Ketosis in dairy cows can lead to poor reproductive success and decreased milk production. Since the serum concentrations of copper (Cu) and zinc (Zn) are closely associated with the health status of cows, we investigated whether serum concentrations of Cu and Zn differed in dairy cows with subclinical ketosis and healthy dairy cows. Blood samples of 19 healthy dairy cows and 15 subclinically ketotic dairy cows were collected from three farms, and the concentrations of β-hydroxybutyrate (BHBA), glucose, non-esterified fatty acids (NEFA), Cu, and Zn were determined. Subclinically ketotic dairy cows had significantly higher BHBA and NEFA levels (p?<?0.01) and lower glucose (p?<?0.01) than healthy dairy cows. Likewise, serum concentrations of Zn were significantly decreased (p?<?0.05) in dairy cows with subclinical ketosis. There was no significant difference observed for serum Cu concentration between healthy and subclinically ketotic dairy cows. This study suggests that a decreased serum Zn concentration could be a cause of decreased reproductive performance in subclinically ketotic dairy cows.  相似文献

The objective of this study was to determine the levels of serum copper and zinc in subclinically hypocalcemic peripartum dairy cows in comparison to healthy animals. Blood samples were taken from 219 multiparous Holstein cows near parturition (from 4 weeks prepartum to 4 weeks postpartum) and 51 cows with subclinical hypocalcemia. The results showed that the serum copper concentration increased gradually at 1 week prepartum and remained high for the first 4 weeks postpartum in the healthy periparturient dairy cows. The serum zinc concentration reached a nadir at 1 week postpartum and subsequently increased gradually to baseline. The serum zinc concentration was significantly decreased (P?<?0.01) in dairy cows with subclinical hypocalcemia compared with healthy cows. There was no significant difference in the serum copper concentration between cows with subclinical hypocalcemia and healthy cows. These data demonstrate that the concentrations of copper and zinc in serum change dramatically during the peripartum period in dairy cows, which is a tremendous challenge for the body and for the maintenance of dairy cow health. The present study further suggests that a decreased serum zinc concentration could be a cause of decreased productive performance and increased susceptibility to other diseases due to immunosuppression in dairy cows with subclinical hypocalcemia. Additionally, this decreased zinc concentration may be involved in the pathogenesis of subclinical hypocalcemia.  相似文献

Initiation of long-term treatment with rbST (Posilac, Monsanto, St. Louis, MO) coincident with first insemination increased pregnancy rates in dairy cattle, but neither the efficacy of using only the initial injection, nor its effects on retention of pregnancy are known. Lactating dairy cows, dairy heifers, and lactating beef cows were assigned at random to treatment (rbST) or control. Dairy cows, dairy heifers, and beef cows received 500 mg rbST (n = 48, 35, 137 inseminations, respectively) at artificial insemination or were left untreated (n = 62, 33, 130 inseminations, respectively). Pregnancy was diagnosed by ultrasonography at 28-36 days. Treatment with rbST at insemination improved conception rates in dairy cows (60.4% versus 40.3%; P < 0.05), but not in dairy heifers or beef cows. Conception rates did not differ in dairy cows at < or =100 days in milk (DIM), but were improved in cows treated with rbST after 100 DIM (64.3% versus 25.8%; P < 0.05). Retention of pregnancy to approximately 60 days and sizes of CL, diameter of follicles > or =5 mm, and crown-rump lengths of embryos were not affected by treatment. The second objective was to examine the effects of rbST at insemination on birth weight and post-natal calf growth in beef cows. However, birth and weaning weights of beef calves were not affected by treatment. In conclusion, a single treatment with rbST at insemination increased conception rates in dairy cows, specifically in those >100 DIM.  相似文献

目的研究奶牛阴道菌群,并从健康奶牛阴道分离出产酸能力很强的乳酸菌。方法采用常规的方法对奶牛阴道进行细菌的分离及鉴定,并进行菌群分析。结果健康奶牛阴道优势菌群主要为乳酸菌(P<0.01),屡配不孕奶牛阴道优势菌群主要为金黄色葡萄球菌(P<0.01);从健康奶牛阴道分离出的乳酸菌为55株,其中产酸能力很强的6株乳酸菌鉴定结果分别为Lactobacillus plantarum、Lactobacillus brevis、Enterococcus faecalis、Lactococcus garvieae、Lactobacillus kitasatonis和Lactobacillus amylovorus。结论奶牛阴道菌群中分离的6株乳酸菌可作为潜在的奶牛阴道微生态制剂进行深入研究。  相似文献

Fungal infection of reproduction system of dairy cattle have not been received much more attention. The aim of this study was to determine the fungal infection of Holstein dairy cows with reproductive disorders or healthy. Fungal isolates of cervicovaginal fluids of 176 Holstein dairy cows were collected by using the double rod swabs for cervix and the sterilized cotton swabs for discharges of vagina. They were evaluated for fungal infections. The treatment group included 70 dairy cows with reproductive diseases, such as abortion, repeat breeder, endometritis, metritis, retention of fetal membrane, dystocia, cervicitis, and vaginitis. The control groups were included 42 healthy non-pregnant cows and 64 pregnant cows. Isolates of fungi were obtained from cervix and vagina of 27.1% and 28.6% of treatment group, 26.7% and 31.2% of pregnant cows, and 33.3% and 21.4% of healthy non-pregnant cows, respectively, indicating no significant differences. It is showed that the cervix and vagina of the treatment group have been infected by six different mycotic isolates. However, the cervix and vagina of pregnant and non-pregnant healthy cows in control group were infected with 5, 6 and 5, 4 different fungal agents, respectively. Penicillium and yeast were the most common isolated agents. Regarding to the result of this study, it is concluded that fungal infections can occur in cervicovaginal cavity of Holstein dairy cows with or without reproductive diseases.  相似文献

Reproductive efficiency is not optimal in high-producing dairy cows. Although many aspects of ovarian follicular growth in cows are similar to those observed in heifers, there are numerous specific differences in follicular development that may be linked with changes in reproductive physiology in high-producing lactating dairy cows. These include: 1) reduced circulating estradiol (E2) concentrations near estrus, 2) ovulation of follicles that are larger than the optimal size, 3) increased double ovulation and twinning, and 4) increased incidence of anovulation with a distinctive pattern of follicle growth in anovular dairy cows. The first three changes become more dramatic as milk production increases, although anovulation has not generally been associated with level of milk production. To overcome reproductive inefficiencies in dairy cows, reproductive management programs have been developed to synchronize ovulation and enable the use of timed AI in lactating dairy cows. Effective regulation of the CL, follicles, and hormonal environment during each part of the protocol is critical for optimizing these programs. This review discusses the distinct aspects of follicular development in lactating dairy cows and the methodologies that have been utilized in the past two decades in order to manage the dominant follicle during synchronization of ovulation and timed AI programs.  相似文献

Early embryonic death of calves due to sub-fertility in cows is of great economic concern to dairy industry. Early pregnancy factor (EPF) is a secretory protein with pregnancy associated immunosuppressive properties. Rosette inhibition test (RIT) was used to detect EPF in inseminated dairy cows. Blood samples were collected at two intervals, 1-3 and 5-7 days after insemination from 23 inseminated and 18 non-inseminated control cows for RIT and pregnancy diagnosis performed between 42 and 45 days on palpation. The study indicates that RIT (P<0.05) has the potential to distinguish pregnant from non-pregnant dairy cows in the first week of pregnancy.  相似文献

Hormone ghrelin and orotic acid accelerate wound healing as well as controlling inflammation and immunity. We have, therefore, investigated the serum and milk levels of ghrelin and orotic acid in dairy cows with (n = 21) or without (n = 21) subclinical mastitis. Acylated and des-acylated ghrelin as well as orotic acid concentration were detected by using high performance liquid chromatography (HPLC). The results revealed that ghrelin level in milk and serum was significantly higher in dairy cows with subclinical mastitis than that of dairy cows without subclinical mastitis. This was also the case when the orotic acid concentrations in dairy cows with subclinical mastitis were compared with those dairy cows without subclinical mastitis. In conclusion, ghrelin and orotic acid occur in particularly high concentrations in subclinical mastitis, and might, therefore, be required in greater amounts for tissue repair and may be also used as a indicator for subclinical mastitis.  相似文献

The objective of this study was to evaluate the effects of GnRH administered at Day 12 post-AI on the reproductive performance of dairy cows. Holstein-Friesian dairy cows (n=103) on a large Hungarian dairy farm were allocated randomly to treated (n=54) or control (n=49) groups. Twelve days after AI, treated cows received a GnRH agonist i.m., while the control group received a placebo (physiological saline). Progesterone radioimmunoassay was used to determine the correct timing of artificial insemination (Day 0) and the incidence of luteal insufficiency on Day 12. Ultrasonography and radioimmunoassay for pregnancy-associated glycoprotein were used to detect pregnancy and late embryonic/fetal mortality between Days 32 and 55 after AI. Three cows from each group were inseminated when progesterone concentrations were >1.0 ng/mL, and six cows (four from the treated and two from the control group) had luteal insufficiency (progesterone<1.0 ng/mL) on Day 12. Late embryonic/fetal mortality occurred in three treated cows and in two control cows. When these cows were removed from the model, calving rates after first service were 59.6% (28/47) and 59.1% (26/44) for treated and control cows, respectively (P>0.05). There was no significant difference between treated and control cows when they were inseminated before or after Day 100 from calving. In summary, administration of a GnRH agonist on Day 12 after AI did not improve reproductive performance in dairy cows. However, our approach may be used for the field evaluation of different treatment protocols.  相似文献

Brucellosis is a bacterial disease caused by brucella; mainly spread by direct contact transmission through the brucella carriers, or indirect contact transmission by the environment containing large quantities of bacteria discharged by the infected individuals. At the beginning of 21st century, the epidemic among dairy cows in Zhejiang province, began to come back and has become a localized prevalent epidemic. Combining the pathology of brucellosis, the reported positive data characteristics, and the feeding method in Zhejiang province, this paper establishes an dynamic model to excavate the internal transmission dynamics, fit the real disease situation, predict brucellosis tendency and assess control measures in dairy cows. By careful analysis, we give some quantitative results as follows. (1) The external input of dairy cows from northern areas may lead to high fluctuation of the number of the infectious cows in Zhejiang province that can reach several hundreds. In this case, the disease cannot be controlled and the infection situation cannot easily be predicted. Thus, this paper encourages cows farms to insist on self-supplying production of the dairy cows. (2) The effect of transmission rate of brucella in environment to dairy cattle on brucellosis spreading is greater than transmission rate of the infectious dairy cattle to susceptible cattle. The prevalence of the epidemic is mainly aroused by environment transmission. (3) Under certain circumstances, the epidemic will become a periodic phenomenon. (4) For Zhejiang province, besides measures that have already been adopted, sterilization times of the infected regions is suggested as twice a week, and should be combined with management of the birth rate of dairy cows to control brucellosis spread.  相似文献

The expression of oestrous behaviour in Holstein Friesian dairy cows has progressively decreased over the past 50 years. Reduced oestrus expression is one of the factors contributing to the current suboptimal reproductive efficiency in dairy farming. Variation between and within cows in the expression of oestrous behaviour is associated with variation in peripheral blood oestradiol concentrations during oestrus. In addition, there is evidence for a priming role of progesterone for the full display of oestrous behaviour. A higher rate of metabolic clearance of ovarian steroids could be one of the factors leading to lower peripheral blood concentrations of oestradiol and progesterone in high-producing dairy cows. Oestradiol acts on the brain by genomic, non-genomic and growth factor-dependent mechanisms. A firm base of understanding of the ovarian steroid-driven central genomic regulation of female sexual behaviour has been obtained from studies on rodents. These studies have resulted in the definition of five modules of oestradiol-activated genes in the brain, referred to as the GAPPS modules. In a recent series of studies, gene expression in the anterior pituitary and four brain areas (amygdala, hippocampus, dorsal hypothalamus and ventral hypothalamus) in oestrous and luteal phase cows, respectively, has been measured, and the relation with oestrous behaviour of these cows was analysed. These studies identified a number of genes of which the expression was associated with the intensity of oestrous behaviour. These genes could be grouped according to the GAPPS modules, suggesting close similarity of the regulation of oestrous behaviour in cows and female sexual behaviour in rodents. A better understanding of the central genomic regulation of the expression of oestrous behaviour in dairy cows may in due time contribute to improved (genomic) selection strategies for appropriate oestrus expression in high-producing dairy cows.  相似文献

Antibodies to Neospora caninum were measured in bovine foetuses, dairy cows and beef cows in Argentina using the IFAT, the N. caninum agglutination test, and the recombinant NCDG1 and NCDG2 ELISA. Serum antibodies (IFAT titre 1:80) were found in 20 of 82 (24.4%) dairy cow foetuses and one of 22 (4.5%) beef cow foetuses. Microscopic lesions suggestive of neosporosis were seen in brains of seven of eight foetuses with IFAT titres of 1:80. Antibodies (IFAT) were found in 122 of 189 (64.5%) dairy cows that aborted. Serum antibody titres (IFAT) of 189 dairy cows that aborted were: < 1:25 (67 cows), 1:25 (four cows), 1:50 (16 cows), 1:200 (seven cows), 1:> or = 800 (95 cows). Of the 87 sera with IFAT titres of < or = 1:50, 57 had no antibodies in 1:40 dilution and 30 had titres of 1:40 in the N. caninum agglutination test. Thus, sera from at least 56 dairy cows which had aborted were seronegative both in the N. caninum agglutination test and the IFAT. The distribution of positive and negative sera was similar when measured by ELISA, except that, depending on cut-off titre, the ELISA indicated a greater number of seropositive cows that were negative by the IFAT and N. caninum agglutination test. These results suggest that transplacental transmission of N. caninum in dairy cows in Argentina is frequent.  相似文献

Calcium homeostasis is crucial for the normal function of the organism. Parathyroid hormone, calcitriol and calcitonin play critical roles in the homeostatic regulation of calcium. Serotonin and prolactin have also been shown to be involved in the regulation of calcium homeostasis. In modern dairy cows, the endocrine pathways controlling calcium homeostasis during non-lactating and non-pregnant physiological states are unable to fully support the increased demand of calcium required for milk synthesis at the onset of lactation. This review describes different endocrine systems associated with the regulation of calcium homeostasis in mammalian species around parturition with special focus on dairy cows. Additionally, classic and novel strategies to reduce the incidence of hypocalcemia in parturient dairy cows are discussed.  相似文献

There is increasing interest in the use of continuous housing systems for dairy cows, with various reasons put forward to advocate such systems. However, the welfare of dairy cows is typically perceived to be better within pasture-based systems, although such judgements are often not scientifically based. The aim of this review was to interrogate the existing scientific literature to compare the welfare, including health, of dairy cows in continuously housed and pasture-based systems. Although summarising existing work, knowledge gaps and directions for future research are also identified. The scope of the review is broad, examining relevant topics under three main headings; health, behaviour and physiology. Regarding health, cows on pasture-based systems had lower levels of lameness, hoof pathologies, hock lesions, mastitis, uterine disease and mortality compared with cows on continuously housed systems. Pasture access also had benefits for dairy cow behaviour, in terms of grazing, improved lying/resting times and lower levels of aggression. Moreover, when given the choice between pasture and indoor housing, cows showed an overall preference for pasture, particularly at night. However, the review highlighted the need for a deeper understanding of cow preference and behaviour. Potential areas for concern within pasture-based systems included physiological indicators of more severe negative energy balance, and in some situations, the potential for compromised welfare with exposure to unpredictable weather conditions. In summary, the results from this review highlight that there remain considerable animal welfare benefits from incorporating pasture access into dairy production systems.  相似文献

The effects of administration of synthetic thyrotropin-releasing hormone (TRH) on circulating growth hormone (GH), PROLACTIN (PRL) and triiodothyronine (T3) levels of lactating dairy cows, non-lactating dairy heifers, and beef cows were studied. Intravenous administration of 0.1, 1, and 5 microgram of TRH per kg of body weight (bw) elevated plasma GH and PRL levels of lactating cows within 5 min. The plasma GH and PRL levels increased in proportion to the dose of TRH and reached a peak 10 to 30 min after TRH injection. Intravenous administration of 1 microgram of TRH per kg of bw to 7 non-lactating heifers, 14 lactating dairy cows, and 5 non-lactating beef cows elevated plasma GH level to peak values after 15 min, the increase rates being 6.9, 5.6, and 3.8 times as high as those in the pretreatment levels. The mean maximum vale was also in that order. Plasma T3 levels of non lactating dairy heifers at pre- and post-injection of TRH were significantly higher than those of lactating cows. The peak values of plasma PRL were obtained between 5 to 30 min after TRH administration. The increase rates of lactating dairy cows, heifers, and beef cows were 19.2, 13.9, and 20.9 times as high as those in the pretreatment. In contrast to GH and T3, plasma PRL levels of both pre- and post-injection with TRH in lactating cows and heifers were significantly higher in May than in October, though the increase rates were similar. Plasma PRL levels of lactating dairy cows at pre- and post-injection with TRH were significantly higher than those of non-lactating heifers. Subcutaneous administration of TRH was also effective to increase plasma TH, rl, and T3 levels in lactating cows. No significant change of GH or PRL response to TRH was observed after a short-term pretreatment of thyroid hormones.  相似文献

The metabolic adaptation of dairy cows during the transition period has been studied intensively in the last decades. However, until now, only few studies have paid attention to the genetic aspects of this process. Here, we present the results of a gene-based mapping and pathway analysis with the measurements of three key metabolites, (1) non-esterified fatty acids (NEFA), (2) beta-hydroxybutyrate (BHBA) and (3) glucose, characterizing the metabolic adaptability of dairy cows before and after calving. In contrast to the conventional single-marker approach, we identify 99 significant and biologically sensible genes associated with at least one of the considered phenotypes and thus giving evidence for a genetic basis of the metabolic adaptability. Moreover, our results strongly suggest three pathways involved in the metabolism of steroids and lipids are potential candidates for the adaptive regulation of dairy cows in their early lactation. From our perspective, a closer investigation of our findings will lead to a step forward in understanding the variability in the metabolic adaptability of dairy cows in their early lactation.  相似文献

The severity of heat stress conditions in high-yielding dairy cows is currently underestimated. The present study aimed to determine the heat load threshold of the temperature-humidity index (THI) on physiological parameters of lactating Holstein-Friesian cows under a continental climatic zone in Germany. Physiological parameter measurements, such as respiration rate (RR), measured hourly, and heart rate (HR) and rectal temperature (RT), both measured twice daily, were performed in a total of 139 multiparous cows on three randomly chosen measurement days per week. In addition, the ambient temperature and relative humidity of the barn were recorded every 5 min to calculate the current THI. The physiological parameter data were linked to the THI, and the heat load thresholds were determined using the broken-stick model. The heat load duration effect of each physiological parameter was obtained by regression analysis. Considering the increases in the physiological parameters, our study provided reliable data to determine heat load thresholds for lactating high-yielding dairy cows in a moderate climatic zone. The heat load threshold could be determined for RR in standing cows (THI = 70) and lying cows (THI = 65) and for HR (THI = 72) and RT (THI = 70) in standing cows. The heat load duration also demonstrated a significant effect on the increases in physiological parameters among dairy cows. In particular, the present study enabled a strategy to be devised to initiate heat mitigation in high-yielding dairy cows when they are exposed to THIs above 65.  相似文献

Early detection of post-calving health problems is critical for dairy operations. Separating sick cows from the herd is important, especially in robotic-milking dairy farms, where searching for a sick cow can disturb the other cows’ routine. The objectives of this study were to develop and apply a behaviour- and performance-based health-detection model to post-calving cows in a robotic-milking dairy farm, with the aim of detecting sick cows based on available commercial sensors. The study was conducted in an Israeli robotic-milking dairy farm with 250 Israeli-Holstein cows. All cows were equipped with rumination- and neck-activity sensors. Milk yield, visits to the milking robot and BW were recorded in the milking robot. A decision-tree model was developed on a calibration data set (historical data of the 10 months before the study) and was validated on the new data set. The decision model generated a probability of being sick for each cow. The model was applied once a week just before the veterinarian performed the weekly routine post-calving health check. The veterinarian’s diagnosis served as a binary reference for the model (healthy–sick). The overall accuracy of the model was 78%, with a specificity of 87% and a sensitivity of 69%, suggesting its practical value.  相似文献
