We investigated emersion-induced nitrogen (N) release from Porphyra umbilicalis Kütz. Thallus N concentration decreased during 4 h of emersion. Tissue N and soluble protein contents of P. umbilicalis were positively correlated and decreased during emersion. Growth of P. umbilicalis did not simply dilute the pre-emersion tissue N concentration. Rather, N was lost from tissues during emersion. We hypothesize that emersion-induced N release occurs when proteins are catabolized. While the δ15N value of tissues exposed to emersion was higher than that of continuously submerged tissues, further discrimination of stable N isotopes did not occur during the 4 h emersion. We conclude that N release from Porphyra during emersion did not result from bacterial denitrification, but possibly as a consequence of photorespiration. The release of N by P. umbilicalis into the environment during emersion suggests a novel role of intertidal seaweeds in the global N cycle. Emersion also altered the physiological function (nitrate uptake, nitrate reductase and glutamine synthetase activity, growth rate) of P. umbilicalis and the co-occurring upper intertidal species P. linearis Grev., though in a seasonally influenced manner. Individuals of the year round perennial species P. umbilicalis were more tolerant of emersion than ephemeral, cold temperate P. linearis in early winter. However, the mid-winter populations of both P. linearis and P. umbilicalis, had similar temporal physiological patterns during emersion.  相似文献

Basic c-type cytochromes homologous with plant and animal mitochondrial cytochrome c have been isolated and purified from Rhodymenia palmata and Porphyra umbilicalis. The N-terminal regions have been analysed using a Beckman 890C automatic sequencer. When compared to animal cytochrome c, the Rhodymenia cytochrome c has an unblocked N-terminal tail of 10 amino acids, whereas Porphyra has an unblocked N-terminal tail of only a single amino acid.  相似文献

Cellulose has been characterized from isolated cell walls of the conchocelis phases of both Porphyra umbilicalis and P. leucostricta. Evidence for cellulose II (regenerated cellulose) in Schweitzer's reagent extracts was provided by X-ray powder analysis and paper chromatography of partial hydrolyzates. The presence of cellulose in the conchocelis phase of species of Porphyra provides evidence for the continuity of cell wall composition within the Rhodophyta. Adoption of a classification scheme incorporating consolidation of all red algal orders under the single class Rhodophyceae is proposed.  相似文献

Molecular biotechnology of marine algae is referred to as the biotechnology on the identification, modification, production and utilization of marine algal molecules. It involves not only the manipulation of macromolecules such as DNA, RNA and proteins, but also deals with low molecular weight compounds such as secondary metabolites. In the last decade, molecular systematic researches to investigate the relationship and to examine the evolutionary divergence among Chinese marine algae have been carried out by Chinese scientists. For example, RAPD has been widely used in several laboratories to elucidate genetic variations of the reds, such as Porphyra, Gracilaria, Grateloupia and the greens such as Ulva and Enteromorpha. Some important data have been obtained. The study on molecular genetic markers for strain improvement is now in progress. In 1990s, genetic engineering of economic seaweeds such as Laminaria, Undaria, Porphyra, Gracilaria and Grateloupia has been studied in China. For Laminaria japonica, the successfully cultivated kelp in China, a model transformation system has been set up based on the application of plant genetic techniques and knowledge of the algal life history. Progress has been made recently in incorporating a vaccine gene into kelp genome. Evidence has been provided showing the expression of gene products as detectable vaccines. In the present paper, the progress of molecular biotechnological studies of marine algae in China, especially researches on elucidating and manipulating nucleic acids of marine algae, are reviewed.  相似文献

Traditional morphological features have formed the basis for distinguishing species of Porphyra. Among these features are number of cell layers, number of chloroplasts per cell, arrangement of reproductive structures on the thallus, and overall morphology. Chromosome number and chromosome morphology have helped corroborate some species identities. A survey of northeast Pacific species of Porphyra using starch gel electrophoresis of 15 soluble proteins has shown that electrophoretic banding patterns provide a reliable diagnostic tool for species identification. Data from starch gel electrophoresis are presented to confirm the identities of species formerly associated with the Porphyra perforata species-complex in British Columbia and northern Washington. Porphyra abbottae, P. fallax, P. kanakaensis, and P. torta are recognized as distinct species, and Porphyra sanjuanensis is synonymized with P. perforata.  相似文献

We investigated the genetic variations of the samples that were tentatively identified as two cultivated Porphyra species (Porphyra yezoensis Ueda and Porphyra tenera Kjellm.) from various natural populations in Japan using molecular analyses of plastid and nuclear DNA. From PCR‐RFLP analyses using nuclear internal transcribed spacer (ITS) rDNA and plastid RUBISCO spacer regions and phylogenetic analyses using plastid rbcL and nuclear ITS‐1 rDNA sequences, our samples from natural populations of P. yezoensis and P. tenera showed remarkably higher genetic variations than found in strains that are currently used for cultivation. In addition, it is inferred that our samples contain four wild Porphyra species, and that three of the four species, containing Porphyra kinositae, are closely related to cultivated Porphyra species. Furthermore, our PCR‐RFLP and molecular phylogenetic analyses using both the nuclear and plastid DNA demonstrated the occurrence of plastid introgression from P. yezoensis to P. tenera and suggested the possibility of plastid introgression from cultivated P. yezoensis to wild P. yezoensis. These results imply the importance of collecting and establishing more strains of cultivated Porphyra species and related wild species from natural populations as genetic resources for further improvement of cultivated Porphyra strains.  相似文献

The genus Porphyra (and its sister genus Pyropia) contains important red algal species that are cultivated and/or harvested for human consumption, sustaining a billion-dollar aquaculture industry. A vast amount of research has been focused on species of this genus, including studies on genetics and genomics among other areas. Twelve novel microsatellite markers were developed here for Porphyra linearis. Markers were characterized using 32 individuals collected from four natural populations of P. linearis with total heterozygosity varying from 0.098 to 0.916. The number of alleles per locus ranged from 2 to 18. All markers showed cross amplification with Porphyra umbilicalis and/or Porphyra dioica. These polymorphic microsatellite markers are useful for investigating population genetic diversity and differentiation in P. linearis and may become useful for other genetic research on the reproductive biology of this important species.  相似文献

Porphyra is one of the world’s most valued maricultured seaweeds and has been cultivated for several hundred years in Asia. The objective of this study was to produce critical information as a guide for the selection of an appropriate Porphyra species from coastal New England for the development of a land-based aquaculture system. Four Northwest Atlantic Porphyra species: P. leucosticta, P. amplissima, P. linearis and P. umbilicalis, were cultivated for 1 and 2 weeks at saturated light intensities (100–150 μmol photons m−2s−1) and six combinations of ammonium (25 and 250 μmoles L−1) and temperature (10, 15 and 20°C). Specific growth rate (SGR) increased with decreasing temperature in P. leucosticta, P. linearis and P. umbilicalis and increased with increasing temperature in P. amplissima. The SGR of all species was greater at the higher ammonium concentration. Porphyra linearis had the highest SGR, increasing in biomass by approximately 16% day−1. Phycoerythrin (PE) content was higher at 10°C and 250 μmoles L−1 in all species except P. amplissima. The PE content, measured as fresh weight (FW), of P. linearis (29 mg g−1 FW−1) and P. umbilicalis (26 mg g−1 FW−1) was significantly higher than the other two species. Tissue nitrogen content of all species measured in dry weight was on average 1.45% higher at 250 μmoles L−1 than at 25 μmoles L−1 ammonium concentration. Porphyra umbilicalis had the highest tissue nitrogen contents (6.76%) at 10°C and 250 μmoles L−1 ammonium. Based on these results, P. linearis and P. umbilicalis should be considered as potential candidates for bioremediation with finfish and shellfish mariculture.  相似文献

Due to their rapid growth and nutrient assimilation,Porphyra spp. are good candidates for bioremediation and polyculture. The production potential of two strains of P. purpurea and P. umbilicalis from north-east USA was evaluated by measuring rates of photosynthesis (as O2evolution) of material grown at 20 °C. Photosynthetic rates of P. umbilicalis were 80%higher than P. purpurea over the temperature range 5–20 °C, at both sub-saturating andsaturating irradiances (37 and 289 μmol photonm-2 s-1). Porphyra umbilicalis was more efficient at low irradiances (higher α) and had a higher Pmax (23.0 vs 15.6 μmolO2 g-1 DW min-1) than P.purpurea, suggesting that P. umbilicalis is a better choice for mass culture, where self-shading maybe severe. We developed a simple, rapid and stable method for extraction of high molecular weight DNA from the marine red alga Porphyra yezoensis Ueda using both guanidium treatment and QIAGEN? kit (Funakoshi, Tokyo, Japan). The method does not require expensive equipment and complex steps. The DNA yield averaged 1.5 μg 100 mg?1 of Porphyra tissue and the A260/A280 and A230/A260 ratios of the DNA were approximately 1.8 and 0.4, respectively. It was of sufficient quality to be used for not only polymerase chain reactions but also other DNA manipulation techniques such as restriction digestion and construction of genomic libraries.  相似文献

We investigated phylogenetic relationships among red algae of the order Bangiales by analysis of sequences of the nuclear gene encoding cytosolic small-subunit ribosomal RNA in Bangia atropurpurea (Roth) C. Ag. and eight samples representing seven species of Porphyra. The ssu-rDNA range from 1818 to 1845 nucleotides in length, with guanosine plus cytosine ratios between 47.0% and 48.6%. A group IC1 intron occurs in the B atropurpurea ssu-rDNAs at the same position as in P. spiralis var. amplifolia Oliveira Filho et Coll and several other eukaryote ssu-rDNAs. The nine sequences form a stable monophyletic group upon phylogenetic analysis. The ssu-rDNA from B. atropurpurea nests stably within the Porphyra group and is closely related to P. amplissima (Kjellm.) Setchell et Hus in Hus, making the genus Porphyra paraphyletic. No correlation is seen between phylogenetic position and number of cell layers in the Porphyra thallus. We discuss possible taxonomic and evolutionary implications of these observations.  相似文献

Foliose species of the Bangiales (Porphyra sensu lato) have a long history of study in the N Atlantic, but there are still regions, especially in the northern parts of the N Atlantic that need more attention. A molecular study using rbcL and cox1 sequences was undertaken to assess the diversity of foliose Bangiales species in Iceland and the Faroe Islands. Herbarium collections from the intertidal and subtidal of Iceland (summer and winter) and the Faroe Islands (all seasons) revealed a total of 13 species (11 common to both areas), which were referred to four of the genera recognized in a recent two-gene global phylogeny. Boreophyllum birdiae, Porphyra dioica, P. linearis, P. purpurea, P. umbilicalis, Pyropialeucosticta’ A, Pyropia njordii Mols-Mortensen, J. Brodie & Neefus, sp. nov., Wildemania amplissima and W. miniata were common to both areas, while Pyropia thulaea and Wildemania abyssicola (Kjellman) A. Mols-Mortensen & J. Brodie, comb. nov. (=Porphyra abyssicola Kjellman) were reported from Iceland but not from the Faroe Islands; Porphyra sp. FO and Pyropia elongata were reported from the Faroe Islands but not from Iceland. Boreophyllum birdiae is reported for the first time for Iceland and Porphyra sp. FO is reported for the first time for the Faroe Islands. Pyropia njordii is described from the Faroe Islands and is also recorded for Iceland, Greenland, New England, USA and Nova Scotia, Canada. A total of 25 foliose Bangiales species are now reported from the N Atlantic and these results demonstrate the importance of investigating as many areas as possible to reach a more complete understanding of species diversity and distribution.  相似文献

Sensory analyses were conducted to determine levels of consumer acceptability of Porphyra yezoensis, P. umbilicalis, and P. amplissima to select appropriate species for aquaculture development in Maine (USA). The subjects included children (n = 67) and adults (n = 84); the children participated in study design by helping to select the 9 point hedonic scale used in the affective sensory tests. Two substrates were used; Porphyra was baked in crackers and also used as a coating for popcorn. No significant differences (p > 0.5) in acceptability of one species over another were observed in either trial, which suggests that native Atlantic species of Porphyra such as P. amplissima and P. umbilicalis have developmental potential in foods for North American consumers. Fatty acids were analyzed in the taste test material and in freshly collected P. umbilicalis; eicosapentaenoic acid [EPA; 20:5 (n-3)] and palmitic acid were the most common fatty acids. Quantitative analysis of EPA determined that freshly collected (January 2005) P. umbilicalis contained 3.2 mg EPA g dry wt−1 (74 mg EPA 100 g fresh wt−1). This concentration is not high enough to make P. umbilicalis a primary source of daily omega-3 fatty acids, but the favorable n-3/n-6 ratio (2-3:1) in these species contributes to their nutritional value.  相似文献

Temperature is one of the major environmental factors that affect the distribution, growth rate, and life cycle of intertidal organisms, including red algae. In an effort to identify the genes involved in the high‐temperature tolerance of Porphyra, we generated 3,979 expression sequence tags (ESTs) from gametophyte thalli of P. seriata Kjellm. under normal growth conditions and high‐temperature conditions. A comparison of the ESTs from two cDNA libraries allowed us to identify the high temperature response (HTR) genes, which are induced or up‐regulated as the result of high‐temperature treatment. Among the HTRs, HTR2 encodes for a small polypeptide consisting of 144 amino acids, which is a noble nuclear protein. Chlamydomonas expressing the Porphyra HTR2 gene shows higher survival and growth rates than the wild‐type strain after high‐temperature treatment. These results suggest that HTR2 may be relevant to the tolerance of high‐temperature stress conditions, and this Porphyra EST data set will provide important genetic information for studies of the molecular basis of high‐temperature tolerance in marine algae, as well as in Porphyra.  相似文献

As the first step in an investigation of the life-history ofPorphyra umbilicalis (L.) Kütz. var. laciniata (Lightf.)J. Ag., spores which have originated as a result of the repeateddivision of the mother-cells have been germinated. When grownon glass their method of germination and growth agrees withthat described by previous investigators, but when germinatedon sterile shell the germ-tubes penetrate the shell and developinto growths identical with Conchocelis rosea Batters. This‘species’, therefore, is a phase in the life-historyof P. umbilicalis var. laciniata and not an autonomous species.The development of the ‘Conchocelis’-phase in cultureis described and also the formation of fertile cell-rows and‘plantlets’. Until the significance of these structuresis understood and a cytological investigation completed, therelationship of the intertidal leafy Porphyra-thallua to thefilamentous, shell-inhabiting, deep-water Conchocelis cannotbe expressed in the terms usually employed to denote the variousphases of the life-histories of algae.  相似文献

The marine red alga Porphyra umbilicalis has high tolerance toward various abiotic stresses. In this study, the contents of floridoside, isofloridoside, and trehalose were measured using gas chromatography mass spectrometry (GC-MS) in response to desiccation and rehydration treatments; these conditions are similar to the tidal cycles that P. umbilicalis experiences in its natural habitats. The GC-MS analysis showed that the concentration of floridoside and isofloridoside did not change in response to desiccation as expected of compatible solutes. Genes involved in the synthesis of (iso)floridoside and trehalose were identified from the recently completed Porphyra genome, including four putative trehalose-6-phosphate synthase (TPS) genes, two putative trehalose-6-phosphate phosphatase (TPP) genes, and one putative trehalose synthase/amylase (TreS) gene. Based on the phylogenetic, conserved domain, and gene expression analyses, it is suggested that the Pum4785 and Pum5014 genes are related to floridoside and isofloridoside synthesis, respectively, and that the Pum4637 gene is probably involved in trehalose synthesis. Our study verifies the occurrences of nanomolar concentrations trehalose in P. umbilicalis for the first time and identifies additional genes possibly encoding trehalose phosphate synthases.  相似文献
