Conducting experiments on very soft biological tissues can be difficult. Traditionally, unconfined compression and shear have been used. Here, an improved method of compression testing, lubricated squeezing flow is described. This gives a uniform compression along the squeezing axis and almost uniform equi-biaxial elongation at right angles to the squeezing axis, with minimal shear deformation due to the constant lubrication of the sample surfaces during testing. Sample results for porcine liver obtained using this method are described here.  相似文献

We present a new constitutive formulation that combines certain desirable features of two previously used approaches (phenomenological and microstructural). Specifically, we assume that certain soft tissues can be idealized as composed of various families of noninteracting fibers and a homogeneous matrix. Both the fibers and the matrix are assumed to follow the gross deformation. Within the usual framework of pseudoelasticity, incompressibility, homogeneity, and the continuum hypothesis, a pseudostrain-energy function (W) is proposed wherein W is expressed in terms of matrix and fibrous contributions. Unlike phenomenological approaches where a W is usually chosen in an ad hoc manner, the present approach can be used to postulate reasonable forms of W based on limited structural information and multiaxial stress-strain data. Illustrative applications of the theory are discussed for visceral pleura and myocardium. Concise structurally motivated constitutive relations result, wherein load-dependent anisotropy, nonlinear material behavior, finite deformations, and incompressibility are accounted for.  相似文献

When lung tissue is subjected to finite deformations, phenomena appear that can only be described using nonlinear models. This paper considers the lung as a material composed of two elements, a continuous phase that acts uninterruptedly and a second phase composed of fiber elements that are recruited progressively into the mechanical process. Each individual fiber participates in the mechanical response of the set only when the deformation is above a certain value. A nine-parameter model was designed adopting standard viscoelastic elements both for the matrix and for each of the fibers. The mechanical behavior of the lung can be reproduced by a fitting process with standard numerical procedures in both dynamic-mechanical measurements and stress relaxation processes. Mechanical stress relaxation tests and dynamic-mechanical measurements have been carried out on subpleural parenchymal strips from rat lung. The model permits the reproduction of lung behavior in both types of measurements. The results show a recruitment ratio that decreases with deformation and the nonparticipation of the parallel matrix fraction in the lung's mechanical response so that a uniaxial transmission of force in the lung occurs via the recruited elements and the matrix series.  相似文献

A theoretical model is developed to predict the elastic properties of very soft tissues such as glands, tumors and brain. Tissues are represented as regular arrays of polyhedral (cubic or tetrakaidecahedral) cells, surrounded by extracellular spaces of uniform width. Cells are assumed to be incompressible, with very low resistance to shear deformation. Tissue shear rigidity is assumed to result mainly from the extracellular matrix, which is treated as a compressible elastic mesh of interconnected fibers. Small-strain elastic properties of tissue are predicted using a finite-element method and an analytical method. The model can be used to estimate the compressibility of a very soft tissue based on its Young's modulus and extracellular volume fraction.  相似文献

A model of the mechanical behaviour of soft connective tissue has been developed by considering the role of the collagen and glycosaminoglycan (GAG) components within the tissue in order to examine the mechanism by which a variation in the GAG components may exert a control over the mechanical properties of the tissue. It is proposed that the strain energy stored within the collagen fibrils of the loaded tissue can be transferred into a potential field created by the charged GAG components and their electrostatic interaction with the collagen fibrils. A fundamental mechanical unit is described to simulate this energy transfer and a combination of such units is used to represent the tissue. The computer implementation of the proposed tissue model shows it to reproduce many features which have been recognised in the rate dependent mechanical behaviour of soft tissues. These include the characteristic non-linearity of the force-deformation behaviour and the approximate invariance of the stress relaxation behaviour with deformation. The model is also consistent with earlier constitutive representations of tissue behaviour.  相似文献

This paper describes a new apparatus and method for measuring swelling dependent electrical conductivity of charged hydrated soft tissues. The apparatus was calibrated using a conductivity standard. Swelling dependent specific conductivity of porcine annulus fibrosis (AF) samples was determined. The conductivity values for porcine AF were similar to those for human and bovine articular cartilage found in the literature. Results revealed a significant linear correlation between specific conductivity and water content for porcine AF tissues tested in phosphate buffered saline (PBS).  相似文献

Evaluation and simulation of the multiaxial mechanical behavior of native and engineered soft tissues is becoming more prevalent. In spite of this growing use, testing methods have not been standardized and methodologies vary widely. The strong influence of boundary conditions were recently underscored by Waldman et al. [2002, J. Materials Science: Materials in Medicine 13, pp. 933-938] wherein substantially different experimental results were obtained using different sample gripping methods on the same specimens. As it is not possible to experimentally evaluate the effects of different biaxial test boundary conditions on specimen internal stress distributions, we conducted numerical simulations to explore these effects. A nonlinear Fung-elastic constitutive model (Sun et al., 2003, JBME 125, pp. 372-380, which fully incorporated the effects of in-plane shear, was used to simulate soft tissue mechanical behavior. Effects of boundary conditions, including varying the number of suture attachments, different gripping methods, specimen shapes, and material axes orientations were examined. Results demonstrated strong boundary effects with the clamped methods, while suture attachment methods demonstrated minimal boundary effects. Suture-based methods appeared to be best suited for biaxial mechanical tests of biological materials. Moreover, the simulations demonstrated that Saint-Venant's effects depended significantly on the material axes orientation. While not exhaustive, these comprehensive simulations provide experimentalists with additional insight into the stress-strain fields associated with different biaxial testing boundary conditions, and may be used as a rational basis for the design of biaxial testing experiments.  相似文献

Indentation using the atomic force microscope (AFM) has potential to measure detailed micromechanical properties of soft biological samples. However, interpretation of the results is complicated by the tapered shape of the AFM probe tip, and its small size relative to the depth of indentation. Finite element models (FEMs) were used to examine effects of indentation depth, tip geometry, and material nonlinearity and heterogeneity on the finite indentation response. Widely applied infinitesimal strain models agreed with FEM results for linear elastic materials, but yielded substantial errors in the estimated properties for nonlinear elastic materials. By accounting for the indenter geometry to compute an apparent elastic modulus as a function of indentation depth, nonlinearity and heterogeneity of material properties may be identified. Furthermore, combined finite indentation and biaxial stretch may reveal the specific functional form of the constitutive law--a requirement for quantitative estimates of material constants to be extracted from AFM indentation data.  相似文献

In this paper the mechanical response of soft collagenous tissues with regular fiber arrangement (RSCTs) is described by means of a nanoscale model and a two-step micro–macro homogenization technique. The non-linear collagen constitutive behavior is modeled at the nanoscale by a novel approach accounting for entropic mechanisms as well as stretching effects occurring in collagen molecules. Crimped fibers are reduced to equivalent straight ones at the microscale and the constitutive response of RSCTs at the macroscale is formulated by homogenizing a fiber reinforced material. This approach has been applied to different RSCTs (tendon, periodontal ligament and aortic media), resulting effective and accurate as proved by the excellent agreement with available experimental data. The model is based on few parameters, directly related to histological and morphological evidences and whose sensitivity has been widely investigated. Applications to simulation of some physiopathological mechanisms are also proposed, providing confirmation of clinical evidences and quantitative indications helpful for clinical practice.  相似文献

The dynamic response of soft human tissues in hydrostatic compression and simple shear is studied using the Kolsky bar technique. We have made modifications to the technique that allow loading of a soft tissue specimen in hydrostatic compression or simple shear. The dynamic response of human tissues (from stomach, heart, liver, and lung of cadavers) is obtained, and analyzed to provide measures of dynamic bulk modulus and shear response for each tissue type. The dynamic bulk response of these tissues is easily described by a linear fit for the bulk modulus in this pressure range, whereas the dynamic shearing response of these tissues is strongly non-linear, showing a near exponential growth of the shear stress.  相似文献

SDS-polyacrylamide gel electrophoresis is a useful technique in bacterial differentiation and identification. A rapid, semi-automated SDS-PAGE system (Phast System) was assessed for identification of formate-fumarate-requiring, asaccharolytic, Gram-negative oral anaerobes. The system permitted loading, separation and staining of gels within 2 h. Percentage similarities between strains were determined using correlation coefficients and cluster analysis. The protein profiles were sufficiently reproducible provided that distorted profiles were disregarded. Strains were successfully separated into their species, with the exception of Bacteroides ureolyticus NCTC 10939, which appeared to be distinct from other strains of that species. Twenty-nine unidentified formate-fumarate-requiring, sub-gingival plaque strains were suitably clustered with the standard strains as verified by a series of physiological and biochemical tests. and accepted 25 July 1989  相似文献

As dura mater has an anisotropic fibrous structure and exists under wet and dynamic stretching conditions in the brain, its mechanical properties have not yet been properly investigated. Here we developed a fluid-assisted mechanical system integrated with a photonic sensor and a pressure sensor in order to measure the elasticity of the dura mater. Porcine dura mater sample was loaded as a stretched diaphragm into a liquid chamber to mimic the in vivo condition. Increasing the flow rate of saline solution into the chamber swelled and deformed the dura mater. The micron-scale deflection of the dura mater was optically detected by the photonic sensor. Fluid pressure and deflection values were then used to calculate the elastic modulus. The average elastic modulus of the porcine dura mater was 31.14 MPa. We further measured the elasticity of a well-known material to further validate the system. We expect that this optofluidic system developed in this study will be useful to measure the elasticity of a variety of thin biological tissues.  相似文献
