Biofilm development on titanium panels immersed in the surface waters of Dona Paula Bay was investigated using molecular biomarkers such as n-alkanes and other chemical and biological parameters. Biofilm biomass measured as organic carbon (OC), organic nitrogen (ON), chlorophyll a, diatoms and bacterial numbers on the titanium panels generally increased over the period of immersion. Total lipids and n-alkane concentration also showed similar trends. n-alkanes from C12 to C30 were detected in the biofilm samples, which showed a bimodal distribution. The first mode consisted of n-alkanes > C23 with a strong even over odd predominance. In the second mode, the n-alkanes < C23 were more abundant with odd carbon number maxima at C15, C17 and C19 and a strong odd over even carbon number predominance (Carbon Preference Index > 2). The predominance of these odd-chain n-alkanes strongly indicates that the organic matter derived from macroalgal sources was the major contributor to the biofilm organic matter developed on the titanium panels over the 15 d period of study. The data suggest that molecular characterization is a useful tool in understanding the sources of biofilm organic matter. The observed abundance of macroalgal organic matter during the 15 d period of biofilm development may play an important role in subsequent fouling by micro- and macrofouling organisms.  相似文献

Mild steel (MS), stainless steel (SS) and copper (Cu) test panels were immersed in the surface water of Dona Paula Bay over a period of 15 d. During the immersion period data on the hydrography, nutrients and suspended matter were also collected. The suspended matter and fouling products on the MS, SS and Cu panels were analysed for organic carbon (OC), organic nitrogen (ON), chlorophyll a (chl a), protein and carbohydrate concentration and composition, and the dry weight (DW) was recorded. Compared to suspended matter, the chemical and biochemical components of the fouling products showed strong temporal and substratum related differences. The microfouling biomass (as DW, OC, ON, chl a and protein) on all the test panels generally increased over the period of immersion. Carbohydrates were more abundant in the suspended matter whereas fouling products were enriched in proteins. The contribution of protein-carbon to the total carbon increased over the period of immersion for the microfouling products on MS and SS whilst it did not show a consistent trend on Cu. Whereas, the carbohydrate-carbon contribution to the total carbon increased for the fouling products on MS, it did not exhibit a particular pattern on SS or Cu over the period of immersion. Capillary gas chromatographic analysis showed the presence of glucose, galactose, mannose, arabinose, xylose, fucose and ribose in both the fouling products and suspended matter. However, there were differences in the relative distribution of these monosaccharides in the suspended matter and the fouling products. Glucose was the most abundant monosaccharide, which showed strong temporal variations in suspended matter. In contrast, the wt % concentrations of individual monosaccharides showed large temporal differences for the fouling products, which were strongly influenced by the period of immersion and the type of test substratum. Glucose and fucose were relatively more abundant in the fouling products on SS and Cu, whilst glucose was the most abundant monosaccharide on MS. The monosaccharide and chemical composition data suggest strong temporal changes in the composition of the fouling products.  相似文献

A growing number of studies have provided insights into the diversity of coral-associated bacteria and their function in the coral holobiont. Yet, information about spatial heterogeneity of bacteria within coral colonies is limited. Using 16S rRNA gene metabarcoding, we analyzed the bacterial community composition across four distinct locations in each of five wild Acropora loripes colonies. Considerable variation within and among colonies was present, which has implications for sampling strategies and data interpretation in coral microbiome research. Bacterial assemblages significantly differed in alpha and beta diversity among colonies, with all corals possessing a high relative abundance of Endozoicomonas. When the same A. loripes colonies were subsequently reared in aquaria over 4 weeks, the relative abundance of Marinobacter initially increased in all colonies. However, no significant alteration in bacterial community composition was observed over time and the colonies maintained distinct bacterial microbiomes.

Changes in bacterial surface hydrophobicity, charge, and degree of irreversible binding to glass surfaces of seven marine isolates were followed during starvation. The degree of hydrophobicity was measured by hydrophobic interaction chromatography and by two-phase separation in a hexadecane-water system, whereas changes in charge were measured by electrostatic interaction chromatography. All isolates underwent the starvation-induced responses of fragmentation, which is defined as division without growth, and continuous size reduction, which results in populations with increased numbers of smaller cells. The latter process was also responsible for a significant proportion of the total drop in cell volume; this was observed by noting the biovolume (the average cell multiplied by the number of bacteria) of a population after various times of starvation. Four strains exhibited increases in both hydrophobicity and irreversible binding, initiated after different starvation times. The most hydrophilic and most hydrophobic isolates both showed a small increase in the degree of irreversible binding after only 5 h, followed by a small decrease after 22 h. Their hydrophobicity remained constant, however, throughout the entire starvation period. On the other hand, one strain, EF190, increased its hydrophobicity after 5 h of starvation, although the degree of irreversible binding remained constant. Charge effects could not be generally related to the increase in irreversible binding. Scanning electron micrographs showed a large increase in surface roughness throughout the starvation period for all strains that showed marked changes in physicochemical characteristics.  相似文献

Body condition and parasite abundance were examined in two size classes of European bitterling Rhodeus amarus during the first overwintering period in two seasons (2007–2008 and 2009–2010). Body condition of large fish did not change during winter, and increased significantly in March. From November to February, small fish showed a decreasing trend in condition. Despite a significant increase in March condition of small fish only reached the same level as before winter. Total parasite abundance increased significantly in winter in both fish size classes, reflecting a seasonal increase in monogenean infection. Large fish were parasitized significantly more than small fish during winter, but only in small fish was a negative correlation between parasite infection and condition found and a significant decrease in parasite abundance recorded after wintering, indicating mortality of heavily infected individuals with low condition during the winter. A trend for higher overwinter mortality in small fish was found under semi‐experimental conditions. The decrease in condition during the winter period in small fish may reflect faster energy depletion generally expected in smaller individuals. The results indicate that parasite infection may contribute to the overwinter mortality of 0+ year R. amarus, with a stronger effect in smaller individuals.  相似文献

Well-defined laboratory strains as well as 72 clinical strains ofStaphylococcus aureus isolated from bovine mastitis were investigated for surface hydrophobicity by the salt aggregation test (SAT).Staphylococcus aureus strain Cowan 1, rich in protein A and fibronectin-binding surface proteins, was found to show high surface hydrophobicity, whereas strain Wood 46, deficient in these surface proteins, showed low surface hydrophobicity. SAT showed a significant difference in surface hydrophobicity (P<0.001) between protein A-positive and A-negative strains measured by 2-test analysis. Comparison of SAT values with results obtained from hydrophobic interaction chromatography (HIC) showed a good correlation (P<0.025). A high-level protein-A-producing mutant (SA 113prA-3) showed increased surface hydrophobicity as compared with the parent strain (SA 113), whereas ten protein-A-negative mutants showed low surface hydrophobicity in SAT. Of the 72 clinical isolates tested by SAT, 47 (65%) showed autoaggregating properties, i.e., the strains aggregated even in isotonic buffers. Tween 80 (1% vol/vol) and ethylene glycol (50% vol/vol) prevented autoaggregation of some hydrophobic strains aggregating in phosphate-buffered saline. However, 2M of a chaotropic agent (NaSCN) was more efficient in preventing autoaggregation of the strains tested. Heating of cell suspensions to 80°C or 100°C as well as trypsin andStreptomyces griseus protease treatment generally caused a decrease in the cell surface hydrophobicity. This indicates that protein A, fibronectin-binding proteins, and probably other as yet unidentified proteins contribute to the high surface hydrophobicity of most strains isolated from bovine mastitis.  相似文献

The role of cell surface hydrophobicity in the adhesion to stainless steel (SS) of 11 wild yeast strains isolated from the ultrafiltration membranes of an apple juice processing plant was investigated. The isolated yeasts belonged to four species: Candida krusei (5 isolates), Candida tropicalis (2 isolates), Kluyveromyces marxianus (3 isolates) and Rhodotorula mucilaginosa (1 isolate). Surface hydrophobicity was measured by the microbial adhesion to solvents method. Yeast cells and surfaces were incubated in apple juice and temporal measurements of the numbers of adherent cells were made. Ten isolates showed moderate to high hydrophobicity and 1 strain was hydrophilic. The hydrophobicity expressed by the yeast surfaces correlated positively with the rate of adhesion of each strain. These results indicated that cell surface hydrophobicity governs the initial attachment of the studied yeast strains to SS surfaces common to apple juice processing plants.  相似文献

This study documents the advancement of laying dates in three species of tits (Paridae) in southernmost Sweden during recent decades, and the absence of a similar response in the pied flycatcher Ficedula hypoleuca. It is based on several different nestbox studies; the oldest one starting in 1969. During 1969 to 2012, mean spring temperatures in the study area increased by between 0.06 and 0.08°C per year, depending on the period considered. Great tits Parus major, blue tits Cyanistes caeruleus and marsh tits Poecile palustris, which generally start egg laying between the last week of April and the first week of May, all advanced laying date at a similar rate during the study period (0.25 d yr–1). This indicates that these species were similarly affected by increasing temperatures. When accounting for mean spring temperature variation, we still found an advancement of laying date over the study period, mostly due to such relationships among marsh and blue tits. This result could reflect ongoing microevolution favouring earlier laying, but could also be a result of other factors such as increased intra‐ or inter‐specific competition for early breeding. Pied flycatchers, which generally lay during the third week of May, did not significantly advance the date of egg laying despite that the long‐term trend in the increase in ambient temperature during the 30‐d period preceding the start of egg laying was similar for pied flycatchers compared to the tit species.  相似文献

The goal of this study was to analyze the variations in abundance of rodent communities over the last 24 years in a rural area, and their relation to possible changes in climatic variables and land use. The principal change in the area observed along the study period was an increase in the area covered by soybean. The habitats studied were crop fields and borders. The total abundance of rodents did not show a significant trend of variation over time in crop fields while in borders the abundance in autumn–winter (A–W) showed a significant trend to decrease over time. The different rodent species showed a differential response over time. While Calomys laucha and C. musculinus showed a decrease between the period before and after the soybean expansion, Akodon azarae did not change its abundance over time. The mean minimum temperature increased over time while the number of days with frost decreased. Total rodent abundance in A–W was positively associated with the cumulated precipitation of the previous spring–summer period and negatively with the cumulated precipitation of the same period. We conclude that rodent abundance variations in crop fields and borders of the study area are influenced by precipitation, but the observed trends of variation over time are better explained by changes in agricultural practices than by meteorological variables.  相似文献

We have studied the physiological and morphological features of 17 isolates ofParacoccidioides brasiliensis in order to define their phenotypes. The isolates were cultured at room temperature on potato dextrose agar (PDA, Difco) slants for mycelial growth and in 1% dextrose brain heart infusion agar (BHIA, Difco) at 37°C for the study of yeast forms. Most mycelial and yeast forms grew well between pH 5.6–9.4. In their response to osmotic pressure the isolates were separated in three groups: intolerant, intermediate and tolerant. They also varied in carbohydrate assimilation tests, which indicated important metabolic variation. No clear differences were observed in phenol oxidase tests, KNO3, starch, casein and arbutin assimilation tests. Only 1 of the isolates, Bt-19, had gelatinase activity. No correlation was observed between the above differences and virulence. Two patterns of growth were observed in the mycelial cultures, glabrous and cottonous, the latter being correlated with increased virulence for ddY mice. Most yeast forms grew as cerebriform colonies, but Pb-HC and Bt-19 colonies had a cobblestone-like surface.  相似文献

To investigate the effects of physiological properties on polycyclic aromatic compound (PAH) degradation, the surface tension and emulsification activities, and cell surface hydrophobicity of five PAH-degrading yeast isolates were compared to Saccharomyces cerevisiae from cultures grown with glucose, hexadecane, or naphthalene as carbon sources. The cell surface hydrophobicity values for the five yeast strains were significantly higher than for S. cerevisiae for all culture conditions, although these were highest with hexadecane and naphthalene. Strains with higher hydrophobicity showed higher rates of naphthalene and phenanthrene degradation, indicating that increased cell hydrophobicity might be an important strategy in PAH degradation for the five strains. Emulsification activities increased for all five yeast strains with naphthalene culturing, although no relationship existed between emulsification activity and PAH degradation rate. Surface tensions were not markedly reduced with naphthalene culturing.  相似文献

To investigate the changes in the morphology and polysaccharide content of Microcystis aeruginosa (Kütz.) Kütz. during flagellate grazing, cultures of M. aeruginosa were exposed to grazing Ochromonas sp. for a period of 9 d under controlled laboratory conditions. M. aeruginosa responded actively to flagellate grazing and formed colonies, most of which were made up of several or dozens of cells, suggesting that flagellate grazing may be one of the biotic factors responsible for colony formation in M. aeruginosa. When colonies were formed, the cell surface ultrastructure changed, and the polysaccharide layer on the surface of the cell wall became thicker. This change indicated that synthesis and secretion of extracellular polysaccharide (EPS) of M. aeruginosa cells increased under flagellate grazing pressure. The contents of soluble extracellular polysaccharide (sEPS), bound extracellular polysaccharide (bEPS), and total polysaccharide (TPS) in colonial cells of M. aeruginosa increased significantly compared with those in single cells. This finding suggested that the increased amount of EPS on the cell surface may play a role in keeping M. aeruginosa cells together to form colonies.  相似文献

Studies of the phytoplankton ecology in different localities in north-Norwegian fjords, the White Sea and the Barents Sea were carried out in spring and early summer to investigate the contribution of single and colonial stages of Phaeocystis pouchetii to phytoplankton abundance. Three different types of flagellated and four colonial cells were observed in all localities. P. pouchetii was rare under the ice of the Barents and White Seas, but their abundance increased rapidly during ice retreat. Single cell C dominated over colonial cell C, often by 50 times or more. The highest share of colonial cells was encountered in April in northern Norwegian fjords, in May in the Barents Sea and in May–June in the White Sea. At times the single cell dominated the total P. pouchetii biomass in Balsfjord (April 1999, 2001) with hardly any colonies present. In the White Sea colonies of P. pouchetii were less abundant than in the other regions. Cell carbon of P. pouchetii colonies appears never to be as dominating in the north-eastern North Atlantic as P. globosa blooms in coastal regions such as the southern North Sea. However, the lobal matrix of P. pouchetii colonies appears to be less solid than that of P. globosa and partly dissolution of the colony matrix during handling and storage of fixes samples induces uncertainty about the absolute numbers of P. pouchetii colonial cell counts. Despite of that, single cells of P. pouchetii seem to dominate significantly over colonial cell biomass at most sites and during some years and in some regions colonial cells seem rare. We speculate that top-down regulation of Phaeocystis spp. blooms possibly determines the ratio between single and colonial cells.  相似文献

Summary The relationship between the annual colony cycle and seasonal patterns of forage availability was investigated for the African honey bee,Apis mellifera scutellata, in the Okavango River Delta, Botswana. The annual cycle occurred in three distinct periods. The swarming season occurred from October-November, following two to three months of intense brood production, and coincided with the end of peak forage abundance. The migration season occurred from November-May and coincided with reduced and variable floral resources. During the migration season, brood production and food storage were generally low but quite variable from month to month, and swarms passing over the study area at this time traveled in an easterly direction. The migration season was followed by the establishment period (June-September), in which large numbers of new colonies traveling from the west moved into the study area. The establishment period coincided with, and slightly preceded, the period of peak forage abundance, and colonies devoted resources collected at this time almost entirely to brood rearing, which culminated in swarm production. The data suggest that honey bee colonies in the Okavango are mobile and gear their reproduction and movement to seasonally shifting resource pattern.  相似文献

Isolates of alder Phytophthora were collected in the southern part of Belgium on riverbanks planted with Alnus glutinosa and A. incana. They were compared with strains isolated in other European countries in terms of maximum temperature for growth, oogonia shape, pathogenicity on Alnus seedlings and genetic traits. Using both molecular techniques [random amplified polymorphic DNA (RAPD) and random amplified microsatellite (RAMS)], two groups of isolates were identified, the first group being further divided into two subgroups, Ia and Ib, using RAPD. Most of the Walloon alder Phytophthora isolates as well as the standard type from UK (formally designated P. alni subsp. alni) fell into group Ia. One isolate was classified in group Ib with the German and Dutch variants (P. alni subsp. multiformis), while three isolates were placed with the Swedish variant (P. alni subsp. uniformis) in group II. In terms of morphological properties, isolates from groups Ia and Ib developed colonies with a felt‐like appearance and usually produced numerous oogonia, varying from wavy to warty after 1 week (group Ia) or 2–3 weeks (Ib) in darkness. In contrast, colonies from group II isolates were generally irregular, and smooth oogonia were produced in low quantities after approximately 1 month in culture. A polymerase chain reaction (PCR) using sequence‐characterized amplification region (SCAR) primers derived from a polymorphic amplification product generated with a RAPD primer was developed for the specific detection of alder Phytophthora. Abstract

Daily counts were made of spores trapped on microscope slides coated with petroleum jelly and placed on the ground at distances up to 200 em from isolated colonies of Atrichum undulatum and Bryum argenteum. The observations continued for 30–34 days during the annual period of spore release in these species. Estimates of the number of spores released by the colonies were obtained as the product of the numbers of capsules and the difference between the mean numbers of spores present in undehisced capsules at the beginning of the experiment and remaining in dehisced capsules at the end.

Spore deposition over the experimental periods showed a steep gradient, deposition per unit area falling from 4740—14, 230 spores cm?2 in the centre of the colonies to less than 10 spores cm?2 200 cm from the edge of the colonies. Despite the steep deposition gradients, however, it was estimated that more than 85% of the spores in A. undulatum, and more than 95% of those in B. argenteum, were dispersed to unknown distances beyond the trapping areas.  相似文献

The effectiveness of two commonly used preservatives, sodium benzoate and potassium disulfite, was evaluated in terms of their bactericidal activity and capacity to induce changes in the surface properties ofEscherichia coli isolated from commercial food preserves. Preservative treatment over a five-week test period resulted in controlling the multiplication of these organisms and causing a decline in cell-surface hydrophobicity, hemagglutinating ability and adherence capacity to rat intestinal cells ofE. coli isolates. Forty-four enterococcal strains isolated from human clinical specimens were investigated for binding of 125I-labeled fibronectin, vitronectin, thrombospondin, lactoferrin, and collagen type I and IV, and for cell surface hydrophobicity. Most strains expressed low binding of iodine-labeled human fibronectin, collagen I and IV, and higher binding of human vitronectin, human lactoferrin, and human thrombospondin. Bacteria grown in Todd-Hewitt broth exhibited increased binding to vitronectin and thrombospondin. In particle agglutination assays (PAA), Enterococcus faecalis strains reacted strongly with coated latex beads in contrast to E. faecium strains, which generally did not react. The ability of enterococci to bind ECM proteins was affected by heating and proteolytic digestion, suggesting that some protein-binding components become surface exposed after treatment with proteases. The binding of 125I-labeled proteins to E. faecalis strain E70 was inhibited when cells were preincubated with unlabeled proteins. Preincubating cells with sulfated polymers such as dextran sulfate (M r 5000 and 8000), pentosan sulfate and heparin decreased binding of vitronectin, lactoferrin, and thrombospondin. The binding of lactoferrin and thrombospondin was also decreased when bacteria were preincubated with galactose, fucose, and mannosamine, but not with mannose. All of 30 E. faecalis strains expressed pronounced surface hydrophobicity, but 10 of 14 E. faecium strains showed hydrophilic cell surface. Received: 22 April 1996 / Accepted: 29 June 1996  相似文献

Aims:  To identify lactic acid bacteria (LAB) colonies isolated from sediments of a coastal marsh by the reduction of 2,3,5‐triphenyltetrazolium chloride (TTC) in MRS medium. Methods and Results:  Single colonies isolated from sediments of a coastal marsh by enrichment in MRS broth were selected from MRS‐TTC plates and classified according to colony phenotype based on TTC reduction. A total of 37 colonies grouped in seven different phenotypes were identified by analysis of its 16S ribosomal gene sequence. Most isolates belonged to the Firmicutes phylum, mainly to orders Bacillales and Lactobacillales. LAB were represented by 20 isolates, 15 of which belong to the genus Weissella. Conclusions:  Enrichment in MRS was highly selective for the isolation of bacteria belonging to phylum Firmicutes. Several different phenotypes were developed by LAB and must be considered during LAB isolation based on TTC reduction. Significance and Impact of the Study:  To our knowledge, this is the first study aimed at determining a relationship between colony phenotype from TTC reduction and a partial identification of isolates based on 16S ribosomal gene sequence similarities. Besides, this is the first report of isolation of W. cibaria from environmental samples.  相似文献
