Aquaculture is one of the fastest growing industries in the world. There is a need for enhanced disease resistance, feed efficiency and growth performance of cultured fish species. The cost of production are likely to be reduced if growth performance and feed efficiency are increased in commercial aquaculture. Also if the survivility of the fishes increase, then their overall production cost would be remarkably reduced. Dietary supplementation of different feed additives e.g. immunostimulants, probiotics and prebiotics usually in small quantities for the purpose of fortifying it with certain nutrients have been found to be beneficial for improving immune status, feed efficiency and growth performance of crustaceans and finfishes. The present article was constructed to highlight the effect of dietary prebiotics on different body growth parameters of fish. However, the application of prebiotics and supplementary enzymes in fish feed is now gradually gaining importance in commercial aquaculture practices, the article stresses on the effect of prebiotics on live body weight gain, dressing percentage, weight of vital organs and muscles and mean villus lengths in digestive tract of fish along with their application as growth promoters in commercial aquafeed (Gatesoupe in Aqua Feeds Formul Beyond 2(3):3–5, 2005; Ganguly et al. in Israeli J Aquac Bamidgeh 62(3):130–138, 2010a, Fish Chimes 30(7):64, 2010b).  相似文献

Probiotic administration can be a nutritional strategy to improve the immune response and growth performance of fish. The current study aimed to evaluate the effects of a probiotic blend (Bacillus sp., Pediococcus sp., Enterococcus sp., Lactobacillus sp.) as a dietary supplement on growth performance, feed utilization, innate immune and oxidative stress responses and intestinal morphology in juvenile Nile tilapia (Oreochromis niloticus). The probiotic was incorporated into a basal diet at three concentrations: 0 g/kg (A0: control), 3 g/kg (A1: 1.0×106 colony forming unit (CFU)/g) and 6 g/kg (A2: 2.3×106 CFU/g diet). After 8 weeks of probiotic feeding, weight and specific growth rate where significantly higher in fish-fed A1 diet than in fish-fed A0. Alternative complement in plasma was significantly enhanced in fish-fed A2 when compared with A0. The hepatic antioxidant indicators were not affected by probiotic supplementation. Villi height and goblet cell counts increased significantly in the intestine of fish-fed A1 and A2 diets compared with A0. The dietary probiotic supplementation was maintained until 20 weeks of feeding. Then the selected immune parameters, digestive enzymes and apparent digestibility of diets were studied. No effect of probiotic feeding was observed after that longer period supplementation. The dietary supplementation of mixed species probiotic may constitute a valuable nutritional approach towards a sustainable tilapia aquaculture. The improvement of the immune responses and intestinal morphology play an important role in increasing growth performance, nutrient absorption and disease resistance in fish, important outcomes in such a competitive and developing aquaculture sector.  相似文献

All metazoans possess innate immune defence system whereas parameters of the adaptive immune system make their first appearance in the gnathostomata, the jawed vertebrates. Fish are therefore the first animal phyla to possess both an innate and adaptive immune system making them very interesting as regards developmental studies of the immune system. The massive increase in aquaculture in recent decades has also put greater emphasis on studies of the fish immune system and defence against diseases commonly associated with intensive fish rearing. Some of the main components of the innate and adaptive immune system of fish are described. The innate parameters are at the forefront of immune defence in fish and are a crucial factor in disease resistance. The adaptive response of fish is commonly delayed but is essential for lasting immunity and a key factor in successful vaccination. Some of the inherent and external factors that can manipulate the immune system of fish are discussed, the main fish diseases are listed and the pathogenicity and host defence discussed. The main prophylactic measures are covered, including vaccination, probiotics and immunostimulation. A key element in the immunological control of fish diseases is the great variation in disease susceptibility and immune defence of different fish species, a reflection of the extended time the present day teleosts have been separated in evolution. Future research will probably make use of molecular and proteomic tools both to study important elements in immune defence and prophylactic measures and to assist with breeding programmes for disease resistance.  相似文献

Bacterium Flavobacterium columnare is the causative agent of columnaris disease in many wild and farmed fish species. Immunostimulants are used with success in aquaculture against many pathogens, but the ability to improve innate resistance to columnaris disease has not been studied. Fingerling rainbow trout were treated with two immunostimulants, yeast β-glucan and β-hydroxy-β-methylbutyrate (HMB). Selected innate immune function parameters, the production of reactive oxygen species (ROS) by whole blood and by isolated head kidney leukocytes, plasma lysozyme activity and complement bacteriolytic activity, were determined to assess the immune status of fish. The fish were then bath challenged with virulent F. columnare bacteria, and the mortality of fish was recorded. Given orally both stimulants raised the levels of immune function parameters, but did not improve survival in challenge at any concentration of the stimulants used. Intra peritoneal injection of β-glucan increased parameter values several fold, but no beneficial effect of injected glucan on survival was noted. As a control, antibiotic medication administered prior to and during the challenge infection prevented the mortality. Innate immune mechanisms, even when induced to high levels with immunostimulants, as evidenced here, were not able to increase resistance against F. columnare. This may be connected to the external character of the infection. The results from the treatments with β-glucan and HMB suggest that there is little prospect of preventing columnaris disease by means of immunostimulants in early life stage of rainbow trout. However, the efficacy of other immune stimulants remains open.  相似文献

The increased resistance of fish pathogens to conventional treatments has lead researchers to investigate the antibacterial properties of natural resources, such as essential oils (EOs) of plants, in an effort to find products that are less harmful to the environment. The objective of this review is to provide an overview of the studies, in vivo and in vitro, that addressed the use of EOs and their major compounds as antimicrobial agents in fish, to identify the best EOs and compounds to investigate considering feasibility of application and suggest possible future studies. To date, studies suggest that the use of EOs in the prevention and/or treatment of infectious diseases in fish may be a promising strategy to reduce the use of conventional antibiotics in aquaculture, since several EOs effectively reduce or avoid the effects of bacterial infections in fish. The use of EOs through nanotechnology delivery systems, especially in dietary supplementation experiments, is promising. This form of application of the EOs allows a potentiation and targeting of the desired effect of the EOs and also allows the protection of EOs active constituents against enzymatic hydrolysis, deserving further study.  相似文献

Infectious hematopoietic necrosis virus (IHNV) is one of the most significant viral pathogens of salmonids and is a leading cause of death among cultured juvenile fish. Although several vaccine strategies have been developed, some of which are highly protective, the delivery systems are still too costly for general use by the aquaculture industry. More cost effective methods could come from the identification of genes associated with IHNV resistance for use in selective breeding. Further, identification of susceptibility genes may lead to an improved understanding of viral pathogenesis and may therefore aid in the development of preventive and therapeutic measures. Genes of the major histocompatibility complex (MHC), involved in the primary recognition of foreign pathogens in the acquired immune response, are associated with resistance to a variety of diseases in vertebrate organisms. We conducted a preliminary analysis of MHC disease association in which an aquaculture strain of Atlantic salmon was challenged with IHNV at three different doses and individual fish were genotyped at three MHC loci using denaturing gradient gel electrophoresis (PCR-DGGE), followed by sequencing of all differentiated alleles. Nine to fourteen alleles per exon-locus were resolved, and alleles potentially associated with resistance or susceptibility were identified. One allele (Sasa-B-04) from a potentially non-classical class I locus was highly associated with resistance to infectious hematopoietic necrosis (p < 0.01). This information can be used to design crosses of specific haplotypes for family analysis of disease associations.  相似文献

The effects of dietary whole cell yeast (Saccharomyces cerevisiae), n-3 HUFA-enriched yeast and treated yeast cells with beta-mercapto-ethanol (2ME) on immunity, growth performance and disease resistance to Yersinia ruckeri were investigated in Oncorhynchus mykiss. During 30 days, juvenile rainbow trout were fed diets supplemented with different forms of yeast at 5 × 10(7) CFU g(-1) or a control diet. After the feeding trial, remaining fish of each treatment were challenged by pathogenic Yersinia ruckeri and kept under observation for 14 days to record clinical signs and daily mortality rate. Yeast supplementation in all treatment groups significantly promoted the growth performance compared to control group. A significantly increase was also observed in immune responses in juvenile fish fed 2ME-treated yeast diet. More ever, the lowest fish mortality was obtained in this treatment group. The present results show that a diet supplemented with 2ME-treated yeast stimulates the immune system and growth of juvenile rainbow trout thus enhancing their resistance against Y. ruckeri.  相似文献

Stress is known to impair immune function and disease resistance in fish. In the present study, repeated handling was employed as a chronic stressor in order to verify whether its attributed immunosuppressive effects could be minimized by dietary arginine supplementation. Therefore, Senegalese sole (Solea senegalensis) were air exposed daily for 3 min during 14 days (handling) or left undisturbed (control). In addition, both control and handled specimens were fed 3 diets with graded levels of arginine (Arg 4.4, Arg 5.7 and Arg 6.9 g 16 g(-1) N). Following the 14 days stress challenge and feeding on those diets, fish were infected with Photobacterium damselae subsp. piscicida (strain PC566.1; LD(50) 5 × 10(3) cfu mL(-1)) and fed the same experimental diets. Respiratory burst activity and nitric oxide production of head-kidney leucocytes increased parallel to dietary arginine supplementation. HIF-1, HAMP-1, MIP1-alpha and gLYS expression values and some humoral parameters augmented in control specimens fed the Arg 5.7 and Arg 6.9 diets. Interestingly, repeated acute stress increased both disease resistance and some innate immune mechanisms in handled fish. The role of dietary arginine and repeated handling on Senegalese sole innate immunity and disease resistance are discussed.  相似文献

Abstract The intensive rearing of various fish species in aquaculture has revealed intimate relationships between fish and bacteria that eventually may affect establishment of a ``normal' mucosal microflora or result in disease epizootics. Interactions between bacteria and mucosal surfaces play important roles both at the egg and larval stages of marine fish. Bacterial adhesion and colonization of the egg surface occur within hours after fertilization. The diverse flora which eventually develops on the egg appears to reflect the bacterial composition and load of the ambient water, but species-specific adhesion at the egg surface may also play a role in development of the egg epiflora. Proteolytic enzymes produced by members of the adherent epiflora may cause serious damage to the developing egg and may also affect further adhesion of the epiflora. Ingestion of bacteria at the yolk sac stage results in establishment of a primary intestinal microflora which seems to persist beyond first feeding. Establishment of a gut microflora is likely to undergo several stages, resulting in an ``adult' microflora weeks to months after first feeding. Ingested bacteria may serve as an exogenous supply of nutrients or essential factors at an early life stage. Early exposure to high bacterial densities is probably important for immune tolerance, and thus for the establishment of a protective intestinal microflora. Successful rearing of early life stages of several marine fish species depends on knowledge of the complex interactions among the cultured organisms and the bacterial communities which develop at the mucosal surfaces and in the ambient water and rearing systems. The routine use of antibiotics during rearing of fish larvae is not advisable, since it may increase the risk of promoting antibiotic resistance and adversely affect the indigenous microflora of the larvae. The use of probiotics has proven advantageous in domestic animal production, and the search for effective probiotics may have a great potential in aquaculture of marine organisms. Bacteria with antagonistic effects against fish pathogens have been successfully administered to several fish species, resulting in decreased mortality or increased growth rate. The use of immunostimulants has received increased attention due to the discovery of Toll-like receptors (TLR) or/and pattern recognition receptors (PRR). These receptors have been found to bind molecules from a range of pathogens including self-molecules. When cell damage has occurred many of the released molecular structures act as so-called "danger" signals possessing pathogen-associated molecular patterns (PAMP). These danger signals often consist of repeating molecular moieties yielding high molecular weight compounds. Examples are beta-glucans and CpG containing DNA, but some danger signals possess low molecular weight structures. It has been found that the PRR bind unit structures of PAMP, and that PAMP-binding involves several other humoral and cell membrane proteins, exemplified by the more or less simultaneous LPS recognition displayed by MD-2, CD-14 and TLR4 on the cell membrane. Also, the binding of beta-glucans has been shown to include several different cell membrane receptors. Several immunostimulants are commercially exploited in aquaculture as feed additives. This applies to beta-glucans, alginates and nucleotides. Despite their use as feed additives no targeted approach has been conducted to include PAMP as adjuvants in fish vaccines. Interestingly, most of the PAMP studied activate antigen-presenting cells together with na?ve T cells into dendritic cells and Th1 or Th2 cells [1]. In turn, this may activate Th1 and Th2 immune responses with production of Th1 or Th2 signature molecules such as IFN-gamma and IL-4, respectively [2-4]. This review will mainly focus on binding characteristics of beta-glucans, their effects on T helper cell differentiation, effects on functional levels, gene expression profiles and application of the commonly used ss-glucan in the aquaculture sector. In addition, ss-glucans show promises in shrimp aquaculture by inducing disease resistance, this review will also highlight the use and the effects of beta-glucans in experimental models.  相似文献

A 4-week feeding trial was conducted to investigate the effects of dietary β-glucan, mannan oligosaccharide (MOS) and their combinations on growth performance, immunity and disease resistance of sea cucumber Apostichopus japonicus. Sea cucumbers (1215 individuals with initial weight of 3.8?±?0.2?g) were fed nine practical diets according to a 3?×?3 factorial design: the basal diet as the control supplemented with three levels of β-glucan (0, 0.075, 0.15%?w/w), crossed with 0, 0.1% (w/w) or 0.2% (w/w) MOS. Immune indices including total coelomocytes count (TCC), phagocytosis, superoxide anion production, superoxide dismutase (SOD) activity and total nitric oxide synthase activity (T-NOS) were measured at days 7, 11, 15, 18, 22, 25 and 29. At the end of the feeding trial, all the sea cucumbers left were weighted to monitor growth, and then were challenged by Vibrio splendidus. Poly-β-hydroxybutyrate (PHB) is a bacterial energy and carbon storage compound which exhibits a controlling effect on the gastrointestinal microbiota. Its beneficial activities for aquaculture have already been shown in terms of increased disease resistance and growth performance in a number of studies. However, the action of PHB on the intestinal microbial community in the treated animals has not yet been studied in depth. In this research, the effects of PHB on the microbiota composition in the intestinal tract of juvenile sea bass were examined. It was found that fish cohabiting in the same tank were on average 87% similar regarding the intestinal microbiota. When subjected to the same treatment and environmental conditions but reared in different tanks, the compositions of the enteric communities diverged. The provision of PHB overruled this tank effect by sustaining a microbial core community in the gut that represented 60% of the total bacterial diversity at the highest PHB level of 10%. The microbial community compositions converged between replicate tanks upon supplementation of PHB and were characterized by high dynamics and increased evenness. The results are discussed in the framework of hypotheses that try to relate the intestinal microbial community composition to the health status of the host organisms.  相似文献

The identification of specific bacterial species influenced by mannan oligosaccharide (MOS) supplementation may assist in the formulation of new and improved diets that promote intestinal health and improve bird performance, offering suitable alternatives to antimicrobials in feed for sustainable poultry production. This study has been conducted to evaluate the use of a MOS compound derived from the yeast cell wall of Saccharomyces cerevisiae on turkey performance, bacterial community structure and their phylogenetic associations. A 42-day turkey trial was carried out on birds fed control and MOS-supplemented diets. Bird performance data (weight gains, feed consumption and feed efficiency ratios) were collected, and caecal contents were extracted from randomly caught poults on days 28, 35 and 42 posthatch. Bird performance data showed no improvements as a result of dietary supplementation. Automated ribosomal intergenic spacer analysis (ARISA) revealed the bacterial community structure to be significantly altered on days 28 and 35 posthatch but not day 42 as a result of dietary supplementation. This technique was coupled with 16S rRNA gene sequence analysis to elucidate phylogenetic identities of bacteria. The dominant bacteria of the caecum on all days in both treatment groups were members of phylum Firmicutes, followed by the Bacteroidetes and Proteobacteria phyla, respectively. Statistical analysis of the 16S rRNA gene libraries showed that the composition of the MOS clone library differed significantly to the control on day 35 posthatch. It can be concluded that MOS alters the bacterial community structure in the turkey caecum.  相似文献

The study was conducted to evaluate effects of dietary supplementation with Ligustrum lucidum (LL, 10 g/kg), Schisandra chinensis (SC, 10 g/kg), LL (10 g/kg) + mannan oligosaccharides (MOS, 50 mg/kg), or SC (10 g/kg) + MOS (50 mg/kg) on growth performance and parameters of antioxidative and immunological status of broilers. The results showed that feeding LL, SC, LL + MOS, or SC + MOS had no significant effect on growth performance of broilers relative to the control. However, compared to the control, LL, SC, LL + MOS, or SC + MOS significantly decreased malondialdehyde concentration in serum, thigh, and heart of broilers. In addition, glutathione reductase activity of heart and sera of the birds were significantly elevated by supplementation LL, SC, LK + MOS, or SC + MOS. Furthermore, LL, SC, LL + MOS, or SC + MOS significantly improved antibody titres against Newcastle disease virus and lymphocyte proliferation of broilers (p < 0.05). Whereas, no cooperating effect between LL (or SC) and MOS on antioxidant status and immunity of broilers were found.  相似文献

Myxozoan parasites are responsible for important economic losses among fisheries and aquaculture industries, and hence the high interest in studying the immune response of fish against them. The most important data available concerning the immune response of fish against myxosporeans are reviewed, with emphasis on the different innate and adaptive immune mechanisms, their relationship with natural and acquired resistance and the strategies to control and prevent myxosporoses. Cellular effectors (lymphocytes, granulocytes, phagocytes, non-specific cytotoxic cells, rodlet cells) and humoral factors (lysozyme, peroxidades, antiproteases, complement, specific antibodies) have been examined for several myxosporoses, and some immune relevant genes have been studied. Genetic considerations permeate the intricateinteractions of marine aquacultural populationswith their wild counterparts. We illustratethese complexities in a matrix where sevencategories of primary aquaculture activity(conservation, supplementation, mitigation,intentional introduction, put-and-take,commercial captive rearing, experimental) arelinked to important genetic considerationsrelated to resident and cultured taxa. We focuson the effects of cultured populations on theirwild native conspecifics. Using publishedexamples, we examine both the frequency ofoccurrence and the biological implications ofsituations in which released cultured fish havehad direct genetic effects throughintrogressive hybridization, as well asindirect genetic effects such as inducedthrough disease transfer, induced over-harvest,and displacement. These examples are contrastedwith those instances where aquacultural activitieshave coexisted in harmony with underlyingconcerns related to issues of conservationgenetics.
