模拟生态系统中噬藻体裂解蓝藻宿主的生态学效应   总被引:1,自引:0,他引:1  
实验采用模拟生态系统,研究了噬藻体裂解蓝藻宿主后营养物质循环变化的过程,以及细菌和漂浮植物对这个过程的影响。结果表明噬藻体裂解宿主所释放的营养元素在细菌的作用下迅速进入新的循环并形成新的水华,而加入浮萍则可以有效吸收水体营养,防止新的水华发生。因此推测单纯地利用包括噬藻体在内的微生物裂解藻类的方法,并非治理水华的有效方法,但如果能够结合漂浮植物等其它治理手段,则有可能同时实现水华的控制和水质的改善。  相似文献

The distribution of the IncFI basic replicons among IncFIV plasmids was assessed by DNA hybridization. In addition these and 20 other plasmids from 16 incompatibility groups were screened for the presence of IncIV, an incompatibility determinant recently found on the IncFIV plasmid R124. The IncIV determinant was found commonly but not universally among the IncFIV plasmids. It was also detected on the IncFI reference plasmid R386 and plasmids from IncB, IncI alpha and IncI gamma. The frequency and distribution of IncFI replicons among the IncFIV plasmids is similar to that observed in other F groups. The similarity of the IncFIV plasmids to plasmids of the other IncF groups and the failure to find replicons unique to IncFIV plasmids indicates that their division into a separate incompatibility group is not justified.  相似文献

A phenotypic matrix was established for 512 marine Vibrio species previously surveyed for plasmid incidence and characteristics. The organisms were subjected to a variety of tests to determined phenetic groups and plasmid functions. Both physiologic and plasmid function tests were used to cluster the strains by numerical taxonomy. The resulting phenotypic clusters showed an uneven distribution of plasmids. Some clusters contained isolates with no plasmids and other clusters contained isolates with a high incidence of plasmids. Traits correlating with plasmid occurrence in clusters included bioluminescence and antibiotic and heavy metal resistance. These characteristics were assumed to be either plasmid-mediated or plasmid-associated traits. Cluster analysis was found to be a successful method for relating the presence of plasmids with specific traits.  相似文献

Ultrastructural studies were made on seven genera of filamentous cyanobacteria from Sections III and IV (in the Rippka classification): Oscillatoria limosa Ag. ex Gomont, Spirulina subsalsa Turp. ex Gomont, Crinalium epipsammum Crow, Nostoc commune Vaucher ex Born. et Flah., Anabaena variabilis Kützinger ex Born. et Flah., Arthrospira maxima Stizenb. ex Gomont, Arthrospira platensis Stizenb. ex Gomont, and Cyanospira rippkae Florenzano. Perforation types and their distribution in the peptidoglycan layer observed in Crinalium epipsammum Crow were similar to those in Oscillatoria limosa and two Arthrospira strains. In Cyanospira and Anabaena no junctional perforations in the peptidoglycan wall layer were observed; however, intracellular perforations were documented. Except for Spirulina , the perforations were organized in rows (up to 10) placed on both sides of the cross wall. In the Spirulina cross sections, perforations were found only in the thickened, inner part of the cell wall. The perforation patterns in Spirulina and Arthrospira strains were clearly different and justify the separation into different genera.  相似文献

Using the cloning of part of the T-DNA of pTiC58 from Agrobacterium tumefaciens as an example, techniques are described which enable recombinant plasmids to be mapped and used as hybridization probes. In all cases the starting material is a colony of cells grown on an agar plate which is then subjected to lysis by lysozyme and Triton X-100 in volumes of the order of 300 microliters thus eliminating the need for handling and centrifuging liquid cultures under restrictive containment conditions.  相似文献

Summary Bacteria isolated from groundwater aquifer core materials of pristine aquifers at Lula and Pickett, OK, and from a site with a history of aromatic hydrocarbon contamination and natural renovation located at Conroe, TX, were screened for the presence of plasmid DNA by alkaline or enzyme lysis and agarose gel techniques. Some of the isolates were also subjected to taxonomic tests in addition to screening for resistance to antibiotics, tolerance to heavy metal salts, and bacteriocin production. There was no significant difference in the distribution of the traits usually associated with plasmid occurrence in isolates from the three sites. These traits, which occurred at low frequencies, were not restricted to plasmid-bearing strains of the communities. Plasmids were found in isolates from all three sites, but on the average there was a significantly higher percentage of isolates containing plasmids in the samples from Conroe (19.4%) than from either Lula (1.8%) or Pickett (7.7%). The sizes of the plasmids found ranged between 3.5 and 202 kilobases but, for the Conroe samples, many more isolates (67%) contained smaller plasmids (<10 kb) rather than larger ones. No plasmids were found in bacteria recovered from naturally renovated aquifer material at the Conroe site.  相似文献

Inteins are genetic elements found inside the coding regions of different host proteins and are translated in frame with them. The intein-encoded protein region is removed by an autocatalytic protein-splicing reaction that ligates the host protein flanks with a peptide bond. This reaction can also occur in trans with the intein and host protein split in two. After translation of the two genes, the two intein parts ligate their flanking protein parts to each other, producing the mature protein. Naturally split inteins are only known in the DNA polymerase III alpha subunit (polC or dnaE gene) of a few cyanobacteria. Analysing the phylogenetic distribution and probable genetic propagation mode of these split inteins, we conclude that they are genetically fixed in several large cyanobacterial lineages. To test our hypothesis, we sequenced parts of the dnaE genes from five diverse cyanobacteria and found all species to have the same type of split intein. Our results suggest the occurrence of a genetic rearrangement in the ancestor of a large division of cyanobacteria. This event fixed the dnaE gene in a unique two-genes one-protein configuration in the progenitor of many cyanobacteria. Our hypothesis, findings and the cloning procedure that we established allow the identification and acquisition of many naturally split inteins. Having a large and diverse repertoire of these unique inteins will enable studies of their distinct activity and enhance their use in biotechnology.  相似文献

RSF1010-derived plasmids are most efficiently transferred by conjugation to the unicellular cyanobacteriaSynechocystis strains sp. PCC6803 and PCC6714 andSynechococcus strains sp. The addition of extra antibiotic resistance determinants to plasmids of a series previously shown to be incompatible in an atypical fashion with group I and group P plasmids, has enabled the existence of a third and major incompatibility function determined by these plasmids to be demonstrated. The in vitro construction of a plasmid consisting of the replication region of one of the plasmids, linked to the genes of the galactose operon, facilitated the identification of this third incompatibility function as similar to IncB plasmids.  相似文献

Abstract We describe the subcloning of plasmids present in Vibrio cholerae 01 strain V58: the P factor, the large cryptic plasmid (lcp) and small cryptic plasmid (scp). Strains harbouring fragments of the P sex factor and the lcp were examined for plasmid encoded proteins by Coomassie blue staining and analysis in Escherichia coli K-12 minicells.
The distribution of these three plasmids in a variety of Vibrio species has been examined using some of the cloned fragments as DNA probes. Most recent clinical isolates of V. cholerae were found to contain the scp. None of the strains contained the lcp. The P factor was only detected in one clinical isolate in addition to the scp. While plasmids appear to be generally uncommon among V. cholerae , and do not appear to differentiate biovars, the presence of plasmids may be a useful epidemiological adjunct.  相似文献

The variation in transposition history of different Ty1-copia group LTR retrotransposons in the species lineages of the Pisum genus has been investigated. A heterogeneous population of Ty1-copia elements was isolated by degenerate PCR and two of these (Tps12 and Tps19) were selected on the basis of their copy number and sequence conservation between closely related species for further in-depth study of their transpositional history in Pisum species. The insertional polymorphism of these elements and the previously characterised PDR1 element was studied by sequence-specific amplification polymorphism (SSAP). Each of these elements reveals a unique transpositional history within 55 diverse Pisum accessions. Phylogenetic trees based on the SSAP data show that SSAP markers for individual elements are able to resolve different species lineages within the Pisum genus. Finally, the SSAP data from all of these retrotransposon markers were combined to reveal a detailed picture of the intra and inter-species relationships within Pisum. The nonheterocystous filamentous CyanobacteriumPlectonema boryanum strain UTEX 594 contains at least two plasmids. A small 145 kb plasmid was cloned in pBR322. It has no homology with the bigger resident plasmid or with chromosomal DNA. A small fraction of the plasmid is present in the form of multimers or concatemers. Copy number and hybridization patterns of the plasmid were similar under dinitrogen-fixing and non-fixing conditions. Restriction site mapping of the plasmid was done to enable its use in the development of cyanobacterial cloning vectors. It is among the smallest natural plasmids reported from bacteria.  相似文献

Distribution of virulence plasmids within Salmonellae   总被引:10,自引:0,他引:10  
The virulence region of the Salmonella dublin 50 MDa plasmid shared homology with 678 of 1021 salmonellae tested in colony hybridization experiments. The majority of S. dublin, S. typhimurium and S. enteritidis isolates tested hybridized with the region whereas, with the exception of S. hessarek, S. pullorum and S. gallinarum, other serotypes did not. Homologous virulence regions were plasmid encoded. In S. typhimurium a common 60 MDa plasmid was present in all phage types tested but not in DT4, DT37 and DT170. Smaller plasmids showing partial homology were found in DT12, DT18, DT193 and DT204C. In S. enteritidis a distinct plasmid profile for each of eight phage types was observed. Hybridizing plasmids were found in DT3, DT4, DT8, DT9 and DT11 whereas DT7, which was plasmid free, and DT10 and DT14, which harboured plasmids, did not hybridize. The extent of homology shared between S. dublin, S. typhimurium and S. enteritidis virulence plasmids was about 10 MDa and appeared conserved. Virulence plasmids from S. typhimurium and S. enteritidis did not show homology with a region of the S. dublin 50 MDa plasmid which was not associated with virulence functions whereas plasmids of about 24 MDa and 38 MDa in some S. typhimurium phage types did. The association of conserved virulence regions upon differing plasmids within salmonellae is discussed with reference to possible mechanisms of distribution and evolution of virulence genes.  相似文献

The members of the Bacillus cereus group, Bacillus anthracis, Bacillus thuringiensis, and B. cereus senso stricto, are largely defined by their content of large plasmids, which encode major virulence factors. Here we offer an easy, fast, and reliable protocol for the isolation and detection of large plasmids up to the size of at least 350kb. Furthermore, using this method, we report that Bacillus mycoides contain large plasmids.  相似文献

Polymerase chain reaction studies showed that naphthalene-utilizing bacteria isolated from various localities of Belarus most often contained Nah plasmids of the P-9 incompatibility group and plasmids of indefinite systematics. The conventional incompatibility test and restriction enzyme analysis revealed three new IncP-9 subgroups: ζ, η, and IncP-9-like. In addition to the known nucleotide sequences of nahG and nahAc, two novel nahG variants were revealed by a restriction enzyme analysis of amplification products. An amplified rDNA restriction enzyme analysis (ARDRA) demonstrated that the native hosts of IncP-9 Nah plasmids were fluorescent bacteria of the genus Pseudomonas (P. fluorescens, P. putida, P. aeruginosa, and Pseudomonas sp.) and nonfluorescent bacteria of indefinite systematics.  相似文献

Summary Linear plasmids are genetic elements commonly found in yeast, filamentous fungi, and higher plants. In contrast to all other plasmids they possess terminal inverted repeats and terminal bound proteins and encode their own DNA and RNA polymerases. Here we present alignments of conserved amino acid sequences of both the DNA and RNA polymerases encoded by those linear plasmids for which DNA sequence data are available. Additionally these sequences are compared to a number of polymerases encoded by related viral and cellular entities. Phylogenetic trees have been established for both types of polymerases. Summary In a survey of 35 strains ofStreptococcus thermophilus, 13 strains were found to harbor plasmid DNA. Most of these strains contained plasmid species varying in size from 2.2 to 7.15 kilobases. Only three strains had more than one plasmid species. Each of the nine distinct types of plasmid DNAs identified had two or more unique recognition sites for restriction endonucleases. The characteristics of the indigenous cryptic plasmids ofS. thermophilus may allow their development as cloning vectors useful in the genetic engineering of this species and other streptococci that are important in food production  相似文献
