In the present study, bioleaching characteristic of cinnabar and process parameters were investigated. The effects of temperature, initial pH of the solution, dilution rate and iron on the cinnabar bioleaching were examined. The relationship between the redox potential and extraction of mercury was evaluated. Based on the S/N ratio, the optimized conditions for bioleaching were established. Iron of the solution was the most effective parameter in comparison with others. A maximum mercury concentration of 1.38 g/L was achieved. The indirect mechanism plays a major role in bioleaching. Some jarosite crystal deposits could be found when the iron concentration was 4.0 g/L and initial pH was 1.9. The bioxidation of sulfur could inhibit the hydrolyzation of ferric iron. Regulating pH value and ion concentration in medium could control the formation of the precipitation.  相似文献

The aim of this paper is to determine the efficiency of bioleaching of arsenic in realgar, a Chinese mineral drug, using pure cultures of Acidithiobacillus ferrooxidans or Acidithiobacillus thiooxidans and a mixed culture of A. ferrooxidans and A. thiooxidans. The experiments were carried out in shaker flasks, at 150 rpm, 30 °C at a culture pH of 1.80. To investigate the mechanism of the bioleaching in realgar, media with and without ferrous iron were chosen for the experiments. The results showed that the leaching rate of arsenic in realgar after 20 days was higher (43%) in A. ferrooxidans cultures with ferrous iron compared to cultures without ferrous iron (10%), and the leaching rate of A. thiooxidans cultures only increased from 21% to 23% in the presence of ferrous iron. The leaching rate of arsenic in mixed culture with ferrous iron was greatly enhanced from 16% to 56%, indicating that bioleaching in mixed culture is preferable for the dissolution of realgar.  相似文献

Bio-leaching studies were carried out in a 2 L bioreactor- BIOSTAT-B® equipped with a PLC based controller at 20–40% (w/v) pulp density using enriched culture of A.ferrooxidans for Turamdih uranium ore (Jharkhand, India). With the enriched culture of A.ferrooxidans adapted on Fe(II) at pH 2.0, 35 °C and 20% (w/v) pulp density, a 98.3% uranium recovery was recorded in 14 days. The leaching of uranium in the bioreactor improved the dissolution rate by reducing the time from 40 days in shake flask as per our earlier studies to 14 days. While investigating the importance of biogenic Fe(III) in the bio-leaching process a maximum recovery of 84.7% U3O8 was observed at pH 2.0 and 20% (w/v) pulp density in 10 h as compared to the uranium leaching of 38.3% in the control experiments. On raising the pulp density to 30%, uranium bio-recovery increased to 87.6% in 10 h at pH 2.0 with <76 μm size material. This showed a distinct advantage because of better mixing of slurry in the bioreactor with auto-controlled conditions that improved the kinetics.  相似文献

In the present study, enzyme assisted extraction of organosulfur compounds, especially cycloalliin, from garlic (Allium sativum L.) was examined using various commercial cellulases, and the changes of the cycloalliin contents in garlic extract were investigated after storage at 40 °C and 60 °C for 30 days. Among the commercial enzymes tested, Ultraflo L showed the greatest yield of cycloalliin compared to other cellulases. The conditions were optimized to include 2.5% (v/w) addition of Ultraflo L, 1 h incubation at 40 °C and a pH of 6.0. Under the optimum conditions, the contents of cycloalliin achieved 1.5-folds increase in the enzyme-assisted garlic extract compared with the non-enzymatic extraction. In addition, the cycloalliin content was also significantly increased 3.8-fold after storage at 60 °C for 15 days. The polyphenol content was also significantly increased by 3-fold after at 60 °C for 30 days. Overall, Ultraflo L proved to be an efficient method to extract cycloalliin from garlic.  相似文献

Constructed treatment wetlands have served the City of Columbia, MO, for fourteen years. Four free water surface wetland units in series, comprised of 23 cells, are an addition to the activated sludge wastewater treatment plant, for the purpose of added biochemical oxygen demand (BOD) and total suspended solids (TSS) control. The system operates year-round, and supplies water to the Eagle Bluffs Conservation Area for wetland maintenance. The cattail wetlands processed an average of 57,000 m3/d, at a water depth of 20 cm. The resulting detention time was approximately 2 days, and the hydraulic loading was 13 cm/d. Water temperatures were warm leaving the treatment plant and in the wetlands in winter, because of the short detention. The period of record average carbonaceous biochemical oxygen demand (CBOD) leaving the wetlands was 5.0 mg/L, and the TSS was 14.7 mg/L. Dissolved oxygen was depressed in summer, likely because of the high sediment demand. Nutrient concentrations were only minimally reduced, total nitrogen (TN) by 22% and total phosphorus (TP) by 6%. However, load reductions were maximal, 98 t/yr for nitrogen, and 3.6 t/yr for phosphorus. Fecal coliforms were reduced by 98%, and E. coli by 95%. First order rate coefficients were high for CBOD (64 m/yr), nitrate (61 m/yr) and organic nitrogen (42 m/yr), but relatively low for ammonia (8 m/yr) and phosphorus (5.7 m/yr). Nitrogen removal was strongly affected by vegetative uptake. Sediment accretion in the wetland inlets was substantial, at 1.6 cm/yr in the inlets to the upstream wetland units. Muskrats caused vegetation damage, and waterfowl use was high in winter, causing TSS excursions.  相似文献

The study aimed at investigating rural domestic wastewater treatment performance through vermifilter enhancement by a converter slag–coal cinder filter. The research was carried out by column experiments in a lab scale. Results showed the average removal rate of TCOD, BOD, ammonia nitrogen (NH4+-N) and phosphorus removal by the system were 78.0%, 98.4%, 90.3%, 62.4%, respectively at a hydraulic loading rate of 4 m3 m?2 day?1. Vermifiltration was effective for insoluble organic matter and suspended solid removal, and the converter slag–coal cinder filter played an important role in phosphorus removal. The molecular weight of particles in the influent and effluents from every unit of the system were distributed in a wide range between 0.1 kDa and 10,000 kDa with predominance of values between 10 kDa and 300 kDa; the major portion of soluble material are high MW compounds. In addition, the optimal design parameters for vermifiltration and converter slag and coal cinder filters, respectively, were studied in the experiments.  相似文献

Three-phase partitioning (TPP) was used to isolate trypsin inhibitors from navy bean (NB), red kidney bean (RK) and adzuki bean (AZ) from the Royal Project Foundation in Thailand. The method was to mix the crude extract with solid ammonium sulfate (30% saturation, w/v) and tert-butanol (t-butanol) in order to obtain the three phases. The trypsin inhibitor was purified to 5-, 14- and 7-fold with 315%, 441% and 228% recovery for NB, RK and AZ, respectively. The SDS-PAGE showed the major inhibitor band with the molecular weights (MWs) of 132, 118 and 13 kDa for NB, RK and AZ, respectively. The fractions from NB and AZ showed higher pH stability compared to that of the RK, and they had the optimum pH ranges of 7–9. The highest relative inhibitory activity of the fractions of NB and RK were found at 50 °C, and all fractions were quite stable at 90 °C for 60 min of incubation. Increasing the concentration of salt (up to 3%, w/v) did not significantly decrease the inhibitory activity of all fractions (p > 0.05). The trypsin inhibitors from the three legumes were unable to inhibit the autolysis of Pacific whiting and arrowtooth flounder muscles.  相似文献

We developed a new pretreatment process for producing high-efficiency bioethanol from a lignocellulosic biomass. Barley straw was pretreated with sodium hydroxide in a twin-screw extruder for continuous pretreatment. The biomass to ethanol ratio (BTER) for optimal pretreatment conditions was evaluated by response surface methodology. Simultaneous saccharification and fermentation (SSF) was conducted to investigate the BTER with 30 FPU/g cellulose of enzyme and 7% (v/v) yeast (Saccharomyces cerevisiae CHY 1011) using 10% (w/v) pretreated biomass under various pretreatment conditions. The maximum BTER was 73.00% under optimal pretreatment conditions (86.61 °C, 0.58 M, and 84.79 mL/min for temperature, sodium hydroxide concentration, and solution flow rate, respectively) and the experimental BTER was 70.01 ± 0.59%. SSF was performed to investigate the optimal enzyme and biomass dosage. As a result, maximum ethanol concentration and ethanol yield were 46.00 g/L and 77.36% at a loading pretreated biomass of 20% with 30 FPU/g cellulose of the enzyme dosage for barley straw to bioethanol. These results are a significant contribution to the production of bioethanol from barley straw.  相似文献

Strain Trichoderma koningii D-64 was improved for enhanced cellulase production. A potential mutant MF6 was obtained and its enzymes contained filter paper cellulase (FPase), carboxymethylcellulase (CMCase), β-glucosidase and xylanase with respective activities of 2.0, 1.3, 2.0 and 3.0 folds of those for the parental strain. MF6 cellulases showed enhanced hydrolysis performance for the treated lignocellulosic biomass. Hydrolysis of treated oil palm empty fruit bunch (OPEFB), horticulture wastes (HW) and wood chips (WC) resulted in cellulose to glucose conversion of 96.3 ± 2.2%, 98.2 ± 3.0% and 81.9 ± 1.4%, respectively. The corresponding conversions of xylan to xylose were 96.9 ± 1.5%, 95.0 ± 2.2% and 76.1 ± 3.1%. Consistently, high sugar yield of 770–844 mg/g biomass was obtained for high-loading (10–16%, w/v) of OPEFB hydrolysis and sugar titer of 135.1 g/L was obtained for 16% (w/v) OPEFB loading at 96 h. In addition, MF6 enzymes alone performed equally well for high-loading OPEFB hydrolysis compared to the enzyme mixture of β-glucosidase from Aspergillus niger and cellulase from T. reesei Rut C30.  相似文献

A novel polyethylene glycol (PEG) gel was fabricated and used as a carrier to immobilize Clostridium sp. LS2 for continuous hydrogen production in an upflow anaerobic sludge blanket (UASB) reactor. Palm oil mill effluent (POME) was used as the substrate carbon source. The optimal amount of PEG-immobilized cells for anaerobic hydrogen production was 12% (w/v) in the UASB reactor. The UASB reactor containing immobilized cells was operated at varying hydraulic retention times (HRT) that ranged from 24 to 6 h at 3.3 g chemical oxygen demand (COD)/L/h organic loading rate (OLR), or at OLRs that ranged from 1.6 to 6.6 at 12 h HRT. The best volumetric hydrogen production rate of 336 mL H2/L/h (or 15.0 mmol/L/h) with a hydrogen yield of 0.35 L H2/g CODremoved was obtained at a HRT of 12 h and an OLR of 5.0 g COD/L/h. The average hydrogen content of biogas and COD reduction were 52% and 62%, respectively. The major soluble metabolites during hydrogen fermentation were butyric acid followed by acetic acid. It is concluded that the PEG-immobilized cell system developed in this work has great potential for continuous hydrogen production from real wastewater (POME) using the UASB reactor.  相似文献

Biopolymer DYU500 produced from Bacillus subtilis DYU1 was found to have excellent flocculating ability. With the addition of 40 mg-DYU500/L and 50 mM CaCO3, the optimum temperature for flocculation performance of DYU500 was 30 °C, giving the highest flocculating activity and rate of 13.5 and 97%, respectively. Analysis with Fourier transform infrared spectrophotometry (FT-IR), nuclear magnetic resonance spectrometry (NMR) and amino acid identification shows that the DYU500 biopolymer mainly possesses the structure of poly-glutamic acid (PGA). The average molecular weight of DYU500 was about (3.16–3.20) × 106 Da as determined by gel permeation chromatography. The major components of biopolymer DYU500 were total sugars, uronic acids, proteins and polyamides (homopolymer of glutamic acid), accounting for a weight ratio of approximate 14.9, 2.7, 4.4 and 48.7% (w/w), respectively. The flocculating activity of DYU500 in the kaolin suspension was markedly stimulated by the addition of bivalent cations Ca2+ or Mg2+ in optimum concentration ranges of about 0.15–0.90 and 0.10–0.90 mM, respectively. The synergistic effect of cations was most effective at a weak acidic or neutral pH (6.0–7.0). The flocculating activity of DYU500 linearly decreased with an increase in incubation temperature and the activity was completely lost when heating upon 120 °C, arising from the destruction of the polyamides structure of DYU500. Moreover, mechanisms describing the flocculation process with DYU500 were proposed based on the experimental observations.  相似文献

This work studied the hydrolysis of lactose using β-galactosidase from Aspergillus oryzae immobilized with a combination of adsorption and glutaraldehyde cross-linking onto the ion exchange resin Duolite A568 as a carrier. A central composite design (CCD) was used to study the effects of lactose concentration and feed flow rate on the average hydrolysis reaction rate and lactose conversion in a fixed bed reactor operating continuously with an upflow at a temperature of 35 ± 1 °C. The optimal conditions for the average hydrolysis reaction rate and the lactose conversion included a lactose concentration of 50 g/L and a feed flow rate of 6 mL/min. The average reaction rate and conversion reached 2074 U and 65%, respectively. The immobilized enzyme activity was maintained during the 30 days of operation in a fixed bed reactor with a 0.3 mL/min feed flow rate of a 50 g/L lactose solution at room temperature. Feed flows ranging from 0.6 to 12 mL/min were used to determine the distribution of residence times and the kinetics of the fixed bed reactor. A non-ideal flow pattern with the formation of a bypass flow in the fixed bed reactor was identified. The conditions used for the kinetics study included a lactose solution concentration of 50 g/L at pH 4.5 and a temperature of 35 ± 1 °C. Kinetic models using a PFR and axial dispersion methods were used to describe the lactose hydrolysis in the fixed bed reactor, thus accounting for the competitive inhibition by galactose. To increase the lactose conversion, experiments were performed for two fixed bed reactors in series, operating in continuous duty with upflow, with the optimal conditions determined using the CCD for a fixed bed reactor. ObjectivesThe aim of this study was to investigate blood lead level and its relationship to essential elements (zinc, copper, iron, calcium and magnesium) in school-age children from Nanning, China.MethodsA total of 2457 children aged from 6 to 14 years were enrolled in Nanning, China. The levels of lead (Pb), zinc (Zn), copper (Cu), iron (Fe), calcium (Ca) and magnesium (Mg) were determined by an atomic absorption spectrometer.ResultsThe mean blood lead level (BLL) was 57.21 ± 35.00 μg/L. 188 (7.65%) asymptomatic children had toxic lead level higher than 100 μg/L. The school-age boys had similar lead level among different age groups, while the elder girls had less BLL. The blood Zn and Fe were found to be increased in the boys with elevated BLL, but similar trends were not observed in the girls. Positive correlations between Pb and Fe or Mg (r = 0.112, 0.062, respectively, p < 0.01) and a negative correlation between Pb and Ca (r = −0.047, p < 0.05) were further established in the studied children.ConclusionsLead exposure in school-age children was still prevalent in Nanning. The boys and girls differed in blood levels of lead and other metallic elements. Lead exposure may induce metabolic disorder of other metallic elements in body.  相似文献

Efficacies of phosphorolytic enzymes (phytase+acid phosphatase), and an enzymic “cocktail” (phytase+acid phosphatase+pectinase+citric acid), were investigated in broilers fed wheat-based diets from day 1 to 43. Broilers were fed the following four diets: (1) a positive control diet (7.1 g total P/kg, 4.1 g non-phytate P/kg, 9.8 g Ca/kg); (2) a low phosphorus diet (4.1 g total P/kg, 1.7 g non-phytate P/kg, 6.0 g Ca/kg) supplemented with phytase (750 units/kg) and acid phosphatase (3156 units/kg); (3), a low phosphorus diet (4.1 g total P/kg, 1.7 g non-phytate P/kg, 6.0 g Ca/kg) supplemented with phytase, acid phosphatase, pectinase (1900 units/g) and citric acid (20 g/kg); and (4) a low phosphorus diet (4.1 g total P/kg, 1.7 g non-phytate P/kg, 8.0 g Ca/kg) supplemented as in diet 3. For the grower period (22–43 days), the contents of P and Ca were lowered by 0.2 and 0.3 g/kg, respectively. The dietary treatments were fed to three floor pen replicates of 50 birds each. For the starter period, there were no differences observed among dietary treatments in terms of body weight gains or feed efficiency. Total body weight gains for the starter and grower periods did not differ among dietary treatments, but total feed efficiency was significantly enhanced in birds fed diet 2. At the completion of the experiment chickens fed phosphorolytic enzymes had the best feed efficiencies, the highest contents of ash in the toes, and the highest carcass yield. Chicken receiving the cocktail of enzymes and 8 g Ca/kg (diet 4) performed as well as birds in the control treatment, but had higher yields of carcass and excreted 56% less phosphorus.  相似文献

Acetoin production by Bacillus amyloliquefaciens was used as a model of product feedback to develop a strategy to enhance the production of acetoin. To enhance the resistance of B. amyloliquefaciens to acetoin, an acetoin-tolerant mutant E-11 was screened by using adaptive evolution with acetoin stress as the selection pressure. When compared with the parent FMME044, the mutant E-11 exhibited superior fermentation performance as follows: (1) the mutant E-11 exhibited increased tolerance to high concentration of acetoin, and the specific growth rate was 265.2% higher than that of the parent FMME044 in medium containing 80 g/L acetoin; (2) acetoin production by the mutant E-11 reached 71.5 g/L at 44 h when cultured in a 7-L fermentor with 173 g/L glucose, and the acetoin concentration and productivity of the mutant E-11 were 39.6% and 14.4% higher than those of the parent FMME044, respectively; (3) the unsaturated fatty acid contents in the mutant E-11 were 64.8%, 37.8%, and 18.4% higher than those in the parent FMME044 when cultured in 0, 40, and 60 g/L acetoin, whereas the saturated fatty acid contents in the mutant E-11 were 9.5%, 13.9%, and 14.1% lower than those in the parent FMME044, respectively.  相似文献

This paper examined the growth and yield performance of Lentinus citrinus on cupuaçu exocarp (Theobroma grandiflorum) mixed with litter (CE + LI) or rice bran (Oryza sativa) (CE + RB) in the ratio of 2:1 (800 g:200 g) to investigate the nutritional composition and proteolytic potential of the fruiting body produced. Significance values of yield were determined on substrate combinations. In CE + LI the biological efficiency of the mushrooms was 93.5% and the content of fat (4.5%), fiber (11.0%), protein (27.0%) and amino acids were higher when compared with CE + RB. Among the amino acids, the amount of glutamic acid, aspartic acid, alanine, arginine and leucine was high. The biological efficiency on CE + RB reduced to 84.2% and based on the nutritional value, carbohydrates (53.59%), energy (324.33 kcal) and minerals such as zinc, iron, copper, potassium and phosphorus were higher in this substrate combination. Protease activity from fruiting body was significant in CE + LI (463.55 U/mL). This protease showed an optimal activity at 50 °C in neutral and alkaline pH with maximum stability at 30 °C at alkaline pH. This is the first report of L. citrinus fruiting body nutritional composition with potential for human food and application in industrial processes.  相似文献

The phosphorus (P) fractions and bioavailable P in the sediments from the Quanzhou Bay Estuarine Wetland Nature Reserve were investigated using chemical extraction methods for the first time to study the distribution and bioavailability of P in the reserve sediments. A hypothesis was presented suggesting that the bioavailable P in the sediments could be evaluated using the P fractions. The total phosphorus (TP), inorganic phosphorus (IP), organic phosphorus (OP), non-apatite phosphorus (NAIP), and apatite phosphorus (AP) contents in the sediments were in the ranges of 303.87–761.59 mg kg−1, 201.22–577.66 mg kg−1, 75.83–179.16 mg kg−1, 28.86–277.90 mg kg−1, and 127.36–289.94 mg kg−1, respectively. The water soluble phosphorus (WSP), readily desorbable phosphorus (RDP), algal available phosphorus (AAP), and NaHCO3 extractable phosphorus (Olsen-P) contents in the sediments were in the ranges of 0.58–357.17 mg kg−1, 80.77–586.75 mg kg−1, 1.09–24.12 mg kg−1, and 54.96–676.82 mg kg−1, respectively. The correlation analysis results showed that the NAIP was the major component of the bioavailable P and that the impact of the AP on the bioavailable phosphorus may be minimal. Due to the low TP content in the sediments of the Quanzhou Bay Estuarine Wetland Nature Reserve, the potential pollution risks of P in the sediments may not be very high. The results also show that the bioavailable P concentrations in the sediments of the Quanzhou Bay Estuarine Wetland Nature Reserve could not be evaluated by measuring the P fractions and that the hypothesis was untenable.  相似文献

The high concentration of total phenolic compounds (TPC) in Psidium guajava leaf extracts (GvEx) is correlated to its anti-hyperglycemic activity. In this study, we established the optimum ultrasound extraction conditions for maximizing TPC yield. The response surface methodology (RSM) was employed for empirical model building. The maximum value of TPC (26.12%) was obtained at solvent to solid ratio (v/w) of 12.1, extraction temperature of 59.8 °C, and extraction time of 5.1 min. The anti-hyperglycemic activity of GvEx was compared to the commonly used diabetic drug acarbose. The IC50 of GvEx for α-amylase and α-glucosidase inhibition was 50.5 μg/mL and 34.6 μg/mL, respectively. However, the IC50 of acarbose for α-amylase and α-glucosidase inhibition was 95.3 μg/mL and 1075.2 μg/mL, respectively. In conclusion, GvEx obtained under optimum extraction conditions had higher anti-hyperglycemic activity than acarbose. In addition, the recommended extraction procedures for GvEx save time and are environmentally friendly.  相似文献

This study investigated the characteristics of nitrous oxide (N2O) emission from intermittently aerated sequencing batch reactors (IASBRs) treating high strength slaughterhouse wastewater at 11 °C, where partial nitrification followed by denitrification (PND) was achieved. N2O generation and emission was examined at three aeration rates of 0.4, 0.6, and 0.8 L air/min in three IASBRs (SBR1, SBR2, and SBR3, respectively). The slaughterhouse wastewater contained chemical oxygen demand (COD) of 6057 ± 172.6 mg/L, total nitrogen (TN) of 576 ± 15.1 mg/L, total phosphorus (TP) of 52 ± 2.7 mg/L and suspended solids (SS) of 1843 ± 280.5 g/L. In the pseudo-steady state, the amount of N2O emission was up to 5.7–11.0% of incoming TN. The aeration rate negatively affected N2O emission and the ratio of N2O emission to incoming TN was reduced by 48.2% when the aeration rate was increased from 0.4 to 0.8 L air/min. Results showed that more N2O was generated in non-aeration periods than in aeration periods. Lower DO concentrations enhanced N2O generation in the aeration periods (probably via nitrifier denitrification) while low DO concentrations (lower than 0.2 mg/L) did not affect N2O generation in the non-aeration periods (probably via heterotrophic denitrification). When PHB was utilized as the organic substrate for denitrification, there was a high N2O generation potential. It was estimated that 1.8 mg N2O-N was generated accompanying per mg PHB consumed.  相似文献

The production of conjugated linoleic acid (CLA) by four strains of lactic acid bacteria isolated from fish, i.e., Leuconostoc mesenteroides H20, Leuconostoc mesenteroides H22, Leuconostoc lactis H24 and Lactobacillus pentosus H16, was evaluated in MRS broth and on MRS agar. The bioconversion and production of CLA by resting cells were also assessed. Linoleic acid was detected in cultures grown on agar at percentages of up to 18.3% (w/w) of total fatty acid, and conjugated isomers were found in the fatty acid profiles of Lactobacillus pentosus H16. The percentage of CLA relative to total fatty acid increased from 5.68 ± 1.65% to 23.69 ± 0.79% when resting cells were removed from agar plates and incubated without the addition of exogenous linoleic acid as a substrate. When Lactobacillus pentosus H16 cells were incubated with linoleic acid, cyclization and changes in monounsaturated fatty acid percentages were observed instead of conjugation. These results show that growth on a solid support is required for CLA production. More significantly, an increase in the CLA content could be achieved by incubating resting cells without exogenous substrate.  相似文献
