Host specificity tests carried out in the laboratory in Australia during 1977, showed thatMicrothrix inconspicuella Ragonot could develop on young apple leaves (Harley et al., 1979). Field studies in unsprayed apple orchards in South Africa showed that some feeding occurred, but fewer than 40% of late instar larvae developed to adults when confined in sleeves on apple tree branches. No feeding or survival occurred in large field cages or in the open. Adults which developed from apple fed larvae were smaller, deformed, occasionally mated and laid fertile eggs but their progeny did not feed or develop on apple fruit or leaves. In conclusion,M. inconspicuella larvae did not develop on apple fruit or leaves in the field, damage was mainly limited to apples already injured and feeding on leaves was minimal. Under normal pest control practicesM. inconspicuella populations did not survive on any part of the apple tree or onE. australis growing under the trees.   相似文献

To develop a damage threshold for the European red mite (ERM), Panonychus ulmi (Koch), the effect of ERM injury on fruit weight and return crop in the apple cultivars (CVs) Golden Delicious and Oregon Spur were studied in the Upper Galilee of Israel for two consecutive seasons. The ERM significantly affected the leaf colour, chlorophyll content, fruit weight and return harvest in both CVs. High population levels of 450 adult female cumulative mite days (ACMDs) reduced the fruit weight on Golden Delicious but not on Oregon Spur. Population levels above approximately 600 and 900 ACMDs in Golden Delicious and Oregon Spur, respectively, caused a substantial reduction in the return harvest. Because no damage was observed at low population levels, an action threshold of 150 ACMDs is recommended.  相似文献

The rate of entry by neonate larvae of the frugivorous codling moth, Cydia pomonella (L.) (Lepidoptera: Tortricidae), into fruit material was investigated. We used no‐choice bioassays in climate‐controlled rooms to assay larval entry across four host plant species (apple, pear, quince, walnut) and three varieties within a single fruit species (apple). Larvae successfully entering apples were reared to adulthood, and we collected tissue samples from apples which were successfully colonized in order to determine sucrose concentrations. This information was used to evaluate differences in adult moth size, development time, and pulp sucrose concentration due to apple variety. Four important findings emerged: (1) neonate larvae had the highest frequency of entry (86% of larvae) into apple fruits, compared with pear (78%), quince (56%), and walnut (32%); (2) the frequency of larval entry into immature apples differed across apple varieties, and larval entry rate was highest in variety Golden Delicious (72%), compared with Granny Smith (46%) and Red Delicious (64%); (3) on average, adult moths were larger and development times were shorter on the variety with the highest entry frequency (Golden Delicious); and (4) apple pulp sucrose concentrations were higher for Golden Delicious (17.5 μg mg?1) than for either Granny Smith (15.9 μg mg?1) or Red Delicious (15.1 μg mg?1) varieties, which correlates positively with entry and development data. We conclude that host fruit species and varietals within a species affect the entry rate of neonate codling moth larvae in no‐choice assays. We hypothesize that larval development is influenced by mean sucrose concentrations or other phytochemical differences associated with host fruit varieties.  相似文献

Previously, we showed that after a female apple maggot fly, Rhagoletis pomonella,arrives on a host hawthorn or apple fruit, its propensity to accept (bore into) or reject that fruit prior to egg deposition can be modified by previous ovipositional experience with one or the other species and, hence, involves learning. Here, we present both field and laboratory evidence indicating that females also are able to learn characteristics of three different apple biotypes or cultivars: Early Macintosh, Red Delicious, and Golden Delicious. We suspect that females learn to discriminate among these three cultivars on the basis of differences in chemical stimuli among cultivars. The effect of fruit cultivar learning was not as strong as the effect of fruit species learning.  相似文献

In field-cage studies, we investigated how the foraging behavior of tephritid fruit flies is modified by experience immediately prior to release on host plants. We observed females of a relatively monophagous species,Rhagoletis mendax (blueberry maggot fly), an oligophagous species,Rhagoletis pomomella (apple maggot fly), and a polyphagous species,Ceratitis capitata (Mediterranean fruit fly). Just prior to release on a host plant, the following kinds of stimuli were supplied: (1) single oviposition in a host fruit, (2) contact with 20% sucrose, (3) contact with a mixture of protein food (bird feces and sucrose), (4) contact with water, and (5) a walk over a host-plant leaf. When flies foraged on host plants without resources, search was most intensive (as measured by number of leaves visited) following a single oviposition in fruit, but residence time generally was the same following exposure to sugar, protein, and fruit stimuli.Rhagoletis mendax andC. capitata females visited the fewest leaves following exposure to water or host leaves, whereasR. pomonella foraged equally intensively following exposure to food stimuli, water, or leaves. On host plants containing resources (fruit and protein food), a single oviposition dramatically increased the number of females of all three species that found fruit compared to females that received experience with food, water or foliar stimuli. We found no significant effect of recent brief experience with any of the stimuli on subsequent attraction to protein food. Overall,C. capitata exhibited a higher propensity to abandon host plants than eitherR. mendax orR. pomonella. We suggest that this may reflect adaptations to differences in distribution of host plants in nature, strategies of dispersal, and host range.  相似文献

磷酸蔗糖合酶(sucrose phosphate synthase,SPS)是植物中蔗糖合成的主要限速酶,影响植物的生长发育和果实中蔗糖的含量。为探明苹果中SPS基因家族特性及其在蔗糖合成中的作用,该研究从苹果基因组中分离了MdSPS家族基因,分析了它们的进化关系以及mRNA表达特性与酶活性和蔗糖含量的关系。结果显示:(1)在苹果基因组中有8个SPS家族基因表达,它们分别属于双子叶植物的3个SPS亚家族。(2)荧光定量PCR分析显示,苹果C类的MdSPS6基因和A类的MdSPS1a/b基因是苹果中表达丰度最高的SPS基因成员,其中MdSPS6在苹果成熟果中表达丰度最高,其次是成熟叶片,而MdSPS1a/b在不积累蔗糖的幼果中表达丰度最高。(3)在果实发育过程中,除MdSPS1a/b之外,其它5个苹果MdSPS家族基因均随果实的生长表达丰度增加,与SPS活性和蔗糖含量明显呈正相关关系。研究表明,C类家族MdSPS6是苹果果实发育后期和叶片中蔗糖合成的主要SPS基因。  相似文献

Summary One of the most controversial putative cases of host race formation in insects is that of the apple maggot fly, Rhagoletis pomonella (Diptera: Tephritidae). A principal cause of the controversy is lack of relevant data. In laboratory and field enclosure experiments, we compared the host acceptance behavior of sympatric populations of flies originating from naturally infested hawthorn (the native host) and apple (an introduced host) in Amherst, Massachusetts or East Lansing, Michigan. In general, hawthorn fruit were accepted for ovipositional attempts nearly equally by apple and hawthorn origin females, whereas apples were accepted much more often by apple than hawthorn origin females. Similarly, males of apple and hawthorn origin exhibited about equal duration of residence on hawthorn fruits as sites at which to acquire potential mates, while males of apple origin tended to reside substantially longer than males of hawthorn origin on apples. Irrespective of fly origin, both sexes always responded more positively to hawthorn fruit than to apples. Because all flies assayed were naive (ruling out effects of prior host experience of adults) and because tests revealed no influence of pre-imaginal fruit exposure on pattern of host fruit acceptance by females, the combined evidence suggests the phenotypic differences we observed in host response pattern between hawthorn and apple origin flies may have an underlying genetic basis. Further tests showed that while larval progeny of flies of each origin survived better in naturally growing hawthorn fruit than in naturally growing apples, there was no differential effect of fly origin on larval survival ability in either host. We discuss our findings in relation to restriction in gene flow between sympatric populations of R. pomonella and in relation to current models of host shifts in insects.  相似文献

Emission rates of volatile organic compounds (VOCs) from Pirus malus L. subsp. mitis (Wallr.) var. Golden Delicious and var. Starking attacked by the phytophagous mite Panonychus ulmi Koch, and their attractiveness to the predatory mites Amblyseius andersoni Chant and Amblyseius californicus McGregor, were studied during three years. A large variability was found in the emission of individual VOCs depending on the infestation, the apple tree variety and the date. There were larger total VOC emission rates and larger total VOC leaf concentrations in apple trees attacked by phytophagous mites, especially in the var. Starking. In infested trees of this variety, there were also more predatory mites. An olfactometer assay showed that predatory mites preferentially chose branches infested by Panonychus ulmi (85% went to infested branches vs 15% to uninfested control branches) indicating that volatiles may be used as cues to find their prey.  相似文献

Four types of experiments were conducted to assess the pathogenicity of entomogenous nematodes against the apple sawfly,Hoplocampa testudinea Klug (Hymenoptera: Tenthredinidae). In Petri dish,Steinernema carpocapsae (Weiser) DD 136 strain,S. carpocapsae All strain,S. feltiae (Filipjev) andHeterorhabditis bacteriophora Poinar caused 100% larval mortality 72 h after the treatments. In semi-field conditions under dwarf apple trees, a single treatment with either 40 or 80S. carpocapsae All strain/cm2 caused significant (>80%) larval mortality. No significant increase in mortality was obtained with a second application. In 1990, late treatment with 80S. carpocapsae All strain/cm2 caused significant (39.1%) pupal mortality. In 50×50 cm quadrats under dwarf apple trees, treatment with 40 and 80S. carpocapsae All strain/cm2 were done in spring 1989. In spring 1990, respectively 22 and 25 apple sawfly adults emerged from quadrats treated with 40 and 80S. carpocapsae All strain/cm2, whereas 78 adults emerged from untreated quadrats. In a foliar application experiment, a single treatment withS. carpocapsae All strain significantly reduced the percent of fruit showing secondary damage. The possibility of using entomogenous nematodes as part of an integrated pest management in apple orchards is discussed, with particular reference to the integration of programs aimed at managing plum curculio,Conotrachelus nenuphar (Herbst) (Coleoptera: Curculionidae) populations.   相似文献

Insecticidal proteins are a potential resource to enhance resistance to insect pests in transgenic plants. Here, we describe the generation and analysis of the apple cultivar ‘Royal Gala’ transgenic for Nicotiana alata (N. alata) proteinase inhibitor (PI) and the impact of this PI on the growth and development of the Epiphyas postvittiana (light-brown apple moth). A cDNA clone encoding a proteinase inhibitor precursor from N. alata (Na-PI) under the control of either a double 35S promoter or a promoter from a ribulose-1,5-bisphosphate carboxylase small sub-unit gene (rbcS-E9 promoter) was stably incorporated into ‘Royal Gala’ apple using Agrobacterium-mediated transformation. A 40.3 kDa Na-PI precursor protein was expressed and correctly processed into 6-kDa proteinase inhibitors in the leaves of transgenic apple lines. The 6-kDa polypeptides accumulated to levels of 0.05 and 0.1% of the total soluble protein under the control of the rbc-E9 promoter and the double 35S promoter, respectively. Light-brown apple moth larvae fed with apple leaves expressing Na-PI had significantly reduced body weight after 7 days of feeding and female pupae were 19–28% smaller than controls. In addition, morphological changes such as pupal cases attached to the wing, deformed wings, deformed body shape, and pupal cases and curled wings attached to a deformed body were observed in adults that developed from larvae fed with apple leaves expressing Na-PI, when compared to larvae fed with the non-transformed apple leaves.  相似文献

Larvae of Epiphyas postvittana and Planotortrix octo were released onto branches cut from apple trees, and allowed to colonize a range of types of artificial nests. Both species exhibited similar strong preferences for nests comprising leaf-leaf or leaf-fruitlet combinations, followed by nests comprising leaf-plastic leaf, leaf-plastic fruitlet or plastic leaf-fruitlet combinations. Nests involving fresh plant material alone (shoot, fruitlet or leaf alone) were also colonized to a lesser extent, but no larvae were found on nests consisting of plastic leaves or fruitlets alone or in combinations of the two. In another experiment, more E. postvittana larvae colonized nests with leaf and fruitlet combinations, compared to leaf and glass ball, or leaf and treated wax ball combinations, where the wax had been in contact with fresh apples or fruitlets. Choice tests, conducted using larval traps, showed that larvae were caught in traps baited with odors collected and released by wax which had been in contact with mature apples and leaves. Chloroform extracts from apple skin also caught larvae in choice tests. These results suggest that both physical and chemical cues are important to leafroller larval establishment.  相似文献

MDH1: an apple homeobox gene belonging to the BEL1 family   总被引:2,自引:0,他引:2  
Differential display was used to isolate genes differentially expressed early in fruit development of apple (Malus domestica Borkh.). This approach resulted in the isolation of MDH1, a homeobox gene with a homeodomain similar to that of BELL1 (BEL1), which is involved in regulation of ovule development in Arabidopsis. However, outside the homeodomain MDH1 is quite different from BEL1. In apple, MDH1 mRNA was predominantly found in flowers, expanding leaves and expanding fruit. In pre-anthesis flowers, in situ hybridization showed that MDH1 mRNA accumulated in ovules. To further investigate the function of this new homeobox gene, MDH1 was transformed into Arabidopsis thaliana under the control of the cauliflower mosaic virus 35S promoter. The transgenic Arabidopsis plants showed dwarfing, reduced fertility and changes in carpel and fruit (silique) shape. The size and shape of the cells in the transgenic fruit was irregular. Both the transgenic phenotypes in Arabidopsis and the expression pattern of this gene in apple are consistent with the idea that MDH1 is likely to play an important role in control of plant fertility.  相似文献

Ceratitis capitata (Wiedemann), the Mediterranean fruit fly, is one of the key pest species affecting deciduous fruit orchards along the Mediterranean coasts. Because of global warming, C. capitata is gradually spreading north and is becoming a major pest of apples. Determining the susceptibility of the main apple varieties grown in the region will serve as a cornerstone to the management of this pest. In this study, we show the results of a field and laboratory no‐choice test conducted to determine the Medfly preferences on different apple cultivars. The seven main varieties of apples (Gala, Red Delicious, Golden Delicious, Granny Smith, Kanzi, Morgen Dallago and Fuji) were tested. The results demonstrate that C. capitata lays eggs on all apple cultivars in both field and laboratory conditions. The Granny Smith, Red Delicious and Morgen Dallago varieties showed the lowest susceptibility in laboratory conditions, (0.75, 1.55, 2 oviposition punctures/fruit, respectively), with significant differences in oviposition compared to the Golden Delicious, Kanzi and Fuji (3.27, 3.31, 3.1 oviposition punctures/fruit, respectively) varieties, which were shown to be the most susceptible to Medfly attack in laboratory conditions. On the other hand, only slight and not statistically significant differences emerged from the field trials. In relation to the physico‐chemical characteristics, the apple cultivars showing the lowest susceptibility (Granny Smith, Red Delicious and Morgen Dallago) had harder peels and pulps and lower sugar contents than the most susceptible cultivars (Golden Delicious, Fuji and Kanzi). These results were also confirmed through evaluation of larval development on different varieties. In fact, Granny Smith, Red Delicious and Morgen Dallago were the three varieties that did not allow adequate larval and adult development and reduced the possibility of the emergence of a new generation.  相似文献

The photosynthate consumption in apple fruits in relation to the leaf/fruit ratio was studied in sections of branches of the Graasten and Golden Delicious varieties by exposure to 14CO2during July and August. A significant, negative correlation was found between the fixation of 14C in the fruits in terms of percent of the total amount of 14C absorbed and the leaf/fruit ratio of the branch sections. The leaf area required for the saturation of one fruit was found to be ca. 190 and 230 cm2 (14 and 17 leaves) in the case of Golden Delicious, and ca. 400 and 670 cm2 (25 and 42 leaves) for Graasten in July and August, respectively, calculated under conditions of large leaf areas per fruit. In such cases a fairly good, reverse proportionality exists between the 14C fixation in the fruit in terms of percent and the leaf area expressed in multiples of saturation area. At low leaf/fruit ratios, however, the actual saturation area is found to be lower than the theoretically computed one. The translocation of the 14C assimilated in the leaves of the extension shoots or of spurs without fruits to fruits on other spurs was on the whole promoted by a decreasing leaf/fruit ratio in the parts in question; similarly the greatest transport took place on the side where the leaf/fruit ratio was lowest. The fixation was often greatest in the fruit closest to the treated leaves, hut in a number of cases the value was higher for the second closest or even further removed fruits. In this connection the importance of the size of the fruit and the vascular connections is discussed.  相似文献

Responses of the apple maggot fly,Rhagoletis pomonella (Walsh), to 8 cm red sticky sphere traps, baited with the synthetic fruit odor butyl hexanoate, were investigated in field-caged apple trees containing green or red Gravenstein apples. Trap capture rate and the probability of oviposition in apples before capture generally increased with female age and number of mature eggs in ovaries. Two days of pre-test exposure of mature females to red hawthorns, green Red Delicious apples or green Gravenstein apples had no significant effect on the likelihood of a fly finding a red sphere. However, before capture on a sphere or departing a tree, hawthorn-exposed females found significantly fewer apples and laid significantly fewer eggs than females exposed to Gravenstein apples. Variation in duration of pre-test exposure (1–4 days) of flies to Gravenstein apples had no detectable influence on female response to apples or to a red sphere in a test tree. The relevance of these findings to effectiveness of sphere traps, forR. pomonella control in commercial orchards is discussed.  相似文献

Biological control and induced resistance are two of the promising approaches to the control of postharvest diseases. This study was conducted to evaluate the efficacy of salicylic acid (SA) alone or in combination with an antagonistic yeast, Cryptococcus laurentii, in controlling the blue mold disease caused by Penicillium expansum on apple fruit wounds. SA alone significantly inhibited the spore germination of P. expansum in vitro when its concentration was increased to 1000 μg ml−1, but it was not effective in controlling the disease in vivo. Simultaneous application of SA and C. laurentii to the wounds on the apple fruit surface showed that SA could improve the efficacy of C. laurentii against P. expansum in a concentration-dependent manner, being most effective at 10 μg ml−1 but less effective at a higher or lower concentrations. Besides reducing the blue mold incidence in the local wound sites, the combination of C. laurentii with SA at 10 μg ml−1 also had a synergistic effect on the induction of fruit resistance to the disease, which might be associated with a rapid increase in peroxidase, phenylalanineamonialyase and lipoxygenase activities. In addition, SA at 100 μg ml−1 or above showed an adverse effect on the growth of C. laurentii in vitro and in vivo, whereas it had no effect when its concentration was decreased to 10 μg ml−1 or lower. This suggested that SA could enhance the biological activity of C. laurentii in apple fruit by inducing resistance to pathogens based on the antagonistic activity of C. laurentii.  相似文献

This paper summarises the information on the occurrence of phenolic compounds in apple Malus domestica leaves and fruits with special reference to their developmental changes and regulation of biosynthesis. Besides the ontogenetic variation in biosynthesis and accumulation, the stress-induced and pathogenesis-related changes are emphasised. Aspects of commercial importance are addressed, ranging from fruit colouration, through disease resistance, to the direct use of apple tissues, as raw material for the extraction of bioactive phenolic compounds.  相似文献

Damage of leaf spot, caused by Mycosphaerella fragariae and gray mold also called Botrytis fruit rot, caused by Botrytis cinerea, average fruit weight and yield were evaluated with regard to cultural methods over 2years. Leaf spot damage decreased significantly by around 90% due to leaf sanitation (removal of dead and leaf spot infected leaves in early spring) and by 50% due to plantation in a one-row-system instead of a two-row-system. When all leaves including the healthy green ones were removed in early spring, average fruit weight decreased significantly by 10%. Fruit sanitation – the third treatment – did not influence any of the measured parameters. Neither leaf sanitation nor fruit sanitation (removal of damaged fruits during harvest) reduced B. cinerea damage significant. Only the combination of a one-row-system, leaf sanitation and fruit sanitation almost halved (not significantly) B. cinerea damage in the first crop year compared to a two-row-system without leaf and fruit sanitation. B. cinerea damage correlated significantly and positively with the biomass of plants by R2= 0.47. According to this study and the cited literature it is suggested for humid Central European conditions to apply a one-row-system combined with leaf sanitation in early spring and fruit sanitation during harvest if fruit density is high, to reduce the risk of damages in larger dimension caused by M. fragariae and B. cinerea.  相似文献

The nucleotide sequence of a cDNA prepared from poly(A)+ RNA from Lycopersicon esculentum fruit codes for a protein, M r 20812, with features representative of the protein core of arabinogalactan proteins. The deduced amino acid sequence resembles that of peptides of arabinogalactan proteins isolated from carrot and rose and is most similar to the sequence of tryptic peptides from Lolium multiflorum (Gleeson et al., Biochem J 264 (1989) 857–862). The similar sequences include a number of Ala-Pro repeats, a feature considered distinctive of arabinogalactan proteins. The amino acid composition is similar to that of the peptide core of the Lolium multiflorum arabinogalactan protein; alanine, serine and proline account for 57% of the polypeptide. The mRNA corresponding to the cDNA sequence was detected in roots, leaves and fruit. The levels of mRNA are reduced in older leaves, in fruit that have commenced ripening and in leaves and fruit that have been wounded.  相似文献
