1. In polymorphic species, two or more discrete phenotypes co‐occur simultaneously. Sex‐limited polymorphism is a particular case of polymorphism, in which several discrete morphs coexist within one of the two sexes only. Several hypotheses were proposed to explain the existence and the maintenance of sex‐limited polymorphism in insects: (i) the morphs have similar fitness, such as similar survival and expected fecundity, and their frequencies vary randomly (i.e. the null hypothesis); (ii) harassment by males is reduced towards the less common female morph, in this case andromorph females (i.e. the male mimicry and learned mate recognition hypotheses); (iii) morphs differ in predation risk (i.e. the predation hypothesis); or (iv) morphs differ in thermoregulation ability (i.e. the thermoregulation hypothesis). 2. Field observations and experiments were employed to compare the relative support of these hypotheses using dimorphic females of the bog fritillary butterfly. Differences were detected between morphs in survival, fecundity, harassment by males, predation pressure and thermal properties, thereby rejecting the null hypothesis. 3. The lifestyle of both morphs is associated with different costs and benefits, with advantages in daily survival and precocious emergence for the gynomorph females, and advantages in fecundity, predation and male harassment for the andromorph females. Besides, as the bog fritillary butterfly is protandrous (i.e. males emerge before females), the longer development of andromorph females puts them at risk of emerging when all the males are dead. The results raise the question as to which mechanisms control the ontogenetic pathways driving the production of the two morphs (i.e. genetic polymorphism or phenotypic plasticity). 相似文献

Sex differences in ageing and lifespan are ubiquitous in nature. The "unguarded‐X” hypothesis (UXh) suggests they may be partly due to the expression of recessive mutations in the hemizygous sex chromosomes of the heterogametic sex, which could help explain sex‐specific ageing in a broad array of taxa. A prediction central to the UX hypothesis is that inbreeding will decrease the lifespan of the homogametic sex more than the heterogametic sex, because only in the former does inbreeding increase the expression of recessive deleterious mutations. In this study, we test this prediction by examining the effects of inbreeding on the lifespan and fitness of male and female Drosophila melanogaster across different social environments. We found that, across social environments, inbreeding resulted in a greater reduction of female than male lifespan, and that inbreeding effects on fitness did not seem to counterbalance sex‐specific effects on lifespan, suggesting the former are maladaptative. Inter‐ and intra‐sexual correlation analyses also allowed us to identify evidence of an underlying joint genetic architecture for inbreeding effects on lifespan. We discuss these results in light of the UXh and other alternative explanations, and suggest that more attention should be paid to the possibility that the “unguarded‐X” may play an important role in the evolution of sex‐specific lifespan. 相似文献

Immunoglobulin G (IgG) plays an important role in clinical diagnosis and therapeutics. Meanwhile, the consensus binding site (CBS) on the Fc domain of IgG is responsible for ligand recognition, especially for Fc‐specific ligands. In this study, molecular simulation methods were used to investigate molecular interactions between the CBS of the Fc domain and seven natural Fc‐specific ligands. The analysis on the binding energy of the Fc–ligand complex indicated that hydrophobic interactions provide the main driving force for the Fc–ligand binding processes. The hot spots on the ligands and Fc were identified with the computational alanine scanning approach. It was found that the residues of tryptophan and tyrosine on the ligands have significant contributions for the Fc–ligand binding, while Met252, Ile253, Asn434, His435, and Tyr436 are the key residues of Fc. Moreover, two binding modes based on tryptophan or tyrosine were summarized and constructed according to the pairwise interaction analysis. Classic theories of ageing evolution predict that increased extrinsic mortality due to an environmental hazard selects for increased early reproduction, rapid ageing and short intrinsic lifespan. Conversely, emerging theory maintains that when ageing increases susceptibility to an environmental hazard, increased mortality due to this hazard can select against ageing in physiological condition and prolong intrinsic lifespan. However, evolution of slow ageing under high‐condition‐dependent mortality is expected to result from reallocation of resources to different traits and such reallocation may be hampered by sex‐specific trade‐offs. Because same life‐history trait values often have different fitness consequences in males and females, sexually antagonistic selection can preserve genetic variance for lifespan and ageing. We previously showed that increased condition‐dependent mortality caused by heat shock leads to evolution of long‐life, decelerated late‐life mortality in both sexes and increased female fecundity in the nematode, Caenorhabditis remanei. Here, we used these cryopreserved lines to show that males evolving under heat shock suffered from reduced early‐life and net reproduction, while mortality rate had no effect. Our results suggest that heat‐shock resistance and associated long‐life trade‐off with male, but not female, reproduction and therefore sexually antagonistic selection contributes to maintenance of genetic variation for lifespan and fitness in this population. 相似文献

Increasing atmospheric carbon dioxide (ab. CO2) and fertilizer‐nitrogen (ab. N) applications may have marked direct effects on the plant growth of agricultural crops, and in turn affect the higher trophic level of insect herbivores. In this study, the effects of elevated CO2 (i.e., 650 µl/L vs. ambient 400 µl/L) and fertilizer‐N (0, 50, 100, 200 kg/ha) on the population abundances and the inter‐specific competition among three co‐occurring species of wheat aphids, Sitobion avenae, Rhopalosiphum padi and Schizaphis graminum, were studied. The grain weight per ear and the 1,000‐grain weight were generally increased when grown under elevated CO2 and showed a significant effect at the 100 kg/ha (grain weight per ear) and 0, 50 and 100 kg/ha (1,000‐grain weight) N. These two yield indexes increased with increasing fertilizer‐N levels within reasonable limits and reached a maximum at 100 kg/ha. Elevated CO2 combined with fertilizer‐N levels formed complex indirect effects on the three wheat aphids through the wheat crops they fed on. Elevated CO2 significantly decreased the niche overlap index (ab. NOI) between S. avenae and R. padi under 0 and 100 kg/ha and that between R. padi and S. graminum under 0 kg/ha, while significantly increased the three NOIs under 50 kg/ha and that between R. padi and S. graminum under 100 and 200 kg/ha. S. avenae and R. padi had the larger population and stronger competition in low‐N condition (0 and 50 kg/ha), which was harmful to wheat yield and quality when combined with its own poor nutrition. Overall, the 100 kg/ha N level was the best option based on the aphid population, competition and wheat yields. Therefore, the balanced relationship formed among fertilizers, plants and insects under 100 kg/ha N was vital for the interactive ecosystem. 相似文献

In this study, Streptococcus gordonii‐specific quantitative real‐time polymerase chain reaction (qPCR) primers, RTSgo‐F2/RTSgo‐R2, were developed based on the nucleotide sequences of RNA polymerase β‐subunit gene (rpoB). The specificity of the RTSgo‐F2/RTSgo‐R2 primers was assessed by conventional PCR on 99 strains comprising 63 oral bacterial species, including the type strain and eight clinical isolates of S. gordonii. PCR products were amplified from the genomic DNAs of only S. gordonii strains. The qPCR primers were able to detect as little as 40 fg of S. gordonii genomic DNA at a cycle threshold value of 33. These findings suggest that these qPCR primers detect S. gordonii with high specificity and sensitivity. 相似文献

Yellow perch, Perca flavescens, is an ecologically and economically important species native to a large portion of the northern United States and southern Canada and is also a promising candidate species for aquaculture. However, no yellow perch reference genome has been available to facilitate improvements in both fisheries and aquaculture management practices. By combining Oxford Nanopore Technologies long‐reads, 10X Genomics Illumina short linked reads and a chromosome contact map produced with Hi‐C, we generated a high‐continuity chromosome‐scale yellow perch genome assembly of 877.4 Mb. It contains, in agreement with the known diploid chromosome yellow perch count, 24 chromosome‐size scaffolds covering 98.8% of the complete assembly (N50 = 37.4 Mb, L50 = 11). We also provide a first characterization of the yellow perch sex determination locus that contains a male‐specific duplicate of the anti‐Mullerian hormone type II receptor gene (amhr2by) inserted at the proximal end of the Y chromosome (chromosome 9). Using this sex‐specific information, we developed a simple PCR genotyping assay which accurately differentiates XY genetic males (amhr2by+) from XX genetic females (amhr2by?). Our high‐quality genome assembly is an important genomic resource for future studies on yellow perch ecology, toxicology, fisheries and aquaculture research. In addition, characterization of the amhr2by gene as a candidate sex‐determining gene in yellow perch provides a new example of the recurrent implication of the transforming growth factor beta pathway in fish sex determination, and highlights gene duplication as an important genomic mechanism for the emergence of new master sex determination genes. 相似文献

Sporadic Creutzfeldt–Jakob disease (sCJD) is characterized by wide clinical and pathological variability, which is mainly influenced by the conformation of the misfolded prion protein, and by the methionine and valine polymorphism at codon 129 of the prion protein gene. This heterogeneity likely implies differences in the molecular cascade that leads to the development of certain disease phenotypes. In this study, we investigated the proteome of the frontal cortex of patients with the two most common sCJD subtypes (MM1 and VV2) using 2D‐DIGE and MS. Analysis of 2D maps revealed that 46 proteins are differentially expressed in the sCJD. Common differential expression was detected for seven proteins, four showed opposite direction of differential expression, and the remaining ones displayed subtype‐specific alteration. The highest number of differentially expressed proteins was associated with signal transduction and neuronal activity. Moreover, functional groups of proteins involved in cell cycle and death, as well as in structure and motility included subtype‐specific expressed proteins exclusively. The expression of Rab GDP dissociation inhibitor alpha, which regulates Rab3a‐mediated neurotransmitter release, was affected in both sCJD subtypes that were analyzed. Therefore, we also investigated as to whether Rab3a recycling is altered. Indeed, we found an accumulation of the membrane‐associated form, thus the active one, which suggests that dysfunction of the Rab3a‐mediated exocytosis might be implicated in sCJD pathology. 相似文献

An experimental methodology that facilitates functional analysis of numerous protein–protein interactions, which have been found in genome‐wide interactome researches, has long been awaited. We propose herein an antagonistic inhibition‐based approach. The antagonizing polypeptide is generated in the course of interaction domain mapping based on yeast 2‐hybrid (Y2H) screening coupled with in vitro convergence of the Y2H‐selected fragments, which is performed in a formatted procedure. Using the coupled methodology, we first performed a high‐resolution mapping of an interdomain interaction network within budding yeast's Dam1 complex. Dam1 complex is a kinetochore protein complex composed of 10 essential subunits including Spc34p and Spc19p. The high‐resolution mapping revealed the overall network structure within the complex for the first time: Dam1 components form into two separated subnetworks on N‐terminal scaffolding domains of Spc34p and Spc19p, and the coiled‐coil interaction in their C‐terminal domains connects the subnetworks. Secondly, we show that the domain fragments converged in the high‐resolution mapping acted as potent inhibitors for the endogenous interactions when episomally overexpressed. The in vivo Dam1 interaction targeting with the fragments conferred a similar phenotype on the host cells; a critical and irreversible damage, which was accompanied with disturbed budding and chromosome mis‐segregation as a result of disorganized spindle. These phenotypes were strongly related to the cellular function of the Dam1 complex. The results and approach we demonstrated herein not only shed light on the Dam1 molecular architecture but also pave the road to reverse‐interactome analysis and discoveries of novel drugs that target disease‐related protein–protein interactions. © 2010 American Institute of Chemical Engineers Biotechnol.
