Interaction of cis-dichloro(dipyridine)platinum(II) (cis-PPC) with calf thymus DNA, calf thymus histone, l-amino acids, poly-l-amino acids, nucleosides, and nucleotides has been evaluated by equilibrium dialysis technics. At least a 28 % decrease in the association of cis-PPC with DNA occurs when the platinum compound is pre-incubated with l-amino acids. The greatest decrease in association is seen upon pre-incubation of the platinum compound with the free amino acids. Glut, Asp, Lys, Arg, and CySH, before the addition of a sack containing a solution of DNA. The low level of association between DNA and the amino acids tends to rule out competition between cis-PPC and amino acids for DNA association sites. cis-PPC was repelled from sacks containing positively charged poly-l-Lys, poly-l-Arg, and calf thymus histone; however, in the presence of poly-l-Glut and poly-l-Asp, cis-PPC associated with these negatively charged polymers to a considerable degree. Enhanced chloride dissociation from cis-PPC was observed in the presence of all of the amino acids and the nucleotides GMP, CMP, UMP, and TMP, but not in the presence of AMP or the nucleosides rG and dG. In the presence of calf thymus histone, the association of cis-PPC with calf thymus DNA was reduced by more than 50% at histone/DNA ratios of 0.8–1.0.These data suggest that cis-PPC or cis-Pt(II) may associate with electron-rich areas of not only nucleic acids and proteins but also with body pools of free nucleotides and amino acids. The presence of positively charged histones shielding DNA strands in vivo suggests that the most probable point of platinum-DNA association would be at de-repressed areas of DNA which are undergoing RNA synthesis. The aquated form of the platinum complex may also associate with acidic proteins which appear to be involved in the positive control of RNA synthesis and, as a result, this interaction may be of pharmacological significance.  相似文献

The influence of substituted nicotine and isonicotine amides and their Pt(IV) metal complexes on activity of cAMP specific phosphodoiesterase (cAMP-PDE), hydrolyzing cAMP, known second messenger, has been investigated. Isonicotine derivatives are effective activators of this enzyme. On the contrary, substituted nicotine amides inhibited cAMP-PDE activity as efficiently as a reference substance, theophylline. In general, the inhibitory effects of nicotine amides and their metal complexes did not differ. However, in contrast to the parent compound, one of the metal complexes of this group competitively inhibited the enzyme while the complex and the ligand itself had the same reversibility.  相似文献

The reaction of H2[PtCl6] · 6H2O and (H3O)[PtCl5(H2O)] · 2(18C6) · 6H2O (18C6 = 18-crown-6) with 9-methylguanine (MeGua) proceeded with the protonation of MeGua forming 9-methylguaninium hexachloroplatinate(IV) dihydrate (MeGuaH)2[PtCl6] · 2H2O (1).The same compound was obtained from the reaction of Na2[PtCl6] with (MeGuaH)Cl.On the other hand, the reaction of guanosine (Guo) with (H3O)[PtCl5(H2O)] · 2(18C6) · 6H2O in methanol at 60 °C proceeded with the cleavage of the glycosidic linkage and with ligand substitution to give a guaninium complex of platinum(IV), [PtCl5(GuaH)] · 1.5(18C6) · H2O (2).Within several weeks in aqueous solution a slow reduction took place yielding the analogous guaninium platinum(II) complex, [PtCl3(GuaH)] · (18C6) · 2Me2CO (3).H2[PtCl6] · 6H2O and guanosine was found to react in water, yielding (GuoH)2[PtCl6] (4) and in ethanol at 50 °C, yielding [PtCl5(GuoH)] · 3H2O (5).Dissolution of complexes 2 and 5 in DMSO resulted in the substitution of the guaninium and guanosinium ligands, respectively, by DMSO forming [PtCl5(DMSO)].Reactions of 1-methylcytosine (MeCyt) and cytidine (Cyd) with H2[PtCl6] · 6H2O and(H3O)[PtCl5(H2O)] · 2(18C6) · 6H2O resulted in the formation of hexachloroplatinates with N3 protonated pyrimidine bases as cation (MeCytH)2[PtCl6] · 2H2O (6) and (CydH)2[PtCl6] (7), respectively. Identities of all complexes were confirmed by 1H, 13C and 195Pt NMR spectroscopic investigations, revealing coordination of GuoH+ in complex 5 through N7 whereas GuaH+ in complex 3 may be coordinated through N7 or through N9. Solid state structure of hexachloroplatinate 1 exhibited base pairing of the cations yielding (MeGuaH+)2, whereas in complex 6 non-base-paired MeCytH+ cations were found. In both complexes, a network of hydrogen bonds including the water molecules was found. X-ray diffraction analysis of complex 3 exhibited a guaninium ligand that is coordinated through N9 to platinum and protonated at N1, N3 and N7. In the crystal, these NH groups form hydrogen bonds N–HO to oxygen atoms of crown ether molecules.  相似文献

Platinum(II) and platinum(IV) complexes with 3-amino-5-methyl-5-(4-pyridyl)-2,4-imidazolidenedione (L) with general formulaе cis-[PtL2X2nH2O and [PtL2Cl4], where X = Cl, Br, I and n = 2-4) were synthesized. The novel compounds were fully characterized by elemental analysis, IR, 1H, 13C, 195Pt NMR spectra, thermal analysis and molar conductivity. The geometry of Pt(II) complexes and of the organic ligand in the gas phase were optimized using the hybrid DFT method B3LYP with LANL2DZ and 6-31G** basis sets. Some physicochemical parameters as dipole moment, HOMO, LUMO energies and ESP charges were calculated. The comparison of the bond length and angles, obtained from the X-ray analysis and DFT calculations is realized. The cytotoxic effects of these complexes in human T-cell leukemia KE-37 (SKW-3) are reported.  相似文献

New tetrazolate complexes trans-[PtCl2(RCN4)2]2−, trans-[PtCl4(RCN4)2]2− with Ph3PCH2Ph+ and (CH3)2NH2+ counterions have been obtained by azidation of nitriles coordinated to Pt(II) and Pt(IV) {trans-[PtCl2(RCN)2] and trans-[PtCl4(RCN)2] (R = Et, Ph)} and characterized. The composition and the molecular structure of the complexes obtained were established by the СHN elemental analyses, 1Н and 13С NMR spectroscopy, IR spectroscopy, mass spectrometry, and X-ray diffraction. The coordination of nitriles to Pt(II) and Pt(IV) is shown significantly activate the azidation: the reaction proceeds with a higher rate and at relatively low temperature compared with the classical 1,3-dipolar addition of azides to nitriles.  相似文献

Reaction of pyridine-2,6-dicarboxylic acid (dipicH2) with ammonium ceric nitrate and Cu(II) salts yielded three heterometallic compounds all of which contain [Ce(dipic)3]2− linked to aquo-Cu(II) complex units. Part of the Ce(IV) gets reduced by solvent during the reaction leading to [(Ce(dipic)3Ce(H2O)8)2Cu(H2O)4][Ce(dipic)3]2·12H2O (1). Other lanthanide(III) ions could take the place of Ce(III) as demonstrated by the preparation of [(Ce(dipic)3La(H2O)8)2Cu(H2O)4][Ce(dipic)3]2·12H2O (4), which is isomorphous with compound 1. [Ce(dipic)3Cu(H2O)4]·8H2O (2) is a one-dimensional coordination polymer in which two types of aquo-Cu(II) complex units which differ in the orientation of the tetragonal axis alternate along the chain. The central Cu(H2O)22+ unit in the trinuclear anion of [Cu(H2O)6][Ce(dipic)3Cu(H2O)2Ce(dipic)3]·8H2O (3) is chelated by two carboxylate groups in trans positions in off-axis mode. In all the four complexes, the Cu(II) centres are magnetically isolated leading well-resolved EPR spectra in polycrystalline samples.  相似文献

A series of Pt(II) complexes containing 1,2-diphenylethylenediamine (stien) isomers were synthesized and tested for their antitumor activity against leukemia L1210. Among the Pt(II) complexes examined water-soluble Pt(II) complexes with sulfate, nitrate and D-glucuronate ions as leaving groups exhibited relatively high antitumor activity. Furthermore, the interactions between calf-thymus DNA and Pt(SO4) (stein) complexes were investigated by means of circular dichroism spectrometry. Dichroism enhancements observed in the interaction between DNA and Pt(SO4) (stien) complexes were analysed to be contributable to two factors: (1) vicinal effects of DNA on the d-d transitions of Pt(II) ions and (2) conformational changes of DNA caused by the coordination of cis-configurational Pt(II) complexes.  相似文献

From the reaction between dihydroxoplatinum(II) and l-ascorbic acid, two types of platinum(II) ascorbate complexes were obtained and structurally characterized with ethylenediamine (en), N,N-dimethylethylenediamine (dmen) and N,N,N′-trimethylethylenediamine (trimen) as stabilizing ligands. In [Pt(en)(asc-C,O)] (1), [Pt(dmen)(asc-C,O)] (2) and [Pt(trimen)(asc-C,O)] (4), the ascorbate dianion forms a five-membered chelate ring, coordinating to the Pt(II) ion at the 2-carbon and the 5-oxygen atoms (C,O-chelate). From the same mother solution, crystals of [Pt(trimen)(asc-O,O′)] (3) were obtained during the precipitation of 4; in 3 the ascorbate is bound to the Pt at the 2- and 3-oxygen atoms (O,O′-chelate). Compounds 3 and 4 are the first well-characterized linkage isomers among the transition-metal ascorbate complexes. The O,O′-chelated 3 slowly changes to the C,O-chelated 4 in an aqueous solution. Bulkiness of the stabilizing ligand, i.e. en, dmen and trimen has an influence on the formation of the C,O-chelated species, 1, 2 and 4.  相似文献

The Pt(II) complexes of 2N1O-donor ligands containing a pendent indole, 3-[N-2-pyridylmethyl-N-2-hydroxy-3,5-di(tert-butyl)benzylamino]ethylindole (Htbu-iepp), and 1-methyl-3-[N-2-pyridylmethyl-N-2-hydroxy-3,5-di(tert-butyl)benzylamino]ethylindole (Htbu-miepp) (H denotes an ionizable hydrogen), were synthesized, and the structure of [Pt(tbu-iepp)Cl] (1) was determined by X-ray analysis. Complex 1 prepared in CH3CN was revealed to have the C2 atom of the indole ring bound to Pt(II) with the Pt(II)-C2 distance of 1.981(3) Å. On the other hand, [Pt(tbu-miepp)Cl] (2) was concluded to have a phenolate coordination instead of the C2 atom of the indole ring by 1H NMR spectra. Reaction of 1 with 1 equiv. of Ce(IV) in DMF gave the corresponding one-electron oxidized species, which exhibited an ESR signal at g = 2.004 and an absorption peak at 567 nm, indicating the formation of the Pt(II)-indole-π-cation radical species. The half-life, t1/2, of the radical species at −60 °C was calculated to be 43 s (kobs = 1.6 × 10−2 s−1).  相似文献

An investigation into the feasibility of removing Cu(II) and Cr(IV) from solution with basidiomycete (Gloeophylum sepiarium, Pleurotus sp.)-colonized sawdust was undertaken. Obeche (Triplochyton scleroxylon) sawdust exposed to the basidiomycetes for 1–3 months reduced the concentration of the metals in the solution to 22.0–84.4 mg/l. The supernatant from the centrifuged mixture of a solution of 100 mg metal ions/l and aqueous extract of a 3-month basidiomycete-degraded obeche sawdust contained lower concentration of the metal ions (38.6–75.4 mg/l). Unextracted sawdust of pigmented tropical timbers, African mahogany (Khaya ivorensis), black afara (Terminalia ivorensis) and camwood (Baphia nitida) exposed to the test basidiomycetes, removed Cu and Cr significantly better than the extracted sawdust. It is hypothesised that some products of basidiomycete wood-degradative activities were ligands which immobilized the test metals.  相似文献

This study details the reactions of the electrophiles CH3Hg(NO3), cis-[PtCl2(NH3)2] (cis-DDP) and trans-[PtCl2(NH3)2] (trans-DDP) with calf thymus DNA using Raman and Raman difference spectroscopy. The order of CH3Hg(II) binding to calf thymus DNA is G > T > C > A. The electrophilic attack of cis- and trans-DDP on calf thymus DNA produces different orders of binding: cis-DDP-G>C approximately AT, trans-DDP-G approximately C approximately AT. The reaction of CH3Hg(II) with DNA results in a decrease in the percentage of B-form DNA. whereas the reactions of cis- and trans-DDP with DNA decrease the percentage of B-DNA and cause the formation of C-DNA structure.  相似文献

In this study, the effects of four novel mononuclear palladium(II) and platinum(II) complexes on the activity of human serum paraoxanase1 were examined. First, four novel mononuclear palladium(II) and platinum(II) complexes were synthesized with a nitrogen donor ligand 1‐(2‐aminoethyl)pyrrolidine and nonsteroidal anti‐inflammatory drugs diclofenac, mefenamic acid. These complexes were characterized by spectroscopic, thermal, and elemental analyses. The crystal structures of complex [Pd(2‐amepyr)2](dicl)2 1 and [Pd(2‐amepyr)2](mef)2 3 were determined by X‐ray crystallography. Then, paraoxonase1 enzyme was purified from human serum. The effects of these complexes on enzyme were evaluated in vitro. The complexes consist of the cationic unit and the counterions. The diclofenac and mefenamic acid acted as a counterion in the complexes. It was observed that all the complexes were stable up to high temperatures. These complexes, even at low doses, inhibited the activity of the enzyme with different inhibition mechanisms.  相似文献

Four palladium(II) and platinum(II) saccharinate (sac) complexes with 2-(hydroxymethyl)pyridine (2-hmpy) and 2-(2-hydroxyethyl)pyridine (2-hepy), namely trans-[Pd(2-hmpy)2(sac)2]·H2O (1), trans-[Pt(2-hmpy)2(sac)2]·3H2O (2), trans-[Pd(2-hepy)2(sac)2] (3) and trans-[Pt(2-hepy)2(sac)2] (4), have been synthesized and characterized by elemental analysis, UV–vis, IR and NMR. Single crystal X-ray analysis reveals that the metal(II) ions in each complex are coordinated by two sac and two 2-hmpy or 2-hepy ligands with a trans arrangement. Anticancer effects of 14 were tested against four different cancer cell lines (A549 and PC3 for lung cancer, C6 for glioblastoma, and Hep3B for liver cancer). Cytotoxicity was first screened by the MTT assay and the results were further confirmed by the ATP assay. The mode of cell death was determined by both histological and biochemical methods. Among the metal complexes, complex 2 resulted in relatively stronger anti-growth effect in a dose-dependent manner (3.13–200 μM), compared to the others, by inducing apoptosis.  相似文献

New mixed polypyridyl {NMIP = 2′-(2″-nitro-3″,4″-methylenedioxyphenyl)imidazo-[4′,5′-f][1,10]-phenanthroline, dmb = 4,4′-dimethyl-2,2′-bipyridine, bpy = 2,2′-bipyridine} ruthenium(II) complexes [Ru(dmb)2(NMIP)]2+ (1) and [Ru(bpy)2(NMIP)]2+ (2) have been synthesized and characterized. The binding of these complexes to calf thymus DNA (CT-DNA) has been investigated with spectroscopic methods, viscosity and electrophoresis measurements. The experimental results indicate that both complexes could bind to DNA via partial intercalation from the minor/major groove. In addition, both complexes have been found to promote the single-stranded cleavage of plasmid pBR 322 DNA upon irradiation. Under comparable experimental conditions compared with [Ru(phen)2(NMIP)]2+, during the course of the dialysis at intervals of time, the CD signals of both complexes started from none, increased to the maximum magnitude, then no longer changed, and the activity of effective DNA cleavage dependence upon concentration degree lies in the following order: [Ru(phen)2NMIP]2+ > complex 2 > complex 1.  相似文献

Syntheses of two novel ligand precursors O,O'-diisopropyl- (1a) and O,O'-diisobutyl-(S,S)-ethylenediamine-N,N'-di-2-propanoate dihydrochloride monohydrate (1b) and the corresponding dichloroplatinum(II) (2a and 2b) and tetrachloroplatinum(IV) complexes (3a and 3b) are described here. The substances were characterized by IR, (1)H and (13)C spectroscopy and elemental analysis. Crystal structures were determined for 1a and the corresponding platinum(IV) complex, 3a. In vitro antiproliferative activity was determined against tumor cell lines: human adenocarcinoma HeLa, human myelogenous leukemia K562, human malignant melanoma Fem-x, rested and stimulated normal immunocompetent cells (human peripheral blood mononuclear PBMC cells) using KBR test (Kenacid Blue Dye binding test). The IC(50)(microM) values for the most active compound 3a were: 30.48+/-2.54; 12.26+/-2.60; 13.68+/-3.22; 80.18+/-24.07 and 71.30+/-21.70, respectively.  相似文献

Six dinuclear platinum(II) complexes with a chiral tetradentate ligand, (1R,1′R,2R,2′R)-N1,N1′-(1,4-phenylenebis(methylene))dicyclohexane-1,2-diamine, have been designed, synthesized and characterized. In vitro cytotoxicity evaluation of these metal complexes against human A549, HCT-116, MCF-7 and HepG-2 cell lines have been carried out. All compounds showed antitumor activity to HepG-2, HCT-116 and A549. Particularly, compounds A1 and A2 exhibited significant better activity than other four compounds and A2 even showed comparable cytotoxicity to cisplatin against HepG-2 cell line.  相似文献

The speciation of several insulin-mimetic/enhancing VO(IV) and Zn(II) complexes in human blood serum was studied and a comparison was made concerning the ability of the serum components to interact with the original metal complexes and the distribution of the metal ions between the low and the high molecular fractions of the serum. It was found that the low molecular mass components may play a larger role in transporting Zn(II) than in the case with VO(IV). Among the high molecular mass serum proteins, transferrin is the primary binder of VO(IV), and albumin is that of Zn(II). The results revealed that protein-ligand interactions may influence the metal ion distribution in the serum.  相似文献

The clinical application of Pt-based anticancer drugs has inspired the development of novel chemotherapeutic metallodrugs with improved efficacies. Pt(IV) prodrugs are one of the most promising successors of Pt(II) drugs and have displayed great anticancer performance. In particular, judicious modification of axial ligands endows Pt(IV) complexes with unique properties that enable them to overcome the limitations of conventional Pt(II) drugs. Herein, we summarize recent developments in Pt(IV) anticancer complexes, with a focus on their axial functionalization with other anticancer agents, immunotherapeutic agents, photosensitive ligands, peptides, and theranostic agents. We hope that this concise view of recently reported Pt(IV) coordination complexes will help researchers to design next-generation multi-functional anticancer agents based on a comprehensive Pt(IV) platform.  相似文献
