The European spruce bark beetle, Dendroctonus micans Kugelann (Coleoptera: Scolytidae), is one of the most serious destructive pests of oriental spruce (Picea orientalis L.). In order to find an effective biocontrol agent against this pest, we determined the effectiveness of entomopathogenic fungi against D. micans. Virulence of nine highly pathogenic strains including Beauveria bassiana (2), Beauveria cf. bassiana (2), Metarhizium anisopliae (2), Metarhizium sp. (1), Isaria fumosorosea (1) and Evlachovaea sp. (1) was evaluated on D. micans larvae and adults under laboratory conditions. Mortality values for both larvae and adults ranged from 83 to 100% and from 23 to 100%, respectively. Larvae were more susceptible to fungi than adults (P<0.05). Based on screening tests, B. cf. bassiana isolate KTU-53 was found the most effective isolate. LC50 values were calculated as 1.77×104 and 2.65×104 conidia mL?1 for isolate KTU-53 against larvae and adults, respectively. Consequently, B. cf. bassiana isolate KTU-53 appears to be the most promising microbial control agent for biocontrol of D. micans.  相似文献

1 Great spruce bark beetle Dendroctonus micans (Kug.) (Coleoptera: Scolytidae) has been subject to an Integrated Crop Management (ICM) regime in Great Britain since its first discovery in 1982. The elements of the ICM approach are sanitation felling of the initial infestations, restriction on movement of potentially infested conifers to prevent spread to uninfested parts of the country and biological control through rearing and release of the specific predatory beetle Rhizophagus grandis Gyll. (Coleoptera: Rhizophagidae). Such a programme carries costs that have to be evaluated against the benefits to the ICM strategy. 2 This paper presents a cost-effectiveness analysis of three options – (i) continue with the current policy of restriction on movement of infested timber and use of the imported predator R. grandis, (ii) do nothing (i.e. drop all attempts to reduce the spread of D. micans), or, (iii) introduce a new east–west Dendroctonus Micans Control Area (DMCA) to define the management area. Assumptions on rates of mortality and natural spread were based on research into D. micans in Britain’s spruce forests. 3 Appraisal indicated that the current policy was the most cost-effective, even when subject to sensitivity analysis to test the limits of the assumptions included in the models. The European spruce bark beetle, Dendroctonus micans Kugelann (Coleoptera, Scolytidae), is one of the most serious pests of oriental spruce (Picea orientalis L.) in Turkey. In this study, we investigated bacterial flora of D. micans collected from different populations of the forests of Eastern Black Sea Region of Turkey from 2002 to 2004. Seven different bacteria were isolated from healthy, diseased and dead specimens based on the color of colony and morphology. According to morphological, physiological and biochemical properties, metobolic enyzme profile by BIOLOG microtiter plate system, and total cellular fatty acid profile by Microbial Identification System (MIS), isolates were identified as Micrococcus luteus, Bacillus thuringiensis subsp. morrisoni, Serratia grimesii, Enterobacter cloaceae, Enterobacter intermedius, Streptococcus sp. and Pseudomonas putida. This is the first study on the bacterial flora of D. micans.  相似文献

The great spruce bark beetle, Dendroctonus micans (Kugelann) (Coleoptera: Curculionidae), has been a potential threat for Turkey and the entire Eurasian spruce forests for many years. Control strategies which have been applied so far are still insufficient to prevent its damage. A previous study has shown that a Beauveria isolate (ARSEF 9271) proved to be an efficient microbial control agent against the great spruce bark beetle. In this study, this isolate was identified as B. pseudobassiana based on the partial sequence of EF1‐α and ITS sequence. A conidial suspension (1 × 108/ml) of this fungus caused 100% mortality on both larvae and adults of D. micans within 5 and 6 days, respectively. Also, it caused 100% mycosis value on both larvae and adults. Mortality values of horizontal transmission experiments between larvae and adults which were contaminated with 1 × 106/ml spore suspension at 25%, 50%, 75% and 100% rates were determined as 100% after 15 days at 20°C under the laboratory conditions. We also determined the decrease of the damage in spruce wood block (15 × 25 cm) when the contamination rate of the larvae was increased. Our results indicate that B. pseudobassiana ARSEF 9271 seems to be a very promising biocontrol agent against D. micans.  相似文献

AIMS: To formulate the entomopathogenic fungus Beauveria bassiana in invert emulsion, then apply it against adults of almond bark beetle (Scolytus amygdali) under laboratory and field conditions. METHODS AND RESULTS: The effect of formulated B. bassiana in invert emulsion against S. amygdali adults was shown by comparing the mortality percentage of adults exposed to the formulated fungus using a Petri dish treatment method and by field applications to infested peach trees with mortality of adults exposed to the unformulated fungus or the untreated control. Results obtained from both exposure methods have indicated that treatment of S. amygdali adults with the formulated fungus resulted in a significantly higher mean mortality percentage (P < 0.05) when compared with the treatment with the unformulated fungus or the untreated control. This mortality ranged from 81.2 to 100%, 10 days after treatment with the formulated fungus when compared with 6.7 to 49.6% mortality, 10 days after treatment with the control or the unformulated fungus, respectively. Viability of the fungus conidia in invert emulsion was assessed by calculating the germination percentage of the conidia over time. Results indicated a high storage stability shown by a small loss of germination percentage for the formulated conidia of both strains (5.8 to 8.4% over a 12-week period) vs a low storage stability shown by a high loss of germination percentage for the unformulated conidia of the same strains (58.9 to 61.0% over the same period). The presence of B. bassiana in the galleries of beetles following the treatment of infested trees was shown in the present research. CONCLUSIONS: The results obtained have demonstrated a significantly higher level of efficacy of formulated B. bassiana in invert emulsion against S. amygdali adults under laboratory and field conditions. The ingredients of invert emulsion used in the formulation of the fungus had a negligible effect on the viability of formulated conidia when compared with the unformulated. SIGNIFICANCE AND IMPACT OF THE STUDY: Results obtained in the present research are promising and may be exploited commercially to control S. amygdali adults on various species of stone fruit trees, especially peach trees. This type of biocontrol of this insect may be used as an alternative means to chemical control for management of the insect. No adverse environmental impacts of the fungus or its formulation have been observed during application.  相似文献

An infection of the great European spruce bark beetle Dendroctonus micans (from Turkey) by the parasitic green alga Helicosporidium is described. This is the first time that Helicosporidium has been found to infect a bark beetle (Scolytidae) and the first time that a naturally infected beetle has been reported for the Eurasian continent. The typical cysts of Helicosporidium contain three ovoid cells and one helical, filamentous cell. The morphological characteristics are revealed by light and electron microscopy. Distinct electron-dense inclusions with a peculiar ultrastructure may represent pyrenoids. Since D. micans is an important pest, the discovery of a natural pathogen may offer a chance for biological control.  相似文献

Larvae of mealworms Tenebrio molitor L. (Coleoptera: Tenebrionidae) have been used as animal feed, but fungal pathogens rapidly downsize the populations, resulting in economic losses. In this work, we established an effective management strategy for fungal pathogens. An entomopathogenic fungus, Beauveria bassiana, was isolated from mealworm cadavers. The bioassay of some isolates of this species at >90% relative humidity revealed that the ERL1575 isolate had the highest virulence. At 20–30% RH, ERL1575 conidia when ingested produced 80% mortality but when sprayed topically produced only <10% mortality. Mealworms that had ingested conidia were exposed to 20, 25, 30 and 35°C and high humidity (>95%) for 5 days. This experiment produced about 90% mortality except at 35°C where mortality was <20%. When 40 fungicides were assayed against ERL1575, fluazinam (1000‐fold) and mancozeb (667‐fold) significantly inhibited conidial germination and/or hyphal growth. When fluazinam and mancozeb were added to the mealworm diet of conidia‐inoculated wheat bran, most were alive 3 days post application. However, 100% mortality resulted 3 days post application in the conidia‐inoculated wheat bran without any fungicides. In conclusion, B. bassiana isolates are pathogenic at <30°C when they are ingested by mealworms but fluazinam and mancozeb can be used for management to control the pathogen in their cultures.  相似文献

Summary The role of lignin as a physical defence against Dendroctonus micans was investigated in laboratory feeding experiments. The effect of lignin is dose-dependent, reducing larval survival, growth rate, and weight, as well as affecting gallery construction. Adults lay fewer eggs in lignified bark and also tend to construct abnormal galleries. The distribution of lignin in trees suggests a role in defence against bark beetles that feed in the thicker bark on the lower bole.  相似文献

This paper provides a simple mathematical model of the aggregation behavior of the gregarious intracortical-feeding larvae of the bark beetle, Dendroctonus micans. The model's assumptions are that each individual produces pheromones at a constant rate and reacts positively to a pheromone gradient. These hypotheses have been tested by comparing experiments and simulations, both of which showed that (1) homogeneously distributed individuals aggregate rapidly at the center of the experimental or theoretical arena and (2) eccentric, preformed groups succeed in attracting dispersed individuals, provided that the initial size of these groups is sufficiently high. There is good agreement between most of the experimental and theoretical results, providing a link among chemical communication, density of larvae, and random events occurring during the development of a spatial structure.  相似文献

Abstract

In this study, Metarhizium anisopliae TR 106 and Beauveria bassiana TR 217 was tested against fourth instar larvae of Thaumetopoea pityocampa. The LT50 and LT90 of 1?×?106 concentration of M. anisopliae against T. pityocampa were 3.60 and 4.11 for direct application, while these were 2.87 and 3.60?days, respectively in leaves application. The LT50 and LT90 of the 1?×?108 concentration of the same isolate were 2.50 and 2.95?days for direct application, and 2.98 and 3.74?days for leaves application. The LT50 of insect and leaves application for 1?×?106 of B. bassiana were 3.75 and 3.49?days, respectively. The LT90 of same concentration for insect application was 4.48?days, while LT90 for leaves application was 4.63?days. Similarly, LT50 of insect and leaves application for 1?×?108 of B. bassiana were 3.03 and 3.31?days, while LT90 were 3.68 and 4.29?days, respectively. We determined the virulence of Beauveria bassiana, Metarhizium anisopliae, Isaria fumosoroseus and Lecanicilium lecanii against larvae of Chilo suppressalis Walker by bioassay and evaluated several enzymatic and non-enzymatic components. LC50 values of the entomopathogenic fungi revealed 90, 32, 45,000, 4600, 42,000 and 1,540,000 spores/larva for isolates BB1–BB3 of B. bassiana, I. fumosoroseus, M. anisopliae and L. lecanii, respectively. Isolate BB3 and I. fumosoroseus had the highest amounts of total protein and hydrophobin and isolates BB3 and M. anisopliae showed the highest activities of lipases and chitinases. In case of proteases, the highest activities were observed for Pr1 of BB1 and Pr2 of L. lecanii. The highest general esterase activities were obtained in I. fumosoroseus and BB1 when 1-naphtyl acetate and 2-naphtyl acetate were used as substrates. The highest activity of glutathione S-transferase (GST) was observed in I. fumosoroseus by using both reagents but BB1 demonstrated the highest activities of alkaline phosphatase and acid phosphatase. Clustering of the fungi using biochemical enzymes revealed BB2 and BB3 as a separate group of entomopathogenic fungi. In another group, I. fumosoroseus and L. lecanii had the most similarity and were separated from BB1 and M. anisopliae. The fungi exhibited different virulence on larvae of C. suppressalis by producing adhering protein and extracellular enzymes. Overall, results of the bioassays and clustering based on enzymatic activities revealed that isolate BB2 was the most effective fungus against larvae of C. suppressalis.  相似文献

Dendroctonus mexicanus is polyphagous within the Pinus genus and has a wide geographical distribution in Mexico and Guatemala. We examined the pattern of genetic variation across the range of this species to explore its demographic history and its phylogeographic pattern. Analysis of the mtDNA sequences of 173 individuals from 25 Mexican populations allowed to us identify 53 geographically structured haplotypes. High haplotype and low nucleotide diversities and Tajima’s D indicate that D. mexicanus experienced rapid population expansion during its dispersal across mountain systems within its current range. The nested clade phylogeographic analysis indicates that the phylogeographic pattern of D. mexicanus is explained by continuous dispersion among lineages from the Sierra Madre Occidental, the Sierra Madre Oriental and the Trans-Mexican Volcanic Belt. However, we also observed isolation events among haplotypes from the Cofre de Perote/Trans-Mexican Volcanic Belt/Sierra Madre Oriental and the Trans-Mexican Volcanic Belt/Sierra Madre del Sur, which is consistent with the present conformation of mountain systems in Mexico and the emergence of geographical barriers during the Pleistocene.  相似文献

红脂大小蠹 Dendroctonus valens LeConte (Red turpentine beetle,RTB)是近年来在中国爆发的入侵生物,在我国主要以老熟幼虫在油松伐桩和立木的根部越冬.室内测定昆虫的过冷却点(SCP)和短时间致死低温(LLT)是评价昆虫耐寒能力的重要方法.实验结果显示,红脂大小蠹越冬幼虫的平均过冷却点为一11.98±2.55℃,是一种耐冰冻的昆虫.红脂大小蠹的过冷却点在不同地理分布区的种群问有明显差异,老熟幼虫的过冷却点明显低于低龄幼虫,在越冬前和越冬后的幼虫问没有明显差异.红脂大小蠹幼虫在冬季至少町以忍受-23.5℃的大气温度安全越冬.从2001年开始引入我国的云杉大小蠹的捕食者大唼蜡甲(Rhizophagus grandis cyll.)幼虫的过冷却点为-18.05±2.76℃,低于红脂大小蠹所有虫态的过冷却点,说明比红脂大小蠹有更强的耐寒能力.  相似文献

为揭示华山松大小蠹和伴生蓝变真菌引起秦岭华山松枯萎的机制,选择秦岭北坡沣峪林场境35年树龄的健康华山松Pinus armandi为研究对象,对接种华山松大小蠹Dendroctonus armandi及与其伴生的蓝变真菌Ceratocystis polonica引起的寄主树木木质部形态变化进行了解剖观察。结果表明:接种致病性蓝变真菌C. polonica 1周后的4株华山松 的木质部组织内,蓝变区域显著增加,4~6周后蓝变区域不再增加; 而在接种无菌琼脂的2株对照华山松的木质部组织内,没有检测到蓝变区域。研究结果提示蓝变真菌C. polonica是致死秦岭华山松的重要病原菌,该伴生菌随华山松大小蠹入侵健康寄主华山松木质部组织,在木质部定居并分解木质部,堵塞树脂道,致使寄主华山松树脂代谢和水分代谢紊乱。该研究结果表明,虽然华山松大小蠹长期以来被认为是致死华山松的毁灭性小蠹虫,但是其共生蓝变真菌C. polonica对成熟华山松的致害作用不应该被忽视。  相似文献

对外来松树害虫红脂大小蠹Dendroctonus valens LeConte的信息化学物质进行了研究。通过GC-MS测定,鉴定出红脂大小蠹后肠挥发性物质中除了含有松树挥发性物质α-蒎烯、β-蒎烯、3-蒈烯、月桂烯和柠檬烯外,还含有马鞭草烯醇和马鞭草烯酮;利用触角电位技术,对马鞭草烯醇、马鞭草烯酮以及在林间引诱效果最好的3-(+)-蒈烯进行了触角电生理测试;利用Y-型双向选择嗅觉仪对这些物质进行了室内趋向实验。实验结果表明: 1 μg的马鞭草烯醇、马鞭草烯酮和3-(+)-蒈烯引起雌雄触角的电位反应分别达416 μV/470 μV、597 μV/630 μV和926 μV/1 099 μV。浓度为1 μL/mL的马鞭草烯酮引起红脂大小蠹的正趋向反应,而在100 μL/mL浓度下引起红脂大小蠹的负趋向反应;在1 μL/mL的浓度下,3-(+ )-蒈烯引起了红脂大小蠹正趋向反应,而马鞭草烯醇则对红脂大小蠹具有驱避作用,说明这些物质在红脂大小蠹搜寻寄主和调节虫口密度方面起着重要作用。  相似文献

An approach to ensure effective pest biocontrol would be to select native isolates of biological control agents (BCAs). A survey to isolate entomopathogenic fungi (EPF) from a hazelnut growing area has been carried out. EPF were recovered from 133 of the 295 soil samples. The main species isolated were Metarhizium anisopliae sensu lato (36%) and Beauveria bassiana sensu lato (42.8%). With regard to controlling abiotic factors, altitude had an effect on the distribution of B. bassiana, but not on M. anisopliae. Cropping system did not have an effect on the occurrence of these EPF, while pH appeared as a predictive variable for both. In addition, we tested the virulence of six of these isolates: three M. anisopliae and three of B. bassiana against larvae of Curculio nucum L. The highest larval mortality (reaching 80%) was due to M. anisopliae (strain 34) when applied in simultaneous combination with four entomopathogenic nematode species: Steinernema carpocapsae (strain B14), Steinernema feltiae (strain D114), Steinernema. sp. (strain D122) and Heterorhabditis bacteriophora (strain DG46). The effect of nematodes was greater and no antagonistic or synergistic effects were observed.  相似文献

The spruce bark beetle, Ips typographus, is a recent new introduction to the Qilian Mountains of China. An outbreak of these beetles has infested over 0.03 million hectares of spruce forests in this area. Although primary attraction to volatiles has been clearly demonstrated for I. typographus, the existence and role of attraction to insect‐produced pheromones have been widely debated. Currently, commercial lures for I. typographus include only the volatiles ipsdienol, cis‐verbenol, trans‐verbenol, 2‐methyl‐3‐buten‐2‐ol and 2‐phenylethanol in Europe. Several potential pheromone candidates have been identified for I. typographus. Our GC–MS and GC–FID analyses volatiles from hindgut extracts of I. typographus in different attack phases demonstrated that the 2‐methyl‐3‐buten‐2‐ol, ipsdienol, cis‐verbenol and trans‐verbenol as major hindgut components, and ipsenol, 2‐phenylethanol, trans‐ myrtenol and verbenone as minor components. We tested various combinations of semiochemical candidates, to determine an optimal blend. Our results suggest that addition of 2‐methyl‐3‐buten‐2‐ol to either ipsenol alone, or to blends of ipsenol and other semiochemical candidates, significantly enhanced attraction of I. typographus. Therefore, a simple lure consisting of ipsenol and 2‐methyl‐3‐buten‐2‐ol would be an optimal blend of I. typographus in the Qilian Mountains, China. We conclude that this optimal semiochemical blend may provide an effective biological pest control method for use in forest ecosystem against I. typographus.  相似文献
