Influence of EDTA (C10H14N2Na2O8.2H2O) and EGTA (C14H24N2O10) on physiological functions homoiothermic organisms at deep hypothermia, was studied. White rats during cooling were in special sections without rigid fixing of head and limbs. In reply to intravenous introduction of EDTA and EGTA solutions, similar answers of the organisms were observed: raised breathing frequency and amplitude, intensity of electrical activity of muscles; these signs of activation of physiological functions lasted 8-10 minutes. Besides, of the 20th-30th minute after introduction of the second dose of preparations (at rectal temperature 17.1 +/- 0.5 degrees C), the secondary activation respiratory and thermoregulatory functions were registered. The termination of the cold shivering in experiments with introduction of EDTA and EGTA solutions occurred at lower temperatures in rectum and in a brain (16.7-17.3 degrees and 17.8-18.2 degrees C, resp.) than in control experiments (18.7 +/- 0.6 degrees C and 20.2 +/- 1.5 degrees C). The authors suppose that the activation of the thermoregulatory and respiratory functions is caused by a decrease in concentration of ions Ca2+ in the blood plasma.  相似文献

In parenteral and intranasal immunization with M. pneumoniae of guinea pigs, along with seroconversion, there was observed development from the 15th-20th day of hypersensitivity of delayed type; this hypersensitivity was revealed by positive skin tests and by the reaction of specific transformation of the peripheral blood lymphocytes. An increased reactivity of the smooth muscles of the intestine of parenterally immunized animals was revealed. With the aid of the test of alteration of blood neutrophils in vitro there was found development of specific sensitization of the organism to M. pneumoniae in the patients with mycoplasma pneumonia.  相似文献

While researching the mechanism underlying the effect of the sympathetic nervous system on the operation of skin receptors, we demonstrated that a tremendous role in this process is played by smooth muscles, whose condition also mediates the sympathetic effect on receptors. The increase in the activity of the sympathetic nerve fibers results in an increase in the tonus of the smooth muscles which in turn alters the mechanical condition of the tissues surrounding the receptors. It was established that the change in the tonus of the smooth muscles in the skin itself affects the reaction of the receptors that is evoked by mechanical stimulation. The change in the tonus of the smooth muscles of the vessels affects the response of receptors caused by cooling of the skin.Nizhegorod Medical Institute, Russian Federation Ministry of Health. Ante- and postnatal development of anatomical structures and cells of lymphatic nodes has been studied in rat offspring subjected to tetracycline effect during placentation and organogenesis (the 8th-14th days) and during fetogenesis (the 15th-20th days). Tetracycline injection during the 8th-14th days of embryogenesis results in certain disturbances of the nodular structure formations, inhibition of lympho- and plasmo-cytopoesis against the background of a sharp increase in number of basophilic granulocytes and tissue basophils. The antibiotic effect on the 15th-20th days of embryogenesis does not disturb the lymph node formation, but produces an increasing number of lymphocytes, plasmocytes, macrophages, eosinophilic granulocytes and tissue basophils in them.  相似文献

Experiments were conducted on rats during the early postnatal period; a study was made of the membrane potential (MP) establishment of the fibers of the skeletal muscles of the neck, the anterior and the posterior limbs. At birth the most mature were the muscles of the neck, and the least -- the muscles of the posterior limb. Establishment of the stationary MP level in the muscles of the neck occurred during the first week after birth, in the muscles of the anterior limbs -- by the 10th-12th day, and of the posterior limbs -- by the 15th-20th day. The order of maturation of various groups of the skeletal muscles was associated with the peculiarities of the neuro-trophic influences at various age periods. Muscles of the neck were characterized at all the developmental stages by a rhythmic low-frequency electromyographic activity. In the muscles of the limbs the rhythmic electromyographic activity was transformed into the discharge high-frequency activity by the period of termination of increase of the MP of the muscles.  相似文献

The deformation of rat skin in response to cooling and stimulation of nerve fibers innervating the piloerector and vasoconstrictor muscles has been studied by optical coherence microscopy. It has been found that the deformation of skin upon cooling is due to the contraction of collagen fiber bundles rather than the smooth muscle of vessels or piloerectors.  相似文献

Effect of a nonspecific beta adrenergic blocker — propranolol (40 mg per os) on thermoregulatory responses of cold water immersed (12.5°C) humans was studied. Propranolol attenuates resting and cold induced thermogenesis, rectal temperature, heart rate and systolic blood pressure, but increases production of adrenaline and cortisol. Propranolol has no effect on the threshold body temperature for induction of cold thermogenesis and on central thermosensitivity. The following conclusions are drawn from consideration of the data presented: During the early phase of cooling (20 min after the start of cooling) the thermogenesis mediated by beta adrenergic receptors may cover about 80% of the total metabolic increase induced by cold. After about 30 min of cooling the relative proportion of beta adrenergic thermogenesis starts to decline, reaching 20% of the total cold thermogenesis at the end of cooling.

It can be suggested from consideration of the data that, in man, the beta adrenergic receptors in the heart, blood vessels, adipocytes and muscles participate in mediating effect of cold on cardiovascular and thermoregulatory responses. Furthermore, these data imply that human adrenergic thermogenesis is produced outside of the brown adipose tissue. Thus, physiological mechanisms mediating adrenergic thermogenesis in humans appear to be different from those in small mammals.  相似文献

The following study describes the synthesis of new benzanilide derivatives and their pharmacological investigation on smooth muscle preparations of guinea pigs. All compounds were synthesized in good yields and showed a spasmolytic activity without significant effect on vascular smooth muscles and heart muscle preparations. Moreover, further pharmacological investigations as well as in silico studies were performed to elucidate the mechanism of action. Compound 3 showed the most potent spasmolytic activity with an IC(50) of 3.25microM.  相似文献

In 40 series of histological sections performed in human embryos and prefetuses from 4 up to 20 weeks of development, as well as in 20 corpses of fetuses and stillborns, it has been stated that the anlage of the thoracic duct appear in 6-7-week-old fetuses as lymphatic clefts surrounded with mesenchymal cells that are situated near large veins in the areas of the most active morphogenesis. Connecting with each other, the clefts form the jugular and retroperitoneal lymph sacs and a well branching network of canals. From the latter, on the 7th-8th week of development a plexus of lymph vessels appear, and later on (on the 8th-9th week)--bilaterally situating trunks of the thoracic duct. Further development of the thoracic duct is connected with the lymph nodes formation, their germs appear on the 9th-10th week along the course of the left trunk, as well as along the ductal branches and anastomoses. The formation of the lymph nodes results in reduction of some trunks and plexuses of the thoracic duct. Owing to this, its form in 14-15-week-old prefetuses resembles the one in newborns. Disturbances in the formation processes of the lymph nodes along the course of the reducing ductal areas, as well as their formation along the course of its main trunk can result in various structural variants of the thoracic duct in children and grown-up persons. Histogenesis of the thoracic duct wall and formation of the lymph nodes are not completed by birth.  相似文献

目的:建立药物生殖毒性研究中孕鼠离体子宫平滑肌张力的测定方法。方法:SD大鼠于妊娠第6~15天(GD6-15)给予受试物,在妊娠第20天(GD20)取子宫平滑肌,分别给予不同浓度缩宫素和硫酸镁溶液刺激,选择试验的最佳条件。在此基础上,测定不同剂量组孕鼠离体子宫平滑肌张力的变化情况,采用RM-6240BD多道生理信号采集处理系统分析数据,统计结果。结果:选择0.007 U/m L缩宫素、0.008 mol/m L硫酸镁为最佳刺激浓度。随着受试物剂量的增加,加入缩宫素后,各组妊娠子宫平滑肌的频率和张力均呈逐渐上升的趋势,而加入硫酸镁后,各剂量组妊娠子宫平滑肌的活动幅度、频率和张力均呈现降低的趋势,但各剂量组与溶媒对照组相比均未见明显差异(P0.05)。结论:本研究建立了孕鼠离体子宫平滑肌张力的测定方法,该方法可以更好的反映受试物对孕鼠子宫肌的毒性作用,更加全面的评价受试物的生殖毒性。  相似文献

A study was made of the effects of insulin on smooth muscle cells (SMC) of the femoral artery of rats aged 6-8 and 24-26 months. Insulin hyperpolarized the SMC membrane and diminished the tone of smooth muscles of the vessels. These effects were dependent on the hormone concentration and perfusate temperature. In old animals, the sensitivity of smooth muscles of the vessels to insulin and the response to higher concentrations were reduced. It is believed that the changes described may play a definite role in the mechanisms by which the vascular tone is controlled.  相似文献

The effect of severe cold (5 to 10 degrees C) on adrenergic neurotransmission was compared in the isolated cutaneous (saphenous) artery and vein of the dog. The vein contracted to sympathetic nerve stimulation at temperatures as low as 10 degrees C; higher temperatures were needed for the artery to contract. Both blood vessels contracted to exogenous norepinephrine at temperatures as low as 5 degrees C. However, the contractile response to exogenous norepinephrine was less in the saphenous artery, and contractions to high K+ solution were depressed by cooling more in the artery than in the vein. During electrical stimulation of the sympathetic nerves in saphenous arteries and veins previously incubated with labeled norepinephrine, progressive cooling from 37 to 5 degrees C caused a sharp decline in overflow of [3H]norepinephrine and its metabolites. However, overflow of labeled norepinephrine in both blood vessels continued at very cold temperatures. Thus the inability of the saphenous artery to contract to sympathetic nerve stimulation at 10 degrees C can be explained by a greater sensitivity of the arterial smooth muscle to the direct depressant effect of cold, rather than to a differential release or metabolism or norepinephrine in the arterial wall or a loss of responsiveness to norepinephrine at very cold temperatures.  相似文献

The effect of melatonin (MLT) on the isolated rat heart was studied using the standard perfusion conditions (Langendorff preparation) and model of calcium paradox (Ca(2+)-paradox). Ca(2+)-paradox was induced by 1 minute perfusion with Ca(2+)-free Krebs-Henseleit (KH) solution and subsequent 20 minutes perfusion with a normal Ca(2+)-containing KH solution. In MLT group, MLT (10 micromol/l) was in the perfusion solution throughout the experiment. In controls, there was no MLT. VARIABLES: heart rate, coronary flow, systolic and diastolic pressure, +dP/dt max (index of contractility) and -dP/dt max (index of relaxation) were measured at the end of stabilization, i.e. after 30 minutes of standard perfusion and then in the 5th, 10th, 15th, 20th minute after perfusion with Ca(2+)-free KH solution. RESULTS: There was no difference between MLT group and controls in the standard perfusion conditions at the end of stabilization. After perfusion with Ca(2+)-free KH solution, systolic-diastolic difference (in the 10th, 15th, 20th minute), +dP/dt max (in the 5th, l0th, 15th, 20th minute) and -dP/dt max (in the 15th minute) were significantly decreased in MLT group in comparison to controls. CONCLUSION: Melatonin didn't influence rat isolated heart in standard perfusion conditions but it made the heart more susceptible to Ca(2+)-paradox.  相似文献

A sesquiterpenoid Bakkenolide (1), and two steroids, (3β, 22E)-Stigmasta-5, 22-diène-3-ol (Stigmasterol) (2) and stigmasterol 3β-glucoside (3), isolated from the Hertia cheirifolia (L.) chloroform extract, were evaluated respectively for their spasmolytic and anti-inflammatory activities. We note that these natural products were isolated and purified for the first time from the specie Hertia cheirifolia. Their structures have been established by spectroscopy (1 and 2D NMR experiences) and mass spectrometry. Chloroform-, ethyl acetate- and methanol-extracts were also tested for their spasmolytic and anti-inflammatory activities. Spasmolytic and anti-inflammatory screening were based respectively on the contractile response effects on rat isolated smooth muscles and on the dose-related carrageenan induced paw edema in rats. screening of the crude extracts showed spasmolytic and anti-inflammatory positive results. The antispasmodic effect of Bakkenolide was found in the same range as that of Alverine, a standard musculotropic spasmolytic agent.  相似文献

Stimulation of alpha-adrenoceptors was simulated by intraarterial injection of 0.6 mmol CaCl2 in acute experiments on resting muscles of the dog hind leg. It has been demonstrated that alpha-adrenergic stimulation of metabolism in skeletal muscles has a vasodilative effect in the muscle-supplying vessels. It has been also shown that imitation of alpha-adrenergic stimulation effect by intraarterial injection of CaCl2 causes Ca- and alpha-adrenergic-dependent vasodilative reactions. It is suggested that disturbance of vasodilative effects caused by alpha-adrenergic stimulation in the effector tissues, counteracting the direct constrictive alpha-adrenoceptor effect in the smooth muscle cells of the vessel wall, can lead to systemic impairment of the vascular tone.  相似文献

This research was conducted to study the effect of three different sowing dates (15th October, 15th November and 15th December) and two vernalization treatments (5 °C and −20 °C) on leaf structure of Betavulgaris L. cv. Univers. The obtained data are summarized as follows:The maximum values of the most studied parameters; lower epidermis + spongy tissue thickness, midrib, mesophyll tissue, vascular bundle, collenchymatous tissue and number of xylem vessels per arm were found as a result of 15th October sowing date treatment compared with the two other sowing dates. Furthermore, effect of the cooling treatments varied according to the recorded character, sowing date and cooling degree. Most of the vernalization treatments at early sowing dates increased the mesophyll tissue, midrib, number of vascular bundles per transverse section, vascular bundle thickness and number of xylem arms per transverse section.The two studied cooling treatments at 15th October sowing date increased both stomatal index and average number of stomata: average number of epidermis cells compared with the control. Furthermore, 15th October under −20 °C treatment led to small epidermal cells and stomata formation, straight epidermal cell walls and closed stomata in comparison to the control.  相似文献

The effects of rapid cooling on the mechanical and electrical activities of the guinea pig taenia coli and circular muscle of the stomach were investigated. Lowering the temperature from 32° to 10°C (cold shock) depolarized the membrane and increased the membrane resistance in both tissues. However, in the taenia coli, an initial reduction of membrane resistance was observed. In both tissues, contracture evoked by cold shock and rapid relaxation after rewarming, preceded the changes of membrane properties. Displacements of the membrane potential did not modify the amplitude of contracture under cold shock. Caffeine and thymol modified the membrane properties, but the effects of cold shock were still observed. The effects of cold shock were also observed on K-induced contracture. It was postulated that at least two different sites of sequestered bound Ca are located in these smooth muscles and are responsible for evoking the mechanical response. One component possesses a close relation to membrane and the other component is presumably sequestered within the muscle.  相似文献

Cerebrospinal ganglia of 11-, 13-, 15-, 18- and 20-day-old embryos have been studied light- and electron-microscopically. On the 11th day of anlage the ganglia are presented exclusively as neuroblasts: the satellite-cells are formed later from lemmoblasts that install into the ganglion on the 13th-15th days both from the surrounding mesenchyme and from the developing roots and trunklets of the cerebrospinal nerves. In the ganglion they undergo proliferation and further differentiation. On the 15th day simple satellite-neuron interrelationships are formed: one satellite-cell is tightly connected with perikaryons of some neuroblasts and young neurons. On the 18th day around some neurons a double layered capsule is formed from the satellite-cells. Multilayered tunics are formed only on the 10th-15th day after birth. During the process of the capsule formation an unusual reactive character of the satellite-neuron interrelationships is revealed, it is manifested as, performed by the satellite-cells, an active microphagocytosis of the perikaryon cytoplasmic processes and the initial segment of the sensitive neuron main process. In embryogenesis the lemmocyte-axon interrelationships develop earlier than the satellite-neuronal ones.  相似文献
