【目的】有关Microbacterium maritypicum在植物线虫生物防治方面的研究较少,探究菌株M. maritypicum Sneb159的杀线虫活性,明确菌株发酵液中具有杀线虫活性的物质,为生物农药的开发提供理论依据。【方法】本研究检测了菌株Sneb159发酵液对大豆胞囊线虫二龄幼虫的触杀活性;并以触杀活性为追踪,采用有机溶剂萃取、硅胶柱层析及半制备高效液相色谱等技术对菌株Sneb159发酵滤液中的活性物质进行分离纯化;采用核磁共振波谱对纯化物进行结构鉴定。【结果】菌株Sneb159具有杀线虫活性,在24 h和48 h处理组中线虫的死亡率均显著高于对照组。从菌株Sneb159发酵滤液中分离得到杀线虫活性物质A6,经结构鉴定确定该物质为苯乙酰胺。【结论】首次发现M. maritypicum对大豆胞囊线虫的触杀作用,并且明确活性物质为苯乙酰胺。结果表明菌株M. maritypicum Sneb159和苯乙酰胺在大豆胞囊线虫的生物防治方面具有较好的应用潜力。  相似文献

Abstract

A simple strategy for designing a cancer immunotherapeutic system involves modification of tumor cells from tumor-bearing animals in vivo in such a way that the host can evoke a specific immune response against them. We have expressed allogeneic class I major histocompatibility complex (MHC) molecules on tumor cells, through ex vivo DNA-mediated gene transfer. These molecules are potent immuno-modulators for the stimulation of strong immune reactions against certain malignancies. In order to achieve efficient gene delivery to tumor cells in vivo we have compared the efficiencies of gene transfer into mammalian tumor cells by the biolistic particle delivery system and cationic liposomes. In this report, we have demonstrated that cationic liposomes prepared by DC-chol and DOPE gives the best efficiency of transfection for tumor cells in vivo. We also showed that a strong anti-H-2Kb allo-reactive cytotoxic T lymphocyte (CTL) response could be generated following in vivo immunization of AKR/J mouse spleens with the H-2Kb gene and DC-chol cationic liposomes. The direct immunization of mouse spleens to induce cell-mediated immunity against exogenous antigens may allow alternative treatment strategies for cancer immunotherapy.  相似文献

A new lumped model of flow driven by pumping without valves is presented, motivated by biomedical applications: the circulation of the human fetus before the development of the heart valves and mechanism of blood flow during the external cardiopulmonary resuscitation (CPR). The phenomenon of existence of a unidirectional net flow around a loop of tubing that consists of two different compliances is called valveless pumping. The lumped parameter model of valveless pumping in this paper is governed by the ordinary differential equations for pressure and flow, with time-dependent compliance, resistance, and inertia. This simple model can represent the essential features of valveless pumping we observed in earlier mathematical models and physical experiments of valveless pumping. We demonstrate that not only parameters of the driving function, such as frequency or amplitude, but also physical parameters, such as wall thickness and tube stiffness, are important in determining the direction and magnitude of a net flow. In this system, we report two new and interesting phenomena of valveless pumping: One is that the shifted peak frequency can be predicted by the pulsewave speed and the other is that time-dependent resistance is a crucial factor in generating valveless pumping. We also demonstrate that this lumped model can be extended to a one-dimensional flow model of valveless pumping and explain why a linear case, the case of the constant compliance, resistance, and inertia, generates almost zero net flow. This emphasizes that the nonlinearity of valveless pumping is also an important factor to generate a net flow in a closed loop model of valveless pumping.  相似文献

ABSTRACT

Background: Mechanisms affecting invasiveness of non-indigenous species have received much attention. Few studies have investigated invasions by native plants. Invasive native species such as common ragwort (Jacobaea vulgaris) may become noxious weeds. They challenge farming and nature conservation by outcompeting fodder plants or rare herbs. One mechanism that can result in outcompeting plants is by allelopathy.

Aim: We evaluated the potential of J. vulgaris to suppress germination in common and rare grassland species by allelopathy.

Methods: In a germination chamber experiment, we exposed 22 species and J. vulgaris itself to ragwort leachate. We controlled for osmotic effects by germination tests in mannitol solution and water. We assessed germination percentage—time —synchrony and radicle length.

Results: Leachate reduced germination percentage, germination time, synchrony and radicle length. These effects were similar for J. vulgaris, indicating autotoxicity. Rare species germinated less than common species but were not more sensitive to the phytotoxic effects of J. vulgaris. Restraining effects of the leachate were similar to the mere osmotic effect.

Conclusion: Our results question allelopathy as the main driving mechanism behind J. vulgaris gaining dominance. However, the impact of J. vulgaris might depend on the composition of the invaded plant community due to species-specific effects.  相似文献

Background: Long-term climate trends in mountain systems often vary strongly with elevation.

Aims: To evaluate elevation dependence in long-term precipitation trends in subalpine forest and alpine tundra zones of a mid-continental, mid-latitude North American mountain system and to relate such dependence to atmospheric circulation patterns.

Methods: We contrasted 59-year (1952–2010) precipitation records of two high-elevation climate stations on Niwot Ridge, Colorado Front Range, Rocky Mountains, USA. The sites, one in forest (3022 m a.s.l.) and the other in alpine tundra (3739 m), are closely located (within 7 km horizontally, ca. 700 m vertically), but differ with respect to proximity to the mountain-system crest (the Continental Divide).

Results: The sites exhibited significant differences in annual and seasonal precipitation trends, which depended strongly on their elevation and distance from the Continental Divide. Annual precipitation increased by 60 mm (+6%) per decade at the alpine site, with no significant change at the subalpine site. Seasonally, trends at the alpine site were dominated by increases in winter, which we suggest resulted from an increase in orographically generated precipitation over the Divide, driven by upper-air (700 hPa) north-westerly flow. Such a change was not evident at the subalpine site, which is less affected by orographic precipitation on north-westerly flow.

Conclusions: Elevation dependence in precipitation trends appears to have arisen from a change in upper-air flow from predominantly south-westerly to north-westerly. Dependence of precipitation trends on topographic position and season has complex implications for the ecology and hydrology of Niwot Ridge and adjacent watersheds, involving interactions among physical processes (e.g. snowpack dynamics) and biotic responses (e.g. in phenologies and ecosystem productivity).  相似文献

Aplysina aerophoba, Nardo 1886 is a common and well examined demosponge in the Mediterranean Sea. For sponges, as sessile, inner filter feeders, the most important function is pumping water through their canal system. This flow through is actively generated, supplies the sponge with oxygen and food particles and washes out waste products. Oxygen measurements ex situ, as a given example, show high oxygen saturations inside pumping sponges and oxygen depletion in non-pumping sponges. Thus, the oxygen situation within the sponge, like the food particle supply, waste washout and others, are directly related to its active pumping. To learn more about the poriferan function, it is important to know more about its undisturbed in situ pumping activity and consequently about the correlated conditions inside the sponge body.We conducted a tracer experiment in situ and tested sponge activity in terms of active pumping. Our technique excluded stress and disturbance to the sponges, hence minimizing experimental artefacts. The results show Aplysina aerophoba to be permanently pumping, which is implying a permanent supply of oxygen and of course food particles as well as a permanent washout of waste products and a permanent, presumably high energy consumption.We therefore conclude, that Aplysina aerophoba is always well supplied with oxygen, and that tissue anoxia or anaerobic metabolisms are of no significant importance in this sponge species. This fact of a permanent flow through in sponges will have to be taken into account for past and future hypotheses on the physiology of the sponge-microbial systems.  相似文献

Genus Jatropha with 172 species having significant economic importance belongs to the family Euphorbiaceae. There are no reports on molecular characterization and phylogenetic relationship among the species of Jatropha. Hence, the present study was undertaken to assess the extent of genetic variability that exist and also to establish phylogenetic relationship among Jatropha curcas, J. glandulifera, J. gossypifolia, J. integerrima, J. multifida, J. podagrica and J. tanjorensis using RAPD and AFLP. The percentage of loci that are polymorphic among the species studied was found to be 97.74% by RAPD and 97.25% by AFLP. The mean percentage of polymorphism (PP) was found to be 68.48 by RAPD and 71.33 by AFLP. The phylogram generated with RAPD and AFLP data showed maximum similarity. With the generated data maximum relatedness was found between J. curcas and J. integerrima this may be the reason for the success of inter hybrid crosses between these two species. Neither RAPD nor AFLP data generated in this study supports the view of J. tanjorensis, a natural interspecific hybrid between J. curcas and J. gossypifolia. The present study concludes that both RAPD and AFLP techniques are comparable in divergence studies of Jatropha species. The markers generated by RAPD and AFLP can be employed efficiently for interspecific hybrids identification, marker assisted selection and genetic resource management.  相似文献

The cytochromebc 1 complex was isolated from yeast mitochondria solubilized with the detergent dodecyl maltoside and reconstituted into proteoliposomes to measure electrogenic proton pumping. Optimal respiratory control ratios of 4.0, obtained after addition of the uncoupler CCCP, and H+/e ratios of 1.6 were obtained when the proteoliposomes were prepared with egg yolk phosphatidylcholine supplemented with cardiolipin. Moreover, it was critical to remove excess dodecyl maltoside in the final concentrated preparation prior to reconstitution to prevent loss of enzymatic activity. The rate of electrogenic proton pumping, the respiratory control ratios, and the H+/e ratios were decreased by incubation of the cytochromebc 1 complex with dicyclohexylcarbodiimide (DCCD) in a time and concentration dependent manner. Maximum inhibitions were observed when 50 nmol DCCD per nmol of cytochromeb were incubated for 30 min at 12°C with the intact cytochromebc 1 complex. Under these same conditions maximum labeling of cytochromeb with [14C] DCCD was reported in a previous study [Beattieet al. (1984).J. Biol. Chem. 259, 10562–10532] consistent with a role for cytochromeb in electrogenic proton movements.  相似文献

Summary The plasma membrane (Mg2+)-dependent adenosine triphosphatase ((Mg2+)-ATPase) from human erythrocytes has been tested for its ability to transport ions. Using a preparation of inside-out vesicles loaded with the pH-sensitive fluorescence probe 1-hydroxypyrene-3,6,8-trisulfonic acid (HPTS), we have demonstrated the absence of proton movement during (Mg2+)-ATPase activity. From the rate of ATP hydrolysis and the passive proton permeability of these vesicles, an upper limit of 0.03 H+ transported per ATP hydrolyzed was calculated. To verify that proton pumping could be detected in this system, the intravesicular pH was monitored during (Ca2+)-dependent adenosine triphosphatase ((Ca2+)-ATPase) activity. Proton efflux associated with (Ca2+)-ATPase activity was observed (in agreement with a recent report of proton pumping by a reconstituted erythrocyte (Ca2+)-ATPase (Niggli, V., Sigel, E., Carafoli, E. (1982)J. Biol. Chem. 257:2350–2356)) and was shown to be stimulated by calmodulin. The ability of the (Mg2+)-ATPase to pump28Mg2+,35SO 4 2– and86Rb+ was also tested, with the results leading to the conclusion that the human erythrocyte enzyme does not function as an ion transport system.  相似文献

Abstract

Associated microorganisms have been described in numerous marine sponges. Their metabolic activity, however, has not yet been investigated in situ. We quantified for the first time microbial processes in a living sponge. Sulfate reduction rates of up to 1200 nmol cm?3d?1 were measured in the cold-water bacteriosponge Geodia barretti . Oxygen profiles and chemical analysis of sponge tissue and canal water revealed steep oxygen gradients and a rapid turnover of oxygen and sulfide, dependent on the pumping activity of the sponge. Identification of the microbial community with fluorescently labelled oligonucleotide probes (FISH) indicates the presence of sulfate-reducing bacteria belonging to the Desulfoarculus/Desulfomonile/Syntrophus -cluster in the choanosome of this sponge. Analysis of lipid biomarkers indicates biomass transfer from associated sulfate-reducing bacteria or other anaerobic microbes to sponge cells. These results show the presence of an anoxic micro-ecosystem in the sponge G. barretti, and imply mutualistic interactions between sponge cells and anaerobic microbes. Understanding the importance of anaerobic processes within the sponge/microbe system may help to answer unsolved questions in sponge ecology and biotechnology.  相似文献

This paper investigates and compares experimentally determined water velocity field above natural macrozoobenthos burrows generated by Chironomus Plumosus larva during their bio-irrigation activity. All experiments were carried out using particle image velocimetry and performed in mesocosms filled with sediment burrowed by larvae, and the water velocity fields near the inlets and outlets of the U-shaped burrows were measured. From water velocity data the average volumetric flow rates between 54.6 and 61.1 mm3/s were calculated. Assuming an average burrow diameter of 2.25 mm, the volumetric flow rates suggest the average flow velocities through burrows during the pumping period between 13.7 and 15.4 mm/s. Two additional interesting phenomena could also be shown by analyzing the flow field generated by the larva. The analysis of the amount of tracers used for visualizations revealed that some of the tracer particles added to the water must have been consumed along their path from the inlet toward the outlet, hinting clearly to the so-called filter-feeding action of C. plumosus. The second phenomenon is due to the form of motion C. plumosus generates. By careful flow visualizations it was found that unlike other organisms such as Urechis caupo that use peristaltic body contractions, C. plumosus worms its body sinusoidally catapulting the fluid far into the overlying water body. This action is of ecological advantage for it avoids generating short oxygen circuits for their respiration and filter feeding.  相似文献

A flow cytometry system was used to evaluate the impact of pulsed laser irradiations from an Nd:YAG laser on two marine coastal water diatoms, Chaetoceros gracilis and Skeletonema costatum. Three flow speeds, i.e. 9, 18 and 27 ml minm 1 and three laser fluences, i.e. 0.025, 0.05 and 0.1 J cmm 2 pulsem 1 were tested during this study. The reduction in cell density and chlorophyll a (chl a) concentrations were monitored by reference to non-irradiated samples as controls. Upon irradiation, the cell density and the chl a concentrations became reduced significantly compared to the control (one way ANOVA p <0.001 for the cell density in both the species and p <0.05 for chl a concentrations in both species). A maximum mortality of 0.77 log10 (about 83%) for C. gracilis and 0.68 log10 (about 78%) for S. costatum was observed at 9 ml minm 1 flow speed and 0.1 J cmm 2 laser fluence. The maximum reduction observed in the chl a concentration was about 26% (control 0.413 and sample 0.306 mg mlm 1) for C. gracilis and 27% (control 0.222 and sample 0.16 mg mlm 1) for S. costatum, when the flow rate was 9 ml minm 1 and the fluence 0.1 J cmm 2. In general, mortality increased with an increase in the laser fluence. The results thus show if the cooling water is laser-irradiated to mitigate biofouling, this could result in significant damage to the planktonic flora of the flowing seawater system, which in turn might reduce algal biofilm formation on industrially important structures. The reduction in the chl a concentration showed that the laser irradiations also could result in a significant reduction in the primary productivity of the cooling water.  相似文献

Abstract

An interactive computer system using a dot matrix approach has been developed and used to determine potentially significant features due to distortions in the B-DNA helix as a result of variations of purine and pyrimidine patterns. Sequences were compared using matrices which were generated using the Calladine-Dickerson rules (C.R. Calladine, J. Mol. Biol. 161, 343–352, 1982 and R.E. Dickerson, J. Mol. Biol. 766, 419–441, 1983). Having control over various parameters to enhance different aspects of the visual appearance of these matrices was helpful in discovering patterns that were not known a priori. Specifically, it was found that a pattern of alternating doublets of purines and pyrimidines appear to exist in regulatory regions. This event is shown to be beyond probabilistic expectation.  相似文献

ABSTRACT

The aim of the study is to test the influence of in vivo magnetostimulation on the rheological properties of blood in neurological patients. Blood circulation in the body depends both on the mechanical properties of the circulatory system and on the physical and physicochemical properties of blood. The main factors influencing the rheological properties of blood are as follows: hematocrit, plasma viscosity, whole-blood viscosity, red cells aggregability, deformability, and the ability of red cells to orient in the flow. The blood samples were collected from neurological patients with pain. Blood samples were collected twice from each patient, that is, before the magnetostimulation and immediately after the therapy. For each blood sample, the hematocrit value was measured using the standard method. Plasma viscosity and whole-blood viscosity were measured by means of a rotary-oscillating rheometer Contraves LS40. Magnetic field was generated by the instrument Viofor JPS® and the magnetostimulation treatments were performed using M1P2 and M1P3 programs. The analysis of the results included estimation of the hematocrit value (Hct), plasma viscosity (ηp), whole-blood viscosity and rheological parameters of Quemada’s model: k0, k, γc. Plasma viscosity values were obtained from the shear rate dependence of shear stress using the linear regression method. The results obtained in the study suggest that the blood rheological properties change in accord with applied magnetostimulation program.  相似文献

Background: Hybridisation in Senecio/Jacobaea is frequent and of significance in speciation and evolution. Moreover, these genera are model systems for studying the evolution of pyrrolizidine alkaloids (PAs). Hybrids between Jacobaea aquatica and J. vulgaris have been reported from north-western Europe.

Aims: Our aim was to determine whether hybrids between Jacobaea aquatica and J. vulgaris also occurred in Austria, and if these hybrids could be responsible for the presumed increase of J. aquatica-like plants in this part of Europe. Furthermore, we tested if such hybrids showed an altered PA spectrum compared with parental species.

Methods: We used amplified fragment length polymorphisms (AFLPs) to analyse genetic diversity and structure, and gas chromatography–mass spectrometry (GC–MS) to identify PAs.

Results: We were able to identify genetically one hybrid population adjacent to a large J. aquatica population. The AFLP phenotypes of hybrids demonstrated a closer relationship to J. aquatica, suggesting frequent backcrossing. All hybrid individuals contained the same set of PAs as the parental species, but in differing amounts, plus four additional PAs, one of which may be novel.

Conclusions: Although hybridisation in Jacobaea is common, we were able to confirm only one hybrid population in Austria. Therefore, it seems unlikely that hybridisation plays a role in the presumed increase of J. aquatica in this area.  相似文献

BackgroundSkin fibrosis is a significant medical problem that leads to a functional, aesthetic, and psychosocial impact on quality-of-life. Light-emitting diode-generated 633-nm red light (LED-RL) is part of the visible light spectrum that is not known to cause DNA damage and is considered a safe, non-invasive, inexpensive, and portable potential alternative to ultraviolet phototherapy that may change the treatment paradigm of fibrotic skin disease.ObjectiveThe goal of our study was to investigate the how reactive oxygen species (ROS) free radicals generated by high fluence LED-RL inhibit the migration of skin fibroblasts, the main cell type involved in skin fibrosis. Fibroblast migration speed is increased in skin fibrosis, and we studied cellular migration speed of cultured human skin fibroblasts as a surrogate measure of high fluence LED-RL effect on fibroblast function. To ascertain the inhibitory role of LED-RL generated ROS on migration speed, we hypothesized that resveratrol, a potent antioxidant, could prevent the photoinhibitory effects of high fluence LED-RL on fibroblast migration speed.MethodsHigh fluence LED-RL generated ROS were measured by flow cytometry analysis using dihydrorhodamine (DHR). For purposes of comparison, we assessed the effects of ROS generated by hydrogen peroxide (H2O2) on fibroblast migration speed and the ability of resveratrol, a well known antioxidant, to prevent LED-RL and H2O2 generated ROS-associated changes in fibroblast migration speed. To determine whether resveratrol could prevent the high fluence LED-RL ROS-mediated photoinhibition of human skin fibroblast migration, treated cells were incubated with resveratrol at concentrations of 0.0001% and 0.001% for 24 hours, irradiated with high fluences LED-RL of 480, 640, and 800 J/cm2.ResultsHigh fluence LED-RL increases intracellular fibroblast ROS and decreases fibroblast migration speed. LED-RL at 480, 640 and 800 J/cm2 increased ROS levels to 132.8%, 151.0%, and 158.4% relative to matched controls, respectively. These LED-RL associated increases in ROS were prevented by pretreating cells with 0.0001% or 0.001% resveratrol. Next, we quantified the effect of hydrogen peroxide (H2O2)-associated ROS on fibroblast migration speed, and found that while H2O2–associated ROS significantly decreased relative fibroblast migration speed, pretreatment with 0.0001% or 0.001% resveratrol significantly prevented the decreases in migration speed. Furthermore, we found that LED-RL at 480, 640 and 800 J/cm2 decreased fibroblast migration speed to 83.0%, 74.4%, and 68.6% relative to matched controls, respectively. We hypothesized that these decreases in fibroblast migration speed were due to associated increases in ROS generation. Pretreatment with 0.0001% and 0.001% resveratrol prevented the LED-RL associated decreases in migration speed.ConclusionHigh fluence LED-RL increases ROS and is associated with decreased fibroblast migration speed. We provide mechanistic support that the decreased migration speed associated with high fluence LED-RL is mediated by ROS, by demonstrating that resveratrol prevents high fluence LED-RL associated migration speed change. These data lend support to an increasing scientific body of evidence that high fluence LED-RL has anti-fibrotic properties. We hypothesize that our findings may result in a greater understanding of the fundamental mechanisms underlying visible light interaction with skin and we anticipate clinicians and other researchers may utilize these pathways for patient benefit.  相似文献

The lymphatic system is an open-ended network of vessels that run in parallel to the blood circulation system. These vessels are present in almost all of the tissues of the body to remove excess fluid. Similar to blood vessels, lymphatic vessels are found in branched arrangements. Due to the complexity of experiments on lymphatic networks and the difficulty to control the important functional parameters in these setups, computational modeling becomes an effective and essential means of understanding lymphatic network pumping dynamics. Here we aimed to determine the effect of pumping coordination in branched network structures on the regulation of lymph flow. Lymphatic vessel networks were created by building upon our previous lumped-parameter model of lymphangions in series. In our network model, each vessel is itself divided into multiple lymphangions by lymphatic valves that help maintain forward flow. Vessel junctions are modeled by equating the pressures and balancing mass flows. Our results demonstrated that a 1.5 s rest-period between contractions optimizes the flow rate. A time delay between contractions of lymphangions at the junction of branches provided an advantage over synchronous pumping, but additional time delays within individual vessels only increased the flow rate for adverse pressure differences greater than 10.5 cmH2O. Additionally, we quantified the pumping capability of the system under increasing levels of steady transmural pressure and outflow pressure for different network sizes. We observed that peak flow rates normally occurred under transmural pressures between 2 to 4 cmH2O (for multiple pressure differences and network sizes). Networks with 10 lymphangions per vessel had the highest pumping capability under a wide range of adverse pressure differences. For favorable pressure differences, pumping was more efficient with fewer lymphangions. These findings are valuable for translating experimental measurements from the single lymphangion level to tissue and organ scales.  相似文献

We earlier described CBS-pyrophosphatase of Moorella thermoacetica (mtCBS-PPase) as a novel phosphohydrolase that acquired a pair of nucleotide-binding CBS domains during evolution, thus endowing the protein with the capacity to be allosterically regulated by adenine nucleotides (J?msen, J., Tuominen, H., Salminen, A., Belogurov, G. A., Magretova, N. N., Baykov, A. A., and Lahti, R. (2007) Biochem. J., 408, 327–333). We herein describe a more evolved type of CBS-pyrophosphatase from Clostridium perfringens (cpCBS-PPase) that additionally contains a DRTGG domain between the two CBS domains in the regulatory part. cpCBS-PPase retained the ability of mtCBS-PPase to be inhibited by micromolar concentrations of AMP and ADP and activated by ATP and was additionally activated by diadenosine polyphosphates (AP n A) with n > 2. Stopped-flow measurements using a fluorescent nucleotide analog, 2′(3′)-O-(N-methylanthranoyl)-AMP, revealed that cpCBS-PPase interconverts through two different conformations with transit times on the millisecond scale upon nucleotide binding. The results suggest that the presence of the DRTGG domain affords greater flexibility to the regulatory part, allowing it to more rapidly undergo conformational changes in response to binding.  相似文献
