The potato tuberworm Phthorimaea operculella (Zeller) is an important agricultural pest that causes significant economic losses to potato growers worldwide. The addition of an effective method of biological control for the potato tuberworm is greatly needed, and is currently unavailable in Brazil. The granulosis virus (Baculoviridae) is a promising biological control agent to protect post-harvest potatoes and in storage from the potato tuberworm. However, the control measure must be economically feasible. Liquid suspensions of a granulosis virus applied alone or in mixture with two commercial neem oil-based products (DalNeem? and NeemAzal?), and a dry powder formulation of viral granules were evaluated for control of potato tuberworm larvae by treating potato tubers under laboratory conditions. High larval mortality (86.7%) was achieved when DalNeem and virus were applied together at 4?mg of azadirachtin/L and 104 occlusion bodies (OBs)/mL, respectively. This combination resulted in ??50% efficacy in relation to their counterparts alone. Conversely, NeemAzal did not enhance virus effectiveness against larvae of the potato tuberworm. The talc-based virus formulation was used for dusting seed tubers at different concentrations and resulted in 100% larval mortality at 5?×?108 OBs/g. Formulated and unformulated virus provided 50% mortality at 166 OBs/g and at 5.0?×?105 OBs/mL, respectively. As a result, talc-based virus formulation had a better control efficiency on potato tuberworm than the aqueous virus suspension. The granulosis virus combined with DalNeem at low rates or formulated with talc powder is a viable option to control the potato tuberworm under storage conditions.  相似文献

The possible involvement of potato (Solanum tuberosum L.) starch-branching enzyme I (PSBE-I) in the in vivo synthesis of phosphorylated amylopectin was investigated in in vitro experiments with isolated PSBE-I using 33P-labeled phosphorylated and 3H end-labeled nonphosphorylated α(1→4)glucans as the substrates. From these radiolabeled substrates PSBE-I was shown to catalyze the formation of dual-labeled (3H/33P) phosphorylated branched polysaccharides with an average degree of polymerization of 80 to 85. The relatively high molecular mass indicated that the product was the result of multiple chain-transfer reactions. The presence of α(1→6) branch points was documented by isoamylase treatment and anion-exchange chromatography. Although the initial steps of the in vivo mechanism responsible for phosphorylation of potato starch remains elusive, the present study demonstrates that the enzyme machinery available in potato has the ability to incorporate phosphorylated α(1→4)glucans into neutral polysaccharides in an interchain catalytic reaction. Potato mini tubers synthesized phosphorylated starch from exogenously supplied 33PO43− and [U-14C]Glc at rates 4 times higher than those previously obtained using tubers from fully grown potato plants. This system was more reproducible compared with soil-grown tubers and was therefore used for preparation of 33P-labeled phosphorylated α(1→4)glucan chains.  相似文献

Ca 2+ transport activity in mitochondria from some plant tissues   总被引:8,自引:0,他引:8  
Mitochondria isolated from some 14 different higher plants and fungi were examined for their capacity to carry out respiration-dependent accumulation of Ca2+. Additions of Ca2+ give little or no stimulation of state 4 respiration of plant mitochondria, although the added Ca2+ was largely accumulated. Accumulation of Ca2+ required phosphate and, in most cases, was stimulated by Mg2+ and ADP or ATP. Ca2+ uptake was abolished by respiratory inhibitors and uncoupling agents. The ratio of Ca2+ ions taken up per pair of electrons per energy-conserving site was normal at about 2.0 for mitochondria from sweet potato and white potato; mitochondria from other plants showed somewhat lower ratios. Accumulated Ca2+ was only very slowly released from previously loaded plant mitochondria. Respiration-inhibited sweet potato mitochondria show both high-affinity and low-affinity Ca2+ binding sites sensitive to uncouplers, La3+, and ruthenium red and thus resemble animal mitochondria. Most other plant mitochondria lack high affinity sites. In general, mitochondria from sweet potato and white potato tubers resemble those from animal tissues, but mitochondria from carrots, beets, turnips, onions, cabbage, artichokes, cauliflower, avocados, mung bean and corn seedlings, and mushrooms show rather low affinity and activity in accumulation of Ca2+, probably due to lack of a specific Ca2+ carrier.  相似文献

A novel potato micro-tuber-inducing compound was isolated from the culture broth of Lasiodiplodia theobromae Shimokita 2. The structure of the isolated compound was determined as (3R,6S)-6-hydroxylasiodiplodin by means of spectroscopic analyses, the modified Mosher method, and chemical conversion. The compound showed potato micro-tuber-inducing activity at a concentration of 10?4 M, using the culture of single-node segments of potato stems in vitro.  相似文献

A memristor is a nonlinear element because its current-voltage characteristic is similar to that of a Lissajous pattern for nonlinear systems. This element was postulated recently and researchers are looking for it in different biosystems. We investigated electrical circuitry of red Irish potato tubers (Solanum tuberosum L.). The goal was to discover if potato tubers might have a new electrical component - a resistor with memory. The analysis was based on a cyclic current-voltage characteristic where the resistor with memory should manifest itself. We found that the electrostimulation by bipolar sinusoidal or triangle periodic waves induces electrical responses in the potato tubers with fingerprints of memristors. Tetraethylammonium chloride, an inhibitor of voltage gated K+ channels, transforms a memristor to a resistor in potato tubers. Our results demonstrate that a voltage gated K+ channel in the excitable tissue of potato tubers has properties of a memristor. Uncoupler carbonylcyanide-4-trifluoromethoxy-phenyl hydrazone decreases the amplitude of electrical responses at low and high frequencies of bipolar periodic sinusoidal or triangle electrostimulating waves. The discovery of memristors in plants creates a new direction in the understanding of electrical phenomena in plants.  相似文献

Plant species and genotypes within the same species may differ in phosphorus efficiency. The objective of this research was to study phosphorus efficiency of cabbage (Brassica oleraceae L.), carrot (Daucus carota L.), and potato (Solanum tuberosum L.) and to quantify the contribution of morphological root characteristics to P uptake of the plant species. An experiment was conducted in a glasshouse with six P levels: 0, 12, 27, 73, 124 and 234 mg P kg–1 soil, and with six replications. Cabbage attained 80% of its maximum yield already at the level of no P supply, whereas carrot and potato reached only 4 and 16% of their highest yields respectively at this level of P supply. This indicated that cabbage was P-efficient compared to carrot and potato. Root/shoot ratio (cm root g–1 shoot d. m.) increased in the order of cabbage < carrot < potato, and was enhanced at lower P levels. Root hair length was not affected by P level, and averaged 0.22, 0.03 and 0.18 mm for cabbage, carrot, and potato, respectively. Predicting P uptake by a mechanistic simulation model revealed that root hairs contributed about 50% to the total P uptake of cabbage and potato, but only 0.3% to that of carrot. The relationship between the observed P uptake and the predicted P uptake of the plants revealed that model parameters explained nearly 4/5th of the total P uptake of carrot and potato, but only 2/5th of that of cabbage. This showed that the P uptake of cabbage was strongly under-predicted, whereas that of carrot and potato was predicted well. Therefore, it was hypothesised that cabbage may have the ability to mobilise and take up soil P additionally by other root mechanisms such as exudation of organic acids.  相似文献

Eight isolates from different potato growing regions in Vietnam were characterized. All were highly pathogenic in some potato cultivars, but did not overcome the extreme resistance of Solanum stoloniferum and Solanum demissum. RT‐PCR analysis revealed that all of these isolates are recombinants. Sequence data for 4 isolates were obtained, and their reaction in potato cultivars harbouring specific N genes was determined. Different phylogenetic analyses of viral sequences confirmed previous results that the recombinant isolates evolved from different parental sequences. One of the Vietnamese isolates investigated had a specific structure. The need for a clear classification of PVYNWi isolates is discussed.  相似文献

Mouse monoclonal antibodies (MAbs) specific for potato virus M (PVM) were prepared and the properties of three of them were studied. MAb M4C1 is IgG2b, it binds with high affinity to PVM coat protein, to purified virus preparations and recognises PVM in infected potato leaves and tubers. MAb M6D5 is IgG2a and also reacts with PVM coat protein, purified PVM and with PVM in potato leaf and tuber extracts. In double-antibody sandwich ELISA (DAS ELISA) MAbs M4C1 and M6D5 reacted with all 17 PVM isolates tested. MAb M7 is IgG2b and recognises PVM only in indirect dot ELISA on nitrocellulose filters and viral coat protein on Western blots. MAbs against PVM were used as capture antibodies and europium-labelled MAbs as conjugates in time-resolved fluoroimmunoassay (EuTRFIA). The standard EuTRFIA curve of PVM detection is approximately linear over a range of PVM concentrations from 0.5 ng/ml to 1000 ng/ml. The lowest PVM concentration detectable in EuTRFIA was 0.5 ng/ml and correspondingly 6 ng/ml in DAS ELISA. The use of the europium chelate label allows PVM detection in potato leaf and tuber sap at dilutions greater than 10--4 with very low background fluorescence. EuTRFIA with MAbs, with either one or two incubations is about 10–20 times more sensitive for PVM detection than is DAS ELISA. PVM and PVX, mixed with healthy potato tuber sap, were simultaneously tested in a single sample at concentrations lower than 10 ng/ml by double-label TRFIA using europium-labelled MAbs to PVM and samarium-labelled MAbs to PVX.  相似文献

The reaction of several cultivated potato varieties (Solarium tuberosum L.) to three strains of tobacco etch potyvirus (TEV-F, TEV-Mex21 and TEV-ATCC) and the reaction of several pepper lines (Capsicum annuum L. and C. chinense L.) to two strains of potato Y potyvirus (PVYO and PVYN) and one strain of potato A potyvirus (PVA-M) was tested. The potato varieties included in this study carried resistance genes against PVY, PVA and potato V potyvirus, but all were susceptible to TEV and developed mottle and mosaic symptoms. TEV was readily transmitted by mechanical inoculation from tobacco and potato to potato, whereas transmission from pepper to potato occurred infrequently. TEV was transmitted through potato tubers, and from pepper to potato plants by aphids. Lack of detectable systemic infection following graft-inoculation indicated extreme resistance to PVYO and PVA in several pepper lines. No pepper line was systemically infected with PVYN following mechanical inoculation (graft-inoculation was not carried out with PVYN). The development of necrotic lesions following mechanical and graft-inoculation indicated hypersensitive response to PVYO in several pepper lines which resembled the resistance responses to these potyvirus strains in potato. Results of this study together with previous work indicate that C. annuum cv. Avelar is resistant to four potyviruses [PVY, PVA, pepper mottle potyvirus (PepMoV) and some isolates of TEV]; C. annuum cv. Criollo de Morelos and C. chinense PI 152225 and PI 159236 are resistant to three potyviruses (PVY, PepMoV and PVA; and PVY, PepMoV and TEV, respectively); C. annuum 9093–1 and 92016–1 are resistant to PVY and PepMoV; and C. annuum cv. Jupiter and C. annuum cv. RNaky are resistant to PVYN and PVA.  相似文献

TCP转录因子是植物特有的一类转录因子,参与植物生物学过程的多个方面。为研究马铃薯TCP转录因子在响应低氮肥胁迫中的作用,该研究以氮肥供应不足(0.05 mmol·L-1)和氮肥供应充足(7.5 mmol·L-1)条件下马铃薯的根和叶片构建4个转录组文库进行测序,并对差异表达的TCP转录因子进行分析。结果表明:(1)在4个转录组文库中共鉴定TCP转录因子24个,它们主要分布在2号、3号、6号染色体上。(2)经结构域分析显示,24个TCP 转录因子均具有典型的basic-Helix-Loop-Helix结构域。(3)经系统进化分析显示,马铃薯与拟南芥TCP蛋白可聚集在一起,分属于10个亚类。(4)转录组测序结果显示,在低氮肥胁迫下,大多数TCP转录因子被抑制表达,有3个TCP转录因子在根中显著性差异表达,5个TCP转录因子在叶中特异性表达。(5)根据GO功能注释分析和马铃薯TCP转录因子与拟南芥TCP转录因子的亲缘关系分析推测,这些TCP转录因子参与了马铃薯对低氮肥胁迫的响应。该研究结果为进一步研究马铃薯与其他粮食作物TCP转录因子响应低氮肥胁迫的分子功能奠定了基础。  相似文献

Field studies on the control of potato common scab were done with non-antibiotic Actinomycete biofertilizer, which was manufactured from swine feces with coprophilous actinomycetes and acidified to pH 5.0 with sulfuric acid. The field soil at pH 5.0 was mixed with 0.56 kg of the acidified biofertilizer per subplot (1.0 x 1.2 m) to a depth of the 15 cm. The scab severity, in terms of the percent coverage of the total surface area of potato tubers, was 10.7%, in the control, while the lesions were slight in the biofertilizer subplot, scab severity being only 2.30/0, and the potato production was increased 3 times with the biofertilizer. The viable counts of Streptomyces scabies in the biofertilizer subplot decreased to 1 × 103 per gram soil from 5 × 105 by 12 weeks, while the inhabitant fluorescent pseudomonads increased to 2.4 × 107 per gram soil from 3 × 104, and the increased fluorescent pseudomonads were antagonistic to the pathogenic strains.  相似文献

RECENT evidence indicates that the agent of the potato spindle tuber disease, previously thought to be a virus, is a replicating RNA with a molecular weight of about 50,000 (refs. 1 and 2). Because there seems to be no helper virus in the system1, the RNA must rely for its replication principally on biosynthetic systems already functioning in the host. This, as well as the absence of virions, distinguishes the potato spindle tuber agent from all known viral agents. I suggested that such agents, which may include the agents of citrus exocortis3 and of chrysanthemum stunt4 diseases, should be called “viroids”1. I now suggest that the agent of scrapie may also be a viroid.  相似文献

The Andean potato weevil Premnotrypes suturicallus Kuschel (Coleoptera: Curculionidae) is one of the most damaging potato (Solanum spp.) pests in the Andes. The objective of this study was to get a better understanding of weevil infestation sources and its distribution in potato fields as well as on the effect of potato cropping intensity and farmers’ harvest practices on weevil infestation to provide better clues for its management. For this purpose, a methodology was developed to assess weevil field densities in potato. A Taylor’s power law indicated that 177 and 69 samples are necessary to assess low (0.5 weevils/m2) and high (5 weevils/m2) weevil densities for a moderate reliability or precision level (D = 0.5). In potato fields, overwintering weevil densities were highest at field borders with 3.5 weevils/m2 at a distance of <2.5 m compared to 0.7 weevils/m2 at distances >10 m from the field border. No effects of time of harvest could be detected on soil overwintering weevil densities. The quantification of the larval density per potato plant after harvest showed that fields after 2‐year potato rotations had eight times more overwintering weevils compared to 1‐year rotations. Weevil infestation sources were mainly potato fields with the highest infestation (89%) followed by olluco (Ullucus tuberosus) and oat (Avena sativa) fields having volunteer potato plants (35%). The results confirm and support previous assumptions about the importance of the cropping systems for Andean potato weevil infestation and management. The confirmation that weevils do not occur or only in neglected numbers on fallow fields supports the use of plastic barriers to effectively exclude migrating flightless adult weevils to potato fields cultivated after fallow. The distribution of overwintering weevils indicates that farmers could concentrate efforts to control adult weevils mainly to the first meters of potato fields.  相似文献

Cultures of potato (Solanum tuberosum) cv. Atlantic, chokecherry (Prunus virginiana L.) cv. Garrington and saskatoon berry (Amelancher alnifolia Nutt.) cv. Northline grown in vitro for 3 weeks at 24/22 °C, 16-h photoperiod, 150 μmol m−2 s−1 photosynthetic photon flux density (PPFD) mixed fluorescent/incandescent light were stored for 6, 9 and 12 weeks at 4 °C under 0 (darkness) and 3 μmol m−2 s−1 PPFD (690 nm red light continuous illumination). Growth regulators free MSMO medium either with or without 30 g l−1 sucrose was used to store the cultures. All cultures retained capacity to re-grow after storage. Tested factors, sucrose, light and the length of the storage period had an impact on shoot quality and re-growth capacity of the cultures. For either light treatment sucrose was essential for the low temperature maintenance of vigorous stock plants of potato, if stored for over 6 weeks. Chokecherry and saskatoon cultures stored well without sucrose; although chokecherry benefited from sucrose in the storage medium when the stock cultures were kept at the low temperature for 12 weeks. Low light significantly improved quality of the stored potato cultures, but had very little effect on both chokecherry and saskatoon berry cultures. The woody plant cultures grew during storage, and the longer the stock plants were stored, the more vigorous cultures they generated. The results indicate that growers can successfully use their existing facilities, small refrigerators and coolers with low light intensity, set at 4 °C, for short term storage of potato, chokecherry and saskatoon berry cultures. The potato cultures, which are known to be sensitive to prolonged low temperature storage, should be frequently monitored and subcultured as required. On the other hand, the woody plant stock cultures do not require any special attention when kept at 4 °C and re-grow the most vigorous shoots if stored for at least 12 weeks. S-ethyldipropylthiocarbamate (EPTC), S-(2,3-dichloroallyl)diisopropylthiocarbamate (diallate) and S-(2,3,3-trichloroallyl)diisopropylthiocarbamate (triallate) inhibited the formation of very long chain fatty acids by aged potato discs. Incorporation of acetate-[14C] into total fatty acids was inhibited 24% by EPTC, 50% by triallate and 55% by diallate at 10?4 M. The relative sensitivity of very long chain fatty acid synthesis to thiocarbamates in potato tuber provides further evidence that these herbicides reduce cuticular wax by inhibiting fatty acid elongation.  相似文献

Summary In search of establishing a system for genetic transformation of Brazilian potato cultivars, Agrobacterium tumefaciens carrying the plasmid pGV1040, was used to transform leaf discs of three cultivars of local importance, i.e., Aracy, Baronesa and Mantiqueira. This plasmid contains marker genes for resistance to kanamycin and phosphinothricin plus the gene for the enzyme -glucuronidase. A two step regeneration/selection procedure produced shoots of potato cultivar Mantiqueira with in vitro resistance to kanamycin and to phosphinothricin. After transfer to the greenhouse, the potentially transgenic plants, sprayed with the herbicide Finale® (20% a.i.; Hoechst®) remained green as compared to control clones that died immediately afterwards. Southern blot analysis and histochemical and fluorimetric assay for -glucuronidase indicated that the gene coding for the enzyme was integrated in the potato genome and could be expressed in potato tissues. Agrotis segetum Schiff granulosis virus (AsGV) propagated in Denmark was supplied against naturally occurring cutworm populations (A. ipsilon and to a less extentA. segetum) in experimental field plots of tobacco, okra, potato and sugar beet in northern Pakistan. AsGV doses varied between 4 × 107 and 4 × 1011 capsules per m2 plot, and no. of applications between 1 and 3. One treatment with AsGV did not reduce cutworm damage significantly to tobacco seedlings and potato plants. Two treatments with AsGV reduced cutworm damage significantly. In tobacco, reduction was 64–82%, in okra and potato 85% and 77% respectively. Damage in sugar beet was reduced 78%. Three treatments with AsGV dis not reduce damage significantly better than two treatments. AsGV and the chemical insecticides Tamaran and Dieldrin, andBacillus thuringiensis (Thuricide) were about equally effective, reducing damage by 85%, 79%, 87% and 69%, respectively. No difference was found between the efficiency of highly purified AsGV to which activated charcoal was added and partially purified AsGV without charcoal.   相似文献

IAA oxidase preparations from fresh sweet potato tuber discs oxidized IAA only in the presence of added phenolic cofactors, and the pH optimum for enzyme activity depended on the cofactor used. Ageing of tuber discs, either by aeration in distilled water or by incubation on moist filter paper, resulted in increased peroxidase and phenol-stimulated IAA oxidase activities, as well as the development of IAA oxidase activity in the absence of added cofactors. High phenolase activity of fresh tuber discs decreased considerably with ageing. Phenol-stimulated IAA oxidase activity reached maximal levels before IAA oxidase activity in the absence of added cofactors. Enzyme preparations from aged tuber discs had double pH optima, similar to those previously described for sweet potato root IAA oxidase preparations. IAA in the concentration range 10?4 to 10?2 M inhibited the increase in peroxidase and IAA oxidase activities with ageing. DCP-stimulated IAA oxidase activities in preparations from both fresh and aged sweet potato tuber discs were inhibited by manganous ion.  相似文献
