The strength of a slippery slope argument is a matter of some dispute. Some see it as a reasonable argument pointing out what probably or inevitably follows from adopting some practice, others see it as essentially a fallacious argument. However, there seems to be a tendency emerging to say that in many cases, the argument is not actually fallacious, although it may be unsubstantiated. I shall not try to settle this general discussion, but merely seek to assess the strength of the slippery slope argument applied to human gene therapy. The structure of my argument will be the following. First, I shall distinguish between three different versions of the slippery slope argument; two logical versions and an empirical one. Next, I will address human gene therapy in terms of each of the three versions, partly relying on slippery slope arguments against this practice that have already surfaced in the literature. I shall argue that neither version pulls through. The logical versions fail primarily because relevant distinctions can be made between different uses of gene therapy, contrary to what the proponents of the arguments claim. The empirical version fails because there seems to be no evidence supporting the claim that we shall in fact slide down the slope if we engage in gene therapy, and because if we accepted the conclusion that we should not allow gene therapy on the basis of the empirical argument, we should have to make very far-reaching and undesirable modifications in health care in general, in order to be consistent. Research on human stem cells and embryos creates ethical issues. Here I discuss ten frequently used arguments against research and point out their weaknesses. These arguments include the possessed potentiality of the embryo per se and, in contrast to other cell systems, the "slippery slope" argument, the right of disposal of parents, totipotency versus pluripotency, the burden of proof for research, natural versus artificial, and three arguments based on the precaution principle (the open biological questions, uncertainty regarding clinically applicable therapies, and the problem solving rule). I finally suggest a different answer to the ethical questions concerning research on human embryos and embryonic stem cells, which takes into consideration their biological context.  相似文献

In mammals, silencing of one of the two X chromosomes is necessary to achieve dosage compensation. The non coding RNA Xist triggers X inactivation. Gene silencing by Xist is only possible in certain developmental contexts that only exist in cells of the early embryo and specific hematopoietic progenitors. Critical silencing factors may only be present in these contexts giving an explanation of why Xist is not operative outside these contexts. It has been demonstrated that Xist is functional in tumor cells, where SATB1 was identified as the first silencing factor for Xist mediated chromosome silencing.  相似文献

A basic consideration in research on human embryos is the controversy about when the embryo acquires moral status. The author refutes the contention that segmentation is the determinant of moral status. She notes that segmentation, as a stage in embryonic development, does not coincide with the development of "irreversible individuality" upon which the segmentation argument depends. Dawson also finds a lack of clarity in the meaning of "individuality." These problems, she maintains, prevent segmentation from being morally important and render the proposed 14-day limit on embryo research unnecessary. Dawson concludes that to introduce a time restriction on embryo research is premature because it is based on an inadequate philosophical argument.  相似文献

Using a cryo-scanning electron microscope, we studied microstructure of the slippery zone in nine Nepenthes taxa. For N. fusca, N. macrophylla, N. mirabilis, N. ventricosa, N. dicksoniana, and N. veitchii, it was examined here for the first time. Three types of the slippery zone were distinguished among the studied taxa: (1) with well-developed crystalline wax coverage, (2) with greatly reduced wax coverage, and (3) without wax crystals. These data were combined with morphometrical measurements of the two pitcher zones primarily relevant to prey catching and retaining: the slippery zone and the peristome. In species with fully developed wax coverage, the slippery zone was longer and the peristome was narrower compared to those with reduced or lacking crystalline wax. We found statistically significant negative correlation between the relative length of the slippery zone and the relative width of the peristome. Based on the analysis of the relationship between the microstructure of the slippery zone and pitcher macromorphology, two main types of pitchers in Nepenthes are proposed: (a) traps based predominantly on the waxy slippery zone and (b) peristome-based traps.  相似文献

In annelids, molluscs, echiurans and sipunculids the establishment of the dorsal-ventral axis of the embryo is associated with D quadrant specification during embryogenesis. This specification occurs in two ways in these phyla. One mechanism specifies the D quadrant via the shunting of a set of cytoplasmic determinants located at the vegetal pole of the egg to one blastomere of the four cell stage embryo. In this case, at the first two cleavages of embryogenesis there is an unequal distribution of cytoplasm, generating one macromere which is larger than the others at the four cell stage. The D quadrant can also be specified by a contact mediated inductive interaction between one of the macromeres at the vegetal pole with micromeres at the animal pole of the embryo. This mechanism operates at a later stage of development than the cytoplasmic localization mechanism and is associated with a pattern of cleavage in which the first two cleavages are equal. An analysis of the phylogenetic relationships within these phyla indicates that the taxa which determine the D quadrant at an early cleavage stage by cytoplasmic localization tend to be derived and lack a larval stage or have larvae with adult characters. Those taxa where the D quadrant is specified by induction include the ancestral groups although some derived groups also use this mechanism. The pulmonate mollusc Lymnaea uses an inductive mechanism for specifying the D quadrant. In these embryos each of the four vegetal macromeres has the potential of becoming the D macromere; however under normal circumstances one of the two vegetal crossfurrow macromeres almost invariably becomes the D quadrant. Experiments are described here in which the size of one of the blastomeres of the four cell stage Lymnaea embryo is increased; this macromere invariably becomes the D quadrant. These experiments suggest that developmental change in relative blastomere size during the first two cleavages in spiralian embryos that normally cleave equally may have provided a route that has led to the establishment of the cytoplasmic localization mechanism of D quadrant formation.  相似文献

Many rodents hoard seeds as a means of ensuring food availability during scarcity. Siberian chipmunks (Tamias sibiricus) feed on acorns of white oak Quercus mongolica and hoard them for future use. Such caches may deteriorate due to the early germination of the acorns in autumn, which can be inhibited by radicle pruning or embryo excision. Siberian chipmunks are found to frequently prune the radicles of germinated acorns of Q. mongolica; however, whether this behavior is a kind of cache management tactic remains unclear. Here, we performed semi-natural enclosure and field experiments to test the role of radicle pruning in cache management of T. sibiricus. We found that T. sibiricus preferred non-germinated acorns and tended to prune radicles of germinating acorns when scatter-hoarding them, but did not perform the behavior of embryo excision by squirrels. Both semi-natural enclosure and field experiments showed that T. sibiricus not only pruned radicles repeatedly but removed radicles varied in length from acorns, which significantly postponed acorn germination and radicle growth. These observations suggest that radicle pruning would be an evolutionary tactic for cache management of Siberian chipmunks.  相似文献

Over the past few years, several proposals aimed at procuring human pluripotent (embryonic‐like) stem cells without involving the destruction of a human embryo have been proposed and widely discussed. This article focuses on a basic aspect of the debate, namely the plausibility of one or more of these new proposals being able to meet the ethical requirements that those who regard the human embryo as sacred have tried to impose on stem cells research in the last ten years. The thesis of the article is that focusing the discussion only on the sources of stem cells has prevented a full understanding of the foundation, meaning and scope of these ethical requirements. To substantiate this thesis, the article takes into consideration two issues: the first has to do with the potential of the cells obtained through some of the new approaches (iPS included), the second (and decisive) with the argument of the ‘indirect complicity’, applied to the use of ‘contaminated’ knowledge.  相似文献

The intensity of interspecific territorial defense should be based upon the degree of competitive overlap. We tested this relationship in two territorial Caribbean damselfish (dusky, Stegastes adustus, and longfin, S. diencaeous) with intruders being the bluehead wrasse (Thalassoma bifascatus) and the slippery dick wrasse (Halichoeres bivittatus). Based on food habits, the slippery dick and the bluehead wrasse should have the same degree of competitive overlap to the two damselfish species. We also predicted that the larger slippery dick wrasse intruder would receive more aggression than the smaller bluehead wrasse intruder. Neither damselfish species distinguished between the two wrasses suggesting that they were ecologically equivalent. We also tested size differences within both species of wrasse and found that size had no influence on the aggression in the dusky damselfish. The longfin also did not show a size preference for the bluehead but did prefer to attack the larger slippery dick wrasse. In spite of the similarities between the dusky and the longfin damselfish and the similar food habits of the bluehead and the slippery dick wrasse, our results suggest that using intensity of interspecific territorial defense alone may not be an adequate measure of competitive overlap.  相似文献

Germinability of isolated embryos from developing fruits of Acer platanoides was high at the earliest developmental stage assessed (90 dpa), but fell subsequently and at seed maturity was very low. These observations showed an inverse correlation with changes in endogenous free abscisic acid (ABA) levels in the embryo, which were low during early ontogeny, but reached maximum levels late in development (150–160 dpa). These observations suggest the possibility that dormancy may be induced during development as a result of ABA accumulation in the embryo, an argument strengthened by the obvious inhibitory effect of added ABA on the germinability of isolated embryos. The cotyledons appear to exert an inhibitory influence on embryo germinability that may result from their free ABA content although the embryonic axis itself possesses an innate dormancy that may reflect its own free ABA content. The increased germinability of isolated embryos resulting form added kinetin serves only to emphasise the complexity of the system and the dangers of simplistic interpretation.The correlation between germinability and ABA content is not complete, however, since much of the reduction in germinability had occurred before any appreciable increase in free ABA levels in the embryo was observed. Indeed the failure of the intact seed to respond to endogenous changes in embryonal ABA levels suggests that even though free ABA in the embryo may influence embryo germinability, it has little effect in the intact seed, where the presence of an intact testa may be a more important factor.The absence of a desiccation phase in the embryo during the late stages of development suggests that the large increases in endogenous free ABA did not cause dormancy by inhibiting water uptake, nor did they result from water stress in the embryonal tissues.  相似文献

Summary Sexual and aposporously apomictic plants of buffelgrass (Cenchrus ciliaris L.) form megaspore tetrads. In sexual plants the chalazal megaspore develops into a single Polygonum type embryo sac. In aposporous plants the megaspores degenerate, and one or more un-reduced nucellar cells form Panicum type embryo sacs. Apospory is conditioned by gene A; the dominant allele of gene B is epistatic to A and preserves sexual reproduction. We recently observed that heavy application of (NH4)2SO4 to the soil induced multiple embryo sacs in a sexual line. Therefore we tested the effect of salt stress on embryo sac formation in sexual and aposporous genotypes. One molar solutions of CaCl2, NaCl, (NH4)2SO4, NH4Cl, NaNO3, or Na2SO4 were applied to the soil of greenhouse plants every day or two starting at the archespore stage. Some of the pistils in salt-treated plants of sexual genotypes AaBb, aaBb, and aabb showed features not seen in untreated controls: (1) multiple Polygonum type embryo sacs in 1%–7% of pistils depending upon the salt; (2) embryo sacs without antipodals (0%–7%); (3) embryo sacs protruding through the micropyle (1%–16%). Some pistils of salt-treated obligately aposporous lines, but not controls, developed Polygonum type embryo sacs (4%–13%) and protruding embryo sacs (0%–6%). There was no ion specificity for induction of abnormal features. We postulate that salt stress suppresses the developmental priority of nucellar embryo sacs over megaspores in aposporous lines and of the chalazal megaspore over other megaspores in all lines. This may permit megaspores of aposporous plants to form reduced Polygonum type gametophytes, and permit more than one megaspore to form reduced embryo sacs in all lines. Abstract

This article summarises the results of a research project that used a scenario about sex selection of embryos for social reasons as a basis for discussion groups with lay people. The aim of the research was to examine the processes by which non-professionals make ethical evaluations in relation to a contested area in medical genetics. We note in particular the role played in the discussions by expressions of instinct; making distinctions; rational argument; reference to principles; use of personal experience; analogies and examples; slippery slope arguments and meta-reflections. Abstract

Seed formation involves not only the embryo and endosperm development, but also the formation of a series of either ephemeral or non-ephemeral structures. In this article, we study several of those structures in Cytisus multiflorus and Cytisus striatus. The endosperm development is first nuclear and later cellular, except for the chalazal area, whose development is always nuclear. It generates, in the early developmental stages, a sac-like haustorium. As the seed develops, two structures seem to be closely related to nutrient mobilization to the embryo sac: on the one hand, a group of cells and a channel, located in the chalazal area and closely related between them and to the endosperm haustorium, which could be interpreted as a hypostase and on the other hand, an endothelium, derived from the inner integument, which later degenerates leaving no trace in the mature seed. All of these structures would be associated with the directionality of assimilates from ovule tissues to embryo sac. In mature seed and surrounding the embryo appears a unicellular layer of cells rich in proteins (aleurone layer), which is the origin of the outermost layer of the cellular endosperm. The seed coat is made up only of the outer integument.  相似文献

Abstract

This paper describes classical experiments of experimental embryology which may be repeated at class level using the early embryos of Xenopus laevis. The experiments demonstrate the effects of lithium on the early development of the nervous system, and the use of hypertonic salt solutions to produce exogastrulation. Both experiments involve the removal of the jelly coats around the embryo, and for this purpose a rapid chemical method using a cysteine hydrochloride/papain solution is described. Both experiments are theoretically important: the effects of lithium are used as evidence for the existence of an anterior-posterior gradient in the early embryo, while exogastrulation provides a convenient system for interfering with the primary inductive effect of the chordo-mesoderm, necessary for ectodermal neuralization.

Maintaining genetic diversity is a crucial goal of intensive management of threatened species, particularly for those populations that act as sources for translocation or re‐introduction programmes. Most captive genetic management is based on pedigrees and a neutral theory of inheritance, an assumption that may be violated by selective forces operating in captivity. Here, we explore the conservation consequences of early viability selection: differential offspring survival that occurs prior to management or research observations, such as embryo deaths in utero. If early viability selection produces genotypic deviations from Mendelian predictions, it may undermine management strategies intended to minimize inbreeding and maintain genetic diversity. We use empirical examples to demonstrate that straightforward approaches, such as comparing litter sizes of inbred vs. noninbred breeding pairs, can be used to test whether early viability selection likely impacts estimates of inbreeding depression. We also show that comparing multilocus genotype data to pedigree predictions can reveal whether early viability selection drives systematic biases in genetic diversity, patterns that would not be detected using pedigree‐based statistics alone. More sophisticated analysis combining genomewide molecular data with pedigree information will enable conservation scientists to test whether early viability selection drives deviations from neutrality across wide stretches of the genome, revealing whether this form of selection biases the pedigree‐based statistics and inference upon which intensive management is based.  相似文献

Conclusion It may be seen, then, that if one was prepared to accept the existence of insulating sheaths on the nerves, all the arguments raised against the proposed identification of the nervous and electrical fluids, except one, could be answered satisfactorily. The single exception involved the question of how an electrical disturbance in the brain could be confined to a single nerve, and, as was indicated earlier, it was scarcely fair to hold this sort of objection against the electrical theory alone. In that case, there remained no convincing argument to show why one should not accept the identification of the two fluids. On the other hand, of course, it remained an open question as to whether there was any convincing argument to show why one should accept the identification either. In cattle, the detection of very early endometrial responses is considered to be hampered by the presence of only a single embryo. Therefore, we have previously developed a model of multiple embryo transfer to circumvent this hindrance. In this work, we analysed embryo–maternal interactions in the bovine uterus on day 8 of development while comparing the presence of multiple v. single embryos using embryo transfer and artificial insemination, respectively. Concentration of proteins (β-actin, NFkB, clusterin and immunoproteosome 20S β5i subunit–i20S), by western blot, and hexoses (glucose and fructose) were measured in paired samples of uterine fluid (UF) from the same animal with and without embryos in the uterus and were compared with UF obtained after artificial insemination. Prostaglandin (PG) F2α and PGE2 concentrations were also analysed in blood plasma. The four proteins analysed and hexoses were unaffected by the presence of one or more embryos in the uterus. However, blood PGF2α showed similar, significant increases with one or more embryos over cyclic animals; such changes were not observed in blood PGE2. Although multiple embryo transfer may appear to be non-physiological, we showed that the uterus, at the very early embryonic stages, does exhibit physiological reactions. Multiple embryo transfer can, therefore, be used for studies of very early embryo–maternal interactions in vivo in monotocous species.  相似文献
