A series of compounds containing dihydroquinazolinone moiety was designed and synthesized. Amine bridge part was changed in comparison with known anthranilic diamides insecticides. Their insecticidal activities against oriental armyworm (Mythimna separata) indicated that most of the compounds showed moderate to high activities at the tested concentrations. In particular, compounds 5a and 5k showed 80% larvicidal activities against oriental armyworm at the concentration of 5 mg/L. The present study also explored the possible effects of target compounds on the high voltage-gated calcium channel and the calcium channels in the endoplasmic reticulum in the central neurons isolated from the third instar larvae of Spodoptera exigua using whole-cell patch clamp and calcium imaging technique. The results showed that compound 5a activated the high voltage-gated calcium channel in the central neurons of S. exigua weakly. The peak currents only increased by 6% of the initial value at the end of the 10-min recording after treated with 0.22 μM 5a, while chlorantraniliprole has an opposite effect. The effects of 5a on the intracellular calcium ion concentration ([Ca2+]i) in neurons were well investigated. The experimental results indicated that these novel compounds have different mechanism compared with chlorantraniliprole.  相似文献

Dose– and time–mortality relationships of baculoviruses in pest insects are important for the determination of effective spraying regimes. A series of experiments with Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) and Spodoptera exigua MNPV (SeMNPV) against synchronized populations of S. exigua larvae in greenhouse chrysanthemum was conducted. Dose– and time–mortality relationships of different virus concentrations and S. exigua target stages were determined and the area foliage consumption was measured. Crop injury was greatly reduced when S. exigua were controlled as second or third instar larvae, whereas virus applications against fourth instar larvae could not prevent considerable crop injury, even at high concentrations. SeMNPV was approximately 10 times as infectious as AcMNPV when applied on greenhouse chrysanthemum. The relative virulence of AcMNPV and SeMNPV corresponded reasonably well with previously published laboratory bioassay data. SeMNPV killed second and fourth instar S. exigua larvae approximately 12 h faster than did AcMNPV in chrysanthemum, but no difference in speed of action was found for third instar larvae. The relative speed of action of AcMNPV and SeMNPV determined in chrysanthemum and in laboratory bioassays did not correspond for third instar S. exigua larvae; laboratory bioassay data can therefore not simply be extrapolated to the crop level.  相似文献

This study examined the physiological effects of joint and separate parasitism and infection by the endoparasitoid Microplitis pallidipes Szépligeti and the nucleopolyhedrovirus (NPV), respectively, on haemolymph 20‐hydroxyecdysone (20‐E) titre in Spodoptera exigua (Hübner) larvae. The results indicated that in parasitized larvae, virus‐infected larvae (5.7 × 103 and 5.7 × 105 OB/ml) and parasitized larvae infected with virus at 5.7 × 105 OB/ml, compared to healthy larvae, the 20‐E all declined during the first 3 days but began to increase from day 4 after treatment, while in jointly parasitized and infected larvae (5.7 × 103 OB/ml), the 20‐E declined during the first 4 days but began to increase on day 5 after treatment. Meanwhile, compared to parasitized larvae, the 20‐E declined during the first 4 days but significantly increased on day 5 in jointly parasitized and infected larvae (5.7 × 103 OB/ml), while significantly increased during the first 2 days but began to decrease from day 3 after treatment in jointly parasitized and infected larvae (5.7 × 105 OB/ml). Finally, in larvae that were both parasitized and virus infected (5.7 × 103 OB/ml), compared to just virus‐infected larvae (5.7 × 103 OB/ml), the 20‐E was lower on days 3 and 4 but higher on other days after treatment; in larvae that were both parasitized and virus infected (5.7 × 105 OB/ml), compared to just virus‐infected larvae (5.7 × 105 OB/ml), the 20‐E was significantly higher at the first 2 days but lower from day 3 after treatment. Our results revealed that 2nd instar larval M. pallidipes in host bodies may release 20‐E into the haemolymph of S. exigua larvae and that NPV infection may stimulate S. exigua to release more 20‐E during its third to fourth instar larval moulting. We found that this stimulatory effect was greater with higher virus concentrations.  相似文献

Abstract. The effects of Bacillus thuringiensis (Bt) Cry1C toxin on the metabolic rate of Cry1C resistant and susceptible Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae) are investigated using closed‐system respirometry. Mechanisms of resistance to the Bt toxin may be associated with an energetic cost that can be measured as an increase in metabolic rate compared with Bt‐susceptible insects. This hypothesis is tested using third‐ and fifth‐instar larvae and 1–7‐day‐old pupae. Metabolic rate is measured as the amount of O2 consumed and CO2 produced. V?O2 and V?CO2 (mL g?1 h?1) of third‐instar Cry1C resistant larvae reared continuously on a diet containing 320 µg Cry1C toxin per g diet (CryonT) are significantly greater than third‐instar Cry1C resistant larvae reared on toxin for 5 days and reared thereafter on untreated diet (Cry5dT), Cry1C resistant larvae reared on untreated diet (CryReg) and the susceptible parental strain (SeA) reared on untreated diet. There are no differences in V?O2 and V?CO2 (mL g?1 h?1) among treatment groups for fifth‐instar larvae. CryonT larvae and pupae weigh significantly less than larvae and pupae receiving other treatments. Smaller body mass may be an important biological cost to individuals exposed continuously to Bt toxin. One‐day‐old pupae of all treatment groups exhibit a high V?O2 (mean approximately 0.174 mL g?1 h?1) with CryonT having a significantly greater value than all other treatments; there are no differences among the other treatments. Pupal metabolic rates of all treatment groups decline to a minimum between days 2 and 4 then increase linearly between days 4 and 7 until adult emergence. These results demonstrate no difference in metabolic rates, and possibly fitness costs, between resistant (CryReg and Cry5dT) and susceptible (SeA) S. exigua except when larvae were reared continuously on toxin (CryonT).  相似文献

以Ba2+为载流子,采用全细胞膜片钳法,研究了在电极液中分别加入G蛋白稳定 激活剂GTPγS(GTP类似物)和抑制剂GDPβS(GDP类似物)对棉铃虫Helicoverpaarmigera 3龄幼虫神经细胞高电压敏感钙通道的调节作用。Ba2+电流记录时间为20 min。对照组 和实验组的Ba2+电流在记录的初期均出现电流的增加现象,随后电流衰减,即“rundown ”。对照组峰电流在第20 min时降为初始值的(72.09±12.80)%。电极内液中加入2 mmol/L GTPγS可缓解电流的衰减现象,在第20 min时,峰电流为初始值的(95.99±7.93)%,明显大 于对照组的峰电流(P<0.01),而且电流 电压(I-V)关系曲线向正电压方向移动。相反 ,电极内液加入2 mmol/L GDPβS则导致峰电流衰减更加严重,第20 min时,峰电流仅为初始水 平的(41.95±9.32)%,显著小于对照组(P<0.01),但未见电流 电压(I-V)关系曲 线的明显漂移。结果表明,棉铃虫神经细胞钙通道活动受G蛋白激活剂GTPγS和G蛋白抑制剂GDPβS的影响,提示G蛋白活动水平的改变调节钙通道的电流幅值和电压依赖性。  相似文献

Chitin synthase (CHS) is an important enzymatic component, which is required for chitin formation in the cuticles and cuticular linings of other tissues in insects. CHSs have been divided into two classes, classes A and B, based on their amino acid sequence similarities and functions. Class A CHS (CHS‐A) is specifically expressed in the epidermis and related ectodermal cells such as tracheal cells, while class B CHS (CHS‐B) is expressed in gut epithelial cells that produce peritrophic matrices. In this study, we cloned the CHS‐A gene from the beet armyworm, Spodoptera exigua (SeCHS‐A). The SeCHS‐A contains an open reading frame of 4,698 nucleotides, encoding a protein of 1,565 amino acids with a predicted molecular mass of approximately 177.8 kDa. The SeCHS‐A mRNA was expressed in all developmental stages and specifically in the epidermis and tracheae tissue by quantitative real‐time‐PCR analysis. Expression of SeCHS‐A gene was suppressed by feeding double‐stranded RNA (dsCHS‐A, 400 ng/larva) in the third instar larvae of S. exigua. Suppression of the SeCHS‐A gene expression significantly increased 35% of mortality on pupation of S. exigua. Also, the third instar larvae fed with dsCHS‐A significantly increased susceptibility to entomopathogenic fungi, Beauveria bassiana ANU1 at 3 days after treatment. These results suggest that the SeCHS‐A gene plays an important role in development of S. exigua and RNA interference may apply to effective pest control with B. bassiana.  相似文献

The present study relates to a methanol extract of the seed coat of Ginkgo biloba, and tested particularly on the third instar larvae of Spodoptera exigua. The extract was found to have an inhibitory effect on the growth of the larvae besides bringing a change in the nutrient reserves in the body of the insect. Topical application of five different doses of the methanol extract resulted in a mortal effect to third instar larvae of S. exigua that is very much dependent on the dose as well as duration of exposure. Lower doses revealed lower mortality after 24 h of application. At doses of 1.00, 2.00, 4.00, 8.00 and 16.00 ng/larva, mortalities were 9.25, 26.07, 50.32, 56.28 and 92.44%, respectively. The dose for 50% mortality (LD50) of methanol extracts by applied by a topical method with 1 µL of acetone solution was 1.92 ng/larva. Nutrient reserves like protein, glycogen and lipid are known to regulate pupation and adult emergence. These reserves have been found to be lower in treated larvae, indicating the insecticidal role of methanol extracts from G. biloba against third instar larvae of S. exigua.  相似文献

As a result of parasitism by Glyptapanteles liparidis in the first, second, third and fourth instar larvae of Acronicta rumicis, the mortality of each larval stage was found to be 46.67, 90, 71 and 16.67%, respectively. The mortality was highest when G. liparidis parasitized the second and third instar larvae. The difference in mortality between the parasitized group and the control group was 72.14% in the second instar larvae. With regards to the food consumption of the parasitized larvae, the first and second instar larvae consumed 6495.58 ± 646.52 mm2 (leaf surface) and 7951.12 ± 4167.36 mm2, respectively, while the third and fourth larvae consumed 13 826.77 ± 3396.66 mm2 and 18 599.85 mm2, respectively, showing that food consumption increased with instar stages of the host larvae. The clutch size of G. liparidis increased in relation to the instar stages of the host: it was 25.25 ± 7.89, 48.65 ± 53.75, 91.09 ± 44.52 and 114 individuals when they were fed with the first, second, third and the fourth instar larvae of the host, respectively.  相似文献

Spodoptera exigua nuclear polyhedrosis virus was produced in late fourth instar S. exigua larvae, reared on semi-artificial diet. A maximum amount of virus, 1–2 × 109 polyhedra/larva, was produced in individually-reared larvae after 7 days of incubation, with an inoculum of 7–5 × 104 polyhedra/cm2 diet surface. Virus yield was slightly reduced to 9 × 108 polyhedra/larva when production was carried out in groups of 400 and 600 larvae per container. Biological activity of virus harvested from living larvae and from dead cadavers was similar. Microbial contaminants, predominantly bacteria, in the virus product numbered 1–6% of the number of polyhedra. Tests for the presence of vertebrate-pathogenic bacteria in the virus product were all negative.  相似文献

Beet armyworm (Spodoptera exigua (Hübner)) (Lepidoptera: Noctuidae) is the most economically important sugar beet (Beta vulgaris) pest worldwide. In this study, a comparison was made between two different diets: one was based on Merkx diet (Holidic diet) and the other was based on sugar beet leaf (Oligidic diet). Results showed that the whole development time from larvae to adult between two diets (Merkx and leaf) was not significantly different. For example, developmental time from first instar larvae to adult in Merkx diet was 11.33?days, whilst developmental time of larvae to adult when larvae fed with sugar beet leaf was 10.33?days. However, analysis of variance showed that in some cases like development time of the first instar, third instar and fifth instar larvae and pupae was significantly different between two treatments (p?<?0.05). Larval weight showed differences when larvae fed on Merkx diet and sugar beet leaves. For example, significant differences were shown between first, third and fourth instar larvae weight when larvae fed on Merkx diet and sugar beet leaves (p?<?0.05). However, significant differences were not observed between weight of second and fifth instar as well as pupae weight when larvae fed on Merkx diet and sugar beet leaves (p?>?0.05).  相似文献

Beet armyworm (Spodoptera exigua Hb.) (Lepidoptera: Noctuidae) is the major pest of sugar beet (Beta vulgaris). Pesticide applications are the main method of the insect control. So, alternative method/s is/are needed to control this insect species. So, in the current study, the effect of Galanthus nivalis agglutinin (GNA) (snowdrop lectin) on beet armyworm α-amylase was studied. Measurement of the amylase activity of the larval midgut fed on artificial diet and sugar beet leaves showed that the enzyme activity was higher when the larvae fed on artificial diet. However, in both cases, the fourth instar larvae had the greatest amylase activity. Thus, fourth instar larvae were offered artificial diet containing 1 and 2% GNA. Both treatments of the lectin significantly reduced the α-amylase activity of the insect. For example, amylase activity of the fourth instar larvae in the control (fed only on artificial diet) was 2.62 Uml?1 whilst the activity of the enzyme in the two treatments including diet containing 2 and 1% lectin was 1.45 and 1.75 Uml?1, respectively. The achieved data showed that lectin, in addition to have toxic effect on the larval growth and development, affects the α-amylase activity of the insect gut.  相似文献

2019年草地贪夜蛾Spodoptera frugiperda入侵我国多个省份和地区,对我国农业生产造成重大影响。化学防治是应对入侵害虫的主要措施,因此,本研究测定了6种不同作用机制杀虫剂对草地贪夜蛾初孵和3龄幼虫的生物活性。结果表明,对初孵幼虫具有较高毒力作用的为氯虫苯甲酰胺、多杀菌素、茚虫威、阿维菌素和甲氧虫酰肼;对3龄幼虫具有较高毒力作用的为氯虫苯甲酰胺、多杀菌素和茚虫威;与初孵幼虫相比,阿维菌素、茚虫威、多杀菌素和甲氧虫酰肼的LC 50分别提高了78.06、1.70、11.73和23.09倍。本研究筛选出了对草地贪夜蛾初孵幼虫和3龄幼虫杀虫效果较好的药剂,证明了低龄幼虫对部分农药抵抗力较弱,而3龄后的幼虫抵抗力显著增加,为田间草地贪夜蛾的化学防治提供了科学的用药指导。  相似文献

In addition to damaging trees, the eastern tent caterpillar is implicated in early fetal loss and late‐term abortion in horses. In a field study, we evaluated the potential biological control of the caterpillar using eastern tent caterpillar nuclear polyhedrosis virus (ETNPV), a naturally occurring virus that is nearly species‐specific. Egg masses were hatched and second instar larvae were fed virus‐inoculated foliage to propagate the virus in vivo. Then, a viral pesticide was formulated at concentrations of 104, 106 and 108 polyhedral inclusion bodies per ml. The pesticide was applied to foliage on which second, third and fourth instar caterpillars were feeding. When the majority of surviving larvae reached the sixth instar, colonies were collected and the surviving caterpillars counted. Mean numbers of surviving caterpillars per treatment were compared via 95% bootstrap confidence intervals. The data indicate second instar caterpillars were highly susceptible to the virus, but only at the highest concentration tested. Third instar caterpillars were also somewhat susceptible to high virus concentrations, while fourth instar caterpillars were fairly resistant. Our data provide the strongest evidence to date that ETNPV can be propagated, harvested and refined for formulation as a biological control agent for eastern tent caterpillar. Its use on this insect may be merited in circumstances where landowners and managers need to protect trees and horses.  相似文献

Abstract Understanding predator–prey interactions has a pivotal role in biological control programs. This study evaluated the functional response of three larval instars of the green lacewing, Chrysoperla carnea (Stephens), preying upon eggs and first instar larvae of the cotton bollworm, Helicoverpa armigera Hübner. The first and second instar larvae of C. carnea exhibited type II functional responses against both prey stages. However, the third instar larvae of C. carnea showed a type II functional response to the first instar larvae of H. armigera, but a type III functional response to the eggs. For the first instar larvae of C. carnea, the attack rate on H. armigera eggs was significantly higher than that on the larvae, whereas the attack rate of the second instar C. carnea on H. armigera larvae was significantly higher than that on the eggs. For the third instar larvae of C. carnea, the attack rate on the larvae was 1.015 ± 0.278/h, and the attack coefficient on the eggs was 0.036 ± 0.005. The handling times of the third instar larvae on larvae and eggs were 0.087 ± 0.009 and 0.071 ± 0.001 h, respectively. The highest predation rate was found for the third instar larvae of C. carnea on H. armigera eggs. Results of this study revealed that the larvae of C. carnea, especially the third instar, had a good predation potential in controlling H. armigera eggs and larvae. However, for a comprehensive estimation of the bio‐control abilities of C. carnea toward H. armigera, further field‐based studies are needed.  相似文献

Dinoflagellate bioluminescence serves as a whole‐cell reporter of mechanical stress, which activates a signaling pathway that appears to involve the opening of voltage‐sensitive ion channels and release of calcium from intracellular stores. However, little else is known about the initial signaling events that facilitate the transduction of mechanical stimuli. In the present study using the red tide dinoflagellate Lingulodinium polyedrum (Stein) Dodge, two forms of dinoflagellate bioluminescence, mechanically stimulated and spontaneous flashes, were used as reporter systems to pharmacological treatments that targeted various predicted signaling events at the plasma membrane level of the signaling pathway. Pretreatment with 200 μM Gadolinium III (Gd3+), a nonspecific blocker of stretch‐activated and some voltage‐gated ion channels, resulted in strong inhibition of both forms of bioluminescence. Pretreatment with 50 μM nifedipine, an inhibitor of L‐type voltage‐gated Ca2+ channels that inhibits mechanically stimulated bioluminescence, did not inhibit spontaneous bioluminescence. Treatment with 1 mM benzyl alcohol, a membrane fluidizer, was very effective in stimulating bioluminescence. Benzyl alcohol‐stimulated bioluminescence was inhibited by Gd3+ but not by nifedipine, suggesting that its role is through stretch activation via a change in plasma membrane fluidity. These results are consistent with the presence of stretch‐activated and voltage‐gated ion channels in the bioluminescence mechanotransduction signaling pathway, with spontaneous flashing associated with a stretch‐activated component at the plasma membrane.  相似文献

Abstract 1 Mnesampela privata (Guenée) has a host list of 40 Eucalyptus and at least one Corymbia species. Larval survival and performance was studied on 19 species to investigate how certain leaf traits influence the suitability of different species. 2 After 7 days, survival on Eucalyptus aggregata and Eucalyptus camphora is greater than 70% even though the toughness of leaves is 0.15–0.19 mg/mm2. However, after the same time, survival on genotypes of Eucalyptus melliodora and Eucalyptus sideroxylon was less than 60%, or even 0%, even though the toughness of some leaves was as low as 0.11 mg/mm2. An unmeasured allelochemical, rather than toughness, may reduce survival on these species. 3 Dry weights of first‐instar larvae were negatively correlated with leaf toughness for 13 of the species studied. Species that produced the heaviest first‐instar larvae were not the same hosts that produced the heaviest second‐instar larvae. 4 Dry weights of female pupae were negatively correlated with total oil content for five of the species studied. 5 Larvae exhibit age‐related changes in feeding behaviour. Neonates skeletonize leaves (avoid leaf veins and oil glands) and post‐third‐instar larvae ingest whole leaf fragments (consume small leaf veins and oil glands). These findings suggest that neonates are sensitive to high leaf toughness and non‐oil plant secondary metabolites whereas older larvae are less sensitive to high leaf toughness and are likely to become larger adults on hosts with lower oil contents.  相似文献

Scolopendra subspinipes mutilans, also known as Chinese red‐headed centipede, is a venomous centipede from East Asia and Australasia. Venom from this animal has not been researched as thoroughly as venom from snakes, snails, scorpions, and spiders. In this study, we isolated and characterized SsmTx‐I, a novel neurotoxin from the venom of S. subspinipes mutilans. SsmTx‐I contains 36 residues with four cysteines forming two disulfide bonds. It had low sequence similarity (<10%) with other identified peptide toxins. By whole‐cell recording, SsmTx‐I significantly blocked voltage‐gated K+ channels in dorsal root ganglion neurons with an IC50 value of 200 nM, but it had no effect on voltage‐gated Na+ channels. Among the nine K+ channel subtypes expressed in human embryonic kidney 293 cells, SsmTx‐I selectively blocked the Kv2.1 current with an IC50 value of 41.7 nM, but it had little effect on currents mediated by other K+ channel subtypes. Blockage of Kv2.1 by SsmTx‐I was not associated with significant alteration of steady‐state activation, suggesting that SsmTx‐I might act as a simple inhibitor or channel blocker rather than a gating modifier. Our study reported a specific Kv2.1‐blocker from centipede venom and provided a basis for future investigations of SsmTx‐I, for example on structure–function relationships, mechanism of action, and pharmacological potential. 为了探讨氯虫苯甲酰胺对亚洲玉米螟Ostrinia furnacalis (Guenée)的抗性风险评估, 为科学使用氯虫苯甲酰胺防治亚洲玉米螟提供理论基础, 本研究在实验室条件下采用饲料混毒法测定了新型杀虫剂氯虫苯甲酰胺对亚洲玉米螟的毒力, 利用生命表技术研究了氯虫苯甲酰胺对亚洲玉米螟 3龄幼虫的亚致死效应。结果表明: 氯虫苯甲酰胺对亚洲玉米螟3龄幼虫的LC10, LC40 和LC50分别为0.038, 0.098 和0.123 mg a. i. /L。以氯虫苯甲酰胺LC10和LC40剂量分别处理亚州玉米螟3龄幼虫后, 幼虫和蛹的发育历期明显延长, 成虫寿命缩短, 产卵量也显著降低, 但对蛹重(雌: P=0.337, 雄: P=0.121)、卵孵化率(P=0.087)和性比(P=0.379)无显著影响。处理种群的净增值率(R0)、内禀增长率(rm)和周限增长率(λ)显著低于对照种群(P<0.0001)。结果提示, 氯虫苯甲酰胺的亚致死浓度可降低亚洲玉米螟的种群增长。  相似文献

Gambierol is a marine polyether ladder toxin derived from the dinoflagellate Gambierdiscus toxicus. To date, gambierol has been reported to act either as a partial agonist or as an antagonist of sodium channels or as a blocker of voltage‐dependent potassium channels. In this work, we examined the cellular effect of gambierol on cytosolic calcium concentration, membrane potential and sodium and potassium membrane currents in primary cultures of cerebellar granule cells. We found that at concentrations ranging from 0.1 to 30 µM, gambierol‐evoked [Ca2+]c oscillations that were dependent on the presence of extracellular calcium, irreversible and highly synchronous. Gambierol‐evoked [Ca2+]c oscillations were completely eliminated by the NMDA receptor antagonist APV and by riluzole and delayed by CNQX. In addition, the K+ channel blocker 4‐aminopyridine (4‐AP)‐evoked cytosolic calcium oscillations in this neuronal system that were blocked by APV and delayed in the presence of CNQX. Electrophysiological recordings indicated that gambierol caused membrane potential oscillations, decreased inward sodium current amplitude and decreased also outward IA and IK current amplitude. The results presented here point to a common mechanism of action for gambierol and 4‐AP and indicate that gambierol‐induced oscillations in cerebellar neurons are most likely secondary to a blocking action of the toxin on voltage‐dependent potassium channels and hyperpolarization of sodium current activation. New silica formulations offer expanded possibilities for use in horticultural crops. However, many crop pests are found on the leaf underside, and this is especially challenging when using silica because the substance must have direct contact with the insect to be effective. Silica a new and safety material was used against the second instar larvae of tomato leafminer, Tuta absoluta (Mayrick) alone and combined with other pesticides (chlorfenapyr, spinetoram and chlorantraniliprole). The results showed that when silica was used alone, the percents of mortalities were 51.7, 26.7 and 13.3%. The results showed also chlorantraniliprole was the most toxic compared with the other compounds. The per cents of mortalities were 90, 65 and 30% with the first, second and third concentrations, respectively. When silica was used in a combination with the tested pesticides, the mixture of silica and chlorfenapyr was the most toxic against the second instar larvae of T. absoluta, the percents of mortalities were 100, 76.7 and 60% with the first, second and third concentrations, respectively. The results recommended that silica was moderate toxic to the second instar larvae of T. absoluta when it used alone, so it can be used when this pest under the economic threshold or with integrated pest management programme. Silica has an activation action on the tested pesticides. Although silica was combined with one-fifth of the field rate of the tested pesticides, the toxicity of these pesticides was increased.  相似文献
