We report the synthesis and study of surfactant-free poly(acrylic acid) (PAA) nanogels using hydroxypropylcellulose (HPC) as a template in aqueous HPC solutions at room temperature or above. Through the hydrogen bonding interaction of acrylic acid (AA) with hydroxypropylcellulose (HPC), AA absorbed on the HPC polymer chains and triggered the phase transition of HPC at a lower temperature, with increasing AA concentration, than the HPC intrinsic phase transition temperature 41 °C. As AA polymerized to form PAA, the much stronger interpolymer hydrogen bonding triggered the phase transition of HPC at a temperature around room temperature, causing HPC coil-global phase transition to collapse and form nanospheres at room temperature, PAA hydrogen-bonded HPC chains collapsed and formed nanogels chemically crosslinked by poly(ethylene glycol) diacrylate (PEGDA) or methylenebisacrylamide (BIS). The results showed that all the PAA nanogels demonstrated a narrow size distribution with diameters ranging from 60 nm to 600 nm.  相似文献

Novel polyelectrolyte hydrogels (coded as GA) based on cationic guar gum (CGG) and acrylic acid monomer by photoinitiated free-radical polymerization were synthesized with various feed compositions. Fourier transform infrared spectra (FTIR), scanning electron microscopy (SEM), and differential scanning calorimetry (DSC) confirmed that the formation of the polyelectrolyte hydrogels was attributed to the strong electrostatic interaction between cationic groups in CGG and anionic groups in poly (acrylic acid) (PAA). Swelling experiments provided important information on drug diffusion properties, which indicated the GA hydrogels were highly sensitive to pH environments. Potential applications of the hydrogels matrices in controlled drug delivery were also examined. The ketoprofen-loaded CGG/PAA matrices were prepared by hydrogels and directly compressed tablets, respectively. Release behavior of ketoprofen relied on the preparative methods of matrices, ratios of CGG/AA and pH environments. The release mechanism was studied by fitting experimental data to a model equation and calculating the corresponding parameters. The result showed that the kinetics of drug release from the hydrogels in pH 7.4 buffer solution was mainly non-Fickian diffusion. However, for tablets, the drug release in pH 7.4 buffer solution was mainly affected by polymer erosion. The pH of the dissolution medium appeared to have a strong effect on the drug transport mechanism. At more basic pH values, Case II transport was observed, indicating a drug release mechanism highly influenced by macromolecular chain relaxation. The ketoprofen release is also tested in the conditions chosen to simulate gastrointestinal tract conditions. The results implied that the GA hydrogels can be exploited as potential carriers for colon-specific drug delivery.  相似文献

Ultrafine cellulose fiber (diameter 200-400 nm) surfaces were grafted with polyacrylic acid (PAA) via either ceric ion initiated polymerization or methacrylation of cellulose with methacrylate chloride (MACl) and subsequent free-radical polymerization of acrylic acid. PAA grafts by ceric ion initiated polymerization increased with increasing reaction time (2-24 h), monomer (0.3-2.4 M), and initiator (1-10 mM) concentrations, and spanned a broad range from 5.5-850%. PAA grafts on the methacrylated cellulose fibers also increased with increasing molar ratios of MACl to cellulosic hydroxyl groups (MACl/OH, 2-6.4) and monomer acrylic acid (AA) to initiator potassium persulfate (KPS) ratios ([AA]/[KPS], 1.5-6), and were in a much narrower range between 12.8% and 29.4%. The adsorption of lipase (at 1 mg/ml lipase and pH 7) and the activity of adsorbed lipase (pH 8.5, 30 degrees C), in both cases decreased with increasing PAA grafts. The highest adsorption and activity of the lipase on the ceric ion initiated grafted fibers were 1.28 g/g PAA and 4.3 U/mg lipase, respectively, at the lowest grafting level of 5.5% PAA, whereas they were 0.33 g/g PAA and 7.1 U/mg lipase, respectively, at 12.8% PAA grafts on the methacrylated and grafted fibers. The properties of the grafted fibers and the absorption behavior and activity of lipase suggest that the PAA grafts are gel-like by ceric-initiated reaction and brush-like by methacrylation and polymerization. The adsorbed lipase on the ceric ion-initiated grafted surface possessed greatly improved organic solvent stability over the crude lipase. The adsorbed lipases exhibited 0.5 and 0.3 of the initial activity in the second and third assay cycles, respectively.  相似文献

Thermosensitive hydrogels are of great interest for in situ gelling drug delivery. The thermosensitive vehicle with a gelation temperature in a range of 30–36°C would be convenient to be injected as liquid and transform into gel after injection. To prepare novel hydrogels gelling near body temperature, the gelation temperature of poloxamer 407 (PX) were tailored by mixing PX with poly(acrylic acid) (PAA). The gelation behaviors of PX/PAA systems as well as the interaction mechanism were investigated by tube inversion, viscoelastic, shear viscosity, DSC, SEM, and FTIR studies. The gelation temperature of the plain PX solutions at high concentration of 18, 20, and 22% (w/w) gelled at temperature below 28°C, which is out of the suitable temperature range. Mixing PX with PAA to obtain 18 and 20% (w/w) PX with 1% (w/w) PAA increased the gelation temperature to the desired temperature range of 30–36°C. The intermolecular entanglements and hydrogen bonds between PX and PAA may be responsible for the modulation of the gelation features of PX. The mixtures behaved low viscosity liquid at room temperature with shear thinning behavior enabling their injectability and rapidly gelled at body temperature. The gel strength increased, while the pore size decreased with increasing PX concentration. Metronidazole, an antibiotic used for periodontitis, was incorporated into the matrices, and the drug did not hinder their gelling ability. The gels showed the sustained drug release characteristic. The thermosensitive PX/PAA hydrogel could be a promising injectable in situ gelling system for periodontal drug delivery.  相似文献

This study describes the formulation and physicochemical characterization of poly(acrylic acid) (PAA) organogels, designed as bioactive implants for improved treatment of infectious diseases of the oral cavity. Organogels were formulated containing a range of concentrations of PAA (3-10% w/w) and metronidazole (2 or 5% w/w, representing a model antimicrobial agent) in different nonaqueous solvents, namely, glycerol (Gly), polyethylene glycol (PEG 400), or propylene glycol (PG). Characterization of the organogels was performed using flow rheometry, compressional analysis, oscillatory rheometry, in vitro mucoadhesion, moisture uptake, and drug release, methods that provide information pertaining to the nonclinical and clinical use of these systems. Increasing the concentration of PAA significantly increased the consistency, compressibility, storage modulus, loss modulus, dynamic viscosity, mucoadhesion, and the rate of drug release. These observations may be accredited to enhanced molecular polymer entanglement. In addition, the choice of solvent directly affected the physicochemical parameters of the organogels, with noticeable differences observed between the three solvents examined. These differences were accredited to the nature of the interaction of PAA with each solvent and, importantly, the density of the resultant physical cross-links. Good correlation was observed between the viscoelastic properties and drug release, with the exception of glycerol-based formulations containing 5 and 10% w/w PAA. This disparity was due to excessive swelling during the dissolution analysis. Ideally, formulations should exhibit controlled drug release, high viscoelasticity, and mucoadhesion, but should flow under minimal stresses. Based on these criteria, PEG 400-based organogels composed of 5% or 10% w/w PAA exhibited suitable physicochemical properties and are suggested to be a potentially interesting strategy for use as bioactive implants designed for use in the oral cavity.  相似文献

Block copolymers containing stimuli-responsive segments provide important new opportunities for controlling the activity and aggregation properties of protein-polymer conjugates. We have prepared a RAFT block copolymer of a biotin-terminated poly(N-isopropylacrylamide) (PNIPAAm)-b-poly(acrylic acid) (PAA). The number-average molecular weight (M(n)) of the (PNIPAAm)-b-(PAA) copolymer was determined to be 17.4 kDa (M(w)/M(n) = 1.09). The PNIPAAm block had an M(n) of 9.5 kDa and the poly(acrylic acid) (PAA) block had an M(n) of 7.9 kDa. We conjugated this block copolymer to streptavidin (SA) via the terminal biotin on the PNIPAAm block. We found that the usual aggregation and phase separation of PNIPAAm-SA conjugates that follow the thermally induced collapse and dehydration of PNIPAAm (the lower critical solution temperature (LCST) of PNIPAAm is 32 degrees C in water) is prevented through the shielding action of the PAA block. In addition, we show that the cloud point and aggregation properties (as measured by loss in light transmission) of the [(PNIPAAm)-b-(PAA)]-SA conjugate also depended on pH. At pH 7.0 and at temperatures above the LCST, the block copolymer alone was found to form particles of ca. 60 nm in diameter, while the bioconjugate exhibited very little aggregation. At pH 5.5 and 20 degrees C, the copolymer alone was found to form large aggregates (ca. 218 nm), presumably driven by hydrogen bonding between the -COOH groups of PAA with other -COOH groups and also with the -CONH- groups of PNIPAAm. In comparison, the conjugate formed much smaller particles (ca. 27 nm) at these conditions. At pH 4.0, however, large particles were formed from the conjugate both above and below the LCST (ca. 700 and 540 nm, respectively). These results demonstrate that the aggregation properties of the block copolymer-SA conjugate are very different from those of the free block copolymer, and that the outer-oriented hydrophilic block of PAA shields the intermolecular aggregation of the block copolymer-SA bioconjugate at pH values where the -COOH groups of PAA are significantly ionized.  相似文献

The graft copolymerization of cassava starch with acrylic acid was investigated using a free radical initiator system (Fe(2+)/H(2)O(2) redox system) in water. A comprehensive understanding of the important variables and their interaction has been obtained by applying an experimental design method. In this approach, two ('high' and 'low') values of selected variables are considered. Important result parameters are add-on and the grafting efficiency. Out of eight reaction variables, it was found that only temperature, starch concentration and the starch to monomer ratio have a pronounced influence on these response parameters. Moderate reaction temperature (40°C) and high starch concentration (10%) give relatively good results of add-on and grafting efficiency. A low starch to monomer ratio favors add-on but decreases grafting efficiency. These findings can be used to optimize the production of cassava starch-acrylate copolymers and to gain insight in the process-product property interactions, for various applications.  相似文献

A new family of block polyelectrolyte networks containing cross-linked poly(acrylic acid) (PAA) and poly(ethylene oxide) (PEO) was synthesized by copolymerization of acrylic acid and bisacrylated PEO (10 kDa). Two materials with different PEO/PAA ratios were compared with a weakly cross-linked PAA homopolymer network. The networks bound a cationic protein, cytochrome C, due to the polyion coupling, leading to the network contraction. After binding the protein the block polyelectrolyte networks were more porous compared to a homopolymer network, facilitating protein absorption within the gel. The protein was released by adding Ca2+ ions or a polycation. Ca2+ ions migrated within the gels and reacted with PAA chains, thus displacing the protein. The polycation transfer into hydrogels, as a result of polyion substitution reactions, was inhibited by the excess of PEO chains in the block polyelectrolyte networks. Overall, these findings advance development of functional polyelectrolyte networks for immobilization and controlled release of proteins.  相似文献

Purified flax waste was obtained from flax processing wastes via subjecting the latter to alkali treatment followed by peracetic acid bleaching. The so obtained purified flax wastes were chemically modified via reacting with propylene oxide in alkaline medium. The resultant hydroxypropyl cellulose (HPC) was incorporated in a polymerization medium containing acrylic acid and potassium bromate/thiourea mixture as initiation system. The polymerization reaction was monitored by determining the total conversion percent and the rheological properties of the resultant polyacrylic acid–hydroxypropyl cellulose composite [poly(AA)–HPC]. Results obtained indicate that the optimum conditions of polymerization process were: 12 mmole KBrO3, 4 mmole thiourea and 100 g acrylic acid/100 g HPC at 50 °C for 2 h using a material to liquor ratio of 1:5.  相似文献

To improve the water-fertilizer utilization ratio and mitigate the environmental contamination, an eco-friendly superabsorbent polymer (SPA), modified sugarcane bagasse/poly (acrylic acid) embedding phosphate rock (MSB/PAA/PHR), was prepared. Ammonia, phosphate rock (PHR) and KOH were admixed in the presence of acrylic acid to provide nitrogen (N), phosphorus (P) and potassium (K) nutrients, respectively. Impacts on water absorption capacity of the superabsorbent polymer (SAP) were investigated. The maximum swelling capacity in distilled water and 0.9wt.% (weight percent) NaCl solution reached 414gg(-1) and 55gg(-1) (water/prepared SAP), respectively. The available NPK contents of the combination system were 15.13mgg(-1), 6.93mgg(-1) and 52.05mgg(-1), respectively. Moreover, the release behaviors of NPK in the MSB/PAA/PHR were also studied. The results showed that the MSB/PAA/PHR has outstanding sustained-release plant nutrients property.  相似文献

The interaction of fractionated poly(acrylic acid)s (PAA) with bovine serum albumin (BSA) has been studied by measuring the hydrolysis rate of p-nitrophenyl acetate catalysed by BSA in the presence of PAA. The binding of PAA with BSA, which prohibits the catalytic action of BSA, increases with increasing molecular weight of PAA. The change in the electronic spectra of BSA-PAA solutions supports this molecular weight dependence. Circular dichroism of BSA shows that the binding of PAA does not induce any conformational change in BSA.  相似文献

Many studies have clarified that poly(gamma-glutamic acid) (PGA) increases the solubility of Ca(2+), suggesting that PGA enhances calcium absorption in small intestine. However, there has been no report on the specific interaction between PGA and Ca(2+) in water. We studied the aqueous solution properties of PGA calcium salt (PGA-Ca complex). The chelating ability and binding strength of PGA for Ca(2+) were evaluated. PGA-Ca complex was soluble in water in contrast with the insolubility of poly(acrylic acid) (PAA) calcium salt and the chelating ability of PGA for Ca(2+) was almost the same than that of PAA. The globular conformation of PGA-Ca complex in water was estimated by SEC and viscosity measurements. The chelation of PGA for Ca(2+) was examined by 1H NMR. The present study showing the characteristics of PGA-Ca complex will provide useful information of the calcium absorption by PGA in vivo.  相似文献

Silicon (Si) composite electrodes are developed with increased cycle lifetimes and reliability through dynamic ionic bonding between active Si nanoparticles and a polymer binder. Amine groups are covalently attached to Si nanoparticles via surface functionalization. Si composite electrodes are fabricated by combining the Si nanoparticles with a poly(acrylic acid) (PAA) binder. The formation of ionic bonds between amine groups on Si particles and carboxylic acid groups on the PAA binder is characterized by X‐ray photoelectron spectroscopy and Raman spectroscopy. Si composite anodes with ionic bonding demonstrate long term cycling stability with capacity retention of 80% at 400 cycles at a current density of 2.1 A g?1 and good rate capability. The dynamic ionic bonds effectively mitigate the deterioration of electrical interfaces in the composite anodes as suggested by stable impedance over 300 cycles.  相似文献

The miscibility and mechanical properties of poly vinyl alcohol (PVA) and poly acrylic acid (PAA)-composited membranes were studied with molecular simulation. The Flory–Huggins parameters (δ) were calculated to prove the good miscibility of PVA and PAA. The radial distribution functions of hydroxyl and carboxyl atoms and the average number of H-bonds were observed to indicate the degree of physical cross-linking between PVA and PAA. The influences of intermolecular physical cross-linking on the glass transition temperature and mechanical properties were estimated. The results revealed that the PVA/PAA membrane with a composition of 2:3 has the best plastic properties, which exhibits a good application value. All of the simulated results showed good agreement with the experimental data. It indicates that the method presented in this work has a promising application prospect.  相似文献

It was aim of the study to investigate the in vivo potential of a novel insulin-thiomer complex nanoparticulate delivery system. Insulin loaded nanoparticles were obtained by the formation of hydrogen bonds between poly(vinyl pyrrolidone) (PVP) and poly(acrylic acid)-cysteine (PAA-Cys) or poly(acrylic acid) (PAA), respectively, in the presence of insulin. Dissolution behavior of insulin from tablets as well as nanoparticulate suspensions was evaluated in vitro. Serum insulin concentrations and reduction of blood sugar values were determined after oral administration of nanoparticles formulated as enteric coated tablets and suspensions. Results displayed a low serum insulin concentration and pharmacological efficacy in terms of blood sugar reduction after oral administration of enteric coated tablets. On the contrary, nanoparticulate suspensions led to significant serum insulin concentrations. Furthermore a 2.3-fold improvement of the AUC of insulin could be achieved due to the use of thiolated PAA instead of unmodified PAA. In addition, a blood sugar reduction of 22% was observed. Results demonstrate that this novel complex nanoparticulate formulation is an encouraging new attempt toward the noninvasive delivery of peptide drugs.  相似文献

The interaction of α-chymotrypsin with poly(acrylic acid)s (PAA) having different stereoregularities and molecular weights has been studied through the effects of α-chymotrypsin on enzymic hydrolysis of p-nitrophenyl acetate (NpOAc). The results show that isotactic PAA inhibits the hydrolysis more strongly than do atactic and syndiotactic PAAs. The inhibition constant or the dissociation constant of the reactio-inhibiting PAA α-chymotrypsin complex decrease with increasing molecular weight of PAA.  相似文献

(Hydroxypropyl)cellulose (HPC)/vinyl polymer networks were synthesized in film form from liquid-crystalline solutions of HPC in a mixed solvent of methacrylate monomer/methanol/water (2:1:2 in weight) containing cross-linking agents, via photopolymerization of the methacrylate monomer. Di(ethylene glycol) monomethyl ether methacrylate (DEGMEM) or 2-hydroxypropyl methacrylate (HPMA) was used as the polymerizing monomer, and tetra(ethylene glycol) diacrylate and glutaraldehyde were the cross-linkers for the monomers and HPC, respectively. The polymer composite films, HPC/PDEGMEM and HPC/PHPMA, prepared at ca. 60-70 wt % concentrations of HPC in the starting solutions, were iridescently colored due to the selective light reflection, originating from the cholesteric helical arrangement carried over successively into the network system. When the cholesteric films were immersed and swollen in water containing an inorganic neutral salt, their coloration and optical turbidity varied according to a strength of 'chaotropicity' of the impregnant ions. This ionic effect may be interpreted as essentially identical with that found formerly in the coexistent salt-sort dependence of the cholesteric pitch and lower critical solution temperature for HPC aqueous solutions. It is also demonstrated that visual appearance of the swollen networks can be changed by application of an electric potential of practical magnitude between both edges of the samples of rectangular shape.  相似文献

The objectives of this study were to prepare push–pull osmotic tablets (PPOT) of felodipine using an interpolymer complex of chitosan (CS) and poly(acrylic acid) (PAA) as an osmopolymer, and to study the mechanisms of drug release from these tablets. The interpolymer complexes were prepared with different weight ratios of CS to PAA. Preparation of PPOT involved the fabrication of bilayered tablets with the drug layer, containing felodipine, polyethylene oxide, and the polymeric expansion layer, containing the CS–PAA complex. The effects of polymer ratios, type of plasticizers, and compression forces on release characteristics were investigated. It was found that drug release from PPOT exhibited zero-order kinetics and could be prolonged up to 12 or 24 h depending on the plasticizer used. PPOT using dibutyl sebacate showed a longer lag time and slower drug release than that using polyethylene glycol 400. In the case of polyethylene glycol 400, an increase in the CS proportion resulted in an increase in the drug release rate. The compression force had no effect on drug release from PPOT. Drug release was controlled by two consecutive mechanisms: an osmotic pump effect resulting in the extrusion of the drug layer from the tablet and subsequent erosion and dissolution of the extruded drug layer in the dissolution medium. The mathematical model (zero-order) related to extrusion and erosion rates for describing the mechanism of drug release showed a good correlation between predicted and observed values.  相似文献
