Cytoarchitectonics of outer nuclei of the pretectal area was studied in four species of the sturgeons: the great sturgeon Huso huso, L., the Russian sturgeon Acipenser güldenst?dti persicus n. kurensis, Belyaeff, the starred sturgeon Ac. stellatus, Pall., and the barbel sturgeon Ac. nudiventris, Lov. Morphological organization of these structures was studied by methods of Nissl and Bielshowskii modified by Viktorov. In this part of the pretectal area, five nuclear structures were described: parvicellular and magnocellular outer pretectal nuclei, lamellar pretectal nucleus, and dorsal pretectal nucleus, as well as accessory optic nucleus. Comparison of the obtained results with literature data has revealed a sufficiently high level of differentiation of this pretectum part of the sturgeons. Besides, a general conclusion is presented on organization of their entire pretectal area.  相似文献

Kurensis Belyaeff, stellate sturgeon, Ac. stellatus Pall. and barbel sturgeon, Ac. nudiventris Lov. Study of morphological organization of the structures was carried out using routine Nissl staining and Bilschowski impregnation technique in Viktorov’s modification. Similar structure of this pretectal area was found in the species. Five nuclear formations were described in this pretectal area: parvocellular and magnocellular external pretectal nuclei, laminiform nucleus of pretectum, dorsal pretectal nucleus, and the accessory optic nucleus. Comparison of the obtained data with the literature revealed a high level of differentiation of the area of pretectum in the sturgeons. Also, a general conclusion is given on the organization of the whole pretectal area in the sturgeons.  相似文献

An immunohistochemical study was carried out to reveal distribution of immunoreactivity (IR) against Ca-binding proteins (CaBPr): calbindin (Calb), calretinin (Calr), and parvalbumin (Parv) in pretectum of the Central Asian terrestrial turtles (Testudo horsfieldi) and the pond turtles (Emys orbicularis). A wide distribution of CaBPr-IR neurons and terminals with a significant overlapping was established in most of pretectal nuclei (Lm, Pd, Pe, Po, and ndCP) with some differences in density of IR elements for various proteins. The alternative distribution of Parv-IR (a high density of IR neurons) and Calb/Calr-IR (a most low content of IR neurons and terminals) was found in the non-retinorecipient nucleus Ptv. Comparison of data obtained in turtles with literature data obtained in other species of reptiles and mammals has revealed a significant variability in the CaBPr distribution in some, presumably homologous, pretectal nuclei among various representatives of amniotes. The conclusion is made that in the process of evolution of amniotes, apart from conservatism of CaBPr-IR characteristics, there occurred their plastic rearrangements due to morpho-functional specialization of pretectal nuclei.  相似文献

Different parameters of diversity and spatial niche amplitude (Piclou, 1975, Pineda et al., 1981b) have been used to describe the spatial organization of mediterranean grasslands in Central Spain. A slope sampled by 480 contiguous 8×8 cm quadrats proved to have a homogeneous floristic distribution when it was divided into 160 parts and maximum heterogeneity when divided into 4 sectors. These sectors corresponded to different geomorphological zones of the slope. The complexity of vegetation distribution on the slope was reflected by differences in organization — measured by diversity parameter A=H(P/E)/log2 N (number of plots) between the parts obtained by division. The presence of low-entropy species — specialists - and high entropy species — generalists — was related to the different scales of slope organization. The spatial distribution of plants was compared for different successional stages. Multivariate analysis of sampling plots confirmed previously identified organizational characteristics and clarified the nature of transitions between communities of different floristic composition or structure.  相似文献

The distribution of neurotensin, neurokinin A, dynorphin A, galanin, somatostatin-28 (1-12), neuropeptide Y, vasoactive intestinal polypeptide, gastrin-releasing peptide, gamma-melanocyte stimulating hormone, alpha-neo-endorphin, angiotensin H, cholecystokinin-8, serotonin and tyrosine hydroxylase has been studied in the pretectal nuclei of the Cyprinus carpio: nuclei pretectalis superficialis parvicellularis and magnocellularis, pretectalis centralis, pretectalis, and pretectalis periventricularis dorsalis and ventralis using an indirect immunoperoxidase technique. We have found neuropeptide Y and serotonin immunoreactive fibres in all pretectal nuclei, whereas gastrin-releasing peptide immunoreactive fibres were visualized in the nuclei pretectalis superficialis parvicellularis and magnocellularis, pretectalis centralis. pretectalis and pretectalis periventricularis dorsalis; neurokinin A immunoreactive fibres in the nuclei pretectalis superficialis parvicellularis and magnocellularis and pretectalis periventricularis dorsalis; galanin immunoreactive fibres in the nuclei pretectalis superficialis parvicellularis, pretectalis centralis and pretectalis periventricularis dorsalis; and neurotensin immunoreactive fibres in the nucleus pretectalis periventricularis dorsalis. Additionally, immunoreactive cell bodies containing neuropeptide Y were observed in the nuclei pretectalis superficialis parvicellularis and pretectalis periventricularis dorsalis, and serotonin and tyrosine hydroxylase cell bodies were found in the nuclei pretectalis periventricularis dorsalis and ventralis respectively. The presence of the neuroactive substances found in the carp pretectal nuclei suggest that they might be involved in the regulation of certain functions within the visual system.  相似文献

Summary Retinofugal and retinopetal projections were investigated in the teleost fish Channa micropeltes (Channiformes) by means of the cobaltous lysine and horseradish peroxidase (HRP) tracing techniques. Retinofugal fibers cross completely in the optic chiasma. A conspicious lamination is present in those parts of the optic tract that give rise to the marginal branches of the optic tract. This layering of optic fibers continues in the marginal branches to mesencephalic levels. Retinal projections to the preoptic and hypothalamic regions are sparse; they are more pronounced in the area of pretectal nuclei. The medial pretectal complex and the cortical pretectal nucleus are more fully differentiated than in other teleostean species. Further targets include the thalamus and the optic tectum. The course of major optic sub-tracts and smaller fascicles is described. Retinopetal neurons are located contralaterally in a rostral and a caudal part of the nucleus olfactoretinalis, and in a circumscribed nucleus thalamoretinalis. The present findings are compared with reports on other teleost species.  相似文献

Using immunohistochemical labeling of the cells containing neuronal NO synthase (nNOS), tyrosine hydroxylase (TH), GABA, and parvalbumin (PA), as well as histochemical marking of choline acetyltransferase-containing neurons, we examined the neurochemical organization of the glomerular nuclei and preglomerular complex in the brain of the masu salmon (Oncorhynchus masou). Injections of the carbocyanine dye DiI allowed us to examine projections of neurons of the preglomerular and mammillary nuclei in the salmon brain. We showed that cholinergic, GABA-, PA-, TH-, and nNOS-immunopositive neurons belonging to different morphological types are present in the glomerular and medial preglomerular nuclei. The analysis of correlations between morphometric characteristics of the cells belonging to different neurochemical types and densitometric estimates of amounts of neurochemical agents present in these cells allowed us to hypothesize that there are close morphofunctional interrelations in cell populations possessing different neurochemical and morphometric characteristics. These interrelations of the cells belonging to different chemotypes are, probably, realized as mediatory/modulatory ones. The presence of a great number of small slightly differentiated cells in the preglomerular and glomerular nuclei allows us to suppose that the growth of the greatest sensory center of the salmon brain is provided by neuroblasts that migrate from the proliferative zones in the course of postembryonal neurogenesis. It is also hypothesized that NO, TH, and GABA are involved in paracrine control of the postnatal morphogenesis of the salmon preglomerular complex. The data obtained by hodological analysis indicate that the nuclei of the preglomerular complex obtain afferent projections from the dorsomedial and ventroventral telencephalic regions, preoptic nucleus, periventricular layer of the tectum, and posterior central thalamic nucleus. Our study demonstrated the existence of reciprocal functional connections between the preglomerular complex (most important diencephalic center for transmission of sensory information) and dorsomedial and ventral regions of the telencephalon in the masu salmon.  相似文献

Summation processes occurring in single neurons of the pretectal area in response to either moving or stationary light stimuli were studied in acute experiments on cats. In most neurons studied (85%), gradual increase of the angular size of stimulus resulted in clearly defined summation. In neurons lacking directional sensitivity (nondirectional neurons) the stimulus movement in two opposite directions caused, as a rule, similar and symmetrical changes in the number of spikes, whereas under the same conditions direction-sensitive neurons, in addition to symmetrical development of summation processes, could exhibit substantial differences in the summation curves. The responses to a preferred movement direction could be significantly inhibited or facilitated, while the responses to a non-preferred direction remained stable or changed reciprocally. Neuronal responses to different directions of the movement of stimulus might change independently of each other. This was also the case whenon andoff responses of theon—off neurons to stationary stimuli were compared. Almost all vertebrates are capable of recognizing biologically relevant stimuli at or shortly after birth, and in some phylogenetically ancient species visual object recognition is exclusively innate. Extensive and detailed studies of the anuran visual system have resulted in the determination of the neural structures and pathways involved in innate prey and predator recognition in these species [Behav. Brain Sci. 10 (1987) 337; Comp. Biochem. Physiol. A 128 (2001) 417]. The structures involved include the optic tectum, pretectal nuclei and an area within the mesencephalic tegmentum. Here we investigate the structures and pathways involved in innate stimulus recognition in avian, rodent and primate species. We discuss innate stimulus preferences in maternal imprinting in chicks and argue that these preferences are due to innate visual recognition of conspecifics, entirely mediated by subtelencephalic structures. In rodent species, brainstem structures largely homologous to the components of the anuran subcortical visual system mediate innate visual object recognition. The primary components of the mammalian subcortical visual system are the superior colliculus, nucleus of the optic tract, anterior and posterior pretectal nuclei, nucleus of the posterior commissure, and an area within the mesopontine reticular formation that includes parts of the cuneiform, subcuneiform and pedunculopontine nuclei. We argue that in rodent species the innate sensory recognition systems function throughout ontogeny, acting in parallel with cortical sensory and recognition systems. In primates the structures involved in innate stimulus recognition are essentially the same as those in rodents, but overt innate recognition is only present in very early ontogeny, and after a transition period gives way to learned object recognition mediated by cortical structures. After the transition period, primate subcortical sensory systems still function to provide implicit innate stimulus recognition, and this recognition can still generate orienting, neuroendocrine and emotional responses to biologically relevant stimuli.  相似文献

Summary The ability of the highly condensed chromatin of small thymocyte nuclei and the more loosely organized chromatin of hepatocyte nuclei to interact with nine DNA-specific fluorochromes was assessed by microfluorometry. Although the results obtained with five of the fluorochromes—mithramycin, 7-aminoactinomycin d, Hoechst 33258, DAPI, and propidium iodide—were found to be virtually unaffected by differences in the degree of condensation of the chromatin, the values obtained with the remaining fluorochromes—proflavine, quinacrine mustard, berberine sulfate, and pyronin Y—appeared to be affected significantly by organizational differences of the chromatin. All of the latter structural probes, except quinacrine mustard, produced fluorescence values which were higher in the 2c nuclei of hepatocytes than in the nuclei of small thymocytes. Quinacrine mustard yielded higher values in thymocyte nuclei; and in the hepatocyte polyploid series (2, 4, and 8c), it did not produce the expected multiples of the 2c value. Pretreatment of the two types of nuclei with RNase affected their total fluorescence in unpredictable ways. While RNase extraction lessened the differences between thymocyte and 2c hepatocyte nuclei stained with propidium iodide, Hoechst 33258, proflavine, and berberine sulfate, it increased the differences between nuclei stained with mithramycin, quinacrine mustard, pyronin Y, and 7-aminoactinomycin d. The ability of RNA-depleted chromatin to interact with various types of fluorochromes might be a useful parameter in subsequent studies of chromatin organization.  相似文献

The estrous cycle of the female rat is understood fairly well. Determinations of pituitary and target gland hormones, and neurochemical and neurophysiologic studies provided some information allowing the following conclusion. Estradiol and progesterone in proper quantity and timing are signals for the central nervous system-pituitary axis to evoke preovulatory gonadotropin release. The steroids most likely interact with neurotransmitter regulating mechanisms at extra- and intrahypothalamic levels. Neural activity in the medial preoptic area — which plays a key role in regulating the estrous cycle—is increased during the time of preovulatory gonadotropin release. This forebrain structure has been shown to receive neural inputs from limbic and midbrain areas that are known to have modulatory functions on gonadotropin release. The neurochemical basis for increased release of hypothalamic hormone (s), which control gonadotropin release in female, male, and immature animals, are well integrated changes in turnovers of different neurotransmitters. Direct actions of hormones at the pituitary level to modulate the action of hypothalamic hormones are also possible. The molecular basis of these interactions is not completely understood.  相似文献

Using histochemical and immunohistochemical techniques, distribution of activity of oxidative mitochondrial enzyme cytochrome oxidase (CO) and of immunoreactivity to calcium-binding proteins has been studied in spiral ganglion and auditory nuclei of brainstem in two turtle species. It has been shown that immunoreactivity to calbindin, parvalbumin, and calretinin in neurons and neuropil of nuclei of cochlear and superior olivary complexes, in nucleus of lateral lemniscus, and in spiral ganglion neurons coincides topographically with the high CO activity. The similarity of the studied metabolic and neurochemical characteristics of these auditory centers in reptiles, birds, and mammals indicates the existence of some common principles of their organization in amniotes in spite of phylogenetic differences and peculiarities of specialization of the auditory system in different species.  相似文献

The cells of origin of the olivary projection from mesodiencephalic structures have been demonstrated in the Japanese monkey (Macaca fuscata) with the horseradish peroxidase (HRP) method. Particular attention has been paid to the pretecto-olivary projection which is entirely ipsilateral and originates from the nucleus of the optic tract (NOT), the posterior (or principal) pretectal nucleus (PPN), the sublentiform nucleus (SL), and the (pretectal) ventral lateral zone (VLZ). Pretecto-olivary cells are small-medium sizes of oval or fusiform types. To facilitate comparison among the findings of several experiments, a diagram of the macaque olive, as visualized unfolded, was constructed (Fig. 1). Although a topographic correlation has not emerged clearly from the present experiments, the pattern of the pretecto-olivary projection in the monkey appears to be similar to that found in the cat. Participation of some of the pretectal nuclei in the optokinetic nystagmus and the vestibulo-ocular reflex is discussed in connection with the pretecto-olivary and olivocerebellar projections.  相似文献

A simple method is presented for the isolation of nuclei from somatic embryos of carrot (Daucus carota L.), which is applicable to small amounts of material in synchronized culture. The method employs buffers containing a high concentration of glycerol to stabilize the structure of the nuclei. Purification was carried out by centrifugation using preformed Percoll gradients. Treatment with cell wall-degrading enzymes prior to homogenization improved the efficiency of isolation and permitted a reproducible yield of nuclei. The pure preparations were obtained with an efficiency of approximately 60%. The isolated nuclei retained their morphological characteristics as demonstrated by phase — contrast and electron microscopy. Nuclear proteins displayed the expected species of histones by two-dimensional gel electrophoresis. The isolated nuclei showed high RNA polymerase activity.  相似文献

Tick anticoagulant peptide (TAP) is a factor Xa-specific inhibitor and is structurally homologous to bovine pancreatic trypsin inhibitor (BPTI). The fully reduced TAP refolds spontaneously to form the native structure under a wide variation of redox buffers. The folding intermediates of TAP consist of at least 22 fractions of one-disulfide, two-disulfide, and three-disulfide scrambled isomers. Three species of well-populated one- and two-disulfide intermediates were isolated and structurally characterized. The predominant one-disulfide species contains TAP-(Cys33—Cys55). Two major two-disulfide isomers were TAP-(Cys33—Cys55, Cys15—Cys39) and TAP-(Cys33—Cys55, Cys5—Cys39). Both Cys33—Cys55 and Cys15—Cys39 are native disulfides of TAP. These three species are structural counterparts of BPTI-(Cys30—Cys51), BPTI-(Cys30—Cys51, Cys14—Cys38), and BPTI-(Cys30—Cys51,Cys5—Cys38), which have been shown to be the major intermediates of BPTI folding. In addition, time-course-trapped folding intermediates of TAP, consisting of about 47% one-disulfide species and 30% two-disulfide species, were collectively digested with thermolysin, and fragmented peptides were analyzed by Edman sequencing and mass spectrometry in order to characterize the disulfide-containing peptides. Among the 15 possible single-disulfide pairings of TAP, 10 (2 native and 8 nonnative) were found as structural components of its one- and two-disulfide folding intermediates. The results demonstrate that the major folding intermediates of TAP bear structural homology to those of BPTI. However, the folding pathway of TAP differs from that of BPTI by (a) a higher degree of heterogeneity of one- and two-disulfide intermediates and (b) the presence of three-disulfide scrambled isomers as folding intermediates. Mechanism(s) that may account for these diversities are proposed and discussed.  相似文献

Summary In the endosperm ofEchinocystis lobata the endopolyploid nuclei show structural changes of chromocentres typical of the process of endomitosis in Angiosperms.The cytological differentiation of the endosperm proper and the chalazal haustorium in three further representatives of theCucurbitaceae: Sicyos angulata, Cyclanthera pedata andCitrullus colocynthis is also connected with endomitotic polyploidisation. The nuclei of the endosperm proper attain in the different species different levels of polyploidy, 384n, as a maximum. In the nuclei of the chalazal haustorium the degree of polyploidy is lower (as a maximum 24n).The structure of polyploid nuclei which did not undergo postendomitotic mitoses shows some differences in the species studied in the course of the present work.Sicyos angulata has prochromosomic diploid nuclei, whereas those ofCitrullus colocynthis andCyclanthera pedata can be assigned to chromomeric nuclei with a dense structure. Endopolyploid nuclei of all three species studied are characterized by the occurence of compact chromocentres of different size; besides in some nuclei ofSicyos angulata andCyclanthera pedata the chromocentres within the resting nucleus reveal their chromomere structure. In addition, inCitrullus colocynthis nuclei with numerous thin threads of various degrees of spiralisation have been observed. — In all three species the endopolyploid nuclei after mitotic divisions have notably higher numbers of nucleoli and small chromocentres.Dedicated to Professor L.Geitler on the occasion of his 70th birthday.  相似文献

In the last 3 oogonial mitoses in Ascaphus truei all daughter nuclei remain in the same cell. The oocyte is 8-nucleate at the start of meiotic prophase and remains so until late in oogenesis when 7 of the nuclei disappear. All 8 nuclei in a single oocyte resemble one another with respect to size and chromatin distribution at all stages of meiotic prophase. Much of the Feulgen-positive material in pachytene nuclei is concentrated into one region of the nucleus. — All of the 8 germinal vesicles of yolky oocytes have a full set of lampbrush diplotene bivalents. Germinal vesicles from oocytes of up to 0.8 mm diameter have less than 100 nucleoli, some of which are multiple nucleoli in the sense that they have more than one core region. Each of the 8 nuclei in oocytes from one animal had about the same volume of nucleolar material. — Two values have been obtained for the amount of DNA in a diploid nucleus from Ascaphus. A biochemical estimate utilizing erythrocyte nuclei and the diphenylamine reaction yielded a value of 7.1 pg per nucleus. Microphotometry of erythrocyte nuclei stained with Feulgen's reagent gave a value of 8.2 pg per nucleus. — Microphotometric measurements of Feulgen-stained nuclei at various stages of meiotic prophase up to diplotene indicate that each nucleus synthesizes up to 5 pg of extrachromosomal DNA during and immediately after pachytene. This DNA is considered to be nucleolar. Autoradiography of nuclei from oocytes which had been incubated for 6h in 3H thymidine showed silver grains over pachytene and early diplotene nuclei only. In pachytene nuclei the silver grains overlaid that part of the nucleus where Feulgen-positive material was most concentrated. Most of the chromosomal material was unlabelled. — The significance of the 8-nucleate condition in Ascaphus oocytes is discussed, and the amount of nucleolar DNA synthesized at pachytene and of nucleolar material present in germinal vesicles is compared with corresponding situations in other amphibians.  相似文献

By using a histochemical method of determination of activity of cytochrome oxidase (CO), the level of metabolic activity in pigeons has been shown to be higher in centers of the tectofugal visual channel (pretectal nuclei: Pr, SP, SP/IPS, thalamic nucleus Rot, telencephalic entopallidum) than in centers of the thalamofugal visual channel (GLd, visual area of the hyperpallium Wulst). These data agree with the concept of the dominating role of the tectofugal visual channel in organization of the bird everyday behavior. The high CO activity is also characteristic of the mesencephalic structures (EM, isthmus nuclei: IMc, IPc, SLu) modulating transduction of visual information in tectum, Rot and GLd. Similar differences in the metabolic activities between two visual system channels have been shown earlier in reptiles, which indicates the evolutionary conservatism of the tectofugal visual channel among the sauropside amniotes. However, in pigeons the level of the CO activity in some GLd nuclei approaches that in Rot, which allows us to suggest a rise in birds of the role of the thalamofugal channel in processing of information necessary for performance of complex visual functions.  相似文献
