In the present study, the EtOAc extract of the persistent calyx of Physalis angulata L. var. villosa Bonati (PA) was tested for its potential quinone reductase (QR) inducing activity with glutathione (GSH) as the substrate using an UPLC–ESI-MS method. The result revealed that the PA had electrophiles that could induce quinone reductase (QR) activity, which might be attributed to the modification of the highly reactive cysteine residues in Keap1. Herein, three new withanolides, compounds 3, 6 and 7, together with four known withanolides, compounds 1, 2, 4 and 5 were isolated from PA extract. Their structures were determined by spectroscopic techniques, including 1H-, 13C NMR (DEPT), and 2D-NMR (HMBC, HMQC, 1H, 1H-COSY, NOESY) experiments, as well as by HR-MS. All the seven compounds were tested for their QR induction activities towards mouse hepa 1c1c7 cells.  相似文献

The main objective of this research is to evaluate the molluscicide activity of Physalis angulata L. Biomphalaria tenagophila specimens under laboratory conditions. Extracts and fractions were supplied by the Laborat rio de Qu mica de Produtos Naturais, Farmanguinhos-Fiocruz. Experiments were performed according to the methodology described by the World Health Organization for molluscicide tests using the concentrations from 0.1 to 500 mg/l of the extracts, fractions and of a pool of physalins modified steroids present in this species. The results show that ethyl acetate and acetone extracts from the whole plant, the ethanolic extracts of the roots and the physalins pool from stems and leaves were active. Only the whole plant extracts were available in sufficient quantity for the determination of LD50 and LD90 values.  相似文献

Physalis angulata is a medicinal plant with valuable pharmacological activities. The physiology of stress in this plant can play an important role in the induction or maximization of the production of physalin F, B, D and G, bioactive secondary metabolites described as immunosuppressive, anti-malarial and anti-leishmanial agents. P. angulata was cultivated from seeds which had been previously primed in PEG 6000 solution and non-primed seeds. After 45 days, the plants were exposed to water restriction and saline stress in the field for 13 days. Seedling emergence and growth after stress treatment were assessed. Seco-steroids were quantified in leaves and stems by HPLC/PDA. The emergence rate was 14% higher in primed seeds. The types of irrigation proved to have a significant influence on the number of leaves and fruits, plant height and stem diameter, irrespective of whether the seeds were primed or not. The biomass of the fruits, stems and roots was also decreased by water restriction and saline stress. Physalin content in ethanol extracts increased in leaves, mainly after saline stress and from primed seeds. Despite the biomass reduction caused by the treatments, stress application led to an increase in the production of bioactive metabolites.  相似文献

BackgroundThe current treatment of Chagas disease, endemic in Latin America and emerging in several countries, is limited by the frequent side effects and variable efficacy of benznidazole. Natural products are an important source for the search for new drugs.Aim/hypothesisConsidering the great potential of natural products as antiparasitic agents, we investigated the anti-Trypanosoma cruzi activity of a concentrated ethanolic extract of Physalis angulata (EEPA).MethodsCytotoxicity to mammalian cells was determined using mouse peritoneal macrophages. The antiparasitic activity was evaluated against axenic epimastigote and bloodstream trypomastigote forms of T. cruzi, and against amastigote forms using T. cruzi-infected macrophages. Cell death mechanism was determined in trypomastigotes by flow cytometry analysis after annexin V and propidium iodide staining. The efficacy of EEPA was examined in vivo in an acute model of infection by monitoring blood parasitaemia and survival rate 30 days after treatment. The effect against trypomastigotes of EEPA and benznidazole acting in combination was evaluated.ResultsEEPA effectively inhibits the epimastigote growth (IC50 2.9 ± 0.1 µM) and reduces bloodstream trypomastigote viability (EC50 1.7 ± 0.5 µM). It causes parasite cell death by necrosis. EEPA impairs parasite infectivity as well as amastigote development in concentrations noncytotoxic to mammalian cells. In mice acutely-infected with T. cruzi, EEPA reduced the blood parasitaemia in 72.7%. When combined with benznidazole, EEPA showed a synergistic anti-T. cruzi activity, displaying CI values of 0.8 ± 0.07 at EC50 and 0.83 ± 0.1 at EC90.ConclusionEEPA has antiparasitic activity against T. cruzi, causing cell death by necrosis and showing synergistic activity with benznidazole. These findings were reinforced by the observed efficacy of EEPA in reducing parasite load in T. cruzi-mice. Therefore, this represents an important source of antiparasitic natural products.  相似文献

Enzyme electrophoretic studies were made in cultivated Solanum melongena L. (eggplant) and similar wild and weedy forms, several of which have been thought to be different species/taxa. Twenty-nine accessions of S. melongena, 33 accessions of weedy forms (referred to as insanum) and 2 accessions of wild forms (referred to as incanum) were surveyed for 29 isozyme loci. In S. melongena, 22 of the 29 loci were monomorphic, and nearly all of its genes were either also monomorphic or in similar frequencies in insanum and incanum. The results demonstrate that the three taxa have a very close genetic relationship. The high genetic identities between them (0.913–0.967) suggests that they are conspecific even though they include extensive morphological diversity.  相似文献

Physalis angulata and P. peruviana are herbs widely used in folk medicine. In this study, the aqueous and ethanol extracts prepared from the whole plant of these species were evaluated for their antihepatoma activity. Using XTT assay, three human hepatoma cells, namely Hep G2, Hep 3B and PLC/PRF/5 were tested. The results showed that ethanol extract of P. peruviana (EEPP) possessed the lowest IC50 value against the Hep G2 cells. Interestingly, all extracts showed no cytotoxic effect on normal mouse liver cells. Treatment with carbonyl cyanide m-chlorophenyl hydrazone, a protonophore, caused a reduction of membrane potential (Deltapsim) by mitochondrial membrane depolarization. At high concentrations, EEPP was shown to induce cell cycle arrest and apoptosis through mitochondrial dysfunction as demonstrated by the following observations: (i) EEPP induced the collapse of Deltapsim and the depletion of glutathione content in a dose dependent manner; (ii) pretreatment with the antioxidant (1.0 microg/ml vitamin E) protected cells from EEPP-induced release of ROS; and (iii) at concentrations 10 to 50 microg/ml, EEPP displayed a dose-dependent accumulation of the Sub-G1 peak (hypoploid) and caused G0/G1-phase arrest. Apoptosis was elicited when the cells were treated with 50 microg/ml EEPP as characterized by the appearance of phosphatidylserine on the outer surface of the plasma membrane. The results conclude that EEPP possesses potent antihepatoma activity and its effect on apoptosis is associated with mitochondrial dysfunction.  相似文献

A series of cobalt(III) mixed ligand complexes of type [Co(en)2L]+3, where L is bipyridine, 1,10-phenanthroline, imidazole, methylimidazole, ethyleimidazole, dimethylimidazole, urea, thiourea, acetamide, thioacetamide, semicarbazide, thiosemicarbazide, or pyrazole, have been isolated and characterized. The structural elucidation of these complexes has been explored by using absorption, infrared, and 1H NMR nuclear magnetic resonance spectral methods. The infrared spectral data of all these complexes exhibit a band at 1450/cm and 1560-1590/cm, which correspond to C=C and C=N, a band at 575/cm for Co-N (en), and a band at 480/cm for Co-L (ligand). All these complexes were found to be potent antimicrobial agents. The antibacterial activity was studied in detail in terms of zone inhibition, minimum bactericidal, and time period of lethal action. Among all, complexes bipyridine, 1,10-phenanthroline, dimethylimidazole, and pyrazole, possess the highest antibacterial activity. Antifungal activity was done by disc-diffusion assay and 50% inhibitory concentrations that possess high antifungal activity.  相似文献

RAPD analysis was carried out on 52 accessions of Solanum melongena (eggplant) and related weedy forms known as insanum. Twenty-two primers amplified 130 fragments. Solanum melongena exhibited 117 of the fragments, all of which were also present in insanum. Insanum displayed an additional 13 fragments not found in S. melongena. Overall, the insanum accessions were more diverse than those of S. melongena. The calculated similarity between them was 0.947. The RAPD results were closely concordant with the results of an electrophoretic isozyme survey performed on the same accessions. The concordance of the results shows that even though S. melongena and insanum are highly diverse morphologically, it is no longer appropriate to distinguish them taxonomically.  相似文献

Phytochemical investigation of the roots of Solanum melongena L. led to the isolation of 16 phenylpropanoid amides (116). Their structures were identified by analysis of spectroscopic data and comparison with those reported in the literature. This is the first report of the natural occurrence of N-trans-sinapoyloctopamine (9) and N-trans-caffeoyloctopamine (10). N-trans-feruloylnoradrenline (12) and N-cis-feruloylnoradrenline (16) were isolated from the genus Solanum for the first time. Four compounds including 3-(4-hydroxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl] acrylamide (5), 3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl] acrylamide (6), N-trans-p-coumaroylnoradrenline (11), and N-cis-p-coumaroyloctopamine (15) were firstly reported from S. melongena. The chemotaxonomic significance of these compounds was summarized.  相似文献

B chromosomes are additional dispensable chromosomes that may be present in some individuals, populations, or species, which have probably arisen from the A chromosomes but follow their own evolutionary pathway. Supposedly, B chromosomes do not contain major genes except for ribosomal DNA (rDNA) sequences that have been mapped on the supernumerary chromosomes of many plants and animals. This paper is a new report of B chromosome occurrence in plants. B chromosomes with nucleolar organizing regions (NORs) were found in a diploid sample of Nierembergiaaristata D. Don (sub nom. N. stricta Miers) (2n = 2x = 16). This is an extreme case in which B chromosomes possess not only strong nucleolar activity, as revealed by conventional staining methods, AgNOR and fluorescence banding, and fluorescent in situ hybridization (FISH), but also show nucleolar competition with the A chromosomes. The observed phenomenon could be analogous to the nucleolar dominance or 'differential amphiplasty' phenomenon that occurs in interspecific hybrids.  相似文献

A morphological analysis ofPhysalis and several closely related genera, with emphasis on a group of species known as “atypicalPhysalis,” showed that two of the atypical species are better treated as a distinct genus.Tzeltalia is described and two new combinations are made:T. amphitricha andT. calidaria. Restricted to the highlands of Chiapas, Mexico, and adjacent Guatemala,Tzeltalia is distinguished fromPhysalis by a combination of characters including shrubby habit; fasciculate flowers; lobed corolla; scabrate pollen; and non-invaginated, partially open, coriaceous, and reticulate-veined fruiting calyx.

Kephir is a fermented carbonated refreshing milk, with a slightly acidic aromatic taste and creamy foam composition which contains lactobacilli, leuconostocci, acetic acid bacteria, lactostreptococci and yeasts. Recent studies have demonstrated its antibacterial, immunostimulating, antitumoral and cholesterol-lowering activities.

Purpose

The purpose of this study was to investigate the antimicrobial activity of kephir against Bacillus subtilis spp. spizizenii ATCC 6633, Staphylococcus aureus ATCC 6538, Enterococcus faecalis ATCC 29212, Escherichia coli ATCC 8739, Salmonella enteritidis ATCC 13076, Pseudomonas aeruginosa ATCC 9027 and Candida albicans ATCC 10231. The kephir fermented for 24 h and 48 h, as well and after 7 days preservation at 4–8 °C was tested by in vitro disk diffusion method. The intensity of the antimicrobial activity was interpreted by comparison with two antibiotics, i.e. ampicillin and neomycin.

Results

The antimicrobial activity of 24 h as well as 48 fermented kephir, fresh or after 7 days preservation at 4–8 °C was similar and observed against B. subtilis, S. aureus, E. coli, E. faecalis and S. enteritidis. For E. coli, E. faecalis and S. enteritidis the antimicrobial activity was superior to both tested antibiotics and for B. subtilis and S. aureus to one antibiotic. The tested products exhibited no activity against P. aeruginosa and C. albicans.

Conclusion

Kephir is exhibiting large spectrum and strong antibacterial activity probably due to the complex viable probiotic strains association producing antimicrobial substances.  相似文献

Seed coat structure in Solanum L. section Solanum (Solanaceae). Seed coat structure in Solanum L. section Solanum was examined using SEM. The seed coat patterns observed were fairly uniform throughout the section, and thus of little diagnostic use. Moreover, all species are characterized by their seeds being covered by hairlike structures, which are bands of lignified thickening in the lateral (and basal) walls of the outer epidermal cells of the testa, and which only become obvious when the seeds are wetted. The development of the seed coat and its characteristic thickening was followed from the unfertilized ovule to the mature seed in vouchered material of Solanum nigrum L. since this species is the generic type, using sections of freshly fixed material.  相似文献

Abstract.— Although insect herbivory is frequently assumed to be responsible for the maintenance of plant secondary metabolites such as alkaloids, the assumption is controversial and experimental evidence for this assumption is sparse. We examined natural selection on the two major alkaloids present in the leaves of Datura stramonium and found that both alkaloids came under active selection. We found negative directional selection for scopolamine (natural selection acting to reduce scopolamine levels) and stabilizing selection for hyoscyamine (natural selection acting to maintain an intermediate level of hyoscyamine). We also present evidence that insect herbivores act as the agents of selection on these alkaloids. Finally, we show that there were no trade-offs in resistance to different species of insects.  相似文献

BackgroundIn the present study, we explored the composition of Cilantro (Coriandrum sativum L. leaves) essential oil (CEO) cultivated in Saudi Arabia (Al-Kharj) and explored its antioxidant, antimicrobial, and anti-inflammatory effects in vitro.MethodsGas chromatography-mass spectroscopy was used to detect the CEO composition. The 2, 2-diphenyl-1-picrylhydrazyl (DPPH)-induced free radical and ferric chloride scavenging methods were used to determine the antioxidant activity. Antimicrobial activity was investigated using the well diffusion method. Anti-inflammatory activity was evaluated using egg albumin and trypsin-induced inflammation methods.ResultsForty-six compounds representing 90.17% of the total aroma were identified in the CEO; the major constituents were found to be 1-decanol (17.85%), decanal (11.04%), trans-2-dodecen-1-ol (7.87%), menthone (6.71%), 2-decen-1-ol, trans- (5.44%), dodecanal (4.76%), trans-tetradec-2-enal (3.14%), sedanolide (3.02), and thymol (3.01%). DPPH-induced free radical and ferric chloride scavenging assays demonstrated low antioxidant effects of CEO, and the antioxidant activity was observed at a high CEO concentration. The antimicrobial activity of CEO was assessed against 5 microorganisms (bacteria and fungi) by using well diffusion methods; CEO was found to possess excellent antimicrobial activity against all microorganisms, except Escherichia coli. Moreover, CEO demonstrated strong anti-inflammatory activity against egg albumin- and trypsin-induced inflammation.ConclusionThe essential oil extracted from C. sativum chemotype grown in Al-Kharj region of Saudi Arabia possesses low antioxidant potential, superior antimicrobial activity, and outstanding anti-inflammatory effects.  相似文献

In this paper, we report studies on morphological, phytochemical, and biological aspects of a population belonging to Aconitum anthora L. Two compounds, quercetin 3-O-((beta-D-glucopyranosyl-(1-->3)-(4-O-(E-p-coumaroyl))-alpha-L-rhamnopyranosyl-(1-->6)-beta-D-galactopyranoside))-7-O-alpha-L-rhamnopyranoside (1) and kaempferol 3-O-((beta-D-glucopyranosyl-(1-->3)-(4-O-(E-p-coumaroyl))-alpha-L-rhamnopyranosyl-(1-->6)-beta-D-galactopyranoside))-7-O-alpha-L-rhamnopyranoside (2), together with two known flavonol glycosides (3-4) were isolated and identified from A. anthora. The antioxidant activity of the four identified flavonoids was screened by three in vitro tests.  相似文献

Plasticity of leaf traits of Solanum incanum was evaluated by comparing its expression in three sites differing in soil moisture (dry, intermediate, humid) in Nguruman, SW Kenya. Leaf size and shape, and leaf traits related to defence against herbivores, varied significantly among sites. The relationship between leaf traits showing among-site variation and insect herbivory was evaluated at a within-site scale. Leaf thickness was correlated with the level of foliar herbivory by a flea beetle ( Epitrix sp.). This correlation was negative at the dry site and positive at the humid site. No relationship was found at the intermediate site. Interestingly, leaf thickness and toughness were positively and significantly correlated at the dry site, but not elsewhere. Finally, overall patterns of herbivory by the flea beetle and unidentified defoliators differed among sites. The importance of scale when evaluating plant plasticity is discussed.  相似文献

This paper reports about a newly developed assay which enables the user to quantify and compare the antimicrobial (anticyanobacterial) activity of both hydrophilic and lipophilic test compounds without any restriction. The assay is characterised by the application of a test compound solution on a porous matrix (silica gel) in well-defined concentrations per unit area, and by the coating of this matrix by a concentrated suspension of the tested living cyanobacterium (using a spraying or a dipping technique). The organism was only applied after the complete evaporation of the solvent used for preparation of the test compound solution to avert potential influences of this solvent on micro-organisms' viability. Toxic concentrations of a test compound caused a clearly contoured regional damage in the blue-green coloured uniform layer of the cyanobacterial test organism. The resulting regional decolourisation was readily identifiable within a species-dependent short time (e.g. for the species Arthrospira laxissima after 24h). This newly developed assay is applied for a patent in Germany.  相似文献

Nolana comprises 89 species almost entirely restricted to the Atacama and Peruvian deserts. These species are adapted to the arid lomas environment of northern Chile and southern Peru, where they depend on marine fog locally known as garúa or camanchaca for their development. Recent molecular phylogenetic studies provide support for the placement of Nolana in the Solanaceae rather than Nolanaceae. Additionally, a sister relationship with the tribe Lycieae has been suggested. The pollen morphology of 111 species from 18 genera of southern South American Solanaceae was investigated under light and scanning electron microscopes in order to establish the palynological affinities of Nolana within the Solanaceae and to investigate if pollen morphology is congruent with the phylogenetic relationships and former taxonomic classification of the species of Nolana. The following genera were studied: Nolana, Cestrum, Combera, Dunalia, Fabiana, Jaborosa, Latua, Lycium (incl. Grabowskia, Phrodus), Nicandra, Nicotiana, Nierenbergia, Petunia, Physalis, Reyesia, Salpiglossis, Schizanthus, Solanum and Vestia. Two pollen types that differ in exine structure were distinguished within Nolana; these pollen types are congruent with recent phylogenetic studies, thus, we propose to reinstate Nolana sect. The present work aimed to investigate the antioxidant, anti-inflammatory and wound healing potential of ethyl acetate fraction from Bauhinia ungulata L. (FABU) on in vitro and in vivo models. Wound healing assay using human lung adenocarcinoma A549 cell line was employed to evaluate the ability of FABU in modulating cell migration. In addition, a surgical wound model in C57BL/6 mice was used to study the healing potential of FABU incorporated into gel carbomer 940 (Carbopol®). Evaluation of lipid peroxidation, inflammatory and anti-inflammatory mediator gene expression, rate of wound closure, and histological analysis were done. FABU significantly reduced the gap area in in vitro wound healing assay, 24 h after treatment. In the animal model, FABU at 0.5% topically applied once-daily for 5 days to the surgical wounds significantly reduced the lesion area. Moreover, it significantly decreased the levels of lipid peroxidation in the lesions and decreased the relative gene expression levels of IL-1β and TNF-α in the injured region. In conclusion, our study suggests that Bauhinia ungulata can effectively promote the wound healing, probably by regulating the inflammatory environment during the early stages of the process.
