Studying 347 sera of 49 patients suffering from Pseudomonas aeruginosa infection the mean titre value (MTV) calculated from the exponent of the binary logarithm of natural antibody (NA) titres, was found suitable to characterize the humoral immune status. As long as the organism is in equilibrium with the infection, the NA level rises. In septic shock, before death or at the development of a massive infection, the NA titre decreases rapidly. The decrease may be due partly to the permeability increasing effect of endotoxin and antigen-antibody reactions exerted on the capillaries. Consequently, in the most severe phase of sepsis, when bacteria enter the circulation less NA is available to fight against them. This might be a cause of the still very high lethality of septic shock due to Gram-negative bacteria. Finally, when applying the MTV one always has to consider that despite its advantages it gives less information than the Backhausz immunogram.  相似文献

The waveforms of live sea urchin sperm flagella were digitized with video-digitizing apparatus. The flagellar waveforms were expressed by the coordinates of 20 points spaced 2 micron apart along the flagella. The waveforms were condensed into simple algebraic expression with four parameters. Each of these parameters showed a systematic variation with the flagellar frequency. These average trends of the parameters made it possible to define an average, idealized, waveshape as a function of the flagella frequency, in the range 8-80 Hz.  相似文献

The clinico-immunological study of 80 patients with chronic prostatitis of different etiology was carried out. As controls, 20 healthy adult males were used. Mucous membranes of the reproductive tract were found to have different mechanisms of antimicrobial protection whose disturbances led to the inflammatory process. The clinico-immunological analysis carried out in the course of this study made it possible to state that, in contrast to healthy adults, pronounced changes in the characteristics of the local immunity status of the reproductive system occur in chronic prostatitis patients.  相似文献

The informative logical analysis has been used as the basis for the construction of the functional models of the immune status in the form of logical formulae. In these models the character of interrelations between immunocompetent cells in health and disease is reflected. Such models make it possible to substantiate the necessity, as well as the differentiated methods, of the correction of disturbances in the immune status, depending on the nature of the pathological case.  相似文献

As a first step toward making an efficient acclimatization methodology for minipigs, the reaction of G?ttingen minipigs, 3-24 months of age, toward humans was investigated. All minipigs were kept in an individual cage, and the reaction toward humans (acclimatization index) was evaluated by simple observations. The acclimatization index was evaluated as the total number of points scored (0-30 points) based on the following criteria: (1) the position of the minipig when the cage door was opened; (2) the reaction of the minipig when the observer approached; and (31 the reaction of the minipig when the observer touched it. Subsequently, each animal was ranked by total points scored: 30 points = AA, 20 points < or = A < 30 points, 10 points < or = B < 20 points, 0 points < or = C < 10 points. Based on this evaluation, the reactions of minipigs under three conditions were investigated. The following findings were confirmed: first, minipig reaction to humans was influenced by monthly age; second, taming was possible under ordinary conditions of care, but we had to wait until 10 months of age on average for this to occur; third, if simple contact was made during care time, minipigs became tame within less than 4 weeks after the commencement of contact. We therefore consider it possible to artificially control the reaction of minipigs toward humans, and to make minipigs more available for experiments by adding control of the hereditary factors that influence this reaction.  相似文献

A total of 145 women with chronic inflammatory diseases of uterus and appendages (IDUA) were examined. Bacterioscopy of smears and culture of vaginal, cervical canal and uterine cavity contents were performed in all patients. Direct immunofluorescence and PCR were used for detection of chlamydiae in scrapes and smears and in biopsy specimens taken from endometrium. The number of T lymphocytes and their subpopulations, B lymphocytes as well as the level of plasmatic cells synthesizing IgA, IgM and IgG and secretory IgA (sIgA), immune complexes and C3 component of the complement were determined in biopsy specimens of the endometrium. Pathomorphological examination revealed structural changes characteristic of the immunocomplex disease (secondary deficiency of sIgA, pronounced suppression of IgA production by plasmocytes along with an essential increase in the number of IgG synthesizing cells and immune complexes fixed to the venule endothelium) were detected. Lectin-histochemical study revealed considerable changes in secretory activity of endometrial epitheliocytes manifested by severe suppression of bactericidal activity of the uterine mucus. The detected disturbances of local protective reactions in the endometrium of women IDUA gave grounds for including immunomodulators into the traditional treatment scheme. The latter made it possible to increase essentially the therapeutic effect.  相似文献

Earlier experience of psychological trauma of a close person can through a transgenerational transfer influence traumatic reactions of a person going through a trauma at present, resulting in a repetition of earlier traumatic experiences and a development of a variety of mental disturbances. Purpose of our study was to evaluate the influence of transgenerational transfer on the development of Posttraumatic stress disorder (PTSD) in women with diagnosed breast cancer that had a family member with diagnosed cancer. The sample mainly consisted of 120 women treated in a Department of Oncology, Osijek University Hospital Center with diagnosis of newly discovered breast cancer, during the conduction of radio therapy having values Hamilton depression scale (HAM-D) from > or = 8 to < or = 24 or values Hamilton anxiety scale (HAM-A) from > or = 17 to < or = 30. Psychotherapeutic interview with a detailed clinical overview and with applying diagnostic criteria according to DSM-IV for mental disorders, specially structured non-standardized questionnaire for etiologic factors evaluation of the beginning of examinees' mental disorder, Los Angeles Symptom Checklist of PTSD symptoms (LASC), Hamilton's scale for anxiety evaluation (HAM-A) and Hamilton's scale for depression evaluation (HAM-D) were used. Results show that 61 (51%) of patients have a family member with diagnosed cancer. The average total value on LASC for examinees that had a family member with diagnosed cancer was slightly higher (22.92) in comparison to those who had no such family member (20.88). No statistically significant connection was found between having a family member with diagnosed cancer and the average total value on LASC. Although no connection was established between having a family member with diagnosed cancer and the average value on LASC in women with diagnosed breast cancer, transgenerational transfer of emotions seems to be important in their traumatic reactions, but it is still insufficiently researched and it is a challenge for future researches leaving many complicated issues open.  相似文献

The work presents the results of the study of the immune status in 32 patients with acute infectious destructions of the lungs; of these, 17 patients were immunized with staphylococcal vaccine, which made it possible to achieve a positive clinical effect in 16 patients, thus shortening their sick-leave. The injection of the preparation was shown to increase the number of E-rosette-forming cells, "active" E-rosette-forming cells, theophylline-resistant and theophylline-sensitive lymphocytes.  相似文献

Specific humoral immunity, total immune status and typing of HLA antigens, class 1, in loci A and B were studied in children with vaccine-associated paralytic poliomyelitis (VAPP). The immune status investigation revealed that changes in the content of serum immunoglobulins were most frequent. Out of 8 examined children, 5 children had IgA deficiency and 1 child had total variable immunodeficiency. In one case disturbances in cell-mediated immunity prevailed. Tissue typing revealed the presence of HLA A2 and B44 in 5 out of 6 examined children, which considerably exceeded their average occurrence among the Belorussian population. In spite of frequent detection of immunological disturbances in VAPP patients, out of 38 serologically examined children 36 (95%) were found to have virus-neutralizing serum antibodies to poliovirus, which was indicative of the capacity of their immune system for response to the administration of vaccine virus. To minimize the risk of VAPP in children and to achieve the goal of poliomyelitis eradication the combined immunization scheme consisting of 1-3 vaccinations with inactivated poliovaccine with subsequent administration of oral vaccine prepared from attenuated Sabin viruses is regarded as most promising.  相似文献

Idiosyncratic drug reactions represent a major problem. In most cases the mechanisms of these reactions are unknown, but circumstantial evidence points to the involvement of reactive metabolites and the characteristics of the reactions suggest involvement of the immune system. If progress is to be made in dealing with these adverse reactions it is essential that we have a better understanding of their mechanisms, and it is hard to imagine testing mechanistic hypotheses without good animal models. Unfortunately, idiosyncratic reactions are also idiosyncratic in animals so few good models exist. The best models, in which a rodent develops a clinical syndrome similar to that which occurs in humans, appear to be penicillamine-induced autoimmunity in Brown Norway rats and nevirapine-induced skin rash in rats. Sulfamethoxazole-induced hypersensitivity in dogs and propylthiouracil-induced autoimmunity in cats are also similar to adverse reactions that occur in people, but they have practical limitations. Halothane-induced liver toxicity in guinea pigs and amodiaquine-induced bone marrow and liver toxicity in rats represent models in which there is an immune response and mild, reversible toxicity. It is possible that the development of immune tolerance is what limits the toxicity in these models, and if this is true, interventions that prevent tolerance might lead to good models. Although the history of developing animal models of idiosyncratic drug reactions is mostly one of failure, such models are essential. A better understanding of immune tolerance may greatly facilitate the development of better models; transgenic technology may also provide an important tool.  相似文献

The signalling lymphocytic activation molecule (SLAM) family of receptors is expressed by a wide range of immune cells. Through their cytoplasmic domain, SLAM family receptors associate with SLAM-associated protein (SAP)-related molecules, a group of cytoplasmic adaptors composed almost exclusively of an SRC homology 2 domain. SAP, the prototype of the SAP family, is mutated in a human immunodeficiency named X-linked lymphoproliferative (XLP) disease. Recent observations indicate that SLAM family receptors, in association with SAP family adaptors, have crucial roles during normal immune reactions in innate and adaptive immune cells. Production of the Th1 cytokine IFN-gamma by T cells is considered crucial for immunity against Mycobacterium tuberculosis infection. We evaluated IFN-gamma production in tuberculosis in the context of signaling molecules known to regulate Th1 cytokines. Two populations of patients who have active tuberculosis were identified, based on their T cell responses to the bacterium. High responder tuberculosis patients displayed significant M. tuberculosis-dependent T cell proliferation and IFN-gamma production, whereas low responder tuberculosis patients displayed weak or no T cell responses to M. tuberculosis. The expression of the signaling lymphocytic activation molecule (SLAM)-associated protein (SAP) on cells from tuberculosis patients was inversely correlated with IFN-gamma production in those individuals. Moreover, patients with a nonfunctional SAP gene displayed immune responses to M. tuberculosis similar to those of high responder tuberculosis patients. In contrast to SAP, T cell expression of SLAM was directly correlated with responsiveness to M. tuberculosis Ag. Our data suggest that expression of SAP interferes with Th1 responses whereas SLAM expression contributes to Th1 cytokine responses in tuberculosis. The study further suggests that SAP and SLAM might be focal points for therapeutic modulation of T cell cytokine responses in tuberculosis.  相似文献

The effect of stress factors on the oculomotor reactions of electric train drivers with various degrees of fatigue was studied in a modelled accidental situation. The spatio-temporal parameters of oculomotor reactions were shown to characterize not always correctly the state of a driver. More informative were found to be the values of asymmetry of saccades obtained by separate recordings of the right and left eye movements. A comparison between the character of the phase saccade disturbances of tired operators and that of patients with a deficiency in cerebral blood supply showed that the latter may be used as a model.  相似文献

Reduced meat intake is often associated with iron deficiency anaemia. Reduced meat intake that arose from the frugality associated with a prolonged period of national economic reorientation policy, known as the "structural adjustment programme" (SAP), may have placed iron-stress on pregnancy in particular. Iron status of pre-SAP and SAP pregnancies were established from measurement of the haematological values of subjects. Indices such as haemoglobin (Hb), packed cell volume, serum Fe were lower in pregnancy especially during SAP. However, SAP pregnancy serum ferritin did not respond significantly to iron depletion. Unlike PCV, Total Iron Binding Capacity (TIBC) and serum transferrin values showed a reverse behaviour. The level of these values, however, portends a toll exerted on body iron status in pregnancy associated with reduced haem iron intake. The changes in these values did not, however, lead to any over symptoms of iron deficiency probably because increased sea food (molluscs) consumption ameliorates the iron-toll from low absorbed non-haem iron. For lower income rural women in non-coastal areas, the absence of seafood in their diets exacerbates the low iron status in Pregnancy.  相似文献

目的:探讨清胰利胆颗粒联合床旁连续性肾脏替代治疗(CRRT)对重症急性胰腺炎(SAP)患者炎性因子、免疫功能及外周血叉状头/翅膀状螺旋转录因子(Foxp3)、维甲酸相关孤独受体γt(RORγt)m RNA表达的影响。方法:选取2017年3月~2019年12月期间我院收治的SAP患者80例,根据随机数字表法分为对照组(n=40)和研究组(n=40),对照组患者予以床旁CRRT疗法治疗,研究组在对照组的基础上联合清胰利胆颗粒治疗,比较两组疗效、炎性因子、免疫功能、外周血Foxp3、RORγt mRNA相对表达量及不良反应。结果:研究组治疗后临床总有效率为87.50%(35/40),高于对照组患者的70.00%(28/40)(P<0.05)。研究组治疗后免疫球蛋白G(Ig G)、免疫球蛋白A(Ig A)、免疫球蛋白M(Ig M)、Foxp3 m RNA相对表达量较治疗前升高,且高于对照组(P<0.05)。两组治疗后白介素-6(IL-6)、白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)、RORγt m RNA相对表达量较治疗前降低,且研究组低于对照组(P<0.05)。两组在治疗过程中均未出现严重不良反应。结论:清胰利胆颗粒联合床旁CRRT疗法治疗SAP,疗效显著,可有效改善患者炎性因子、免疫功能,其主要作用机制可能与外周血Foxp3、RORγt m RNA相对表达量有关。  相似文献

On the basis of clinico-anatomo-electroencephalographic studies it was shown, that in early terms of cerebral trauma, at gross disturbances of the cortical functions and safety in some cases of only vital regulation, the parameters of the EEG stability or variability are of distinct information value for estimation of the functional state of patients. It was established, that changes of stability of the frequency, power, and coherent EEG characteristics correlate with different parameters of changes of the structural-functional brain organization. At this stage the greatest connection with the disease outcome reveals the EEG coherence parameters of the cortical symmetrical points reflecting the state of predominantly median formations and general brain reactions to traumatic action. In the process of restoration of disturbed neuromental functions priority prognostic significance is acquired by parameters of intrahemispheric coherence as well as by frequency-regional properties of interhemispheric asymmetry of coherence of the brain electrical processes, characterizing functional features of the lesion focus.  相似文献

A mass flux balance-based stoichiometric model of Bacillus licheniformis for the serine alkaline protease (SAP) fermentation process has been established. The model considers 147 reaction fluxes, and there are 105 metabolites that are assumed to be in pseudo-steady state. Metabolic flux distributions were obtained from the solution of the model based on the minimum SAP accumulation rate assumption in B. licheniformis in combination with the off-line extracellular analyses of the metabolites that were the sole carbon source citrate, dry cell, organic acids, amino acids, and SAP; variations in the intracellular fluxes were demonstrated for the three periods of the batch bioprocess. The flux distribution maps showed that the cells completed the TCA cycle and utilized the gluconeogenesis pathway, pentose phosphate pathway, and anaplerotic reactions throughout the fermentation; however, the glycolysis pathway was inactive in all the periods of the fermentation. The flux values toward SAP increased throughout the bioprocess and slightly decreased in the last period; however, SAP selectivity values were almost the same in Periods II and III and higher than Period I. The diversions in the pathways and certain metabolic reactions depending on the bioprocess periods are also presented and the results indicated that the intracellular amino acid fluxes played an important role in the SAP fermentation process.  相似文献

A robust model matching control of immune response is proposed for therapeutic enhancement to match a prescribed immune response under uncertain initial states and environmental disturbances, including continuous intrusion of exogenous pathogens. The worst-case effect of all possible environmental disturbances and uncertain initial states on the matching for a desired immune response is minimized for the enhanced immune system, i.e. a robust control is designed to track a prescribed immune model response from the minimax matching perspective. This minimax matching problem could herein be transformed to an equivalent dynamic game problem. The exogenous pathogens and environmental disturbances are considered as a player to maximize (worsen) the matching error when the therapeutic control agents are considered as another player to minimize the matching error. Since the innate immune system is highly nonlinear, it is not easy to solve the robust model matching control problem by the nonlinear dynamic game method directly. A fuzzy model is proposed to interpolate several linearized immune systems at different operating points to approximate the innate immune system via smooth fuzzy membership functions. With the help of fuzzy approximation method, the minimax matching control problem of immune systems could be easily solved by the proposed fuzzy dynamic game method via the linear matrix inequality (LMI) technique with the help of Robust Control Toolbox in Matlab. Finally, in silico examples are given to illustrate the design procedure and to confirm the efficiency and efficacy of the proposed method.  相似文献

In our previous studies a rather substantial difference between the initial values of the cerebral blood flow was found. On the other hand the brain monoamine content varies in different months of the year when studied. Comparative analysis of these parameters in rabbit brain was the aim of this paper. The content of noradrenaline (NA), dopamine (DA) and serotonin (5-HT) in cortical and subcortical structures and the local cerebral blood flow (ICBF), the systemic arterial pressure (SAP) and pulse rate (PR) were studied. There were found seasonal variations in all parameters. A certain LCBF retardation in subcortical structures and cortex and the weakest effect of the stimulation was observed in May. There was a drop in SAP and some PR increase in the spring (April-May). Brain NA and 5-HT content showed seasonal changes with the lowest values near the winter months and reaching maximum in May as the PR does. Content of DA was low in May. So it seams possible that the ion ICBF in May can be explained by the decrease of brain DA at that time. The ICBF and the reactivity of cerebral microvessels seem to depend on the monoamine content and show seasonal variability.  相似文献

The feasibilities of the Kelvin probe mode were shown to estimate a surface potential of lymphocytes from the patients with leukemia. Having compared the values of the surface potential obtained by the traditional method of microelectrophoresis and by the Kelvin probe mode, it was established that the use of the atomic force microscope made it possible to obtain data which were of specified diagnostic importance while studying lymphocyte functional status at different stages in leucopoiesis and indicated progression of the pathological process.  相似文献
