Studies were conducted tosearch for fungal strains with potentialpathogenicity against Diabrotica speciosa(Germar) (Coleoptera: Chrysomelidae).Among sixteen fungal isolates screenedthe most virulent was a Beauveria bassiana(Balsamo) Vuillemin isolate (FHD13) thatcaused 70% mortality of D. speciosathird instar larvae. The LC50 value ofB. bassiana isolate FHD13 was3.48 × 1010 conidia/ml.Different temperatures (4, 17 and 26 °C)and vegetable oils (corn, sunflower and canola)used for storage did not significantly affectviability of conidia. A pathogenicity trialagainst D. speciosa larvae performed withthe corn oil formulation (1 × 108 conidia/mlof oil) caused 65% of mortality.  相似文献

Since 1992, seed production of mangrove crab,Scylla serrata, has been attempted at the Gondol Research Station for Coastal Fisheries, Bali, Indonesia. During the production process, almost all of the larvae have died due to fungal infection. Fungi isolated from the larvae with fungal infection were classified into three species in the order Lagenidiales:Lagenidium callinectes, Haliphthoros milfordensis andHalocrusticida baliensis sp. nov. based on detailed morphological characteristics. In Australia, fungi associated with larvae of the biological control agent Cactoblastis cactorum may contribute to the control of the exotic weed pricklypear (Opuntia inermis). C. cactorum larvae were assessed for their ability to vector pathogenic fungi into O. inermis by the infestation of larvae with fungal suspensions. Six fungal isolates caused disease after being carried into the host on external surfaces of larvae, and propagules of one isolate (UQ5109) initiated disease after being transferred from the cladode epidermis into the host by larvae feeding on the plant. Scanning electron microscopy revealed extensive hyphal growth on the external surfaces of larvae infested with several of the isolates. Fungi isolated from field-grown O. inermis cladodes were tested for pathogenicity to this plant in an in vivo plant assay. In total, 152 isolates were screened, 22 of which infected the host in pathogenicity tests. Only 1 (UQ5115) infected undamaged host tissue, whereas the remainder required the host to be wounded before infection could proceed. The majority of isolates were only weakly pathogenic, even when inoculated via wounds, suggesting that most were either saprophytes or weak parasites. This study demonstrates that it is possible for larvae of C. cactorum to transmit fungal pathogens into O. inermis tissue and it has provided a sound basis for future field work to determine the contribution that fungi make to the control of O. inermis.  相似文献

The eulophid parasitoid, Euplectrus maternus, is a gregarious ectoparasitoid of the larvae of thefruit-piercing moth, Eudocima (Othreis) fullonia. This parasitoid is indigenous to India and was released into Guam in 1998–99 to aid in the biological control of E. fullonia, an important pest of ripeguava, banana, mango, pomegranate, litchi,papaya, tomato, orange and other fruit crops.In the laboratory, biology and rearing of E. maternus on the larvae of E. fulloniawere studied. In no-choice tests, ovipositionby E. maternus was significantly greateron first and second instar larvae than on thirdinstar larvae. However, in free-choice tests,oviposition by E. maternus wassignificantly greater on second instar larvae,followed by first and third instar larvae. Thisparasitoid did not lay eggs on later instars,either in no-choice or free-choice tests.Females laid a significantly greater number ofeggs (>80%) on the dorsal surface of thelarvae than on either lateral or ventralsurfaces (1 to 8%). In general, more eggs werelaid on abdominal segments 1–3 on larvalinstars one to three. The survival of theimmature stages (from egg to adult) of E. maternus on second instar E. fullonialarvae was 61%, which was significantlygreater than the survival rates on first andthird instar larvae (32% and 26%), indicatingthat second instar host larvae are ideal formass rearing of the parasitoid. Mated femaleE. maternus continuously laid eggs on thesecond instar larvae of E. fullonia forup to 30 days, but the greatest number of eggswere laid during the first week after exposure.When E. maternus laid more than two eggson host larva, more female progeny wereproduced, indicating female-biasedreproduction. Adult parasitoids lived longerwhen fed with a honey: water (50% w/v)solution than with pure honey. Similarly, thefecundity of females increased significantlywhen fed with the honey-water solution whencompared to feeding with pure honey. To date,E. maternus has not established in Guam.  相似文献

Three species of entomopathogenicnematodes, a combination of two nematodespecies, an entomopathogenic fungal species,and a combination of a nematode and fungalspecies were evaluated against the white grubsEctinohoplia rufipes and Exomalaorientalis (Coleoptera: Scarabaeidae) in the field. The nematodes were acommercial formulation of Steinernemacarpocapsae (BioSafe) and S. glaseri from Dongrae and from Hanrim, and Heterorhabditis bacteriophora from Hamyang,Republic of Korea. The entomopathogenic funguswas Beauveria brongniartii, produced onSabouraud maltose agar plus 1% yeast (SMAY),rice bran, or compost. The combinationtreatment was S. carpocapsae with H.bacteriophora or B. brongniartii. Fieldapplications were made in August or Septemberagainst third instars at a golf course infestedwith E. rufipes in Gyeongnam Province in1991, and one in Pusan with E. orientalisin 1992 and 1993. In 1991, a significantreduction of 70.2 to 79.4% of E. rufipeslarvae was observed in the nematode, fungal andchemical (fenitrothion) treatments comparedwith a 15.7% reduction in the control. In1992, the E. orientalis larval populationwas reduced between 62.7 and 82.8% in thetreatments compared to 10.7% in the control.In 1993, larval reductions in plots treatedwith nematodes (78.3 to 97%) and B.brongniartii propagated on rice bran (84.5%)were significantly better than in plots treatedwith B. brongniartii propagated on SMAY(63.6%) or compost (59.6%). Combining twonematode species did not enhance the efficacycompared to treatments with one nematodespecies alone, but combining S.carpocapsae with B. brongniartiiproduced on SMAY resulted in a significantincrease in grub mortality over the applicationof the fungus alone produced on SMAY orcompost. The high efficacy of the nematode andmost fungal treatments was attributed to theclose proximity of the white grubs to the soilsurface which allowed for excellentpathogen-host contact and to favorable soiltemperatures, sandy soil, post irrigationapplication and/or rain and a minimal thatchlayer in the turfgrass.  相似文献

Larvae of the cedar web-spinning sawfly, Cephalcia tannourinensis Chevin (Hymenoptera: Pamphiliidae), infected with a white fungus were collected from the Tannourine-Hadath El-Jebbeh cedar forest. Macro- and micro-morphological data based on the examination of colonies, conidiophores, and conidial shape of the fungus suggested a Beauveria species. Sequence analysis of the internal transcribed spacer regions of the isolated fungus showed that it is most closely related to isolates of B. bassiana Clade C. The present study showed that the isolated B. bassiana is a naturally occurring entomopathogenic fungus parasitizing the larvae of C. tannourinensis in Lebanon. Laboratory bioassays showed that B. bassiana caused high mortality of eggs and larvae. The infected eggs turned brownish in color, while larvae of the first instar ceased feeding and showed immobility and rigidity within 5 days and before sporulating conidial mat appeared on their cuticle. Second and third larval instars took longer time to show fungal sporulation: mortalities ranged between 85 and 100% within 7 days when treated with different conidial concentrations. The efficacy of control of C. tannourinensis using B. bassiana was higher or equal to the reference insect growth regulator, diflubenzuron, suggesting the possibility of its success as a biological control agent.  相似文献

The predaceous larvae of Chrysoperla rufilabris (Burmeister) exhibited some success-motivated searching, particularly when feeding on Heliothis virescens (F.) eggs, but handling time did not decrease with experience. Handling time for H. virescens larvae was more than twice that for eggs. H. virescens larvae were preferred to cotton aphids (Aphis gossypii (F.)) while aphids were preferred to H. virescens eggs. Laboratory studies were done to measure predation by adult damsel bugs,Nabis roseipennis Reuter [Heteroptera: Nabidae], on 3rd instar larvae of Mexican bean beetle (MBB),Epilachna varivestis Mulsant [Coleoptera: Coccinellidae], and to measure longevity and body weight of the nabids. In the 1st experiment, field-collected nabids were isolated for 24h in 9 cm Petri dishes with lima bean foliage (Phaseolus lunatus L.) and were assigned to one of 3 prey treatments: either 4 3rd-instar MBB larvae, 4 3rd-instar larvae of boll weevil (BW),Anthonomus grandis Boheman [Coleoptera: Curculionidae], or 2 larvae of each species. No MBB larvae were attacked in either the MBB treatment or 2-species treatment. In contrast, BW larvae were attacked in both BW and 2-species treatments. Significantly more BW larvae were attacked in the BW treatment than in the 2-species treatment, and both were greater than the number of MBB larvae attacked. Nabids that did not attack prey lost weight during the 24 h, whereas those that attacked prey gained weight. In the 2nd experiment, nabids that had attacked prey were isolated with lima foliage, and nabids that had not attacked prey were kept with MBB and lima foliage until an attack or death. In no instances were MBB attacked. Longevity and the pattern of weight loss did not differ between nabids that did or did not attack prey. We discuss possible reasons for the failure ofN. roseipennis to attack MBB larvae, as well as the implications for using nabids to influence pest populations in the field.   相似文献

Previous studies which have tested the feeding preferences of shredders for fungal species and the food quality of fungi used detritus uniformly colonized by a fungus, which is not the case for decaying leaves in streams. It is not known whether shredders in different development stages exhibit variations in feeding preference and larval performance. This study examined the feeding preferences and the growth of the third and the fifth instars of Pycnopsyche gentilis larvae using fungal-colonized patches and whole leaves, respectively, having different fungal species compositions (Alatospora acuminata, Anguillospora filiformis, Articulospora tetracladia, Tetrachaetum elegans, and all species combined). The aquatic hyphomycetes used were co-dominant on leaves in the stream inhabited by the caddisfly. During 14 d of feeding, the larvae of both instars did not show significant differences in feeding preferences for the patches growing on oak leaves, although the third instar larvae were slightly more selective than the fifth instar larvae. When fed with maple leaves for 18 d, larval growth rates, gross growth efficiencies, and survivorship were not significantly different among the fungal treatments. However, the larval growth of both instars fed with fungal-colonized leaves was always significantly greater than the growth of larvae fed with diets of uncolonized leaves. The third instar larvae grew faster than the fifth instar larvae, but the growth efficiencies of the two instars were similar. These results suggest that P. gentilis larvae exhibit less selectivity in their feeding than other caddisfly shredders that have been examined and that the dominant fungi colonizing leaves in their habitat are similar in palatability and food quality for this shredder. The in vivo development of the entomogenous hyphomycetePaecilomyces farinosus inSpodoptera exigua (beet armyworm) larvae was examined using light and electron microscopic techniques. Blastospores injected into larval hemocoels (500 blastospores/larva) were immediately ingested by phagocytic hemocytes, and no fungal cells were detected in the hemolymph until 36 h post-injection. As indicated by immunocytochemical methods, the in vivo-produced blastosopres, in contrast to in vitro blastospores, lacked a galacto-mannan surface layer required for opsonization by aS. exigua humoral lectin. Therefore, these in vivo cells were not recognized by phagocytic granulocytes and were freely-circulating in the hemolymph. Hyphae differentiating from the blastospores were recognized by the hemocytes and induced formation of multicellular hemocytic nodules. By 72 h post-injection, mycelia were observed emerging from the nodules and by 96 h, larvae had become mummified due to extensive proliferation of the fungus throughout host tissues. Neither phagocytosis of the initially injected in vitro-produced blastospores nor nodule formation around hyphal cells later in the infection process was effective in stopping fungal growth. The in vivo development ofP. farinosus was similar to that of another hyphomycete,Beauveria bassiana except that in the latter case, extensive nodule formation was inhibited by the production of fungal metabolites.  相似文献

The stem borer, Busseola fusca (Fuller), is an important pest of maize Zea mays L. and sorghum Sorghum bicolor (L.) in eastern and southern Africa. To control this pest, biological control methods including the use of entomopathogenic fungi are being considered. The pathogenicity of one isolate of Metarhizium anisopliae (Metsch.) Sorok. and one isolate of Beauveria bassiana Bals. (Vuill.) were first tested on different developmental stages of B. fusca including eggs, neonate, 2nd and 3rd-instar larvae. Both fungal isolates were pathogenic to all the stages tested. However, differences in mortality were observed among larvae that hatched from treated egg masses. Experiments were conducted thereafter to test whether B. fusca males could serve as a vector for fungal conidia to contaminate B. fusca females and subsequently eggs and larvae. Results demonstrated that B. fusca males successfully transferred inoculum to females during copulation, which in turn transmitted it to the eggs they laid on maize plants, resulting in the decrease of leaf damages.  相似文献

Studies on interactions between a larval parasitoid, Pteromalus cerealellae (Boucek) and one of its hosts, Callosobruchus maculatus (F.) were carried out in the laboratory. The number of host larvae parasitized by P. cerealellae increased with host larvae at low densities and tended to a plateau at a density of 25 larvae per female parasitoid. Each parasitoid was able to parasitize more hosts and produced more offspring at 20 and 25 °C than at 30 °C. The number of non-infested seeds mixed with seeds infested with the last instar of C. maculatus did not preclude P. cerealellae from identifying infested seeds and attacking larvae inside them. When infested seeds were tightly packed, several host larvae escaped parasitism. P. cerealellae may be a useful biological control agent in newly harvested cowpea with low C. maculatus infestation, and lowering the temperature of the storage system may enhance the effectiveness of this parasitoid.  相似文献

The foraging behaviors of larvae of the ladybird, Coccinella septempunctata L., towards both the ant-tended aphid, Aphis craccivora Koch, and the non-ant-tended aphid, Acyrthosiphon pisum Harris, were investigated in the field and in laboratory experiments. Although there were no differences in the development and growth of the ladybird larvae that preyed on either Ac. pisum or Ap. craccivora, the foraging efficiency of the ladybird larvae that preyed on Ap. craccivora was higher than that of the ladybird larvae that preyed on Ac. pisum in the absence of ants. This result was explained by the fact that the number of Ac. pisum that escaped by dropping off the plant was conspicuously larger than the number of Ap. craccivora that escaped in this fashion and derived from the non-ant-attendance associated with Ac. pisum. In the laboratory experiments, fewer ladybird larvae climbed onto a plant with Ap. craccivora in the presence of ants than onto a plant with Ac. pisum in the absence of ants. The ladybird larvae did not switch from foraging for Ap. craccivora to foraging for Ac. pisum, even after suffering attacks by ants on a plant with Ap. craccivora, and it would appear that ladybird larvae are unable to remember where they have previously been attacked by ants. These results could explain why the ladybird larvae in the field more frequently visited Vicia angustifolia plants with Ap. craccivora than those with Ac. pisum and made more visits when ants were absent than when they were present.  相似文献

Reproducing Varroa jacobsoni obtained from brood cells of Apis mellifera L. with 13–16 day old bees (pupae) and Varroa mites kept on adult bees for at least 8 days were simultaneously tested for their choice in three host types. Comparisons were made of attractiveness of Varroa jacobsoni to nurse bees, pollen foragers as to larvae from nearly capped brood cells. Host choices were observed in Petri dishes and in an Y-shaped olfactometer. Varroa jacobsoni obtained from capped brood cells showed a stronger preference for nurse bees in Petri dish simultaneous choice tests with pollen foragers or larvae than did mites which were previously kept on adult bees. In olfactometer simultaneous choice tests, the two mite test groups showed no clear difference in preferences for bees of different ages. The preference of Varroa jacobsoni for bees of different ages is therefore not only influenced by host factors but also by intrinsic factors in female mites that depend on the mite's reproductive stage.  相似文献

The dynamics of the fungal symbionts in the gallery system and the mycangia of the ambrosia beetle,Xylosandrus mutilatus, were studied in relation to its life history using both isolation experiments and scanning electron microscopy (SEM). In the galleries,Ambrosiella sp. was predominant during the larval stages but its relative dominance gradually decreased during the development of the larvae. In contrast, yeasts (mainlyCandida sp.) andPaecilomyces sp. dominated continuously in the galleries after eclosion.Ambrosiella sp. was consistently stored in the mycangia in all adult stages, except in the teneral and overwintering adults when the other fungi were dominant. No fungal spores occurred in the mycangia of the adult beetles reared under aseptic conditions from the pupal stage, while onlyAmbrosiella sp. was stored in those reared from the teneral-adult stage. These results suggest that: (i) Xmutilatus is associated with at least three fungal species, among whichAmbrosiella sp. is the most essential food resource for development of the broods; (ii) immediately after eclosion, new female adults may take at least four associated fungal species, with no or incomplete selection, into their mycangia from the walls of the cradles; and (iii) conditions may well be produced in the mycangia of both matured and dispersing beetles whereby only the spores ofAmbrosiella sp. can proliferate.  相似文献

A Sebacinales species was recovered from a clone library made from a pooled rhizosphere sample of Nicotiana attenuata plants from 14 native populations. Axenic cultures of the related species, Piriformospora indica and Sebacina vermifera, were used to examine their effects on plant performance. Inoculation of N. attenuata seeds with either fungus species stimulated seed germination and increased growth and stalk elongation. S. vermifera inoculated plants flowered earlier, produced more flowers and matured more seed capsules than did non-inoculated plants. Jasmonate treatment during rosette-stage growth, which slows growth and elicits herbivore resistance traits, erased differences in vegetative, but not reproductive performance resulting from S. vermifera inoculation. Total nitrogen and phosphorous contents did not differ between inoculated and control plants, suggesting that the performance benefits of fungal inoculation did not result from improvements in nutritional status. Since the expression of trypsin proteinase inhibitors (TPI), defensive proteins which confer resistance to attack from Manduca sexta larvae, incur significant growth and fitness costs for the plant, we examined the effect of S. vermifera inoculation on herbivore resistance and TPI activity. After 10 days of feeding on S. vermifera-inoculated plants, larval mass was 46% higher and TPI activity was 48% lower than that on non-inoculated plants. These results suggest that Sebacina spp. may interfere with defense signaling and allow plants to increase growth rates at the expense of herbivore resistance mediated by TPIs.  相似文献

The susceptibility of the tick Boophilus microplus to Beauveria bassiana was evaluated by inoculating eggs, larvae and engorged females of the tick with five fungal isolates at concentrations of 106, 107 and 108 conidia/ml. Tick eggs (0.25 g) were immersed in 1 ml of a suspension of the different conidial concentrations for 1 min. Similar exposure was performed by immersion of 2000 larvae and homogeneous groups of nine engorged females in 2 and 20 ml of conidial suspension, respectively. Treated eggs, larvae and adults were placed in an incubator at 27 ± 1 °C and relative humidity above 80% for evaluation of the fungal action. All fungal isolates applied at all conidial concentrations reduced the hatching rate of larvae from treated eggs by 1.36–65.58% and increased the mortality rate of inoculated larvae by 0.8–70.49%. In the bioassay with engorged females, oviposition period was reduced by 9.69–47.80%, egg mass weight by 4.71–53.87%, estimated reproduction by 8.3–60.62%, egg production index by 5.03–54.20%, percent larval hatching by 0.27–13.96%, and the mortality rate of treated females was increased by 96.60–100%. The reduction of the estimated reproduction obtained for the treated groups ranged from 8.37 to 64.52%. The sporulation of the pathogen on dead females ranged from 3.70 to 88.88% depending on the isolate and concentration used. Isolates AM 09, CB 7 and JAB 07 were the most effective and effectiveness increased with increasing concentrations of conidia in the suspensions.  相似文献

Dose– and time–mortality relationships of baculoviruses in pest insects are important for the determination of effective spraying regimes. A series of experiments with Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) and Spodoptera exigua MNPV (SeMNPV) against synchronized populations of S. exigua larvae in greenhouse chrysanthemum was conducted. Dose– and time–mortality relationships of different virus concentrations and S. exigua target stages were determined and the area foliage consumption was measured. Crop injury was greatly reduced when S. exigua were controlled as second or third instar larvae, whereas virus applications against fourth instar larvae could not prevent considerable crop injury, even at high concentrations. SeMNPV was approximately 10 times as infectious as AcMNPV when applied on greenhouse chrysanthemum. The relative virulence of AcMNPV and SeMNPV corresponded reasonably well with previously published laboratory bioassay data. SeMNPV killed second and fourth instar S. exigua larvae approximately 12 h faster than did AcMNPV in chrysanthemum, but no difference in speed of action was found for third instar larvae. The relative speed of action of AcMNPV and SeMNPV determined in chrysanthemum and in laboratory bioassays did not correspond for third instar S. exigua larvae; laboratory bioassay data can therefore not simply be extrapolated to the crop level.  相似文献

The western corn rootworm, Diabrotica virgifera virgifera LeConte(Coleoptera: Chrysomelidae), is the mostdestructive pest of maize (Zea mays L.)in North America, and began to successfullyinvade Central Europe in the early 1990's. Thispaper reports a three-year field surveyconducted in Hungary, Yugoslavia, and Croatia,which are currently the focal points ofinvasion, with the aim to determine theoccurrence of indigenous natural enemies ofD. v. virgifera in Europe. A total of9,900 eggs, 550 larvae, 70 pupae and 33,000adults were examined for the occurrence ofparasitoids, nematodes, and fungal pathogens. It can be concluded from the survey resultsthat effective indigenous natural enemies arenot attacking any of the life stages of D.v. virgifera in Europe. The exception is theoccurrence of the fungi Beauveriabassiana (Bals.) Vuill. (Mitosporic fungi;formerly Deuteromyces) and Metarhiziumanisopliae (Metsch.) Sorok (Mitosporic fungi)attacking adults of D. v. virgifera at anextremely low level (< 1%). However no otherentomopathogenic fungal pathogens,entomopathogenic nematodes, or parasitoids werefound on eggs, larvae, pupae or adults. Whileseveral natural enemies in North and CentralAmerica are known to attack D. v.virgifera, it is apparent that indigenousnatural enemies in Europe have not adapted tothe high population density of the alieninvasive species D. v. virgifera. Classical biological control may provide anopportunity to reconstruct the natural enemycomplex of an invading alien pest, and itsapplication to manage D. v. virgiferapopulations in Europe should be considered.  相似文献

The insect-parasitic rhabditoid nematodes,Steinernema feltiae andHeterorhabditis bacteriophora, released a compound/s/ toxic to larvae of the greater wax moth,Galleria mellonella, that caused paralysis and death of the insect. Larvicidal substances appeared in wax moth larvae during parasitism and after inoculation with the primary form of the bacterial associates of the nematodes. The nematodeS. feltiae and its associate,Xenorhabdus nematophilus, excreted much less toxic activity within larval body thanH. bacteriophora. The secondary form ofXenohabdus did not produce toxin in parasitized larvae, butX. luminescens, the bacterium associated withH. bacteriophora, released detectable titer of toxin activity in broth cultures. Both nematode toxins were sensitive to heat and produced a specific type of proteolytic activity. Preliminary identification of the compounds responsible for larval toxicity revealed similarities to immune inhibitors produced by some bacterial pathogens of insects.   相似文献
