Agaricus blazei Murrill, a native mushroom in Brazil, has been widely consumed in different parts of the world due to its medicinal power. Its anticarcinogenic activity has been shown in experimental animals, and antimutagenic activity has been demonstrated only in Salmonella. In this work, the mutagenic and antimutagenic activities of mushroom teas of strains AB96/07, AB96/09 and AB97/11 were evaluated in Chinese hamster V79 cells, using the comet assay and the micronucleus test. The cells were treated with three different concentrations (0.05, 0.1 and 0.15) of teas prepared from a 2.5% aqueous solution, under three different temperatures: (1) room (20-25 degrees C); (2) ice-cold (2-8 degrees C); and (3) warm (60 degrees C). The teas were applied in co-, pre- and post-treatments in combination with the mutagen methyl methanesulfonate (MMS; 1.6x10(-4) and 4x10(-4)M). The duration of the treatment was 1h in the comet assay and 2h in the micronucleus test. The results showed that the mushroom was not mutagenic itself. Nevertheless, the mushroom is an efficient antimutagen against the induction of micronuclei by MMS in all concentrations and preparations tested. The observed reductions in the frequencies of micronuclei ranged from 61.5 (room temperature 0.1% tea in post-treatment) to 110.3% (co-treatment with warm and ice-cold 0.15% tea). In the comet assay, the antimutagenic activity was detected only when the cells were pre-treated with the following teas: warm 0.1 and 0.15%, room temperature 0.05% and ice-cold 0.1%. The results indicate that the mushroom A. blazei extracts are antimutagenic when tested in V79 cells.  相似文献

报道了溶壁酶系统、酶浓度、不同菌龄、脱壁促进剂、渗透压稳定剂和酶解温度对姬松茸原生质体释放率及不同再生培养基、渗稳剂种类、菌丝酶解时间和单双层平板对原生质体再生的影响。结果表明 ,菌龄为3~ 5d的菌丝以 1.5 %溶壁酶、0 .5 %蜗牛酶和 0 .5 %纤维素酶组成的酶系统在 30℃以KCl为渗透压稳定剂时 ,形成率为 1.4~ 1.5ⅹ 10 7/mL酶液 ;以蔗糖为渗透压稳定剂 ,菌丝酶解 1.5~ 3h ,以SMY和MYP为再生培养基 ,姬松茸原生质体再生率为 1.1‰~ 1.3‰。  相似文献

The influence of beta-carotene on the clastogenicity of the indirect-acting mutagen cyclophosphamide (CPA) was investigated in mice, in vivo, for the induction of chromosome aberrations in bone marrow cells (BM). beta-Carotene (0.5, 1.0, 2.0, 5.0, 10, 25, 50, 100 and 200 mg/kg) was administered by gavage for 5 consecutive days. 4 h after the last treatment with beta-carotene, the mice were injected intraperitoneally with CPA, and the BM cells were fixed after 16, 24 and 32 h for the evaluation of the frequency of chromosome aberrations. The results showed that beta-carotene was effective in reducing chromosomal damage induced by CPA with the increase of its concentration up to a level after which this effect was not observed. This anticlastogenicity was better detected when the cells were fixed at 32 h, although a tendency in reducing the CPA clastogenicity was already observed at 16 and 24 h. Our results suggest that beta-carotene provides significant protection against the genotoxicity of CPA, although no dose-effect relationship on the frequencies of cells with chromosomal aberrations was observed.  相似文献

The effects of Agaricus blazei intake on the lipid profile of animals fed a hypercholesterolemic diet were evaluated. Thirty-two female Fisher rats were divided into four groups and given the standard AIN-93 M diet (C), this diet?+?1 % A. blazei (CAb), a hypercholesterolemic diet with 25 % soybean oil and 1 % cholesterol (H) or this diet?+?1 % A. blazei (HAb) for 6 weeks. Food intake, weight gain, liver and serum lipid profiles, activity of aminotransferases [alanine aminotransferase (ALT) and aspartate aminotransferase (AST)], and creatinine and urea levels as well as abdominal fat weight were measured. Histological analysis of kidney and liver tissue was also performed. The HAb group had a higher food intake, but a lower weight gain as compared to group H. This resulted in a significant decrease in abdominal fat weight, to values close to those of groups C and CAb. Supplementing the hypercholesterolemic diet with A. blazei promoted a significant reduction in total and non-HDL cholesterol, as well as in the atherogenic index, as compared to group H, and this effect was more pronounced in the serum. There was no hepatotoxic effect caused by the supplementation of the diets with the mushroom. We conclude that in our experimental model and in the concentration used, A. blazei was effective in improving the lipid profile of the animals.  相似文献

The medicinal mushroom Agaricus blazei produced high amounts of laccase (up to 5,000 units l–1) in a complex, agitated liquid medium based on tomato juice, while only traces of the enzyme (<100 units l–1) were detected in synthetic glucose-based medium. Purification of the enzyme required three chromatographic steps, including anion and cation exchanging. A. blazei laccase was expressed as a single protein with a molecular mass of 66 kDa and an isoelectric point of 4.0. Spectroscopic analysis of the purified enzyme confirmed that it belongs to the blue copper oxidases. The enzymes pH optimum for 2,6-dimethoxyphenol (DMP) and syringaldazine was pH 5.5; but for 2,2-azino-bis(3-ethylthiazoline-6-sulfonate) (ABTS) no distinct pH optimum was observed (highest activity at the lowest pH tested). Purified laccase was stable at 20°C, pH 7.0 and pH 3.0, but rapidly lost its activity at 40°C or pH 10. Sodium chloride strongly inhibited the enzyme activity, although the inhibition was completely reversible. The following kinetic constants were determined (Km, kcat): 63 M, 21 s–1 for ABTS, 4 M, 5 s–1 for syringaldazine, 1,026 M, 15 s–1 for DMP and 4307 M, 159 s–1 for guaiacol. The results show that—in addition to the wood-colonizing white-rot fungi—the typical litter-decomposing basidiomycetes can also produce high titers of laccase in suitable liquid media.  相似文献

巴氏蘑菇(姬松茸)原菌株J1和辐射突变株J3子实体粗多糖经水提、醇沉、Sevage法去除蛋白,透析得精多糖ABJ-1和ABJ-3;经紫外扫描和凝胶色谱法检测表明,ABJ-1和ABJ-3均具有较高的纯度。以苯酚-硫酸法测得了ABJ-1和ABJ-3的糖含量分别为47%和76.68%,凯氏定氮法测定其含氮量分别为5.77%和21%。,以凝胶过滤法测定了其分子量均在4万左右。完全酸水解后气相色谱分析显示ABJ-1含有甘露糖、葡萄糖、半乳糖等单糖,ABJ-3含有大量的甘露糖及少量的葡萄糖等单糖。红外光谱和Smith降解后气相色谱分析显示ABJ-1和ABJ-3糖基连接键均以a,1-4糖苷键为主。  相似文献

Anti-viral activities of Agaricus blazei Murill were investigated. The water extracts of the cultured mycelia and fruiting bodies were fractionated with different concentrations of ethanol. To several viruses which have cytopathic effects (CPE) on VERO cells, inhibition of these effects by the ethanol fractions was tested. Strong inhibition of CPE induced by western equine encephalitis (WEE) virus was observed in the mycelial fractions but not those of fruiting bodies.  相似文献

Basidiocarp of Agaricus blazei (=Agaricus brasiliensis; =Agaricus subrufescens) is used as teas or capsules due to its antineoplastic effect but there are few reports of using mycelium for this purpose. The objective of this study was to evaluate the antineoplastic activity on sarcoma 180 cells implanted in mice of two forms of preparation of the mycelium from two A. blazei strains grown in culture medium with different concentrations of isolated soy protein. Mycelia were grown in Pontecorvo medium with different concentrations of isolated soybean protein (ISP). Mycelial hot water extract, moistened mycelial powder, hot water extract of green tea, Ifosfamida® (ifosfamide drug), and saline solution were administered daily by gavage in mice with sarcoma 180 cells to evaluate antineoplastic activity. It was concluded that antineoplastic activity was the same for both strains, except when used as moistened mycelial powder, which rules out the use of mycelial powder in capsules. Mycelial hot water extract had high antineoplastic activity with lower metabolic demand on the spleen and maintenance of normal blood parameters. Mycelial growth in different ISP concentrations had the same antineoplastic activity. Also the vegetative mycelium was as effective as the basidiocarp for sarcoma 180 tumor inhibition. The effects of crude extracts of the mushroom Agaricus blazei Murrill (Agaricaceae) on both DNA damage and placental form glutathione S-transferase (GST-P)-positive liver foci induced by diethylnitrosamine (DEN) were investigated. Six groups of adult male Wistar rats were used. For two weeks, animals of groups 3 to 6 were treated with three aqueous solutions of A. blazei (mean dry weight of solids being 1.2, 5.6, 11.5 and 11.5 mg/ml, respectively). After this period, groups 2 to 5 were given a single ip injection 200 mg/kg DEN and groups 1 and 6 were treated with 0.9% NaCl. All animals were subjected to 70% partial hepatectomy at week five and sacrificed 4, 24 and 48 h or 8 weeks after DEN or 0.9% NaCl treatments (10th week after the beginning of the experiment). The alkaline comet assay and GST-P-positive liver foci development were used to evaluate the influence of the mushroom extracts on liver cell DNA damage and on the initiation of liver carcinogenesis, respectively. Previous treatment with the highest concentration of A. blazei (11.5 mg/ml) significantly reduced DNA damage, indicating a protective effect against DEN-induced liver cytotoxicity/genotoxicity. However, the same dose of mushroom extract significantly increased the number of GST-P-positive liver foci.  相似文献

以姬松茸原生质体为诱变材料,用不同诱变剂紫外线(UV),钴60(Co^60),亚硝基胍(NTG)进行多次反复诱变处理,获得2株活性多糖产量较高,遗传稳定的变异株C811/N516。与出发菌株各项发酵指标比较,突变株其他各发酵特性变化不明显,而单位发酵液活性多糖含量分别提高260%和300%以上。  相似文献

The protective effects of carnosine as a natural dipeptide were investigated in mouse bone marrow cells against genotoxicity induced by cyclophosphamide. Mice were injected with solutions of carnosine at three different doses (10, 50 and 100?mg kg(-1) bw) for five consecutive days. On the fifth day of treatment, mice were injected cyclophosphamide and killed after 24?h. The frequency of micronuclei in polychromatic erythrocytes and the ratio of polychromatic erythrocyte/polychromatic erythrocyte?+?normochromatic erythrocyte [PCE/(PCE?+?NCE)] were evaluated by May-Grunwald/Giemsa staining. Histopathology of bone marrow was examined in mice treated with cyclophosphamide and carnosine. Carnosine significantly reduced micronucleated polychromatic erythrocytes (MnPCEs) induced by cyclophosphamide at all three doses. Carnosine at dose of 100?mg kg(-1) bw reduced MnPCEs 3.76-fold and completely normalized the PCE/(PCE?+?NCE) ratio. Administration of carnosine inhibited bone marrow toxicity induced by cyclophosphamide. It appeared that carnosine with protective activity reduced the oxidative stress and genotoxicity induced by cyclophosphamide in bone marrow cells of mice. 对发酵法生产富铁姬松茸菌丝体进行了产业化研究.结果表明,姬松茸的耐铁能力和富铁能力都很强,在铁浓度为0.1~1.5g/L的液体培养基上,菌丝均能生长,当铁浓度超过1.5g/L 时,菌丝生长受到一定影响;采用液体深层培养,最佳的铁源为FeSO4·7H2O ,最佳铁添加量为0.4~0.5g/L.此时,姬松茸菌丝体的生物量得率和富铁率达到最高,同时,菌丝体含铁量和多糖含量也较高.另外,扩大到10t罐进行工业化生产,富铁姬松茸干菌粉得率达到2.82%,菌丝体含铁量可达14.77mg/g.姬松茸菌丝体产品产量高,质量稳定, 已推向市场,提供给一些企业,初步实现了产业化.  相似文献

The growth-promoting effect of the thermophilic fungus Scytalidium thermophilum in mushroom compost on the mycelium of the edible mushroom Agaricus bisporus was investigated. Results obtained by others were confirmed by showing that S. thermophilum leads to an increased hyphal extension rate of the mushroom mycelium. However, it was demonstrated that hyphal extension rates were not clearly related to mushroom biomass increase rates. A number of experiments pointed strongly towards CO2 as the determinant of hyphal extension rates. In compost, CO2 is produced mainly by thermophilic fungi. Several experiments did not reveal any other specific compound produced by S. thermophilum that increases the hyphal extension rate of the mushroom mycelium.  相似文献
