Summary In the present study, synaptic ribbons were studied morphologically and quantitatively in hamster pineal gland. The number of ribbons and spherules of hamster pinealocytes was counted over a 24-h period. The 24-h variations in the quantity of synaptic ribbons were found to parallel fluctuations in pineal melatonin concentrations. No significant circadian changes were observed for synaptic spherules, indicating different roles for these two structures.  相似文献

Summary Myotendinous junctions in the myotomal tail muscles of the tadpole of Rana rugosa were examined by electron microscopy. At the site of the myotendinous junction, the sarcolemma is covered on its sarcoplasmic aspect by the connecting filament layer and the attachment layer, and on the extracellular aspect by the intermediary layer and the external lamina, with associated collagen fibrils. The intermediary layer consists of filamentous structures which closely resemble microfibrils (Hanak and Böck, 1971), spine-like or thread-like profiles (Korneliussen, 1973) and intermediary layer (Nakao, 1975a, b) in the myotendinous junctions of other vertebrate skeletal muscles.Particularly interesting is the fact that all the coverings and linings of the sarcolemma, including the external lamina, are completely absent in the terminal segment of the finger-like sarcolemmal invagination characteristic of the myotendinous junction. Furthermore, special types of coupling between a sac of sarcoplasmic reticulum and a part of the sarcolemmal invagination are frequently observed. These couplings always occur along the region of the sarcolemma where the external lamina is absent. The couplings show features similar to those of the triad, such as SR feet , scalloped SR membranes and granular content of the SR sac, suggesting that they are analogous and functionally similar to the triad and other equivalent structures.  相似文献

The organic-matter carbon isotope discrimination () of lichens with a wide range of photobiont and/or cyanobiont associations was used to determine the presence or absence of a carbon-concentrating mechanism (CCM). Two groups were identified within the lichens with green algal photobionts. One group was characterised by low, more C4-like values ( < 15), the other by higher, more C3-like values ( > 18). Tri-partite lichens (lichens with a green alga as the primary photobiont and cyanobacteria within internal or external cephalodia) occurred in both groups. All lichens with cyanobacterial photobionts had low values ( < 15). The activity of the CCM, organic-matter values, on-line values and gas-exchange characteristics correlated with the presence of a pyrenoid in the algal chloroplast. Consistent with previous findings, lichens with Trebouxia as the primary photobiont possessed an active CCM while those containing Coccomyxa did not. Organic values for lichens with Stichococcus as the photobiont varied between 11 and 28. The lichen genera Endocarpon and Dermatocarpon (Stichococcus + pyrenoid) had C4-like organic values ( = 11 to 16.5) whereas the genus Chaenotheca (Stichococcus — pyrenoid) was characterised by high C3-like values ( = 22 to 28), unless it associated with Trebouxia ( = 16). Gas-exchange measurements demonstrated that Dermatocarpon had an affinity for CO2 comparable to those species which possessed the CCM, with K0.5 = 200–215 1 · 1–1, compensation point () = 45–48 l · l–1, compared with K0.5 = 195 1 · 1–1, = 441 · 1–1 for Trebouxioid lichens. Furthermore, lichens with Stichococcus as their photobiont released a small pool (24.2 ± 1.9 to 34.2 ± 2.5 nmol · mg–1 Chl) of inorganic carbon similar to that released by Trebouxioid lichens [CCM present, dissolved inorganic carbon (DIC) pool size = 51.0 ± 2.8 nmol · mg–1 Chl]. Lichens with Trentepohlia as photobiont did not possess an active CCM, with high C3-like organic values ( = 18 to 23). In particular, Roccella phycopsis had very high on-line values ( = 30 to 33), a low affinity for CO2 (K0.5 = 400 1 · 1–1, = 120 1 · –1) and a negligible DIC pool. These responses were comparable to those from lichens with Coccomyxa as the primary photobiont with Nostoc in cephalodia (organic = 17 to 25, on-line = 16 to 21, k0.5 = 388 1 · 1–1, = 85 1 · 1–1, DIC pool size = 8.5 ± 2.4 nmol · mg–1 Chl). The relative importance of refixation of respiratory CO2 and variations in source isotope signature were considered to account for any variation between on-line and organic . Organic was also measured for species of Anthocerotae and Hepaticae which contain pyrenoids and/or Nostoc enclosed within the thallus. Cultured grape cells accumulate anthocyanins in vacuoles rather than secreting them into the nutrient medium. Therefore, grape cells that contain tonoplast segments in their plasmalemma should be capable of excreting anthocyanins rather than sequestering them in their vacuoles. In initial attempts to construct such novel cells, small vacuoles were fused with the plasmalemma of cultured plant cells. Protoplasts were isolated from grape calluses that produce and accumulate anthocyanins. Small vacuoles were formed by gently rupturing vacuoles isolated from grape protoplasts. Although small vacuoles and protoplasts became aligned in an AC field, the tonoplast and plasmalemma did not readily fuse when subjected to 3 DC pulses of 1200 V cm–1 for 50 s each. Changes in the intensity, number and/or duration of the DC pulses had no effect on the fusion process. When 1.0% polyethylene glycol was added to the electrofusion buffer, however, small vacuoles and protoplasts fused within a few minutes after the DC pulses were applied. Synopsis Oocytes of the slug,Limax flavus, display cytoplasmic and nucleolar fluorescence in the presence of 8-anilino-1-naphthalene sulphonic acid, a fluorescent probe for hydrophobic groups. When nuclei are isolated in a balanced salt solution, or in 0.25 m sucrose, chromosomes also become fluorescent, but chromosomes never display fluorescence in intact cells. In the complex multiform nucleolus characteristic of slug oocyte nuclei, the nucleolar cap displays conspicuously high levels of fluorescence and patterns of differential fluorescence are obvious. It would appear that this and similar reagents could be of significant value in the investigation of nucleoli in supravital and unfixed preparations of isolated nuclei.  相似文献

Summary Aluminum toxicity limits root growth in acid subsoils that are difficult to lime. An alternative to subsoil liming is the development of plants having greater tolerance to Al. Alfalfa (Medicago sativa L.) is considered an Al-susceptible species. Preliminary studies indicated that alfalfa cultivars differ in Al tolerance, but the extreme plant-to-plant variation within cultivars prevented the establishment of clearcut cultivar differences.Tolerant and susceptible plants were selected from each of six cultivars (DuPuits, Atlantic, Team, Buffalo, Grimm, and Sirsa 9) grown on an Al-toxic Bladen soil at pH 4.1 to 4.3. The tolerant selections were repotted and interpollinated to form one population of polycross seed. Susceptible selections were treated similarly to form a second population. These two populations, tolerant and susceptible, were subjected to an additional cycle of recurrent phenotypic selection for tolerance and susceptibility, respectively, to Al-toxic Bladen soil at pH 4.6.Plants from the population selected for tolerance to the acid Bladen soil were significantly higher in both root and top vigor on Al-toxic Tatum soil than plants from the population selected for susceptibility. The results indicated that Al tolerance is a heritable trait in these alfalfa populations and that recurrent selection can be used effectively to develop strains having differential tolerance to Al-toxic soils. The observation that only 2% of the plants from the tolerant population were in the most tolerant class suggests a good opportunity for more progress in selecting toward Al tolerance.  相似文献

Effects of various concentrations of Dormex (a.i. 49% hydrogen cyanamide) on fruit thinning of Rome Beauty apple (Malus domestica Borkh.), Friar and Simka plums (Prunus salicina Lindley) were studied. A full bloom application of Dormex at all tested concentrations decreased Rome Beauty apple fruit set and yield, and increased fruit weight. Dormex at 0.25% (v/v) resulted in adequate apple thinning, indicated by production of an optimum fruit weight (320 g). Prebloom and full bloom applications of Dormex at greater than 0.75% reduced plum fruit set and yield in Friar. Full bloom application of Dormex at 0.50% showed a satisfactory fruit set, yield, and fruit weight in Friar plum. Prebloom Dormex application had no significant effect on `Simka' plum fruit set or yield, but full bloom application decreased fruit set and yield.  相似文献

Four methoxypolyethylene glycols (MPEG, molecular masses 350, 750, 2000 and 5000 Da), each activated by nitrophenyl chloroformate, were used to modify trypsin. Compared with the native trypsin, the MPEG-modified trypsin was more stable against temperature between 30°C and 70°C, longer chain of MPEG moiety corresponding to higher thermal stability. The T for the native and the modified trypsin (0.4 mg ml–1) was increased from 47°C to 66°C. The stabilization effect caused by MPEG modification was the result of decreasing in both the autolysis rate and the thermal denaturation rate. The thermodynamic analysis of the thermal denaturation process showed that the activation free energy (G*) of the native and the modified trypsin at 60°C was increased from 102.9 to 109.3 kJ mol–1; the activation enthalpy (H*) was increased from 57.4 to 86.9 kJ mol–1; the activation entropy (S*) was increased from –136 to –67 J molK–1. A possible explanation for the decreased thermal denaturation rate caused by MPEG modification was also discussed.  相似文献

Summary An approximate method to determine sample size for the estimation of population variance, 2, is given. The estimate of 2 is denoted as s2 . Based on the assumption of a normal distribution for (s2/2–1), the sample size is approximately equal to 20,000 z2 p,/k2; where z is a standard normal deviate, p is the probability that s2 ( 100¦s22¦/2) is less than, or equal to, a critical value k, and k (measured as gDs2) is the desired precision of s2 .The expected value of s2, with respect to sample size, and the expected cumulative frequencies of s2 over sample size for various k values are given. Their goodness of fit to the observed results was satisfactory except for populations that were different from normal. The observed values were taken from a study on four yield components in five sugarcane polycross progenies, grown in two contrasting environments over 2 years in three selection stages.The expected s2 was found to be independent of the population coefficient of variance.Research suppoted in part by USDA, ARS, grant #12-14-5001-34. Summary The mucus content in the mormyromast ofGnathonemus petersii (Mormyridae) has been studied with specific histochemical methods for mucopolysaccharides; P.A.S. reaction, toluidine blue and alcian blue. The present work reports observations concerning the use of buffered toluidine blue according to Lison's technique.The existence of the polysaccharides of different constitutions is demonstrated: in the intraepidermal cavity and on the top of the sensory cells (cs 1) the mucus is P.A.S. negative and metachromatic. On the contrary, the mucus is P.A.S. positive in the canal which joins the intraepidermal cavity and in the sensory cavity of the sensory cells (cs 2). The metachromatic reaction of mucopolysaccharides filling the intraepidermal cavity is due to the sulfuric acid radicals; the reaction occurs at very low pH (0.6) and is suppressed by methanol HCL. The successive use of P.A.S. reaction and alcian blue demonstrates the coexistence of acid and neutral mucus.The structure situated on the top of the sensory cells (cs 2) and considered by Szabo and Barets (1963) as a statolith does not contain any calcium salts. It is constituted of polysaccharides much similar to the mucopolysaccharides of the intraepidermal cavity.The mucus identified in the mormyromasts are different from those present in the ampullary organs (Denizot, 1970) and in the Knollenorgan. This indicates a specificity of the mucopolysaccharides for each type of organ.Résumé Les mucus présents au niveau des mormyromastes deGnathonemus petersii (Mormyridés) ont été étudiés avec des techniques histochimiques spécifiques des mucopolysaccharides: réaction au P.A.S., au bleu alcian et au bleu de toluidine. Dans le présent travail, nous donnons des précisions quant à l'emploi du bleu de toluidine en solution tampon selon la méthode de Lison.Ces méthodes nous ont permis de démontrer que les polysaccharides existant dans cet organe sensoriel sont de constitution différente: au niveau de la cavité intraépidermique et des cellules sensorielles (cs 1), le mucus est P.A.S. négatif et métachromatique; par contre, au niveau du canal reliant la cavité intraépidermique et la loge sensorielle des cellules sensorielles (cs 2) le mucus est P.A.S. positif. La métachromasie se manifeste même aux pH très bas (0,6) pour les mucopolysaccharides de la cavité; elle est détruite par l'alcool méthylique acide, la métachromasie étant due à des radicaux acides sulfuriques. L'emploi successif de la réaction au P.A.S. et au bleu alcian montre qu'au niveau de la cavité intraépidermique et du canal, les mucus acides et neutres coexistent.Les différents fixateurs conservent la structure située à l'apex des cellules sensorielles (cs 2) qui a été, en raison de sa position, assimilée par Szabo et Barets (1963) à un statolithe; cependant, ce dernier n'est pas lié à des sels calcaires, mais à des polysaccharides présentant beaucoup de similitudes avec les mucopolysaccharides de la cavité intraépidermique. Les mucus identifiés dans le mormyromaste diffèrent de ceux présents dans l'organe ampullaire (Denizot, 1970) et au niveau du Knollenorgan (Denizot, en préparation). Ceci indique une spécificité des mucopolysaccharides pour chaque type d'organe.

Summary In order to characterize a -amylase deficiency in the endosperm of mutant rye lines, homologous cDNA probes were prepared. A rye cDNA library was constructed from a normal line and screened with a barley -amylase probe. Three partial cDNA clones specific for endosperm -amylase in rye were isolated and characterized. The largest of these clones was used to investigate the expression of endosperm -amylase in mutant and normal lines by Northern hybridization. These experiments, as well as in vitro translation experiments, demonstrate the absence of endosperm -amylase mRNA in mutant lines. Sequencing of three different cDNA clones revealed a single nucleotide difference, which suggests that two genes encoding endosperm -amylase genes might exist in rye. From Southern blots we anticipate that these two genes are tightly linked. Results of these experiments and previous data indicating that the mutation was located within the -amylase locus on chromosome 5 are consistent with the hypothesis that the mutation results from a deletion simultaneously affecting the two genes. However, due to extensive polymorphism within normal lines used as control, additional experiments will be required to further substantiate this conclusion. The deduced amino acid sequence reveals the occurrence of three short glycine-rich repeats containing 11 or 12 residues close to the carboxyl terminus of the protein. A comparison of the nucleotide sequence between rye and previously described barley cDNA clones revealed ca. 90% homology at the amino acid level, except in this C-terminal repeated part, where it drops to 45%.  相似文献

Spin-state selective experiments, HSQC-/ and CT-HMQC-/, are proposed for the simple and rapid measurement of scalar one-bond coupling constants in two-dimensional,1 H-detected 15N-1H or13 C-1H correlation experiments based on HSQC and HMQC schemes. Pairs of subspectra are obtained, containing either the high-field or the low-field component of the doublet representing the one-bond coupling constant. The subspectral editing procedure retains the full sensitivity of HSQC and HMQC spectra recorded without heteronuclear decoupling during data acquisition, with a spectral resolution similar to that of decoupled spectra.  相似文献

Summary Determination of the free and bound aminoacids during microsporogenesis of Datura metel showed that the principal amino-acids were proline, glutamic acid, aspartic acid, threonine-serine and alanine. Of these, only proline showed a consistent increment during pollen development. In contrast, aspartic acid and lysine decreased in the later stages of microsporogenesis. The amounts of other amino-acids did not show any consistent pattern. Four amino-acids, namely proline, glutamic acid and threonine-serine constituted nearly 85% of the free amino-acid pool in the developed anther (stage IV). Proline accumulation, relative to the total free amino-acid pool in mature anthers, was correlated with the water-content. To establish a protocol suited for the molecular characterization of root induction the influences of explant position, etiolation state and orientation as well as temperature and light intensity on root and callus formation were investigated. Depending on these factors stem discs of Malus incubated on indole-3-butyric acid containing medium and subsequently on hormone-free medium regenerated roots and callus of filamentous and smooth texture to a varying extent. Concentration and incubation duration of indole-3butyric acid strongly affected rooting and the production of smooth callus. Moreover smooth callus was profuse at the low light levels applied during rooting. Rooting efficiency decreased and filamentous callus increased between 19°C and 25°C. Explants close to the shoot apex displayed reduced rooting efficiency and profuse filamentous callus. Summary Psathyrostachys juncea (synonymous to Elymus junceus; 2n=2x=14, NN) has unique biotic and abiotic attributes that could contribute towards wheat improvement. The effectiveness of such an intergeneric hybridization program depends greatly on being able to establish diagnostic markers of the alien chromosomes. Isoelectric focusing (IEF) analyses of six enzyme systems have identified five biochemical markers — malate dehydrogenase (MDH), esterase (EST), shikimate dehydrogenase (SKDH), phosphoglucomutase (PGM), and -amylase (-AMY) — to be of positive diagnostic value; glucosephosphate isomerase (GPI) banding profiles were of no definite value in the background of Triticum aestivum cvs Chinese Spring and Seri-82, the potential recipients of Ps. juncea chromosomes. The Giemsa C-banding karyotype distinctively separates the Ps. Juncea chromosomes from each other and from those of T. aestivum with little banding site polymorphisms prevalent among its accessions analyzed, indicating the usefulness of C-bands as cytological markers.  相似文献

Summary To explain the high frequency of partial and intermediate back-mutants and the back-mutation of deletions, a model of mutation production based on unequal sister strand exchange in multi-repeat loci is proposed.The recovery of complementary products of mutation which the model predicts should be produced from single nuclei is cited as evidence for its plausability.Back-mutation of the mutant used (ahis-3 mutant,Gg59) is associated with a high frequency of simultaneous switch mutations of other characters in the same genome.It is suggested that unequal sister strand exchange may also contribute to the production of spontaneous mutations.The validity of citing ability to back-mutate as critical evidence for classifying a mutant as a point (single base pair substitution?) mutation is queried.With 2 Figures in the Text  相似文献

There is now abundant evidence that the structure of the mammalian visual cortex is not innately determined but can be altered by visual experiences during a certain period at an early age. A model based on the evidence is proposed that will explain some aspects of the developmental process in the visual cortex. The model self-organizingly forms a simple cell which responds to bars and edges of a certains orientation and retinal position. The total system of the model corresponds to a hyper column which contains a complete cycle of orientational selectiviy. The model has the following three original points. i) A hyper column is finally formed in the model. ii) Most of the cells do not have orientation selectivity in the initial state. iii) It is hypothesized that the orientation continuity of the hyper column is caused by similar orientations of successive input stimuli. The results of computer simulation show that the model has the expected performance.  相似文献

Résumé Nos méthodes expérimentales permettent l'isolement d'une larve de sexe déterminé par hôte de l'ectoparasite grégaire Dinarmus vagabundus et du solaitire, D. basalis. Des hôtes porteurs de 3 à 8 larves par hôte de D. vagabundus sont aussi isolés. Dans ces conditions la quantité de nourriture disponible est la même pour toutes les densités larvaires étudiées.Les larves élevées en solitaire des deux espèces assimilent une quantité de nourriture significativement supérieure à celle assimilée par les . Ceci conduit à des adultes de poids moyen supérieur à celui des . Le poids moyen des et des de D. vagabundus diminue significativement aux fortes densités larvaires. L'intensité de la liaison entre la quantité de nourriture assimilée et la biomasse produite s'affaiblit au fur et à mesure que la densité larvaire par hôte augmente.Les de D. vagabundus de poids moyen (0,42 mg) engendrent deux fois et demi plus de descendants que les lilliputiennes (0,20 mg) émergées d'hôtes à forte densité larvaire. Celles de D. basalis (0,65 g) sont moins prolifiques que les de D. vagabundus.  相似文献

The gene encoding 5 3 exonuclease domain-deleted Tfi DNA polymerase, named 5 3 Exo Tfi fragment, from Thermus filiformis was expressed in Escherichia coli under the control of the tac promoter on a high-copy plasmid, pJR. The expressed enzyme was purified 27-fold with a 19% yield and a specific activity of 2621 U mg–1 protein. The 5 3 exonuclease domain of Tfi DNA polymerase was removed without significant effect on enzyme activity and stability. PCR conditions for the 5 3 Exo Tfi fragment were more tolerant to the buffer composition as compared to the full-length enzyme.  相似文献

Résumé On a effectué une étude ultrastructurale sur les corps de Call et d'Exner. Ceux-ci se montrent constitués, sous leur aspect le plus typique, par une couronne de cellules de la granulosa disposées autour d'une cavité pleine d'un liquide semblable au liquor folliculi .Les cellules, de forme irrégulière, accolées dans leur partie proche du corps et écartées dans celle éloignée, déterminent des fentes dans lesquelles s'insinuent des groupes d'autres cellules voisines; la membrane cytoplasmique, parfois présente sous forme de microvilloités, parfois incisée, montre de fréquentes interruptions; le cytoplasme riche en ribosomes libres et adhérant aux parois du réticule endoplasmique (ergastoplasme), possède en outre un appareil de Golgi modérément développé et de nombreuses mitochondries; le noyau, très volumineux, présente à l'intérieur un nucléole grossièrement réticulé.Le corps de Call et d'Exner proprement dit est, au contraire, constitué par une cavité unitaire d'aspect à peu près sphérique d'environ 15–30 de diamètre, pleine d'un liquide dont les caractéristiques sont comparables à celles du liquor folliculi; dans cette cavité on voit un fin réseau à mailles irrégulières, tandis que sa surface montre une mince zone de condensation.L'aspect ultrastructural des cellules disposées autour des corps de Call et d'Exner est caractérisé par des cellules en élaboration, ce qui laisse penser qu'une partie de cette élaboration peut être versée à l'intérieur du corps, en contribuant ainsi à l'augmentation de son volume.En ce qui concerne, enfin, le réticule que l'on voit à l'intérieur du corps il est vraisemblable qu'il soit du à des phénomènes de condensation et consécutif à un état physicochimique particulier du liquide endocavitaire.
Summary The so-called Call-Exner bodies have been found to be formed, in their most typical shape, by a ring of granulosa cells enclosing a cavity filled with a fluid comparable to the liquor folliculi. The cells, irregular in shape, lying side by side in their portions closer to the body and far apart in their distal portions leave open spaces which are occupied by prolongations of adjacent cells. The cytoplasmic membrane, sometimes forming mierovilli, sometimes incisions, presents a discontinuous aspect; the cytoplasm is very rich in ribosomes which can be free or sticking to the walls of the endoplasmic reticulum (ergastoplasm). It has a fairly developed Golgi's apparatus and numerous mitochondria, sometimes open. The very voluminous nucleus shows internally a roughly reticulated nucleolus.The Call-Exner body, on the other hand, is a unitary cavity approximately spherical in shape, having about a 15–30 micron diameter filled with a fluid characteristically similar to the liquor folliculi, which shows internally a fine reticulation of irregular meshwork, while a thin condensation zone can bee seen peripherically.The ultrastructural aspect of the cells radiating from the Call-Exner bodies is typical of cells in formation. Therefore we cannot exclude the possibility that a portion of the substances elaborated by the cells may be thrown into the center of the body, thus contributing to its increase in volume. The mesh-like structure visible inside the body may be due to condensation phenomena following a particular physico-chemichal change in the endocavitary luid.
