The expression of estrogen receptors, ESR1 (ERα) and ESR2 (ERβ), and androgen receptors (AR) in the thyroid gland has been reported in few vertebrate species other than a few mammals. This study reports the presence of sex steroid hormone receptors and thyroid receptors (ERα, ERβ, AR, TRα, and TRβ) in the thyroid gland of the American alligator at several life stages. It provides a semiquantification and distribution of ERα in the thyroid follicle cells using an immunohistochemical approach as well as reports quantitative differences in mRNA expression of ERα, ERβ, TRα, TRβ, and AR in the same tissue using quantitative real time‐PCR (Q‐PCR) with primers designed specifically for alligators. The thyroid tissue of the American alligator expresses ERα, ERβ, and AR at all of the life stages examined here although no statistically significant differences were observed between male and female in thyroid mRNA expression for any of the genes analyzed. No sexual dimorphism was observed in ERα immunostaining. No statistical analysis across life stages were performed due to confounding factor of season. J. In zebrafish, like many other teleost species, the development and differentiation of many major organs continue unabated into the yolk-sac larval stage before culminating in a free-swimming larva capable of exogenous feeding. We investigated the role of thyroid hormone (TH) in this important embryonic to larval transitory phase. Thyroid hormone receptor (TR) alpha and beta mRNAs are expressed during the early stages of zebrafish embryonic development. Beginning from the midblastula stage, the level of TR beta mRNAs increases dramatically and is maintained until the end of the transitory phase. Excessive exogenous thyroxine (T4; 30 nM) is toxic and causes severe developmental defects. Cotreatment of embryos with amiodarone, an antagonist of TR, and goitrogen methimazole (MMI) lead to severe retardation in the maturation of the gastrointestinal system, swim bladder, and the lower jaw cartilages and the resorption of the yolk sac. The developmental arrest is lethal, and treated larvae do not survive beyond 7 day postfertilization (dpf), but can be completely rescued by the presence of 10 nM T4. We propose that the embryonic to larval transitory phase in many teleost species is characterized by its dependency on the timely synthesis of TH and the concomitant autoinductive increase in TR beta mRNA levels.  相似文献

Thyroid hormones and their receptors (TRs) have critical functions in development. Here we show that a chicken TR beta cDNA clone encodes a receptor with a novel, short N-terminal domain. In vitro-expressed TR beta protein bound thyroid hormone with similar affinity as the chicken TR alpha. Comparison of expression of TR alpha and TR beta mRNAs throughout chicken development until 3 weeks post-hatching revealed ubiquitous expression of TR alpha mRNAs (in 14 different tissues) with some variations in levels, from early embryonic stages. In contast, expression of TR beta mRNA was restricted, occurring notably in brain, eye, lung, yolk sac and kidney, and was subject to striking developmental control, especially in brain where levels increased 30-fold upon hatching. Levels also sharply increased in late embryonic lung, but were relatively high earlier in embryonic eye and yolk sac. RNase protection analyses detected no obvious mRNAs for alpha and beta TRs with variant C-termini as demonstrated previously for the rat TR alpha gene. The data suggest a general role for TR alpha and specific developmental functions for TR beta, and that thyroid-dependent development involves temporal and tissue-specific expression of the TR beta gene.  相似文献

We have isolated and sequenced a cDNA clone encompassing the entire protein coding region of the Xenopus laevis estrogen receptor (xER). The Xenopus ER, the first steroid hormone receptor to be sequenced from a cold-blooded organism, exhibits two regions of striking amino acid homology with the human and avian ERs. In the putative DNA binding region, the amino acid sequence of the xER differs from those of the human and avian ERs at only one of 83 amino acids. The putative hormone binding region contains 44 and 46 amino acid blocks in which the sequence is identical in the Xenopus and human ERs. Blot hybridizations of Xenopus liver RNA suggest that the xER is encoded by four mRNAs with lengths of approximately 9, 6.5, 2.8, and 2.5 kilobases. In contrast, hybridization of human RNA to a human ER cDNA clone reveals only a single major ER RNA, approximately 6.7 kilobases in length.  相似文献

We have recently reported that prolactin (PRL) inhibits both morphogenesis and cell death in thyroid hormone (T3)-induced amphibian metamorphosis (Tata et al., 1991), and that the autoinduction of T3 receptor (TR alpha and beta) mRNA is among the most rapid responses of premetamorphic Xenopus tadpoles to T3 (Kawahara et al., 1991). We now demonstrate that PRL prevents the rapid T3-induced upregulation of TR alpha and beta mRNAs in stages 50-54 Xenopus tadpoles and in organ cultures of tadpole tails. This effect is followed by the inhibition of the de novo activation of 63-kDa keratin gene by T3. We present an experimentally testable model whereby PRL exerts its juvenilizing action by preventing the amplification of TR by its autoinduction by T3.  相似文献

The accumulation of mRNA encoding for hepatic and intestinal T3-receptor (TR) and body and liver masses were measured in fed and 3-week fasted juvenile and swim up stage rainbow trout embryos. Plasma and total body thyroid hormone (TH) levels were measured for juvenile and swim up stages, respectively. Fasted juveniles exhibited a lower hepatosomatic index (HSI), liver mass and plasma T4 and T3 concentrations than fed animals, but there were no changes in body mass or the accumulation of mRNA encoding for either of the TR(alpha) or TR(beta) isoforms in liver or intestine. TR(beta) mRNA accumulation was greater than TR(alpha) mRNA accumulation in both tissues. Fasted embryos had lower whole body TH levels and body, liver and intestinal tract masses, in addition to a lower intestinosomatic index. However, there was no change in HSI. Fasting did not affect whole body or hepatic TR(alpha) and TR(beta) mRNA accumulation, although intestinal tract TR(alpha) and TR(beta) mRNA accumulation was lower in the fasted embryos. The HSI and body mass changes in fasted juvenile and embryo stages, respectively, indicated that both developmental stages were impacted by fasting. Both stages also showed evidence of decreased TH production. The lower TR gene expression in the intestinal tract of fasted embryos may suggest a role for THs in the transitional stage of intestinal development during this period of development.  相似文献
