Temporal and spatial variability in the kinetic parameters of uptake of nitrate (NO3), ammonium (NH4+), urea, and glycine was measured during dinoflagellate blooms in Changjiang River estuary and East China Sea coast, 2005. Karenia mikimotoi was the dominant species in the early stage of the blooms and was succeeded by Prorocentrum donghaiense. The uptake of nitrogen (N) was determined using 15N tracer techniques. The results of comparison kinetic parameters with ambient nutrients confirmed that different N forms were preferentially taken up during different stages of the bloom. NO3 (Vmax 0.044 h−1; Ks 60.8 μM-N) was an important N source before it was depleted. NH4+ (Vmax 0.049 h−1; Ks 2.15 μM-N) was generally the preferred N. Between the 2 organic N sources, urea was more preferred when K. mikimotoi dominated the bloom (Vmax 0.020 h−1; Ks 1.35 μM-N) and glycine, considered as a dominant amino acid, was more preferred when P. donghaiense dominated the bloom (Vmax 0.025 h−1; Ks 1.76 μM-N). The change of N uptake preference by the bloom-forming algae was also related to the variation in ambient N concentrations.  相似文献

The nitrogen (N) uptake kinetic parameters for Microcystis field assemblages collected from the San Francisco Bay Delta (Delta) in 2012 and non-toxic and toxic laboratory culture strains of M. aeruginosa were assessed. The 15N tracer technique was used to investigate uptake of ammonium (NH4+), nitrate (NO3), urea and glutamic acid over short-term incubations (0.5–1 h), and to study inhibition of NO3, NH4+ and urea uptake by NH4+, NO3 and NH4+, respectively. This study demonstrates that Delta Microcystis can utilize different forms of inorganic and organic N, with the greatest capacity for NH4+ uptake and the least for glutamic acid uptake, although N uptake did not always follow the classic Michaelis–Menten hyperbolic relationship at substrate concentrations up to 67 μmol N L−1. Current ambient N concentrations in the Delta may be at sub-saturating levels for N uptake, indicating that if N loading (especially NH4+) were to increase, Delta Microcystis assemblages have the potential for increased N uptake rates. Delta Microcystis had the highest specific affinity, α, for NH4+ and the lowest for NO3. In culture, N uptake by non-toxic and toxic M. aeruginosa strains was much higher than from the field, but followed similar N utilization trends to those in the field. Neither strain showed severe inhibition of NO3 uptake by NH4+ or inhibition of NH4+ uptake on NO3, but both strains showed some inhibition of urea uptake by NH4+.  相似文献

This study represents the most comprehensive assessment of kinetic parameters for Karenia brevis to date as it encompasses natural populations sampled during three different bloom years in addition to cultured strains under controlled conditions. Nitrogen (N) uptake kinetics for ammonium (NH4+), nitrate (NO3), urea, an amino acid mixture, individual amino acids (glutamate and alanine), and humic substrates were examined for the toxic red tide dinoflagellate, K. brevis, during short term incubations (0.5–1 h) using 15N tracer techniques. Experiments were conducted using natural populations collected during extensive blooms along the West Florida Shelf in October 2001, 2002, and 2007, and in cultured strains (CCFWC 251 and CCFWC 267) obtained from the Florida Fish and Wildlife Institute culture collection. Kinetic parameters for the maximum uptake velocity (Vmax), half-saturation concentration (Ks), and the affinity constant (α) were determined. The affinity constant is considered a more accurate indicator of substrate affinity at low concentrations. K. brevis took up all organic substrates tested, including N derived from humic substances. Uptake rates of the amino acid mixture and some NO3 incubations did not saturate even at the highest substrate additions (50–200 μmol N L−1). Based upon the calculated α values, the greatest substrate preference was for NH4+ followed by NO3  urea, humic compounds and amino acids. The ability of K. brevis to utilize a variety of inorganic and organic substrates likely helps it flourish under a wide range of nutrient conditions from bloom initiation in oligotrophic waters offshore to bloom maintenance near shore where ambient nutrient concentrations may be orders of magnitude greater.  相似文献

The toxic HAB dinoflagellate Karenia brevis (Davis) G. Hansen & Ø. Moestrup (formerly Gymnodinium breve) exhibits a migratory pattern atypical of dinoflagellates: cells concentrate in a narrow (∼0–5 cm) band at the water surface during daylight hours due to phototactic and negative geotactic responses, then disperse downward at night via non-tactic, random swimming. The hypothesis that this daylight surface aggregation behavior significantly influences bacterial and algal productivity and nutrient cycling within blooms was tested during a large, high biomass (chlorophyll a >19 μg L−1) K. brevis bloom in October of 2001 by examining the effects of this surface layer aggregation on inorganic and organic nutrient concentrations, cellular nitrogen uptake, primary and bacterial productivity and the stable isotopic signature (δ15N, δ13C) of particulate material. During daylight hours, concentrations of K. brevis and chlorophyll a in the 0–5 cm surface layer were enhanced by 131% (±241%) and 32.1% (±86.1%) respectively compared with an integrated water sample collection over a 0–1 m depth. Inorganic (NH4, NO3+2, PO4, SiO4) and organic (DOP, DON) nutrient concentrations were also elevated within the surface layer as was both bacterial and primary productivity. Uptake of nitrogen (NH4+, NO3, urea, dissolved primary amines, glutamine and alanine) compounds by K. brevis was greatest in the surface layer for all compounds tested, with the greatest enhancement evident in urea uptake rates, from 0.08 × 10−5 ng N K. brevis cell−1 h−1 to 3.1 × 10−5 ng N K. brevis cell−1 h−1. These data suggests that this surface aggregation layer is not only an area of concentrated cells within K. brevis blooms, but also an area of increased biological activity and nutrient cycling, especially of nitrogen. Additionally, the classic dinoflagellate migration paradigm of a downward migration for access to elevated NO3 concentrations during the dark period may not apply to certain dinoflagellates such as K. brevis in oligotrophic nearshore areas with no significant nitricline. For these dinoflagellates, concentration within a narrow surface layer in blooms during daylight hours may enhance nutrient supply through biological cycling and photochemical nutrient regeneration.  相似文献

The nitrogen uptake capabilities of the toxigenic diatom Pseudo-nitzschia australis (Frenguelli), freshly isolated from Monterey Bay California, were examined in unialgal laboratory cultures at saturating photosynthetic photon flux densities (100 μmol photons m−2 s−1) and 15 °C. The kinetics of nitrogen (nitrate, ammonium, urea and glutamine) uptake as a function of substrate concentration were estimated from short (20.5 min) incubations using the 15N-tracer technique. Based on the estimated maximum specific uptake rates and measures of N affinity (the initial slope of the uptake versus nutrient concentration curve), nitrate is the preferred nitrogen substrate, followed by glutamine and ammonium, which are equivalent. Rates of urea uptake by P. australis did not saturate at concentrations as high as 36 μg-at N L−1, and urea uptake as a function of concentration could not be described by Michaelis–Menten kinetics over the concentration gradient tested. Although there is a clear preference for nitrate at equivalent concentrations (compared to ammonium, urea, and glutamine), these laboratory results demonstrate the capability of this pennate diatom to utilize both inorganic and organic forms of nitrogen, supporting field observations that P. australis blooms during both upwelling and non-upwelling conditions off the west coast of North America. Substantial differences in the nitrogenous nutrition of P. australis can be expected in these environments, and anthropogenic inputs of N substrates such as ammonium and urea can support its growth, and may contribute significantly to both harmful diatom blooms and the maintenance of seed populations at non-bloom abundances, particularly during periods of reduced or absent upwelling.  相似文献

The effects of NH4+ or NO3 on growth, resource allocation and nitrogen (N) uptake kinetics of two common helophytes Phragmites australis (Cav.) Trin. ex Steudel and Glyceria maxima (Hartm.) Holmb. were studied in semi steady-state hydroponic cultures. At a steady-state nitrogen availability of 34 μM the growth rate of Phragmites was not affected by the N form (mean RGR = 35.4 mg g−1 d−1), whereas the growth rate of Glyceria was 16% higher in NH4+-N cultures than in NO3-N cultures (mean = 66.7 and 57.4 mg g−1 d−1 of NH4+ and NO3 treated plants, respectively). Phragmites and Glyceria had higher S/R ratio in NH4+ cultures than in NO3 cultures, 123.5 and 129.7%, respectively.Species differed in the nitrogen utilisation. In Glyceria, the relative tissue N content was higher than in Phragmites and was increased in NH4+ treated plants by 16%. The tissue NH4+ concentration (mean = 1.6 μmol g fresh wt−1) was not affected by N treatment, whereas NO3 contents were higher in NO3 (mean = 1.5 μmol g fresh wt−1) than in NH4+ (mean = 0.4 μmol g fresh wt−1) treated plants. In Phragmites, NH4+ (mean = 1.6 μmol g fresh wt−1) and NO3 (mean = 0.2 μmol g fresh wt−1) contents were not affected by the N regime. Species did not differ in NH4+ (mean = 56.5 μmol g−1 root dry wt h−1) and NO3 (mean = 34.5 μmol g−1 root dry wt h−1) maximum uptake rates (Vmax), and Vmax for NH4+ uptake was not affected by N treatment. The uptake rate of NO3 was low in NH4+ treated plants, and an induction phase for NO3 was observed in NH4+ treated Phragmites but not in Glyceria. Phragmites had low Km (mean = 4.5 μM) and high affinity (10.3 l g−1 root dry wt h−1) for both ions compared to Glyceria (Km = 6.3 μM, affinity = 8.0 l g−1 root dry wt h−1). The results showed different plasticity of Phragmites and Glyceria toward N source. The positive response to NH4+-N source may participates in the observed success of Glyceria at NH4+ rich sites, although other factors have to be considered. Higher plasticity of Phragmites toward low nutrient availability may favour this species at oligotrophic sites.  相似文献

Environmental factors that shape dynamics of benthic toxic blooms are largely unknown. In particular, for the toxic dinoflagellate Ostreopsis cf. ovata, the importance of the availability of nutrients and the contribution of the inorganic and organic pools to growth need to be quantified in marine coastal environments. The present study aimed at characterizing N-uptake of dissolved inorganic and organic sources by O. cf. ovata cells, using the 15N-labelling technique. Experiments were conducted taking into account potential interactions between nutrient uptake systems as well as variations with the diel cycle. Uptake abilities of O. cf. ovata were parameterized for ammonium (NH4+), nitrate (NO3) and N-urea, from the estimation of kinetic and inhibition parameters. In the range of 0 to 10 μmol N L−1, kinetic curves showed a clear preference pattern following the ranking NH4+ > NO3 > N-urea, where the preferential uptake of NH4+ relative to NO3 was accentuated by an inhibitory effect of NH4+ concentration on NO3 uptake capabilities. Conversely, under high nutrient concentrations, the preference for NH4+ relative to NO3 was largely reduced, probably because of the existence of a low-affinity high capacity inducible NO3 uptake system. Ability to take up nutrients in darkness could not be defined as a competitive advantage for O. cf. ovata. Species competitiveness can also be defined from nutrient uptake kinetic parameters. A strong affinity for NH4+ was observed for O. cf. ovata cells that may partly explain the success of this toxic species during the summer season in the Bay of Villefranche-sur-mer (France).  相似文献

The large diatom Coscinodiscus wailesii is one of the problematic species which indirectly cause bleaching damage to “Nori” (Porphyra thalli) cultivation through competitive utilization of nutrients during its bloom. In the present study, we experimentally investigated the nitrate (N) and phosphate (P) uptake kinetics of C. wailesii, Harima-Nada strain. Maximum uptake rates (ρmax), obtained by short-term experiments, were 58.3 and 95.5 pmol cell−1 h−1 for nitrate and 41.9 and 59.1 pmol cell−1 h−1 for phosphate at 9 and 20 °C, respectively. The half saturation constants for uptake (Ks) were 2.91 and 5.08 μM N and 5.62 and 6.67 μM P at 9 and 20 °C, respectively. The ρmax values of C. wailesii, much higher than those of other marine phytoplankton species, suggest that C. wailesii is able to take up large amounts of nutrients from the water column. On the other hand, Vmax/Ks (Vmax; Vmax = ρmax/Q0, Q0; minimum cell quota) values of C. wailesii, which is a better measure to evaluate the competitive ability for nutrient uptake, were low in dominant diatom species. This parameter indicates that C. wailesii is disadvantaged compared to other diatom species in competing for nutrients, and the decreasing nutrient concentrations from winter to spring is an important factor limiting C. wailesii blooming in early spring.  相似文献

The kinetic data obtained from the action of a cathepsin D-like enzyme from Biomphalaria glabrata hepatopancreas (digestive gland) on MOCAc-Gly-Lys-Pro-Ile-Leu-Phe-Phe-Arg-Leu-Lys(DNp)-D–Arg-NH2, was studied as a data prototype, generated by means of a fluorogenic substrate. An initial fluorescence, due to incomplete energy transfer, of about 8% of the values attained after complete substrate hydrolysis; a non-linear standard curve even at μM concentrations and an exponential decay of the steady state fluorescence of reaction product of the order of 10 4 × s 1 were the main analytical problems encountered. The standard curves for fluorescence of the substrate reaction product after 48 h of hydrolysis, and the reference compound MOCAc-Pro-Leu-Gly-NH2, were fitted by polynomial approximation and the point derivates used as calibration factors. Time dependence of the calibration factor for the reaction product was − 2.96 × 10 4 a.u μM 1 × s 1 that is, in the same order of observed enzymic reaction rates. A mathematical treatment was devised for obtaining rates corrected for errors derived from the three analytical problems indicated. The method is of general application in continuous fluorometric assays, irrespective of the particular enzyme used, but of special value for substrates that present significant initial fluorescence. The reaction rates were 11% higher; as calculated by means of the calibration factor [substrate] ÷ (final  initial fluorescence intensities), which is the prevalent procedure in the literature; leading to underestimation of Km and overestimation of Vmax.  相似文献

The responses of soil-atmosphere carbon (C) exchange fluxes to growing atmospheric nitrogen (N) deposition are controversial, leading to large uncertainty in the estimated C sink of global forest ecosystems experiencing substantial N inputs. However, it is challenging to quantify critical load of N input for the alteration of the soil C fluxes, and what factors controlled the changes in soil CO2 and CH4 fluxes under N enrichment. Nine levels of urea addition experiment (0, 10, 20, 40, 60, 80, 100, 120, 140 kg N ha−1 yr−1) were conducted in the needle-broadleaved mixed forest in Changbai Mountain, Northeast China. Soil CO2 and CH4 fluxes were monitored weekly using the static chamber and gas chromatograph technique. Environmental variables (soil temperature and moisture in the 0–10 cm depth) and dissolved N (NH4+-N, NO3-N, total dissolved N (TDN), and dissolved organic N (DON)) in the organic layer and the 0–10 cm mineral soil layer were simultaneously measured. High rates of N addition (≥60 kg N ha−1 yr−1) significantly increased soil NO3-N contents in the organic layer and the mineral layer by 120%-180% and 56.4%-84.6%, respectively. However, N application did not lead to a significant accumulation of soil NH4+-N contents in the two soil layers except for a few treatments. N addition at a low rate of 10 kg N ha−1 yr−1 significantly stimulated, whereas high rate of N addition (140 kg N ha−1 yr−1) significantly inhibited soil CO2 emission and CH4 uptake. Significant negative relationships were observed between changes in soil CO2 emission and CH4 uptake and changes in soil NO3-N and moisture contents under N enrichment. These results suggest that soil nitrification and NO3-N accumulation could be important regulators of soil CO2 emission and CH4 uptake in the temperate needle-broadleaved mixed forest. The nonlinear responses to exogenous N inputs and the critical level of N in terms of soil C fluxes should be considered in the ecological process models and ecosystem management.  相似文献

The South Nation River Watershed, in eastern Ontario, Canada, is an agricultural watershed impacted by excess nutrient loading primarily from agricultural activities. A constructed wetland for the treatment of agricultural wastewater from a 150-cow dairy operation in this watershed was monitored in its eighth operating season to evaluate the proportion of total nitrogen (TN) (approximated by total Kjeldahl nitrogen (TKN) due to low NO3) and total phosphorus (TP) removal that could be attributed to storage in Typha latifolia L. and Typha angustifolia L., which dominate this system. Nutrient loading rates were high, with 16.2 kg ha−1 d−1 N and 3.4 kg ha−1 d−1 P entering the wetland and loading the first wetland cell. Plant uptake accounted for 0.7% of TKN removal when the vegetated free water surface cells were considered together. However, separately, in the second wetland cell with lower N and P loading rates, plants accounted for 9% of TKN, 21% of NH4+ and 5% of TP removal. Plant uptake was significant to overall removal given wetland age and nutrient loading. Nutrient storage during the growing season at this constructed wetland helped reduce the nutrient load entering the watershed, already stressed by intensive local agriculture.  相似文献

Domoic acid (DA) poisoning in the southern part of the California Current System has been associated typically with blooms of Pseudo-nitzschia australis. The environmental variables that promote growth and DA production in the Mexican part of this system have not been identified. The present study investigated the effect of temperature and two nutrient ratios on the growth characteristics and DA content of two (BTS-1, BTS-2) P. australis strains isolated from the Pacific coast of northern Baja California peninsula, México. Of the different temperatures assayed (10, 12, 14, 15, 18 and 20 °C), the maximum cell abundance was detected at 12 °C for BTS-2 and 14 °C for BTS-1. The highest maximum specific growth rate (1.69 day−1) was measured at 15 °C for BTS-2. With the exception of cells maintained at 15 °C, growth characteristics were similar in P. australis cultured in a high Si:NO3 (2.5) or low Si:NO3 (0.5) ratio at each temperature. Dissolved (dDA) and cellular (cDA) DA content measured at the stationary phase of growth was similar in cells cultivated at the different temperatures. No difference in cDA (between 0.11 and 1.87 pg DA cell−1) was observed in cells cultivated at the two nutrient ratios. To evaluate if P. australis accumulates DA (cDA + dDA) at different stages of the culture and not only during the stationary phase of growth, the BTS-1 strain was cultivated at 14 °C and the content of this toxin was measured during culture development. The cultures were maintained at high (HL; 200 μmol quanta m−2 s−1) and low light (LL; 30 μmol quanta m−2 s−1) and in the two nutrient ratios to evaluate the effect of these variables on DA content. The photosynthetic performance and pigment concentration were measured as indicators of the physiological condition of the cells. cDA was detected in all culture conditions and during the different stages of growth. The highest DA content was measured during the lag phase of growth and it was present mainly in the medium (dDA = 70.83 pg DA cell−1). Cells cultivated at HL produced more DA than LL cultured cells. P. australis cultured in HL presented lower photosynthetic rates than LL cells and had similar concentrations of photoprotective pigments and the highest maximum photosynthetic rates were detected during the lag phase of growth in all culture conditions. The results demonstrate that P. australis from northern Baja California peninsula presents a narrow temperature range for optimal growth under batch culture conditions. P. australis produce DA at different stages of growth, and DA content was related to the light intensity at which the cells were cultivated.  相似文献

Chara hispida forms dense beds (0.78–0.95 kg DW m−2) in Colgada Lake. The ability of Chara meadows to act as a nitrogen source or sink was evaluated by the following methods: (1) investigating Chara growth, (2) nitrogen incorporation and decomposition laboratory experiments and (3) relating experimental results to field conditions. Sediment oospores were germinated in large aquaria and observed growth rates were 0.001 m day−1 (shoot length) and 0.0002 g day−1 (dry weight). Nitrogen uptake rates were determined by addition of K15NO3 during two different periods of Chara growth and the rates were 1.21 and 3.86 μM g DW−1 h−1 when charophytes were 166 days old (not sexually mature) and 323 days old (sexually mature), respectively. After the uptake experiments, the same charophytes were allowed to decompose within two types of litter bags (3 mm-pore litter bags and entire, non-porous plastic litter bags). Decomposition rates of Ch. hispida were 0.016 and 0.009 day−1 in perforated and non-perforated bags, respectively, and fit a negative exponential model. The nitrogen release rate, calculated as the disappearance of N content from Chara tissues, was 0.012 day−1 and there were no statistically significant differences between the values from the two different bag types. The dissolved organic nitrogen concentrations in aquarium and non-perforated litter bags waters increased linearly with time due to the leaching of soluble compounds from Chara. The rate of N loss from Chara tissues, total nitrogen and dissolved organic nitrogen release rates and the decrease in initial dry weight rate were all lower than the daily rate of Chara N uptake. By extrapolating laboratory data to field situations, we determined that approximately 38% of the N taken up by charophytes in Colgada Lake during the growth period is retained. Given the high charophyte biomass in the lake, its ability to incorporate nitrogen, its low decomposition rate and its ability to over-winter, we conclude that Chara beds could be acting as nitrogen sinks in this ecosystem.  相似文献

Aquatic macroinvertebrates have been among the principal biological communities used for freshwater monitoring and assessment for several decades, but macroinvertebrate biomonitoring has not incorporated nutrient measures into assessment strategies. Two nutrient biotic indices were developed for benthic macroinvertebrate communities, one for total phosphorus (NBI-P), and one for nitrate (NBI-N). Weighted averaging was used to assess the distributions of 164 macroinvertebrate taxa across TP and NO3 gradients and to establish nutrient optima and subsequent nutrient tolerance values. Both the NBI-P and NBI-N were correlated with increasing mean TP and NO3 values (r = 0.68 and r = 0.57, respectively, p < 0.0001). A three-tiered scale of eutrophication for TP and NO3 (oligotrophic: ≤0.0175 mg/l TP, ≤0.24 mg/l NO3, mesotrophic: >0.0175 to ≤0.065 mg/l TP, >0.24 to ≤0.98 mg/l NO3, eutrophic: >0.065 mg/l TP, >0.98 mg/l NO3) was also established through cluster analysis of invertebrate communities using Bray–Curtis (quantitative) similarity. Significant differences (p < 0.0001) were detected between median NBI-P and NBI-N scores among the three trophic states. Therefore, the nutrient biotic indices (NBIs) appear to accurately reflect changes in stream trophic state. Multimetric water quality assessments were also used to identify thresholds of impairment among the three trophic states. Hodges-Lehman estimation indicated that the greatest change in assessment results occurred between the mesotrophic and eutrophic states. The eutrophic state also represented the highest percentage of overall impairment. Therefore, the suggested threshold for nutrient impairment is the boundary between mesotrophic and eutrophic (0.065 mg/l TP and 0.98 mg/l NO3). The corresponding NBI-P score (6.1) and NBI-N score (6.0) for this threshold incorporate predictive capabilities into the NBIs. The NBI and index score thresholds of impairment will provide monitoring programs with a robust measure of stream nutrient status and serve as a useful tool in enforcing regional nutrient criteria.  相似文献

The effect of NH4+/NO3 availability on nitrate reductase (NR) activity in Phragmites australis and Glyceria maxima was studied in sand and water cultures with the goal to characterise the relationship between NR activity and NO3 availability in the rhizosphere and to describe the extent to which NH4+ suppresses the utilization of NO3 in wetland plants.The NR activity data showed that both wetland helophytes are able to utilize NO3. This finding was further supported by sufficient growth observed under the strict NO3 nutrition. The distribution of NR activity within plant tissues differed between species. Phragmites was proved to be preferential leaf NO3 reducer with high NR activity in leaves (NRmax > 6.5 μmol NO2 g dry wt−1 h−1) under all N treatments, and therefore Phragmites seems to be good indicator of NO3 availability in flooded sediment. In the case of Glyceria the contribution of roots to plant NO3 reduction was higher, especially in sand culture. Glyceria also tended to accumulate NO3 in non-reduced form, showing generally lower leaf NR activity levels. Thus, the NR activity does not necessarily correspond with plant ability to take up NO3 and grow under NO3-N source. Moreover, the species differed significantly in the content of compounds interfering with NR activity estimation. Glyceria, but not Phragmites, contained cyanogenic glycosides releasing cyanide, the potent NR inhibitor. It clearly shows that the use of NR activity as a marker of NO3 utilization in individual plant species is impossible without the precise method optimisation.  相似文献

The positional distributions in potential of lateral root budding and oxygen uptake rate were examined using the segments of madder and horseradish hairy roots with a length of 5.0×10−3 m obtained at different mean distances from the root tips of l=7.5×10−3–47.5×10−3 m. The average rate of lateral root budding and oxygen uptake rate of the roots with smaller l values were higher and both the rates gradually decreased with increase in l value. Positive relations were observed between the rates of lateral root budding and oxygen uptake of both the hairy roots. The relation indicated that the potential of lateral root budding was suppressed at the oxygen uptake rates of 0.15×10−5 and 0.32×10−5 mol O2/(h m) for madder and horseradish hairy roots, respectively.  相似文献

A monitoring program with a weekly sampling frequency over a 15-month period indicates that urea concentrations above a certain threshold level may trigger the blooms of Alexandrium catenella in Thau lagoon. However, urea concentrations were also sometimes related to ammonium and dissolved organic nitrogen concentrations, indicating that the role of urea may not be a direct one. An original approach is used to assess the relative contribution of several nitrogen sources (nitrate, nitrite, ammonium, urea) to growth of A. catenella by comparing nitrogen uptake rates to nitrogen-based growth rates estimated from dilution experiments during four blooms over a 4-year period (2001–2004) in Thau lagoon. Nitrate and nitrite contributed 0.1–14% and 0.1–5% respectively of growth requirements. Ammonium and urea were the main N sources fueling growth of A. catenella (30–100% and 2–59%, respectively). Indirect estimates indicated that an unidentified N source could also contribute significantly to growth at specific times. Concerning ammonium and urea uptake kinetics, half-saturation constants varied between 0.2 and 20 μgat N L−1 for ammonium and between 0.1 and 44 μgat N L−1 over the 4-year period, indicating that A. catenella can have a competitive advantage over other members of the phytoplankton even under low concentrations of ammonium and urea. However, the observed large changes in ammonium and urea uptake kinetics on a short time scale (days) during blooms preclude more precise estimates of those contributions to growth and require further investigation.  相似文献

A high spatial resolution sampling of Alexandrium pacificum cysts, along with sediment characteristics (% H2O, % organic matter (OM), granulometry), vegetative cell abundance and environmental factors were investigated at 123 study stations in Bizerte Lagoon (Tunisia). Morphological examination and ribotyping of cells obtained from a culture called ABZ1 obtained from a cyst isolated in lagoon sediment confirmed that the species was A. pacificum. The toxin profile from the ABZ1 culture harvested during exponential growth phase was simple and composed of the N-sulfocarbamoyl toxins C1 (9.82 pg toxin cell−1), the GTX6 (3.26 pg toxin cell−1) and the carbamoyl toxin Neo-STX (0.38 pg toxin cell−1). The latter represented only 2.8% of the total toxins in this strain.High abundance of A. pacificum cysts correlated with enhanced percentages of water and organic matter in the sediment. In addition, sediment fractions of less than 63 μm were examined as a favorable potential seedbed for initiation of future blooms and outbreaks of A. pacificum in the lagoon. A significant difference in the cyst distribution pattern was recorded among the lagoon's different zones, with the higher cyst abundance occurring in the inner waters. Also, no correlation due to the specific hydrodynamics of the lagoon was observed in the spatial distribution of A. pacificum cysts and vegetative cells.  相似文献

The main goal of this study was to test the effect of [CO2] on C and N management in different plant organs (shoots, roots and nodules) and its implication in the responsiveness of exclusively N2-fixing and NO3-fed plants. For this purpose, exclusively N2-fixing and NO3-fed (10 mM) pea (Pisum sativum L.) plants were exposed to elevated [CO2] (1000 μmol mol−1 versus 360 μmol mol−1 CO2). Gas exchange analyses, together with carbohydrate, nitrogen, total soluble proteins and amino acids were determined in leaves, roots and nodules. The data obtained revealed that although exposure to elevated [CO2] increased total dry mass (DM) in both N treatments, photosynthetic activity was down-regulated in NO3-fed plants, whereas N2-fixing plants were capable of maintaining enhanced photosynthetic rates under elevated [CO2]. In the case of N2-fixing plants, the enhanced C sink strength of nodules enabled the avoidance of harmful leaf carbohydrate build up. On the other hand, in NO3-fed plants, elevated [CO2] caused a large increase in sucrose and starch. The increase in root DM did not contribute to stimulation of C sinks in these plants. Although N2 fixation matched plant N requirements with the consequent increase in photosynthetic rates, in NO3-fed plants, exposure to elevated [CO2] negatively affected N assimilation with the consequent photosynthetic down-regulation.  相似文献

Aerobic production-scale processes are constrained by the technical limitations of maximum oxygen transfer and heat removal. Consequently, microbial activity is often controlled via limited nutrient feeding to maintain it within technical operability. Here, we present an alternative approach based on a newly engineered Escherichia coli strain. This E. coli HGT (high glucose throughput) strain was engineered by modulating the stringent response regulation program and decreasing the activity of pyruvate dehydrogenase. The strain offers about three-fold higher rates of cell-specific glucose uptake under nitrogen-limitation (0.6 gGlc gCDW−1 h−1) compared to that of wild type, with a maximum glucose uptake rate of about 1.8 gGlc gCDW−1 h−1 already at a 0.3 h−1 specific growth rate. The surplus of imported glucose is almost completely available via pyruvate and is used to fuel pyruvate and lactate formation. Thus, E. coli HGT represents a novel chassis as a host for pyruvate-derived products.  相似文献
