Le pré-analytique en hémostase

La phase pré-analytique est celle de tous les dangers, en particulier lorsque le biologiste n’a pas la ma?trise du recueil de l’échantillon et de son acheminement au laboratoire. En hémostase thrombose, elle est essentielle. Les paramètres pré-analytiques sont d’importance inégale selon les différents examens et leur retentissement peut varier d’un réactif à l’autre et selon les patients. De fait, il existe un risque de rendre un résultat erroné, même avec une technique irréprochable, ainsi qu’un problème de responsabilité technico-légale. La littérature sur les conditions pré-analytiques est pauvre et de nouvelles études sont nécessaires, même si elles sont difficiles à mener et peuvent para?tre de peu d’intérêt. En réalité, leur utilité est aujourd’hui indéniable, en raison notamment du regroupement des laboratoires.     

Fluctuations morphologiques et hétérochronies chez Toxaster (échinides,Crétacé inférieur)

Morphological fluctuations and heterochronies of Toxaster (echinoid, Lower Cretaceous). Several steps of the diversification of the spatangoid Toxaster are described with an analysis of six species distributed over the north-west Tethyan margin, and ranging from the Lower Valanginian to the Barremian. Morphological variation was investigated with the analysis of two parameters (angles), which allowed recognition of three modules on the test. Comparison of ontogenetic trajectories of two reference species (T. granosus kiliani and T. retusus) suggests a peramorphic heterochronic pattern. However, no peramorphocline can be attested when comparisons are extended to adults of the six species. Evolution among these species is partially driven by allometric changes in one module, indicating an important influence of size. On the temporal and spatial scale of the study, the evolution of Toxaster appears globally quite conservative. To cite this article: É. François et al., C. R. Palevol 2 (2003).

Faut-il éradiquer Phragmanthera capitata,parasite des hévéas en Afrique ?

The effect of Phragmanthera capitata (Sprengel) Balle, an epiphytic Loranthaceae, on latex production of three clones of rubber tree, Hevea brasiliensis (Müll. Arg.), was studied in the industrial plantation of Mitzic, in Gabon. The 22-year-old trees, presenting the largest trunk circumference, were the most parasitized and at the same time the best producers of latex. At first, this would suggest that mistletoe parasitism could have a favourable effect on latex yield. However, an accurate statistical analysis showed that, in fact, parasitism had no significant effect on latex yield. These data, discussed in relation to the biology of flowering parasitic plants, suggest that there is no urgency to eradicate Phragmanthera capitata for the moment.     

Analyse du statut spermatique, érectile et hormonal chez des patients hémodialysés

Introduction

Our work was to establish the hormonal, semen and erectile profile among haemodialysed patients and to seek the impact of hormonal disturbances on erectile function and semen parameters.

Patients and methods

We conducted a cross-sectional study in haemodialysed patients in whom a semen, in parallel with hormonal analyses including FSH (follicle stimulating hormone) and testosterone. Erectile function was assessed by the study of the international index of erectile function in its French version (IIEF). Analyses of sex hormones were done by radio immunoassay and semen analyses according to WHO guidelines.

Results

Two patients had a semen analysis and a normal hormone balance; the IIEF was normal in a one patient. Azoospermic patients (16%) showed a hypergonadotrophic eugonadism. Forty percent of the patients had hypospermia without any correlation between hypospermia and hormonal values. One third of patients showed severe oligospermia associated with high levels of FSH in 77% of cases. Sperm motility and morphology were altered in 96% and 50% of the cases respectively. Hormonal analyses showed an elevated FSH (> 8.5 mUI/ml) in 40% of the cases and testosterone was decreased (< 3.2 ng/ml) in 25% of the cases.

Discussion

Semen volume was significantly decreased in patients over 30 years. Erectile function was disturbed in 73% of the patients with a mean IIEF score of 15. Several authors have shown a correlation between gonadal dysfunction and high levels of gonadotropins in men with chronic renal insufficiency, with or without testicular atrophy. In our series, testosterone was normal despite the absence of androgen. Are haemodialysis sessions effective in preserving the endocrine function?

Conclusion

Patients in chronic haemodialysis for a period exceeding one year had a hypergonadotrophic eugonadism and a severe erectile dysfunction. The state of the genital tract was relatively preserved. The duration of haemodialysis did not significantly affect sperm and erectile function. Patients older than 30 years showed a significant decrease in semen volume, which could be a marker to determine the impairment of erectile and reproductive functions.

Dysplasia of epidermal ridges, with autosomal dominant transmission

Summary A large family is described where 15 individuals on 4 generations are affected with a dysplasia of the digital, palmar and plantar epidermal ridges. In this family, the anomaly is transmitted according to the autosomal dominant mode of inheritance with complete penetrance and variable expressivity.

Étude de la variabilité de la clairance hépatique de la mébrofénine-99mTc en scintigraphie hépatobiliaire

AimThe purpose of this study was to investigate some of the parameters likely to influence mebrofenin-^99mTc hepatic clearance calculation and inter-and intra-observers reproducibility.Materials and methodsHepatic clearance (%/min m²) of 30 scintigraphies was calculated from the values of hepatic, cardiac, and total activities, according to the method recommended in the literature. We studied: 1) impact of injection–acquisition delay variations; 2) acquisition type: anterior face only (FA) or geometric mean (GM); 3) clearances calculated according to four different body surface area (BSA) formulas; 4) intra-and inter-observers reproducibility for three observers (two evaluations for each observer).Results1) Clearance differences between different studied intervals were statistically significant, more important if the studied interval was far from reference interval (150–350 secondes) and even more when the interval studied was too early (110–310 secondes). 2) There was a statistically significant difference between clearance calculated using either FA or GM datasets (0.85 %/min m²). 3) There were small but statistically significant differences for four of the clearance comparisons using different BSA formulas. 4) Despite differences in size of cardiac and hepatic regions of interest (ROI), intra-observer reproducibility of hepatic clearance was excellent for each observer. Inter-observers reproducibility was also excellent (r = 0.982).ConclusionHepatic clearance of mebrofenin-^99mTc appears to be a highly reproducible method provided that acquisition and clearance calculation are standardized. It provides additionnal functional information to morphological and biological data usually performed before major hepatectomy. Thereby, the definition of a standardized protocol would enable realization of multicentric studies.     

Caractérisation d'une hémopexine sérique de truite par utilisation d'une protéine recombinante

Rainbow trout hemopexin was produced by Sf9 cells through the Baculovirus system; the protein is released into the culture medium. It has been purified through two successive chromatographic steps: ion exchange (MonoQ) and chelation (Zn⁺ affinity). Treatment of the protein by N-glycosidase F reduced its relative mass by 3 000, demonstrating N-glycosylation. Spectrophotometric analysis in the Soret band of protein + hemin mixture revealed characteristics of a hemoprotein (oxidized: λ = 413 nm, dithionite-reduced: λ = 422 nm); the titration curve suggested a 1:1 molar ratio. An anti tHx antiserum was produced; it detected an intense band in the trout serum, but also in the serum of two perciforms: gilt-head sea bream and sea bass; it did not cross-react with the serum of the phylogenetically distant eel.
