B型烟粉虱Bemisia tabaci Gennadius是一种重要的世界性农业害虫。玫烟色棒束孢Isaria fumosorosea是防治烟粉虱的一种重要的虫生真菌。对不同温度(20、23、26、29、32℃)、不同湿度(53%、65%、75%、85%、95%)下玫烟色棒束孢IF‐1106菌株分生孢子萌发及其对烟粉虱的致病力进行了测定。结果表明:温度对孢子的萌发有显著影响,26℃时孢子萌发率最高。当相对湿度低于75%时,孢子不萌发或萌发率较低;当相对湿度为85%–95%时,孢子萌发率显著升高。26℃时,烟粉虱2龄若虫的累计死亡率最高。随着相对湿度增大,病菌的致病力增强。当相对湿度为53%–95%时,烟粉虱2龄若虫累计死亡率从54.55%增加到88.89%。玫烟色棒束孢的致病力与孢子萌发率呈正相关,但温度比相对湿度对其影响更明显。结果表明,玫烟色棒束孢IF‐1106菌株侵染烟粉虱的最佳条件是温度26℃和相对湿度大于85%。  相似文献

The strain IfB01 of Isaria fumosorosea (Paecilomyces fumosoroseus) is a promising myco-insecticide candidate for control of Bemisia tabaci. Joint action of strain IfB01 and four chemical insecticides [spirotetramat (Spi), acetamiprid (Ace), imidacloprid (Imi) and thiamethoxam (Thi)] against the whitefly B. tabaci was evaluated using the cooperative virulence index (c.f.). Substantial synergistic actions were found in the mixtures of IfB01 conidia with Spi, Imi and Thi 2–4 days after treatment. The greatest synergism was recorded in the mixture of IfB01 (2.5 × 106 conidia/L) and Imi (12.5 mg/L), which had a c.f. value of 184 at day 3 after treatment. Furthermore, the shorter LT50 values recorded in mixtures indicated that effectiveness improved when the conidia were applied with the chemical insecticides. However, no substantial synergisms were recorded with Ace and from day 5 onwards.  相似文献

We determined host plant effect on susceptibility of whitefly Bemisia tabaci to the entomopathogenic fungus Beauveria bassiana under controlled conditions. Insects were reared on cucumber, eggplant, tomato or cabbage. Fungal suspensions of 1×104, 105, 106, 107 and 108 conidia/mL were applied on second-instar nymphs. Nymphal survival significantly differed among different host plant species on which the nymphs were reared. Ten days after inoculation with 1×108 conidia/mL, percent survival was 4.2±0.7, 9.6±0.4, 13.4±0.8, and 24.3±0.9% on cucumber, eggplant, tomato and cabbage, respectively. Average survival times of nymphs were also significantly influenced by host plant species. After inoculation with 1×108 conidia/mL, survival times were 4.8±0.15, 6.0±0.11, 5.7±0.13, and 6.2±0.08 days for nymphs reared on cucumber, eggplant, tomato, and cabbage, respectively. Virulence also differed depending on host plant species; 10 days after inoculation, LC50 values were 4.6×104, 1.6×105, 4.2×105 and 2.1×106 conidia/mL on cucumber, eggplant, tomato and cabbage, respectively. Abstract The sweetpotato whitefly, Bemisia tabaci, has been a destructive pest in China for over the past two decades. It is an extremely polyphagous insect, being recorded feeding on hundreds of host plants around the world. Potential host plants and natural enemies of B. tabaci in the south, southeast, middle, north and northwest of China were investigated during the last decade. In total 361 plant species from 89 families were recorded in our surveys. Plants in the families Compositae, Cruciferae, Cucurbitaceae, Solanaceae and Leguminosae were the preferred host species for B. tabaci, which therefore suffered much damage from this devastating pest due to their high populations. In total, 56 species of parasitoids, 54 species of arthropod predators and seven species of entomopathogenic fungi were recorded in our surveys. Aphelinid parasitoids from Encarsia and Eretmocerus genera, lady beetles and lacewings in Coleoptera and Neuroptera were found to be the dominant arthropod predators of B. tabaci in China. The varieties of host plant, their distribution and the dominant species of natural enemies of B. tabaci in different regions of China are discussed.  相似文献

Isaria fumosorosea is an entomopathogenic fungus that is used as a control alternative for nymphs and adults of Bemisia tabaci. Currently there are some commercial products, however, in greenhouse or field, these do not reach the levels of control as in the laboratory because the viability of the spores decreases as a result of the conditions of application of these products in situ. The objective of this work is to implement, through agroecological data, a system of biotechnological traps based on I. fumosorosea to increase the control efficiency mainly of adults of B. tabaci in strawberry greenhouses. One way to quantify the degree of infestation of a crop is the use of yellow traps, likewise to determine the spatial distribution of adults. The Taylor method [(1984). Assessing and interpreting the spatial distributions of insect populations. Annual Reviews of Entomology, 29, 321–357) was used in five different strawberry cultivation models, finding aggregate and regular distributions. Finally, once the crop model with the highest degree of infestation was selected, the designed traps were tested and mortalities were obtained between 50% and 90% in both the laboratory and the greenhouse. The biotechnological traps based on I. fumosorosea both in the laboratory and in the greenhouse had statistically the same effect as those used under the traditional method used in the field that is aspersion; therefore, this alternative method of application can be a tool important for the biological control of this pest.  相似文献

Recently, the Q biotype of tobacco whitefly has been recognized as the most hazardous strain of Bemisia tabaci worldwide because of increased resistance to some insecticide groups requiring alternative strategies for its control. We studied the susceptibility of this biotype of B. tabaci to 21 isolates of Beauveria bassiana, three isolates of Isaria fumosorosea, one isolate of I. cateni, three isolates of Lecanicillium lecanii, one isolate of L. attenuatum, and one isolate of Aschersonia aleyrodis. These isolates were evaluated on pruned eggplant seedlings, at a concentration of 108 conidia/mL (deposited at 6000±586 conidia mm?2). The mortality based on mycosis varied from 18 to 97% after 6 days. Isaria fumosorosea isolate Pf04, B. bassiana isolates Bb06, Bb12, and L. lecanii L14 were found the most effective. Furthermore, five isolates were chosen for concentration–mortality response assays and compared to B. bassiana GHA as a standard. The numbers of nymphs infected by fungi were correlated with the spore concentration. L. lecanii L14 and I. fumosorosea Pf04 had the shortest LT50 at 3.5 and 3.3 days at 6000±586 conidia mm?2. Mortality declined and LT50 values were longer as the concentration of conidia was reduced. The LD50 values were calculated as 87, 147, 191, 263, and 269 conidia mm?2 for isolates L14, GHA, AS1260, Bb13, and Pf04, respectively. These results indicated that the Q biotype of sweetpotato whitefly was susceptible to the five isolates of entomopathogenic fungi and these isolates have potential to be developed as microbial pesticides for whitefly control.  相似文献

A laboratory bioassay was developed to evaluate strains of Isaria fumosorosea Wize, against Diaphorina citri. Up to 100% of adult psyllids were killed at concentrations between 106 and 107 blastospores/ml after 12 days, with derived LC50 values (at 7 days post treatment) between 1.4 × 105 and 2.0 × 106 blastospores/ml for strains ARSEF 3581, FE 9901 and Apopka-97. A significantly higher value (1.5 × 107) was obtained with a conidial formulation of Apopka-97. Average survival times were dosage dependent, i.e. between 10.2 days at 103 blastospores/ml and 3.5 days at 108 blastospores/ml. Rates of mycosis were also dosage dependent, with up to 100% sporulation on cadavers at 108 blastospores/ml but declining at lower concentrations. The Apopka-97 strain (commercially available as PFR-97) was tested against established D. citri infestations in potted citrus in greenhouse cages. Treatments at label rates reduced psyllid populations by approximately 50% over 3 weeks. The combination of PFR-97 with emulsifiable oils (0.25% v/v) did not increase psyllid mortality compared with either agent alone. Imidacloprid applied as a drench killed 100% of psyllids within 3 weeks. Subsequent greenhouse tests during humid conditions were hampered by natural dissemination of I. fumosorosea to untreated psyllids, suggesting that this fungus is spread by air movement and may be highly effective under very humid conditions. In later tests, a Cladosporium sp. rapidly colonised psyllid cadavers and leaf surfaces, but was not pathogenic in laboratory tests. Our studies confirm the potential of I. fumosorosea to be used in IPM strategies for D. citri that rely on other tactics, such as insecticidal oils and native or introduced biological control agents.  相似文献

Isaria fumosorosea is one of important entomopathogenic fungi showed a good potential in controlling Bemisia tabaci. The effects of I. fumosorosea Ifchit1 mutant (Ifchit1 gene deletion mutant) on the mortality, oviposition, and host immunological response of B. tabaci, on Brassica campestris L. plant, were evaluated under laboratory conditions. The wild-type fungal strain infection significantly increased insect mortality and reduced the oviposition effeciency of B. tabaci, whereas the Ifchit1 mutant was much less effective, resulting in higher survival and ovipositing of B. tabaci. The activities of four insect enzymes were examined during a time course of fungal infection. Insect phenoloxidase, perioxidase, and catalase activities were decreased in whiteflies treated with the wild type and mutant I. fumosorosea strain at 12–36?h post treatment. However, these enzyme activities increased in fungal-treated whiteflies as compared to controls between 36 and 60?h post-infection, reaching peak values. Superoxide dismutase activity in fungal-treated whiteflies was higher than that in controls during the entire experimental time course examined. The overall enzyme activity profiles in Ifchit1 mutant-treated whiteflies were significantly different from wild-type strain treatments. Our results showed that loss of the Ifchit1 gene in I. fumosorosea affects whitefly mortality, ovipositioning and various antioxidant enzyme activities, providing new insights into the role of chitinases in I. fumosorosea-insect host–pathogen interactions.  相似文献

The effects of eight commonly used fungicides: propamocarb, fenarimol, triadimefon, procimidone, azoxystrobin, carbendazim, cooper oxychloride and Trichoderma harzianum on germination, mycelial growth and virulence of Isaria fumosorosea (Wize) Brown & Smith (Ascomycota: Hypocreales) was studied. The greatest effect on germination was observed by azoxystrobin, followed by carbendazim, triadimefon and procimidone. Propamocarb, fenarimol, cooper oxychloride and T. harzianum did not affect conidial germination with germinations of 95, 93, 79 and 84%, respectively. Virulence was evaluated against early fourth instar nymphs of Trialeurodes vaporariorum (Hemiptera: Aleyrodidae). The mix containing fungal conidial suspensions plus cooper oxychloride or fenarimol presented a significant reduction in the mortality in comparison with spore suspension itself. This study suggests that the most appropriate fungicides for use in Integrated Pest Management Programs against T. vaporariorum in combination with I. fumosorosea are propamocarb and T. harzianum.  相似文献

烟粉虱Bemisia tabaci(Gennadius)成虫对寄主植物选择行为的实验结果表明,烟粉虱是通过植物广谱的绿色气体识别植物材料,并进一步受到绿色的诱导。通过烟粉虱在适宜与非适宜寄主植物上滞留时间的差异,可以看出烟粉虱是通过口针刺探的寄主评估过程逐渐实现寄主偏好性的,存在一个短的时滞。综合来看,烟粉虱的寄主选择是通过嗅觉、视觉和味觉共同参与的一个决策过程,以寄主植物的气味、颜色和质量为线索,逐步定位到适宜的寄主植物上,因此在寄主选择过程中存在很大的可塑性。  相似文献

Effects of entomopathogenic fungus Isaria fumosorosea on biological characteristics and life table parameters of Axinoscymnus cardilobus, a predator of whiteflies, were studied using five different conidial concentrations under laboratory conditions. Non-significant differences were observed among different fungal concentrations on the percent survival of all immature stages. The survival of A. cardilobus on different host plants did not differ significantly. The prey consumption of third instar A. cardilobus on Bemisia tabaci treated with different fungal concentrations differed significantly at different time intervals. The developmental periods for all immature stages (from eggs, first, second, third, fourth instar nymph and pupae up to emergence) among the treatments were not statistically significant when compared to that of control. In the present study, fecundity, longevity, egg viability and life table parameters of females were not statistically significant over the different concentrations. It can be concluded that control strategies tested are compatible to a greater extent and incorporation of these have promising prospects for control of whitefly.  相似文献

为探讨利用趋避作用防治烟粉虱的可能性,进行了3方面试验:1)烟粉虱Bemisia tabaci (Gennadius) 对黄瓜、花椰菜、油菜和莴苣等4种蔬菜的寄主选择性; 2)互喷烟粉虱不同嗜好的寄主植物汁液对其寄主选择性的影响; 3)不同间作方式对黄瓜叶上烟粉虱成虫密度的影响。结果表明,烟粉虱对供试植物有明显的选择性,其嗜好程度为花椰菜>黄瓜>油菜>莴苣。喷施莴苣和花椰菜汁液对烟粉虱的寄主选择性均有显著影响,喷施莴苣原液后,花椰菜和黄瓜上的成虫数分别减少79.6%和87.4%,花椰菜上的着卵量减少84.3%;喷施花椰菜原液后,莴苣上的成虫数和着卵量分别增加82.4%和79.2%,表明不同蔬菜中的内含物是引起烟粉虱对寄主选择性的重要原因。在黄瓜中间作莴苣可使黄瓜叶上烟粉虱成虫数平均减少76.5%;间作花椰菜时,通过诱集作用,也可使黄瓜叶上的成虫数平均降低69.7%。这些结果为利用喷施蔬菜汁液和间作方式控制烟粉虱危害提供了理论依据。  相似文献

【目的】明确烟粉虱Bemisia tabaci MED隐种抑制蛋白基因Btarrestin是否参与吡虫啉耐药性。【方法】根据已有烟粉虱转录组数据,利用RT-PCR克隆Btarrestin的全长序列,进行生物信息学分析。通过qPCR分析Btarrestin在烟粉虱MED隐种各个龄期(卵,1-2龄、3龄、4龄若虫,雌、雄成虫)以及吡虫啉处理(100 mg/L)后成虫中的表达量变化,明确其时空表达模式。利用RNAi技术沉默Btarrestin,观察沉默前后Btarrestin表达量和烟粉虱MED隐种成虫死亡率变化。【结果】成功克隆烟粉虱MED隐种Btarrestin的cDNA序列(GenBank登录号: MK204377),编码区全长1 227 bp,编码409个氨基酸,预测所编码的蛋白分子量约为45.33 kD,理论等电点(pI)为8.38。保守结构域分析表明,Btarrestin具有Arrestin_N和Arrestin_C两个超家族保守结构域,符合抑制蛋白家族特征。分子系统树分析表明,Btarrestin与褐飞虱Nilaparvata lugens的Arrestin亲缘关系最近。Btarrestin的表达量随烟粉虱MED隐种的生长发育逐渐升高,成虫期表达量最高。100 mg/L吡虫啉处理成虫24 h后Btarrestin的表达量较对照增加了2.39倍。RNAi干扰Btarrestin后进行生物测定,烟粉虱MED隐种成虫的死亡率上升了31.27%。【结论】Btarrestin可能与烟粉虱MED隐种对吡虫啉的耐药性有关。  相似文献

Lipases are important cuticle degrading enzymes involved in the infection process of entomopathogens by hydrolysing the ester bonds of lipoproteins, fats and waxes present in the insect integument. Production of extracellular lipase by Isaria fumosoroseus (Cordycipitaceae; Hypocreales) isolate IF28.2 was investigated using different combinations of basal medium components. The effect of different vegetable oils added to a basal medium at different concentrations to improve enzyme production was evaluated. Maximum lipase activity (125.33±2.96 U/mL) as well as maximum biomass production (22.36±0.99 mg/mL) was observed for olive oil when used at a concentration of 2% (v/v) of the basal medium. In the presence of surfactants, the highest lipase activity occurred when SDS and Tween 80 were added at the time of fungal inoculation. SDS proved to be the best surfactant having 110.66±3.52 U/mL lipase activity. The effects of the divalent metal ions (iron and magnesium) on lipase activity were also studied. Iron inhibited, whereas magnesium slightly increased lipase activity. The optimum pH for lipase production was 5.7 while 32°C proved to be the best temperature for lipase production.  相似文献

An assessment of the influence of water nanosilver suspension was made at a concentration of 10 mg·L?1 on biological material (i.e., vegetative mycelium and conidia of Isaria fumosorosea entomopathogen) on a background of the silver nitrate ionic form used. Conidia of I. fumosorosea treated with silver nitrate for more than 168 h were completely deactivated. The application of nanosilver on Isaria hyphae resulted in a quantitative limitation of mycelium growth and its weaker sporulation after culturing compared to the control. The pathogenicity of a conidial suspension obtained from such culturing towards test insects did not diverge from that observed in the standard culture. No obvious toxic effects of nanosilver were observed on I. fumosorosea conidia. Isaria conidia, after exposure to nanosilver over a period between 1 and 800 h, initially demonstrated weaker vegetative mycelium formation in culture on solid medium and, as a consequence, this mycelium often sporulated in a poorer manner. In one case, there was a significant stimulation of the sporulation process for nanosilver treatment before culture for 168 h. Concurrently, conidial suspensions obtained from the culture after exposure of over 168 h to nanosilver exhibited enhanced pathogenicity towards test insects, which may be considered a beneficial phenomenon in terms of the function played by Isaria in the whole environment. The reaction of conidia with nanosilver indicates the deactivation of conidia cells in suspensions and a possibility of selection in increased pathogenicity.  相似文献

双生病毒科(Geminiviridae)菜豆金黄花叶病毒属病毒是一类重要的植物病毒,主要危害番茄、烟草、棉花等多种经济作物,自然条件下主要由介体昆虫烟粉虱Bemisia tabaci传播。菜豆金黄花叶病毒属病毒在田间的暴发受多种因素的影响,其中一个最重要的因素就是其介体昆虫烟粉虱。因此,明确烟粉虱在田间传播和扩散特定病毒中的作用及影响因素,对于解析病毒病流行的生物学基础具有重要意义。本文综述了烟粉虱对菜豆金黄花叶病毒属病毒的传播及影响因素,并讨论了病毒对媒介昆虫的适应及其机制。菜豆金黄花叶病毒属病毒和烟粉虱都为全球分布的有害生物,通过生物信息学的分析发现,两者都呈现地域相关的遗传多样性。而在生物学的研究中发现,烟粉虱隐存种往往对与其起源于同一地区的病毒具有较高的传播效率。这些发现为进一步解析烟粉虱对菜豆金黄花叶病毒属病毒的传播提供了重要的参考。  相似文献

烟粉虱Bemisia tabaci是一个复合种,它具有的生物型分化、较强的传播病毒的能力和抗药性、较快的繁殖速率等特征使其成为我国农业生产中重要害虫之一。本研究利用细胞色素线粒体氧化酶Ⅰ基因,对采集自江苏、广东和云南三省的烟粉虱样本进行了生物型鉴定,并对烟粉虱生物型与寄主植物之间的关联性开展了调查。结果表明,在广东和云南省,都存在未鉴定的土著种群与入侵的B型、Q型共存的现象;同时,在本研究中广东省尚未采集到Q型烟粉虱,而在江苏采集到的粉虱样本全部为入侵型。研究结果还表明,相对于入侵种而言,土著种群显示出更强的寄主植物趋同性;丰富的寄主植物以及本身具有的多食性特性有助于B型、Q型等生物型在世界各地的广泛入侵。  相似文献

The silverleaf whitefly is one of the most important pests found commonly in Iran and in the world. The development of alternative methods instead of chemical applications is necessary in pest management for human health and environment protection. The objective of this research was to assess the effects of extracts of two medicinal plant species: Calotropis procera and Allium sativum, and a formulation containing azadirachtin on fecundity and fertility of the silverleaf whitefly, grown on greenhouse tomato plants. The effects were compared to that of pymetrozine, a synthetic insecticide. According to the results, there was a significant difference among treatments for all reproductive parameters. Gross fecundity rates for pesticides control, herbal extract control, C. procera extract, A. sativum extract, azadirachtin and pymetrozine were 184.75, 146.72, 80.11, 82.18, 63.06 and 55.96 eggs, respectively. These herbal extracts were effective against this pest and they can be the suitable choices for replacing these chemical insecticides.  相似文献
