Current models that describe the extension of fungal hyphae and development of a mycelium either do not describe the role of vesicles in hyphal extension or do not correctly describe the experimentally observed profile for distribution of vesicles along the hypha. The present work uses the n-tanks-in-series approach to develop a model for hyphal extension that describes the intracellular transport of nutrient to a sub-apical zone where vesicles are formed and then transported to the tip, where tip extension occurs. The model was calibrated using experimental data from the literature for the extension of reproductive aerial hyphae of three different fungi, and was able to describe different profiles involving acceleration and deceleration of the extension rate. A sensitivity analysis showed that the supply of nutrient to the sub-apical vesicle-producing zone is a key factor influencing the rate of extension of the hypha. Although this model was used to describe the extension of a single reproductive aerial hypha, the use of the n-tanks-in-series approach to representing the hypha means that the model has the flexibility to be extended to describe the growth of other types of hyphae and the branching of hyphae to form a complete mycelium.  相似文献

The rate of hyphal elongation and the number of branches per hypha were measured on short sporelings of Aspergillus nidulans growing at different rates. The rate of elongation was proportional to total length in unbranched and branched hyphae. At each growth rate, the number of branches per hypha increased with increasing length and gave approximately straight-line graphs when plotted against length. The average number of branches per unit of hyphal length was quite different for the various growth rates and increased in direct proportion to the growth rate. The results are interpreted to mean that (i) growing tips have a maximum rate at which they can elongate and which is reached at hyphal lengths characteristic of the particular growth rate and (ii) a new branch is formed when the capacity of the hypha to elongate exceeds that of the existing tips.  相似文献

Filamentous fungi like Aspergillus nidulans can effectively colonize their surroundings by the formation of new branches along the existing hyphae. While growth conditions, chemical perturbations, and mutations affecting branch formation have received great attention during the last decades, the mechanisms that regulates branching is still poorly understood. In this study, a possible relation between cell cycle progression and branching was studied by testing the effect of a nuclei distribution mutation, cell cycle inhibitors, and conditional cell cycle mutations in combination with tip-growth inhibitors and varying substrate concentrations on branch initiation. Formation of branches was blocked after inhibition of nuclear division, which was not caused by a reduced growth rate. In hyphae of a nuclei distribution mutant branching was severely reduced in anucleated hyphae whereas the number of branches per hyphal length was linearly correlated to the concentration of nuclei, in the nucleated hyphae. In wild type cells, branching intensity was increased when the tip extension was reduced, and reduced when growing on poor substrates. In these situations, the hyphal concentration of nuclei was maintained and it is suggested that branching is correlated to cell cycle progression in order to maintain a minimum required cytoplasmic volume per nucleus and to avoid the formation of anucleated hyphae in the absence of nuclear divisions. The presented results further suggest the hyphal diameter as a key point through which the hyphal element regulates its branching intensity in response to the surrounding substrate concentrations.  相似文献

Fluorescence microscopy was used to describe the distribution of nuclei and the organization of the microtubule network in hyphae of Pleurotus ostreatus. Dikaryotic hyphae of P. ostreatus N001 grow by tip extension with two closely spaced nuclei moving slowly forward with the growing hyphal tip. During vegetative growth of the hyphae, cytoplasmic microtubules are found as long filaments oriented longitudinally within fungal hyphae. When the apical cell reaches a length of approximately 150 μm, the two nuclei divide synchronously. Mitosis occurs in association with clamp connection formation, with one of the nuclei dividing in the hook of the developing clamp connection and the other in the main hypha. After mitosis, two daughter nuclei move forward to approximately the center of the apical cell, while the other two move backward to a central position in the subapical cell. Two septa are formed, one in the clamp and the other across the main axis of the hypha to delimit the apical cell. The use of fluorescence microscopy made it possible to examine the changes in the cytoplasmic microtubules, the configuration of the mitotic apparatus, the site of septation and the post-mitotic nuclear migrations during conjugate division in P. ostreatus dikaryotic hyphae.  相似文献

In the fungal pathogen Fusarium oxysporum, vegetative hyphal fusion triggers nuclear mitotic division in the invading hypha followed by migration of a nucleus into the receptor hypha and degradation of the resident nucleus. Here we examined the role of autophagy in fusion-induced nuclear degradation. A search of the F. oxysporum genome database for autophagy pathway components identified putative orthologs of 16 core autophagy-related (ATG) genes in yeast, including the ubiquitin-like protein Atg8, which is required for the formation of autophagosomal membranes. F. oxysporum Foatg8Δ mutants were generated in a strain harboring H1-cherry fluorescent protein (ChFP)-labeled nuclei to facilitate analysis of nuclear dynamics. The Foatg8Δ mutants did not show MDC-positive staining in contrast to the wild type and the FoATG8-complemented (cFoATG8) strain, suggesting that FoAtg8 is required for autophagy in F. oxysporum. The Foatg8Δ strains displayed reduced rates of hyphal growth, conidiation, and fusion, and were significantly attenuated in virulence on tomato plants and in the nonvertebrate animal host Galleria mellonella. In contrast to wild-type hyphae, which are almost exclusively composed of uninucleated hyphal compartments, the hyphae of the Foatg8Δ mutants contained a significant fraction of hyphal compartments with 2 or more nuclei. The increase in the number of nuclei per hyphal compartment was particularly evident after hyphal fusion events. Time-lapse microscopy analyses revealed abnormal mitotic patterns during vegetative growth in the Foatg8Δ mutants. Our results suggest that autophagy mediates nuclear degradation after hyphal fusion and has a general function in the control of nuclear distribution in F. oxysporum.  相似文献

Radioactive conidiospores of Aspergillus nidulans were prepared by growing a purine-requiring mutant with tritiated adenine. When these spores germinated in a nonradioactive medium, the dispersion of the original chromosome set could be followed by treating the hyphae with ribonuclease and preparing radioautograms. Germinating spores with four or eight nuclei contained two highly labeled nuclei and two or six nuclei with much less or no radioactivity. Successive mitotic divisions thus distributed the deoxyribonucleic acid (DNA) of the eight spore chromosomes among only two of the progeny nuclei. The two nuclei containing the original chromosome set were not dispersed at random along the linear hypha but were usually located near the growing tip. These results are compatible with the view that chromatids containing DNA strands of identical age segregate as a unit during mitosis. They further indicate that the mechanism which disperses newly formed nuclei in the growing hypha can distinguish between nuclei containing DNA strands of different ages.  相似文献

Summary The studies on the kinetics of nucleoid destruction reported here showed that destruction of chloroplast nucleoids (ct nucleoids) of male origin began to occur at about 30 minutes after mixing of male (mt) and female (mt+) gametes. The timing of initiation of the destruction differed among zygotes but usually occurred during 50–120 minutes after mixing. About 10 minutes was required for complete digestion of the ct nucleoids. UV irradiation on young zygotes or addition of an RNA-synthesis inhibitor, actinomycin D, to the incubation medium during the first 0–30 minutes after mixing almost completely inhibited the incorporation of3H uridine into the cell nuclei and the preferential destruction without inhibiting cell nuclear fusion. These results suggest that soon after mating,de novo RNA synthesis is concerned for the preferential destruction of ct nucleoids. To determine in which of the two cell nuclei in the zygotes the RNA is synthesized, each gamete (mt, mt+) was irradiated with UV and mated with unirradiated gametes of opposite mating type. This treatment of the male gametes had no effect on the incorporation of3H uridine into cell nuclei and the preferential destruction of ct nucleoids but UV irradiation of female gametes almost completely inhibited the incorporation of3H uridine into cell nuclei and the preferential destruction of ct nucleoids. Similar phenomena occurred in other crosses. The UV effect was photoreactivated in about 50% by white light, suggesting that the UV target is DNA. Thus, RNA synthesized in the cell nucleus of female origin soon after mating may be responsible for the preferential destruction of ct nucleoids of male origin  相似文献

The dimorphic yeast Yarrowia lipolytica forms true hyphae in a medium containing N-acetylglucosamine. We made a new finding that serum is a very effective inducer of hypha formation of Y. lipolytica: serum induced its hyphal growth very quickly compared to N-acetylglucosamine (4 h vs. 10 h). Osmotic and oxidative stresses (0.2 M NaCl and 20 mM H2O2) inhibited the hypha formation induced by N-acetylglucosamine, but did not suppress the hypha formation triggered by serum. Serum-specific morphological mutants, which formed hyphae in the N-acetylglucosamine medium but not in serum medium, could be isolated. These results suggest that the signal triggered by serum may be transduced through a different pathway, at least in part, from that used for the N-acetylglucosamine signal in Y. lipolytica.  相似文献

Matings between sister single spore lines of Coprinus disseminatusshowed a cryptic tetrapolar pattern. The two groups of matingsthat resulted in formation of mycelia with clamp connections(apparent dikaryons) differed in rate of nuclear penetrationduring mating. In one group penetration occurred at rates comparablewith nuclear migration in other species and in the other groupit was often extensive but at a rate similar to, or less than,the dikaryon growth-rate. No differences were detected betweenthese two groups in stability, colony extension rate, frequencyof clamp connections, proportions of true clamp connectionsand pseudoclamps, or number of nuclei per hyphal tip cell. Cytological studies and the isolation of hyphal tips showedthat both groups of apparent dikaryons were heterokaryotic di-or trikaryons. The di- and trikaryotic conditions co-existedin the same mycelium, but adjacent cells of individual branchingsystems usually contained equal numbers of nuclei. Within apparentdikaryons the number and kinds of nuclei per cell were similarin hyphae with clamp connections and those with simple septa.Treatments that prevented clamp connection formation did notalter the nuclear status of most of the hyphae. Irregularities in nuclear distribution were infrequent and mostwere associated with pseudoclamps. Forty per cent of nodes withprobable pseudoclamps yielded homokaryotic branches, which wereof either constituent mating type. There was some indicationthat irregularities in nuclear distribution could also occurduring divisions associated with simple septa.  相似文献

A micro-compartment culture method was devised in which a single hypha of Rhizopus stolonifer growing on an agar section traversed an antifungal non-diffusible barrier to another agar section; thus the local environment of the distal or proximal part of the hypha could be controlled independently. The responses in terms of hyphal extension of the test fungus to local application of amphotericin B in this culture system were estimated by using an automatic analysing system. After hyphae had traversed the barrier, distal application of amphotericin B caused no appreciable effect on the proximal hyphae. In contrast, proximal application of amphotericin B caused inhibition of the extension of distal hyphae. The reversal of polarized cytoplasmic streaming also occured during the inhibition of distal hyphal extension. The extents of inhibition of the distal hyphal extension and the cytoplasmic streaming were dependent upon the hyphal distance between the amphotericin B application site and the hyphal tip. These results show that the effect of an antifungal agent on a hypha depends on the region of the hypha exposed. Cytoplasmic streaming may play key role in the transmission of antifungal effects within a single hypha.  相似文献

Nuclear dynamics can vary widely between fungal species and between stages of development of fungal colonies. Here we compared nuclear dynamics and mitotic patterns between germlings and mature hyphae in Fusarium oxysporum. Using fluorescently labeled nuclei and live-cell imaging, we show that F. oxysporum is subject to a developmental transition from a uninucleate to a multinucleate state after completion of colony initiation. We observed a special type of hypha that exhibits a higher growth rate, possibly acting as a nutrient scout. The higher growth rate is associated with a higher nuclear count and mitotic waves involving 2 to 6 nuclei in the apical compartment. Further, we found that dormant nuclei of intercalary compartments can reenter the mitotic cycle, resulting in multinucleate compartments with up to 18 nuclei in a single compartment.  相似文献

Summary During the intercellular nuclear migration of the basidiomycete Schizophyllum commune cytoplasmic microtubules were frequently observed scattered in the hyphae around interphase nuclei and connected with a semiglobular structure at the poles of mitotic and postmitotic nuclei. Thus it seems possible that microtubules, which have been demonstrated to participate in the intracellular nuclear movements in the dikaryotic hyphae of the basidiomycetes, are also involved in the intercellular nuclear movements of these fungi. During hyphal fusion microtubules close to an interphase nucleus were connected with electron-dense structures. Intracellular hyphae and vesicles in mycorrhizal roots of yellow poplar were examined by electron microscopy. An investing layer of host wall material and cytoplasm enclosed the endophyte within the cells. Young developing hyphae contained abundant cytoplasm and few vacuoles. As hyphae matured, they became highly vacuolated and accumulated carbohydrate (glycogen) and lipid reserves. Mature vesicles were engorged with lipid droplets, possessed a trilaminate wall and were also enclosed by host wall material and cytoplasm. Compared with uninfected cells, infected cortical cells showed an increase in cytoplasmic volume, enlarged nuclei, and a reduction of starch reserves. Host nuclei were always proximal to the hyphae during hyphal development and deterioration. While other cytoplasmic components of infected and uninfected cells were comparable large electron-dense bodies occurred in vacuoles of most cells containing hyphae. Deterioration of intracellular hyphae occurred throughout the samples examined. Septa separated functional and degenerating portions of the hyphae. Hyphal deterioration involved degeneration and ultimate disappearance of fungal cytoplasm as well as collapse of hyphal walls. Based on these observations, the authors hypothesize that deterioration of the endophyte may release significant quantities of mineral nutrients, via hyphal contents, which are absorbed by the host.  相似文献

Streptomyces brasiliensis ATCC 23727 showed extensive sporulation when cultured in a liquid medium containing galactose and glutamic acid as carbon and nitrogen sources. Sporogenic hyphae formed under these conditions were morphologically similar and developmentally equivalent to aerial hyphae and metamorphosed into chains of spores by following a sequence of ultrastructural changes similar to that observed during growth on solid media. In addition, our electron microscopy study revealed two previously unrecognized aspects of hyphal development in streptomycetes: the formation of sporogenic hyphae was always preceded by changes in the structure of the nucleoid, and the sheath that characteristically covered these hyphae was not deposited coincidently with wall formation in the apical growing portion of the hypha.  相似文献

Morphological and cultural studies of Polyporus maculatissimus and P. portentosus are presented. Neolentiporus gen. nov. is proposed to accommodate P. maculatissimus , a species with a striking macromorphological similarity with the north temperate P. squamosus . The new genus is characterized by medium to large stipitate fruitbodies with a poroid hymenophore, circular to flabellate pilei with a scaly surface and an excentric or lateral stipe that sometimes is reduced to a short, robust umbo. The hyphal system is dimitic with clamped, irregularly thick-walled generative hyphae that do not react with cresyl-blue, and terminal, unbranched, thick-walled skeletal hyphae that are strongly metachromatic in cresyl-blue. Spores are cylindric, hyaline, thin-walled, inamyloid and binucleate. The sexuality is bipolar, the nuclear behavior is astatocoeno-cytic and the associated wood-rot is brown. The new genus is regarded as the poroid counterpart of the agaricoid Neolentinus. Polyporus portenrosus is included in Laetiporus on the basis of its yellowish fruitbodies, its soft, punky context, its hyphal system composed of simple septate generative hyphae and binding hyphae, its holocoenocytic nuclear behavior and its associated brown wood-rot. A new code symbol, i.e. '9s' is proposed to codify the presence, in cultures, of simple-septate generative hyphae with irregularly thickened walls.  相似文献

Compatible matings of Schizophyllum commune were performed on glucose-peptone-yeast extract medium appended with gelatin (18%) and studied by phase contrast microscopy during nuclear migration. Three categories of nuclear migration were observed. Type I involved a pulsatile jerking of the entire cytoplasmic contents of the hypha, changed direction periodically, and, during periods of cytoplasmic tranquility, the nucleus continued to migrate. Type II A migration of nuclei occurred in the absence of visible cytoplasmic flow. Both Type I and Type II A nuclear movements exceeded the hyphal growth rate by 10--20-fold. Type II B nuclear migration also occurred in the absence of visible cytoplasmic flow and the velocity was within the range of the hyphal growth rate. No specific organelles that were detected either directed or facilitated Type II A or Type II B nuclear movements. The nucleolus could either lead or trail relative to the direction of nuclear movement. Nuclear migration can be attributed to both cytoplasmic flow and self motility, depending upon the particular regions of the migration hypha in which it occurs.  相似文献

Pulse labeling of deoxyribonucleic acid (DNA) and radioautography have been used to study the effect of growth rate on nuclear replication in Aspergillus nidulans. When conidia were germinated in media supporting a fast growth rate, the radioactive pulse labeled either all of the nuclei in a cell or none of them. At slower growth rates, hyphae contained both labeled and unlabeled nuclei. Altering the growth rate thus changed nuclear replication from simultaneous to sequential. The time taken to duplicate the DNA in a nucleus, estimated from the ratio of labeled to total nuclei, remained constant at the different doubling times. The distribution of label showed that nuclei in the same hypha spent unequal times in both the postmitotic gap (G1) and the premitotic gap (G2) periods when grown at slow rates. These unequal G1 and G2 periods are considered to cause asynchrony. Once DNA synthesis was out of phase through growth on a poor medium, transferring the hypha to a rich medium did not resynchronize the nuclei. To interpret the data, two initiator mechanisms, one starting DNA synthesis and the other mitosis, are postulated to control nuclear replication in A. nidulans.  相似文献

Nuclei of multinucleate hyphal tip cells divided almost synchronously. Nuclear division was followed by multiple septation during which the elongated hyphal tip cell was divided into several, generally multinucleate, cells. Within the same hypha a distinct growth pattern was not observed with respect to number, length, and nuclear number of cells formed with each successive nuclear division-septation cycle.
