To examine the influence of vesicular-arbuscular (VA) mycorrhizal fungi on phosphorus (P) depletion in the rhizosphere, mycorrhizal and non-mycorrhizal white clover (Trifolium repens L.) were grown for seven weeks in a sterilized calcareous soil in pots with three compartments, a central one for root growth and two outer ones for hyphae growth. Compartmentation was accomplished by a 30-μm nylon net. The root compartment received a uniform level of P (50 mg kg−1 soil) in combination with low or high levels of P (50 or 150 mg kg−1 soil) in the hyphal compartments. Plants were inoculated withGlomus mosseae (Nicol. & Gerd.) Gerd. & Trappe or remained uninfected. Mycorrhizal inoculation doubled P concentration in shoot and root, and increased dry weight, especially of the shoot, irrespective of P levels. Mycorrhizal contribution accounted for 76% of total P uptake at the low P level and 79% at the high P level, and almost all of this P was delivered by the hyphae from the outer compartment. In the non-mycorrhizal plants, the depletion of NaHCO3-extractable P (Olsen-P) extended about 1 cm into the outer compartment, but in the mycorrhizal plants a uniform P depletion zone extended up to 11.7 cm (the length of the hyphal compartment) from the root surface. In the outer compartment, the mycorrhizal hyphae length density was high (2.5–7 m cm−3 soil) at the various distances (0–11.7 cm) from the root surface. Uptake rate of P by mycorrhizal hyphae was in the range of 3.3–4.3×10−15 mol s−1 cm−1.  相似文献

 The effect of arbuscular mycorrhiza (AM) on white clover and ryegrass grown together in a soil spiked with polycyclic aromatic hydrocarbons (PAH) was assessed in a pot experiment. The soil was spiked with 500 mg kg–1 anthracene, 500 mg kg–1 chrysene and 50 mg kg–1 dibenz(a,h)anthracene, representing common PAH compounds with three, four and five aromatic rings, respectively. Three treatments and two harvest times (8 and 16 weeks) were imposed on plants grown in spiked soil: no mycorrhizal inoculation, mycorrhizal inoculation (Glomus mosseae P2, BEG 69) and mycorrhizal inoculation and surfactant addition (Triton X-100). Pots without PAH were also included as a control of plant growth and mycorrhizal colonization as affected by PAH additions. The competitive ability of clover vis-à-vis ryegrass regarding shoot and root growth was enhanced by AM, but reduced by PAH and the added surfactant. This was reflected by mycorrhizal root colonization which was moderate for clover (20–40% of total root length) and very low for ryegrass (0.5–5% of total root length). Colonization of either plant was similar in spiked soil with and without the added surfactant, but the PAH reduced colonization of clover to half that in non-spiked soil. P uptake was maintained in mycorrhizal clover when PAH were added, but was reduced in non-mycorrhizal clover and in mycorrhizal clover that received surfactant. Similar effects were not observed on ryegrass. These results are discussed in the context of the natural attenuation of organic pollutants in soils. Accepted: 12 June 2000  相似文献

 High salt levels in soil and water can limit agricultural production and land development in arid and semiarid regions. Arbuscular mycorrhizal fungi (AMF) have been shown to decrease plant yield losses in saline soils. The objective of this study was to examine the growth and mineral acquisition responses of greenhouse-grown tomato to colonization by the AMF Glomus mosseae [(Nicol. And Gerd.) Gerd. and Trappe] under varied levels of salt. NaCl was added to soil in the irrigation water to give an ECe of 1.4 (control), 4.7 (medium) and 7.4 dS m–1 (high salt stress). Plants were grown in a sterilized, low P (silty clay) soil-sand mix. Mycorrhizal colonization was higher in the control than in saline soil conditions. Shoot and root dry matter yields and leaf area were higher in mycorrhizal than in nonmycorrhizal plants. Total accumulation of P, Zn, Cu, and Fe was higher in mycorrhizal than in nonmycorrhizal plants under both control and medium salt stress conditions. Shoot Na concentrations were lower in mycorrhizal than in nonmycorrhizal plants grown under saline soil conditions. The improved growth and nutrient acquisition in tomato demonstrate the potential of AMF colonization for protecting plants against salt stress in arid and semiarid areas. New information on N uptake and transport of inorganic and organic N in arbuscular mycorrhizal fungi is reviewed here. Hyphae of the arbuscular mycorrhizal fungus Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe (BEG 107) were shown to transport N supplied as 15N-Gly to wheat plants after a 48 h labelling period in semi-hydroponic (Perlite), non-sterile, compartmentalised pot cultures. Of the 15N supplied to hyphae in pot cultures over 48 h, 0.2 and 6% was transported to plants supplied with insufficient N or sufficient N, respectively. The increased 15N uptake at the higher N supply was related to the higher hyphal length density at the higher N supply. These findings were supported by results from in vitro and monoxenic studies. Excised hyphae from four Glomus isolates (BEG 84, 107, 108 and 110) acquired N from both inorganic (15NH4 15NO3, 15NO3 or 15NH4 +) and organic (15N-Gly and 15N-Glu, except in BEG 84 where amino acid uptake was not tested) sources in vitro during short-term experiments. Confirming these studies under sterile conditions where no bacterial mineralisation of organic N occurred, monoxenic cultures of Glomus intraradices Schenk and Smith were shown to transport N from organic sources (15N-Gly and 15N-Glu) to Ri T-DNA transformed, AM-colonised carrot roots in a long-term experiment. The higher N uptake (also from organic N) by isolates from nutrient poor sites (BEG 108 and 110) compared to that from a conventional agricultural field implied that ecotypic differences occur. Although the arbuscular mycorrhizal isolates used contributed to the acquisition of N from both inorganic and organic sources by the host plants/roots used, this was not enough to increase the N nutritional status of the mycorrhizal compared to non-mycorrhizal hosts. Two experiments were carried out in a growth chamber and a naturally lit glasshouse to investigate the influence of seed phosphorus (P) reserves on growth and P uptake by wheat plants (Triticum aestivum cv Krichauff), and their association with arbuscular mycorrhizal (AM) fungi. Increased seed P reserves improved plant growth at a range of P supply up to over 100 mg P kg–1 soil. Plants grown from seeds with high P reserves tended to accumulate more P from soil, which was mainly attributed to better root system development. Mycorrhizal colonisation did not significantly affect P uptake of plants grown with low irradiance (in growth chamber). However, in the naturally lit glasshouse, mycorrhizal plants had significantly higher P concentrations than non-mycorrhizal plants. Furthermore, mycorrhizal plants grown from seeds low in P accumulated similar amounts of P compared with those grown from seeds with high P, indicating that mycorrhizal colonisation may overcome the disadvantage of having low seed P reserves in the field.  相似文献

Few field studies have investigated the contribution of arbuscular mycorrhizal fungi (AMF) to agricultural systems. In this study, the role of AMF in nutrition and yield of dryland autumn-sown wheat and field pea was examined through a 2-year crop sequence experiment on a red loam (Kandosol) in SE Australia. The soil was P-deficient and had low levels of root pathogens. In Year 1, levels of AMF were increased by growing subterranean clover or LinolaTM and decreased by growing canola or through maintenance of bare fallow with herbicides or tillage. In Year 2, hosts of AMF (wheat and field pea) and non-mycorrhizal canola were grown with 0 P or 20 kg ha–1 of P as superphosphate. Yields of all Year 2 crops were increased by P-fertiliser. Year 1 treatment led to 2–3 fold variation in colonisation by AMF at each P-level for Year 2 wheat and field pea. High colonisation did not correspond with greater crop growth, yield, or uptake of P, K, Ca, Cu or S in wheat or field pea. However, total crop Zn-uptake and grain Zn concentration were positively correlated with colonisation by AMF, due to enhanced Zn-uptake after anthesis. For wheat, high colonisation also corresponded with reduced Mn-uptake and lower grain Mn concentrations. In a glasshouse experiment using a second P-deficient Kandosol, inoculation of wheat with Glomus intraradices and Scutellospora calospora enhanced uptake of Zn and P when no P-fertiliser was applied. We conclude that high colonisation by AMF is unimportant for productivity of the major field crops grown on the Kandosol soils that occupy large areas of cropland in temperate SE Australia, even under P-limiting conditions. Investigation of the factors that control functioning of arbuscular mycorrhizae under field conditions, especially temperature, is required.  相似文献

Alginate production by Azotobacter vinelandii was studied in batch and continuous cultures under microaerobic conditions. In batch culture at a pO2 of 2–3% (air saturation) alginate production was enhanced by decreasing the PO3− 4 level in the medium. Alginate yield from biomass (Y P/X) reached the highest value of 0.66 g/g at the lowest phosphate level (100 mg/l), compared to 0.40 g/g and 0.25 g/g at higher phosphate levels (200 mg/l and 400 mg/l, respectively). In contrast, biomass formation behaved differently and the growth yield (Y X/S) decreased with decreasing PO4 3− concentrations. Moreover, the respiratory quotient (RQ) of the culture was dependent on the initial phosphate concentration, especially in the phosphate-limited phase of growth. As the initial phosphate level decreased from 400 mg/l to 100 mg/l, the average RQ value of the culture declined from 1.46 to 0.89. The low RQ value is very close to the theoretical optimum RQ, calculated to be 0.8 on the basis of the stoichiometry of the metabolic pathways for alginate formation from sucrose. This optimum RQ was also confirmed in continuous culture at different dilution rates. Independent of the dilution rate, a pO2 value of 2–5% (air saturation) was found to be optimal for alginate production, the corresponding RQ values being 0.80–0.84. In addition, the molecular mass and composition of alginate were also found to be affected by both phosphate and oxygen concentrations. In conclusion, the RQ appears to be a useful parameter for optimum control of alginate production with this microorganism. In order to characterise the effect of ectomycorrhiza on Na+-responses of the salt-sensitive poplar hybrid Populus × canescens, growth and stress responses of Paxillus involutus (strain MAJ) were tested in liquid cultures in the presence of 20 to 500 mM NaCl, and the effects of mycorrhization on mineral nutrient accumulation and oxidative stress were characterised in mycorrhizal and non-mycorrhizal poplar seedlings exposed to 150 mM NaCl. Paxillus involutus was salt tolerant, showing biomass increases in media containing up to 500 mM NaCl after 4 weeks growth. Mycorrhizal mantle formation on poplar roots was not affected by 150 mM NaCl. Whole plant performance was positively affected by the fungus because total biomass was greater and leaves accumulated less Na+ than non-mycorrhizal plants. Energy dispersive X-ray microanalysis using transmission electron microscopy analysis of the influence of mycorrhization on the subcellular localisation of Na+ and Cl in roots showed that the hyphal mantle did not diminish salt accumulation in root cell walls, indicating that mycorrhization did not provide a physical barrier against excess salinity. In the absence of salt stress, mycorrhizal poplar roots contained higher Na+ and Cl concentrations than non-mycorrhizal poplar roots. Paxillus involutus hyphae produced H2O2 in the mantle but not in the Hartig net or in pure culture. Salt exposure resulted in H2O2 formation in cortical cells of both non-mycorrhizal and mycorrhizal poplar and stimulated peroxidase but not superoxide dismutase activities. This shows that mature ectomycorrhiza was unable to suppress salt-induced oxidative stress. Element analyses suggest that improved performance of mycorrhizal poplar under salt stress may result from diminished xylem loading of Na+ and increased supply with K+.  相似文献

 Plant phosphorus uptake via external hyphae of arbuscular mycorrhizal fungi has been measured using compartmented systems where a hyphal compartment is separated from a rooting compartment by a fine mesh. By labelling the soil within the hyphal compartment with a radioactive phosphorus (P) isotope, hyphal uptake of P into the plant can be traced. The objective of this growth chamber study was to test two hyphal compartments of different design with respect to their suitabilities for measurement of hyphal P uptake. One hyphal compartment was simply a nylon mesh bag filled with 32P-labelled soil. The labelled soil in the other hyphal compartment was completely surrounded by an 8–10 mm layer of unlabelled soil that served as a buffer zone. Mycorrhizal and non-mycorrhizal subterranean clover plants were grown in pots with a centrally positioned hyphal compartment. Uptake of radioactive P by non-mycorrhizal control plants was 25% of that by mycorrhizal plants with the mesh bag but only 3% when including the buffer zone. Based on this good control of non-mycorrhizal P uptake from within the hyphal compartment and its greater ease of handling once produced, we judged the hyphal compartment including a buffer zone to be superior to the mesh bag. Pedunculate oak (Quercus robur L.) was germinated and grown at ambient CO2 concentration and 650 μmol mol?1 CO2 in the presence and absence of the ectomycorrhizal fungus Laccaria laccata for a total of 22 weeks under nonlimiting nutrient conditions. Sulphate uptake, xylem loading and exudation were analysed in excised roots. Despite a relatively high affinity for sulphate (KM= 1.6 mmol m?3), the rates of sulphate uptake by excised lateral roots of mycorrhizal oak trees were low as compared to herbaceous plants. Rates of sulphate uptake were similar in mycorrhizal and non-mycorrhizal roots and were not affected by growth of the trees at elevated CO2. However, the total uptake of sulphate per plant was enhanced by elevated CO2 and further enhanced by elevated CO2 and mycorrhization. Sulphate uptake seemed to be closely correlated with biomass accumulation under the conditions applied. The percentage of the sulphate taken up by mycorrhizal oak roots that was loaded into the xylem was an order of magnitude lower than previously observed for herbaceous plants. The rate of xylem loading was enhanced by mycorrhization and, in roots of mycorrhizal trees only, by growth at elevated CO2. On a whole-plant basis this increase in xylem loading could only partially be explained by the increased growth of the trees. Elevated CO2 and mycorrhization appeared to increase greatly the sulphate supply of the shoot at the level of xylem loading. For all treatments, calculated rates of sulphate exudation were significantly lower than the corresponding rates of xylem loading of sulphate. Radiolabelled sulphate loaded into the xylem therefore seems to be readily diluted by unlabelled sulphate during xylem transport. Allocation of reduced sulphur from oak leaves was studied by flap-feeding radiolabelled GSH to mature oak leaves. The rate of export of radioactivity from the fed leaves was 4–5 times higher in mycorrhizal oak trees grown at elevated CO2 than in those grown at ambient CO2. Export of radiolabel proceeded almost exclusively in a basipetal direction to the roots. From these experiments it can be concluded that, in mycorrhizal oak trees grown at elevated CO2, the transport of sulphate to the shoot is increased at the level of xylem loading to enable increased sulphate reduction in the leaves. Increased sulphate reduction seems to be required for the enhanced allocation of reduced sulphur to the roots which is observed in trees grown at elevated CO2. These changes in sulphate and reduced sulphur allocation may be a prerequisite for the positive effect of elevated CO2 on growth of oak trees previously observed.  相似文献

The contribution of influx and efflux of NO3 - on NO3 - net uptake has been studied in excised mycorrhizae of 18–20 week old beech (Fagus sylvatica L.) trees. Net uptake rates of NO3 - followed uniphasic Michaelis-Menten kinetics in the concentration range between 10 μM and 1.0 mM external NO3 -, with an apparent Km of 88±7 μM, and a Vmax of 110±7 nmol g-1 root f.wt. h-1. The relative xylem loading of N, i.e. the portion of NO3 - taken up that was loaded into the xylem vessels as NO3 - plus reduced N, was constant over the concentration range tested (4.6–7.7%). NO3 - influx proceeded linearly with increasing external NO3 - supply. When the assumed regulators of net NO3 - uptake, i.e. NH4 + or L-glutamate, were applied together with NO3 -, net uptake rates of NO3 - decreased. This inhibitory effect was caused by a reduction of NO3 - influx rather than an enhanced efflux. The comparison of the present data with a recent study with non-mycorrhizal beech roots (Kreuzwieser et al., 1997; J. Exp. Bot. 48, 1431–1438) revealed that mycorrhization leads to reduced rates of NO3 - net uptake. This effect is caused by reduced influx plus enhanced efflux of NO3 - as compared with non-mycorrhizal beech roots. Sustainability of soil-plant systems requires, among other things, good development and function of mycorrhizal symbioses. The effects of P and micronutrient levels on development of an arbuscular mycorrhizal fungus (AMF) and uptake of Zn, Cu, Mn and Fe by maize (Zea mays L.) were studied. A pot experiment with maize either inoculated or not with Glomus intraradices was conducted in a sand:soil (3 :1) mix (pH 6.5) in a greenhouse. Our goal was to evaluate the contribution of mycorrhizae to uptake of Cu, Zn, Mn and Fe by maize as influenced by soil P and micronutrient levels. Two levels of P (10 and 40 mg kg−1 soil) and three levels of a micronutrient mixture: 0, 1X and 2X (1X contained, in mg kg−1 soil, 4.2 Fe, 1.2 Mn, 0.24 Zn, 0.06 Cu, 0.78 B and 0.036 Mo), were applied to pots. There were more extraradical hyphae at the low P level than at the high P level when no micronutrients were added to the soil. Root inoculation with mycorrhiza and application of micronutrients increased shoot biomass. Total Zn content in shoots was higher in mycorrhizal than non-mycorrhizal plants grown in soils with low P and low or no micronutrient addition. Total Cu content in shoots was increased by mycorrhizal colonization when no micronutrients were added. Mycorrhizal plants had lower Mn contents than non-mycorrhizal plants only at the highest soil micronutrient level. AMF increased total shoot Fe content when no micronutrients were added, but decreased shoot Fe when plants were grown at the high level of micronutrient addition. Psidium guajava L.) plantlets was determined during acclimatization and plant establishment. Guava plantlets were asexually propagated through tissue culture and grown in a glasshouse for 18 weeks. Half of the plantlets were inoculated with a mixed endomycorrhiza isolate from Mexico, ZAC-19, containing Glomus diaphanum, G. albidum and G. claroides. Plantlets were fertilized with modified Long Ashton nutrient solution that supplied 11 μg P ml−1. Gas exchange measurements were taken at 2, 4, 8, and 18 weeks after inoculation using a portable photosynthesis system. All micropropagated guava plantlets survived transplant shock. After 6 weeks, mycorrhizal plantlets had greater shoot growth rates and leaf production than non-mycorrhizal plantlets. This also corresponded with increased photosynthetic rates and stomatal conductance of mycorrhizal plants. By 18 weeks, mycorrhizal plantlets had greater shoot length, leaf area, leaf, stem, and root dry mass. However, gas exchange was comparable among treatments, in part because the container size was restricting growth of the larger mycorrhizal plantlets. Non-mycorrhizal plantlets had greater leaf area ratios and specific leaf areas than mycorrhizal plantlets. Increased leaf tissue mineral levels of P, Mg, Cu, and Mo also occurred with mycorrhizal plantlets. Roots of inoculated guava plantlets were heavily colonized with arbuscules, vesicles and endospores. Guava plantlets were highly mycotrophic with a mycorrhizal dependency index of 103%. Previous reports have indicated positive effects of enriched rhizosphere dissolved inorganic carbon on the growth of salinity-stressed tomato (Lycopersicon esculentum L. Mill. cv. F144) plants. In the present work we tested whether a supply of CO2 enriched air to the roots of hydroponically grown tomato plants had an effect on nitrogen uptake in these plants. Uptake was followed over periods of 6 to 12 hours and measured as the depletion of nitrogen from the nutrient solution aerated with either ambient or CO2 enriched air. Enriched rhizosphere CO2 treatments (5000 μmol mol-1) increased NO3 - uptake up to 30% at pH 5.8 in hydroponically grown tomato plants compared to control treatments aerated with ambient CO2 (360 μmol mol-1). Enriched rhizosphere CO2 treatments had no effect on NH3 + uptake. Acetazolamide, an inhibitor of apoplastic carbonic anhydrase, increased NO3 - uptake in ambient rhizosphere CO2 treatments, but had no effect on NO3 - uptake in enriched rhizosphere CO2 treatments. Ethoxyzolamide, an inhibitor of both cytoplasmic and extracellular carbonic anhydrase, decreased NO3 - uptake in ambient rhizosphere CO2 treatments. In contrast, a CO2 enriched rhizosphere increased NO3 - uptake with ethoxyzolamide, although not to the same extent as in plants without ethoxyzolamide. It is suggested that a supply of enriched CO2 to the rhizosphere influenced NO3 - uptake through the formation of increased amounts of HCO3 - in the cytosol. The impacts of various nitrogen sources, i.e. NO 3, NH4 + or NH4NO3 in combination with gaseous NH3, on nitrogen-, carbon- and water-use efficiency and 13C discrimination (δ13C) by plants of the C3 species Triticum aestivum L. (wheat) and the C4 species Zea mays L. (maize) were studied. Triticum aestivum and Z. mays were hydroponically grown with 2 mol · m−3 of N supplied as NO 3, NH4 + or NH4NO3 for 21 and 18 d, respectively, and thereafter exposed to gaseous NH3 at 320 μg · m−3 or to ambient air for 7 d. In T. aestivum and Z. mays over a 7-d growth period, nitrogen-use efficiency (NUE) values were influenced by N-sources in the decreasing order NH4NO3-N > NO 3-N > NH4 +-N and NO 3-N > NH4NO3-N > NH4 +-N, respectively. Fumigation with NH3 decreased the NUE values of plants grown with any of the N-forms. During 28- and 7-d growth periods, N-sources affected water-use efficiency (WUE) values in the decreasing order of NH4 +-N > NO 3-N≈NH4NO3-N in non-fumigated T. aestivum, while fumigation with NH3 increased the WUE of NO 3-grown plants. There were insignificant effects of N-sources on WUE values of Z. mays over 25- and 7-d growth periods. Furthermore, δ13C values in plant tissues (leaves, stubble and roots) were higher (less negative) in NH4 +-grown plants of T. aestivum and Z. mays than in those supplied with NH4NO3 or NO 3. Regardless of the N-form supplied to the roots of the plant species, exposure to NH3 caused more-positive δ13C values in the plant tissues. These results indicate that the variations in N-source were associated with small but significant variations in δ13C values in plants of T. aestivum and Z. mays. These differences in δ13C values are in the direction expected from differences in WUE values over long or short growth periods and with differences in the extent of non-Rubisco (ribulose-1,5-bisphosphate carboxylase-oxygenase, EC 4.1.1.39) carboxylate contribution to net C acquisition, as a function of N-source.  The influence of an arbuscular mycorrhizal (AM) fungus on phosphorus (P) and iron (Fe) uptake of peanut (Arachis hypogea L.) and sorghum (Sorghum bicolor L.) plants was studied in a pot experiment under controlled environmental conditions. The plants were grown for 10 weeks in pots containing sterilised calcareous soil with two levels of Fe supply. The soil was inoculated with rhizosphere microorganisms only or with rhizosphere microorganisms together with an AM fungus (Glomus mosseae [Nicol. & Gerd.] Gerdemann & Trappe). An additional small soil compartment accessible to hyphae but not roots was added to each pot after 6 weeks of plant growth. Radiolabelled P and Fe were supplied to the hyphae compartment 2 weeks after addition of this compartment. After a further 2 weeks, plants were harvested and shoots were analysed for radiolabelled elements. In both plant species, P uptake from the labelled soil increased significantly more in shoots of mycorrhizal plants than non-mycorrhizal plants, thus confirming the well-known activity of the fungus in P uptake. Mycorrhizal inoculation had no significant influence on the concentration of labelled Fe in shoots of peanut plants. In contrast, 59Fe increased in shoots of mycorrhizal sorghum plants. Atmospheric CO2 concentration continues to rise. It is important, therefore, to determine what acclimatory changes will occur within the photosynthetic apparatus of wheat (Triticum aestivum L. cv. Yecora Rojo) grown in a future high-CO2 world at ample and limited soil N contents. Wheat was grown in an open field exposed to the CO2 concentration of ambient air [370 μmol (CO2) mol−1; Control] and air enriched to ∼200 μmol (CO2) mol−1 above ambient using a Free-Air CO2 Enrichment (FACE) apparatus (main plot). A High (35 g m−2) or Low (7 and 1.5 g m−2 for 1996 and 1997, respectfully) level of N was applied to each half of the main CO2 treatment plots (split-plot). Under High-N, FACE reduced stomatal conductance (g s) by 30% at mid-morning (2 h prior to solar noon), 36% at midday (solar noon) and 27% at mid-afternoon (2.5 h after solar noon), whereas under Low-N, g s was reduced by as much as 31% at mid-morning, 44% at midday and 28% at mid-afternoon compared with Control. But, no significant CO2 × N interaction effects occurred. Across seasons and growth stages, daily accumulation of carbon (A′) was 27% greater in FACE than Control. High-N increased A′ by 18% compared with Low-N. In contrast to results for g s, however, significant CO2 × N interaction effects occurred because FACE increased A′ by 30% at High-N, but by only 23% at Low-N. FACE enhanced the seasonal accumulation of carbon (A′′) by 29% during 1996 (moderate N-stress), but by only 21% during 1997 (severe N-stress). These results support the premise that in a future high-CO2 world an acclimatory (down-regulation) response in the photosynthetic apparatus of field-grown wheat is anticipated. They also demonstrate, however, that the stimulatory effect of a rise in atmospheric CO2 on carbon gain in wheat can be maintained if nutrients such as nitrogen are in ample supply. Castor bean (Ricinus communis L.) plants were grown for 5–7 weeks in a controlled environment at 350 μl l−1 or 700 μl l−1 CO2. Carbon assimilation, assimilate deposition, dark respiration and assimilate mobilization were measured in leaves 2, 3 and 4 (counted from the base of the plant), and a balance sheet of carbon input and export was elaborated for both CO2 concentrations. Carbon dioxide assimilation was nearly constant over the illumination period, with only a slight depression occurring at the end of the day in mature source leaves, not in young source leaves. Assimilation was ca. 40% higher at 700 μl l−1 than at 350 μl l−1 CO2. The source leaves increased steadily in weight per unit area during the first 3 weeks, more at 700 μl l−1 than at 350 μl l−1 CO2. On top of an irreversible weight increase, there was a large gain in dry weight during the day, which was reversed during the night. This reversible weight gain was constant over the life time of the leaf and ca. 80% higher at 700 μl l−1 than at 350 μl l−1. Most of it was due to carbohydrates. The carbon content (as a percentage) was not altered by the CO2 treatment. Respiration was 25% higher in high-CO2 plants when based on leaf area, but the same when based on dry weight. The rate of carbon export via the phloem was the same during the daytime in plants grown at 350 μl l−1 and 700 μl l−1 CO2. During the night the low-CO2 plants had only 50% of the daytime export rate, in contrast to the high-CO2 plants which maintained the high export rate. It was concluded that the phloem loading system is saturated during the daytime in both CO2 regimes, whereas during the night the assimilate supply is reduced in plants in the normal CO2 concentration. Two-thirds of the carbon exported from the leaves was permanently incorporated as plant dry matter in the residual plant parts. This “assimilation efficiency” was the same for both CO2 regimes. It is speculated that under 350 μl l−1 CO2 the growing Ricinus plant operates at sink limitation during the day and at source limitation during the night. Nitrite influx into crayfish showed saturation kinetics, supporting a carrier-mediated uptake. Addition of 4,4′-diisothiocyanatostilbene-2,2′-disulfonate (DIDS: at 10−5, 10−4 and 10−3 M) and bumetanide (at 10−5 M and 10−4 M) to the ambient water did not significantly affect nitrite influx. Rather than suggesting that neither Cl/HCO3 exchange nor K+/Na+/2Cl cotransport were involved in the transport, this may reflect that the gill cuticle has a low permeability to the pharmacological agents, or that the sensitivity of the transport mechanism to the inhibitors is low. Nitrite accumulation in the haemolymph was significantly decreased during hypercapnic conditions compared to normocapnic conditions. This supports the idea that an acid–base regulatory decrease in Cl(influx)/HCO3 (efflux) induced by hypercapnia should decrease NO2 uptake if NO2 and Cl share this uptake route. The respiratory acidosis caused by exposure to hypercapnia alone was partially compensated by HCO3 accumulation in the haemolymph. Combined exposure to hypercapnia and nitrite improved pH recovery, mainly by augmenting the [HCO3 ] increase, but also by decreasing haemolymph PCO2. Exposure to nitrite in normocapnic water induced an initial increase in haemolymph [HCO3 ] and later also a decrease in PCO2. Thus, the improved acid-base compensation during combined hypercapnia and nitrite exposure was an amplification of this nitriteinduced response. Haemolymph base excess rose much more than haemolymph [Ca], suggesting that transfer of acid-base equivalents between animal and water was more important than H+ buffering by exoskeletal CaCO3 in mediating the increase in haemolymph [HCO3 ]. We tested the hypotheses that increased belowground allocation of carbon by hybrid poplar saplings grown under elevated atmospheric CO2 would increase mass or turnover of soil biota in bulk but not in rhizosphere soil. Hybrid poplar saplings (Populus×euramericana cv. Eugenei) were grown for 5 months in open-bottom root boxes at the University of Michigan Biological Station in northern, lower Michigan. The experimental design was a randomized-block design with factorial combinations of high or low soil N and ambient (34 Pa) or elevated (69 Pa) CO2 in five blocks. Rhizosphere microbial biomass carbon was 1.7 times greater in high-than in low-N soil, and did not respond to elevated CO2. The density of protozoa did not respond to soil N but increased marginally (P < 0.06) under elevated CO2. Only in high-N soil did arbuscular mycorrhizal fungi and microarthropods respond to CO2. In high-N soil, arbuscular mycorrhizal root mass was twice as great, and extramatrical hyphae were 11% longer in elevated than in ambient CO2 treatments. Microarthropod density and activity were determined in situ using minirhizotrons. Microarthropod density did not change in response to elevated CO2, but in high-N soil, microarthropods were more strongly associated with fine roots under elevated than ambient treatments. Overall, in contrast to the hypotheses, the strongest response to elevated atmospheric CO2 was in the rhizosphere where (1) unchanged microbial biomass and greater numbers of protozoa (P < 0.06) suggested faster bacterial turnover, (2) arbuscular mycorrhizal root length increased, and (3) the number of microarthropods observed on fine roots rose.
