Yeasts and filamentous fungi carried by the gynes of leaf-cutting ants   总被引:1,自引:1,他引:0  
Insect-associated microbes exhibit a wide range of interactions with their hosts. One example of such interactions is the insect-driven dispersal of microorganisms, which plays an essential role in the ecology of several microbes. To study dispersal of microorganisms by leaf-cutting ants (Formicidae: Attini), we applied culture-dependent methods to identify the filamentous fungi and yeasts found in two different body parts of leaf-cutting ant gynes: the exoskeleton and the infrabuccal pocket. The gynes use the latter structure to store a pellet of the ants’ symbiotic fungus during nest founding. Many filamentous fungi (n = 142) and yeasts (n = 19) were isolated from the gynes’ exoskeleton. In contrast, only seven filamentous fungi and three yeasts isolates were recovered from the infrabuccal pellets, suggesting an efficient mechanism utilized by the gynes to prevent contamination of the symbiotic fungus inoculum. The genus Cladosporium prevailed (78%) among filamentous fungi whereas Aureobasidium, Candida and Cryptococcus prevailed among yeasts associated with gynes. Interestingly, Escovopsis, a specialized fungal pathogen of the leaf-cutting ant-fungus symbiosis, was not isolated from the body parts or from infrabuccal pellets of any gynes sampled. Our results suggest that gynes of the leaf-cutter ants Atta laevigata and A. capiguara do not vertically transmit any particular species of yeasts or filamentous fungi during the foundation of a new nest. Instead, fungi found in association with gynes have a cosmopolitan distribution, suggesting they are probably acquired from the environment and passively dispersed during nest foundation. The possible role of these fungi for the attine ant–microbial symbiosis is discussed.  相似文献

Summary The phosphorylation activities of glucose and levoglucosan (1,6-anhydro-β-d-glucopyranose, LG) were studied in 26 types of LG-assimilating microorganisms isolated from four types of soil in China. The results showed that activities of LG kinase production in most of the yeasts were lower than those of filamentous fungi, and that the highest filamentous fungal activities (0.61 U/mg protein) was approximately twice that of yeast. The filamentous fungi and yeast with highest LG kinase production activity were identified by DNA sequence analysis of 18S or 26S rRNA gene fragments respectively. The identities of them were Alternaria alternata, Eupenicillium javanicum, Aspergillus niger, Penicillium herquei, Cryptococcus laurentii, Cryptococcus flavescens, Cryptococcus luteolus and Rhodotorula aurantiaca, respectively. In addition, the LG kinases from both types of organisms were purified. The effects of temperature and pH on LG kinase activity were also studied. There was very little change in activity between pH 7 to 10 and at temperatures below 30 °C. The apparent K m values for ATP were also similar in different microorganisms, but K m values for LG ranged from 48 to 102 mM.  相似文献

Summary The ultrastructure of the endosymbionts of several populations of whitefly (Homoptera: Aleyrodidae) was examined using transmission electron microscopy. Consistent differences in morphology and relative number of endosymbionts were observed between species and biotypes of whitefly within the Bemisia taxon.Bemisia argentifolii (=B. tabaci B biotype) individuals from Hawaii, Florida, and Arizona contained two morphological types of microorganisms housed within the mycetocyte cells of immature whiteflies. In contrast, individuals from populations ofB. tabaci A biotype from Arizona and Mexico, andB. tabaci Jatropha biotype from Puerto Rico, consistently contained three distinct morphological types of microorganisms within their mycetocytes. Organisms fromB. tabaci A and Jatropha biotypes differed from each other in the relative frequency of each type of microorganism. These observations suggest that different whitefly biotypes may have variable combinations of micro-fauna, with some possibly unique to each group, and furthers the hypothesis that variation in whitefly endosymbionts may be associated with the development of biotypes.  相似文献

Mitochondrial variability was investigated in natural populations of wild carrot (Daucus carota ssp carota) in different regions: South of France, Greece, and various sites in the Mediterranean Basin and Asia. Total DNA was digested with two restriction endonucleases (EcoRV and HindIII) and probed with three mitochondrial DMA-specific genes (coxI, atp6, and coxII). Twenty-five different mitochondrial types were found in 80 analyzed individuals. Thirteen mitotypes were found among the 7 French populations studied. On average, 4.4 different mitotypes were observed per population, and these mitotypes were well-distributed among the populations. All of the mitochondrial types were specific to a single region. However, the proportion of shared restriction fragments between 2 mitotypes from different regions was not particularly lower than that which occurred among mitotypes from a single region. On the basis of the sexual phenotype [male-sterile (MS) or hermaphrodite] of the plants studied in situ and that of their progeny, 2 mitotypes were found to be highly associated with male sterility. Eighty percent of the plants bearing these mitotypes were MS in situ, and all of these plants produced more than 30% MS plants in their progeny. This association with male sterility was consistent in several populations, suggesting an association with a cytoplasmic male-sterility system. Moreover, these two mitotypes had very similar mitochondrial DNA restriction patterns and were well-differentiated from the other mitotypes observed in wild plants and also from those observed in the two CMS types already known in the cultivated carrot. This suggests that they correspond to a third cytoplasmic sterility.  相似文献

Probiotics are defined as live microorganisms, which when administered in adequate amount confer a health benefit to the host. Most of studied or commercialized probiotics contain bacteria and very few of them present yeast in its composition. In this last case, the microorganisms almost always belong to Saccharomyces genus. In the present study, it was of interest to screen among 103 non-Saccharomyces yeasts a candidate for probiotic by using in vitro and in vivo criteria. In vitro assays included growth at 37°C and production of antagonistic compounds against enteropathogenic indicators, and the in vivo assays evaluated the colonization ability of mouse gastrointestinal tract without pathologic consequences and the protective ability in mice experimentally challenged with Clostridium difficile. In conclusion, Pichia kluyveri strain 898 showed to be a potential candidate for probiotic use, based on the criteria cited above, particularly as demonstrated by its protective effect against experimental infection in mice. Interestingly, an in vivo inhibition against C. difficile observed in the animal models did not correlate with the results obtained with the in vitro assays.  相似文献

Summary Several types of filamentous microorganisms were observed and identified in samples of poorly settling (bulking) activated sludge. The major types encountered and the frequency (percentage) of appearance in the total of all treatment plants sampled were: Eikelboom type 0041 (60), type 1701 (45), Haliscomenobacter hydrossis (35), type 021N (30), Thiothrix spp. (20), and Sphaerotilus natans (20). Isolation techniques and culture media were developed and used to recover 42 axenic strains of filamentous bacteria from sludge samples collected. The isolates were identified as strains of Thiothrix, Beggiatoa, S. natans, and Eikelboom types 021N, 1701, 0041, and 0803. Nutritional and differential characterization of the bacteria was important to the differentiation of groups which could not be easily distinguished on the basis of morphology. Although certain treatment plant operating parameters (organic loading) seemed associated with the presence of specific filamentous organism types, possible interaction among factors precluded definite establishment of a cause and effect relationship for most of the treatment plant characteristics and organisms observed.  相似文献

为探讨巴西橡胶树(Hevea brasiliensis)自根幼态无性系与供体间差异产生的原因,应用甲基化敏感扩增多态性扩增技术,对巴西橡胶树体细胞胚发生过程中基因组DNA 胞嘧啶甲基化程度和模式进行了分析。结果表明,在巴西橡胶树体细胞胚发生过程中不同阶段的DNA 甲基化程度不同,以花药的DNA 甲基化程度最高,体细胞胚的DNA 甲基化水平最低。在体细胞胚发生过程中出现了I、Ⅱ和Ⅲ 3 种类型的甲基化多态性带型的改变,包括他们的出现与消失。因此,橡胶树体细胞胚发生过程中可能通过DNA 甲基化甲基化和去甲基化来调控基因的表达。  相似文献

The study evaluated different macroalgal invasions in the main Mediterranean coastal habitats on hard bottom. Biodiversity, species composition and structure of macroalgal assemblages were compared among non-invaded areas and areas invaded by the Chlorophyta Caulerpa racemosa var. cylindracea and by the turf-forming Rhodophyta Womersleyella setacea in three different habitats: shallow rocky bottom, deep rocky bottom and dead matte of the seagrass Posidonia oceanica. Results showed that alien macroalgae constituted a relevant component of benthic assemblages in invaded areas of the Mediterranean Sea. Assemblages invaded by Womersleyella setacea and Caulerpa racemosa showed lower values of diversity and large differences in the structure and species composition related to non-nvaded assemblages. The species that mostly suffered from invasion were erect species reproducing sexually; moreover, the dominance of W. setacea led to low abundance of native filamentous algae, while C. racemosa colonization seemed particularly threatening for encrusting algae. All the studied habitats appeared highly invasible by alien macroalgae, even if W. setacea appeared more invasive in deeper habitats, while colonization of C. racemosa seemed more serious in shallower habitats; the dead matte of P. oceanica represented a suitable substrate for the spread of both species. Differences among assemblages in different habitats were reduced in invaded areas.  相似文献

Great genetic diversity was revealed among 75 rhizobal isolates associated with Vicia faba grown in Chinese fields with AFLP, ARDRA, 16S rDNA sequencing, DNA–DNA hybridization, BOX-PCR and RFLP of PCR-amplified nodD and nodC. Most of the isolates were Rhizobium leguminosarum, and six isolates belonged to an unnamed Rhizobium species. In the homogeneity analysis, the isolates were grouped into three clusters corresponding to (1) autumn sowing (subtropical) region where the winter ecotype of V. faba was cultivated, (2) spring sowing (temperate) region where the spring ecotype was grown, and (3) Yunnan province where the intermediate ecotype was sown either in spring or in autumn. Nonrandom associations were found among the nod genotypes, genomic types and ecological regions, indicating an epidemic symbiotic gene transfer pattern among different genomic backgrounds within an ecological region and a relatively limited transfer pattern between different regions. Conclusively, the present results suggested an endemic population structure of V. faba rhizobia in Chinese fields and demonstrated a novel rhizobium associated with faba bean. Lactic acid bacteria (LAB) are among the integral microflora of ragi tape, a dry starter of Balinese rice wine, brem. The species diversity and population level of LAB present in different types of ragi tape were studied by colony hybridization using 16S and 23S rDNA targeted oligonucleotide probes. These probes were DB6, Lbc, Wgp, and Rpt, which were specific for Enterococcus faecium, Lactobacillus curvatus, Weissella spp., and Pediococcus pentosaceus, respectively. Results revealed that P. pentosaceus and Weissella spp. were the predominant LAB in ragi tape, whereas L. curvatus and E. faecium were associated specific to types of ragi tape. A 21-mer species-specific oligonucleotide probe, Rpt, that targets the 16S rDNA of P. pentosaceus was developed in this study and found to be highly specific to be used as an effective tool to enumerate population of this species in ragi tape and its population changes during rice wine production. It was detected that LAB showed active growth during the early stage of brem fermentation. A succession of growth of LAB population during the fermentation was observed in which the heterofermentative LAB, Weissella spp., grew first, followed by the proliferation of P. pentosaceus.  相似文献

【背景】垫状点地梅作为青藏高原最具代表性的垫状植物,其叶际和内生微生物对适应极端环境有重要意义,同时也是一种独特的资源。【目的】探究垫状点地梅叶际和叶内可培养微生物多样性,以及不同生存状态个体之间的微生物差异。【方法】采用纯培养方法分离和纯化3个不同地区垫状点地梅叶际和叶内的细菌、酵母菌和丝状真菌,并用16S rRNA基因和ITS区域序列进行分析鉴定。【结果】最终得到叶际微生物350株,鉴定为22属49种,优势种为Penicillium sajarovii;内生微生物274株,鉴定为19属45种,优势种为Bacillusmycoides;两者的优势属均为Penicillium。垫状点地梅叶际和叶内之间及不同生存状态个体之间微生物的α多样性大多无显著差异,各群落间的成员也有重叠,但物种组成存在显著的空间异质性。【结论】垫状点地梅叶际和叶内有着丰富的可培养微生物资源,来源于不同生存状态的个体或不同部位的微生物物种组成差别较大,微生物对不同环境的选择偏好形成了不同的群落模式。但这些不同来源的微生物群落之间同样存在高比例的共有菌株,这些共有菌株的异养方式和生态位并不固定,可兼共生和腐生生存,生...  相似文献

Summary The ultrastructure of the intercellular bridges in the ovaries of Orectochilus villosus has been studied. In young egg chambers the inner filamentous coating of the bridge is thin. Characteristic vesicles are observed in close contact with this coating. The inner coating of the vitellogenic chambers is fully developed although vesicles are only sporadically associated with it. The accumulation of cadmium (Cd) by Tetraselmis chuii and Spirulina maxima was studied with dead and growing cells. Results indicated that the 2 microorganisms accumulated Cd by 2 different means according to the mechanisms involved—metabolism-dependent or metabolism-independent sorption. The mechanism involved in Cd accumulation on Tetraselmis chuii was restricted to surface phenomena, while in Spirulina maxima, Cd was accumulated on different layers of the cyanobacterium surface. In order to select a suitable immobilization support for the cells, several seaweeds were tested. Two types of seaweed were selected for experiments, using a small continuous pilot unit: Sargassum sp., a strong Cd adsorber, and Ulva sp., a poor one. The column reactors of the continuous system were filled with the algal supports and covered with dense microbial biofilms of Tetraselmis chuii or Spirulina maxima. The results obtained proved the success of the association between living microbial cells and dead seaweeds for operation of the continuous system.  相似文献

【目的】研究新疆艾比湖湿地不同季节盐角草根际和非根际土壤固氮微生物的多样性和丰富度与环境因子的相关性,以期探究在荒漠化和盐渍化不断严重的艾比湖湿地中随着季节变化的固氮微生物群落对恢复生态功能起到的潜在作用,为后续的湿地保护和退化恢复工作提供理论支持和数据基础。【方法】应用Illumina HiSeq PE250测序技术,分析6个土壤样本固氮微生物的多样性,结合相关的理化因子并利用RDA分析法探究土壤理化性质和固氮微生物菌落结构及丰富度的相关性。【结果】艾比湖湿地盐角草植物根际土壤的固氮微生物多样性高于非根际土壤,7月的土壤固氮微生物多样性高于10月和4月的土壤。土杆菌属(Geobacter)、假单胞菌属(Pseudomonas)、固氮菌属(Azotobacter)和慢生根瘤菌属(Bradyrhizobium)等为盐角草根际和非根际土壤中的共同优势菌属。这些固氮微生物优势菌属隶属于变形菌门(Proteobacteria)和蓝藻门(Cyanobacteria),且相对丰富度占比为85%和10%,其余各菌门共占比较少,仅为5%。土壤中固氮微生物的优势菌群与碱解氮(AN)、全氮(TN)、速效钾(AK)和有效磷(TP)呈显著相关。【结论】随着时间的推移土壤样本中固氮微生物的多样性和群落结构也发了改变,同一时期植物根际与非根际土壤中固氮微生物的群落结构并不相同。土壤的环境因子与固氮细菌的群落结构和丰富度的相关性研究可以为艾比湖湿地的退化恢复提供数据基础和理论支持。  相似文献

California exports tree nuts to countries where they face stringent standards for aflatoxin contamination. Trade concerns have stimulated efforts to eliminate aflatoxins and Aspergillus flavus from almonds, pistachios and walnuts. Incidence of fungi on tree nuts and associations among fungi on tree nuts were studied. Eleven hundred pistachios, almonds, walnuts and brazil nuts without visible insect damage were plated on salt agar and observed for growth of fungi. Samples came both from California nut orchards and from supermarkets. To distinguish internal fungal colonization of nuts from superficial colonization, half the nuts were surface-sterilized before plating. The most common genera found were Aspergillus , Rhizopus and Penicillium . Each species of nut had a distinct mycoflora. Populations of most fungi were reduced by surface sterilization in all except brazil nuts, suggesting that they were present as superficial inoculum on (rather than in) the nuts. In general, strongly positive associations were observed among species of Aspergillus ; nuts infected by one species were likely to be colonized by other species as well. Presence of Penicillium was negatively associated with A. niger and Rhizopus in some cases. Results suggest that harvest or postharvest handling has a major influence on nut mycoflora, and that nuts with fungi are usually colonized by several fungi rather than by single species. In the mangrove surrounding the coastal lagoon of La Mancha, Veracruz, Mexico, we studied litter fall, litter standing crop, and turnover rates in four different mangrove settings, based on the ecological classification of Lugo and Snedaker (1974). We studied those three prominent ecological processes at the basin, fringe and riverine mangrove settings, being the last one a relict riverine stand. The aim was to describe and compare litter dynamics among mangrove types in a lagoon with an ephemeral inlet, as a way of understanding functional heterogeneity within this coastal ecosystem. The daily average values of litter fall were different (P < 0.01) among mangrove site basin I, formed by Avicennia germinans and Rhizophora mangle (2.35 g/m2/day); basin II, formed by Laguncularia racemosa, Avicennia germinans, and Rhizophora mangle (2.93 g/m2/day); fringe with Rhizophora mangle (2.13 g/m2/day); and relic riverine, also with R. mangle (4.70 g/m2/day). The amount of litter standing crop was different among sites (P < 0.001), and also between the dry and rainy season, for each mangrove type (P < 0.001). Turnover ratios were higher in basin I and basin II sites (6.34 and 7.44 times per year) than in relic riverine and fringe mangroves (1.49 and 2.39 times per year). Interstitial salinity and sediment nutrients varied among mangrove types and could influence litter production. Since this lagoon has an ephemeral inlet, continuous inundation throughout 7–8 months per year has an important effect on litter dynamics.  相似文献

Ultrastructure and morphogenesis of extremely halophilic neutrophilic (Halobacteriam distributum, Halococcus turkmenicus) and alkaliphilic (Natronobacterium pharaonis, Natronococcus occultus) archaeobacteria were studied. The H. distributum culture was rather polymorphous and produced cells of four types. Due to the irregular cell fission in different planes packages of various numbers of cells surrounded by a common capsule were formed. Resting forms (halocysts) with multilayer covers were present in the population. The N. pharaonis culture consisted of rod-like cells and cyst-like forms. Besides, under conditions of carbon limitation, multicellular aggregated forms were found in the culture. Encapsulated single cells and aggregated forms with a common capsule were observed in H. turkmenicus and N. occultus cultures.  相似文献

The acquired freeze-thaw tolerance was investigated forLactococcus lactis ssp.diacetylactis. Pretreatment of microorganisms at less severe temperatures to initiate cold tolerance gaveL. lactis ssp.diacetylactis improved cell viability after successive freezings and thawings. The ability of cells to survive freeze-thaw was dependent on factors experienced prior to freezing. Factors affecting lactic acid bacteria survival during freeze-thaw cycles were found to be different diluents, growth phase, and different cold temperatures. Viability experiments showed that this strain displaying cold shock cryotolerance had an improved survival capacity in stationary phase. The plasmid contents of lactic acid bacteria isolated from different types, DRC-2 and DRC-2C, were examined and compared with the plasmid contents of culture collection strains both before and after cold shock treatment. Using agarose gel electrophoresis, no obvious correlation between the cold shock response and the number of plasmids in the cell could be observed.  相似文献

The cry toxin encoding plasmid pHT73 was transferred from Bacillus thuringiensis subspecies kurstaki KT0 to six B. cereus group strains in three lepidopteran (Spodoptera exigua, Plutella xyllostella and Helicoverpa armigera) larvae by conjugation. The conjugation kinetics of the plasmid was precisely studied during the larval infection using a new protocol. The infections were performed with both vegetative and sporulated strains. However, larval death only occurred when infections were made with spore and toxin preparations. Likewise, spore germinations of both donor and recipient strains were only observed in killed larvae, 44–56 h post-infection. Accordingly, kinetics showed that gene transfer between B. thuringiensis strain KT0 and other B. cereus strains only took place in dead larvae among vegetatively growing bacteria. The conjugational transfer ratios varied among different strain combinations and different larvae. The highest transfer ratio reached 5.83 × 10−6 CFU/donor between the KT0 and the AW05R recipient in Helicoverpa armigera, and all transconjugants gained the ability to produce the insecticidal crystal. These results indicated that horizontal gene transfer among B. cereus group strains might play a key role for the acquisition of extra plasmids and evolution of these strains in toxin susceptible insect larvae.  相似文献

The antibiotic resistance profile of 17 poultry isolates of Salmonella was studied against 24 different antibiotics. 69–88% of the Salmonella isolates displayed a high level of resistance, particularly against penicillin, rifampicin, erythromycin, clarithromycin, clindamycin, sulphamethoxazole and vancomycin. In contrast, a relatively low or moderate level of resistance was observed against furazolidone, spectinomycin, ciprofloxacin, chloramphenicol, cefepime, carbenicillin, nalidixic acid, streptomycin, oxacillin and cephalothin (11–59%). Moreover, resistance to multiple antibiotics (2–5) was also observed among the Salmonella strains, and none of the isolates was found susceptible to all the antibiotics used. Similarity coefficient among Salmonella strains by RAPD-PCR analysis varied from 0.60 to 0.86, and all the salmonellae could be classified into seven groups on the basis of dendrogram analysis. Generally, a very high level of concordance between RAPD-PCR profile and antibiotic profile was not observed, which indicates that genes for antibiotic resistance may not always be present on genomic DNA rather may be plasmid-borne.  相似文献
