Relationships between the predispositions to pendulum movements (PMs), catalepsy, and audiogenic epilepsy in the course of breeding for "nervousness" and freezing have been studied. A positive genetic relationship between PMs, catalepsy, and audiogenic epilepsy has been demonstrated. Eighty to ninety percent of rats selected for enhanced PMs (strain PM+) exhibit predisposition to audiogenic seizures, whereas this frequency in the PM-free strain (PM-) is 10-40%.  相似文献

Non-inbred Wistar rats were bred for predisposition to catalepsy for 14 generations. The percentage of cataleptic rats, beginning from the F1, was about 50%, while in the control population it was about 9%. This, together with the data obtained after comparison of the proportion of cataleptic animals in the progeny from homogeneous crossings between rats of normal and cataleptic phenotypes from the group selected for catalepsy (16 and 48%, respectively), makes one suppose predisposition to catalepsy to be an oligogenic character. The later onset of stereotype-like reactions to administrations of methylphenidate, and their longer persistence in cataleptic animals points to inertness of dopaminergic systems. At the same time, the increased frequency of "hyperactivity"-like reactions to methylphenidate, as well as higher arterial pressure and lower frequency of defecations seem to reflect an increased excitability of noradrenergic brain systems in rats predisposed to catalepsy.  相似文献

Typological characteristics of behavior were studied in rats bred for enhancement (PM+) and absence (PM-) of pendulum movements. Excitement in different test situations was manifest in PM+ rats, whereas passive defensive reactions were characteristic of PM- rats. Increased excitability of PM+ rats was expressed in their greater predisposition to audiogenic epilepsy (83% in PM+ versus 40% in PM- rats). On the contrary, PM- rats were found to be more prone to freezing (61% in PV- versus 11% in PM+). In PM+ rats, noradrenaline and serotonin contents were decreased in hypothalamus (as compared to PM- and control Wistar stain), whereas in PM- rats, serotonin content was increased in striatum, hypothalamus and midbrain as compared to control strains.  相似文献

Mongolian gerbils (Meriones unguiculatus) have an incomplete circle of Willis (CoW), as a result of which approximately 30-40% of these animals develop focal cerebral ischemia after unilateral carotid occlusion (UCO). There are four types of patterns of the anterior and posterior communicating arteries (ACoAs and PCoAs, respectively) of the CoW and they determine the severity of the ischemic symptoms. We used 398 gerbils from five generations, including a selectively bred ischemia-prone group, to investigate post-UCO ischemic symptoms and possible correlations of ACoA and PCoA patterns between parents and their progeny. We observed that if the parents had complete ACoAs, their progeny also had complete ACoAs, and we found significant differences when the parents' ACoAs were incomplete: in 60.4% of offspring the type of ACoA was consistent with that of the mother and in 48.2% it was consistent with that of the father. The severity of the neurological symptoms after UCO was significantly related to the patterns of the ACoAs when PCoAs were absent. The proportion of UCO ischemia in gerbils with incomplete ACoAs was significantly higher than in those with complete ACoAs. After selectively breeding five generations, the proportion of UCO ischemia increased from 40% in the F1 animals to 75% in the F5 animals. Our results suggest that variations in the CoW are genetic and demonstrate that we successfully established an ischemia-prone group of gerbils.  相似文献

The hybrids between Krushinsky-Molodkina (KM) inbred strain, selected for high predisposition to audiogenic epilepsy (AE), and Wistar rats non-prone to audiogenic seizure were the initial population for selection. Rats were selected for the trait "the absence of audiogenic seizure proneness". The creation of such strain in which the significant proportion of animals develop no AE in response to sound and share partly the genetic background of the KM strain is very important for the correct use of RV strain as the laboratory model of seizure states. As alleles which determine the AE proneness are recessive the selection for the "opposite" trait proceeds necessarily slow.  相似文献

Systematic studies of cloned animals generated from adult somatic cell nuclei are critical in assessing the utility of somatic cell cloning in various applications, including the safety of food products from cloned animals and their offspring. Previously, we compared somatic cell derived cloned pigs with naturally bred control pigs on a series of physiological and genetic parameters. We have extended our studies to the F1 progeny of these clones to see whether these phenotypic differences are transmitted to the next generation. There were no differences in the average litter size between litters from cloned gilts and naturally bred controls (7.78 +/- 2.6 and 7.40 +/- 3.0, respectively; mean +/- SD) or in the degree of litter size variation (coefficients of variation of 33.4% and 40.5% for litters of clones and controls, respectively). Similarly there were no statistical differences between sex ratios of cloned litters (51-49%, M:F) and control litters (59-41%, M:F). Blood profiles between cloned pigs, control pigs, and their progeny were compared at two time points (i.e., 15 and 27 weeks) to quantify the effect of cloning on various blood parameters and their transmission to the next generation. Although the range of values for all traits overlapped between different classes, the variability between all the traits in F1 progeny of clones and the control pigs was similar at 15 and 27 weeks, with one exception. Combined, our data and previous results in mice strongly support the hypothesis that offspring of clones are similar to offspring of naturally bred animals, and as such there should not be any increased risks associated with consumption of products from these animals.  相似文献

Predispositions to pendulum movements (PMs) and catalepsy was studied in rat strains selected for predisposition to catalepsy and for enhanced or suppressed PMs. Positive correlation between PMs and catalepsy at earlier stages of selection changed to negative at later stages. Based on these data, it was hypothesized that the genes that determine the predisposition to catalepsy affect the expression of the gene determining PMs in such a way that different PM alleles become dominant in the cases of moderate and strong effects of the catalepsy genes.  相似文献

Loss of genetic diversity and increased population differentiation from source populations are common problems associated with translocation programmes established from captive-bred stock or a small number of founders. The bridled nailtail wallaby is one of the most endangered macropods in Australia, having been reduced to a single remnant population in the last 100 years. A translocated population of bridled nailtail wallabies was established using animals sourced directly from the remnant population (wild-released) as well as the progeny of animals collected for a captive breeding programme (captive-bred). The aims of this study were to compare genetic diversity among released animals and their wild-born progeny to genetic diversity observed in the remnant population, and to monitor changes in genetic diversity over time as more animals were released into the population. Heterozygosity did not differ between the translocated and remnant population; however, allelic diversity was significantly reduced across all released animals and their wild-born progeny. Animals bred in captivity and their wild-born progeny were also significantly differentiated from the source population after just four generations. Wild-released animals, however, were representative of the source population and several alleles were unique to this group. Both heterozygosity and allelic diversity among translocated animals decreased over time with the additional release of captive-bred animals, as no new genetic stock was added to the population. Captive breeding programmes can provide large numbers of animals for release, but this study highlights the importance of sourcing animals directly from remnant populations in order to maintain genetic diversity and minimise genetic drift.  相似文献

Some changes in the brain serotonergic system were found in rats bred for predisposition to catalepsy, and in those bred for its absence. The genetic predisposition for catalepsy was found to be characterized by an increased tryptophan hydroxylase activity in the striatum, and an increased serotonin content in the midbrain. No changes in 5-hydroxyindoleacetic acid level were found. A selection for predisposition to catalepsy turned out to entail a decrease in the sensitivity of postsynaptic serotonin receptors as estimated by the "head twitch" test after 5-hydroxytryptophan administration, while a selection for the absence of catalepsy increased the sensitivity of serotonin receptors.  相似文献

The rat brain thermal fields were studied using the thermoencephaloscopic technique in three experimental conditions: the genetic catalepsy (GC rat strain), cataleptic phase of an audiogenic epileptic seizure (Krushinski?-Molodkina strain), and pharmacological catalepsy produced by haloperidol injection (Wistar rats). Irrespective of the experimental conditions, the state of catalepsy, accompanied by a decrease in the muscle tone and inhibition of motor reactions, was characterized by total asymmetric cooling of the brain cortex with the dominance of the right hemisphere. Temperature difference between the parieto-occipital areas of the right and left hemispheres reached 0.3-0.6 degree C.  相似文献

Epilepsy is one of the most common but genetically complex neurological disorders in humans. Identifying animal models that recapitulate human epilepsies is important for pharmacological studies of anticonvulsants, dissection of molecular and biochemical pathogenesis of epilepsy, and discovery of epilepsy susceptibility genes. We discovered that the PL/J inbred mouse strain is susceptible to handling- and rhythmic tossing–induced seizure. The tonic–clonic and generalized seizures observed after induction were accompanied by abnormal EEGs, similar to seizures observed in EL and SWXL-4 mice. PL/J mice also had an extremely low threshold to electroconvulsive seizures compared to other strains and showed variable sensitivity to pentylenetetrazole-induced seizures. Gross neurostructural abnormalities were not found in PL/J mice. Crosses with the seizure-resistant C57BL/6 J strain revealed semidominant inheritance of the rhythmic tossing seizure trait with low penetrance. F2 progeny indicated that the genetic inheritance of seizure susceptibility in PL/J is non-Mendelian. We crossed DBA/2 J mice, which are resistant to rhythmic tossing seizure but susceptible to audiogenic seizures, to PL/J. We found that seizure penetrance in (DBA/2 J × PL/J)F1 mice was similar to the penetrance in (C57BL/6 J × PL/J)F1 mice but the severity and frequency of seizure were higher in (DBA/2 J × PL/J)F1 mice. The PL/J strain serves as an interesting new model for studying the genetics, neurobiology, and pharmacology of epilepsy.  相似文献

The role of brain and liver mitochondria at epileptic seizure was studied on Krushinsky-Molodkina (KM) rats which respond to sound with an intensive epileptic seizure (audiogenic epilepsy). We didn't find significant changes in respiration rats of brain and liver mitochondria of KM and control rats; however the efficiency of АТР synthesis in the KM rat mitochondria was 10% lower. In rats with audiogenic epilepsy the concentration of oxidative stress marker malondialdehyde in mitochondria of the brain (but not liver) was 2-fold higher than that in the control rats. The rate of H2O2 generation in brain mitochondria of КМ rats was twofold higher than in the control animals when using NAD-dependent substrates. This difference was less pronounced in liver mitochondria. In KM rats, the activity of mitochondrial ATP-dependent potassium channel was lower than in liver mitochondria of control rats. The comparative study of the mitochondria ability to retain calcium ions revealed that in the case of using the complex I and complex II substrates, permeability transition pore is easier to trigger in brain and liver mitochondria of KM and КМs rats than in the control ones. The role of the changes in the energetic, oxidative, and ionic exchange in the mechanism of audiogenic epilepsy generation in rats and the possible correction of the epilepsy seizures are discussed.  相似文献

Approximately 5% of pigs produced in Australia is believed to be traded at livestock sales. Interviews and focus group discussions were conducted with producers (106 and 30 producers, respectively), who traded pigs at livestock sales. The purpose of the study was to gather information on how producers identified their pigs in order to evaluate how these practices may impact the ability to trace pig movements in the event of an emergency animal disease outbreak or food safety hazard. Results were analyzed according to herd size (0 to 150 sows, 150+ sows) and location (peri-urban, regional) as prior studies suggested a higher biosecurity risk among smaller farms and due to perceptions that peri-urban farms pose additional risk. Most producers (91.5%) had less than 150 sows and a high proportion (70.8%) resided in regional areas compared with only 29.2% residing in peri-urban areas. A higher proportion of large-scale producers identified their pigs than small-scale producers. A third of small-scale producers reported not identifying breeding stock and most did not identify progeny. The most common forms of on-farm identification used were ear tags for breeding stock and ear notches for progeny. Producers identified breeding stock to assist with mating management and genetic improvement. Ear notches were used to determine the litter of origin of progeny. All large-scale producers owned a registered swine brand and used the official body tattoo for post-farm-gate identification. However, approximately 15% of small-scale producers did not own a registered swine brand, and an additional 8% did not identify their pigs post-farm-gate. Producers were satisfied with tattoos as a methodology for post-farm-gate identification of pigs and considered other methodologies cost-prohibitive. However, variations in the maintenance of the branding equipment, the type of ink used and the time of tattoo application in relation to the animal sale were highlighted during focus group discussions. These results suggest that there is a need for education and extension activities, especially among small-scale pig producers, regarding the benefits of identifying animals on-farm. In addition, increased awareness of the traceability legislation that exists in Australia to meet the National Performance Standards for Livestock Traceability in this country is required.  相似文献

To investigate the basis of the sperm abnormality assay, studies have been made of the frequency of sperm abnormalities in the genital tracts of the progeny of irradiated males. Male C57BL/6 mice were irradiated (75-600 rad X-rays to the testes) and were then bred in the pre-sterile period to untreated C57BL/6 females. The sperm of their male progeny were examined for the frequency of sperm abnormalities. Variant males with clearly elevated levels of sperm abnormalities were more frequently seen amongst the progeny of irradiated fathers than with the progeny of sham-irradiated controls (10 in 170 compared with 2 in 188; P less than 0.02). Although no clear dose-response relation could be discerned with the number of animals studied, similar differences were observed with irradiated male SWR, C3H/He in inbred crosses and with C57BL/6 in hybrid crosses with C3H/He females. In contrast, matings of males made at longer times following irradiation did not lead to a significant increase in the number of affected progeny for the number tested. Breeding experiments with the affected F1 males showed that the sperm morphology defect could be transmitted in 7 of 19 cases. Parallel cytogenetic studies showed that 3 of the initial affected progeny had detectable reciprocal translocations and that in 2 of these cases the translocation was transmitted with the sperm defect. The studies thus showed that radiation can induce mutations that affect the levels of sperm abnormalities and that these mutations can be associated with reciprocal translocations.  相似文献

The levels of dopamine (DA) was determined by intracerebral microdialysis in vivo in KM rats selected for high audiogenic epilepsy, and in Wistar rats selected for nonsusceptibility to loud sound. The basal level of dopamine was 25% higher in the KM rats (P < 0.05). A single amphetamine injection (1 mg/kg body weight, intraperitoneously) caused a significant increase in the DA basal level up to 250-260% in animals of both genotypes. However, in Wistar rats, the level of DA reached maximum as soon as 20 min after amphetamine administration, whereas in KM rats, this happened only after 120 min. After a single injection of the antagonist of D2 and D3 dopamine receptors raclopride (1.2 mg/kg of body weight, intraperitoneously), an increase in the level of DA was similar in amplitude in rats of both genotypes (up to about 210%); however, this occurred 20-30 and 100 min after raclopride administration to Wistar and KM rats, respectively. This evidence suggests that the genetic defect of KM rats, namely, the high level of audiogenic epilepsy, is caused by abnormalities of the neuromediator brain systems and presumably accompanied by the regulatory gene dysfunction.  相似文献

Physiological, biochemical and genetic studies of audiogenic epilepsy in Krushinsky-Molodkina (KM) rat strain are analyzed. In brief, the history of KM strain selection and its current state are reviewed. Physiological mechanisms of the typical audiogenic seizure development and the audiogenic myoclonus (audiogenic kindling phenomena) are considered. Minimum coancestry mating with a maximum of one offspring per mating pair (MC1) is compared with random mating schemes for populations with overlapping generations. Optimum contribution selection is used, whereby ΔF is restricted. For schemes with ΔF restricted to 0.25% per year, 256 animals born per year and heritability of 0.25, genetic gain increased with 18% compared with random mating. The effect of MC1 on genetic gain decreased for larger schemes and schemes with a less stringent restriction on inbreeding. Breeding schemes hardly changed when omitting the iteration on the generation interval to find an optimum distribution of parents over age-classes, which saves computer time, but inbreeding and genetic merit fluctuated more before the schemes had reached a steady-state. When bulls were progeny tested, these progeny tested bulls were selected instead of the young bulls, which led to increased generation intervals, increased selection intensity of bulls and increased genetic gain (35% compared to a scheme without progeny testing for random mating). The effect of MC1 decreased for schemes with progeny testing. MC1 mating increased genetic gain from 11–18% for overlapping and 1–4% for discrete generations, when comparing schemes with similar genetic gain and size.  相似文献
