Although the parathyroid water-clear cell is very rare, it has clinical significance because of its association with parathyroid hyperplasia or adenoma. SAMP6, a substrain of senescence-accelerated mouse, was developed as an animal model for senile osteoporosis. We investigated the morphology of the parathyroid glands in SAMP6 and age-matched normal mouse SAMR1. The parathyroid water-clear cells, which contained numerous vacuoles and the crystalloid inclusions, were found in SAMP6 mice at 5, 8 and 12 months of age. It was noted that the number of water-clear cells increased with aging, which are fairly consistent with the change of the serum parathyroid hormone (PTH) level. We did not find any water-clear cells in the parathyroid glands of SAMR1 mice. The existence of water-clear cells may represent hyperfunction of the parathyroid glands in SAMP6.  相似文献

Mass of floccular substance was observed in the parathyroid glands of fetal, newborn and infantile golden hamsters. Mass composed of floccular substances was spherical with no limiting membrane around it. It was located near the nucleus and the Golgi area, but was also observed in the peripheral cytoplasm. The ultrastructure of the parathyroid glands of adult Japanese lizards (Takydromus tachydromoides) in the spring and summer season was examined. The parenchyma of the gland consists of chief cells arranged in cords or solid masses. Many chief cells contain numerous free ribosomes and mitochondria, well-developed Golgi complexes, a few lysosome-like bodies, some multivesicular bodies and relatively numerous lipid droplets. The endoplasmic reticulum is mainly smooth-surfaced. Cisternae of the rough endoplasmic reticulum are distributed randomly in the cytoplasm. Small coated vesicles of 700-800 Å in diameter are found occasionally in the cytoplasm, especially in the Golgi region. The chief cells contain occasional secretory granules of 150-300 nm in diameter that are distributed randomly in the cytoplasm and lie close to the plasma membrane. Electron dense material similar to the contents of the secretory granules is observed in the enlarged intercellular space. These findings suggest that the secretory granules may be discharged into the intercellular space by an eruptocrine type of secretion. Coated vesicles (invaginations) connected to the plasma membrane and smooth vesicles arranged in a row near the plasma membrane are observed. It is suggested that such coated vesicles may take up extracellular proteins. The accumulation of microfilaments is sometimes recognized. Morphological evidence of synthetic and secretory activities in the chief cells suggests active parathyroid function in the Japanese lizard during the spring and summer season.  相似文献

Summary The fine structure of the interstitial gland of the rat ovary was studied at estrus and on Days 4, 6, 10, 14 and 18 of pregnancy. At estrus, ovarian interstitial cells have small nuclei with dense irregular clumps of heterochromatin. Mitochondria are small and rod-shaped and have predominantely lamellar cristae. Numerous osmiophilic lipid droplets are present. At Days 4 and 6, nuclear heterochromatin is reduced, and nucleoli are larger and complex. Mitochondria are enlarged and often bizarre-shaped and have tubular cristae. Golgi and smooth endoplasmic reticulum are more conspicuous. At Day 10, prominent ultrastructural features include nuclei with conspicuous heterochromatin, smaller mitochondria with both lamellar and tubular cristae, numerous ribosomes and lipid droplets with decreased osmiophilia. At Days 14 and 18, nuclei have increased heterochromatin, mitochondria are small and have lamellated cristae and an increase in the size and number of lipid droplets occurs. These observations suggest that steroidogenic activity of interstitial cells is highest during the first half of pregnancy and regresses during the last half. It is suggested that the interstitial gland is an important ovarian source of pregnancy hormone(s) during the first half of gestation and that LH may modulate steroidogenic activity in this ovarian component.  相似文献

Summary Some features of the structure and histochemistry of the rat parathyroid gland have been studied using a variety of histological and histochemical staining methods. The gland is composed of a compact parenchyma consisting essentially of a single cell type. The parenchyma is encapsulated by a connective tissue layer which extends into it and holds abundant branches of blood vessels. In the parenchymal cells four types of protein granules are demonstrated by the tetrazonium, DDD-diazo blue B, alkaline tetrazolium and HNAH-diazo blue B methods respectively. The DDD-diazo blue B reactive granules are considered to relate to the secretory activity of the parenchymal cells in view of their morphology and recent biochemical data on the nature of parathyroid hormone. The cytophysiological significance of the other three types of protein granules was discussed on the basis of their morphological features. Besides protein granules the parenchymal cells contain glycogen, RNA and DNA, and are reactive for alkaline phosphatase. The quantity of these substances and the enzyme activity seem to reflect to a certain extent different states of cellular activity. The parenchymal cells show scanty sudanophilic lipid content in paraffin sections and are devoid of basophilic mucopolysaccharides. Further, the cells are negative for acid phosphatase. In the connective tissue stroma of the gland the vascular endothelial cells show high alkaline phosphatase activity, and a hypothesis is presented as to the physiological significance of this high enzyme activity.  相似文献

The ultrastructure of the parathyroid glands of fetal and pregnant golden hamsters exposed to 5-gravity environment was studied. In the centrifuged fetal animals the Golgi complexes associated with some prosecretory granules were significantly increased compared with those of the control fetal animals, and several secretory granules were located in a peripheral position just beneath the plasma membrane. In the centrifuged pregnant animals the Golgi complexes associated with numerous prosecretory granules were significantly increased, cisternae of the granular endoplasmic reticulum appeared to be increased compared with those of the control pregnant animals, many secretory granules were located in the peripheral cytoplasm and several granules were situated close to the plasma membrane. These findings suggest that in the parathyroid glands of the fetal golden hamsters as well as the pregnant animals the synthesis and release of parathyroid hormone may be stimulated in response to a hypergravity environment.  相似文献

Summary The parathyroid glands of young male rats given 150 ppm fluoride in their drinking water for 10 weeks were examined by transmission electron microscopy. As a result of fluoride ingestion, the parathyroid chief cells of the experimental animals accumulated glycogen in excess of that seen in control animals given distilled drinking water for the same time period. In the majority of active chief cells, glycogen granules were diffusely spread throughout the cytoplasm as single granules or in small deposits. Large aggregations of glycogen granules were also seen within intercellular spaces. Accompanying the increase in glycogen was a rise in the number and development of the organelles associated with protein synthesis and secretion. The accumulation of glycogen is similar to that in hyperparathyroidism caused by chronic stimulation and prolonged secretory activity of the parathyroid gland. The results of this study suggest that increased amounts of glycogen occur in hyperactive chief cells of the parathyroid in response to the ingestion of large doses of fluoride.  相似文献

Summary A phenylethanolamine N-methyltransferase-(PNMT)-immunoreactivity, present without the other catecholamine-synthesizing enzymes tyrosine hydroxylase (TH) and dopamine--hydroxylase (DBH), has been previously detected in the central nervous system and in endocrine cells of the islets of Langerhans and the pituitary intermediate lobe of the rat. In the present study a similar PNMT-like immunoreactivity is demonstrated in the rat parathyroid gland. The immunoreactivity was distinctly localized to the cell periphery, and present in all glandular cells. The thyroid gland was negative. In the parathyroid TH- and DBH-immunoreactivity was seen only in vascular nerve fibers; no glandular cells were stained.The functional significance of the PNMT-like immunoreactivity is not known. The absence of TH- and DBH-immunoreactivity and the low level of adrenaline detected in the parathyroid, and the peripheral localization of the immunoreactivity may indicate an alternative enzyme function or the detection of an immunologically related protein common to pancreatic, pituitary and parathyroid endocrine cells.  相似文献
