One new pentaoxygenated free xanthone and a new pentaoxygenated xanthone-O-glucoside have been isolated and characterized from the flowering top of a fresh batch of Canscora decussata. The structure previously assigned to ‘xanthone 9’ has now been confirmed by application of NOE. The biochemical significance of xanthone formation and glucosidation in plants is appraised.  相似文献

Natural pigments are important sources for the screening of bioactive lead compounds. This article reviewed the chemistry and therapeutic potentials of over 570 colored molecules from plants, fungi, bacteria, insects, algae, and marine sources. Moreover, related biological activities, advanced extraction, and identification approaches were reviewed. A variety of biological activities, including cytotoxicity against cancer cells, antioxidant, anti-inflammatory, wound healing, anti-microbial, antiviral, and anti-protozoal activities, have been reported for different pigments. Considering their structural backbone, they were classified as naphthoquinones, carotenoids, flavonoids, xanthones, anthocyanins, benzotropolones, alkaloids, terpenoids, isoprenoids, and non-isoprenoids. Alkaloid pigments were mostly isolated from bacteria and marine sources, while flavonoids were mostly found in plants and mushrooms. Colored quinones and xanthones were mostly extracted from plants and fungi, while colored polyketides and terpenoids are often found in marine sources and fungi. Carotenoids are mostly distributed among bacteria, followed by fungi and plants. The pigments isolated from insects have different structures, but among them, carotenoids and quinone/xanthone are the most important. Considering good manufacturing practices, the current permitted natural colorants are: Carotenoids (canthaxanthin, β-carotene, β-apo-8′-carotenal, annatto, astaxanthin) and their sources, lycopene, anthocyanins, betanin, chlorophyllins, spirulina extract, carmine and cochineal extract, henna, riboflavin, pyrogallol, logwood extract, guaiazulene, turmeric, and soy leghemoglobin.  相似文献

A new xanthone and twelve known compounds were isolated from the aerial parts of Gentiana corymbifera. The new xanthone was shown to be 3-methylcorymbiferin (1,8-dihydroxy-3,4,5-trimethoxy-9H-xanthen-9-one) by spectral and chemical procedures. The chemotaxonomic implications are discussed.  相似文献

Six different varieties of date-palm viz. Sukhari, Saggae, Rotana, Kholasi, Rashoodia and Nabtat Ali, were screened for seed-borne fungi. Eleven species belonging to nine different genera of fungi were isolated. The genera isolated were Alternaria, Eurotium, and Fusarium (two species), Aspergillus, Drechslera, Penicillium, Rhizopus, and Curvularia (one species each). This is the first record of seed-borne fungi from Phoenix dactyliera L. in Saudi Arabia.  相似文献

A rapid analytical method has been developed to determine xanthone and secoiridoid glycoside in in vitro and in vivo Swertia chirayita extracts. Ultra performance liquid chromatography–electrospray ionization mass spectrometry (LC-ESI/MS) was applied and validated for the analysis of xanthone and secoiridoid glycoside a potential active component isolated from methanolic extracts of in vitro and in vivo Swertia chirayita plantlets. Chromatographic separation was achieved on a RP-C18 column using gradient elution. Mangiferin (Xanthone), Amarogentin and Swertiamarin (Secoiridoid glycosides) were identified in both the extracts. In the LC/ESI-MS spectra, major [M + H] + and [M + Na] + ions were observed in positive ion mode and provided molecular mass information. An ultra-performance liquid-chromatography in combination with electrospray ionization tandem mass spectrometry involving metal cationisation was successfully utilized for the rapid identification of xanthone and secoiridoid glycosides. This method is suitable for the routine analysis, as well as for the separation and identification of known and novel secoiridoid glycoside and xanthone.  相似文献

A new biflavanone has been isolated from the stem bark of Garcinia mannii (Guttiferae) and identified as I-3′-II-3,3′-I-4′-II-4′-I-5-II-5-I-7-II-7-nonahydroxy-I-3-II-8-biflavanone. Structural assignments for this compound, which has been named manniflavanone, were made on the basis of spectral studies and simple degradation. Drechslera maydis causes severe leaf blight disease of Costus speciosus. One compound, drechslerol-A, isolated from the culture filtrate of this fungus was elucidated as (cis) hentetracont-10-ene-12-hydroxymethyl-4-ol on the basis of spectral analysis. The production of necrotic lesion was observed on the leaves of Costus speciosus with 1.6 × 10−4 M concentration. In the most sensitive reaction drechslerol-A shows complete killing effect on the root growth of wheat seedlings at the concentration of 3.2 × 10−5 M.  相似文献

Microluside A [4 (19-para-hydroxy benzoyloxy-O-β-d-cellobiosyl), 5 (30-para-hydroxy benzoyloxy-O-β-d-glucopyranosyl) xanthone (1)] is a unique O-glycosylated disubstituted xanthone isolated from the broth culture of Micrococcus sp. EG45 cultivated from the Red Sea sponge Spheciospongia vagabunda. The structure of microluside A was determined by 1D- and 2D-NMR techniques as well as high resolution tandem mass spectrometry. The antimicrobial activity evaluation showed that 1 exhibited antibacterial potential against Enterococcus faecalis JH212 and Staphylococcus aureus NCTC 8325 with MIC values of 10 and 13 μM, respectively.  相似文献

Parnafungins, natural products containing an isoxazolidinone ring, have been isolated from Fusarium larvarum and have been shown to be potent inhibitors of the fungal polyadenosine polymerase. The extraction and analysis of fermentation broths of taxonomically related organisms identified as closely related Fusarium spp. produce not only parnafungin A and B, but also significant quantities of two related components. These members of the paranfungin family of natural products have been isolated and the structure of each has been elucidated. While structurally analogous to parnafungin A, parnafungin C is further elaborated by methylation of a phenolic hydroxyl group, and parnafungin D has both the methyl phenol ether as well as an epoxide in the xanthone ring system. Parnafungin C and D have potent, broad spectrum antifungal activity and also have been shown to target fungal mRNA cleavage and polyadenylation.  相似文献

One new tetraoxygenated xanthone, merguensinone (1), along with one known xanthone, 1,5,6-trihydroxy-2-prenyl-6′,6′-dimethyl-2H-pyrano(2′,3′:3,4)xanthone (2) and five known biflavanoids, (?)-GB-1a (3), (?)-GB-2a (4), (+)-morelloflavone (5), (+)-volkensiflavone (6), and amentoflavone (7) were isolated from the methanol extract from the twigs of Garcinia merguensis. 8-Deoxygartanin and two new xanthones, 1,5-dihydroxy-6′,6′-dimethyl-2H-pyrano(2′,3′:3,2)-6″,6″-dimethyl-2H-pyrano(2″,3″:6,7)xanthone and 1,5,6-trihydroxy-6′,6′-dimethyl-2H-pyrano(2′,3′:3,2)-7-(3-methylprop-2-enyl)xanthone, have been isolated from the roots of Rheedia gardneriana. The latter of the two new xanthones has been assigned the trivial name 7-prenyljacareubin while the former has the structure erroneously assigned to pyranojacareubin reported from Garcinia densivenia. This work evaluated the activity and ultrastructural and morphological alterations induced by the xanthone 1,3,7-trihydroxy-2-(3-methylbut-2-enyl)-xanthone (C23) isolated from Kielmeyera coriacea against Trypanosoma cruzi. This xanthone had inhibitory activity against the three forms of this protozoan and did not induce toxicity in mammalian cells. The best activity of this xanthone was against the intracellular amastigote form. Additionally, the mitochondrion was the main target of this compound, reflected by electronic microscopy and rhodamine 123 assays. Our MitoSOX assay results also indicated that C23 increased O2? production in mitochondrion. C23 might be a promising chemotherapeutic agent against T. cruzi because its trypanocidal action involves the disruption of mitochondrion, a specific target of Trypanosomatides.  相似文献

The bark of Calophyllum calaba var. calaba contains a new xanthone, 2,8-di-(3-methylbut-2-enyl)-1,3-dihydroxy-7-methoxyxanthone (calocalabaxanthone), the precursor of 5-hydroxy-8-methoxy-2,2-dimethyl-7-(3-methylbut-2-enyl)-2H,6H-pyano-(3,2-b)-xanthen-6-one (calabaxanthone). The aim of this study was to isolate fungi able to produce pigments. Fifty strains were isolated from the Amazon soil by the conventional technique of serial dilution. Submerged fermentation was performed in Czapeck broth in order to select strains able to synthesise pigments. Five strains were able to produce pigments and were identified by sequencing the rDNA (ITS regions). These fungi were identified as Penicillium sclerotiorum 2AV2, Penicillium sclerotiorum 2AV6, Aspergillus calidoustus 4BV13, Penicillium citrinum 2AV18 and Penicillium purpurogenum 2BV41. P. sclerotiorum 2AV2 produced intensely coloured pigments and were therefore selected for chemical characterisation. NMR identified the pigment as sclerotiorin. In this work, the influence of nutrients on sclerotiorin yield was also studied and it was verified that rhamnose and peptone increased production when used separately. These results indicate that Amazonian fungi bioprospecting is a viable means to search for new sources of natural dyes.  相似文献

Xanthone production in Hypericum perforatum (HP) suspension cultures in response to elicitation by Agrobacterium tumefaciens co-cultivation has been studied. RNA blot analyses of HP cells co-cultivated with A. tumefaciens have shown a rapid up-regulation of genes encoding important enzymes of the general phenylpropanoid pathway (PAL, phenylalanine ammonia lyase and 4CL, 4-coumarate:CoA ligase) and xanthone biosynthesis (BPS, benzophenone synthase). Analyses of HPLC chromatograms of methanolic extracts of control and elicited cells (HP cells that were co-cultivated for 24 h with A. tumefaciens) have revealed a 12-fold increase in total xanthone concentration and also the emergence of many xanthones after elicitation. Methanolic extract of elicited cells exhibited significantly higher antioxidant and antimicrobial competence than the equivalent extract of control HP cells indicating that these properties have been significantly increased in HP cells after elicitation. Four major de novo synthesized xanthones have been identified as 1,3,6,7-tetrahydroxy-8-prenyl xanthone, 1,3,6,7-tetrahydroxy-2-prenyl xanthone, 1,3,7-trihydroxy-6-methoxy-8-prenyl xanthone and paxanthone. Antioxidant and antimicrobial characterization of these de novo xanthones have revealed that xanthones play dual function in plant cells during biotic stress: (1) as antioxidants to protect the cells from oxidative damage and (2) as phytoalexins to impair the pathogen growth.  相似文献

Five new metabolites, including the xanthone derivative wentixanthone A (1), the benzophenone wentiphenone A (2), the diastereomeric mixtures of the bianthrones wentibianthrone A (3a, b) and wentibianthrone B (4a, b), as well as (10R,10′S)-wentibianthrone C (5a) and (10R,10′R)-wentibianthrone C (5b) were obtained from the fungus Aspergillus wentii, isolated from soil of the hypersaline lake El Hamra in Wadi El-Natrun, Egypt. The structures of the isolated compounds were established by one and two-dimensional NMR and MS spectroscopic analysis. The relative configuration of bianthrones (35) was elucidated by comparison of experimental and computed 1H NMR chemical shifts. Results of biological assays are reported.  相似文献

Background

Natural products are well recognized as sources of drugs in several human ailments. In the present work, we carried out a preliminary screening of six natural compounds, xanthone V1 (1); 2-acetylfuro-1,4-naphthoquinone (2); physcion (3); bisvismiaquinone (4); vismiaquinone (5); 1,8-dihydroxy-3-geranyloxy-6-methylanthraquinone (6) against MiaPaCa-2 pancreatic and CCRF-CEM leukemia cells and their multidrug-resistant subline, CEM/ADR5000. Compounds 1 and 2 were then tested in several other cancer cells and their possible mode of action were investigated.

Methodology/Findings

The tested compounds were previously isolated from the Cameroonian medicinal plants Vismia laurentii (1, 3, 4, 5 and 6) and Newbouldia laevis (2). The preliminary cytotoxicity results allowed the selection of xanthone V1 and 2-acetylfuro-1,4-naphthoquinone, which were then tested on a panel of cancer cell lines. The study was also extended to the analysis of cell cycle distribution, apoptosis induction, caspase 3/7 activation and the anti-angiogenic properties of xanthone V1 and 2-acetylfuro-1,4-naphthoquinone. IC50 values around or below 4 µg/ml were obtained on 64.29% and 78.57% of the tested cancer cell lines for xanthone V1 and 2-acetylfuro-1,4-naphthoquinone, respectively. The most sensitive cell lines (IC50<1 µg/ml) were breast MCF-7 (to xanthone V1), cervix HeLa and Caski (to xanthone V1 and 2-acetylfuro-1,4-naphthoquinone), leukemia PF-382 and melanoma colo-38 (to 2-acetylfuro-1,4-naphthoquinone). The two compounds showed respectively, 65.8% and 59.6% inhibition of the growth of blood capillaries on the chorioallantoic membrane of quail eggs in the anti-angiogenic assay. Upon treatment with two fold IC50 and after 72 h, the two compounds induced cell cycle arrest in S-phase, and also significant apoptosis in CCRF-CEM leukemia cells. Caspase 3/7 was activated by xanthone V1.

Conclusions/Significance

The overall results of the present study provided evidence for the cytotoxicity of compounds xanthone V1 and 2-acetylfuro-1,4-naphthoquinone, and bring supportive data for future investigations that will lead to their use in cancer therapy.  相似文献
