Broomrapes (Orobanche spp.) are parasitic angiosperms, which attach to the roots of the hosts to take water and nutrients from them. No complete control measures are available to date, but breeding for resistance remains as one of the most feasible and environmentally friendly methods. However, the mechanisms governing the interaction between these parasites and the host are not yet well understood. We studied the cellular changes associated with the resistance to Orobanche crenata in faba bean as mechanisms involved or responsible for resistance. Two cultivars of faba bean, resistant and susceptible to O. crenata infection, were used. The evolution of the infection and the changes in the cell and tissue organisation and wall components of the host cells were followed and evaluated in both genotypes. Samples of compatible and incompatible interactions were fixed and sectioned, and specific cytochemical methods for different cell components were applied, results being analysed under light and epifluorescence microscopy. A higher proportion of O. crenata seedlings unable to penetrate the root was found on the resistant genotype. Reinforcement of cell walls by callose deposition hampers parasite penetration through the cortex. Lignification of endodermal cells prevents further penetration of the parasite into the central cylinder.  相似文献

Field bean (Vicia faba L.) cv. Maris Bead seeds were inoculated with Rhizobium Catalogue No. 1001, supplied by Rothamsted Experimental Station and grown in sand culture supplied with 15N-labelled nitrate at two concentrations. Plants were sampled at intervals throughout their growth for 15N and total N analysis. The rate of nitrate uptake was almost uniform up to pod-fill and was proportional to the nitrate concentration. Nodule weight was slightly depressed by the larger nitrate concentration at all samplings, and there was a corresponding reduction in the amount of atmospheric nitrogen fixed. However, at harvest the bean seeds from plants given most nitrate contained slightly more total N, as the enhanced nitrate uptake outweighed the reduction in fixation.  相似文献

Differences in seed vigour of zero- and high-tannin faba beans were investigated using 25 seed lots of 12 cultivars following earlier reports of poor emergence in the zero-tannin types. Field emergence ranged from 54–96% indicating differences in seed vigour between cultivars all having high laboratory germination (>91%). Seed from zero-tannin accessions with poor emergence had a higher incidence of testa and cotyledon cracking, a smaller percentage of hard seeds, more rapid water uptake, a lower percentage of vital staining of cotyledons and a greater leaching of solutes than high-tannin types. Nevertheless, variation in these characteristics existed between cultivars and lines of both types. Seeds with more cracks in the seed coat and fewer hard seeds imbibed water more rapidly and consequently showed lower levels of vital staining and more cracks in the abaxial surface of the cotyledons. Slower imbibition in polyethylene glycol lessened the incidence of these deleterious characteristics and may provide a practical resolution to the problem of poor field emergence in zero-tannin lines of faba bean with low seed vigour.  相似文献

Bacteria of the faba bean (Vicia faba L.)/Orobanche spp. root environment were evaluated for their potential use as biocontrol agents for the parasitic weed. Bacteria were isolated mainly from the rhizosphere of faba bean as well as from diseased Orobanche underground structures and an Orobanche-suppressive soil from three districts of northern Tunisia. Out of 351 bacterial isolates, 337 were tested for pathogenicity in an inverted pyramidal-shape screening programme including a Lactuca sativa L. seedlings bioassay, root-chamber and pot experiments. In pre-selection screening on L. sativa seedlings, 37 isolates (11%) showed a strong growth inhibitory effect, of which 70 and 84% also had a significant suppressive activity on the pre-emergence structures of O. foetida and O. crenata, respectively, in root-chamber experiments. Among five bacterial isolates selected for pot trials, strain Bf7-9 of Pseudomonas fluorescens showed high biocontrol activity against both species of Orobanche and positively influenced faba bean growth. The bacterium reduced shoot emergence of O. crenata and O. foetida by 64 and 76% and their dry weight by 39 and 63%, respectively, compared with non-inoculated controls. Pseudomonas marginalis strain Nc1-2 exhibited also a tendency to reduce incidence of O. crenata and to improve faba bean performance. Results of the present study suggest that application of naturally occurring rhizosphere bacteria offers an additional approach for biocontrol of Orobanche spp. that can supplement current methods of control in an integrated weed management strategy.  相似文献

Field bean (Vicia faba L.) cv. Maris Bead seeds were inoculated with Rhizobium Catalogue No. 1001, supplied by Rothamsted Experimental Station, and grown in sand culture supplied with a complete nutrient solution which included nitrate at either 1.5 or 6.0 mM. Nodules were detached from the roots at intervals during plant development and their rates of nitrogen fixation estimated by both acetylene reduction and 15N gas technique. There was a constant relationship, independent of nitrate supply, between the results obtained by these two methods at all samplings. The amounts of acetylene reduced divided by a factor of 5.75 gave the amount of true nitrogen fixation; this factor is about twice the theoretical value. It is suggested that this discrepancy arose because, with acetylene, all the electrons available to the nitrogenase were used to form ethylene, whereas during normal fixation only about half the electron supply was used to fix nitrogen, the remainder having been consumed in the production of hydrogen gas.  相似文献

Summary Homozygous mutant lines of field bean selected for (a) improved yielding potential and (b) altered plant architecture and/or physiological response were tested for symbiotic nitrogen fixing ability under field conditions in comparison with their parent cultivar.15N-tracer techniques were applied to determine %N derived from atmosphere. Data were collected on assimilate and nitrogen accumulation and distribution among various plant parts during two stages of reproductive growth.Symbiotic nitrogen fixation was closely correlated with total plant top biomass and nitrogen yield. A similar close association was found between crop yield and nitrogen harvest index. Both harvest indices tended to be negatively correlated with stage of maturity and with the amount of N derived from air per unit of area. The generally high %N derived from symbiotic N2 fixation and its comparatively small variability implies that this parameter may be difficult to improve inVicia faba under field conditions.It is concluded, that the main genetic potential for improving the amount of biological nitrogen fixation in this crop depends upon factors that promote high photosynthetic productivity and efficient N-use under appropriate agronomic conditions and with effective rhizobial associations. The establishment of rational ideotypes with a possitive impact on yield appears to be of practical significance for increasing the amount of symbiotically fixed nitrogen.  相似文献

利用蚕豆根尖研究环磷酰胺的遗传毒性效应, 结果表明:环磷酰胺(0.1~5.0 mg/mL)能够降低蚕豆根尖细胞有丝分裂指数, 使根尖细胞中具有微核、核出芽及核固缩的细胞明显增多, 并诱发染色体结构和行为异常, 产生染色体断片、滞后和桥。环磷酰胺处理组根尖中具有核固缩和微核的细胞数呈剂量依赖性增加, 且与作用时间呈正相关, 而分裂指数的降低也具有剂量和时间效应关系。研究结果表明, 低浓度长时间接触或高浓度短时间接触环磷酰胺均可产生遗传毒害, 因此, 有关的作业人员应注意防护。  相似文献

用激光扫描共聚焦显微技术,初步研究广谱性蛋白激酶抑制剂星型孢菌素(STS)对蚕豆气孔运动的调控效应.结果表明:(1)光下STS对气孔开度无影响但暗中显著促进气孔开放,表明蛋白激酶参与光/暗对气孔运动的调控,光下蛋白激酶活性低而暗中高;(2)与H2O2清除剂抗坏血酸(ASA)和NO清除剂羧基-2-苯-4,4,5,5-四甲基咪唑-1-氧-3-氧化物(cPTIO)一样,STS既降低暗处理和光下外源H2O2、硝普钠(SNP)处理保卫细胞H2O2、NO水平,也促进气孔开放,表明暗中蛋白激酶通过抑制H2O2、NO清除机制提高保卫细胞内源H2O2、NO水平并促进气孔关闭.  相似文献

A procedure for multiple shoot formation from cotyledonary node explants of faba bean (Vicia faba L.cv.S.Ghdar) cultured on MS medium containing benzyladenine (BA) and thidiazuron (TDZ) was developed. Explants on medium with TDZ in combination with BA produced a higher number of shoots than with either cytokinin alone. The highest number of shoots was obtained when explants from 7-day-old seedlings were cultured on MS medium supplemented with TDZ and BA (2 mgl–1 each) for 31 days before transfer to hormone-free MS medium for elongation. Shoots produced in vitro were rooted on half-strength agar-solidified MS basal medium or with 0.25 or 0.5 mgl–1 naphthalenacetic acid (NAA) prior to transfer to green house conditions. This procedure was found to be applicable to seven other cultivars of faba bean from widely diverse provenances. Thus, it can be advantageously applied to the production of transgenic faba bean plants.  相似文献

BACKGROUND AND AIMS: The aim of this study was to investigate the effects of the interactions between the microbial symbionts, Rhizobium and arbuscular mycorrhizal fungi (AMF) on N and P accumulation by broad bean (Vicia faba) and how increased N and P content influence biomass production, leaf area and net photosynthetic rate. METHODS: A multi-factorial experiment consisting of four different legume-microbial symbiotic associations and two nitrogen treatments was used to investigate the influence of the different microbial symbiotic associations on P accumulation, total N accumulation, biomass, leaf area and net photosynthesis in broad bean grown under low P conditions. KEY RESULTS: AMF promoted biomass production and photosynthetic rates by increasing the ratio of P to N accumulation. An increase in P was consistently associated with an increase in N accumulation and N productivity, expressed in terms of biomass and leaf area. Photosynthetic N use efficiency, irrespective of the inorganic source of N (e.g. NO3- or N2), was enhanced by increased P supply due to AMF. The presence of Rhizobium resulted in a significant decline in AMF colonization levels irrespective of N supply. Without Rhizobium, AMF colonization levels were higher in low N treatments. Presence or absence of AMF did not have a significant effect on nodule mass but high N with or without AMF led to a significant decline in nodule biomass. Plants with the Rhizobium and AMF symbiotic associations had higher photosynthetic rates per unit leaf area. CONCLUSIONS: The results indicated that the synergistic or additive interactions among the components of the tripartite symbiotic association (Rhizobium-AMF-broad bean) increased plant productivity.  相似文献

Nodulated faba-beans ( Vicia faba L. var. minor) exhibiting high rates of N2 fixation (133 μmol C2H4 g−1 dry weight h−1), were subjected to water restriction. A loss of C2H2 reduction due to water stress was always associated with a decline of the leghemoglobin content for each of the 4 decreasing values of Ψmod. Electron micrographs showed ultrastructural alterations of the fixing tissue, which affected both partners and increased with the severity of water stress. In the nodule cytosol, the alkaline proteolysis approximately doubled when Ψmod decreased from −0.55 MPa to −1.55 MPa. Concomitantly, an increase of the nodule intracellular pH from 6.3 to 7.0 was observed. Proteolysis was due to serine proteases, exhibiting a pH-optimum of 8 and which actively degraded purified leghemoglobin in vitro (Km=100 μ M ). The degradation of leghemoglobin during water stress may contribute to the loss of C2H2 reduction and may affect the pattern of recovery upon rewatering.  相似文献

BACKGROUND AND AIMS: A brassinosteroid-deficient mutant faba bean (Vicia faba 'Rinrei') shows dwarfism in many organs including pods and seeds. 'Rinrei' has normal-sized seeds together with dwarf seeds, suggesting that dwarfism in the seed may be indirectly caused by brassinosteroid deficiency. The mechanism of seed size reduction in this mutant was investigated. METHODS: The associations between seed orientation in the pod, seed numbers per pod and pod lengths with seed sizes were analysed in 'Rinrei' and the wild-type plant. KEY RESULTS: 'Rinrei' seeds are tightly arranged in pods containing two or three seeds. Seed size decreased as the number of seeds per pod increased or as the length of the pod decreased. Where no physical restriction occurred between seeds in a pod, the wild-type faba bean seeds had a nearly constant size regardless of seed number per pod or pod length. 'Rinrei' seeds in pods containing single seeds were the same size as wild-type seeds. Brassinolide treatment increased the seed size and the length of pods containing three seeds in 'Rinrei'. CONCLUSION: Seed size of 'Rinrei' is mainly regulated through a reduction of pod length due to brassinosteroid deficiency; physical restriction within pods causes a reduction in seed size. These results suggest a possible mechanism for increasing faba bean yields to optimal levels.  相似文献

Abstract. Gas exchange rates of 4-week-old faba bean plants were measured after exposure to ozonc (120 μg m3) in an open top chamber for 8 h per day over a period of 2 weeks. The exchange rates were compared with those of control plants. Plants exposed between mid-May and the end of July 1987 showed a minor negative effect on stomatal conductance, while there was no effect on photosynthesis and respiration. Plants exposed between the end of August and early October showed a negative effect on both stomatal conductance and photosynthesis. In addition, the dark respiration rate was slightly increased. It is concluded that ozone can have a direct effect on the stomata as well as on the photosynthetic system and that the stomata are more sensitive than the photosynthetic system.  相似文献

Faba bean (Vicia faba) plants were inoculated with rhizobia and then their sap was infected with broad bean mottle bromovirus (BBMV) or bean yellow mosaic potyvirus (BYMV) in a field experiment. Both viral infections significantly decreased shoot and root dry weight, number of nodules, nodule dry weight, numbers of flowers and pods/plant, total plant N, grain yield and N2 fixation. However, inoculation withRhizobium leguminosarum significantly increased all these parameters, both in healthy and virus-infected plants. Two field experiments were conducted during 2004 and 2005 cropping seasons at Adet Agricultural Research Center, Ethiopia to assess yield losses caused by chocolate spot(Botrytis fabae) of faba bean in sole and mixed cropping systems using two cultivars. Cropping systems were sole faba bean (FB), faba bean mixed with field pea (FB: FP), barley (FB: BA) and maize (FB: MA). Mancozeb was sprayed at the rate of 2.5 kg a.i/ha at 7-, 14- and 21-day interval to generate different levels of chocolate spot disease in all the four cropping systems, and unsprayed control was also included. The treatments were arranged in a randomised complete block design (RCBD) with four replications. FB: MA mixed cropping significantly reduced disease severity and the area under disease progress curve (AUDPC) and increased faba bean grain yield. The highest faba bean grain yield among the three mixed croppings under different spray schedules was obtained from FB: MA mixed cropping in both 2004 and 2005 (2.56 and 3.74 t/ha, respectively) cropping seasons. There were highly significant yield differences (P < 0.05) among the spray intervals of mancozeb in both seasons. The highest grain yield (4.9 t/h) was recorded from the 7-day spray interval in 2005. The unsprayed faba bean had a lower grain yield (1.9 t/ha in 2004 and 2.3 t/ha in 2005) compared to the sprayed plots. The highest relative yield loss (67.5%) was calculated in 2005 from FB: FP mixed cropping in unsprayed plots. The relative yield losses in the unsprayed plots were in the range of 35.8–41.5% in 2004 and 52.6–67.5% in the 2005 cropping season. Severity and AUDPC were inversely correlated with faba bean grain yield. Significant differences were recorded in the 100-seed weight and days to maturity (DM). The unsprayed plots had shorter DM ranging from 126 to 128.5 day (except FB: MA mixed cropping) in 2004 and 122–123.9 days in 2005. In the sprayed plots DM was relatively longer than the unsprayed plots. A higher seed weight was recorded in the sole FB (56 g) and FB: MA (55 g) mixed cropping, and the lowest value of 100-seed weight was recorded from FB: FP (53 g) mixed cropping. The productivity of the mixed cropping evaluated by land equivalent ratio (LER) exceeded that of sole cropping. Faba bean grain yield was highly influenced by the severity of chocolate spot. The disease affects the DM, forcing early maturing of the plants.  相似文献

The possibility that the herbicide glyphosate (N-phosphonomethylglycine) may be taken up in plant cells via a phosphate transporter of the plasma membrane was investigated using protoplasts of broad bean leaves ( Vicia faba L.). Phosphonoformic acid, a powerful inhibitor of phosphate transport in animal cells, was first demonstrated to be a competitive inhibitor of phosphate uptake inbroad bean protoplasts. Glyphosate was able to inhibit phosphate uptake into the protoplasts, and to protect partially the phosphate transporter from inhibition by phosphonoformic acid. Concentration dependence studies showed that glyphosate uptake exhibited a saturable phase at low glyphosate concentrations (0. 5 to 3 μ M ), superimposed by a linear uptake at higher concentrations (up to 100 μ M ). Inhibition of glyphosate uptake by para -chloromercuribenzene sulphonic acid, sodium azide and carbonyl-cyanide- m -chlorophenylhydrazone was much stronger at 1 than at 100 μ M glyphosate. Kinetics indicated that the saturable component of glyphosate transport was competitively inhibited by either phosphate or phosphonoformic acid. The effects of fluridone on guard cell morphology, chloroplast ultrastructure and accumulation of drought stress-induced abscisic acid (ABA) were studied in Vicia faba L. plants grown under different light conditions. Drought stress was induced by allowing the leaves to lose 12% of their fresh weight. The appearance of defective and undeveloped stomata, and chloroplasts with a destroyed thylakoid membrane system was found in fluridone-treated plants grown at a photosynthetic photon flux (PPF) of 600 μmol m-2 s-1. Plants grown at a PPF of 40 μmol m-2 s-1 had diminished levels of ABA after imposition of dehydration. Fluridone treatment reduced the level of ABA in both unstressed and dehydrated leaves. Accumulation of ABA in the control plants was considerably reduced when they were exposed to dark periods of 24, 48 and 72 h just before imposition of the stress. Twenty-four hours after the dark treatment dehydration of the leaves resulted in a 3-fold decrease in the level of stress-induced ABA, and 72 h after dark treatment the amount of stress-induced ABA approximated the prestressed values. Fluridone-treated plants failed to accumulate ABA under water stress. In addition to functionally active chloroplasts, well-developed and functional stomata are required for drought stress to elicit a rise in ABA.  相似文献

Leaflets of Vicia faba L. (faba bean) were used to determine whether the mechanical forces resulting from the turgor potentials (Φp) of the larger epidermal cells neighbouring guard cells play a significant role in regulating stomatal aperture. When Φp, of epidermis and Φp of bulk leaflet tissue were compared at midday, Φp of epidermis were only 15–25% those of bulk leaflet tissue at all but the most negative leaflet water potentials (Φ). When plants were bagged to increase Φ by reducing vapour pressure differences between leaflets and air, Φp of bulk leaflet tissue increased to predawn values, but Φp, of epidermis increased to only = 20% of predawn values and stomata opened to their widest apertures. Stomatal apertures were positively correlated with Φp of bulk leaflet tissue but they were not correlated with Φp of epidermis. Reductions in epidermal Φp, began predawn, before stomata were open, and reached minimum values at midday, when stomata were open. We conclude that, in Vicia faba, (1) reduction of Φp of epidermal cells begins predawn, reducing the counterforce to stomatal opening that would exist if full epidermal turgor were maintained throughout the day, and (2) changes in Φp, of leaf epidermal cells do not play a significant role in regulating stomatal aperture.  相似文献

应用蚕豆根尖微核技术研究了Cd(1mgL^-1~25mgL^-1)、Pb(1mgL^-1~25mgL^-1)及其复合污染对根尖细胞的毒害作用,分析了重金属及复合重金属污染对蚕豆根尖细胞有丝分裂和微核及染色体畸变的影响。结果表明:Cd、Pb及其复合处理对蚕豆根尖细胞有丝分裂及染色体行为存在不同程度的影响。具体表现为:低浓度促进细胞分裂,高浓度则抑制细胞分裂;对染色体畸变的影响表现为:当浓度为1.0mgL^-1时Cd即产生显著效应,而Pb浓度达10.0mgL^-1时,表现出稳定的,强度较大的诱变效应,且随浓度的增加毒害增强。Cd、Pb间存在交互作用,Cd达低浓度时与Pb的交互作用明显而高浓度则不明显,且Cd、Pb间存在部分拮抗作用。  相似文献
