A triazole-resistant mutant, cysK1358, showing novel properties was isolated. Biochemical analysis of this strain suggests a regulatory character of the cysK1358 mutation. The cysK1358 mutation has a pleiotropic effect: the expression of the other gene, cysA, is also altered.  相似文献

Cloning of cysB mutant alleles of S. typhimurium   总被引:1,自引:0,他引:1  
Two cysB mutant alleles of S. typhimurium have been cloned onto pBR vectors. The product of the constitutive cysBc 1352 allele present on the plasmid was found to fulfill regulatory functions: as an activator of the cysteine regulon and as an autorepressor. CysB70 auxotrophic mutation impairs both regulatory functions cysB protein. Transfer of the clones cysBc 1352 allele from E. coli to S. typhimurium and from S. typhimurium to E. coli and biochemical analysis of transformants suggest involvement of a restriction-modification system in the constitutive expression of the cysteine regulon.  相似文献

Summary In Salmonella typhimurium, streptomycin resistance can occur by mutation at the strA or the strB mutants have altered ribosomes which are refractory to the drug in cell-free amino acid incorporation systems, and in 3H-dihydrostreptomycin binding studies. StrB mutants, unlike the strA mutants, are resistant to several aminoglycoside antibiotics and resistance is not due to a mutational change in the cell's protein synthetic machinery. Spectinomycin resistant mutants of S. typhimurium also fall into two classes, only one of which is ribosomal in mechanism. The spcA and spcB loci are closely linked to strA, aroC, and argG on the S. typhimurium linkage map.  相似文献

Salmonella typhimurium SR-11 is extremely virulent at a dose as low as 105 colony forming units (cfu) when administered perorally to BALB/c mice. Utilizing mini-transposon mutagenesis, a mutant of S. typhimurium SR-11 was isolated that was unable to utilize oleate and citrate as carbon sources. This mutant, designated S. typhimurium SR-11 Fad (Fatty acid), was found to utilize sugars under cya/crp control as sole carbon sources, suggesting that the mutation is not in either of these genes. In addition, SR-11 Fad utilized pyruvate and succinate, but was unable to utilize either acetate or isocitrate as sole carbon source. In contrast to SR-11, SR-11 Fad was found to be avirulent, i.e. BALB/c mice were completely healthy after oral infection with 109 S. typhimurium SR-11 Fad cells. Moreover, 21 days after SR-11 Fad infection, BALB/c mice were found to be protected against an oral challenge with 109 cells of S. typhimurium SR-11.  相似文献

The study of Salmonella virulent strains has revealed that the characteristic feature of such strains is the presence of plasmids with a molecular weight of 90.2-91.5 kb for S. typhimurium and 77.2-78.5 kb for S. dublin. From Salmonella strains harboring only a single plasmid, variants with no plasmid at all have been obtained. These variants possess lower virulence for mice infected through enteral and intraperitoneal routes; besides, they lose their capacity for penetration into epithelial cells of HeLa line. S. typhimurium and S. dublin have shown decreased multiplication rate in vivo in comparison with the parent strains, while the multiplication rates in vitro were similar. These results suggest that the products of plasmid genes are either responsible for the virulent properties of salmonellae, or they have regulatory functions, thus controlling the work of chromosomal genes.  相似文献

Hepatitis A virus (HAV) has an immunodominant neutralization antigenic site. By using a panel of monoclonal antibodies targeted against the HAV neutralization antigenic site, it was shown that three epitopes within this site are present on 14S subunits (pentamers of the structural unit). In contrast, two other epitopes within this site are formed upon assembly of 14S subunits into capsids. Thus, the epitopes recognized by these two monoclonal antibodies are formed either by a conformational change in the antigenic site or by the juxtaposition of epitope fragments present on different 14S subunits during assembly of 14S into 70S particles. Both 14S and 70S particles elicited HAV-neutralizing antibodies in mice; thus, these particles may be useful for HAV vaccine development.  相似文献

Gamma-irradiation of S. typhimurium cells up to a dose of 500 krad significantly reduces their toxicity. However, the antigenicity of these cells is not altered, which suggests that these cells could be used as a vaccine. The protection offered by the irradiated cells is comparable to that of formalin-treated cells. The radio-vaccine, however, offers an additional advantage of significant detoxification of the endotoxin, thereby minimizing side effects. The lipopolysaccharide extracted from the irradiated S. typhimurium cells offered cross-protection against other Salmonella species tested.  相似文献

目的:比较猪链球菌2型强毒株S.suis 05ZY和弱毒株S.suis 1940毒力相关基因转录水平的差异,为进一步研究强毒株S.suis 05ZY毒力增强的原因提供实验基础。方法:分别提取S.suis 05ZY和S.suis 1940的RNA,反转录成cDNA并纯化,用Cy5或Cy3标记,与猪链球菌全基因组DNA芯片进行杂交,扫描芯片进行数据分析,比较二者在转录水平上的差异基因。结果:编码溶血素、精氨酸氨基肽酶的基因分别上调4.4和6.0倍,参与荚膜多糖合成的相关基因cps2H、cps2I、cps2J和一些可能的毒力相关基因ofs、dpr、SSU050196、SSU050272、SSU051408-1409均发生转录水平的上调。结论:溶血素、荚膜多糖、精氨酸氨基肽酶及一些可能的毒力因子在转录水平的上调很可能与S.suis 05ZY的毒力增强有关。  相似文献
