Characterization of dairy cattle manure/wallboard paper compost mixture   总被引:5,自引:0,他引:5  
The aim of this research was to evaluate the use of manufacturing wallboard paper scraps as an alternative bulking agent for dairy cattle manure composting. The characteristics of the composting process were studied based on the changes in physico-chemical parameters and final compost quality. Composting of dairy cattle manure with wallboard paper was performed in a 481-L cylindrical reactor with vacuum-type aeration. Rapid degradation of organic matter was observed during the thermophilic stage of composting due to high microbial activity. High temperature and alkaline pH conditions promoted intense ammonia emission during the early stage of composting. The number of mesophilic and thermophilic microorganisms were found to be affected by changes in temperature at different composting stages. The total nitrogen (N), phosphorus (P), potassium (K), and sodium (Na) concentrations of the mixture did not change significantly after 28days of composting. However, the presence of gypsum in the paper scraps increased the calcium content of the final compost. The wallboard paper had no phyto-inhibitory effects as shown by high germination index of final compost (GI=99%).  相似文献

Changes in temperature, pH, moisture, C/N ratio, and bacterial community were monitored in Istanbul full-scale composting plant. C/N ratio steadily decreased during composting and final mature compost products had a C/N ratio of less than 20. During the composting process, temperature was mostly above 55 °C and decreased to mesophilic conditions in the matured stages. Different types of bacteria were dominant in every stage of composting and bacterial diversity changed mainly by temperature. Bacillus species were dominant in early stages of composting while Acinetobacter and Sphingobacterium strains were detected in thermophilic and maturing stages. Bacteria were mainly active in the degradation of cellulose and toxic organics while some strains had denitrification ability. Generally, thermophilic stages were more rich in bacterial diversity than mesophilic and hyperthermophilic conditions significantly changed the bacterial community.  相似文献

Ten composting facilities (CF) treating source-sorted organic fraction of municipal solid waste (OF) were sampled to study the relationship between the bulk density and the composting process (CP) development. Hundred and fourteen samples from different stages of the CP were considered (organic fraction, initial mixture, final decomposition, final maturation and compost), including the reject materials coming from the densimetric table at postprocessing. Total organic matter (TOM), moisture content (MC), wet bulk density and dry basis (BDd) were determined. Significant differences were detected for MC, TOM and BDd between some stages of the CP. The BDd increased along the CP while TOM decreased. Correlation studies showed a significant negative relationship between TOM and bulk density, especially BDd, during the CP, as a result of the biological activity. Moreover, a clear relationship was also found between TOM and BDd in samples related to reject materials. The results indicate that bulk density could be a simple and useful tool to evaluate the CP, in addition to the others parameters commonly used. At the same time, BDd could be an easy way to infer TOM lost within rejects.  相似文献

为了明确鸡粪好氧堆肥过程中细菌群落结构和功能的变化,采用高通量测序技术测定了好氧堆肥前、中、后3个时期样品的16S rRNA基因序列,并进行了生物信息学分析。结果表明, 3个堆肥阶段中仅有10%左右的分类操作单元(operational taxonomic units, OTUs)具有阶段特异性;不同发酵阶段细菌α多样性指数ACE、Chao1和Simpson均呈现先升高后降低的趋势,但各阶段间差异不显著(P<0.05)。3个发酵阶段优势菌门类相同,但丰度存在差异。线性判别分析[line discriminant analysis (LDA) effect size, LEfSe]法对细菌生物标志分析表明,从门到属水平共有49个物种,堆肥前期样品组中显著富集的物种最多,后期最少。原核分类群功能注释(functional annotation of prokaryotic taxa,FAPROTAX)对细菌功能多样性分析表明,堆肥前期细菌功能多样性最高,而随着发酵进行细菌功能富集程度增加、多样性降低。该研究为畜禽粪污好氧堆肥过程调控提供理论支撑和技术指导。  相似文献

Dehydrogenase enzymatic activity was determined to monitor the biological activity in a composting process of organic fraction of municipal solid waste. Dehydrogenase activity is proposed as a method to describe the biological activity of the thermophilic and mesophilic stages of composting. The maximum dehydrogenase activity was detected at the end of the thermophilic stage of composting, with values within 0.5-0.7mgg dry matter(-1)h(-1). Also, dehydrogenase activity can be correlated to static respiration index during the maturation mesophilic stage.  相似文献

Aims: The microbiota at industrial full‐scale composting plants has earlier been fragmentarily studied with molecular methods. Here, fungal communities from different stages of a full‐scale and a pilot‐scale composting reactors were studied before and after wood ash amendment. Methods and Result: The portion of fungal biomass, determined using phospholipid fatty acid analysis, varied between 6·3% and 38·5% in different composting phases. The fungal internal transcribed spacer (ITS) area was cloned and sequenced from 19 samples representing different stages of the composting processes. Altogether 2986 sequenced clones were grouped into 166 phylotypes from which 35% had a close match in the sequence databases. The fungal communities of the samples were related with the measured environmental variables in order to identify phylotypes typical of certain composting conditions. The fungal phylotypes could be grouped into those that dominated the mesophilic low pH initial phases (sequences similar to genera Candida, Pichia and Dipodascaceae) and those found mostly or exclusively in the thermophilic phase (sequences clustering to Thermomyces, Candida and Rhizomucor), but a few were also present throughout the whole process. Conclusions: The community composition was found to vary between suboptimally and optimally operating processes. In addition, there were differences in fungal communities between processes of industrial and pilot scale. Significance and Impact of the Study: The results of this study reveal the fungal diversity with molecular methods in industrial composting process. This is also one of the first studies conducted with samples from an industrial biowaste composting process.  相似文献

Serological evidence of hepatitis E virus infection (HEV) has been observed in both humans and different animal species living in non-endemic areas, suggesting that animals could be important reservoir for virus transmission to man. Antibodies to HEV have been detected in some Brazilian population groups. Nevertheless, sporadic cases of acute HEV infection have never been reported. We collected 271 serum samples from several domestic animals and also from pig handlers from Southeast of Brazil in order to investigate the seroprevalence of HEV infection. Anti-HEV IgG was detected in cows (1.42%), dogs (6.97%), chickens (20%), swines (24.3%), and rodents (50%), as well as in pig handlers (6.3%). The recognition of swine HEV infections in pigs in many countries of the world led us to investigate a larger sample of pigs (n = 357) from the same Brazilian region with ages ranging from 1 to > 25 weeks. IgG anti-HEV was detected in 100% of 7-day old pigs. Following a gradual decline between weeks 2 and 8 (probably due to loss of maternal IgG), the prevalence then steady increased until it reached 97.3% of animals older than 25 weeks. Besides the detection of anti-HEV antibodies in different animal species, the results showed that swine HEV infection seems to be almost universal within this Brazilian pig population. This is the first report that shows evidences of HEV circulation in Brazilian animal species and pig handlers.  相似文献

Large-scale Hand, Foot, and Mouth Disease (HFMD) outbreaks have frequently occurred in China since 2008, affecting more than one million children and causing several hundred children deaths every year. The pathogens of HFMD are mainly human enteroviruses (HEVs). Among them, human enterovirus 71 (HEV71) and coxsackievirus A16 (CVA16) are the most common pathogens of HFMD. However, other HEVs could also cause HFMD. To rapidly detect HEV71 and CVA16, and ensure detection of all HEVs causing HFMD, two real-time hybridization probe-based RT-PCR assays were developed in this study. One is a multiplex real-time RT-PCR assay, which was developed to detect and differentiate HEV71 specifically from CVA16 directly from clinical specimens within 1–2 h, and the other is a broad-spectrum real-time RT-PCR assay, which targeted almost all HEVs. The experiments confirmed that the two assays have high sensitivity and specificity, and the sensitivity was up to 0.1 TCID50/ml for detection of HEVs, HEV71, and CVA16, respectively. A total of 213 clinical specimens were simultaneously detected by three kinds of assays, including the two real-time RT-PCR assays, direct conventional RT-PCR assay, and virus isolation assay on human rhabdomyosarcoma cells (RD cells). The total positive rate of both HEV71 and CVA16 was 69.48% with real-time RT-PCR assay, 47.42% with RT-PCR assay, and 34.58% with virus isolation assay. One HFMD clinical specimen was positive for HEV, but negative for HEV71 or CVA16, which was identified as Echovirus 11 (Echo11) by virus isolation, RT-PCR, and sequencing for the VP1 gene. The two real-time RT-PCR assays had been applied in 31 provincial HFMD labs to detect the pathogens of HFMD, which has contributed to the rapid identification of the pathogens in the early stages of HFMD outbreaks, and helped to clarify the etiologic agents of HFMD in China.  相似文献

The objectives of this study were to investigate the fate of microorganisms by using cultivation methods as well as DNA analyses in a commercial microbiological additive (MA) in the course of the composting. Almost all the predominant species in the microbial succession during composting process determined by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) were in disagreement with those determined by the clone library method. None of the microbial species in the composting stages corresponded to the microorganisms identified in the MA either by the cultivation method or DNA analysis. The results in regard to predominant microorganisms of the MA detected from the liquid medium by the PCR-DGGE did not correspond with those detected from the MA itself and composting processes. Although no evidence was found that predominant species in the MA itself dominate in the composting process, predominant species diversity in the MA itself was markedly changed after culturing at different thermophilic temperatures. These results suggested that cultivable microorganisms in the MA did not become predominant in the composting process: however, some microorganisms that are detected from the MA itself by the DNA analysis may act effectively in the composting process.  相似文献

戊型肝炎病毒(Hepatitis E Virus,HEV)感染是一个重要的全球公共卫生问题,而猪被认为是HEV的天然宿主。HEV可以跨种间传播,且已经证实生吃感染的猪肉会导致人感染。在中国西藏许多地区仍然有生吃猪肉、猪肝等的习惯,且不同种家畜混合饲养,极易造成HEV感染和传播。然而中国西藏地区猪HEV流行情况报道甚少。文中对中国西藏5个地区市(拉萨、日喀则、山南、那曲和昌都)猪血清进行HEV Immunoglobulin-M(Ig M)和Ig G抗体检测,并通过逆转录巢氏PCR(RT-n PCR)进行HEV RNA检测和定量RT-PCR(q RT-PCR)进行病毒拷贝计算,首次报道了藏猪血清HEV RNA阳性率。结果显示,在西藏猪中HEV有较高的流行趋势。猪血清HEV Ig M抗体阳性率高达7.6%(26/340),HEV Ig G抗体阳性率为1.8%(6/340),HEV RNA阳性率高达7.6%(26/340),血清中病毒拷贝高达1.7×107 copies/m L,而且5个地区有不同的流行趋势。结果表明西藏猪HEV感染情况严重。有关部门应加强管理,以避免人与动物之间的交叉感染和暴发。  相似文献

Municipal sludge handling is a major problem facing wastewater treatment plants due to the high costs of treatment and disposal. This issue is of special importance in Jordan because of the critical economic situation as well as the lack of financial support for such nonprofit projects. This study investigates the possibility of solving this problem by testing a method of sludge stabilization that requires minimum initial and operating costs. The method tested here is sludge stabilization by composting which is an attempt to transform sludge into a safe, nuisance free, humus like product that can be applied safely to land and can become a source of income that would recover the costs of processing. Two types of composting systems were tested in this study, aerated static pile and windrow. Results obtained indicate that composting of dried sludge was not possible due to the extremely low moisture content; which was overcome by sludge seeding and mixing with amendment and bulking agents. This resulted in efficient stabilization and reduction of the amount of organic matter in the final compost. The experimental results obtained also indicate that both systems (aerated static pile and windrow) are efficient. The organic content of the sludge was reduced in the windrow system by 46% and in the aerated static pile by 66%. In addition, the total volatile solids had decreased in the windrow and the aerated pile by 26 and 73%, respectively. The heavy metals content of the final compost was examined and found to comply with the international standards.  相似文献

AIMS: To assist in the development of safe piggery effluent re-use guidelines by determining the level of selected pathogens and indicator organisms in the effluent ponds of 13 south-east Queensland piggeries. METHODS AND RESULTS: The numbers of thermotolerant coliforms, Campylobacter jejuni/coli, Erysipelothrix rhusiopathiae, Escherichia coli, Salmonella and rotavirus were determined in 29 samples derived from the 13 piggeries. The study demonstrated that the 13 final effluent ponds contained an average of 1.2 x 10(5) colony-forming units (CFU) 100 ml(-1) of thermotolerant coliforms and 1.03 x 10(5) CFU 100 ml(-1) of E. coli. The Campylobacter level varied from none detectable (two of 13 piggeries) to a maximum of 930 most probable number (MPN) 100 ml(-1) (two of 13 piggeries). Salmonella was detected in the final ponds of only four of the 13 piggeries and then only at a low level (highest level being 51 MPN 100 ml(-1)). No rotavirus and no Erysip. rhusiopathiae were detected. The average log10 reductions across the ponding systems to the final irrigation pond were 1.77 for thermotolerant coliforms, 1.71 for E. coli and 1.04 for Campylobacter. CONCLUSIONS: This study has provided a baseline knowledge on the levels of indicator organisms and selected pathogens in piggery effluent. SIGNIFICANCE AND IMPACT OF THE STUDY: The knowledge gained in this study will assist in the development of guidelines to ensure the safe and sustainable re-use of piggery effluent.  相似文献

Livestock manures contain numerous microorganisms which can infect humans and/or animals, such as Escherichia coli O157:H7, Listeria monocytogenes, Salmonella spp., and Mycobacterium avium subsp. paratuberculosis (Mycobacterium paratuberculosis). The effects of commonly used manure treatments on the persistence of these pathogens have rarely been compared. The objective of this study was to compare the persistence of artificially inoculated M. paratuberculosis, as well as other naturally occurring pathogens, during the treatment of dairy manure under conditions that simulate three commonly used manure management methods: thermophilic composting at 55 degrees C, manure packing at 25 degrees C (or low-temperature composting), and liquid lagoon storage. Straw and sawdust amendments used for composting and packing were also compared. Manure was obtained from a large Ohio free-stall dairy herd and was inoculated with M. paratuberculosis at 10(6) CFU/g in the final mixes. For compost and pack treatments, this manure was amended with sawdust or straw to provide an optimal moisture content (60%) for composting for 56 days. To simulate liquid storage, water was added to the manure (to simulate liquid flushing and storage) and the slurry was placed in triplicate covered 4-liter Erlenmeyer flasks, incubated under ambient conditions for 175 days. The treatments were sampled on days 0, 3, 7, 14, 28, and 56 for the detection of pathogens. The persistence of M. paratuberculosis was also assessed by a PCR hybridization assay. After 56 days of composting, from 45 to 60% of the carbon in the compost treatments was converted to CO2, while no significant change in carbon content was observed in the liquid slurry. Escherichia coli, Salmonella, and Listeria were all detected in the manure and all of the treatments on day 0. After 3 days of composting at 55 degrees C, none of these organisms were detectable. In liquid manure and pack treatments, some of these microorganisms were detectable up to 28 days. M. paratuberculosis was detected by standard culture only on day 0 in all the treatments, but was undetectable in any treatment at 3 and 7 days. On days 14, 28, and 56, M. paratuberculosis was detected in the liquid storage treatment but remained undetectable in the compost and pack treatments. However, M. paratuberculosis DNA was detectable through day 56 in all treatments and up to day 175 in liquid storage treatments. Taken together, the results indicate that high-temperature composting is more effective than pack storage or liquid storage of manure in reducing these pathogens in dairy manure. Therefore, thermophilic composting is recommended for treatment of manures destined for pathogen-sensitive environments such as those for vegetable production, residential gardening, or application to rapidly draining fields.  相似文献

Aims: The purpose of this study was to determine whether the methods used in compost operations of small and medium‐sized poultry forms resulted in the production of an amendment free of foodborne pathogens. Methods and Results: Nine compost heaps on five South Carolina poultry farms were surveyed at different stages of the composting process. Compost samples were analysed for coliforms and enriched for Escherichia coli O157:H7, Salmonella spp. and Listeria monocytogenes. The waste materials and composting practices differed among the surveyed farms. On two farms, new materials were added to heaps that had previously completed the active composting phase. Five compost heaps did not reach an internal temperature of 55°C, and c. 62% of all internal samples in the first composting phase contained moisture contents <40%. Escherichia coli was detected in 63% of the surface samples (n = 38) and 9·8% of the internal samples (n = 82) from the first composting phase, as compared with 16·7% of the surface samples (n = 12) and 0% internal samples (n = 24) from the second composting phase. Salmonella was detected in 26 and 6·1% of all surface and internal samples collected from heaps in the first composting phase, respectively, but was absent in all compost samples undergoing a second composting phase. The predominant Salmonella serotypes were Thompson, Montevideo and Anatum. Neither E. coli O157:H7 nor Lmonocytogenes was detected in any of the samples. Conclusions: Our results indicate that the conditions at the compost surface are suitable for pathogen survival, and the complete composting process can result in the elimination of pathogens in poultry wastes. Significance and Impact of the Study: This research provides information regarding the effectiveness of the composting practices and microbiological quality of poultry compost produced by small‐ and medium‐sized farms. Ensuring the safety of compost that may be applied to soils should be an integral part of preharvest food safety programme.  相似文献

Background

Hepatitis E virus (HEV) has been reported in the human population and pigs are a recognized reservoir for HEV and a possible source of HEV transmission to humans. Spray-dried porcine plasma (SDPP) is an ingredient commonly used in feed for pigs around the world. Even though processing conditions used to produce SDPP should be adequate to inactivate HEV, it was of interest to analyze commercial SDPP samples for presence of genome and antibodies (AB) against HEV and to retrospectively analyze serum samples collected from pigs used in past experiments that had been fed diets containing either 0% or 8% SDPP to detect potential transmission of HEV as determined by seroconversion.

Results

Eighty-five commercial SDPP samples were analyzed by ELISA and 100% of them contained AB against HEV, while 22.4% (11 of 49 samples analyzed) were positive for HEV RNA.Frozen sera samples (n?=?140) collected from 70 pigs used in past experiments that had been fed diets containing either 0% or 8% commercial SDPP was analyzed by ELISA for AB against HEV. Age of pigs at sera sampling ranged from 3 to 15 weeks and feeding duration of diets ranged from approximately 4 to 9 weeks. One lot of SDPP used in one experiment was analyzed and confirmed to contain HEV RNA. Regardless of the diet fed, some sera samples collected at the beginning of an experiment contained AB titer against HEV. These sera samples were collected from weaned pigs prior to feeding of the experimental diets and the HEV titer was probably from maternal origin. However, by the end of the experiments, HEV titer was not detected or had declined by more than 50% of the initial titer concentration.

Conclusions

To our knowledge, this is the first study reporting presence of HEV AB titer and RNA in SDPP. Retrospective analysis of serum collected from pigs fed diets with SDPP revealed no indication of seroconversion to HEV. The results indicate that feeding SDPP in diets for pigs does not represent a risk of transmitting HEV, even though HEV genome may be detected in SDPP.

Background  

Composting is an aerobic microbiological process that is facilitated by bacteria and fungi. Composting is also a method to produce fertilizer or soil conditioner. Tightened EU legislation now requires treatment of the continuously growing quantities of organic municipal waste before final disposal. However, some full-scale composting plants experience difficulties with the efficiency of biowaste degradation and with the emission of noxious odours. In this study we examine the bacterial species richness and community structure of an optimally working pilot-scale compost plant, as well as a full-scale composting plant experiencing typical problems. Bacterial species composition was determined by isolating total DNA followed by amplifying and sequencing the gene encoding the 16S ribosomal RNA.  相似文献

In the Netherlands peat was excavated for fuel until 1950. This gave rise to waterbodies (called turf ponds) which were then colonized by aquatic plants. Succession resulted in different aquatic plant communities and more terrestrialized stages such as floating fens. Nature conservation authorities started to excavate new turf ponds in 1990(ca. 2 ha y–1) with the aim to restore calcareous, mesotraphent ecosystems by totally setting back succession. A sequence of new species was revealed by mapping the aquatic vegetation from 1990 onwards.Chara spp. proved early colonizers, which was not expected because they have not been present in ditches and ponds in the area for the last 20 years. The denseChara vegetation prevents the resuspension of organic soil and contributes to keep the water column nutrient-poor and clear. ability of species such asStratiotes aloides to colonise the ponds from adjacent waterbodies is not possible because no open contact exists between a turf pond and a ditch. Management measures, such as re-introduction, have to be considered if the full-range of aquatic plant communities remains the goal.  相似文献
