sativa increased with loss of viability. The largest concentration of spermine was found in non-viable embryos. Spermine was absent in the husks of all the three categories of seeds. Arginine decarboxylase was greatest in high vigoured seeds and its activity gradually declined with loss of viability. However, diamine oxidase and polyamine oxidase activities gradually increased with the loss of viability of the seeds while DNA, RNA and protein contents decreased. The total content of polyamines increased on kinetin treatment but declined on ABA treatment. DNA, RNA and protein followed the same trend as polyamines. The polyamine contents increased by ca 3- and 4-fold, respectively, in high vigoured and low vigoured seeds on 10?4 M kinetin treatment. The activity of ADC followed the same change as that of the polyamines in both cases, but the reverse was observed for the activities of diamine and polyamine oxidases.  相似文献

The maximum respiration rate of tomato fruit during the climacteric period was markedly increased when the plants were grown under potassium-deficient conditions. Whereas potassium deficiency had no effect on cytoplasmic glutamate-oxoloacetate transaminase, there was a significant increase in the activity of this enzyme from mitochondria once the fruit began to change colour. Malate dehydrogenase was reduced in activity by potassium deficiency. It is suggested that the augmented mitochondrial transiminase levels, coupled with reduced malate dehydrogenase activity in low potassium fruit, result in reduced levels of oxaloacetic acid which is a potent inhibitor of Krebs cycle oxidations, thus leading to higher respiration rates for the intact fruit.  相似文献

【背景】嗜盐四联球菌(Tetragenococcus halophilus)是一类存在于发酵食品中的耐盐乳酸菌,研究其精氨酸(arginine,Arg)代谢对解析食品发酵过程中氨基甲酸乙酯(ethyl carbamate,EC)前体积累机制和保障食品安全具有重要意义。【目的】研究酱醪来源嗜盐四联球菌精氨酸脱亚氨基(arginine deiminase,ADI)途径的基因构成,揭示这些基因对菌株精氨酸代谢和氨基甲酸乙酯前体瓜氨酸(citrulline,Cit)利用与积累的影响。【方法】采用PCR扩增与测序分析不同菌株的ADI途径基因组成,通过比较ADI途径关键基因转录水平和关键酶活性,探究环境因素对嗜盐四联球菌代谢氨基酸能力的影响及各拷贝基因参与氨基酸代谢的功能。【结果】酱醪来源嗜盐四联球菌基因组中ADI途径基因类型主要有两大类:以菌株R23为代表含有完整arc操纵子(operon)基因且具有最多基因拷贝数;以菌株C3为代表缺失arcAarcB但含有多拷贝arcBarcC。基因组中有arcA的菌株才具有利用精氨酸能力,并通过利用精氨酸生成瓜氨酸。体系中精氨酸含量和乙醇与脂肪酸的存在均可影响嗜盐四联球菌利用精氨酸积累中间产物瓜氨酸。当精氨酸含量大于5 g/L或体系中含有乙醇与脂肪酸时,嗜盐四联球菌会利用精氨酸积累中间产物瓜氨酸。脂肪酸和乙醇对ADI途径的3个关键酶均有显著抑制作用,可使精氨酸脱亚氨基酶(arginine deiminase,ADI)、鸟氨酸氨甲酰基转移酶(ornithine transcarbamylase,OTC)和氨甲酰磷酸激酶(carbamate kinase,CK)的活性分别降低41.0%、46.4%和60.0%。嗜盐四联球菌中arcB转录水平分别是其拷贝arcB1arcB2的10.5倍和29.8倍,arcC的转录水平分别是arcC1arcC2arcC3的17.6、20.3、23.9倍,说明arcBarcC在瓜氨酸代谢中起主要作用。【结论】精氨酸含量和乙醇加脂肪酸是影响嗜盐四联球菌代谢精氨酸能否积累瓜氨酸的关键环境因素。嗜盐四联球菌arc operon的多拷贝基因中,arcBarcC基因在瓜氨酸代谢中起主要作用。  相似文献

We previously reported two free D-amino acids, D-2-aminopimelic acid (D-APA) and trans-3,4-dehydro-D-2-aminopimelic acid (D-Δ-APA), from Asplenium unilaterale. In the present work we isolated 4-hydroxy-2-aminopimelic acid (OH-APA) from the same plant and determined it to be the α-L-form. We also investigated the configurations of these amino acids isolated from A. prolongatum and A. wilfordii which are morphologically distinct from A. unilaterale. In A. prolongatum, APA was the D- and OH-APA was the L-isomer. In contrast, APA from A. wilfordii was partially racemized and the degree of racemization was significantly different in plant material collected in July and November, L:D = 3:2 and 3:7, respectively. In A. wilfordii OH-APA was almost pure L- and Δ-APA was mostly the D-isomer.  相似文献

The biosynthesis of certain amines in Lathyrus sativus seedlings was studied in isolated shoots and cotyledons. In shoots, arginine was about 14 times more efficient than ornithine for the synthesis of agmatine, putrescine, spermidine and spermine. Isotope dilution experiments, and the changes in specific activities of the 4 amines with time when 14C-arginine served as the precursor, indicated that putrescine and the polyamines were formed mainly from arginine, via agmatine. Similar experiments showed that cadaverine was formed at least in part from homoarginine, though lysine was ca 4 times more effective as a precursor. The pattern of changes in specific activity of homoagmatine and cadaverine with time when 14C-homoarginine served as the precursor support the conclusion that homoarginine and arginine follow analogous metabolic routes in the biosynthesis of putrescine and cadaverine respectively.  相似文献

In order to clarify the biosynthetic origin of 2-geranyl-1,4-naphthoquinone and its biogenetically related anthraquinone, which are possible intermediates of anthrasesamones, [1-13C]glucose was administered to a hairy root culture of Sesamum indicum. The labeling patterns of these quinone derivatives indicated that the naphthoquinone ring and geranyl side-chain of geranylnaphthoquinone were respectively biosynthesized through the shikimate and methylerythritol phosphate pathways, and that these quinone derivatives have the same biosynthetic origin.  相似文献

3-(3-Carboxyphenyl)alanine, (3-carboxyphenyl)glycine, 3-(3-carboxy-4-hydroxyphenyl)alanine and (3-carboxy-4-hydroxyphenyl)glycine occur in all parts of Reseda luteola. The concentrations of the two diastereoisomers of 2(S)-4-hydroxy-4-methylglutamic acid undergo seasonal variation, the highest concentrations occurring in the first part of the summer. Highest concentrations are found in the inflorescences. The two diastereoisomers of 2(S)-4-hydroxy-2-aminopimelic acid occur in appreciable amounts in all parts of the plant. They are easily transformed into two structurally different lactones, one of which is very unstable. The structures of these amino acids have been confirmed by synthesis. Green parts of R. luteola also contain substantial quantities of γ-glutamylglutamic acid and glutathione.  相似文献

Arginine metabolism by wine lactic acid bacteria (LAB) may lead to wine quality degradation. While arginine is essential for growth of the wine relevant LAB Oenococcus oeni , it remains unclear whether it also stimulates its growth. This study evaluated the effect of arginine and citrulline, the partially metabolized intermediate of the arginine deiminase pathway, on the growth of two commercial O. oeni strains in comparison with a Lactobacillus buchneri strain in wine and at wine pH values. Neither arginine nor citrulline increased growth of both O. oeni strains in comparison with the L. buchneri strain. However, arginine and citrulline were partially degraded in all incubations. The extent of citrulline degradation correlated with lower pH values in oenococcal cultivations but with higher pH values in those of the L. buchneri strain. The degradation kinetics of O. oeni and L. buchneri for malic acid and arginine differed and the latter grew in sterile filtered post-malolactic fermentation wine. This study shows that arginine and citrulline did not stimulate growth of the two O. oeni strains studied, and that their physiological role differed among the wine LAB considered. While arginine may play a role in wine microbiological stability, other nutrients should be investigated for their suitability to create a selective ecological advantage for O. oeni strains in wine.  相似文献

In Phycomyces blakesleeanus the total level of free amino acids and their relative distributions change strikingly during the development of sporangiophores. Very high levels of serine, glutamate and histidine are found. The variety of amino acids becomes more restricted in sporangiophores grown on nitrogen-rich media; levels of arginine increase dramatically. The distribution differs in mycelia. Free amino acids in the seeds of 80 species of Acacia have been determined. In addition to confirming the known difference in seed amino acid ‘patterns’ shown by species of the series Gummiferae on the one hand and species of the series Phyllodineae and Botryocephalae on the other, we have found that species of the series Vulgares Benth. show two additional ‘patterns’. One of these is common to some of the Afro-Asian species of the series which have been analysed and the other is common to species from America and Africa. One of the ‘marker’ amino acids of the Afro-Asian group is the neurotoxic lathyrogen α-amino-β-oxalylaminopropionic acid. The possible taxonomic and phylogenetic significance of these results is discussed, with special reference to five species A. albida, A. confusa, A. heterophylla, A. coulteri and A. kauaiensis.  相似文献
