The influence of the amount of red light relative to far red light (red/far red ratio) on leaf-sheath elongation, leaf length, tillering, assimilate partitioning to shoots and roots, and chlorophyll content in a dwarf wheat (Triticum aestivum L. cv Tibet Dwarf) and seven other dwarf and standard cultivars was determined. All cultivars tested showed far red-stimulated leaf and leaf-sheath elongation except Tibet Dwarf. Tibet Dwarf was also unresponsive to a brief end-of-day exposure to far red light, although the other cultivars exhibited increased leaf-sheath and leaf length. In these cultivars, the effects of an end-of-day 5-min far red exposure were reversible by a subsequent 5-min red light exposure. Shoot/root ratios were higher and tillering was suppressed by increased far red irradiance in all cultivars except Tibet Dwarf. In addition, Tibet Dwarf was less responsive to dark-induced leaf and leaf-sheath elongation (etiolation) and retained chlorophyll in dark-adapted leaves longer than control cultivars. Tibet Dwarf did not differ from the other cultivars in leaf chlorophyll content. Western blot analysis, using an antibody against phytochrome A, showed that dark-grown Tibet Dwarf shoots contained at least twice the amount of detectable phytochrome A protein present in the other wheat cultivars. These results, plus the short, thick-stemmed, dark-pigmented phenotype of Tibet Dwarf suggest the possibility of a mutation in the phytochrome/signal transduction pathway.  相似文献

Eight apple genotypes, including cultivars and breeding selections resistant and susceptible to Venturia inaequalis on foliage, were screened for shoot infection and the development of wood pustules following inoculation of shoot tips of 1–year maiden trees in the greenhouse. Where genotypes were highly resistant in terms of foliar symptoms (cvs Prima and Gavin), no shoot infection was observed. Where genotypes were highly susceptible in terms of foliar symptoms, then shoot tissue was either resistant (cv. Shinko) or susceptible (cv. Starking). In one experiment, shoot tips were inoculated sequentially as shoots extended. No pustules developed where inoculations were made later than May. In a further experiment, two leaf internode positions were inoculated on one occasion. Pustules were only observed where inoculations were made above the youngest unrolled leaf. The results suggested that shoot tips were more susceptible during early extension growth of the shoot. Pustules were noted in abundance on petioles of susceptible cultivars, and these probably contributed to early leaf abscission.  相似文献

A study was conducted to find out the role of ascorbic acid (AsA) in modulating growth and different physio-biochemical attributes of canola plants under well-watered as well as water-deficit conditions. Drought stress imposed on 60 % field capacity significantly decreased the shoot and root fresh and dry weights, leaf chlorophyll contents, shoot and root P, root K+, and activity of CAT enzyme, while increased chlorophyll a/b contents, MDA, NPQ, leaf total phenolics, free proline and GB contents in both canola cultivars. Foliar-applied varying levels (50, 100 and 150 mg L?1) of AsA enhanced shoot and root fresh and root dry weights, qN, NPQ, shoot and root P, AsA as well as the activity of POD enzyme particularly under drought stress conditions. Of both canola cultivars, cv. Dunkeld was higher in shoot fresh weights, ETR and F v /F m, MDA, proline and GB contents, and POD activity, however, cv. Cyclone in total phenolics and qN under well-watered and water-deficit conditions. Overall, the foliar-applied AsA had a positive effect, though not marked, on salt sensitive cv. Cyclone in terms of improved growth and other attributes, whereas exogenously applied AsA had a non-significant effect on relatively salt tolerant cv. Dunkeld.  相似文献

Some factors influencing in vitro cultures of potted Hibiscus rosa-sinensis L. using nodal cuttings were investigated. A protocol using a modified MS medium helped to overcome chlorosis, shoot tip necrosis (STN) and leaf drop. These disorders have been caused by mineral imbalance associated with calcium and iron deficiency. STN and leaf drop were overcome by increasing calcium level from 3 mM (MS standard concentration) to 9 mM, and this increase, in addition, enhanced shoot dry weight and shoot extension. The chlorophyll content and leaf area increased by increasing the iron concentration 3-fold from 98 μM to 295 μM. Furthermore, substituting Fe-EDTA with Fe-EDDHA resulted in an increase in chlorophyll content, leaf area and shoot extension. The most suitable multiplication medium for H. rosa-sinensis L. was demonstrated to be a modified MS medium containing 2.2 μM BAP and increased concentrations of calcium at 9 mM and iron at 295 μM provided as Fe-EDDHA. The shoots were rooted in half-strength modified MS medium containing 2.7 μM NAA. Acclimatization was successful with all shoots with or without roots.  相似文献

Effects of total irradiance on growth and flowering were studied in cineraria cv. Cindy Blue grown under warm (mean 21°C) glasshouse conditions. Efficiency of light conversion for leaf and shoot dry weight increase were reduced from 0.08 to 0.02 as the mean daily light integral increased from 0.9 to 4.4 MJ m-2 day-1 but no significant difference in leaf area were associated with this. Specific leaf area decreased exponentially from 0.07 to 0.02 m2g1 over the cumulative irradiance range 23 to 127 MJ m-2 after the start of treatments and thereafter remained stable. A light integral of 19.2 MJ m-2 were required for initiation of one leaf in plants grown under a daily integral of 4.4 MJ m-2 day-1, as compared with only 5.1 MJ m-2day-1 required per leaf in plants grown at less than 0.9 MJ m-2day-1. Neither chronological duration of juvenile development nor leaf number below the flower was affected by irradiance. However, as the rate of leaf initiation increased with irradiance up to 2.4 MJ m-2day-1 so the rate of progress to flower visibility increased linearly with irradiance over the same range. This rate then remained constant from 2.4 to 4.4 MJ m-2day-1. Length of the main flowering shoot decreased and the number of flowering shoots increased as irradiance increased from 0.9 to 2.4 MJ m-2 day-1 and then remained unchanged by further increases in irradiance.  相似文献

The objective of this research was to develop an efficient protocol for shoot regeneration from leaf segments of the Chrysanthemum cv. Vivid Scarlet by examining the effects of plant growth regulators, dark incubation period, gelling agents, and silver nitrate. The highest number of shoots per explant (12.3) was regenerated from leaf explants cultured on Murashige and Skoog (MS) medium supplemented with a combination of 1 mg L−1 of 6-benzyladenine (BA) and 2 mg L−1 of α-naphthaleneacetic acid (NAA) under light conditions without any initial dark period. Gelrite was the most effective gelling agent for shoot regeneration among those tested, whereas the presence of silver nitrate distinctly inhibited shoot regeneration. Superior plant growth and rooting was observed on a hormone-free MS medium solidified with Gelrite. Flow cytometry analysis revealed no ploidy variation between the regenerated plants and the mother plant grown under greenhouse conditions. The established protocol was applicable to shoot regeneration for four out of six cultivars tested. This research will facilitate the genetic transformation and micropropagation of Chrysanthemum cultivars.  相似文献

This paper describes multiple shoot regeneration from leaf and nodal segments of a medicinally important herb Centella asiatica L. on Murashige and Skoog’s (MS) medium supplemented with a range of growth regulators. The highest number of multiple shoots was observed on MS augmented with 3.0 mg dm−3 N6-benzylaminopurine (BAP) and 0.05 mg dm−3 α-naphthaleneacetic acid (NAA). Leaf explant showed maximum percentage of cultures regenerating shoots (81.6 %), with the highest shoot number (8.3 shoots per explant) and the shoot length (2.1 cm) whereas, nodal explant showed less number of shoots with callus formation at the base cut end. Successive shoot cultures were established by repeatedly sub-culturing the original explant on a fresh medium. Rooting of in vitro raised shoots was best induced on half strength MS supplemented with 0.5 mg dm−3 indole-3-butyric acid (IBA) with highest percentage of shoot regenerating roots (76.8 %) with 3–4 roots per shoot. Plantlets were acclimated in Vermi-compost and eventually established in soil. Contents of chlorophyll, total sugars, reducing sugars and proteins were estimated in leaf tissue from both in vivo and in vitro raised plants. Chlorophyll content was higher in in vivo plants, whereas other three components were higher in in vitro plants.  相似文献

测定槐树实生树和嫁接树叶片的光合光响应特性、比叶重和叶绿素含量及枝梢生长的结果表明,嫁接树枝梢基径和高度增长趋势与实生树一致,但嫁接树一次、二次梢基径和高度增长量均大于实生树。嫁接树的最大净光合速率、光饱和点、比叶重和单位重量的叶绿素含量均显著高于实生树,高的光合能力与其单位面积叶片重量增加和单位重量叶绿素含量增高有关。嫁接槐树的类胡萝卜素与叶绿素比值高于实生槐树,叶绿素a,b比值和单位重量类胡萝卜素含量显著高于实生槐树,表明其对强光的适应性强。  相似文献

Slow growth storage has been achieved for Castanea sativa (cv. ‘Montemarano’) shoot cultures over a duration of 48 mo at a temperature of 8°C, where 82% of explants survived and were able to resume normal growth after transfer to standard culture conditions at 23°C. The evaluation of the chlorophyll content of leaves also showed no differences between material stored for 48 mo and control material subcultured at 23°C. With a storage temperature of 4°C, the survival of shoots was significantly lower at approximately 56% after 12 mo, and no plants recovered after 24-mo storage. The presence of 6-benzyladenine 0.44 μM in the culture medium proved to be necessary for the recovery of healthy shoots, while pre-treatments with different concentrations of abscisic acid did not significantly influence the survival of shoots following storage conditions. A low level of light during slow growth storage resulted in positive effects on the rate of shoot survival over the longest preservation periods.  相似文献

Stress tolerance of two Egyptian cotton cultivars (Gossypium barbadense L.) (Giza 45 and Giza 86) exposed to various doses (40, 80, 160, and 320 min) of artificial ultraviolet-A (UV-A) radiation (366 nm) was investigated. Seed germination of cv. Giza 86 was promoted at 40 min, progressively inhibited at 80 and 160 min, and completely suppressed at 320 min irradiation. However, seed germination of cv. Giza 45 was not promoted but inhibited by UV-A light and stopped at the dose of 160 min. In contrast to seed germination, seedling growth of cv. Giza 86 was negatively stressed even at 40 min-dose. UV-A radiation reduced leaf carbohydrate content and shoot growth of both cultivars, but the response was comparatively higher in cv. Giza 45. UV-A radiation decreased chlorophyll (Chl) and carotenoid contents in parallel with an increase in the Chl a/b ratio, diminished Hill reaction activity, and quenched Chl a fluorescence independent of the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea, suggesting an inhibitory effect on both the water-splitting system and electron transport from the primary to the secondary acceptors of photosystem II (PSII) (acceptor side). UV-A radiation also decreased the ratio of unsaturated to saturated fatty acids in thylakoid membranes, thus indicating that the inhibition of PSII activity was followed by lipid peroxidation and changes in thylakoid membrane fluidity. These changes reflect the disturbance of structure, composition, and functioning of the photosynthetic apparatus, as well as the sensitivity of PSII to UV-A stress. Both cultivars developed adaptive mechanisms for damage alleviation involving the accumulation of flavonoids, total lipids, and total soluble proteins, as well as development of smaller and thicker leaf blades. Since cv. Giza 86 showed comparatively higher level of adaptation, it tolerates UV-A stress more successful than cv. Giza 45.  相似文献

The physiological basis of salt tolerance of two cultivars of blackgram, cv Candhari Mash (relatively salt tolerant) and cv Mash 654 (salt sensitive), was assessed in salinized sand culture at the flowering stage. Increasing NaCl concentration in the rooting medium significantly reduced the chlorophyll a, chlorophyll b, and total chlorophyll, leaf water potential (Ψw), leaf solute potential (Ψs), and leaf turgor potential (Ψp) in both the cultivars. Leaf protein and proline content was increased as a result of increasing salt concentration in both cultivars. High salt concentrations had no significant effect on the seed protein content of both cultivars. At high salinities, cv Candhari Mash had significantly greater chlorophyll a, chlorophyll b and total chlorophyll, leaf water potential, solute potential, and turgor potential than cv Mash 654, but the latter had greater leaf proline content than cv Candhari Mash. Cultivars did not differ significantly for both leaf and seed protein contents. The relatively salt tolerant cv Candhari Mash maintained high leaf water potential and turgor potential to resist salt injury. Leaf proline content had negative correlation with salt tolerance in blackgram.  相似文献

Leaf shape and size, stomata dimensions and density, chlorophyll content and photosynthetic rate of the common oak leaves were investigated in shoots of various growth phases (polycyclic growth). Mean and maximal daily net photosynthetic rates, shoot length, leaf area, stomatal density and chlorophyll contents were significantly higher in shoots of the second growth phase. Growth, sun/shade acclimation and nitrogen nutrition were examined in Tradescantia fluminensis to gain greater understanding of why this species is so successful in New Zealand native forest remnants. Over a two year period, the rate of shoot extension of T. fluminensis in a New Zealand mixed mahoe (Melicytus ramiflorus) coastal forest remnant showed a similar pattern to monthly mean values for mean daily air temperature and day length. Growth at the shoot apex was balanced by death at the shoot base. During the first year, nitrate (NO3-)content of the plant in the field was always > 250 mu mol per g dry weight. On high NO3- supply in pot experiments, in a glasshouse or outdoors, total plant dry weight increased with increased relative irradiance from 1 to 30-50% (open ground photosynthetically active radiation = 100% relative irradiance). Changes in shoot to root dry weight ratio (S:R), specific leaf area (SLA) and leaf chlorophyll, carotenoid and protein content associated with decreased irradiance from 50 to 1% were similar to those associated with increased distance into the forest remnant and are discussed in relation to shade acclimation. Values for S:R (>30.1) and SLA (approximate to 900 cm(2) per g dry weight) were extremely high at low irradiance. These results support earlier conclusions that irradiance level is likely to be the primary factor limiting the extent of colonisation of forest remnants by T. fluminensis. Under glasshouse conditions, the growth response of T. fluminensis to different ammonium and NOS concentrations was similar to that previously reported for herbaceous species capable of rapid growth. Leaf nitrate reductase activity was within the range previously reported for fast growing species. Tradescantia fluminensis accumulated substantial amounts of NO3- in shoots with no depression in growth. This NO3- was utilised when nitrogen became limiting to growth. An 'invasion strategy' of T. fluminensis into N.Z. native forest remnants is proposed.  相似文献

Two cultivars of Digitaria eriantha (cold-sensitive cv. Sudafricana and cold-resistant cv. Mejorada INTA) were exposed to cold stress (5?°C) for 0, 6, 24, or 72?h, and compared in terms of leaf and root growth, recovery period, shoot and leaf anatomy, and levels of chlorophyll, auxin (indole-3-acetic acid, IAA) and cytokinins (CKs). In Sudafricana, cold treatment caused reduced growth, slight changes in chlorophyll level, reduced levels of IAA and CK iso-pentenyladenine (iP), and reduced leaf dry weight (DW) and fresh weight (FW) during the recovery period. Anatomical damage was observed in chloroplasts, main stem, and axillary buds. Ultrastructural study showed reduced numbers of starch grains in chloroplasts of the bundle sheath and mesophyll. In Mejorada, cold treatment had no significant effect on growth or chlorophyll level. Leaf DW and FW quickly returned to normal levels during the recovery period. Anatomy of ground meristem was affected, but ultrastructure of bundle sheath and mesophyll chloroplasts was not. The cold tolerance of cv. Mejorada appears to be related to the stability of chlorophyll and CK levels, increase of IAA, and maintenance of normal shoot and leaf anatomy and ultrastructure.  相似文献

We have been examining the importance of the root system on shoot growth and development using a developmentally disabled hybrid of the common bean Phaseolus vulgaris L. Parental cultivars (P. Vulgaris cv. Redkloud of Mesoamerican origin, and P. vulgaris cv. Batt of Andean origin) grow normally, but crosses produce F1 hybrids exhibiting hybrid weakness associated with reduced root and shoot growth. In this study, applications of benzylaminopurine (BAP) to roots of F1 hybrids increased the number of root tips and leaves. Reciprocal grafting was used to study the effects of the root system on shoots. Grafting of roots of the Mesoamerican cultivar onto shoots of F1 hybrids increased the cytokinin concentrations in leaves of F1 hybrids and removed the characteristics associated with hybrid weakness. To determine whether factors in the xylem sap enhanced leaf growth, leaf discs were incubated on sap collected from Mesoamerican and Andean cultivars. Sap from Mesoamerican plants enhanced the growth of leaf discs excised from F1 hybrids more than sap collected from Andean cultivars. Estimates of the transport of zeatin riboside (ZR)–type cytokinins from roots of F1 hybrids indicated that transport out of hybrid roots was reduced compared with those transported out of Mesoamerican or Andean roots. Results suggest that ZR-type cytokinins are involved in hormonal integration between roots and shoots of P. vulgaris and that one of the barriers to hybridization between Andean and Mesoamerican landraces is related to hormone transport. Conditions were defined for precocious differentiation and improved growth of corms at the base of gladiolus shoots. Shoots were derived from explants cultured on agar solidified media, and corm regeneration was obtained in subsequent liquid shake cultures. Benzyladenine (BA), at 10-7 M, was found to have a stimulating effect mainly when provided to the shoots prior to manifestation of corm growth. Paclobutrazol and sucrose promoted corm formation when supplemented to the liquid media. Paclobutrazol, at 10 mg l-1, shifted assimilate allocation towards the growing corm. A differential promotion of corm development by sucrose was not observed, and the concentration of sucrose at which the sugar demand for maximal shoot and corm growth is satisfied (60 g l-1) was unaltered by the presence of paclobutrazol. The rate of corm growth on shoots cultured in a liquid medium supplemented with paclobutrazol and a saturating sucrose concentration, was a function of the length of the shoot's leaf blades, and was similar in light and in dark.  相似文献

We examined the effect of FeEDDHA on multiplication via axillary branching and adventitious shoot regeneration from leaf explants in five red raspberry cultivars. When applied during multiplication, FeEDDHA reduced chlorosis, increased content of chlorophyll and iron but had no effect on the number of side shoots in four of the five cultivars. Addition of FeEDDHA to regeneration medium increased the percentage of regenerating leaves in some cultivars and the number of adventitious shoots in all five cultivars.  相似文献

Deciduous and evergreen species are segregated on northeast and southwest slopes of the southern Appalachian Mountains. The segregated distributions of three ericaceous shrubs (Rhododendron maximum valley positions; Rhododendron periclymenoides on northeast slopes; Kalmia latifolia on southwest slopes) were compared to the respective irradiance environments. Growth patterns of field plants, and photosynthetic acclimation of each species to three irradiance treatments in a phytotron were studied. Rhododendron maximum, an evergreen species, was found to be most sensitive to high radiation. In phytotron experiments, quantum yield, light saturated photosynthetic capacity, photosynthesis per chlorophyll, and water use efficiency decreased at high ambient irradiance for R. maximum. These characteristics limit the growth of R. maximum on high irradiance southwestern slopes. Both K. latifolia and R. periclymenoides were able to improve their photosynthetic performance at high ambient irradiance. Rhododendron periclymenoides, a deciduous species, was found to continue increasing leaf conductance at high irradiance without an increase in photosynthesis indicating a possible limitation by water in high light environments such as southwest slopes. Kalmia latifolia, an evergreen species, had reduced photosynthetic capacity and reduced water use efficiency when grown in low irradiance conditions which coincides with the higher K. latifolia abundance on high light, southwestern slopes.  相似文献

Seedlings of wheat (Triticum aestivum L.) cultivars Jing 411, Jinmai 30 and Yangmai 10 were exposed to 0, 10, 20, 30, 40 or 50 μM of CdCl2 in a solution culture experiment. The effects of cadmium (Cd) stress on wheat growth, leaf photon energy conversion, gas exchange, and Cd accumulation in wheat seedlings were investigated. Gas exchange was monitored at 3, 9, 24 days after treatment (DAT). Growth parameters, chlorophyll content, leaf chlorophyll fluorescence, and Cd concentration in shoot and root were measured at 24 DAT. Seedling growth, gas exchange, chlorophyll content, chlorophyll fluorescence parameters were generally depressed by Cd stress, especially under the high Cd concentrations. Cd concentration and accumulation in both shoots and roots increased with increasing external Cd concentrations. Relationships between corrected parameters of growth, photosynthesis and fluorescence and corrected Cd concentrations in shoots and roots could be explained by the regression model Y = K/(1 + exp(a + bX)). Jing 411 was found to be Cd tolerant considering parameters of chlorophyll content, photosynthesis and chlorophyll fluorescence in which less Cd translocation was from roots into shoots. The high Cd concentrations were in shoots and roots in Yangmai 10 which has been found to be a relative Cd tolerant cultivar in terms of most growth parameters.  相似文献

Summary The effects of irradiance during growth on biomass allocation, growth rates, leaf chlorophyll and protein contents, and on gas exchange responses to irradiance and CO2 partial pressures of the evergreen, sclerophyllous, chaparral shrub, Ceanothus megacarpus were determined. Plants were grown at 4 irradiances for the growth experiments, 8, 17, 25, 41 nE cm-2 sec-1, and at 2 irradiances, 9 and 50 nE cm-2 sec-1, for the other comparisons.At higher irradiances root/shoot ratios were somewhat greater and specific leaf weights were much greater, while leaf area ratios were much lower and leaf weight ratios were slightly lower than at lower irradiances. Relative growth rates increased with increasing irradiance up to 25 nE cm-2 sec-1 and then leveled off, while unit leaf area rates increased steeply and unit leaf weight rates increased more gradually up to the highest growth irradiance.Leaves grown at 9 nE cm-2 sec-1 had less total chlorophyll per unit leaf area and more per unit leaf weight than those grown at 50 nE cm-2 sec-1. In a reverse of what is commonly found, low irradiance grown leaves had significantly higher chlorophyll a/b than high irradiance grown leaves. High irradiance grown leaves had much more total soluble protein per unit leaf area and per unit dry weight, and they had much higher soluble protein/chlorophyll than low irradiance grown leaves.High irradiance grown leaves had higher rates of respiration in very dim light, required higher irradiances for photosynthetic saturation and had higher irradiance saturated rates of photosynthesis than low irradiance grown leaves. CO2 compensation irradiances for leaves of both treatments were very low, <5 nE cm-2 sec-1. Leaves grown under low and those grown under high irradiances reached 95% of their saturated photosynthetic rates at 65 and 85 nE cm-2 sec-1, respectively. Irradiance saturated rates of photosynthesis were high compared to other chaparral shrubs, 1.3 for low and 1.9 nmol CO2 cm-2 sec-1 for high irradiance grown leaves. A very unusual finding was that leaf conductances to H2O were significantly lower in the high irradiance grown leaves than in the low irradiance grown leaves. This, plus the differences in photosynthetic rates, resulted in higher water use efficiencies by the high irradiance grown leaves. High irradiance grown leaves had higher rates of photosynthesis at any particular intercellular CO2 partial pressure and also responded more steeply to increasing CO2 partial pressure than did low irradiance grown leaves. Leaves from both treatments showed reduced photosynthetic capability after being subjected to low CO2 partial pressures (100 bars) under high irradiances. This treatment was more detrimental to leaves grown under low irradiances.The ecological implications of these findings are discussed in terms of chaparral shrub community structure. We suggest that light availability may be an important determinant of chaparral community structure through its effects on water use efficiencies rather than on net carbon gain.  相似文献
